Abstract: Hypertension affects more than one billion people worldwide. Here we identify 113 novel loci, reporting a total of 2,103 independent genetic signals (P< 5 × 10−8) from the largest single-stage blood pressure (BP) genome-wide association study to date (n= 1,028,980 European individuals). These associations explain more than 60% of single nucleotide polymorphism-based BP heritability. Comparing top versus bottom deciles of polygenic risk scores (PRSs) reveals clinically meaningful differences in BP (16.9 mmHg systolic BP, 95% CI, 15.5–18.2 mmHg, P= 2.22 × 10−126) and more than a sevenfold higher odds of hypertension risk (odds ratio, 7.33; 95% CI, 5.54–9.70; P= 4.13 × 10−44) in an independent dataset. Adding PRS into hypertension-prediction models increased the area under the receiver operating characteristic curve (AUROC) from 0.791 (95% CI, 0.781–0.801) to 0.826 (95% CI, 0.817–0.836, ∆AUROC, 0.035, P= 1.98 × 10−34). We compare the 2,103 loci results in non-European ancestries and show significant PRS associations in a large African-American sample. Secondary analyses implicate 500 genes previously unreported for BP. Our study highlights the role of increasingly large genomic studies for precision health research.

4. Anomaly detection using electrical signature analysis and machine learning:application to a CNC mill.

Abstract: Electrical Signature Analysis (ESA) is a powerful tool that uses the voltage and current signals of a machine to infer its health status. ESA can serve as a predictive maintenance tool for detecting common faults at an early stage, thus preventing expensive catastrophic failures and production outages, and extending equipment lifetime. In this study, a novel application of ESA and Machine Learning (ML) for working condition monitoring and health status assessment of a CNC mill is presented. Experimental results show the effectiveness of the proposed approach.

5. Synthesis and characterization of novel fluorene–based green copolymers and their potential application in organic light-emitting diodes

Abstract: Fluorene-benzothiadiazole (PFBT), commercially known as F8BT, and fluorene-thiophene (PFT) copolymers were synthesized by the Suzuki-Miyaura coupling polycondensation technique in the presence of a phase transfer catalyst (Aliquat 336). These novel copolymers showed higher molar masses, lower dispersivity, and thermal stability above 400 °C when synthesized under the modified (M) action of Aliquat 336 (F8BT-M). Polymer Light-emitting diodes (PLEDs) were assembled with the novel synthesized copolymers as the emitter layer. For F8BT-M, a maximum luminance of 327 cd/m2was achieved, while a diode containing just the commercial F8BT resulted in only 210 cd/m2. As for the PFT copolymers, it was observed that for the Aliquat 336 sample synthesized in a 60h extended (E) (PFT-ME), the luminance of the PLED was much higher than that where the reaction time was 48 h (PFT-M), with a luminance of 168 cd/m2and 54 cd/m2, respectively. Without Aliquat 336, the PFT copolymer did not show an electroluminescent signal. The improved electroluminescence in the copolymers with Aliquat 336 was attributed to the improved synthesis conditions, i.e., reaction time and catalytic system employed. The results suggest that copolymers with luminescent and morphological properties, more suitable for light-emitting devices, might be obtained with simple modifications during synthetic polycondensation reactions.

Abstract: Plastic pollution is a notable environmental issue, being plastic widespread and characterized by long lifetime. Serious environmental problems are caused by the improper management of plastic end-of-life. In fact, plastic litter is currently detected in any environment. Biodegradable Polymers (BPs) are promising materials if correctly applied and managed at their end of life, to minimize environmental problems. However, poor data on the fate and toxicity of BPs on marine organisms still limit their applicability. In this work we tested the effects of five biodegradable polymers (polybutylene succinate, PBS; polybutylene succinate-co-butylene adipate, PBSA; polycaprolactone, PCL; poly (3-hydroxybutyrates, PHB; polylactic acid, PLA) widely used for several purposes. Adult individuals of the isopod Idotea balthica basteri were fed on these polymers for twenty-seven days by adding biodegradable microplastic polymers (BMPs) to formulated feeds at two concentrations, viz. 0.84 and 8.4 g/kg feed. The plastic fragments affected the mortality rates of the isopods, as well as the expression levels of eighteen genes (tested by Real Time qPCR) involved in stress response and detoxification processes. Our findings confirmed that I. balthica basteri is a convenient model organism to study the response to environmental pollution and emerging contaminants in the aquatic environment, and highlighted the need for the correct use of BMPs. Abstract: The mechanisms regulating the urinary excretion of uromodulin remain mostly unknown. A meta-GWAS conducted in 29,315 individuals from 13 cohorts identified two novel, genome-wide significant loci, KRT40and WDR72, in addition to the previously known UMOD-PDILTlocus, to be associated with urinary uromodulin. KRT40 colocalizes with uromodulin in TAL cells and functional studies showed that its expression affects the processing and apical excretion of uromodulin. WDR72, which does not colocalize with uromodulin, has been associated with kidney function, urinary acidification, and kidney stones. These studies provide novel insights into the biology of uromodulin and keratins and into the influence of the UMOD-PDILTlocus on kidney function.

