Sexual health is important for the quality of life of patients who have received haematopoietic stem-cell transplantation (HSCT). Sexual dysfunction and couple dissatisfaction can seriously affect a patient's recovery and treatment process. However, this aspect of post-transplantation recovery is still usually neglected in clinical practice. In this Series paper, we aim to elucidate the emotional and psychosocial factors affecting the sexual function in these patients, with a special focus on the partner's role and the psychological consequences of some adverse effects of HSCT. Moreover, we provide an overview of the management approaches and assessment tools of psychological issues associated with sexual dysfunction reported in the literature. These tools can help clinicians in this field to plan essential lifestyle and clinical interventions to help their patients. In conclusion, screening for psychological issues is indispensable when approaching sexual dysfunction in patients with HSCT. Health-care teams in transplantation units should be trained to discuss this aspect of recovery and provide the required treatment and follow-up plan.

Letermovir is the first approved drug for cytomegalovirus (CMV) infection prophylaxis in adult patients who are CMV positive undergoing allogeneic hematopoietic cell transplantation (allo-HCT). Because CMV infection risk varies from patient to patient, we evaluated whether a risk-based strategy could be effective. In this single-center study, all consecutive adult patients who were CMV positive and underwent allo-HCT between 2015 and 2021 were included. During period 1 (2015-2017), letermovir was not used, whereas during period 2 (2018-2021), letermovir was used in patients at high risk but not in patients at low risk, except in those receiving corticosteroids. In patients at high risk, the incidence of clinically significant CMV infection (csCMVi) in period 2 was lower than that in period 1 (P < .001) by week 14 (10.5% vs 51.6%) and week 24 (16.9% vs 52.7%). In patients at low risk, although only 28.6% of patients received letermovir in period 2, csCMVi incidence was also significantly lower (P = .003) by week 14 (7.9% vs 29.0%) and week 24 (11.2% vs 33.3%). Among patients at low risk who did not receive letermovir (n = 45), 23 patients (51.1%) experienced transient positive CMV DNA without csCMVi, whereas 17 patients (37.8%) experienced negative results. In both risk groups, the 2 periods were comparable for CMV disease, overall survival, progression-free survival, relapse, and nonrelapse mortality. We concluded that a risk-based strategy for letermovir use is an effective strategy which maintains the high efficacy of letermovir in patients at high risk but allows some patients at low risk to not use letermovir.

•The risk-based letermovir use strategy is effective for CMV infection prophylaxis and allows some patients at low risk not to use letermovir.•Among patients at low risk without letermovir, most had negative or transient positive CMV DNA without clinically significant CMV infection.

Reduced intensity conditionings (RIC) before after allogeneic hematopoietic stem cell transplantation (HSCT) allow older or unfit patients of being transplanted, but survival expectancy and burden of late complications are poorly described in this setting. All patients (N= 456) who were alive and relapse-free 2 years after HSCT following RIC were included. Cumulative incidences (CI), standardized incidence, or mortality, ratio (SIR or SMR), and competing risk models were used. The 10-year CIs of relapse and non-relapse mortality incidences were 13.9 and 13.4%, respectively. Seventy-eight patients died, late relapse being the most frequent cause of death leading to a SMR of 6.38 (95% CI, 5.1–8.0; p< 0.001). Among non-relapsing patients (n= 412), 30 died (SMR 4.38; 95% CI, 3.3–5.8: p< 0.001). A total of 37 patients developed 41 SM leading to a 10-year cumulative incidence of 12.9%, and a significant SIR relative to the general population (1.4). Finally, we found high CI of cardiovascular (CVC) and venous thromboembolic complications (VTE) (10-year CI; 15.1% and 11.7%, respectively). Older age was the only significant risk factor for CVC and VTE in multivariable analysis. In conclusion, with life expectancy rate of 70%, late survivors after RIC warrants long-term follow-up and active intervention on averting cardiovascular disease and screening cancers.

