Amaç: Bu çalışmada Japon bıldırcını (Coturnix coturnix japonica) rasyonuna nane yağı eklenerek karaciğer üzerine histopatolojik etkileri ışık mikroskobu ile incelenerek gelecekteki benzer çalışmalara ışık tutması amaçlanmıştır. Gereç ve Yöntem: Çalışmada rasyonlarına artan oranda nane yağı ilave edilen, 10 günlük, 40 adet Japon bıldırcını kullanıldı. Her birinde 5 erkek 5 dişi olacak şekilde 10’ar civcivlik 4 gruba ayrıldı, Gruplar; rasyonları- na izokalorik ve izonitrojenik olacak şekilde, kontrol grubu %0.3 ayçiçek yağı (AY)+%0.0 nane yağı (NY), diğer üç grup; %0.2(AY)+ %0.1(NY); %0.1(AY)+%0.2(NY) ve %0.0(AY)+%0.3 (NY) ad libitum ilave edilerek düzenlendi. Kesim sonrası, karaciğerden rutin doku takibi ile hazırlanan parafin bloklardan, rotary mikrotomla kesitler alındı. Hematoksilen-Eozin ile boyanan preparatlar ışık mikroskobunda incelendi. Bulgular: Karaciğerde septum interlobaris ile bölünen lobcuk yapısı görülmedi. Az miktarda inter lobuler intersitisyum ve buna bağlı olarak, bilinenin aksine, hepatik lopçuklarda birarada olması beklenen, üçlü yapı(trias hepatica) dağınıktı. Rasyonda artan yağ oranına paralel olarak basit hepatosteatoz meydana geldiği görüldü. Öneri: Antimikrobiyal ve immunmodülatör etkisiyle sağlıklı bir yetiştiricilik imkanı sunmasının yanı sıra protein sentezi ve yemden yararlanmayı arttırarak verimin yükseltilmesine katkı sağlayan nane yağının sayılan bu olumlu özelliklerinden istifade edilebilmesi için uygun dozda verilmesi gerektiği görülmüştür., Aim: In the present study, peppermint oil was added to the ration of Japanese quail (Coturnix coturnix japonica) and it was aimed to shed light on similar studies in the future by examining its effect on liver tissue by light microscopy. Materials and Methods: In the study, 10-day old, 40 Japanese quails were used. There were 4 groups of 10 chicks, each with 5 males and 5 females. Groups; Control group 0.3% sunflower oil (SO)+0.0% peppermint oil (PO), the other three groups 0.2% (SO) +0.1% (PO); 0.1% (SO)+ 0.2% (PO) and 0.0% (SO)+0.3% (PO). Created by adding oil ad libitum. From the paraffin blocks prepared by routine tissue follow-up from the liver, sections were taken with a rotary microtome. Hematoxylin-Eosin stained preparations were examined under a light microscope. Results: The lobule structure divided by the septum interlobaris was not seen in the liver. A small amount of inter lobuler interstitium and consequently the triple structure (trias hepatica) expected to coexist in the hepatic lobes, contrary to what is known, was scattered. It was observed that simple hepatosteatosis occurred in parallel with the increasing fat ratio in the diet. Conclusion: It has been observed that peppermint oil, which contributes to increase the yield by increasing protein synthesis and feed utilization, as well as providing a healthy breeding opportunity with its antimicrobial and immunomodulatory effect, should be given at an appropriate dose in order to benefit from these positive properties.