13. Is the use of fully covered metal stents effective in the treatment of difficult lithiasis of the common bile duct?

Abstract: Background: The management of difficult lithiasis of common bile duct (CBD) often requires a multimodal approach. Fully Covered Metal Stents (FCSEMS) could fragment the stones, dilate strictures of the CBD and progressively enlarge the papillary orifice. The aim of the study was to evaluate the efficacy of FCSEMS in the treatment of difficult lithiasis of CBD. Methods: All consecutive cases of difficult lithiasis of CBD after an attempt of clearance with complete sphincterotomy were retrospectively considered from 2009 to 2019. Intrahepatic lithiasis cases were excluded. Multivariable analysis (stepwise logistic regression if pvalue < 0.1 at univariable analysis) was made to understand factors associated with efficacy of FCSEMS in difficult lithiasis of CBD. Results: 163 cases of difficult lithiasis over 1516 cases of lithiasis of CBD (incidence of 11%) were considered. Among eligible patients, 96 (mean age 78 ± 11.9 years, 43% males) placed a FCSEMS during the first ERCP. Mean diameter of stone was 17.5 ± 5.5 mm. An attempt of dilation during the same procedure was made in 18% cases. After removal of the stent, 52 (54%) patients had complete clearance of CBD at cholangiography. The rest of patients (44), subsequently underwent multimodal treatment (final success of 95%). After logistic regression, variables positively associated with clearance of CBD were sex male (OR 3.5) and stent permanence ≥ 30 days (OR 3.2). Diameter of the stone ≥ 2 cm was associated with failure (OR 0.2). 7 post-ERCP pancreatitis, 6 post-sphincterotomy bleeding and 3 cholangitis occurred. Conclusions: FCSEMS are useful in the approach of difficult lithiasis of CBD with no significant adverse events associated.

14. Work-related musculoskeletal disorders among endoscopic transsphenoidal surgeons: a systematic review of prevalence and ergonomic interventions

Abstract: Endoscopic transsphenoidal surgery is a surgical technique introduced in the last 20 years for the treatment of skull base pathologies and, in particular, pituitary tumours. Although the prevalence of work-related musculoskeletal disorders is usually significantly higher in endoscopists and minimally invasive surgeons compared to other surgical and medical specialties, reviews on the prevalence of disorders among neurosurgeons dedicated to endoscopic transsphenoidal surgery are not available. This article performs a systematic review to identify the work-related musculoskeletal disorders among transsphenoidal neurosurgeons, their prevalence and the ergonomic interventions proposed to reduce risk factors and prevent disorders. The results show that the ergonomics of transsphenoidal neurosurgeons is an under-investigated topic. Indeed, specific prevalence data are not available. In addition, only a few papers suggest interventions and guidelines, but without objective assessment of the outcomes to confirm the ergonomic benefit. Based on these gaps in the literature, a future research agenda is proposed.

Abstract: Common single-nucleotide polymorphisms (SNPs) are predicted to collectively explain 40–50% of phenotypic variation in human height, but identifying the specific variants and associated regions requires huge sample sizes1. Here, using data from a genome-wide association study of 5.4 million individuals of diverse ancestries, we show that 12,111 independent SNPs that are significantly associated with height account for nearly all of the common SNP-based heritability. These SNPs are clustered within 7,209 non-overlapping genomic segments with a mean size of around 90 kb, covering about 21% of the genome. The density of independent associations varies across the genome and the regions of increased density are enriched for biologically relevant genes. In out-of-sample estimation and prediction, the 12,111 SNPs (or all SNPs in the HapMap 3 panel2) account for 40% (45%) of phenotypic variance in populations of European ancestry but only around 10–20% (14–24%) in populations of other ancestries. Effect sizes, associated regions and gene prioritization are similar across ancestries, indicating that reduced prediction accuracy is likely to be explained by linkage disequilibrium and differences in allele frequency within associated regions. Finally, we show that the relevant biological pathways are detectable with smaller sample sizes than are needed to implicate causal genes and variants. Overall, this study provides a comprehensive map of specific genomic regions that contain the vast majority of common height-associated variants. Although this map is saturated for populations of European ancestry, further research is needed to achieve equivalent saturation in other ancestries.

In detail, a new capping system based on a BTA–silver coordination complex, able to dissolve in acid and alkaline conditions and to simultaneously tailor the BTA release and the capture of chloride ions, was properly designed and realized. Acrylic coatings embedding the engineered MSNs were deposited onto iron rebar samples and tested for their protective capability in acid and alkaline environments. Results highlighted the high potential of the proposed system for the protection of metals, due to the synergistic effect of the mesoporous structure and the capping system, which guaranteed both the sequestration of chloride ions and the on-demand release of the effective amount of anticorrosive agents able to ensure the enhanced protection of the substrate.

