The impact of ageing on the body and its association with skin harm and frailty in relation to multimorbidity, comorbidity and polypharmacy is clearly described in the literature. The ageing population globally brings with it the challenge for healthcare professionals of managing individuals with increasingly complex and inter-related needs. This article considers ageing skin and skin tears within the context of frailty as a syndrome. It discusses what frailty is understood to be, how to recognise and assess for it, and how to consider risk and prevention of harm to the skin of the ageing person with frailty. Aging leads to biochemical and biomechanical changes in skin, with biological and functional consequences. Despite extensive literature on skin aging, there is a lack of studies which investigate the maturation of the tissue and connect the microscopic changes in the skin to its macroscopic biomechanical behavior as it evolves over time. The present work addresses this knowledge gap using multiscale characterization of skin in a murine model considering newborn, adult and aged mice. Monotonic uniaxial loading, tension relaxation with change of bath, and loading to failure tests were performed on murine skin samples from different age groups, complemented by inflation experiments and atomic force microscopy indentation measurements. In parallel, skin samples were characterized using histological and biochemical techniques to assess tissue morphology, collagen organization, as well as collagen content and cross-linking. We show that 1-week-old skin differs across nearly all measured parameters from adult skin, showing reduced strain stiffening and tensile strength, a thinner dermis, lower collagen content and altered crosslinking patterns. Surprisingly, adult and aged skin were similar across most biomechanical parameters in the physiologic loading range, while aged skin had lower tensile strength and lower stiffening behavior at large force values. This correlates with altered collagen content and cross-links. Based on a computational model, differences in mechanocoupled stimuli in the skin of the different age groups were calculated, pointing to a potential biological significance of the age-induced biomechanical changes in regulating the local biophysical environment of dermal cells. Skin microstructure and the emerging mechanical properties change with age, leading to biological, functional and health-related consequences. Despite extensive literature on skin aging, only very limited quantitative data are available on microstructural changes and the corresponding macroscopic biomechanical behavior as they evolve over time. This work provides a wide-range multiscale mechanical characterization of skin of newborn, adult and aged mice, and quantifies microstructural correlations in tissue morphology, collagen content, organization and cross-linking. Remarkably, aged skin retained normal hydration and normal biomechanical function in the physiological loading range but showed significantly reduced properties at super-physiological loading. Background: Various techniques have been employed in aesthetic medicine to combat skin aging, in particular that of the facial region. Hyaluronic acid is utilized to enhance moisture levels and extracellular matrix molecules. This study aims to histologically assess the effects of low molecular weight hyaluronic acid fragments combined with amino acids (HAAM) on facial skin rejuvenation through intradermal microinjections. Methods: A total of twenty women, with an average age of 45 and ranging from 35 to 64 years old, participated in the study, including 8 in menopause and 12 in the childbearing age group. Mesotherapy was used to administer HAAM to the patients. Prior to and three months after the treatment, each patient underwent small circular punch biopsies. Ultrasound examinations were conducted using B-mode, capturing 2D images in longitudinal or transverse orientations with frequencies ranging from 5 to 13 Mega-hertz (MY LAB X8, ESAOTE, Genova, Italy). A total of 60 ultrasound examinations were taken, with 30 collected before treatment and 30 after treatment. Results: The histological analysis demonstrates an increase in fibroblast activity resulting in the production of Type III reticular collagen, as well as an increased number of blood vessels and epidermal thickness. However, the analysis of ultrasound data before and after treatment showed no statistical difference in skin thickness in malar area, chin and mandibular angle. Background: Changes develop on the facial skin as a person ages. Other than chronological time, it has been discovered that gender, ethnicity, air pollution, smoking, nutrition, and sun exposure are notable risk factors that influence the development of skin ageing phenotypes such as wrinkles and photo-ageing. These risk factors can be quantified through epidemiological collection methods. We previously studied wrinkles and photo-ageing in detail using photo-numeric scales. The analysis was performed on the ethnic Chinese skin by three trained assessors. Recent studies have shown that it is possible to use self-reported data to identify skin-related changes including skin colour and skin cancer. In order to investigate the association between risk factors and skin ageing phenotypic outcomes in large-scale epidemiological studies, it would be useful to evaluate whether it is also possible for participants to self-report signs of ageing on their skin. Aim: We have previously identified several validated photo-numeric scales for wrinkling and photo-ageing to use on ethnic Chinese skin. Using these scales, our trained assessors grade