2. Deep Learning-Assisted Single-Molecule Detection of Protein Post-translational Modifications with a Biological Nanopore

- Abstract
Protein post-translational modifications (PTMs) play a crucial role in countless biological processes, profoundly modulating protein properties on both spatial and temporal scales. Protein PTMs have also emerged as reliable biomarkers for several diseases. However, only a handful of techniques are available to accurately measure their levels, capture their complexity at a single molecule level, and characterize their multifaceted roles in health and disease. Nanopore sensing provides high sensitivity for the detection of low-abundance proteins, holding the potential to impact single-molecule proteomics and PTM detection, in particular. Here, we demonstrate the ability of a biological nanopore, the pore-forming toxin aerolysin, to detect and distinguish α-synuclein-derived peptides bearing single or multiple PTMs, namely, phosphorylation, nitration, and oxidation occurring at different positions and in various combinations. The characteristic current signatures of the α-synuclein peptide and its PTM variants could be confidently identified by using a deep learning model for signal processing. We further demonstrate that this framework can quantify α-synuclein peptides at picomolar concentrations and detect the C-terminal peptides generated by digestion of full-length α-synuclein. Collectively, our work highlights the advantage of using nanopores as a tool for simultaneous detection of multiple PTMs and facilitates their use in biomarker discovery and diagnostics.

3. Real-World Populational-Based Quality of Life and Functional Status After Stroke.

- Abstract
This study aimed to describe health-related quality of life (HRQoL) 3 months and 1 year after stroke, compare HRQoL between dependent (modified Rankin scale [mRS] 3-5) and independent (mRS 0-2) patients, and identify factors predictive of poor HRQoL. Patients with a first ischemic stroke or intraparenchymal hemorrhage from the Joinville Stroke Registry were analyzed retrospectively. Using the 5-level version of the EuroQol-5D questionnaire, HRQoL was calculated for all patients 3 months and 1 year after stroke, stratified by mRS score (0-2 or 3-5). One-year HRQoL predictors were examined using univariate and multivariate analyses. Three months after a stroke, data from 884 patients were analyzed; 72.8% were categorized as mRS 0-2 and 27.2% as mRS 3-5, and the mean HRQoL was 0.670 ± 0.256. At 1-year follow-up, 705 patients were evaluated; 75% were classified as mRS 0-2 and 25% as mRS 3-5, and the mean HRQoL was 0.71 ± 0.249. An increase in HRQoL was observed between 3 months and 1 year (mean difference 0.024, P <.0001), both in patients with 3-month mRS 0-2 (0.013, P =.027) and mRS 3-5 (0.052, P <.0001). Increasing age, female sex, hypertension, diabetes, and a high mRS were associated with poor HRQoL at 1 year. This study described the HRQoL after a stroke in a Brazilian population. This analysis shows that the mRS was highly associated with HRQoL after stroke. Age, sex, diabetes, and hypertension were also associated with HRQoL, although not independently of mRS. • Health-related quality of life (HRQoL) utility after stroke is currently unknown in Brazil. Using population-based data, we found that the 1-year HRQoL after stroke is 0.71, significantly lower in dependent (0.40) than independent patients (0.81). • These real-world findings are helpful for many future studies, such as those examining the burden of stroke and economic evaluations that include stroke as an outcome or seek to improve functional status after stroke. • The study also confirms that the modified Rankin scale (mRS) is a reliable HRQoL surrogate and provides mapping scores to convert mRS to utility, which is helpful for stroke studies that include the mRS but not the EQ-5D. Nowadays, there is a growing need for rehabilitation associated with demographic changes and health trends, with an increase in the prevalence of non-communicable diseases and an aging population. Although well-being is classically associated with a set of physical activities, the reality of today's society often shows that there is a lack of time available to develop activities that promote well-being. This fact is aggravated in aging populations by less mobility and greater isolation. In this domain, information and communication technologies have greatly contributed to helping healthcare providers to develop new understanding, measurement and action strategies that promote the physical and emotional well-being of their patients. Accordingly, the role of Virtual Reality has been shown to be promising for the generation of well-being. This article, as part of a broader funding project, reviews the state of the art of the role that Virtual Reality has played in the promotion of well-being, being an exercise of conceptualization that converges in the proposal of a conceptual model - an Index-Engine of Physical and Emotional Sustainability, which will be prototyped and field tested in the future. 6. Optimization of an in SilicoProtocol Using Probe Permeabilities to Identify Membrane Pan-Assay Interference Compounds