17. Oxygen deficiency hazard in confined spaces in the steel industry: assessment through predictive models

Abstract: Objective.In the steel industry, performing activities in confined spaces where potential oxygen displacement can occur may expose workers to fatal consequences. To the best of our knowledge, no quantitative exposure assessment of oxygen deficiency in steel industry confined spaces is available in the literature. To overcome this gap, we performed oxygen deficiency hazard (ODH) assessments in real confined spaces using two existing models to identify the most critical parameters responsible for ODH, and suggest controls for mitigating the asphyxiation risk. Methods.We applied a well-mixed model and a near field–far field approach to estimate the indoor oxygen level with time during and following release of simple asphyxiants. Model inputs were mainly gathered thanks to audits and instrumental tests in three firms. Results.The most severe ODH exposures are posed in spaces with restricted volume and where accidental releases of inert gases can occur. Such exposures can be controlled through early release detections and augmented reality systems. Conclusions.ODH assessments in confined spaces of steel firms allow the identification of the most critical parameters from an oxygen depletion perspective, focusing on which data need careful measurement, and help to establish controls compatible with the operations conducted in these areas.

Abstract: Phthalates are widely employed plasticizers blended to plastic polymers that, during plastic aging and weathering are prone to leach in the surrounding environment. Thus, phthalates were proposed to indirectly evaluate MPs contamination in marine environments, with still uncertain and scarce data, particularly for wildlife. This study investigates simultaneously microplastics (MPs) and phthalates (PAEs) occurrence in wild Actinia equina and Anemonia viridis , two common and edible sea anemone species. Both species had a 100 % frequency of MPs occurrence, with similar average concentrations. PAEs were detected in 70 % of samples, with concentrations up to 150 ng/g in A. equina and 144.3 ng/g for A. viridis. MPs and PAEs present in sea anemone tissues appear to reflect seawater plastic contamination conditions in the study area. Abstract: Plastic and microplastics, including polyethylene (PE), polypropylene (PP), and polystyrene (PS), are major contributors to environmental pollution. However, there is a growing recognition of the need to investigate a wider range of plastic polymers to fully understand the extent and impacts of plastic pollution. This study focuses on the comprehensive characterization of true-to-life nanoplastics (T2LNPs) derived from polyethylene terephthalate (PET) and polyamide (PA) to enhance our understanding of environmental nanoplastics pollution. T2LNPs were produced through cryogenic mechanical fragmentation of everyday items made from these polymers. A solid methodological framework incorporating various characterization techniques was established. Attenuated total reflection Fourier transform infrared (ATR-FTIR) spectroscopy and thermogravimetric analysis (TGA) were employed to study the chemical composition and confirm the absence of chemical modifications possibly occurring during fragmentation. Atomic force microscopy (AFM), scanning electron microscopy (SEM), and transmission electron microscopy (TEM) were used to analyze the morphology of the T2LNPs. Additionally, AFM image analysis compared to dynamic light scattering (DLS) measurements provided insights into the size distribution and the stability of the T2LNP suspensions. The results revealed the heterogeneity of T2LNPs derived from PET and PA, emphasizing the importance of studying different plastic compositions to comprehensively understand nanoplastics pollution. Lastly, the distinctive characteristics and morphology of T2LNPs were translated into the realm of biological interactions, offering initial insights into the influence of these disparities on the formation of the protein corona on the surface of T2LNPs. By proposing T2LNPs as test materials and establishing a comprehensive characterization approach, this study aims to bridge the knowledge gap regarding the behavior and toxicity of nanoplastics. 22. KIAA0753-related skeletal ciliopathy: a ninth case, extending the phenotype and reporting a novel variant

Abstract: KIAA0753-related skeletal ciliopathy is a recently described recessive disorder causing skeletal dysplasia and overlapping features of certain ciliopathies; Joubert, Jeune and Oro-facial-digital syndromes. We describe a ninth case that expands the phenotype; a 10-year-old girl with rhizomelic short stature (−5.6 SD), macrocephaly, developmental delay, CNS anomalies (thin corpus callosum, bilateral ventriculomegaly), cone-rod dystrophy, nystagmus, mild conductive hearing loss and recurrent chest infections secondary to confirmed ciliary dyskinesia. Testing for FGFR3achondroplasia-related hotspots and mucopolysaccharidosis were negative. Whole-exome sequencing, aged eight, via skeletal dysplasia panel analysis and subsequent whole-genome sequencing (via the 100,000 genomes project) found no cause. WGS data reanalysis using exomiser uncovered compound heterozygous pathogenic KIAA0753variants (frameshift and splice site). Further clinical and radiological surveys were consistent with the expected phenotype. We discuss the emerging phenotype of this uncommon disorder. This report details the sixth published case of skeletal dysplasia in all cases of KIAA0753-related disease and the first case to describe a novel c.1830-2A>G splice variant. Our case is the eldest woman reported to date (aged ten years) and the only known case to report associated hearing loss, leg-length discrepancy, pectus carinatum, respiratory ciliary dyskinesia and late-onset (9 years old) neuro-degenerative regression.