Amaç: Karbapenemler, Acinetobacter baumannii infeksiyonlarının tedavisinde kullanılan en önemli geniş spektrumlu antibiyotiklerdir. Fakat dünya genelinde karbapenemlere direnç gittikçe artmaktadır. Bu çalışmamızda, karbapenem direnci saptanan A. baumannii suşlarında gözlenen OXA tipi karbapenemaz genlerinin dağılımını, gerçek zamanlı polimeraz zincir reaksiyonu (PCR) yöntemiyle incelemeyi ve epidemiyolojik veri sunmayı amaçladık. Yöntemler: Ocak 2016-Ocak 2018 arasında klinik örneklerden izole edilen karbapeneme dirençli 20 A. baumannii suşu çalışmaya dahil edildi. Bu suşlardan DNA izolasyonları gerçekleştirildi ve OXA-23, OXA-24, OXA-51 ve OXA-58 karbapenemazlarının genlerine spesifik primer ve TaqMan probları kullanılarak gerçek zamanlı PCR yöntemiyle OXA karbapenemaz genlerinin dağılımı incelendi. Bulgular: Karbapeneme dirençli A. baumannii suşlarının 16 (%80)’sının duyarlı olduğu tigesiklin bu suşlara bağlı infeksiyonlar için en iyi seçenekti. Tüm suşlarda blaOXA-51 saptanırken, blaOXA-24 sadece 1 (%5) suşta saptandı. Yirmi suştan 10 (%50)’unda blaOXA-23 ve blaOXA-51 birlikteliği vardı. Sonuçlar: Karbapeneme dirençli A. baumannii suşlarındaki OXA tipi karbapenemazların dağılımında blaOXA-23 ve blaOXA-51 birlikteliğinin daha sık olması dikkati çekmiştir. Gerçek zamanlı PCR gibi hızlı ve güvenilir sonuç verebilecek tekniklerle, böyle dirençli suşların rutin sürveyansları yapıldığında değerli epidemiyolojik veriler elde edilebilir. The objectives of this study was to investigate the presence of Staphylococcus aureus, distribution of classical staphylococcal enterotoxin (SE) SEA to SEE, relevant gene/s and antimicrobial resistance pattern of S. aureus isolated from traditionally produced raw milk cheeses. A total of 106 fresh white cheese samples were examined. The 25 (23.6 %) of 106 cheese samples were found to be contaminated with coagulase positive staphylococci (CPS). From 52 isolates identified as S. aureus, one or more SEs was detected in 38.4 % of the isolates by ELISA whereas one or more se genes were detected in 50 % of the isolates by RT PCR. SEE (75 %) and see gene (61.5 %) were detected most frequently, whereas SED and sed gene were not detected in any isolates. Overall, 63.5 % of isolates were resistant to antimicrobial agents with 59.6 %, 13.5 %, 5.8 %, 5.8 % and 3.8 % of the isolates were resistant to penicillin, erythromycin, tetracycline, cefoxitin and kanamycin, respectively. The results of this study have revealed that cheeses made from raw milk were highly contaminated with S. aureus, therefore, creates a risk for public health due to the presence of enterotoxins as well as resistant strains against antimicrobial agents.

Amaç: S.epidermidis biyofilmoluşumu, bu bakterilerin antibakteriyel ilaçlara ve bağışıklık sistemisavunmasına karşı koruyan önemli bir bariyer görevi oluşturmaktadır. IcaA genibunun oluşumu için önemlidir. Lactobacillus türlerinin probiyotik etkileribilinmektedir. Bizde çalışmamızda, CRA yöntemi ile biyofilm pozitifliğisaptanan S. epidermidis kökenlerinin icaA gen ekspresyonuna, L. acidophilus ATCC 4356 kökenininin-vitro etkisinin incelenmesi amaçladık.Materyal ve metod: Haziran – Aralık 2017 döneminde klinik örneklerdenizole edilmiş olan ve CRA metodu ile biyofilm üretimi pozitif olarak saptananyirmi (20) S. epidermidis kökeniçalışmamıza dahil edildi. CRA’da siyah pozitif kolonilerden, 2 McFarland’lıksüspansiyonundan 250 mLalınarak, L. acidophilus içeren ve içermeyen 2 tüpe ilave edildi ve %5CO2’liortamda 37oC’da inkübasyona bırakıldı. Staphylococcus aureus, kommensal bir bakteri olarak hem insanla birlikte yaşayan, hem de uygun fırsatı bulduğunda ciddi fırsatçı enfeksiyonlara neden olabilen önemli bir patojendir. Bifidobacterium cinsi bakteriler probiyotik olarak kullanılan ana mikroorganizmalardır. Bu cinslere ait çok sayıda türün, konağın sağlığını iyileştirmede güvenli ve etkili olduğu bildirilmiştir. Bizim çalışmamızın amacı, probiyotik etkili Bifidobacterium longum ATCC 15707’nin, Staphylococcus aureus ATCC 29213’e suşları üzerine etkisinin Real-Time PCR yöntemi ile araştırmaktır. 3 tane brain–heartin fusion broth (Oxoid) besiyeri içeren tüp hazırlandı. Tüm tüplere 103 CFU/mL olacak şekilde Staphylococcus aureus ATCC 29213 kökeninden ilave edildi. 1. tüp kontrol olarak kalırken, 2. tüpe 103 CFU/mL olacak şekilde, 3. tüpe de 106 CFU/mL olacak şekilde Bifidobacterium longum ATCC 15707 kökeni ilave edildi. İnkübasyonun 6., 12., 24. saatlerinde tüplerden alınan numunelerden DNA izolasyonu yapıldı ve bu numunelerden LightCycler 480 sisteminde Real-Time PCR ile kantitatif olarak S. aureus miktarları saptandı. Birinci tüpte 6. saatte 523.333+8993 kopya/mL düzeyinde saptanan S. aureus, farklı tüplerde sırasıyla 103 ve 106 CFU/mL miktarlarında B. longum kökeni ile birlikte inkübe edildiğinde bu miktarların düştüğü olduğu bulundu. Sonuç olarak, çalışmamızda, probiyotik etkili Bifidobacterium longum’un, Staphylococcus aureus’un in vitro ortamda üremesini etkilediği ve bakteri sayısının etki düzeyini arttırdığı saptanmıştır. Probiyotiklerin etkilerinin gnotobiyotik hayvan modellerinde veya daha kapsamlı çalışmalarla incelenerek, ilaç dirençlerinin çok yoğunlaştığı günümüzde, önemli patojenlere karşı antimikrobiyal olarak yarar sağlayabileceği düşüncesindeyiz.

