Recent studies on the composition of edible insects have placed more emphasis on the quantity and quality of macro- and micronutrients and less emphasis on their bioactive components. This study aimed to assess the effect of 0-100% ethanol solvent ratio considering 5-hydroxyindolin-2-one (5-HI) content on the extraction efficiency in Protaetia brevitarsis seulensis larvae (PBSL) extract and to investigate the effect of PBSL on the induction of cell adhesion molecules (CAMs) in tumour necrosis factor (TNF)-α-activated human umbilical vein endothelial cells (HUVECs). A reversed-phase high-performance liquid chromatography method coupled to a diode array detector (HPLC-DAD) was validated for the quantification of 5-HI. The HPLC-DAD method was validated for its specificity, linearity, precision, accuracy, limit of detection, and limits of quantitation. The calibration curve of 5-HI showed significant linearity (R2>1). The limits of detection and quantitation were 0.016 μg/ml and 0.048 μg/ml, respectively. The relative standard deviation of the precision and intraday tests was less than 0.5%. In the recovery test, the accuracy ranged from 94.48-105.5% and the relative standard deviation value was less than 0.13%. The method validation parameters indicate the applicability of the HPLC method for quality control of food or pharmaceutical formulations containing PBSL. Comparative analysis of the 5-HI content by extraction solvent revealed that it was the highest at 357.7 μg/ml in 70% ethanol. Additionally, the analysis result of 5-HI content according to the growth conditions was the highest at 439 μg/ml at 31 °C for 30 days. Furthermore, in the condition where PBSL was at 31 °C for 30 days inhibition of the TNF-α-induced upregulation of protein expression of vascular cell adhesion molecule-1 (VCAM-1), intracellular adhesion molecule-1 (ICAM-1), and endothelial-leukocyte adhesion molecule-1 (E-selectin) was observed. Therefore, PBSL may interfere with early atherosclerotic events involving endothelial CAM induction.

Translation factors have traditionally been viewed as proteins that drive ribosome function and ensure accurate mRNA translation. Recent discoveries have highlighted that these factors can also moonlight in gene regulation, but through functions distinct from their canonical roles in protein synthesis. Notably, the additional functions that translation factors encode are diverse, ranging from transcriptional control and extracellular signaling to RNA binding, and are highly regulated in response to external cues and the intrinsic cellular state. Thus, this multifunctionality of translation factors provides an additional mechanism for exquisite control of gene expression.

Jensen et al . ( 1 ) question evidence presented of a chambered heart within placoderms, citing its small size and apparently ventral atrium. However, they fail to note the belly-up orientation of the placoderm within one nodule, and the variability of heart morphology within extant taxa. Thus, we remain confident in our interpretation of the mineralized organ as the heart.

Brodmann Area 10 (BA10) is the largest cytoarchitectonic region of the human cortex, performing complex integrative functions. BA10 undergoes intensive adolescent grey matter pruning around the average age of onset for Bipolar disorder (BP) and Schizophrenia (SCHIZ), and its dysfunction is likely to underly aspects of their shared symptomology. In this study, we investigated the role of BA10 neurotransmission-related gene expression in BP and SCHIZ. We performed qPCR to measure the expression of 115 neurotransmission-related targets in control, BP, and SCHIZ post-mortem samples (n=72). We chose this method for its high sensitivity to detect low-level expression. We then bolstered our findings by performing a meta-analysis of publicly-released BA10 microarray data (n=101) and identified sources of convergence with our qPCR results. To improve interpretation, we compiled an unusually large database of clinical metadata for our samples. We used this data to explore the relationship between BA10 gene expression, therapeutics, substances of abuse, and symptom profiles, and validated these findings with publicly-available datasets. Using these convergent sources of evidence, we identified 20 neurotransmission-related genes that were differentially expressed in BP and SCHIZ in BA10. These results included a large diagnosis-related decrease in two important therapeutic targets with low-levels of expression, HTR2B and DRD4, as well as other findings related to dopaminergic, GABA-ergic and astrocytic function. We also observed that therapeutics may produce differential expression that opposes the effects of diagnosis. In contrast, substances of abuse showed similar effects on BA10 gene expression as BP and SCHIZ, potentially amplifying diagnosis-related dysregulation.

