1. Cilostazol Mediates Immune Responses and Affects Angiogenesis During the Acute Phase of Hind Limb Ischemia in a Mouse Model

Abstract

Background: Cilostazol is a drug of choice for the treatment of intermittent claudication that also affects innate and adaptive immune cells. The purpose of our study was the evaluation of cilostazol’s impact on the immune and angiogenic response in murine models of hind limb ischemia. Methods: We used 108 immunodeficient NOD.CB17-Prkdcscid/J mice and 108 wild-type CB17 mice. At day 0 (D0), all animals underwent hind limb ischemia. Half of them in both groups received daily cilostazol starting at D0 and for the next 7 postoperative days, while the rest of them served as controls, receiving vehicle. Interleukin (IL) 2, IL-4, IL-6, IL-10, IL-17A, tumor necrosis factor α (TNF-α), and interferon γ (IFN-γ) serum concentrations were measured by flow cytometry on postsurgery days D1, D3, D5, and D7. 5. Ingredients of institutional reputations and citizen engagement with regulators

Abstract

© 2015 Wiley Publishing Asia Pty Ltd. The purpose of this study is to examine the link between the reputational components of efficacy and moral reliability of institutions, and citizens' compliance with institutional recommendations. Research on bureaucratic reputations highlights the significance of positive political reputations based on credibility and legitimacy, but the impact of these components is not systematically isolated and studied. We draw insights from political psychology to move beyond a positive-negative valence-based approach of reputation, and highlight the different effect of efficacy and moral reliability components of reputation on citizens' cooperation, engagement in water saving activities, and levels of complaints. We use the Cypriot Water Authority as a case study and inquire how its institutional reputation influences Cypriot citizens' behavior regarding water use. Our data was collected via a representative national survey administered to a random sample of 800 Cypriots in the spring of 2009 and show that favorable perceptions of particular components of institutional reputation shape the levels of satisfaction with specific organizational outputs.

Abstract

Benthic macrofauna is a biological quality element required for the lagoons’ ecological status assessment in application of the water framework directive. In the Mediterranean area, indices were developed to asses the ecological status of saline lagoons; however they are not relevant in oligohalin to mesohalin ones. The aim of this study was (i) to characterise the benthic macrofauna of six oligomesohalin lagoons, (ii) to explore the possibility to calculate benthic macrofauna metrics commonly included in indices dedicated to freshwater lakes assessment, iii) to analyse the responses of these metrics to physico-chemical stressors. Benthic macrofauna was sampled in spring, in the central part of lagoons and taxa were identified in the laboratory. About 50 metrics were then calculated to characterise composition and diversity of the communities. In the same time, water and sediment were sampled to assess the main physicocharacteristics of the lagoons. The pressures occurring in the catchment were collected in the literature. Graphical analyses were then conducted to highlight metrics responses to these environmental parameters. The variability between lagoons of benthic macrofauna is high but always dominated by freshwater taxa; consequently it has been possible to calculate most of the candidate metrics commonly used in freshwater systems. The natural and anthropogenic parameters of the six lagoons are also very different. No sound relationship was found between the intensity of pressures and the values of the metrics. These one-off samplings are not sufficient to get a robust description of the fauna communities; additionally, scarcity of the sites classified in oligohalin or mesohalin and the high environmental variability of these sites, prevent robust analyses. Some perspectives are discussed.; La macrofaune benthique est un des éléments de qualité biologique entrant dans la caractérisation de l’état écologique des lagunes. Sur les milieux oligohalins à mésohalines méditerranéens, les indices classiquement utilisés en milieux lagunaires plus salés ne sont pas applicables ; Dans ce contexte, nous avons exploré la possibilité de sélectionner des métriques issues des travaux conduits en milieux d’eau douce, pour rendre compte d’une altération de la qualité physico-chimique de 6 lagunes oligohalines à mésohalines du pourtour méditerranéen. Les prélèvements biologiques ont été effectués au printemps dans la zone centrale des lagunes et les taxons identifiés au laboratoire. A partir des compositions faunistiques observées, une cinquantaine de métriques candidates à la bioindication décrivant la composition et la diversité des communautés ont été calculées. Parallèlement, des prélèvements et mesures ont été faits afin de définir les principales caractéristiques physico-chimiques de l’eau et des sédiments à même d’influencer les peuplements d’invertébrés. Des données de pressions/forces motrices s’exerçant sur ces milieux ont aussi été collectées dans la littérature. Les données biologiques ont ensuite été confrontées aux données de milieux. Les résultats montrent de fortes variabilités dans les communautés biologiques, d’un point de vue qualitatif et quantitatif. Ils mettent en évidence aussi d’importantes différences dans les conditions environnementales naturelles et de pressions des 6 lagunes. Les communautés observées sont dominées par des taxons communément rencontrés en eaux douces ; de ce fait, une partie importante des métriques utilisées en bioindication sur ces milieux ont pu être calculées. La mise en relation des valeurs prises par ces métriques avec les variables de pression ne montre cependant pas de tendance nette et/ou écologiquement interprétable. Outre le fait que ces prélèvements ponctuels ne sont pas suffisants pour caractériser les lagunes, le nombre limité de sites et leur forte variabilité environnementale représente une difficulté majeure dans la démarche. Des solutions alternatives sont évoquées en perspectives.