- Abstract
Membrane pan-assay interference compounds (PAINS) are a class of molecules that interact nonspecifically with lipid bilayers and alter their physicochemical properties. An early identification of these compounds avoids chasing false leads and the needless waste of time and resources in drug discovery campaigns. In this work, we optimized an in silicoprotocol on the basis of umbrella sampling (US)/molecular dynamics (MD) simulations to discriminate between compounds with different membrane PAINS behavior. We showed that the method is quite sensitive to membrane thickness fluctuations, which was mitigated by changing the US reference position to the phosphate atoms of the closest interacting monolayer. The computational efficiency was improved further by decreasing the number of umbrellas and adjusting their strength and position in our US scheme. The inhomogeneous solubility-diffusion model (ISDM) used to calculate the membrane permeability coefficients confirmed that resveratrol and curcumin have distinct membrane PAINS characteristics and indicated a misclassification of nothofagin in a previous work. Overall, we have presented here a promising in silicoprotocol that can be adopted as a future reference method to identify membrane PAINS.

7. Prediction of body fat in adolescents: validity of the methods relative fat mass, body adiposity index and body fat index

- Abstract
Objective: To verify the validity of anthropometric methods body adiposity index (BAI), relative fat mass (RFM) and body fat index (BFI) to estimate body fat percentage (%BF) in adolescents. Methods: A cross-sectional study was carried out with 420 Brazilian adolescents aged 15–19 years, stratified by age (< 18 years, n= 356; ≥ 18 years, n= 64) and sex (boys, n= 216; girls, n= 204). The Anthropometric measurements height, body weight, hip circumference and waist circumference were collected to calculate the %BF by BAI, RFM, BFI methods. Subsequently, %BF was measured by dual emission X-ray absorptiometry (DXA), adopted as a reference method. In the statistical analysis of the data, the Pearson correlation test and the paired ttest between %BF obtained by the equations and by the DXA were performed. The method validation criterion was that 68% of individuals should be within an acceptable error range of ± 3.5% of BF and Cohen's Kappa index ≥ 0.61. Additionally, the Bland–Altman graphical analysis was performed. Results: All methods showed a high correlation with DXA. For the Kappa index, only the RFM reached the criterion in the total sample (0.67) and in the sample < 18 years (0.68). None of the methods reached the criterion of 68% of the sample within the error range of ± 3.5% of BF. Conclusion: The BAI, RFM and BFI equations were not valid for predicting BF in the studied sample according to the criteria adopted regardless of sex or age. 8. Ideological extremism, perceived party system polarization, and support for democracy

- Abstract
AbstractDoes ideological polarization undermine or strengthen people’s principled support for democracy? In this study, we suggest that different manifestations of ideological polarization have different implications in this respect. Using data from 11 surveys conducted with representative samples of the adult populations of a group of liberal democratic countries, part of the Comparative National Elections Project, we look at how people’s level of ideological extremism and their perceptions of ideological polarization in their countries’ party systems are related with their support for democracy. We show that citizens who hold more extreme ideological positions are indeed less supportive of democracy and that such a negative relationship is strengthened as citizens’ extremism increases. However, we also show that the citizens who display higher levels of principled support for democracy are those who perceive parties to be neither too distant nor too close to each other in ideological terms. In other words, while a very polarized partisan supply seems to undermine popular commitment with democracy, very low polarization may have similar consequences.