İnsan norovirüsü (HNoV), çevresel etkenlere oldukça dirençli bir RNA virüsüdür ve akut viral gastroenteritin nedeni olan ana etkenlerden birisidir. Hızlı evrim yeteneği nedeniyle 7 genogrubu vardır ve bunlardan GI, GII ve GIV insan enfeksiyonları ile ilişkilidir. Sular HNoV için salgın aracı olarak tanımlamaktadır. Bu bilgiler ışığında, çalışmamızda, lokal kuyular ve derelerden alınan su numunelerinde özellikle insanlarda enfeksiyonları ile ilişkili HNoV genogrup (G)I, GII ve GIV varlığının gerçek zamanlı polimeraz zincir reaksiyonu (real-time PCR) ile tanımlanarak moleküler epidemiyolojik bir veri sağlanması amaçlanmıştır. Çalışma için lokal kuyulardan ve derelerden, Ocak 2017 – Ocak 2018 döneminde toplanan 60 adet su numunesi çalışmamıza dahil edildi. RNA izolasyonu ve cDNA sentezi sonrası HNoV GI, GII ve GIV spesifik primer problar ile LightCycler 480 sisteminde real-time PCR yöntemi ile çalışıldı ve sonuçlar değerlendirildi. Çalışmamıza dâhil edilen 60 numunede, HNoV GII’nin %10 yüzeyinde saptandığı, bunu sırasıyla GI (%5) ve GIV (%1.67) varlığının takip ettiği tespit edildi. 10 numunede (%16.67) HNoV GI, GII ve GIV pozitifliği bulundu. Lokal kuyulardan 3 (%8.57) tanesinde ve derelerden alınan numunelerden de 7 (%28) tanesinde pozitiflik saptandı. Sonuç olarak, ülkemizde ilk defa kuyu suları ve derelerden alınan sularla yaptığımız çalışmamızla, moleküler epidemiyolojik olarak HNoV varlığını saptadık. HNoV’lar arasında GII’nin ön planda tutulması gerektiğini ama GI ve GIV’ünde bulunduğunu tespit ettik. HNoV için salgınlarında sular göz önünde bulundurulmalı ve gelişen moleküler tekniklerle, sular gibi önemli enfeksiyon kaynaklarından epidemiyolojik veriler sağlanarak durum ortaya konabilir ve bu bilgiler ile bölgesel aşı geliştirme çalışmaları içinde ön veriler sağlanabileceği kanaatindeyiz.