wrinkling and photo-ageing with moderately high inter-assessor concordance and agreement. The main objective of this study involves letting participants grade self-reported wrinkling and photo-ageing using these same scales. We aim to compare the concordance and agreement between signs of skin ageing by the participant and signs of ageing identified by our assessors. Method: Three trained assessors studied facial photo-ageing on 1081 ethnic Chinese young adults from the Singapore/Malaysia Cross-sectional Genetics Epidemiology Study (SMCGES) cohort. Self-reported facial photo-ageing data by the same 1081 participants were also collated and the two sets of data are compared. Results: Here, we found that self-reported signs of photo-ageing are concordant with photo-ageing detected by our assessors. This finding is consistent whether photo-ageing is evaluated through studying wrinkle variations (Spearman's rank correlation (ρ) value: 0.246–0.329) or through studying dyspigmentation patterns (Spearman's rank correlation (ρ) value 0.203–0.278). When studying individual wrinkles, both participants and assessors often detect the presence of the same wrinkle (Spearman's rank correlation (ρ) value 0.249–0.366). A weak-to-fair level of agreement between both participants and assessors (Cohen's kappa (κ) values: 0.041–0.233) persists and is statistically significant after accounting for agreements due to chance. Both the participant and the assessor are largely consistent in evaluating the extent of photo-ageing (area under curve (AUC) values 0.689–0.769) and in discerning between the presence or absence of a given facial wrinkle (area under curve (AUC) values 0.601–0.856). Conclusion: When we analyse the overall appearance of the face, our results show that signs of photo-ageing identified by the participant are concordant with signs of photo-ageing identified by our assessors. When we focused our analysis on specific areas of the face, we found that participants were more likely to identify and self-report the same wrinkles that our assessors have also detected. Here, we found that self-reported signs of skin ageing provide a satisfactory approximation to the signs of skin ageing identified by our assessors. The ability to use self-reported signs of skin ageing should also be evaluated on scales beyond the ones discussed in this study. Currently, there are not as many photo-numeric scales for quantifying dyspigmentation patterns as there are for quantifying wrinkle variations. Skin aging is of immense societal and, thus, scientific interest. Because mechanics play a critical role in skin's function, a plethora of studies have investigated age-induced changes in skin mechanics. Nonetheless, much remains to be learned about the mechanics of aging skin. This is especially true when considering sex as a biological variable. In our work, we set out to answer some of these questions using mice as a model system. Specifically, we combined mechanical testing, histology, collagen assays, and two-photon microscopy to identify age- and sex-dependent changes in skin mechanics and to relate them to structural, microstructural, and compositional factors. Our work revealed that skin stiffness, thickness, and collagen content all decreased with age and were sex dependent. Interestingly, sex differences in stiffness were age induced. We hope our findings not only further our fundamental understanding of skin aging but also highlight both age and sex as important variables when conducting studies on skin mechanics. Our work addresses the question, "How do sex and age affect the mechanics of skin?" Answering this question is of both scientific and societal importance. We do so in mice as a model system. Thereby, we hope to add clarity to a body of literature that appears divided on the effect of both factors. Background: The aging of the skin is considered a cumulative process that is classed as intrinsic or extrinsic. Environmental factors like sun exposure and air pollution are considered the main cause of extrinsic aging. Mainly, intrinsic aging reflects the genetic background and depends on time. Spirulina platensis unicellular blue–green algae have a variety of biological and nutritional activities because of their high level of nutrients. The current study aims to investigate the mechanism by which spirulina extract (SE) may act anti-aging in female albino rats. Spirulina extract (20 mg/mL) was injected subcutaneously before UVA irradiation (2.16 J/cm2), daily for 7 days. The epidermal thickness and the collagen fibers layer were stained utilizing hematoxylin and eosin and Masson. Results: SE induced significant improvements in the activities of antioxidants including superoxide dismutase and reduced glutathione, down-regulating in expressions of inflammatory cytokines: interleukin-1β and tumor necrosis factor-α and reverses excessive ROS levels. As well as , the recovery of collagen density and reduction in the production of matrix metalloproteinases were presented. Conclusions: The results found that spirulina extract may delay the signs of skin aging by enhancing collagen as well as antioxidant activities and inhibiting collagen degradation and inflammation. Skin and cellular aging are impacted by various toxins in our environment such as air pollution, water contamination, an increased prevalence of light-emitting diodes, electromagnetic frequencies, various yeast and other fungi, parasitic infections, and mold and heavy metal toxicity. Basic topical skin care is insufficient to adequately protect an individual’s integumentary system and other organs from these types of daily cellular stressors. These stressors