10. Device size selection can enhance Y-stentrieving efficacy and safety as a rescue strategy in stroke thrombectomy

- Abstract
BackgroundDespite advancements in stroke treatment, refractory clots are relatively common, prompting the exploration of alternative techniques. Bifurcation occlusions pose specific intraprocedural challenges, occasionally dealt with by two stentrievers deployed in Y-configuration. Previous studies have portrayed this strategy as feasible, yet little is known about its safety and efficacy, and how to best select retrievers.ObjectiveTo determine whether device selection influences the efficacy and safety of Y-stentrieving.MethodsWe performed a multicentric, retrospective analysis of patients undergoing Y-stentrieving rescue for bifurcation occlusions. Demographics, devices, procedural metrics, neurological severity, reperfusion, disability, and safety were assessed.ResultsY-configuration stents were used as a rescue maneuver after 2.16±1.5 failed attempts with other techniques in 20 patients. Successful reperfusion (modified Thrombolysis in Cerebral Infarction score 2b–3) was achieved in 70% of patients after the first Y-stentrieving attempt. The first stentriever more often had a larger diameter (5.15±0.92 vs 3.67±0.57 mm, p=0.017) and longer length (33.12±5.78 vs 20.67±1.15 mm, p=0.002) in successfully reperfused cases. Also, the diameter of the first device was associated with both any parenchymal (6.0 vs 4.71±0.99 mm, p=0.045) and symptomatic (6.0 vs 4.86±1.02 mm, p<0.001) hemorrhages. Exact logistic regression demonstrated that a longer length first stentriever independently predicted better angiographic outcomes (OR=1.26, p=0.036), and a 6 mm diameter first stentriever independently predicted more intracranial hemorrhages (OR=15.28, p=0.044). No periprocedural mortality was recorded.ConclusionY-stentrieving is an effective and safe bail-out strategy for refractory bifurcation clots. Longer stents may promote better angiographic outcomes, whereas avoidance of disproportionately large retrievers may mitigate intracranial hemorrhage. Future studies should account for these factors when evaluating alternative stentriever techniques.

Abstract: Some plant seeds and berries have gained increased recognition due to their widevariety of bioactive compounds. Many of these foods are rich in lipids with high nutritionalvalue, of which n-3 and n-6 essential fatty acids stand out. However, knowledge of the chemicalcomposition and biological activity of these compounds and their value as a source ofhealthy lipids is far from being fully explored. The benefits of the intake of hemp seeds, flaxseed, and açaí berries, considered functionalfoods, have been compiled in this review. Likewise, their general chemical composition will bedescribed to evaluate their relevance in a healthy diet, highlighting their lipid components andhealth lipid indices. The fibres and essential fatty acids of hemp seeds and flaxseed and the antioxidant propertiesof açaí berries provide them protective roles against several chronic non-communicable diseasesand represent significant beneficial effects that add value to these healthy natural products.More research is needed to deepen the knowledge on their lipids molecular composition andbioactivity.

12. Normal limits of the electrocardiogram in Angolans.

- Abstract
Introduction: Studies on the normal electrocardiogram limits in African populations are limited, especially in sub-Saharan Africa. There is no literature describing normal ECG limits in Angolans.Objectives: The aim of this study is to establish the normal ECG limits for adult Angolans, without established heart disease, stratified by gender and age.Methods: A cross-sectional study was performed, involving 2179 participants from a population in northern Angola, without established heart disease, aged between 15 and 74 years. A 12‑lead ECG and a rhythm strip were recorded for all participants and analysed and processed by the University of Glasgow software and encoded by the Minnesota Code. The normal range of the electrocardiographic parameters were established as the 2nd and 98th percentiles of the measurement distribution per age group and gender. Mann-Whitney and Kruskal-Wallis tests were used for two independent groups and Bonferroni adjustments were used for multiple testing. GAMLSS models were used to obtain the continuous age-dependent percentile curves.Results: The normal range of the ECG differed between men and women: heart rate 49 to 100 bpm vs. 55 to 108 bpm, P wave duration 81 to 130 ms vs. 84 to 130 ms, PR interval 119 to 210 ms vs. 120 to 202 ms, QRS duration 74 to 110 ms vs. 70 to 104 ms, QT interval 324 to 441 ms vs. 314 and 438 ms, P-wave axis - 29 to 850 vs. -18 to 810, QRS-wave axis - 13 to 850 vs. -180 and 820, T-wave axis 0 to 720 vs. -8 to 730, Sokolow-Lyon index 2.13 to 6.21 mV vs. 1.60 to 4.87 mV, Cornell index 0.17 to 6.24 mV vs. 0.14 mV to 4.35 mV.Conclusions: The values described for the electrocardiographic measurements above can act as a reference framework for Angolan adults without established heart disease. Our study suggests that the normal range of most ECG parameters vary according to age and sex and the ECG diagnostic criteria must therefore be specific for these demographic measures. - Abstract
Although a rare disease, bilateral congenital anomalies of the kidney and urinary tract (CAKUT) are the leading cause of end stage kidney disease in children. Ultrasound-based prenatal prediction of postnatal kidney survival in CAKUT pregnancies is far from accurate. To improve prediction, we conducted a prospective multicenter peptidome analysis of amniotic fluid spanning 140 evaluable fetuses with CAKUT. We identified a signature of 98 endogenous amniotic fluid peptides, mainly composed of fragments from extracellular matrix proteins and from the actin binding protein thymosin-β4. The peptide signature predicted postnatal kidney outcome with an area under the curve of 0.96 in the holdout validation set of patients with CAKUT with definite endpoint data. Additionally, this peptide signature was validated in a geographically independent sub-cohort of 12 patients (area under the curve 1.00) and displayed high specificity in non-CAKUT pregnancies (82 and 94% in 22 healthy fetuses and in 47 fetuses with congenital cytomegalovirus infection respectively). Change in amniotic fluid thymosin-β4 abundance was confirmed with ELISA. Knockout of thymosin-β4 in zebrafish altered proximal and distal tubule pronephros growth suggesting a possible role of thymosin β4 in fetal kidney development. Thus, recognition of the 98-peptide signature in amniotic fluid during diagnostic workup of prenatally detected fetuses with CAKUT can provide a long-sought evidence base for accurate management of the CAKUT disorder that is currently unavailable.