Amaç: Giardia intestinalis flagellalı, Giardiyaz’a neden olan bir protozoondur ve dünya çapında önemli bir sorundur. Moleküler yöntemlerle sekiz farklı genotipi saptanan G. intestinalis’de, A ve B genotipinin, insan ve memelilerde hastalıklarla ilişkili olduğu ve farklı genotiplerin, farklı klinik tablolar meydana getirebildiği bildirilmektedir. Biz de bu bilgiler ışığında, giardiyaz tanısı almış ve G. intestinalis pozitif saptanan dışkı örneklerinde bulunan G. intestinalis genotiplerinin dağılımını real-time PCR yöntemi ile belirlemeyi ve moleküler epidemiyolojik bir veri sunmayı amaçladık. Gereç ve Yöntem: Ocak 2016-Ocak 2018 tarihleri arasında, hem nativ hem de lugol ile mikroskobik olarak incelenen dışkı örnekleri içinde G. intestinalis kist ve/veya trofozoit’i pozitif bulunan 50 G. intestinalis pozitif hastanın dışkı numuneleri çalışmaya dâhil edildi. Dışkı örneklerinden DNA izolasyonu gerçekleştirildikten sonra genotip A ve genotip B için spesifik primerler kullanılarak real-time PCR ile analiz edildi. Bulgular: Çalışmamıza dâhil edilen 50 giardiyaz tanılı hastanın dışkı örneklerinde, 28’inde (%56) A genotipi saptanırken, 17’sinde (%34) B genotipi, 5’inde (%10) ise hem A, hem de B genotipi bulundu. Cinsiyete göre saptanan genotipler incelendiğinde, erkeklerde ve kadınlarda sırasıyla 25 (%50) ve 25 (%50) olarak bulundu. Sonuç: Sonuç olarak, çalışmamız ile ülkemizde giardiyaza neden olan ama ayrımı yalnızca moleküler yöntemlerle ortaya konabilen G. intestinalis genotipleri incelenerek, G. intestinalis’in A genotipinin, B genotipine oranla biraz daha fazla olduğu belirlendi. Ülkemizde G. intestinalis’in moleküler epidemiyolojisine yönelik veriler sınırlıdır. Bu çalışmanın buna katkı sağlayacağı düşüncesindeyiz., Objective: As a causative agent giardiasis, Giardia intestinalis is a flagellated protozoa, and it is a major problem worldwide. Eight different genotypes have been identified in G. intestinalis by molecular method. It has been reported that the genotypes A and B are related to diseases in human and mammals and different genotypes cause different clinical pictures. In this study, we aimed to determine the distribution of G. intestinalis genotypes in stool specimens of G. intestinalis positive patients by realtime PCR and to present a molecular epidemiological data. Material and Methods: Between January 2016 and January 2018, stool samples from fifty G. intestinalis cyst and / or trophozoite positive patients were included in the study. Native andlugol-treated stool specimens were microscopically examined. DNA isolation from stool specimens was performed and then analyzed by real-time PCR using specific primers for genotypes A and B. Results: In stool samples of fifty patients with diagnosis of giardiasis included in our study, genotype A (n=28: 56%), genotype B (n=17: 34%) and both genotype A and genotype B (n=5: 10%) were detected. These genotypes were found in 25 (50%) male, and 25 (50%) female patients. Conclusion: In conclusion, we investigated G. intestinalis genotypes which cause giardiasis in our country but these genotypes can only be distinguished by molecular methods. We found that G. intestinalis genotype A is slightly more frequent than genotype B. In our country, the data on the molecular epidemiology of G. intestinalis genotypes are very limited. We believe this work will contribute to this.

Bu çalışmada Şanlıurfa'da satışa sunulan yoğurtlarda Listeria spp. varlığını ve sıklığını belirlemek ve real-time PCR kullanılarak identifikasyonu amaçlanmıştır. Semt pazarları ve marketlerden toplanan, toplam 62 adet yoğurt örne-ğinde Listeria spp. varlığını belirlemek için ISO 11290-1/A1-2004 tarafından önerilen iki aşamalı bir seçici zenginleştir-me kullanılmıştır. Listeria spp. olarak izole edilen kolonilerden elde edilen DNA’lar kullanılarak yapılan real-time PCR yöntemi ile Listeria monocytogenes identifikasyonu gerçekleştirilmiştir. İncelenen yoğurt örneklerinde Listeria spp. pre-valansı %3.2 (2 örnek) olarak bulunmuştur. Şüpheli kolonilerin ön zenginleştirme broth’tan izole edilen DNA’larla yapı-lan real-time PCR çalışmasında, bu iki Listeria spp. izolatı L. monocytogenes olarak identifiye edilmiştir. Yoğurt örnek-leri 4oC’de 10 gün süreyle muhafaza edilmiş ve pozitif örnekler inkübasyonun birinci, üçüncü ve onuncu günlerinde pH, izolasyon ve real-time PCR ile identifikasyon tekrarlanmıştır. Çalışmamız; Şanlıurfa ilindeki semt marketleri ve pazar-larda satılan yoğurtlarda Listeria spp. türlerinin yaygınlığını göstermesinin yanında, Listeria spp. türleri moleküler yön-temle daha hızlı identifiye edilmiş ve insan sağlığı açısından çok tehlikeli olan L. monocytogenes’in yoğurtta varlığını ve bu yolla insanlarda ciddi gıda kaynaklı hastalıklara neden olabileceği gerçeğini göstermiştir.