Reports of SARS-CoV-2 coronavirus (COVID-19) vaccine-related myocarditis, particularly after mRNA vaccines, have raised concerns amongst the general public. This review examined the literature regarding myocarditis post COVID-19 vaccination, drawing from vaccine safety surveillance databases and case reports.Combinations of search terms were used in PubMed and COVID-19-specific repositories - LitCovid and the Cochrane COVID-19 Study Register - between 1 October 2020 and 31 October 2021. Manual searches of GoogleScholar and screening of article bibliographies were also performed.Information was obtained from five vaccine safety surveillance databases. Fifty-two (52) case reports totalling 200 cases of possible COVID-19 vaccine-related myocarditis were summarised. Vaccine surveillance databases differed in reporting formats and vaccination rates; however, gross estimates suggested low overall incidence rates of 2-5 per million mRNA vaccines. The incidence appeared to be higher in younger male populations, with onset of symptoms within a few days, usually after the second dose. Some with prior COVID-19 infections had onset after the first dose. Cases with prior unrelated myocarditis were also noted. Almost all presented with chest pain (98.0%). Troponin elevation was universally described and cardiac magnetic resonance imaging was commonly reported based on the updated Lake Louise criteria. Clinical course was mild in the majority, with response to anti-inflammatory treatment.COVID-19 vaccine-related myocarditis is an important but rare adverse event. More research is needed into its pathogenesis and reasons for its predominance in young males, while gaps in data exist in those aged16 years, as well as those with prior COVID-19 infections and prior myocarditis.

The complete chloroplast (cp) genome sequence of Pearl millet (Pennisetum glaucum [L.] R. Br.), an important grain and forage crop in the family Poaceae, is reported in this study. The complete cp genome sequence of P. glaucum is 138,172 bp in length with 38.6% overall GC content and exhibits a typical quadripartite structure comprising one pair of inverted repeat (IR) regions (22,275 bp) separated by a small single-copy (SSC) region (12,409 bp) and a large single-copy (LSC) region (81,213). The P. glaucum cp genome encodes 110 unique genes, 76 of which are protein-coding genes, 4 ribosomal RNA (rRNA) genes, 30 transfer RNA (tRNA) genes and 18 duplicated genes in the IR region. Nine genes contain one or two introns. Whole genome alignments of cp genome were performed for genome-wide comparison. Locally collinear blocks (LCBs) identified among the cp genomes showed that they were well conserved with respect to gene organization and order. This newly determined cp genome sequence of P. glaucum will provide valuable information for the future breeding programs of valuable cereal crops in the family Poaceae.

Diabetic peripheral neuropathy (DPN) is the most common complication of diabetes, with several underlying pathophysiological mechanisms, some of which are still uncertain. The cornea is an avascular tissue and sensitive to hyperglycemia, resulting in several diabetic corneal complications including delayed epithelial wound healing, recurrent erosions, neuropathy, loss of sensitivity, and tear film changes. The manifestation of DPN in the cornea is referred to as diabetic neurotrophic keratopathy (DNK). Recent studies have revealed that disturbed epithelial-neural-immune cell interactions are a major cause of DNK. The epithelium is supplied by a dense network of sensory nerve endings and dendritic cell processes, and it secretes growth/neurotrophic factors and cytokines to nourish these neighboring cells. In turn, sensory nerve endings release neuropeptides to suppress inflammation and promote epithelial wound healing, while resident immune cells provide neurotrophic and growth factors to support neuronal and epithelial cells, respectively. Diabetes greatly perturbs these interdependencies, resulting in suppressed epithelial proliferation, sensory neuropathy, and a decreased density of dendritic cells. Clinically, this results in a markedly delayed wound healing and impaired sensory nerve regeneration in response to insult and injury. Current treatments for DPN and DNK largely focus on managing the severe complications of the disease. Cell-based therapies hold promise for providing more effective treatment for diabetic keratopathy and corneal ulcers.