20. Low-cost bus business models and the case of Brazil

- Abstract
Purpose: The purpose of this paper is to identify low-costbus business models from different parts of the world and check their applicability in the Brazilian market. It also identifies crucial factors for the development of that kind of business and investigates the relationship between low-cost buses and other modes of transport. This research analyzes every relevant aspect to the applicability of low-cost business models in Brazil, driving to discussions and conclusions. The gains on the development of low-cost bus systems in Brazil may have a wide reach, from personal to general public benefits. Design/methodology/approach: Business models for low-cost bus systems are used to analyze in a qualitative approach. The data are collected through semi-structured interviews, direct observations and documental basis. In addition, innovations over the previous five years are evaluated in order to establish a comparative pattern between companies. Findings: There is a great potential in the Brazilian passenger market for the entrance of low-cost bus companies. The only question is just when it is the right time to enter that market. Most of the negative points presented for the implementation of a low-cost company are related to the current economical and political crisis in Brazil. It was identified as a potential cause for the overall decrease of the passengers market in recent years, and specifically of the bus passengers market. Originality/value: The recent regulation changes, the high demand for passengers and even the similarity of possible routes in Brazil to the ones in Europe and in the USA make Brazil a fertile soil for the development of that kind of business. A similar price mechanism to the ones applied worldwide was also identified as doable in Brazil.