Cell lines are essential models for biomedical research. However, they have a common and important problem that needs to be addressed. Cell lines can be misidentified, meaning that they no longer correspond to the donor from whom the cells were first obtained. This problem may arise due to cross-contamination: the accidental introduction of cells from another culture. The contaminant, which is often a rapidly dividing cell line, will overgrow and replace the original culture. The end result is a false cell line, also known as a misidentified or imposter cell line. False cell lines may come from an entirely different species, tissue, or cell type than the original donor. If undetected, false cell lines produce unreliable and irreproducible results that pollute the biomedical literature and threaten the development of reliable drug discovery and meaningful patient treatments.The goal of this study was to ascertain how widespread this problem is and how it affects the literature, as well as to estimate how much funding has been used to produce pools of scientific literature of questionable value. We focus on HEp-2 [HeLa] and Intestine 407 [HeLa], two false cell lines that are widely used in the scientific literature but were shown to be cross-contaminated in 1967. These two cell lines have been used in 8497 and 1397 published articles and extensively described as laryngeal cancer and normal intestine, respectively, rather than their true identity: the cervical cancer cell line HeLa. Discussed are tools, approaches, and resources that can address this issue—both retrospectively and prospectively.

Although metformin is increasingly being used in women with type 2 diabetes during pregnancy, little data exist on the benefits and harms of metformin use on pregnancy outcomes in these women. We aimed to investigate the effects of the addition of metformin to a standard regimen of insulin on neonatal morbidity and mortality in pregnant women with type 2 diabetes.

Research in toxicology relies on in vitromodels such as cell lines. These living models are prone to change and may be described in publications with insufficient information or quality control testing. This article sets out recommendations to improve the reliability of cell-based research.

Although still experimental, natural orifice translumenal endoscopic surgery (NOTES) aims to use the natural orifices for intraabdominal surgery. Pure transvaginal umbilical hernia repair has been reported. However, mesh protection devices were used to minimize mesh contamination during mesh insertion. The authors believe that before widespread implementation of this technique, more foundational research is indicated to establish the sterility of hernia mesh insertion through this route. This prospective study aimed to compare transvaginal ventral hernia mesh insertion sterility with laparoscopic trocar-site insertion sterility to establish baseline data to help promote the safety of NOTES tranvaginal hernia repair. This was a prospective descriptive study (Canadian Task Force classification 2A). With institutional review board approval, 10 patients undergoing laparoscopic surgery for benign gynecologic disease were enrolled in the study. Atrium Prolite mesh (polypropylene monofilament) was inserted into the vagina before and after standard surgical preparation with 10 % povidone–iodine. As a control, mesh also was inserted through a prepped laparoscopic port site. The mesh was cultured for bacterial, fungal, and viral contamination. All patients received standard infection prophylaxis that included preoperative intravenous cefazolin and metronidazole. The unprepped vaginal canal was cultured and demonstrated normal multiorganism vaginal flora in all 10 cases. Of the 10 skin incision mesh samples, 3 (30 %) grew bacteria, including Staphylococcus lugdunensis, a potentially pathogenic organism. In contrast, none of the prepped vaginal mesh specimens yielded any growth of microorganisms or potential pathogens. This study showed that a surgically prepped vaginal canal can be a sterile conduit for insertion of polypropylene mesh for transvaginal ventral hernia repair without the use of additional mesh protection. Surprisingly, the prepped vaginal conduit in our patients was more sterile than a prepped skin incision.

The land subsidence which occurs at the Larissa Basin (Thessaly Plain, Central Greece) is due to various causes including aquifer system compaction. Deformation maps of high spatial resolution deduced by the Persistent Scattering Interferometry (PSI) technique (using radar scenes from ERS and ENVISAT satellites) for the period 1992–2006 were produced to study the spatial and temporal ground deformation.

Hunting high and low: A cyanide‐bridged rhombic mixed‐spin FeII–FeIIcomplex that features low‐spin (LS) and high‐spin (HS) FeIIcenters was prepared by self‐assembly including the decomposition of SeCN. The structural motif represents a hybrid [2×2]grid/metallocyclic architecture (see picture).