Methicillin resistant Staphylococcus aureus (MRSA) is one of the most important pathogenic microorganisms that display multiple drugresistance, causing severe infections in animals and humans. Contaminated or raw milk products, such as yoghurt, are one of the root causesof such MRSA infections for the consumers. Recent studies on clonal investigation revealed the same clonal isolates of animals in humaninfections. Thus, the epidemiology of these bacteria in food products and animals has gained importance. Therefore, the aim of this study wasto epidemiologically detect MRSA in yoghurt samples from Urfa via real-time PCR. To detect MRSA via real-time PCR, the mecA gene wasinvestigated on the DNA obtained from the yoghurt samples. Among 64 yoghurt samples, of which 55 were unpackaged and 9 were packaged,the MRSA mecA gene was only detected in 2 (3.13 %) unpackaged samples via real-time PCR. These results showed that it was possible todetect and epidemiologically investigate MRSA, an important pathogen for human and animal health, via real-time PCR from yoghurt sampleswith direct DNA isolation. In addition, this pathogen can be found in the products sold in Şanlıurfa, which indicates that it may lead to severediseases in case of consumption, if not conformed to hygienic conditions.

Scleroderma is a rarely seen chronic collagen tissue disease which has been first described in the 18th century. Bu çalışmada; Çukurova Üniversitesi Tıp Fakültesi Çocuk Sağlığı ve Hastalıkları Anabilim Dalı ve Pediatrik Hematoloji Bilim Dalına Eylül 2009-2011 tarihleri arasında başvuran hematolojik malignensi’li hastalarda JAK2V617F mutasyonu dağılımı incelenmiştir. Pediatri grubunda sitomorfolojik, immunohistokimyasal ve immün akımsitometrik çalışmalar ile Akut Lösemi tanısı alan hastalardan 216’sı (164’ü ALL, 52 AML’li) çalışmaya alınmıştır. Ayrıca Eylül 2009-2011 tarihleri arasında Dahiliye Hematoloji bölümüne başvuran PV, ET, KML ve PMF’li 176 yetişkin hasta çalışmaya dahil edilmiştir. JAK2V617F mutasyonunu saptamak amacı ile tam kandan DNA izolasyonu yapılmıştır. DNA izolasyonu manuel yöntemle High Pure PCR Template Preperation Kit (Roche) kullanılarak gerçekleştirilmiş ve DNA’lar -80oC’de saklanmıştır. JAK2V617F gen mutasyonları RealTime PCR kullanılarak, genotiplendirmeler ise erime eğrileri analizleri ile saptanmıştır. Mutant JAK2617F/F için Tm=53ºC, yaban tip JAK2617V/V için Tm=62ºC ve Heterezigot allelleri JAK2617V/F için Tm=53ºC ve Tm=62ºC’dir. Sonuç olarak; ALL’li 164 hastanın hiçbirinde (% 0), AML’li 52 hastanın 1 (% 1,92), PV’li 79 hastanın 71’i (% 89,8), ET’li 51 hastanın 22’si (% 43,1), KML’li 22 hastanın 1’i (% 4,5) ve PMF’li 24 hastanın 15’inde (% 62,5) JAK2V617F mutasyonu saptanmıştır. KML, PV, PMF’li yetişkin hasta gruplarında trombosit değerleri ile JAK2V617F mutasyonu arasında herhangi bir anlamlı ilişki bulunmaz iken beklendiği gibi ET’de hastalığın temel patogenomik bulgusu olan trombositoz nedeni ile JAK2V617F mutasyonu ve trombosit değerleri arasında anlamlı bir ilişki saptanmıştır. ET ve PV’li yetişkin hasta gruplarında, genotip ve allel dağılımı arasında istatiksel olarak önemli bir fark bulunmuş (p=0,000) ve allel dağılımı açısından T allelinin hastalık riski oluşturabileceği saptanmıştır (p=0,000). JAK2 V617F mutasyonu saptanan ve saptanmayan PV ve ET hastaları arasında hemoglobin (Hb), hematokrit (Hct), lökosit (Wbc) değerleri açısından istatistiksel bir fark bulunmamıştır (p>0.05). Erişkinler ile karşılaştırıldığında çocukluk çağı lösemilerinde JAK2V617F mutasyonu hematolojik malignensilerde yok denecek kadar az olduğu sonucu çıkarılmıştır. Bu çalışma; pediatrik yaş grubunda bu kadar yüksek hasta sayısı ile Türkiyede yapılan ilk araştırmadır. Dünya literatüründe ki çalışmalar ile paralel sonuçlar elde edilmiştir. Bunun yanında, ilk kez, lösemilerde önemli bir sınıflama aracı olan immünofenotipleme için yapılan akımsitometrik verileri ile JAK2V617F mutasyon ilişkisi değerlendirilmiştir. Literatürde