Abstract: Educational attainment is widely used as a surrogate for socioeconomic status (SES). Low SES is a risk factor for hypertension and high blood pressure (BP). To identify novel BP loci, we performed multi-ancestry meta-analyses accounting for gene-educational attainment interactions using two variables, “Some College” (yes/no) and “Graduated College” (yes/no). Interactions were evaluated using both a 1 degree of freedom (DF) interaction term and a 2DF joint test of genetic and interaction effects. Analyses were performed for systolic BP, diastolic BP, mean arterial pressure, and pulse pressure. We pursued genome-wide interrogation in Stage 1 studies (N= 117 438) and follow-up on promising variants in Stage 2 studies (N= 293 787) in five ancestry groups. Through combined meta-analyses of Stages 1 and 2, we identified 84 known and 18 novel BP loci at genome-wide significance level (P< 5 × 10-8). Two novel loci were identified based on the 1DF test of interaction with educational attainment, while the remaining 16 loci were identified through the 2DF joint test of genetic and interaction effects. Ten novel loci were identified in individuals of African ancestry. Several novel loci show strong biological plausibility since they involve physiologic systems implicated in BP regulation. They include genes involved in the central nervous system-adrenal signaling axis (ZDHHC17, CADPS, PIK3C2G), vascular structure and function (GNB3, CDON), and renal function (HAS2and HAS2-AS1, SLIT3). Collectively, these findings suggest a role of educational attainment or SES in further dissection of the genetic architecture of BP.

Abstract: Rapid decline of glomerular filtration rate estimated from creatinine (eGFRcrea) is associated with severe clinical endpoints. In contrast to cross-sectionally assessed eGFRcrea, the genetic basis for rapid eGFRcrea decline is largely unknown. To help define this, we meta-analyzed 42 genome-wide association studies from the Chronic Kidney Diseases Genetics Consortium and United Kingdom Biobank to identify genetic loci for rapid eGFRcrea decline. Two definitions of eGFRcrea decline were used: 3 mL/min/1.73m2/year or more (“Rapid3”; encompassing 34,874 cases, 107,090 controls) and eGFRcrea decline 25% or more and eGFRcrea under 60 mL/min/1.73m2at follow-up among those with eGFRcrea 60 mL/min/1.73m2or more at baseline (“CKDi25”; encompassing 19,901 cases, 175,244 controls). Seven independent variants were identified across six loci for Rapid3 and/or CKDi25: consisting of five variants at four loci with genome-wide significance (near UMOD-PDILT(2), PRKAG2, WDR72, OR2S2) and two variants among 265 known eGFRcrea variants (near GATM, LARP4B). All these loci were novel for Rapid3 and/or CKDi25 and our bioinformatic follow-up prioritized variants and genes underneath these loci. The OR2S2locus is novel for any eGFRcrea trait including interesting candidates. For the five genome-wide significant lead variants, we found supporting effects for annual change in blood urea nitrogen or cystatin-based eGFR, but not for GATMor LARP4B. Individuals at high compared to those at low genetic risk (8-14 vs. 0-5 adverse alleles) had a 1.20-fold increased risk of acute kidney injury (95% confidence interval 1.08-1.33). Thus, our identified loci for rapid kidney function decline may help prioritize therapeutic targets and identify mechanisms and individuals at risk for sustained deterioration of kidney function.