Photocatalysis was recognised as a suitable process for the photoabatement of atmospheric pollutants. The photooxidation mechanism on TiO 2 has been widely studied. However, recent studies demonstrated that the very often-assumed photooxidation intermediated by the hydroxyl radical cannot explain all the experimental observations. Indeed, this study contributes for a new understanding of NO photooxidation. First, the adsorption equilibrium isotherms of NO, NO 2 and H 2 O on the photocatalyst, Aeroxide ® P25 from Evonik Industries, were obtained. Also, the concentration of hydroxyl radicals was determined by photoluminescence. A comprehensive design of experiments was then followed; NO conversion and selectivity were obtained as a function of the relative humidity, irradiance, NO inlet concentration and residence time, following a response surface methodology (RSM). The present study evaluated whether a natural dietary additive, dried Artemisia annua leaves, may be useful to control Rhipicephalus (Boophilus) microplus on naturally infested cattle. Twenty heifers of the Canchim breed, weighing around 250 kg, were divided into two equally sized experimental groups: 1) control animals and 2) animals receiving 200 g/day of dried A. annua leaves for two months. Before treatment began, the animals were homogeneously distributed in control and treatment groups based on their pre-treatment weight and tick infestation level. Counts of engorged female ticks then occurred weekly during the two-month experimental period. We also monitored cattle weight gain and packed cell volume (PCV). Artemisinin (0.96%) was quantified in the plant material by high-performance liquid chromatography with refractive index detector (HPLC-IR). No statistical differences between the control and treatment groups were observed for engorged female counts (log averages of 1.3 ticks and 1.4 ticks per animal, respectively), daily cattle weight gain (0.910 kg and 0.888 kg, respectively) or PCV (33.5% and 33.0%, respectively). We conclude that the oral supplementation of cattle feed with dried A. annuna leaves did not control natural infestation of R. (B.) microplus . This paper describes a method specially devoted to quantitatively measure the success of collaborative university-industry R&D funded contracts, which could be managed as a singular project or a program of projects. The method aims to measure the success throughout the program/project lifecycle, combining both retrospective (lagging) and prospective (leading) performance indicators. The method uses tangible/specific outcomes as performance indicators, like patents or publications, as well as intangible/subjective performance indicators such as social relationships, organizational arrangements or motivations. The proposed method was conceived by conducting a thorough review of the published literature on this area, and by analyzing, as case studies, two consecutive R&D collaborative funded programs, between University of Minho and Bosch Car Mutimedia, amounting to an overall investment of over 70 million Euros, from 2012 to 2018. In this article we analyze the evolution of the attitudes towards Europe of Portuguese parliamentary elites in the context of the sovereign debt crisis. Our analysis relies on interviews to a total of 227 MPs in the context of the ENEC project in 2014 and of the Intune Project in 2009 and 2007. Our principal finding is that an important gap has risen between the attitudes of the MPs and of the masses. Indeed, while the Portuguese are becoming increasingly less pro- European, this is not the case of their deputies. We show that, in 2014, the percentage of deputies that believed that EU membership benefited the country has decreased a little, but it stays as high as 89%. Our data, however, show that MPs now exhibit lower levels of trust towards the European institutions. Interestingly, the drop in the levels of trust towards EU institutions does not affect all institutions equally: trust towards the European Parliament remains constant regarding 2007, whereas the levels of trust in the remaining two institutions decrease. Finally, we show that it is in the Socialist Party (PS) that there is the steepest decline in the level of overall trust, which is rooted in declining levels of confidence in the European Commission. Given that PS has traditionally portrayed itself as the most pro-Europe of the Portuguese parties, this is solid evidence of a growing discontent with the European Commission. [ABSTRACT FROM AUTHOR]

The stagnation temperatures experienced by glazed photovoltaic-thermal (PV-T) collectors pose a threat to their performance and longevity, in part due to the limited temperature stability of ethylene vinyl acetate (EVA) encapsulants. In order to identify suitable solutions for application in residential solar domestic hot water (SDHW) PV-T systems, a comparative analysis of known overheating prevention and stagnation handling measures was conducted and dynamic simulations were used to support the analysis. While no measure was found to comply with all desirable goals including reliability, implementation and operation costs, integral venting mechanisms were identified as the most promising among the control systems reviewed. Moreover, active collector heat dumping and automatic collector shading led to minimal electrical efficiency increases and decreases, respectively, while purging tank water when the collector overheats was found to be ineffective due to delays in the start of circulation. Purpose Hemorrhagic cystitis is an important dose limiting side effect of ifosfamide based cancer chemotherapy. Despite chemoprophylaxis inflammation can still be found in cystoscopy guided biopsies. Previous studies confirmed the role of TNF-α and IL-1β. We evaluated the protective effect of the IL-1R antagonist anakinra and the anti-TNF-α antibody infliximab in experimental ifosfamide induced hemorrhagic cystitis. Materials and Methods Hemorrhagic cystitis was induced by an injection of ifosfamide (400 mg/kg intraperitoneally) in Swiss wild-type C57Bl/6, IL-1R –/– , TNFR1 –/– or TNFR1/R2 –/– mice. Mice were treated 30 minutes before ifosfamide with anakinra (100 mg/kg intraperitoneally), infliximab (5 mg/kg intraperitoneally) or vehicle. Visceral nociception was evaluated after hemorrhagic cystitis induction. At 12 hours the animals were sacrificed. Bladders were harvested to assess bladder wet weight, vascular permeability, macroscopic and microscopic findings, muscle contractility, and for cystometrography. Inflammatory cell infiltration was assessed by myeloperoxidase assay and flow cytometry. Results Anakinra attenuated hemorrhage, edema, neutrophil infiltration, visceral hyperalgesia and bladder dysfunction. IL-1R –/– mice also showed milder hemorrhagic cystitis. Infliximab inhibited bladder edema and visceral hyperalgesia without preventing hemorrhage, bladder dysfunction, neutrophils or accumulation. Additionally, the lack of TNFR1 decreased bladder edema but not cell infiltration whereas concomitant deficiency of TNFR1 and TNFR2 resulted in worse hemorrhagic cystitis. Conclusions Anakinra is effective for preventing experimentally ifosfamide induced hemorrhagic cystitis. It seems that neutrophil and macrophage infiltration in this circumstance depends on IL-1 signaling through IL1R. Possibly TNFR2 has a protective role in hemorrhagic cystitis. [ABSTRACT FROM AUTHOR]

