Although accumulating evidence has highlighted the molecular mechanisms by which hTERT promotes tumour cell invasion and metastasis, the molecular mechanisms of the properties enabling hTERT to contribute to invasion and metastasis have not been clearly illustrated. Here, we report that hTERT promotes gastric cancer invasion and metastasis by recruiting p50 to synergistically inhibit PLEKHA7 expression. We observed that the expression of PLEKHA7 in gastric cancer was significantly negatively associated with the TNM stage and lymphatic metastasis and that decreased PLEKHA7 expression dramatically increased invasion and metastasis in gastric cancer cells. Further mechanistic research showed that hTERT directly regulates PLEKHA7 expression by binding p50 and recruiting the hTERT/p50 complex to the PLEKHA7 promoter. Increased hTERT dramatically decreased PLEKHA7 expression and promoted invasion and metastasis in gastric cancer cells. The hTERT-mediated invasion/metastasis properties at least partially depended on PLEKHA7. Our work uncovers a novel molecular mechanism underlying invasion/metastasis in gastric cancer orchestrated by hTERT and p50.

COVID-19 has resulted in a significant impact on individual lives, bringing a unique challenge for face retrieval under occlusion. In this paper, an occluded face retrieval method which consists of generator, discriminator, and deep hashing retrieval network is proposed for face retrieval in a large-scale face image dataset under variety of occlusion situations. In the proposed method, occluded face images are firstly reconstructed using a face inpainting model, in which the adversarial loss, reconstruction loss and hash bits loss are combined for training. With the trained model, hash codes of real face images and corresponding reconstructed face images are aimed to be as similar as possible. Then, a deep hashing retrieval network is used to generate compact similarity-preserving hashing codes using reconstructed face images for a better retrieval performance. Experimental results show that the proposed method can successfully generate the reconstructed face images under occlusion. Meanwhile, the proposed deep hashing retrieval network achieves better retrieval performance for occluded face retrieval than existing state-of-the-art deep hashing retrieval methods.

Several clinical trials have reported that patients with cancer cachexia can benefit from n-3 polyunsaturated fatty acids (n-3 PUFAs) supplements; however, the results have been conflicting. This systematic review and meta-analysis aimed to evaluate the effect of n-3 PUFAs on cancer cachexia. A search of the PubMed, Embase, and Cochrane Library databases was performed to identify the included randomized controlled trials. Trials including patients with cancer cachexia who were administered a course of n-3 PUFAs were included. A meta-analysis on body weight, lean body weight, proinflammatory factors, quality of life, and median duration of survival was conducted. A total of 12 randomized controlled trials with 1184 patients were included. No effect on body weight (standard mean difference [SMD], 0.10; 95% CI, -0.06 to 0.26; P = .236), lean body weight (SMD, -0.17; 95% CI, -0.36 to 0.03, P = .095), or proinflammatory factors (interleukin-6: SMD, 0.31; 95% CI, -0.14 to 0.75; P = .18; tumor necrosis factor-α: SMD, -0.85; 95% CI, -2.39 to 0.69; P = .28) was observed. The use of n-3 PUFAs was associated with a significant improvement in quality of life (SMD, 0.70; 95% CI, 0.01-1.40; P = .048) and median duration of survival (median survival ratio, 1.10; 95% CI, 1.02-1.19; P = .014). For patients with cancer cachexia, our meta-analysis indicated that n-3 PUFAs improved quality of life and survival, but not body weight.

It is debated whether cortical responses matching the time scales of phrases and sentences mediate the mental construction of the syntactic chunks or are simply caused by the semantic properties of words. Here, we investigate to what extent delta-band neural responses to speech can be explained by semantic relatedness between words. To dissociate the contribution of semantic relatedness from sentential structures, participants listened to sentence sequences and paired-word sequences in which semantically related words repeated at 1 Hz. Semantic relatedness in the 2 types of sequences was quantified using a word2vec model that captured the semantic relation between words without considering sentential structure. The word2vec model predicted comparable 1-Hz responses with paired-word sequences and sentence sequences. However, empirical neural activity, recorded using magnetoencephalography, showed a weaker 1-Hz response to paired-word sequences than sentence sequences in a word-level task that did not require sentential processing. Furthermore, when listeners applied a task-related rule to parse paired-word sequences into multi-word chunks, 1-Hz response was stronger than that in word-level task on the same sequences. Our results suggest that cortical activity tracks multi-word chunks constructed by either syntactic rules or task-related rules, whereas the semantic relatedness between words contributes only in a minor way.