impact the level of oxidative stress status (OSS). OSS is measurable through biomarker analysis of various body fluids including blood, saliva, urine, and breath. This poses a unique assessment challenge for aesthetic practitioners as the OSS of a patient impacts their overall aging process. Aesthetic practitioners observe the aging process through visual assessment of a patient’s skin quality, skin barrier function, and the presence of solar lentigines, erythema, edema, telangiectasia, loss of collagen and elastin, bone density, and redistribution of subcutaneous tissue. Mitigating a patient’s daily exposure to OS and its impacts on the skin, other organ tissues, and metabolism poses a unique challenge in medical aesthetic treatment planning. For this reason, the use of stem cells and exosomes is gaining popularity in aesthetic medicine. The purpose of this literature review is to highlight currently available research, applications, limitations, and mitigation strategies in reducing OSS on the integumentary system and aging processes. The collagen had been widely used as a promising source of functional food supplements for skin care. Here, we developed a novel animal-derived collagen that displayed multiple functions for protecting the human skin cells under UV irradiation. Different evaluations were performed to explore the protective effects of this collagen on human skin fibroblasts and keratinocytes. Specifically, we found that our collagen could induce the production of collagen I, elastin, and hyaluronic acid in fibroblasts and could also enhance the skin wound healing ability. Besides, it could elevate the expression of aquaporin-3 and cluster of differentiation 44 in keratinocytes. Moreover, this collagen had been demonstrated to alleviate the generation of reactive oxygen species and the malondialdehyde content in UVA-treated fibroblasts, as well as the secretion of inflammation factors in keratinocytes. These data indicated that the novel animal-derived collagen was a hopeful material for the comprehensive protection of the skin cells and the prevention of skin aging. [ABSTRACT FROM AUTHOR]

The article reviews the ageing changes of the midfacial and maxillary bones, the mandible, the overlaying soft tissues and the smile, and presents clinical guidelines aiming to rejuvenate older faces by means of orthodontic therapy. With regard to the ageing changes, the maxillary skeleton appears to rotate clockwise inferior to the orbit and becomes retrusive, and as a general pattern the midface contracts and deteriorates with age. Resorption below the mental foramen, reduction in alveolar height, loss of bone at the chin region, and relative increase in size and shape are signs of an aged mandible. Epidermal thinning and decrease in collagen in combination with the effect of gravity and various external factors contribute to the ageing of the skin. Atrophy of the superficial and deep fat, changes in ligamentous tissues and changes in muscle structure, position and tone, all contribute to the stigmata of the aged face. In the article, two late adulthood orthodontic cases are discussed as examples, and general guidelines for orthodontic management of the older face aiming at reversing the 'shrinkage' of the tissues by restoring the facial shape and tightening the soft tissue mask are described. The possible mechanisms explaining the changes observed on the faces of the clinical cases are also discussed. A properly planned and executed orthodontic intervention reversing changes from the inside-out before embarking on cosmetic surgery might have a synergistic effect multiplying the benefits for adult patients. Various studies have reported the antimicrobial, antifungal, and antioxidant effects of Asphodelus species. These species are used in the treatment of various skin-related issues such as acne, abscesses, psoriasis, burns, alopecia, wound healing, and eczema in folk medicine. The purpose of the present study is to determine the antioxidant, antityrosinase, anticollagenase and cytotoxic effects of Asphodelus aestivus and Asphodelus fistulosus as potential dermocosmetic agent, as well as the phenolic substance profiles of the species by Reversed phase high performance liquid chromatography (RP-HPLC). Antioxidant capacity of the species was determined by total phenolic and flavonoid content determination, cupric ion reducing power (CUPRAC), ferric ion reducing power (FRAP) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity assays. Enzyme inhibition studies were conducted spectrophotometrically. The cytotoxic activity was analyzed by the 3-(4, 5-dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide (MTT) assay. As a result, it was determined that the methanolic extracts obtained from the species showed strong antioxidant activity. Based on the CUPRAC and DPPH results, A. aestivus was found to be more effective while according to the FRAP results, A. fistulosus exhibited greater effectiveness. A. fistulosus was discovered to be abundant in phenolic substances, while A. aestivus was found to be richer in flavonoids. It has also been observed that the methanolic extracts have a high inhibitory effect on collagenase and tyrosinase enzymes. It is believed that the species can be used as a potential resource in the treatment of skin aging and hyperpigmentation problems in the dermocosmetic industry. It is thought that the data obtained in the study will lead to further studies. Air pollution, a global health concern, has been associated with adverse effects on human health. In particular, particulate matter (PM), which is a major contributor to air pollution, impacts various organ systems including