25. Natural human knockouts and Mendelian disorders: deep phenotyping in Italian isolates

Abstract: Whole genome sequencing (WGS) allows the identification of human knockouts (HKOs), individuals in whom loss of function (LoF) variants disrupt both alleles of a given gene. HKOs are a valuable model for understanding the consequences of genes function loss. Naturally occurring biallelic LoF variants tend to be significantly enriched in “genetic isolates,” making these populations specifically suited for HKO studies. In this work, a meticulous WGS data analysis combined with an in-depth phenotypic assessment of 947 individuals from three Italian genetic isolates led to the identification of ten biallelic LoF variants in ten OMIM genes associated with known autosomal recessive diseases. Notably, only a minority of the identified HKOs (C7, F12, and GPR68genes) displayed the expected phenotype. For most of the genes, instead, (ACADSB, FANCL, GRK1, LGI4, MPO, PGAM2, and RP1L1), the carriers showed none or few of the signs and symptoms typically associated with the related diseases. Of particular interest is a case presenting with a FANCLbiallelic LoF variant and a positive diepoxybutane test but lacking a full Fanconi anemia phenotypic spectrum. Identifying KO subjects displaying expected phenotypes suggests that the lack of correct genetic diagnoses may lead to inappropriate and delayed treatment. In contrast, the presence of HKOs with phenotypes deviating from the expected patterns underlines how LoF variants may be responsible for broader phenotypic spectra. Abstract: Fluorescent organic molecules have received great attention due to their largest applications, for example, in DNA and RNA spectroscopies studies, development of new photoluminescence bioprobes, and applications in fluorescence spectroscopy. In specific, purine base analog molecules present high fluorescence quantum yields and significant Stokes shift. Furthermore, the addition of push–pull structures at the purine core could increase the photoluminescence properties, making candidates for photoluminescence bioprobes. To consider this, a complete spectroscopic study was performed on nine push–pull purines, distinguished by different push–pull structures. In specific, for this research, the two-photon absorption (2PA) study showed that the compounds present induced two-photon fluorescence at the therapeutic window, desired for fluorescence microscopy. The brightness property was evaluated, indicating that all chromospheres are fluorescent by a 2PA process. Additionally, ultrafast transient absorption was performed to elucidate contribution of the excited states on the 2PA spectra, and quantum chemistry calculations were performed to corroborate the experimental results.

Abstract: The genomic variation of the Italian peninsula populations is currently under characterised: the only Italian whole-genome reference is represented by the Tuscans from the 1000 Genome Project. To address this issue, we sequenced a total of 947 Italian samples from three different geographical areas. First, we defined a new Italian Genome Reference Panel (IGRP1.0) for imputation, which improved imputation accuracy, especially for rare variants, and we tested it by GWAS analysis on red blood traits. Furthermore, we extended the catalogue of genetic variation investigating the level of population structure, the pattern of natural selection, the distribution of deleterious variants and occurrence of human knockouts (HKOs). Overall the results demonstrate a high level of genomic differentiation between cohorts, different signatures of natural selection and a distinctive distribution of deleterious variants and HKOs, confirming the necessity of distinct genome references for the Italian population.

Abstract: Magnetic nanoparticles based on manganese–zinc ferrite presenting spherical and cubic shapes, verified by small-angle X-rays scattering, were studied by means of the hyper-Rayleigh scattering, the Z-Scan technique in the open-aperture configuration, and a spectrally resolved femtosecond transient absorption setup. Hyper-Rayleigh scattering and Z-Scan experiments were performed with applied magnetic field parallel to the laser polarization state and in the perpendicular direction. The hyperpolarizability of spherical nanoparticles was greater than the cubic ones due to the higher volume of spherical nanoparticles and, therefore, the increased influence of the spin-disoriented layer at the surface on cubic nanoparticles, resulting in a smaller orientational average contribution to the hyperpolarizability. The two-photon absorption cross section was the same for both samples, since the basic constituents are the same and the light absorption is not influenced by the surface anisotropy present on magnetic nanoparticles. For both nanoparticles, the measured optical second harmonic and the nonlinear absorption increased during experiments performed in the presence of external field in the parallel configuration, while a decrease was verified for experiments in the perpendicular case, which demonstrates a crystalline anisotropy on the nonlinear optical properties. Transient absorption measurements revealed an ultrafast relaxation process containing at least two separated dynamical processes, the faster with characteristic time below 1 ps and a longer one, much higher than 100 ps.

30. Microfiber Release to Water, Via Laundering, and to Air, via Everyday Use: A Comparison between Polyester Clothing with Differing Textile Parameters

Abstract: Textiles are one of the major sources of microplastic pollution to aquatic environments and have also been reported in dry and wet atmospheric deposition. There is still a lack of information on the direct release of microfibers from garments to the air and on the influence of textile characteristics including structure, type of yarn, and twist. The present study examines microfiber emissions directly to the air and to water as a consequence of laundering. Polyester garments with different textile characteristics were examined including various material compositions, fabric structure, yarn twist, fiber type, and hairiness. Scaling up our data indicates release of microfibers per person per year to the air is of a similar order of magnitude to that released to wastewater by laundering. The lowest releases to both air and water were recorded for a garment with a very compact woven structure and highly twisted yarns made of continuous filaments, compared with those with a looser structure (knitted, short staple fibers, lower twist). Our results demonstrate for the first time that direct release of microfibers from garments to air as a consequence of wear is of equal importance to releases to water. Currently there is considerable interest in interventions focused on capture from wastewater. However, our results suggest more effective interventions are likely to result from changes in textile design that could reduce emissions to both air and water.