- Abstract
The austerity policies pursued in several countries during the Eurozone crisis began to call attention to the role played by courts as relevant actors in the context of budgetary and financial stress. The case of the Portuguese Constitutional Court has often been singled out in national and international forums as one characterized by particularly intense activism in this respect. Allegedly, political conflict around austerity policies and the demand for their judicial review had fundamentally changed the role of the Portuguese Constitutional Court and the behavior of its judges. However, after examining these claims empirically, we find that, when properly assessed with scrutiny of comparable legislation in other periods, the judicial behavior on austerity policies exhibits a much less exceptional pattern than often argued. Constitutional review in Portugal seems to respond to institutional arrangements (i.e., those fostering a central role for ideological preferences and party loyalty) and not to specific business cycles.

This study examined whether sleep disturbance predicted or moderated responses to psychotherapy in participants who participated in STEP-BD, a national, multisite study that examined the effectiveness of different treatment combinations for bipolar disorder. Participants received either a brief psychosocial intervention called collaborative care (CC; n= 130) or intensive psychotherapy (IP; n= 163), with study-based pharmacotherapy. Participants (N= 243) were defined as current (past week) short sleepers (<6 hours/night), normal sleepers (6.5–8.5 hours/night), and long sleepers (≥9 hours/night), according to reported average nightly sleep duration the week before randomization. Sleep disturbances did not predict the likelihood of recovery nor time until recovery from a depressive episode. There was no difference in recovery rates between IP versus CC for normal sleepers, and medium effect sizes were observed for differences in short and long sleepers. In this study, sleep did not play a major role in predicting or moderating response to psychotherapy in bipolar disorder.

41. Relationship between treatment delay and type of reperfusion therapy and mechanical complications of acute myocardial infarction

Objectives: The incidence of mechanical complications after acute myocardial infarction has markedly declined with the advent of reperfusion. Nevertheless there is some controversy about the equal effectiveness of the different reperfusion therapies in preventing these complications. We aimed to analyse how reperfusion therapy and treatment delay relate to the incidence of mechanical complications in a population of ST-elevation myocardial infarction (STEMI) patients.Methods: We analysed all STEMI patients included in the second phase of the Portuguese Registry on Acute Coronary Syndromes, between October 2010 and July 2015. We compared both conservative medical treatment with reperfusion therapy and thrombolysis with primary percutaneous coronary intervention for mechanical complications. We also evaluated the impact of treatment delay on mechanical complications.Results: Among 5230 STEMIs we observed 77 mechanical complications (1.5%). These were significantly more frequent in the non-reperfused patients (3.3% vs. 1.1%, P<0.001) and they were numerically higher in thrombolysis than in primary percutaneous coronary intervention patients (1.6% vs. 1.0%, respectively, P=0.282). Patients with mechanical complications had higher times from symptom onset to hospitalisation and to reperfusion. In multivariate analysis performing reperfusion therapy (odds ratio 0.52, 95% confidence interval 0.29–0.93) and a time from symptom onset to hospitalisation ?6 hours (odds ratio 2.44, 95% confidence interval 1.37–4.33) were independent predictors of mechanical complications. The type of reperfusion did not influence the occurrence of mechanical complications.Conclusion: A longer time from symptom onset to hospitalisation was associated with an increased number of mechanical complications. Timely reperfusion therapy prevented mechanical complications and no significant difference was found between thrombolysis and primary percutaneous coronary intervention.