the skins. In fact, PM has been suggested as a culprit for accelerating skin aging and pigmentation. In this study, using single-cell RNA sequencing, IL-24 was found to be highly upregulated among the differentially expressed genes commonly altered in keratinocytes and fibroblasts of ex vivo skins exposed to PM. It was verified that PM exposure triggered the expression of IL-24 in keratinocytes, which subsequently led to a decrease in type I procollagen expression and an increase in MMP1 expression in fibroblasts. Furthermore, long-term treatment of IL-24 induced cellular senescence in fibroblasts. Through high-throughput screening, we identified chemical compounds that inhibit the IL-24-STAT3 signaling pathway, with lovastatin being the chosen candidate. Lovastatin not only effectively reduced the expression of IL24 induced by PM in keratinocytes but also exhibited a capacity to restore the decrease in type I procollagen and the increase in MMP1 caused by IL-24 in fibroblasts. The deterioration of the skin is caused by dermatological disorders, environmental conditions, and aging processes. One incisive strategy for supervising the skin aging process is implementing healthy nutrition, preserving a balanced diet, and a good supply of food supplements. Here, we compared H-Pro-Hyp-OH peptide, hydrolyzed collagen, and an original mixture of six amino acids (we named 6aa)--including glycine, l -alanine, l -proline, l -valine, l -leucine, and l -lysine--effects on the production of extracellular matrix (ECM) components, particularly the elastin, fibronectin, collagen 1, and collagen 4. Treatment of BJ human skin fibroblasts with the 6aa mixture upregulated elastin, fibronectin, and collagen 1 gene expression, without affecting the expression of anti-reactive oxygen species enzymes. Moreover, the mammalian target of rapamycin (mTOR) signaling pathway seems to be involved, at least in part. There has been a noticeable increase in the number of patients who present to oral and maxillofacial surgery (OMFS) departments for treatment of post-operative complications of dermal fillers and botulinum toxin. These complications are more likely when the treatments are performed by untrained persons or self-administered by patients. Occasionally, these adverse outcomes cannot be avoided, even when products are administered by medically trained professionals. This review includes two cases and discusses some of the key issues surrounding these products so that patients can be informed of the benefits and risks with these types of non-surgical aesthetic procedures and, where possible, reduce the risk of complications occurring. CPD/Clinical Relevance: Appropriate knowledge of facial anatomy and different types of dermal fillers is necessary to reduce the risk of complications associated with injection of these products. One of the essential organs and protective barricades, the skin, needs to be taken care of early. Skin is affected by several intrinsic and extrinsic factors, and despite their morphological and pathological differences, they have many molecular similarities. As of today, various mechanisms and theories have been recommended for aging, such as cellular anility, reduced proliferative tendency, reduction in length of telomere, mutations in DNA, theory of free radical generation, and many others. In today's society, skin health is often considered an important indicator of health, which has led to an increased demand for anti-aging products. However, numerous conventional cosmetics and phytocompounds (curcumin, Vitamin E, resveratrol) utilized in anti-aging products have inimical physical and chemical attributes, including insufficient chemical stability and inadequate skin penetration bound their effectuality after topical administration. So recently, new novel nanotechnological approaches for preventing skin aging, such as liposomes, niosomes, solid lipid nanoparticles, transferosomes, ethosomes, nanostructured lipid carriers, and carbon nanotubes, are being used. The goal of this systematic review was to explore emerging perspectives on the role of skin microbiota in acne vulgaris, skin aging, and rosacea. We searched the literature for published clinical trials, randomized controlled trials, cross-sectional studies, and cohort studies, both experimental and observational, whose primary main purpose was to ascertain the associations between the skin microbiome and chronic skin disease, acne vulgaris, rosacea, and skin aging, using the Embase and PubMed databases. Fifty-one relevant published articles were identified for systematic review (up to December 2021). The possible roles of the skin microbiome in these skin diseases were explored to shed light on its development and to identify potential therapeutic targets for treatment. However, the mechanisms of microbial interaction in these diseases are still under-studied. The results of this evidence-based review suggest that it may be possible to develop individualized therapies targeting the pathogenic strains within the skin microbiome involved in these diseases. This alternative therapeutic approach, involving modifications of the microbiome, may form the basis of the next generation of treatment, known collectively as "ecobiological" anti-inflammatory therapies. ETT (Glu-Met-Phe-Gly-Thr-Ser-Ser-Glu-Thr) isolated from Isochrysis zhanjiangensis was an antioxidant nonapeptide with the highest active site on Met 2. In this study, the investigated the anti-skin aging effect of ETT on both keratinocytes and fibroblasts, took ultraviolet B (UVB)-induced human immortalized keratinocytes (HaCaT) and hydrogen peroxide (H 2 O 2)-induced human skin fibroblasts (BJ cells). The results showed the anti-photoaging effect of ETT on UVB-induced HaCaT cells by declining an increasing level of intracellular reactive oxygen species (ROS), activating antioxidant system via nuclear factor erythroid 2 (Nrf2)/ heme oxgenase-1 (HO-1) signaling pathway, promoting autophagy to enhancing mitochondrial membrane potential and inhibiting apoptosis. Moreover, ETT also against skin aging on H 2 O 2 -induced BJ cells by attenuating senescence and improving collagen generation via transforming growth factor β1 (TGF-β1)/Smad signaling pathway. In general, these results indicated ETT is potential to against skin aging by maintaining keratinocytes homeostasis, reducing apoptosis, attenuating fibroblasts senescence and enhancing content of collagen. Skin undergoes mechanical alterations due to changes in the composition and structure of the collagenous dermis with aging. Previous studies have conflicting findings, with both increased and decreased stiffness reported for aging skin. The underlying structure--function relationships that drive age-related changes are complex and difficult to study individually. One potential contributor to these variations is the accumulation of nonenzymatic crosslinks within collagen fibers, which affect dermal collagen remodeling and mechanical properties. Specifically, these crosslinks make individual fibers stiffer in their plastic loading region and lead to increased fragmentation of the collagenous network. To better understand the influence of these changes, we investigated the impact of nonenzymatic crosslink changes on the dermal microstructure using discrete fiber networks representative of the dermal microstructure. Our findings suggest that stiffening the plastic region of collagen's mechanical response has minimal effects on network-level stiffness and failure stresses. Conversely, simulating fragmentation through a loss of connectivity substantially reduces network stiffness and failure stress, while increasing stretch ratios at failure. [ABSTRACT FROM AUTHOR]

Prolonged exposure to sun radiation may result in harmful skin photoaging. Therefore, discovering novel anti-photoaging treatment modalities is critical. An active component isolated from Salvia miltiorrhiza (SM), Salvianolic acid B (Sal-B), is a robust antioxidant and anti-inflammatory agent. This investigation aimed to discover the therapeutic impact and pathways of salvianolic acid B for UVB-induced skin photoaging, an area that remains unexplored. We conducted in vitro experiments on human dermal fibroblasts (HDFs) exposed to UVB radiation, assessing cellular senescence, superoxide dismutase (SOD) activity, cell viability, proliferation, migration, levels of reactive oxygen species (ROS), and mitochondrial health. The potential mechanism of Sal-B was analyzed using RNA sequencing, with further validation through Western blotting, PCR, and nuclear factor erythroid 2-related factor 2 (NRF2) silencing methods. In vivo , a model of skin photoaging induced by UVB in nude mice was employed. The collagen fiber levels were assessed utilizing hematoxylin and eosin (H&E), Masson, and Sirus red staining. Additionally, NRF2 and related gene and protein expression levels were identified utilizing PCR and Western blotting. Sal-B was found to significantly counteract photoaging in UVB-exposed skin fibroblasts, reducing aging-related decline in fibroblast proliferation and an increase in apoptosis. It was observed that Sal-B aids in protecting mitochondria from excessive ROS production by promoting NRF2 nuclear translocation. NRF2 knockdown experiments established its necessity for Sal-B's anti-photoaging effects. The in vivo studies also verified Sal-B's anti-photoaging efficacy, surpassing that of tretinoin (Retino-A). This study aimed to investigate the impact of three winemaking techniques on the volatile compounds and sensory profile of La Mancha Chelva wines, including wine elaborated by traditional white winemaking process (CW), wine enzymatically treated (WE), wine from must with skin contact treatment (SCW) and wine aging on lees (WL). A total of 32 varietal aroma compounds and 44 volatile compounds formed principally during alcoholic fermentation were identified and quantified by GC-MS independently of the winemaking technique and vintage year. WE showed the highest contents of terpenes, C 13 -norisoprenoids and benzenic compounds, SCW displayed major concentrations of C 6 compounds whereas WL exhibited the major concentrations of the principal groups of volatile compounds formed mainly during alcoholic fermentation. All wines showed the same sequence of aromatic series, with the highest aroma contribution being those of the fruity, sweet and fatty series. The differences found in volatile compounds modified the sensory aroma profile of CW enhanced the main attributes. These results reveal that the winemaking process used had an important effect on the volatile composition of Chelva wines and can be used for optimizing the elaboration method to ensure wines in concordance with consumer preferences and that preserved the distinctive aromatic typicity of Cheva wines. To assess factors for canine skin extensibility, our study investigated associations between the dogs' skin extension index and the following factors, gender, age, neuter status, weight, coat color and six coat color related gene polymorphisms. Swab samples were collected from 69 toy poodles to extract DNA. The skin extension indices of the lower back and the neck were measured using the following formula: vertical height of the skin fold divided by body length multiplied by 100. The dogs' age, weight, gender, neuter