8. Incidence of infectious disease during a spa therapy in overrates vulnerable population. A longitudinal controlled study on 738 patients

Abstract

OBJECTIF : Le but de ce travail est de détecter les facteurs de risque de survenue des infec- tions au cours d’une cure thermale. MÉTHODE : Les facteurs de risques potentiels ont été définis par consensus entre un pneumologue, un infectiologue et un rhumatologue thermal. Les pathologies présumées à risque étaient cancer ou hémopathie évolutifs, greffe d’organe, insuffisance respiratoire chronique, infection à virus de l’immunodéficience humaine. Les traitements potentiellement à risque étaient : méthotrexate ou néflunomide, traitement anti-TNF alpha, corticothérapie générale, traitement immunosuppresseur. Nous avons recruté prospectivement et consécutivement tous les curistes présentant au moins un facteur de risque et noté l’ensemble des événements survenant au cours de leur cure thermale. Les témoins étaient les curistes suivant immédiatement le curiste à risque sur la liste de rendez-vous. Les statistiques ont utilisé le test de Chi2 pour les comparaisons de variables qualitatives et le test U de Mann et Whitney pour les variables numériques. RÉSULTAT : Entre 2002 et 2004, les 10 médecins participant à l’étude ont surveillé 39150 cures sur les 98208 suivis aux Thermes nationaux d’Aix-les-Bains. Ils ont recruté 376 patients à risque et 362 témoins. Il y a eu 31 infections dans le groupe à risque et 20 dans le groupe témoins (7,6 % vs 4,9 %, p=0,14). Les analyses en sous-groupes de traitement ou de pathologie à risque ne montrent pas de différence significative avec les témoins, même si les chiffres sont proches du seuil pour les insuffisants respiratoires (24 % vs 7 % ; n=29, p=0,14). La réalisation d’une cure voies respiratoires (VR) est associée à un risque d’infection significatif chez les sujets à risque (29,1 % vs 0,0 % ; n=24, p=0,01). Le risque d’infection est particulièrement élevé dans le sous-groupe de patients effectuant une cure VR et présentant une insuffisance respiratoire (55 % vs 0 %, n=9 ; p=0,03). CONCLUSION : Les patients ayant une bronchite chronique ou une insuffisance respiratoire chronique, s’ils font une cure ORL associée à la cure rhumatologique ont un risque d’infection supérieur. Ce groupe semble donc présenter un véritable sur-risque d’infection et il faudra bien peser le rapport béné- fice /risque avant de leur proposer un séjour thermal., OBJECTIVE : The purpose of this study is to detect risk factor for incident infections during a spa therapy. METHODS : Potential risk factors were defined by consensus between a pneumologist, a specialist of infectious diseases and a senior rheumatologist practitioner of the spa center. Risk pathologies were cancer, hemopathy, organ graft, chronic bronchitis, respiratory deficiency and VIH infection. Risk treatments were methotrexate, neflunomide, TNF alpha blockers, corticoids, immunosupressors. We recruited prospectively and consecutively all patients treated in the spa center presenting almost one risk factor and note all diseases occurring during the spa stay. Controls were the patients following the risk patients on the appoint- ment list. Statistical anlysis used chi2 fort qualitative variable and U Mann Whitney test for continuous variables. RESULTS : Between 2002 and 2004, the 10 rheumatologist practitioners examined 39150 patients for spa therapy on the 98208 treated in the Aix-les-Bains spa centre. They recruited 376 risk patients and 362 controls. They were 31 infections in the risk group and 20 in the control group (7,6 % vs 4,9 %, p=0,14). Treatment or pathology subgroup analysis detected no significant difference with control even if, results are close of statistical significance for respiratory deficiency and chronic bronchitis (24 % vs 7 % ; n=29, p=0,14). Spa therapy for respiratory tract is associated with a significant increase of risk infection (29,1 % vs 0,0 % ; n=24, p=0,01). It is higher with patient subgroup associating spa for respiratory tract and chronic bronchitis or respiratory deficiency (55 % vs 0 %, n=9 ; p=0,03). CONCLUSION : Patients with chronic bronchitis or respiratory deficiency, if they have a spa therapy for respi- ratory tract have a higher incidence of infection. This group seems to present an over risk of infection and benefit/risk ratio have to be weighted before proposing them a spa stay.