Objective: To assess the level of knowledge of primary school teachers in the public school network of Northeastern Brazil with respect to management of dental trauma and its relationship with prognosis. Methods: A questionnaire was applied to 195 school teachers of public schools in Northeastern Brazil. The questionnaire comprised 12 objective questions about dental trauma and methods for its prevention and management. Data were submitted to chi-square test and Poisson regression test (P > 0.05). Results: Out of the 141 teachers who responded the questionnaires, the majority were women (70.2%) and most of them had experienced previous dental accidents involving a child (53.2%). The majority (84.4%) had incomplete college education and few were given some training on how to deal with emergency situations during their undergraduate course (13.5%) or after it (38.3%). Their level of knowledge about dental trauma and emergency protocols showed that unsatisfactory knowledge level was associated with the male sex: 46% higher for men in comparison to women (P = 0.025). Conclusions: Approximately half of teachers evaluated had unsatisfactory knowledge about dental trauma and emergency protocols, with female teachers showing more knowledge than men. Abstract: Background/purpose: The mechanism of fetal gastric dilation in gastroschisis is controversial. This study was designed to characterize changes in the contractile profile of strips of stomach from rats following experimental gastroschisis. Methods: Pregnant Wistar rats were operated on day 18.5. Fetuses were divided into three groups: gastroschisis (G), sham (S), and control (C). On day 21.5, gastric fundus and antrum strips were obtained and suspended to a force transducer connected to a digital data acquisition system. They were submitted to increasing concentrations of carbachol (CCh) and weighed at the end of each procedure. Frequency and amplitude of each contraction were evaluated. Results: Under basal conditions, spontaneous oscillatory contractions of antrum and fundus strips of G, S, and C were similar (P>0.05; ANOVA). However, cumulative concentrations of CCh (0.01–100μM) produced different effects in all groups and were characterized by a significant increase in amplitude and frequency of spontaneous contractions in antral smooth muscle and a sustained increase in tonus in fundic strips. Upon analysis, no significant difference in frequency or amplitude was noted in antral tissues comparing C to G and to S (P>0.05). No significant contractility difference was noted in fundic smooth muscle (comparing all groups, P >0.05), with the CCh-induced curve following a typical sigmoidal format, dependent on increasing concentrations (P<0.001). Conclusions: Gastric contractile responses to CCh are preserved in experimental gastroschisis. These results do not support the theory that gastric dilation occurs secondary to intestinal inflammation alone. [Copyright &y& Elsevier]

Abstract: Objective: Many studies have documented the high rates of functional impairment among patients with schizophrenia. The majority of the available instruments used to assess functioning, however, focus on global measures of functional recovery rather than specific domains of psychosocial functioning. Most of these instruments have important limitations regarding use in psychiatry. The aim of the present study was to evaluate the psychometric properties of the Brazilian version of the Functioning Assessment Short Test (FAST) in patients with schizophrenia. Methods: A convenience sample of 107 chronic outpatients with schizophrenia and 108 controls was assessed in a university hospital (Hospital de Clínicas de Porto Alegre, Brazil). Psychometric properties of FAST (internal consistency, concurrent validity, and test-retest reliability) were analyzed. Results: The internal consistency obtained was high; the Cronbach’s alpha was 0.89. FAST total score was higher in patients than in the control group (Z = 11.95; P<0.001). FAST test-retest agreement was excellent (intraclass correlation coefficient = 0.93; 95% confidence interval 0.81–0.97). In addition, FAST displayed a positive correlation with the Brief Psychiatric Rating Scale (ρ = 0.41; P<0.001) and a negative correlation with the Global Assessment of Functioning scale (ρ =−0.71; P = 0.001). Conclusions: Psychotic symptoms, comorbidity, and functional and cognitive impairment contribute to the decreased quality of life of patients with schizophrenia. It is important to obtain a valid and reliable instrument that is capable of evaluating the functional domains in this pathology. In this context, FAST showed accurate psychometrics properties and was able to detect functional differences between patients with the diagnosis of schizophrenia and healthy subjects. Abstract: Background: The use of crack cocaine is a major public health concern in Brazil and internationally. Recent data suggest that childhood trauma is associated with worse outcomes among cocaine users. This study had the objective of evaluating the relationship of childhood trauma with executive functioning and impulsivity in outpatients with crack cocaine use disorders. Methods: This is a cross-sectional study of 84 consecutive outpatients with a primary crack cocaine use disorder who sought treatment in Porto Alegre, Brazil. Childhood trauma was evaluated with the Childhood Trauma Questionnaire; executive functioning, with the Wisconsin Card Sorting Test; and impulsivity, with the Barratt Impulsivity Scale. Results: Childhood trauma was strongly associated with executive dysfunction and impulsivity, even when controlled for possible confounders. Conclusions: Childhood trauma may be associated with executive dysfunction and impulsivity in crack cocaine users. The full impact of trauma needs to be further investigated in longitudinal studies.