status and coat color were also recorded, as well as polymorphisms of the following six selected coat color related genes, Melanocortin 1 receptor, Tyrosinase-related protein 1, Melanophilin, Canine β-defensin-1, Major Facilitator Superfamily Domain Containing 12 and Agouti-signaling protein (ASIP). Univariable analysis showed there was a meaningful association between the lower back skin extension index and both gender and age (P<0.001 and P=0.048, respectively). Also, there was a possible association between the lower back skin extension index and ASIP Single nucleotide polymorphism (SNP) (R96C) (P=0.078). Linear model analysis showed there was a significant association between the lower back skin extension index and gender (P<0.001), and there was a tendency of the association between the lower back skin extension index and ASIP SNP (R96C) (P=0.098). In addition, there was an association between gender and age for the skin extension index. (P=0.048). Therefore, these results suggest that a greater risk of skin extensibility in toy poodle could be related to being female and the ASIP SNP (R96C), because these factors were associated with higher lower back skin extension index. Sulfonylureas are widely used oral anti-diabetic drugs. However, its long-term usage effects on patients' lifespan remain controversial, with no reports of influence on animal longevity. Hence, the anti-aging effects of chlorpropamide along with glimepiride, glibenclamide, and tolbutamide were studied with special emphasis on the interaction of chlorpropamide with mitochondrial ATP-sensitive K+ (mitoK-ATP) channels and mitochondrial complex II. Chlorpropamide delayed aging in Caenorhabditis elegans , human lung fibroblast MRC-5 cells and reduced doxorubicin-induced senescence in both MRC-5 cells and mice. In addition, the mitochondrial membrane potential and ATP levels were significantly increased in chlorpropamide-treated worms, which is consistent with the function of its reported targets, mitoK-ATP channels. Increased levels of mitochondrial reactive oxygen species (mtROS) were observed in chlorpropamide-treated worms. Moreover, the lifespan extension by chlorpropamide required complex II and increased mtROS levels, indicating that chlorpropamide acts on complex II directly or indirectly via mitoK-ATP to increase the production of mtROS as a pro-longevity signal. This investigation studied the effect of concentrated growth factor and nanofat on aging skin of nude mice induced by D-galactose. BALB/c mice were randomly divided into five groups: 5 mice in the control group were fed normally without any intervention, 9 mice were treated with concentrated growth factor (CGF), 9 mice were treated with nanofat (NF), 9 mice were treated with CGF+NF, and 9 mice in the model group (no treatment after subcutaneous injection of D-galactose). Relevant indicators are measured and recorded. In skin and serum, SOD and GSH content in the model group were significantly lower than those in other groups (P<0.05), and the MDA of the three treatment groups was significantly lower than that of the model group (P<0.05). Compared with the control group, the contents of total collagen, type I collagen and type III collagen in the NF group and model group were decreased in different degrees (P<0.05); the contents of elastin and elastic fiber in the skin of nude mice in the model group and NF group were significantly decreased. Compared with the model group, he number of CD31 and VEGF in the treatment group was significantly increased (P<0.01); the skin AGE content of three treatment groups was significantly lower (P<0.05). The "gut-skin" axis has been proved and is considered as a novel therapy for the prevention of skin aging. The antioxidant efficacy of oligomannonic acid (MAOS) makes it an intriguing target for use to improve skin aging. The present study further explored whereby MAOS-mediated gut-skin axis balance prevented skin aging in mice. The data indicated the skin aging phenotypes, oxidative stress, skin mitochondrial dysfunction, and intestinal dysbiosis (especially the butyrate and HIF-1α levels decreased) in aging mice. Similarly, fecal microbiota transplantation (FMT) from aging mice rebuild the aging-like phenotypes. Further, we demonstrated MAOS-mediated colonic butyrate-HIF-1α axis homeostasis promoted the entry of butyrate into the skin, upregulated mitophagy level and ultimately improving skin aging via HDAC3/PHD/HIF-1α/mitophagy loop in skin of mice. Accurate segmentation of wrinkles from facial images forms the basis of wrinkle analysis and acts as an important process in skin aging evaluation. However, Wrinkles have high intraclass variability while shallow wrinkles more closely resemble the texture of the skin. Accurate segmentation of both deep wrinkles and shallow fine lines is therefore still a significant challenge. In this paper, we propose a deep learning-based automatic wrinkle segmentation model named Striped WriNet. Its central module is Striped Attention Module (SAM), which consists of Multi-Scale Striped Attention (MSA) and Global Striped Attention (GSA). The ability of the model to extract contextual information about deep folds and shallow fine lines is improved by involving the geometry of the folds to design multi-scale structures and attention mechanisms. In this paper, we conducted experiments on both the public and private datasets, where the images are finely labeled. For Accuracy (Acc), Dice Score (Dice), and Jaccard Similarity Index (JSI), the proposed method is superior to the current state-of-the-art methods. Ablation experiments and coupling tests are performed to demonstrate the effectiveness of each