Abstract: Growing evidence supports the potential role of intestinal microbiota in the pathophysiology of inflammatory bowel diseases (IBD) even if the literature does not reveal uniform alterations. The aim of the study was to evaluate the mucosal (MM) and faecal microbiota (FM) composition in a cohort of IBD patients compared to healthy controls (CTRLs). Faecal and mucosal samples were collected from 14 IBD patients and 11 CTRLs. The V1-V3 region of 16S rRNA locus was amplified on a 454-Junior Genome Sequencer. Reads were grouped into operational taxonomic units (OTUs) at a sequence similarity level of 97% for taxonomic assignment, and aligned for OTUs matching against Greengenes database. Irrespective of disease localization and activity, in the MM of IBD patients a statistically significant increase of Proteobacteria (especially Enterobacteriaceae, Acidaminococcus , Veillonella dispar) and decrease of Firmicutes (especially Roseburia and Faecalibacterium prausnitzii) and Actinobacteria was found compared to CTRLs. In the colon district some specific bacterial biomarkers were identified: Enterobacteriaceae for IBD stools, Bacteroides for IBD biopsies, Mogibacteriaceae, Ruminococcaceae and Prevotella for CTRL stools, Ruminococcaceae for CTRL biopsies. The profiles of FM were more similar to CTRLs, suggesting that microbiota adhering to the gut mucosa better discriminates patients from controls, with the identification of some interesting biomarkers. Abstract: Abstract In this work we consider the design of a Free Floating Car Sharing (FFCS) system based on Electric Vehicles. We face the problems of finding the optimal placement of charging stations, and the design of smart car return policies, i.e, how many and where to place charging stations, and whether to ask or not customers to return the car to a charging pole. We leverage actual data containing rentals performed by Car2Go customers. We obtain information for several months worth of actual trips in the city of Turin, our use case. Via trace driven simulations, we replay the exact same trips while simulating electric car based FFCS, to accurately gauge battery discharging and recharging. With this, we compare different charging station placements, also driven by optimisation algorithms. Moreover, we observe the impact of collaborative or selfish car return policies. Results are surprisingly: just as few as 13 charging stations (52 poles) guarantee a fleet of 377 vehicles running in a 1 million inhabitant city to work flawlessly, with limited customer's discomfort. We believe our data driven methodology helps researchers and car sharing providers discerning different design solutions. Abstract: OBJECTIVE: Nasal administration of Streptococcus salivarius 24SMB and Streptococcus oralis 89a has been proposed to reduce the risk of new episodes of adenoiditis, tonsillitis and acute rhinosinusitis in children. PATIENTS AND METHODS: We enrolled 202 children with a recent diagnosis of recurrent upper respiratory tract infection. All the patients were treated twice daily for 7 days each month for 3 consecutive months with a nasal spray whose active agents were two specific bacterial strains: Streptococcus salivarius 24SMB and Streptococcus oralis 89a. Evaluation was performed at the end of treatment and at follow-up at 3, 6, and 12 months. RESULTS: Patients who completed the entire 90-day course of bacteriotherapy and the follow-up period showed a 64.3% reduction in their episodes of upper respiratory tract infections compared to the number of episodes recorded in the previous year. Treatment decreased the reported incidence of infection events by 52.4% in the first 3 months, 31.2% at 6-month follow-up, and 20.8% after 12 months. Enrolled patients tolerated the product well, and there were no dropouts. CONCLUSIONS: Prophylactic bacteriotherapy by administration of Streptococcus salivarius 24SMB and Streptococcus oralis 89a in children with a history of recurrent upper respiratory tract infection could reduce the number of episodes of otolaryngologic infections. Bacteriotherapy can be even more clinically important due to increasing difficulty in finding new effective antibiotic compounds. New alternative therapeutic approaches must be found with, in comparison to antibiotics, greater specificity and safety with respect to patients’ native beneficial flora; lack of drug interactions; the ability to leverage complementary systemic modes of action; and drastically reduced risk of developing resistance within the patient population and the environment. Abstract: This study summarizes the data of multiple surveys carried out at the Isalo Massif (southwestern Madagascar), both within and outside the National Park limits between 2004 and 2014, providing an updated list of the amphibians and non-avian reptiles present within the area. We used an integrative taxonomic approach, combining morphological and molecular data sets to identify collected samples as either described species, or as undescribed candidate species. We here report on one new amphibian candidate species (Mantidactylus sp. aff. multiplicatus Ca65 "Isalo"), and two new amphibian distribution records (Mantella sp. aff. expectata "South", Scaphiophryne sp. aff. calcarata Ca02) compared to the former inventories of the area. Additionally, we provide molecular reference sequences for 23 of the 24 amphibian taxa and for 40 of the 47 reptile taxa known to occur in the area, combining all available sources. With this study, we validate Isalo as an important centre of biodiversity and micro-endemism within Madagascar, with four described endemic amphibian (Gephyromantis azzurrae, Mantella expectata, Mantidactylus noralottae, and Scaphiophryne gottlebei) and one reptile species (Trachylepis nancycoutuae). Of the taxa here listed for Isalo, seven amphibians and six reptiles are new candidate species, and among them at least one amphibian (Mantidactylus sp. aff. multiplicatus Ca65 "Isalo") and three reptiles (Lygodactylus sp. aff. tuberosus Ca02 "Isalo", Paroedura sp. aff. bastardi Ca02 "Isalo" and P. sp. aff. bastardi Ca03 "Ilakaka") are currently known only from Isalo. The unique biodiversity of the Isalo Massif was likely favoured by the confluence of different biomes in the area and/or its role as a refugium during palaeoclimatic oscillations. Our study highlights the crucial importance of applying molecular taxonomic identification in updating the species lists even of previously well surveyed areas, and of maintaining and increasing museum voucher collections as a permanent and accessible testimony of the biodiversity inventory. Abstract: A new and more alarming source of marine contamination has been recently identified in micro and nanosized plastic fragments. Microplastics are difficult to see with the naked eye and to biodegrade in marine environment, representing a problem since they can be ingested by plankton or other marine organisms, potentially entering the food web. An important source of microplastics appears to be through sewage contaminated by synthetic fibres from washing clothes. Since this phenomenon still lacks of a comprehensive analysis, the objective of this contribution was to investigate the role of washing processes of synthetic textiles on microplastic release. In particular, an analytical protocol was set up, based on the filtration of the washing water of synthetic fabrics and on the analysis of the filters by scanning electron microscopy. The quantification of the microfibre shedding from three different synthetic fabric types, woven polyester, knitted polyester, and woven polypropylene, during washing trials simulating domestic conditions, was achieved and statistically analysed. The highest release of microplastics was recorded for the wash of woven polyester and this phenomenon was correlated to the fabric characteristics. Moreover, the extent of microfibre release from woven polyester fabrics due to different detergents, washing parameters and industrial washes was evaluated. The number of microfibres released from a typical 5 kg wash load of polyester fabrics was estimated to be over 6,000,000 depending on the type of detergent used. The usage of a softener during washes reduces the number of microfibres released of more than 35%. The amount and size of the released microfibres confirm that they could not be totally retained by wastewater treatments plants, and potentially affect the aquatic environment. [ABSTRACT FROM AUTHOR]