Acetylcholine (ACh), as a classical neurotransmitter, plays great roles in the nervous system. There is increasing evidence of its non-neuronal roles in regulating basic cell functions in vertebrates. However, knowledge about the non-neuronal cholinergic system in insects is scarce.A comparative transcriptome analysis was performed to investigate differences in the key molecular components of the cholinergic system between the head and ovary. The results showed that expression levels of most cholinergic system-related genes were higher in the head than in the ovary, and some cholinergic components were absent in the ovary. ACh contents ranged from 0.1 to 1.3 μg mgThis study reveals the existence of the non-neuronal cholinergic system in the ovary of M. separata, whose synthesis and release mechanisms are different from those of the head. These results provide novel insights into the non-neuronal cholinergic system in insects, and will be valuable in the discovery of new target genes and the future development of green pest control. © 2022 Society of Chemical Industry.

Oral squamous cell carcinoma (OSCC) is the most common head and neck cancer with a poor prognosis. Therefore, it is crucial to explore molecular prognostic biomarkers for OSCC. ZEB1 (also known as δEF1) is a member of the zinc finger E-box binding protein family of transcription factors involved in various biological processes, including tumorigenesis, progression, and metastasis. Recent evidence suggests that ZEB1 has a role in the tumorigenicity of oral epithelial cells, although its mode of action needs to be investigated further. To better understand the relationship between ZEB1 and OSCC, we transfected the ZEB1-overexpressing oral squamous cell lines SCC9 and SCC25 with lentivirus and then extracted RNA from the cells for gene expression analysis. Predictions are constantly generated from diverse sources to optimize cognitive functions in the ever-changing environment. However, the neural origin and generation process of top-down induced prediction remain elusive. We hypothesized that motor-based and memory-based predictions are mediated by distinct descending networks from motor and memory systems to the sensory cortices. Using functional magnetic resonance imaging (fMRI) and a dual imagery paradigm, we found that motor and memory upstream systems activated the auditory cortex in a content-specific manner. Moreover, the inferior and posterior parts of the parietal lobe differentially relayed predictive signals in motor-to-sensory and memory-to-sensory networks. Dynamic causal modeling of directed connectivity revealed selective enabling and modulation of connections that mediate top-down sensory prediction and ground the distinctive neurocognitive basis of predictive processing.

The ability to maintain events (i.e. interactions between/among objects) in working memory is crucial for our everyday cognition, yet the format of this representation is poorly understood. The current ERP study was designed to answer two questions: How is maintaining events (e.g., the tiger hit the lion) neurally different from maintaining item coordinations (e.g., the tiger and the lion)? That is, how is the event relation (present in events but not coordinations) represented? And how is the agent, or initiator of the event encoded differently from the patient, or receiver of the event during maintenance? We used a novel picture-sentence match-across-delay approach in which the working memory representation was ‘pinged’ during the delay, in two ERP experiments with Chinese and English materials. First, we found that maintenance of events elicited a long-lasting late sustained difference in posterior-occipital electrodes relative to non-events. This effect resembled the negative slow wave reported in previous studies of working memory, suggesting that the maintenance of events in working memory may impose a higher cost compared to coordinations. Second, in order to elicit a hallmark for agent vs. patient representation in working memory, we pinged agent or patient characters during the delay. Although planned comparisons did not reveal significant differences in the ERPs elicited by the agent pings vs. patient pings, we found that the ping appeared to dampen the ongoing sustained difference, suggesting a shift from sustained activity to activity silent mechanisms. These results represent one of the uses of ERPs to elucidates the format of neural representation for events in working memory.

The computational principles underlying attention allocation in complex goal-directed tasks remain elusive. Goal-directed reading, i.e., reading a passage to answer a question in mind, is a common real-world task that strongly engages attention. Here, we investigate what computational models can explain attention distribution in this complex task. We show that the reading time on each word is predicted by the attention weights in transformer-based deep neural networks (DNNs) optimized to perform the same reading task. Eye-tracking further reveals that readers separately attend to basic text features and question-relevant information during first-pass reading and rereading, respectively. Similarly, text features and question relevance separately modulate attention weights in shallow and deep DNN layers. Furthermore, when readers scan a passage without a question in mind, their reading time is predicted by DNNs optimized for a word prediction task. Therefore, attention during real-world reading can be interpreted as the consequence of task optimization.