The application of conventional SAR Interferometry (InSAR) together with the two techniques of subcentimeter accuracy, the Stacking and the Permanent Scatterers (PS) Interferometry, were used to study the ground deformation in the broader area of Athens for the period 1992 to 2002. Using the Stacking interferometric method, 55 ERS-1&2 SAR scenes, between 1992 and 2002, were acquired producing 264 differential interferograms. Among these only 60 were finally selected as fulfilling certain criteria. The co-seismic deformation associated with the Athens Earthquake (Mw= 5.9, September 7, 1999) was excluded from the analytical procedure in an attempt to present results of only aseismic character. In total ground subsidence results of about 12 mm in the southern suburbs of Athens, but higher value of about 40 mm in the northern ones for the period 1992–2002. Based on the PS technique, a precise average annual deformation rate-map was generated for the period 1992–1999, ending just before the Athens earthquake event. Both circular and elongated-shape areas of subsidence are recognizable especially in the northern part of the Athens Basin (3–4 mm/yr), as well as at its southern part (1–3 mm/yr). In addition, a rate of 2–3 mm/yr is also yielded for some part of the Athens city center. Subsidence rates of 1–2 mm/yr are measured at the western part of the basin over an area of old mining activities, and around the newly built Syntagma Metro Station. The correlation of the observed deformation patterns with respect to the spatial distribution of water pumping, older mining activities, metro line tunneling and other local geological parameters is examined and discussed.

We present a simple and versatile method for linking single wall carbon nanotubes (SWNT) together through DNA by non-covalent chemistry using streptavidin-biotin recognition complex. Streptavidin coated SWNTs are reacted with biotin or bis-biotin ended DNA double strands leading to SWNT-DNA and SWNT-DNA-SWNT adducts in high yield. This method avoids strong acidic treatment of SWNTs prior to functionalization as usually required in covalent routes. Complementary characterizations by gel electrophoresis and AFM demonstrated the efficiency of the present binding method. In addition, SWNTs bound to DNA can be aligned on a substrate using the combing properties of DNA strands, bringing a new tool into the toolkit for self-assembling SWNTs onto surfaces. Research has looked at refining scaffold architecture to promote more directed growth of tissues through well-defined anisotropy in the pore structure. In many cases it is also desirable to incorporate therapeutic ingredients, such as growth factors, into the scaffold so that their release occurs as the scaffold degrades. Therefore, scaffold fabrication techniques must be found to precisely control, not only the overall porosity of scaffolds, but also the pore size, shape and spatial distribution.The design of tissue engineering scaffolds must take into account many factors including successful vascularisation and the growth of cells. Materials presenting a stable and reversible switch of physical properties in the solid state are of major interest either for fundamental interests or potential industrial applications. In this context, the design of metal complexes showing a light‐induced crossover from one spin state to another, leading to a major change of magnetic and optical properties, is probably one of the most appealing challenges. The so‐denoted spin‐crossover materials undergo, in some cases, a reversible photoswitch between two magnetic states, but, unfortunately, lifetimes of the photomagnetic states for compounds known so far are long enough only at low temperatures; this prohibits any applications. We have measured and collected the temperatures above which the photomagnetic effect disappears for more than sixty spin‐crossover compounds. On the basis of this large data base, a correlation between the nature of the coordination sphere of the metal and the photomagnetic lifetime can be drawn. Such correlation allows us to propose here a general guideline for the rational design of materials with long‐lived photomagnetic lifetimes. This result clearly opens the way towards room‐temperature photonic materials, based on the spin‐crossover phenomenon, which will be of great interest for future communication devices.

Doublecortin (DCX) is a microtubule‐associated protein widely expressed in the developing mammalian nervous system and important for neuronal migration. DCX is known to belong to a novel protein family defined by sequence homology and the presence of a conserved microtubule‐binding domain, but the functions of other members of this family are still undefined. In this study, we describe the cloning of the chick ortholog of doublecortin‐like kinase (DCLK), a member of this family, and assess the expression of DCX and DCLK in the layered regions of the developing chick brain. DCX and DCLK are widely expressed in pallial and subpallial structures, including the telencephalon, optic tectum, and cerebellum, in similar distribution patterns. In addition to their expression in migrating cells, both proteins were also detected in the ventricular zone and in postmigratory Purkinje cells. Finally, DCX and DCLK were found to be coexpressed in all areas examined. In postmigratory Purkinje cells, DCX and DCLK both colocalized to the cell membrane, although DCLK was also distributed more generally throughout the cell soma. These data are consistent with multiple roles for DCX and DCLK in the developing chicken brain and suggest that the chick cerebellum will be an intriguing system to explore the effects of DCX and DCLK on postmigratory neuronal function. Doublecortin (DCX) is a 40 kDa microtubule-associated protein required for normal neural migration and cortical layering during development. Mutations in the human DCX gene cause a disruption of cortical neuronal migration. Defects in cdk5 (cyclin-dependent kinase 5) also cause defects in neural migration and cortical layering. DCX is a substrate for cdk5 in vitro and in vivo and the major site of in vitro phosphorylation is Ser-297. We used a highly developed MS strategy to identify the cdk5 phosphorylation sites and determine the major and minor sites. Several phosphopeptides were identified from a tryptic digest of 32P-labelled, cdk5-phosphorylated DCX using a combination of off-line HPLC and matrix-assisted laser-desorption ionization-MS with alkaline phosphatase treatment. Tandem MS/MS enabled the identification of seven phosphorylation sites for cdk5. Monitoring of 32P label indicated that there was one major site, Ser-28, at the N-terminus, and a major site, Ser-339, in the serine/proline-rich domain at the C-terminus. Five other sites, Ser-287, Thr-289, Ser-297, Thr-326 and Ser-332, were also found in the tail. Site-directed mutagenesis largely supported these findings. Single mutation of Ser-28 reduced but did not abolish phosphorylation. Double, rather than single, mutation for Ser-332 and Ser-339 was required to reduce overall phosphorylation, suggesting an interaction between these sites. Truncations of the tail produced a significant reduction in cdk5 phosphorylation of DCX. These results do not support Ser-297 as the major cdk5 phosphorylation site in DCX, but indicate that DCX is subject to complex multisite phosphorylation. This illustrates the importance of a well-developed MS strategy to identify phosphorylation sites.