Abstract: Elevated serum urate levels cause gout and correlate with cardiometabolic diseases via poorly understood mechanisms. We performed a trans-ancestry genome-wide association study of serum urate in 457,690 individuals, identifying 183 loci (147 previously unknown) that improve the prediction of gout in an independent cohort of 334,880 individuals. Serum urate showed significant genetic correlations with many cardiometabolic traits, with genetic causality analyses supporting a substantial role for pleiotropy. Enrichment analysis, fine-mapping of urate-associated loci and colocalization with gene expression in 47 tissues implicated the kidney and liver as the main target organs and prioritized potentially causal genes and variants, including the transcriptional master regulators in the liver and kidney, HNF1Aand HNF4A. Experimental validation showed that HNF4A transactivated the promoter of ABCG2, encoding a major urate transporter, in kidney cells, and that HNF4A p.Thr139Ile is a functional variant. Transcriptional coregulation within and across organs may be a general mechanism underlying the observed pleiotropy between urate and cardiometabolic traits.

Abstract: Chronic kidney disease (CKD) is responsible for a public health burden with multi-systemic complications. Through trans-ancestry meta-analysis of genome-wide association studies of estimated glomerular filtration rate (eGFR) and independent replication (n= 1,046,070), we identified 264 associated loci (166 new). Of these, 147 were likely to be relevant for kidney function on the basis of associations with the alternative kidney function marker blood urea nitrogen (n= 416,178). Pathway and enrichment analyses, including mouse models with renal phenotypes, support the kidney as the main target organ. A genetic risk score for lower eGFR was associated with clinically diagnosed CKD in 452,264 independent individuals. Colocalization analyses of associations with eGFR among 783,978 European-ancestry individuals and gene expression across 46 human tissues, including tubulo-interstitial and glomerular kidney compartments, identified 17 genes differentially expressed in kidney. Fine-mapping highlighted missense driver variants in 11 genes and kidney-specific regulatory variants. These results provide a comprehensive priority list of molecular targets for translational research.