BackgroundGrowing evidence suggests that nutritional status and inflammation are associated with survival in various cancers. This study aimed to evaluate the prognostic value of the prognostic nutritional index (PNI), geriatric nutritional risk index (GNRI), and systemic inflammatory indexes (neutrophil/lymphocyte ratio [NLR], monocyte/lymphocyte ratio [MLR], and platelet/lymphocyte ratio [PLR]) in patients with stage IIB–III cervical cancer receiving radiotherapy.ResultsThe ideal cutoff values for the PNI, GNRI, NLR, MLR, and PLR were 48.3, 97.04, 2.8, 0.41, and 186.67, respectively. Low PNI and GNRI scores were associated with poor OS and PFS. High NLR, MLR, and PLR also predicted inferior 5-year OS and PFS rates in patients with stage IIB–III cervical cancer. Multivariate Cox regression analysis identified tumor size, histological type, stage, number of metastatic lymph nodes, PNI, GNRI, NLR, PLR, and MLR as significant prognostic factors for OS and PFS.ConclusionsThe current findings suggest that the PNI, GNRI, NLR, PLR, and MLR are essential parameters for predicting prognosis in patients with stage IIB–III cervical cancer receiving radiotherapy.

To realize the heat transfer between granular flow and sCO2 cycle, moving bed heat exchangers have been widely applied in Concentrating Solar Power systems. This work developed a prediction model for heat transfer enhancements and dynamic control strategies in a shell-and-plate moving bed heat exchanger. For fast evaluation of heat transfer performances in granular flow, the continuum model was adopted with different thermal resistances. The wall effect of near-wall region was modified by contact thermal resistance, and the equivalent heat diffusion in bulk region was reflected by penetration thermal resistance. The results showed that, the decrement of penetration thermal resistance can improve heat transfer effectively. To weaken the penetration thermal resistance, the thinner thermal boundary layer and the higher effective thermal conductivity are urgently required. The staggered arrangement is a valid way to limit the development of thermal boundary layer. The bank of heat exchanger plates offset horizontally from the previous bank, and the re-evolution of temperature distributions is equal to the equivalent thermal mixing between different banks. Meanwhile, the effective thermal conductivity can be improved by particle self-diffusions perpendicular to the major flow direction. For the particle migrations, the local disturbances can be applied. The relatively decrement of penetration thermal resistance can achieve 45.7% at a disturbance intensity of 5 × 10−7 m2/s. The outlet temperature of sCO2 can be well controlled by the transient temperature–velocity feedback method with relaxation coefficients. The relatively decrement of temperature fluctuation is larger than 80% with a relaxation coefficient of 40.

An elliptical-like combined tube (ELCT) is proposed for the shell-and-tube moving packed bed heat exchanger. The heat transfer of particle flow around the ELCT is numerical studied. The kinetic energy ratio, contact number, contact time and heat transfer coefficient are analyzed. It is found that, the heat transfer performance of the ELCT is similar to that of the elliptical tube (ET) and better than the circular tube (CT). Compared with the CT, the ELCT significantly reduces the stagnation zone and the cavity zone, and improves the heat transfer at the top and the bottom of the tube (average increase of 42% and 53%). Furthermore, compared with the ET and the CT, the ELCT has better inside-tube heat transfer performance (average increase of 5% and 29%), various inside-tube heat transfer enhancement techniques (same with the CT) and better flexural capacity (average increase of 8% and 36% under same mass).