module and the decoupling of SAM. Finally, this paper innovatively proposes the wrinkle index as a computational method for wrinkle evaluation. The experiments were performed on 2000 wrinkle pictures, and the Pearson coefficient of wrinkle index and the expert clinical score was 0.9399, which proving the robustness of the proposed method in assessing wrinkles under different individuals. Both pro-inflammatory cytokines and ultraviolet (UV) radiation can cause skin aging by reducing collagen synthesis and enhancing the activity of matrix metalloproteinases (MMPs). Sargahydroquinoic acid (SHQA), a meroterpenoid purified from Sargassum macrocarpum , has demonstrated a range of pharmacological properties, including anti-senescence, anti-oxidant and anti-inflammatory. Despite these properties, the protective effect of SHQA against skin aging has not been thoroughly examined. In this study, we investigated the impact of SHQA on collagen and MMPs using human skin fibroblast culture Hs68 subjected to tumor necrosis factor (TNF)-α challenge or UVB irradiation. Our findings reveal that both TNF-α (20 ng/mL) and UVB (10 mJ/cm2) induced mRNA expression and extracellular activity of MMP-2 and MMP-9, which was significantly inhibited by SHQA. While SHQA reduced the MMP-1 mRNA level, its impact on MMP-1 protein levels, both intracellular and extracellular, was not significant. Furthermore, SHQA mitigated the TNF-α-induced reduction in collagen type I alpha 1 (COL1A1), whereas low-dose UVB did not significantly affect COL1A1 protein expression. The inhibition of Akt and p38 signaling pathways by SHQA is implicated in this process. In summary, our findings demonstrate that SHQA effectively attenuates skin aging induced by both TNF-α and low-dose UV irradiation in dermal fibroblasts, through attenuating MMP hyperactivation and maintaining the level of COL1A1. Objective: To determine the effect of Lentinula edodes extract on ultraviolet (UV) A and UVB-induced changes in matrix metalloproteinase (MMP) and type I procollagen expression using human immortalized HaCaT keratinocytes. Methods: Lentinula edodes ethanol extract (LEE) was obtained by extraction with 80% ethanol for 4 h at 80 °C. Effect of LEE on UVinduced alteration on the expression and production of MMPs and type I procollagen in keratinocytes was investigated using ELISA, RT-PCR, and Western blotting assay. To determine the underlying mechanism of LEE-mediated effects, mitogen-activated protein kinase (MAPK) and activator protein 1 signaling pathways were analysed by Western blotting assay. Results: LEE significantly inhibited the expression of MMP-1 and MMP-9 and increased the expression of type I procollagen in UVA and UVB-irradiated HaCaT keratinocytes. The phosphorylation levels of p38 were significantly inhibited by LEE whereas it did not affect c-Jun N-terminal kinase and extracellular signal-regulated kinase phosphorylation. Suppression of p38 phosphorylation was also accompanied by downregulation of UVA and UVB-induced increase in c-Fos. Conclusions: LEE effectively inhibits the expression of MMP-1 and MMP-9 and increases type I procollagen production through the p38 MAPK/c-Fos signaling pathway in UVA and UVB-irradiated HaCaT keratinocytes. Skin fragility caused by structural and functional deterioration is ubiquitous especially in older people, and it determines a significant impact on quality of life. Dermatoporosis is a relatively new term used to describe the chronic skin insufficiency and fragility syndrome marked by morphological and functional manifestations. It is much more than a merely cosmetic problem. Dermatoporosis' manifestations may determine an increased risk of morbidity and mortality. Severe stages of dermatoporosis may determine skin lacerations, deep dissecting hematomas, and skin necrosis. Most patients affected by dermatoporosis are between 70 and 90 years old, but first clinical signs may start at around 40–60 years. This aim of this study was to observe the effect of Yang Yan Qing E Wan (YYQEW) on senescent phenotypes and the expression of β-catenin and p16INK4a in the hydrogen peroxide (H 2 O 2)-induced premature senescence of normal human skin fibroblasts (NHSFs). Primary normal human skin fibroblasts were randomly divided into a normal group, a blank group, a model group, and a YYQEW group. The cells of the model group and the YYQEW group were exposed to 150 μmol/L H 2 O 2 for 2 h. The morphological changes of the cells were analyzed by microscopy and by kits used to estimate the activities of the senescence-associated β-galactosidase (SA-β-gal), reactive oxygen species (ROS), and superoxide dismutase (SOD). The outcomes revealed that dyeing rate proportion of SA-β-gal was 2.78% ± 0.22% in the normal group, 2.83% ± 0.29% in the blank group, 37.58% ± 2.56% in the model group, and 28.39% ± 0.93% in the YYQEW group. The number of SA-β-gal positive cells was thus significantly higher in the model group than in the normal or blank group. There were also fewer SA-β-gal positive cells in the YYQEW group compared with the model group. The expression of ROS and p16INK4a in the model group increased significantly compared with that in the normal or blank groups, while the expression of ROS and p16INK4a in the YYQEW group decreased significantly compared with that in the model group. The expression of SOD and β-catenin in the model group decreased significantly compared with that in the normal or blank group, and the expression of SOD and β-catenin in the YYQEW group increased significantly compared with that in the model group. Skin fulfills several vital functions, many of which are dependent on its mechanical properties. Therefore, as