belirtildiği gibi, hematolojik malignitelerde özellikle PV ve ET ayırıcı tanısında ve tedaviye yanıtın takibinde, JAK2V617F mutasyonun Realtime PCR ile saptanmasının, tarama testi olarak faydalı olduğu birkez daha gösterilmiştir. Yakın zamanda MPDs hastalarında tedavide önemli olan seçici JAK1/2 inhibitorü içeren bir ilaç geliştirilmiştir. Bu da JAK2V617F mutasyonun incelenmesinin ne kadar önemli olduğunu göstermiştir. In this study we have examined the deviation of JAK2V617F mutation among the patients who applied to the departments of child health and disease and pediatric hematology of Çukurova University Medicine Faculty between September 2009 and September 2011. 216 patients in the pediatric group that were diagnosed with acute leukemia by cytomorphological and immunohistochemical studies have been examined. In addition, 176 adults -PV, ET, CML and PMF patients- who applied to Hematology clinic between the dates September 2009 and 2011 have been studied. In order to identify the JAK2V617F mutation, DNA has been isolated from the whole blood. DNA isolation has been carried out with manual methodology, using the High Pure PCR Template Preparation Kit (Roche), and the extracted DNAs have been preserved at -80ºC. Gene mutations have been identified by analyzing the Tm curves obtained throughout the RealTime PCR. The mutant JAK2617F/F Tm value is 53ºC, the wild type JAK2617V/V Tm value is 62ºC, and heterozygosis allels JAK2617V/F Tm values are 53ºC and 62ºC. As a result, the rates of the identified JAK2V617F mutation are as follows: none (0%) among the 164 ALL patients, 1 (1,92%) among the 52 AML patients, 71 (89,8%) among the 79 PV patients, 22 (43,1%) among the 51 ET patients, 1 (4,5%) among the 22 CML patients, 15 (62,5%) among the 24 PMF patients. Although no significant relation has been detected between the platelet value and JAK2V617F mutation for the CML, PV and PMF adult patient groups, a significant relation has been detected between the platelet value and JAK2V617F mutation for the ET patients -as expected- because of thrombocytosis, which is the main pathogenomic sign of the disease. A significant statistical difference (p=0,000) has been found between the allele and genotype distribution. T allele is identified as a risk for the disease according to allele distribution (p0.05) has been found for hemoglobin (Hb), hematocrit (Hct), white blood cell (WBC) values for the PV and ET patients with and without JAK2V617F mutation. It is concluded that the JAK2V617F mutation at the hematological malignancies is very little or negligible in the childhood leukemia when compared to the adults. This study examines the highest number of patients in pediatric age group in Turkey and in the literature. The results obtained are in accordance with the literature. Moreover, this is the first study to evaluate the relationship between JAK2V617F mutation and flowcytometric findings for the immunophenotypic examination, which is an important classification tool of leukemia. It is once again shown that determination of the JAK2V617F mutation with Realtime PCR is a useful screening test for definitive diagnosis of hematologic malignancies especially PV and ET and follow-up of patients’ response to therapy as reported in the literature. A medicine has been recently developed consisting of JAK1/2 inhibitor to cure MPDs patients. This reveals the importance of examination of JAK2V617F mutation. Bu çalışma Ç.