Cellulose nanocrystals (CNCs), as the promising reinforcing fillers in the rubber industry, their surface chemical groups have vital effects on the vulcanization kinetics, cross-linking densities, and mechanical properties of rubber composites. Herein, CNCs with acidic carboxyl (CCA) and alkaline amino groups (CCP) were produced by modifying the sulfonic CNCs (CCS) in environment-friendly ways. Studies found the CCS and CCA with acid groups have obvious inhibiting effects on the vulcanization of natural rubber (NR), while CCP with alkaline amino groups accelerates the vulcanization of NR. Differential scanning calorimeter, Fourier transform infrared spectroscopy, and Electron paramagnetic resonance, etc. were performed to clarify the effecting mechanisms of CNCs surface groups on NR vulcanization. It was found that NR/CCS and NR/CCA nanocomposites vulcanize through radical reactions, and the surface acidic groups of CCS and CCA, i.e., hydroxyl, sulfonate, and carboxyl groups inactivate the sulfur radicals generated during vulcanization and depress the vulcanization activity. Cognitive impairments are core aspects of schizophrenia and are highly related to poor outcomes. However, the effect of therapy on cognitive impairments remains unsatisfactory as its biological mechanisms are not fully understood. The purpose of this study was to investigate the disrupted intrinsic neural activity of the frontal areas and to further examine the functional connectivity of frontal areas related to cognitive impairments in schizophrenia. We collected brain imaging data using a 3T Siemens Prisma MRI system in 32 patients with schizophrenia and 34 age- and sex-matched healthy controls. The mean fractional amplitude of low-frequency fluctuation (mfALFF) in the frontal regions was calculated and analyzed to evaluate regional neural activity alterations in schizophrenia. Seed regions were generated from clusters showing significant changes in mfALFF in schizophrenia, and its resting-state functional connectivity (rs-FC) with other brain regions were estimated to detect possible aberrant rs-FC indicating cognitive impairments in schizophrenia. We found that mfALFF in the bilateral frontal cortices was increased in schizophrenia. mfALFF-based rs-FC revealed that decreased rs-FC between left middle frontal gyrus (MFG) and left medial superior frontal gyrus (MFSG) was associated with poor delayed memory (r = 0.566, Bonferroni-corrected p = 0.012). These findings demonstrate increased neural activity in the frontal cortices in schizophrenia. FC analysis revealed a diminished rs-FC pattern between the left MFG and left MSFG that was associated with cognitive impairments. These findings have provided deeper insight into the alterations in brain function related to specific domains of cognitive impairment and may provide evidence for precise interventions for cognitive deficits in schizophrenia.

Welding properties of three alloys with different Cu contents (0.1, 0.4 and 0.9 wt.%) is studied by means of melt inert-gas welding (MIG). The results showed that, with the increase of Cu content from 0.1 wt.% to 0.9 wt.%, the strength and elongation increased obviously. When the Cu content was 0.9 wt.%, the strength and elongation reached 375MPa and 15%, respectively, which is a better match for bearing structure part of vehicles application. Besides, the adding of Cu element provided an increase limit for welding joint strength. Due to abundant coarse phases in fusion zone (FZ) and welding zone (WZ), the strength slightly increased and the failure fracture was different compared with that of 0.1Cu and 0.4Cu alloy. Present result was meaningful and valuable for pushing aluminum application automobile structural parts and other engineering components.

Moving bed heat exchangers have been the economic alternatives for the granular flow in industries. To predict the heat transfer coefficients, a better understanding is required of the contact resistance (1/hws) and the penetration resistance (1/hso). In this work, the heat transfer coefficients were simulated using the finite volume method to investigate these resistances in both ordered and random packing sphere particles. It was found that, the 1/hws is determined by the effective thermal conductivity in the wall region, of which the thickness is around 3/22 particle diameters. Moreover, the 1/hso is affected by the heat capacity, thermal conductivities and particulate structures. The effect is reflected by two modified equivalent heat absorption coefficients (1/p1 and 1/p3) in granular flows. Generally, p1 is smaller in the ordered structure because the peak local solid fraction is higher, while p3 is smaller in the random structure due to the lower average porosity.

Discrete element method (DEM) is a powerful tool for solid granular, while the related heat transfer model requires more calibrations due to the over-simplified heat transfer path. In the present study, the calibration of overall thermal resistances (Rp) was conducted between a particle and the wall, where heat conduction in the particle interior and gap is more remarkable. It was found that, the existing DEM models should be improved in a wide range of the solid-to-gas ratio of thermal conductivity (ks/kg) and the dimensionless body distance (Sx/dp). The ideal heat transfer path may result in larger deviations as ks/kg > 250 or Sx/dp > 0, which can be only mathematically modified by the empirical factor. The general descriptions for modification are still required. To overcome the limit, an effective thermal conductivity model is proposed with certain formulas, where Rp is divided into two parts in the mixing phase zone and the gas zone.