Abstract: In this work, we propose the synthesis of a novel bromine chalcone (E)-3-(2-bromophenyl)-1-(2-((phenylsulfonyl)amine))phenyl)prop-2-en-1-one (BRC) that has been crystallized by the slow evaporation technique. The second-order molecular optical scattering and two-photon absorption(2PA) spectrum of the BRC molecule dissolved in dimethyl sulfoxide (DMSO) were evaluated by using hyper-Rayleigh scattering and the femtosecond tunable Z-scan techniques. The first-order hyperpolarizability of BRC dissolved in DMSO was estimated by using a simplified two-level model, in which one- and two-photon absorption parameters were used as input information to the model. The BRC crystal was characterized from single-crystal X-ray diffraction (XRD) and spectroscopy analyzes. Also, the thermogravimetric analyses and the fluorescence spectra were obtained. In addition, an ab initiocalculation method, which includes the Møller–Plesset perturbation theory (MP2) and the density functional theory (DFT) at the CAM-B3LYP level, was used to estimate the crystal linear refractive index and the third-order electric susceptibility. Also, the average first hyperpolarizability of BRC molecules dissolved in DMSO was calculated and compared with the experimental results. The obtained values are good and qualify the BRC crystal as a potential candidate for application in nonlinear optical devices.

Abstract: Plastic pollution is a remarkable environmental issue. In fact, plastic is widespread in the lifetime and serious environmental problems are caused by the improper management of plastic end of life, being plastic litter detected in any environment. Efforts are put to implement the development of sustainable and circular materials. In this scenario, biodegradable polymers, BPs, are promising materials if correctly applied and managed at the end of life to minimize environmental problems. However, a lack of data on BPs fate and toxicity on marine organisms, limits their applicability. In this research, the impact of microplastics obtained from BPs, BMPs, were analyzed on Paracentrotus lividus. Microplastics were produced from five biodegradable polyesters at laboratory scale by milling the pristine polymers, under cryogenic conditions. Morphological analysis of P. lividus embryos exposed to polycaprolactone (PCL), polyhydroxy butyrate (PHB) and polylactic acid (PLA) showed their delay and malformations, which at molecular level are due to variation in expression levels of eighty-seven genes involved in various cellular processes, such as skeletogenesis, differentiation and development, stress, and detoxification response. Exposure to poly(butylene succinate) (PBS) and poly(butylene succinate-co-adipate) (PBSA) microplastics showed no detectable effects on P. lividus embryos. These findings contribute with important data on the effect of BPs on the physiology of marine invertebrates. 47. Protective effect of Rumenic acid rich cow's milk against colitis is associated with the activation of Nrf2 pathway in a murine model.

Abstract: Dietary supplementation with pure cis9, trans11 isomer of Conjugated Linoleic Acid –known as Rumenic Acid (RA)– improves cytoprotective defenses downstream through the activation of nuclear factor-E2-related factor-2(Nrf2). This capability, when Rumenic Acid is consumed in the form of foods, is still unknown. The ability of standard ( St ) or cow milk naturally-enriched in RA ( En ) to activate Nrf2 pathway and its impact on dextran sodium sulfate (DSS)-induced colitis was comparatively evaluated. Activity of Nrf2 pathway was investigated in colonic tissue of BALB/c mice, receiving 4-week supplement with skimmed milk ( SK ), St or St reinforced with pure RA ( RSt ) providing increasing RA dose (0, 124 or 404 mg RA/kg -1 b.w, respectively). Next, the anti-oxidant/ anti-inflammatory effect produced by St or En treatment (383 mg RA/kg -1 b.w.) was explored. Finally, macroscopic and histomorphologic features of colitis were evaluated in animals challenged with 5% (w/v) DSS, at the end of St or En treatment. Significant activation of Nrf2 pathway is associated with RSt and En intake (P<0.05), but not with SK or En treatment. En pre-treatment offers better protection, in comparison with St , against pro-oxidant, pro-inflammatory signs (P<0.01) and macroscopic signs triggered by DSS. It can be concluded that Nrf2 activation by higher RA amount contained in En is, at least in part, responsible for the improved protection associated with En intake against DSS-induced colitis. Abstract: Background Altered body composition is frequently observed in Crohn’s disease (CD) patients. Aims To investigate the nutritional status, and the effect of different therapeutic regimes in adult CD patients. Methods Fat free mass (FFM) and BIA-derived phase angle (PhA) were assessed in 45 CD patients, 22 on conventional therapy (CT) and 23 on maintenance therapy with infliximab (MT). Nutritional status was also assessed in 12 CD patients before and following the induction protocol with infliximab. BIA data of CD patients were compared with those of 20 healthy asymptomatic volunteers. In CD patients C Reactive Protein (CRP) and albuminaemia dosage were obtained. Results The mean values of PhA and of FFM were significantly lower in CT patients when compared with control group and MT patients. Following infliximab treatment FFM increased, although not significantly, while mean phase angle value significantly increased from 4.6 ± 0.3 to 6.2 ± 0.4 (p < 0.05). CRP was significantly lower in MT patients compared to that in CT patients. Conclusion CD patients on conventional therapy showed a lower FFM and a lower mean phase angle score compared to those on infliximab therapy. Following infliximab treatment the mean phase angle score normalized. PhA is a reliable nutritional indicator in IBD patients and could be considered as an additional tool for assessing response to treatment. [ABSTRACT FROM AUTHOR]