Background & Aims: Antiviral treatment from hepatitis B envelope antigen (HBeAg)-positive status may attenuate the integration of hepatitis B virus DNA into the host genome causing hepatocellular carcinoma (HCC). We investigated the impact of HBeAg status at the onset of antiviral treatment on the risk of HCC. Methods: The incidence of HCC was evaluated in Korean patients with chronic hepatitis B who started entecavir or tenofovir in either HBeAg-positive or HBeAg-negative phase. The results in the Korean cohort were validated in a Caucasian PAGE-B cohort. Results: A total of 9143 Korean patients (mean age, 49.2 years) were included: 49.1% were HBeAg-positive and 49.2% had cirrhosis. During follow-up (median, 5.1 years), 916 patients (10.0%) developed HCC. Baseline HBeAg positivity was not associated with the risk of HCC in the entire cohort or cirrhotic subcohort. However, in the non-cirrhotic subcohort, HBeAg positivity was independently associated with a lower risk of HCC in multivariable (adjusted hazard ratio [aHR], 0.41; 95% confidence interval [CI], 0.26–0.66), propensity score-matching (aHR, 0.46; 95% CI, 0.28–0.76), and inverse probability weighting analyses (aHR, 0.44; 95% CI, 0.28–0.70). In the Caucasian cohort (n = 719; mean age, 51.8 years; HBeAg-positive, 20.3%; cirrhosis, 34.8%), HBeAg-positivity was not associated with the risk of HCC either in the entire cohort or cirrhotic subcohort. In the non-cirrhotic subcohort, none of the HBeAg-positive group developed HCC, although the difference failed to reach statistical significance (aHR, 0.21; 95% CI, 0.00–1.67). Conclusions: This multinational cohort study implies that HBeAg positivity at the onset of antiviral treatment seems to be an independent factor associated with a lower risk of HCC in patients with chronic hepatitis B without cirrhosis, but not in those with cirrhosis. © 2021 AGA Institute

This paper presents design challenges and solutions for the fifth generation (5G) phased-array transceiver ICs in millimeter-wave (MMW) frequency bands. A 28nm bulk CMOS device technology is selected to integrate multiple RF phased-array elements in a single-chip to achieve a high-level of TX EIRP and RX sensitivity. Several design approaches of gain, P OUT , stability, reliability and linearity enhancement techniques are applied to enable CMOS as a key device solution for 5G applications in MMW frequency bands. A 39GHz band 16-channel CMOS RF phased-array transceiver IC is designed and can support 4T/4R MIMO base-station applications including ×64 RF phased-array ICs (total 1,024 phased-array elements). T/RX paths have gain dynamic ranges of >30/40dB for flexibility and scalability. The TX path shows P OUT /Ch. of >6.0dBm at EVM of -34dB (800MHz) and P DC /Ch. of 105mW. The RX path performs NF of 4.2dB, EVM of -38dB (100MHz) and P DC /Ch. of 39mW. These state-of-the-art results lead to TX EIRP of >55dBm and RX sensitivity of

Lightning talks of EduHPC are a venue where HPC educators discuss work in progress. This paper summarizes the EduHPC 2020 lightning talks, which cover four very different areas: (i) The simulation-based pedagogy of the EduWRENCH project, including motivations for using simulation to teach High Performance Computing, the design principles underlying EduWRENCH modules, a survey of the available modules, a look at a particular module, plus a conclusion including lesson learned thus far and future plans. (ii) The use of the software-tuning component from Student Cluster Competitions in the HPC master's program at the University of Liverpool. (iii) Steps being taken by the Computer Systems Genome Project at Virginia Tech to foster a community atmosphere among the diverse students working to catalog the lineage of computer system performance over time. (iv) A 3-semester master's degree program titled Computational Engineering, focused on HPC training, being offered at the University of Warsaw.