10. Variability of the horizontal velocity structure in the upper 1600 m of the water column on the equator at 10 degrees W

Abstract

This paper presents initial results from new velocity observations in the eastern part of the equatorial Atlantic Ocean from a moored current-meter array. During the "EQUALANT" program (1999-2000), a mooring array was deployed around the equator near 10 W that recorded one year of measurements at various depths. Horizontal velocities were obtained in the upper 60 m from an upward-looking acoustic Doppler current profiler (ADCP) and at 13 deeper levels from current meters between 745 and 1525 m. To analyze the quasiperiodic variability observed in these records, a wavelet-based technique was used. Quasiperiodic oscillations having periods between 5 and 100 days were separated into four bands: 5-10, 10-20, 20-40, and 40-100 days. The variability shows (i) a strong seasonality (the first half of the series is dominated by larger periods than the second one) and (ii) a strong dependence with depth (some oscillations are present in the entire water column while others are only present at certain depths). For the oscillations that are present in the entire water column the origin of the forcing can be traced to the surface, while for the others the question of their origin remains open. Phase shifts at different depths generate vertical shears in the horizontal velocity component with relatively short vertical scales. This is especially visible in long-duration events (>100 days) of the zonal velocity component. Comparison with a simultaneous lowered acoustic Doppler current profiler (LADCP) section suggests that some of these flows may be identified with equatorial deep jets. A striking feature is a strong vertical shear lasting about 7 months between 745 and 1000 m. These deep current-meter observations would then imply a few months of duration for the jets in this region.

Abstract

Temperature, salinity, velocity, and wind from a mooring at 0 degrees N, 23 degrees W are used along with satellite data for sea surface temperature and sea level to examine the contribution of tropical instability waves ( TIWs) to the energy and heat balance of the equatorial Atlantic mixed layer. The TIWs appear as periodic 20 - 30 day fluctuations of currents, temperature, and salinity, which intensify beginning in June and peak in late boreal summer. The intensification occurs in phase with strengthening of the southeasterly trade winds and the seasonal appearance of the equatorial tongue of cold mixed layer temperatures. In 2002 these waves, which warm the mixed layer by 0.35 degrees C during summer months, are maintained by both barotropic and baroclinic conversions that are of comparable size. Salinity fluctuations, previously neglected, increase the magnitude of baroclinic energy conversion.

Abstract

Nous présentons des résultats d'expériences d'assimilation séquentielle de données altimétriques dans un modèle linéaire de l'océan Atlantique tropical. Nous assimilons des champs d'anomalies du niveau de la mer issus des mesures altimétriques du satellite Geosat. En dépit des faibles amplitudes des anomalies, nous observons des améliorations très nettes des simulations du modèle grâce aux assimilations. Nous montrons de ce fait que les données altimétriques peuvent se révéler très utiles pour améliorer les simulations des modèles en utilisant des techniques d'assimilation adaptées. Abstract

L'utilisation conjointe de données in situ et de données satellitaires altimétriques est appliquée à une région limitée de l'Atlantique central nord contenant la 3e boîte de l'expérience Topogulf. Cette application se divise en trois phases. La première est une "inversion" des données hydrographiques de cette 3ème boîte par modélisation variationnelle. La seconde consiste à utiliser la procédure de Ménard pour estimer la variabilité des courants géostrophiques de surface à partir des données des traces répétitives de Seasat. Enfin la synthèse de ces deux approches nous permet de discuter deux types très différents de solutions obtenus par inversion. Toutefois, nous devons nous garder d'interpréter hâtivement la variabilité obtenue par Seasat. Nous nous interrogeons également sur le concept de circulation générale. (Résumé d'auteur)