mice have become an invaluable model for skin research, determining murine skin's mechanical properties is important. Specifically, skin's mechanical properties are important for functional tests as well as for prognostic and diagnostic purposes. Additionally, computational simulations of skin behavior are becoming commonplace, rendering accurate models of murine skin's constitutive behavior necessary. To date, our knowledge of mouse skin mechanics shows significant gaps. For example, there are no comprehensive reports correlating skin's mechanical properties with region, age, and direction. Moreover, mouse skin's residual strain behavior has not been reported on. In our current work, we set out to fill these gaps. Based on histology, 2-photon microscopy, and planar biaxial testing, while accurately tracking various reference configurations, we report on differences in gross structure, microstructural organization, and constitutive response of skin, and cast those properties into a versatile Fung-type hyperelastic constitutive law for three reference configurations. Our data is the most comprehensive report contrasting the mechanical properties of young (12 weeks) and aged (52 weeks) mouse skin and will, thus, be valuable to basic science as control data, and provide accurate constitutive laws for mouse skin modeling. Our findings are significant as they fill several gaps in our understanding of mouse skin mechanics. This is particularly important as mouse skin is becoming a frequent and critical model of human skin for cosmetic and medical science. Specifically, we quantified how mechanical properties of mice skin vary with age, with location, and with direction. Premature skin-aging is a disorder in human skin tissue caused by the effects of environmental factors. It is important to prevent and treat this disorder using natural ingredients. One of them is the Molineria latifolia plant, which is reportedly anti-aging. This study aims to characterize the chemical content of M. latifolia extract and evaluate its biological activity as a skin anti-aging. The 70% ethanol extract of M. latifolia roots was fractionated by a chromatography column using a solvent based on a gradient. The fractions obtained were evaluated for antioxidant, anti-tyrosinase, and anti-elastase activities. The active fraction was purified, and its chemical structure was characterized. The compounds were assessed for anti-aging activity in silico using 2Y9X and 3HGN proteins and continued with in vitro evaluation. The isolation results obtained six secondary metabolite compounds, namely 2,6 dimethoxy-benzoic acid (1) , curculigoside-A (2) , curcapital (3) , syringic acid (4) , 2-hydroxy-4-methoxybenzyl-2,3-dihydroxy-5-methoxybenzoate or curlatifolia-SN (5) , and luteolin (6). Compound 5 is newly phenolic discovered and has not been reported in previous research. Compounds 3, 5, and 6 interact best with 2Y9X (tyrosinase) and 3HGN (elastase) proteins in silico based on the obtained binding affinity. In vitro activity shows that compounds 3, 5, and 6 have strong to very strong activity inhibiting 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals, tyrosinase, and elastase. Compounds 3, 5, and 6 inhibit tyrosinase through a mixed inhibition mechanism, while elastase inhibits competitively. After 12 weeks, skin parameters showed improvement, and participants reported relief from dry eyes and vulvar issues. Werner syndrome (WS) is an autosomal recessive disorder characterized by physical signs and symptoms, including premature aging and scleroderma-like skin changes. The gene responsible for WS is the WRN gene. A significant proportion of WS-related malignant tumours are non-epithelial types, and the incidence of oral squamous cell carcinoma (SCC) is rare. A case of oral SCC of the lower alveolus and gingiva arising in a 63-year-old woman with WS is reported here. Biopsy confirmed moderately differentiated SCC. Surgical resection was performed and there was no recurrence or metastasis at the 3-year follow-up. Mutation analysis using next-generation sequencing, detected no mutations in the genes encoding the molecules strongly involved in the development of oral SCC, such as TP53 or PIK3CA. No obvious mutations were detected. Based on the results of the study, the results of mutation analysis suggest that this case might be genetically different from the common mechanisms of SCC in the oral cavity. [ABSTRACT FROM AUTHOR]

Background: Coenzyme Q10 is a fat-soluble antioxidant that can help to prevent collagen and elastin damage and avoid wrinkles. Coenzyme Q10 has several disadvantages to be formulated in topical dosage forms, such as low water solubility and large molecular weight. These make coenzyme Q10 retained in the stratum corneum and cause low skin penetration, so proper formulation is required to get products that can penetrate the skin layer. A nanostructured lipid carrier (NLC) consists of a matrix of solid lipids and liquid lipids in a certain amount with nanoparticle size; it may help increase the penetration of active ingredients. Methods: For the antiaging activity test, mice were grouped into four treatment groups and killed on the 14th day; then the back of the skin was stained with Masson trichrome staining. For the irritation test, the mice were grouped into three groups and killed after 24 h; then the back of the mice was stained with hematoxylin-eosin staining. Results: The number of fibroblasts in mice with NLC coenzyme Q10 is highest from all test groups. The irritation test results after 24 h of application preparation showed that NLC coenzyme Q10 did not irritate the skin of the back of male mice.