Line shape spectroscopy is a valuable tool both for diagnostic purposes, and for understanding the basic atomic processes in the boundary region of magnetically confined fusion plasmas. We report on the present state of a versatile line shape model including Stark, Zeeman and Doppler broadening for an arbitrary emitter in the divertor or other edge plasmas. Several examples of temperature or density diagnostics based on the use of this model are presented for neutral and ionized carbon, and for hydrogen isotopes. In the case of the Dα line, fitting the model to the experimental spectra involves the determination of several parameters and we have thus used an efficient fitting technique based on a genetic algorithm. The results of this analysis, suggesting the existence of a population of neutrals of several hundreds of electronvolts, demand a more detailed study of the line wings. With the help of a simple model, we show that this part of the spectrum might indeed be sensitive to the turbulent fluctuations ubiquitous in edge plasmas.

Passive spectroscopy of the Dα line emission is used for the investigation of the Tore-Supra edge plasma in front of a neutralizer plate of the Ergodic Divertor. Two kinds of spectra are observed: symmetric and asymmetric. Symmetric spectra are fitted considering two isotropic populations of deuterium representing the main neutral production mechanisms. The fit of the asymmetric spectra, requires the addition of an anisotropic population resulting from the reflection of ions as neutrals on the V-shaped notches of the neutralizer plate and the inclusion of the relaxation of the deuterium populations through elastic collisions with the plasma ions.

The deuterium Dα line emitted in front of an equatorial neutraliser plate in Ergodic Divertor Tore Supra plasmas is a common diagnostic of recycling particles in present Tokamaks. First, the possible perturbation of the Dα spectral lineshape by the nearby HeII line Brβ has been studied. Although its effect on the Dα line profile shape is evaluated to be non negligible in plasmas with large Helium contents, it is concluded that it should be of no influence in usual fusion plasmas. Second, a detailed study of the Dα spectral lineshape has been performed to obtain temperatures and densities of the different neutral deuterium species. A synthetic profile method taking into account Zeeman and Doppler effects has been used. This shows that two deuterium populations with specific behaviours have to be considered: the first one (‘cold atoms’) is composed of atomic deuterium coming from D2 desorbed from the neutraliser plate, while the second one (‘warm atoms’) is composed of charge exchange (CX) and reflected deuterium. The Doppler temperature of cold atoms increases with the edge electron density, n^edgee (1.4 eV &lt; T^cD &lt; 2.5 eV). The possible heating of cold atoms by D⁺ ions is evaluated to explain this effect. Molecular and atomic deuterium densities behaviour versus edge electron temperature Te are analysed with the use of the warm and cold Dα brightnesses coupled to a collisional radiative deuterium model. The results are compared to the numerical code EDCOLL. An important molecular density (> 10¹⁸ m^-3) is found for the whole range of edge electron temperature (10 eV &lt; Te &lt; 40 eV) while the ratio of the atomic to the molecular densities is around 1.8. The density of atoms coming from molecular dissociation is ~ 0.8 times the density of atoms created by charge exchange or re.ection of deuterons on the neutraliser.