Abstract

Vaccination is a useful option to control infection with porcine reproductive and respiratory syndrome virus (PRRSV), and several modified live-PRRSV vaccines have been developed. These vaccines have shown some efficacy in reducing the incidence and severity of clinical disease as well as the duration of viremia and virus shedding but have failed to provide sterilizing immunity. The efficacy of modified live-virus (MLV) vaccines is greater against a homologous strain compared with heterologous PRRSV strains. The objective of this study was to evaluate the efficacy of Fostera PRRS MLV vaccine in protecting against challenge with a heterologous field strain widely circulating in the swine herds of eastern Canada. Forty-six piglets were divided into 4 groups: nonvaccinated-nonchallenged; nonvaccinated-challenged; vaccinated-challenged; and vaccinated-nonchallenged. The animals were vaccinated at 23 d of age with Fostera PRRS and challenged 23 d later with a heterologous field strain of PRRSV (FMV12-1425619). Overall, the vaccine showed some beneficial effects in the challenged animals by reducing the severity of clinical signs and the viral load. A significant difference between nonvaccinated and vaccinated animals was detected for some parameters starting 11 to 13 d after challenge, which suggested that the cell-mediated immune response or other delayed responses could be more important than pre-existing PRRSV antibodies in vaccinated animals within the context of protection against heterologous strains.La vaccination est une option utile pour limiter l’infection par le virus du syndrome reproducteur et respiratoire porcin (VSRRP), et plusieurs vaccins VSRRP vivants modifiés ont été développés. Ces vaccins ont démontré une certaine efficacité à réduire l’incidence et la sévérité de la maladie clinique ainsi que la durée de la virémie et de l’excrétion virale mais ont failli à produire une immunité stérilisante. L’efficacité des vaccins vivants modifiés (VVM) est supérieure contre une souche homologue comparativement à des souches hétérologues de VSRRP. L’objectif de la présente étude était d’évaluer l’efficacité du vaccin Fostera, un VVM contre le VSSRP, à protéger contre une infection défi avec une souche de terrain hétérologue circulant librement dans les troupeaux porcins de l’est du Canada. Quarante-six porcelet ont été répartis en quatre groupes : non vaccinés-non infectés; non vaccinés-infectés; vaccinés-infectés; et vaccinés-non infectés. Les animaux ont été vaccinés à 23 jours d’âge avec Fostera SRRP et infectés 23 jours plus tard avec une souche de terrain hétérologue du VSRRP (FMV12-1425619). De manière générale, le vaccin a démontré quelques effets bénéfiques chez les animaux infectés en réduisant la sévérité des signes cliniques et la charge virale. Une différence significative entre les animaux non vaccinés et ceux vaccinés a été détectée pour quelques paramètres et débutant 11 à 13 j suite à l’infection, ce qui suggère que la réponse de l’immunité à médiation cellulaire ou d’autres réponses retardées pourraient être plus importantes que la présence d’anticorps anti-SRRP existants chez des animaux vaccinés dans le contexte d’une protection contre des souches hétérologues.(Traduit par Docteur Serge Messier).

Abstract

We present results of sequential data assimilation of altimetric data in a linear tropical Atlantic model, using an analysis technique that we have developed. We use sea level anomaly fields obtained from altimetric data of the satellite Geosat. Despite the weak amplitudes of the anomaly fields, we obtain marked improvements of the model simulations due to assimilations. Thus, we show that altimetric data may be very useful for improving model simulations when adapted assimilation techniques are applied.

Abstract

International Altimetry Team., In 2018 we celebrated 25 years of development of radar altimetry, and the progress achieved by this methodology in the fields of global and coastal oceanography, hydrology, geodesy and cryospheric sciences. Many symbolic major events have celebrated these developments, e.g., in Venice, Italy, the 15th (2006) and 20th (2012) years of progress and more recently, in 2018, in Ponta Delgada, Portugal, 25 Years of Progress in Radar Altimetry. On this latter occasion it was decided to collect contributions of scientists, engineers and managers involved in the worldwide altimetry community to depict the state of altimetry and propose recommendations for the altimetry of the future. This paper summarizes contributions and recommendations that were collected and provides guidance for future mission design, research activities, and sustainable operational radar altimetry data exploitation. Recommendations provided are fundamental for optimizing further scientific and operational advances of oceanographic observations by altimetry, including requirements for spatial and temporal resolution of altimetric measurements, their accuracy and continuity. There are also new challenges and new openings mentioned in the paper that are particularly crucial for observations at higher latitudes, for coastal oceanography, for cryospheric studies and for hydrology. The paper starts with a general introduction followed by a section on Earth System Science including Ocean Dynamics, Sea Level, the Coastal Ocean, Hydrology, the Cryosphere and Polar Oceans and the “Green” Ocean, extending the frontier from biogeochemistry to marine ecology. Applications are described in a subsequent section, which covers Operational Oceanography, Weather, Hurricane Wave and Wind Forecasting, Climate projection. Instruments’ development and satellite missions’ evolutions are described in a fourth section. A fifth section covers the key observations that altimeters provide and their potential complements, from other Earth observation measurements to in situ data. Section 6 identifies the data and methods and provides some accuracy and resolution requirements for the wet tropospheric correction, the orbit and other geodetic requirements, the Mean Sea Surface, Geoid and Mean Dynamic Topography, Calibration and Validation, data accuracy, data access and handling (including the DUACS system). Section 7 brings a transversal view on scales, integration, artificial intelligence, and capacity building (education and training). Section 8 reviews the programmatic issues followed by a conclusion., At the forefront of this support, we must obviously mention the space agencies CNES, ESA and NASA which have played and still play a decisive role in the development and launch of several prominent altimetry missions from the outset. Other agencies such as DLR, EUMETSAT, ISRO, NOAA, NSOAS and organizations such as CMEMS, also contribute significantly to developments in all forms of altimetry.