Ü. Rabies is a fatal disease of all warm blooded animals. In this investigation the distribution of the rabies virus was determined by real time PCR, and comparison of PCR against histopathology, Seller reaction and florescent antibody test for diagnosis, was made. The material of the research was composed of 25 head of the cattle with rabies infection that was brought to Adana Veterinary Control and Research Institute between the years 2007-2009. All of the cases were determined viral nucleic acid by real time PCR. Eleven of them were positive by Seller, 15 were positive by histopathologically and 21 was positive by fluorescent antibody reaction for rabies. In this research it was observed that the amount of the viral nucleic acid was higher in cerebellum than the other parts of the head. A positive correlation was found between the viral load on the different areas of the head and viral load on origin of the nerves innerved these areas. The results of this study confirmed the real time PCR can be used in cases that rabies cannot be diagnosed by routine methods and absence of cerebrum and cerebellum., Kuduz bütün sıcakkanlı canlılarda görülen, öldürücü zoonoz bir hastalıktır. Bu çalışmada Real Time PCR tekniği ile kuduz virüsünün sığır kafasındaki lokalizasyonları tespit edilerek, kuduz hastalığının teşhisinde Real Time PCR ile histopatoloji, Seller boyama ve floresan antikor testi (FAT) karşılaştırıldı. Çalışmanın materyalini Adana Veteriner Kontrol ve Araştırma Enstitüsüne 2007-2009 yılları arasında getirilen kuduz pozitif 25 adet sığır kafası oluşturdu. Real Time PCR ile kuduz olduğu belirlenen 25 olgunun 21’inde FAT ile 11’inde Seller boyama ile 15’inde histopatolojik olarak kuduza ait bulgular tespit edildi. Real Time PCR ile cerebellum’larda belirlenen viral nükleik asit miktarının diğer bölgelere göre daha fazla olduğu, kafanın farklı bölgelerinde ve bu bölgeleri inerve eden sinirlerin orijin yerlerinde belirlenen viral nükleik asit miktarları arasında pozitif korelâsyonun bulunduğu belirlendi. Beyin ve beyinciğin klasik yöntemlerle muayeneye uygun olmadığı durumlarda Real Time PCR'nun teşhis için kullanılabileceği ve yine beyin ve beyinciğin bulunamadığı durumlarda kafanın farklı bölgelerinden alınan örneklerden Real Time PCR ile kuduz hastalığının teşhisinin yapılabileceği kanaatine varıldı.

İnsanlarda Salmonella bakterileri Tifo, Paratifo, Sepsis, Salmonella'lı Gastro-Enteritler ve Besin zehirlenmeleri etkeni olabilmektedirler. Kişinin yaşam standartları, yaşadığı evin tipi, kalabalık olması, sosyoekonomik durumu, çevre şartları ve en önemlisi kullanılan suyun kaynağı, hastalığın yayılmasında en önemli etkenlerdir. Bu çalışmamızda İstanbul ili sınırlarında yaşayan ve belirli sektörlerde çalışan personelden kan örnekleri alınarak Salmonella taşıyıcılığı açısından araştırılmıştır. Kan örnekleri Vi-antijeni ile Salmonella taşıyıcılık kontrolünden sonra taşıyıcı bireylerin dışkısından kronik taşıyıcı olup olmadığının tespiti için dışkı örnekleri tekrar incelenmiştir. İstanbul ilinde yapılan bu çalışmada 10.000 denek üzerinde tarama yapılmıştır. Bu 10.000 kişinin 22'sinin taşıyıcı olduğu ve bunlardan da 2'sinin kronik taşıyıcı olduğu tespit edilmiştir. Bunun sonucunda kronik tifo taşıyıcılığının yıllara oranla azaldığı fakat tümüyle kaybolmadığı saptanmış ve tarama sonunda taşıyıcı oldukları saptanan bu bireylere kontrollü bir proflaksi uygulanmıştır.Temmuz,2002 Akın YİĞİNSalmonella bacteria can be the reasons of Typhoid, Paratyphoid, Gasro-Enteriditis and Nutrition poisonings. Person's living standarts, the accommodation type, the crowd of the house, socioeconomic conditions, environmental conditions and the water sources, the most importantly, are the main factor spreading of the disease. Within this study we used blood specimen that were taken from the people who live and work in certain working sectors in Istanbul. After the carrier people's feces were again controlled for chronic carrier. Furthermore, medical treatment was supplied to the carriers to maintain the isolation of these people from the society.With this study, 10.000 people are investigated throughout the city of Istanbul It is determined that 22 of these 10.000 people are carrier and 2 of them are chronic carrier.As a conclusion it is determined that chronic typhoid carrier has been decreasing throughout the years but it is not terminated totally. After the investigation the controlled proflaxi is applied to the infected people.