Due to the normal BMI, non-obese NAFLD are easily to be ignored and eventually lead to potential liver injuries.Methods: A population-based cross-sectional study was conducted on 1608 cases with normal serum alanine aminotransferase (ALT) levels who were divided into an obese group (BMI ≥ 25 kg/m2) and a non-obese group (BMI < 25 kg/m2). NAFLD was diagnosed by ultrasound and Fibro Scan examination. Non-obese populations were divided into NAFLD group (CAP ≥ 240 db/m) and non-NAFLD group (CAP < 240 db/m). The incidence of NAFLD in the obese and non-obese populations and constituent ratios of genders, age, and serum levels of triglycerides (TG), cholesterol (CHOL), and blood glucose were compared. Risk factors of NAFLD in non-obese people were analyzed by multivariate logistics regression.Results: The occurrence of NAFLD was higher in the obese group than in the non-obese group, regardless of gender (P < 0.001). In the non-obese group, the occurrence of NAFLD in female patients was lower than that in male (P=0.001). The occurrence of NAFLD increased with age, with 50– 59 years being the peak age of incidence in both male and female. The peak age of NAFLD occurrence in non-obese male patients was more delayed than that in obese male patients. BMI (OR=1.311, P=0.000) and TG (OR=2.545, P=0.000) were risk factors for NAFLD in the non-obese population.Conclusion: Compared with obese population, the incidence of NAFLD in non-obese population was relatively low and more frequently in male than in female, the peak age of NAFLD occurrence in non-obese male patients was also delayed. This study aims to investigate the role of lipopolysaccharide (LPS), a glycolipid compound found in the outer membrane of gram-negative bacteria, on the migration and invasion of PCa cells, and to evaluate the protective effect of melatonin.Materials and Methods: DU145, PC-3 and LNCaP cells were incubated with LPS in the presence or absence of melatonin. Wound healing and Transwell assays were used to analyze migration and invasion of PCa cells. RT-PCR and Western blotting were used to assess the mRNA and protein levels, respectively. Co-IP was used to analyze β-catenin ubiquitination.Results: Our results showed that LPS promoted migration and invasion of PCa cells. In addition, LPS stimulated inflammatory reaction and induced epithelial–mesenchymal transition (EMT) in PCa cells by activating several TLR4 downstream pathways. Specifically, LPS promoted NF-κB/IL-6/STAT3 signal transduction. In addition, LPS upregulated phosphorylation levels of cytoplasmic AKTSer473 and GSK-3βSer9. Moreover, LPS induced phosphorylation of GSK-3βSer9 in the “disruption complex”, and then inhibited phosphorylation and ubiquitination of cytoplasmic β-catenin, leading to β-catenin nuclear translocation. Interestingly, melatonin inhibited invasion and migration not only in LPS-stimulated but also in LPS-unstimulated PCa cells. Melatonin suppressed PCa cells migration and invasion by blocking EMT mediated by IL-6/STAT3, AKT/GSK-3β and β-catenin pathways.Conclusion: This study provides evidence that melatonin inhibits migration and invasion through blocking multiple TLR4 downstream EMT-associated pathways both in LPS-stimulated and -unstimulated PCa cells. The article established a modelling that predicted and measured the quenching and pre-stretching stress in 7050 aluminum alloy thick plate in order to make the alloy suitable enough for manufacturing in the aerospace industries. The results show that both the rolling direction component stress and the transverse direction component stress have a “M” distribution along the plate thickness of 7050 aluminum alloy thick plate, but the level is slightly different, but the level is slightly different. The maximum value of quenching residual stress is about 187MPa in the 80mm thick of 7050 plate. Pre-stretching of the heat treated and quenched alloy to 0.5%, 1.0%, 1.5%, 2.0%, 2.5%, 3.0% and 3.5% to reduce the residual stresses. It has been recorded that more than 90% reduction of residual stresses were observed in a pre-stretching rage of 2-3%. Measuring the surface residual stresses of the alloy by ultrasonic methods and measuring the internal residual stresses of the alloy by crack compliance methods. The two methods are used to verify the model. The difference between the measurement and the simulation is within 40MPa. The experimental data of the ultrasonic and the crack compliance methods were found to correspond well with the simulated models. This shows that the simulation model can effectively predict the evolution of residual stress in 7050 aluminum alloy thick plate, and the simulation model can also be reconstructed and inspected through the experimentally measurement.
