Abstract

In chicken, the ultimate pH (pHu) of meat is largely determined by the muscle glycogen content at slaughter age.A divergent selection on the Pectoralis major pHu allowed the creation of the pHu+ and pHu- lines, whichrepresent a unique model for studying the genetic and physiological control of energy reserves and the meatquality. Several characteristics have been measured in the two lines at hatch then at 5 days of age: animal weight,muscle yields, muscular and hepatic glycogen content and several plasma parameters. At hatch, the pHu+ andpHu- lines present equivalent body weights and muscle yields. However, the pHu- line shows a higher glycemiathan the pHu+ line, a difference that will persist until the slaughter age. In addition, primary myoblasts culturesfrom both lines show differences in insulin response. After 5 days, the pHu+ and pHu- lines are alreadydivergent in terms of glycogen (and pHu) and muscle yield, for a similar body weight. Differences between bothlines observed at hatch and at 5 days may have been associated with regulation of signaling pathways involvedin protein synthesis and muscle glycogen turnover. In conclusion, pHu+ and pHu- chicks already have at hatchthe ability to respond differently to nutrients and hormones consistent with the fact that after only 5 days offeeding, significant differences in yield and muscular glycogen contents exist between the two lines.; Chez le poulet, le pH ultime (pHu) de la viande est en grande partie déterminé par la teneur en glycogène dumuscle à l'abattage. Une sélection divergente sur le pHu du filet a permis la création des lignées pHu+ et pHu-,qui représentent un modèle unique pour étudier le contrôle génétique et physiologique des réserves énergétiqueset de la qualité de la viande. Plusieurs caractéristiques ont été mesurées dans les deux lignées à l’éclosion puis à5 jours d’âge: le poids vif, les rendements en muscles, le glycogène musculaire et hépatique et plusieursparamètres plasmatiques. A l’éclosion, les lignées pHu+ et pHu- présentent des poids vifs et des rendementsmusculaires équivalents. Toutefois, la lignée pHu- présente une glycémie supérieure à celle de la lignée pHu+,différence qui va persister jusqu'à l'âge d’abattage. Par ailleurs, les cultures primaires des myoblastes issues desdeux lignées révèlent des différences de réponse à l'insuline. Après 5 jours, les lignées pHu+ et pHu- sont déjàdivergentes en terme de glycogène (et de pHu) et de rendement en muscle, pour un poids vif similaire. Lesdifférences entre lignées observées à l'éclosion et à 5 jours ont pu être associées à des régulations au niveau desvoies de signalisation impliquées dans la synthèse protéique et le turn-over du glycogène musculaire. Enconclusion, les poussins pHu+ et pHu- présentent dès l’éclosion des capacités de réponses différentes auxnutriments et aux hormones cohérentes avec le fait qu’après seulement 5 jours d’alimentation, des différencessignificatives de rendement et de teneur en glycogène musculaire existent entre les deux lignées.

Abstract

The processing ability and sensory quality of chicken breast meat are highly related to its ultimate pH (pHu), which is mainly determined by the amount of glycogen in the muscle at death. To unravel the molecular mechanisms underlying glycogen and meat pHu variations and to identify predictive biomarkers of these traits, a transcriptome profiling analysis was performed using an Agilent custom chicken 8 × 60 K microarray. The breast muscle gene expression patterns were studied in two chicken lines experimentally selected for high (pHu+) and low (pHu−) pHu values of the breast meat. Across the 1,436 differentially expressed (DE) genes found between the two lines, many were involved in biological processes related to muscle development and remodelling and carbohydrate and energy metabolism. The functional analysis showed an intensive use of carbohydrate metabolism to produce energy in the pHu− line, while alternative catabolic pathways were solicited in the muscle of the pHu+ broilers, compromising their muscle development and integrity. After a validation step on a population of 278 broilers using microfluidic RT-qPCR, 20 genes were identified by partial least squares regression as good predictors of the pHu, opening new perspectives of screening broilers likely to present meat quality defects.

Abstract

The enzyme beta, beta-carotene-15,15'-monooxygenase (BCMO1) cleaves provitamin A carotenoids into active vitamin A principally in liver and intestine. The BCMO1 gene is expressed at low level in the muscle tissue but little is known about its function. In the chicken muscle, we observed that various BCMO1 expression levels are associated with different carotenoids contents. To investigate the potential role of BCMO1 on skeletal muscle, we assessed the impact of beta-carotene (BC, the prototype substrate of the BCMO1 enzyme) supplementation in vitro on proliferative avian myoblasts. Proliferation was evaluated by BrdU incorporation and by flow cytometry. The BrdU incorporation index was reduced and th e proportion of G0/G1 cells increased following BC supplementation. Cell differentiation was evaluated by immunolabelling of sarcomeric myosin heavy chain (MHC). In this proliferative environment, the proportion of MHC positive cells increased following BC supplementation. The effects of BC were inhibited in the presence of DEAB, an inhibitor of retinaldehyde dehydrogenase, supporting the hypothesis that the BCMO1 enzyme is active in myoblasts and can contribute to retinoic acid production from BC. Our data suggest that provitamin A carotenoids could be used as nutritional regulators of skeletal muscle growth

Abstract

Avian gametes present specific features related to their internal long-term mode of fertilization. Among other central actors of energetic metabolism control, it has been suspected that 5'-AMP-activated protein kinase (AMPK) influences sperm functions and thus plays a key role in fertilization success. In the present work, we studied AMPK localization and function in chicken sperm incubated in vitro. Effects of the pharmacological AMPK activators (AICAR, metformin) and the AMPK inhibitor compound C were assessed by evaluating AMPKalpha (Thr(172)) phosphorylation (by Western blotting), semen quality (by viability, motility, and ability to perform acrosome reaction), and energetic metabolism indicators (lactate, ATP). Localization of AMPK in subcellular sperm compartments was evaluated by immunocytochemistry. Total AMPK was found in all compartments except for the nucleus, but the phosphorylated form phospho-Thr(172)-AMPK was essentially localized in the flagellum and acrosome. AMPK activators significantly improved AMPK phosphorylation, sperm motility (increased by 40% motile, 90% progressive, and 60% rapid sperm), acrosome reaction and lactate production (increased by 40%) and viability. The AMPK inhibitor significantly reduced AMPK phosphorylation and percentages of motility (decrease by 25%), progressive energy (decrease by 35%), and rapid sperm (decreased by 30%), acrosome reaction, lactate production, and ATP release. The two activators differed in their effect on ATP concentration: AICAR stimulated ATP formation, whereas metformin did not. Our results indicate that AMPK plays a key role in the regulation of chicken sperm functions and metabolism. This action differs from that suggested in mammals, mainly by its crucial involvement in the acrosome reaction process.

The aim of this study was to determine the long-lasting effects of heat manipulation of the embryo on the physiology, body temperature (Tb), growth rate and meat processing quality of broiler chickens reared in floor pens. Broiler chicken eggs were incubated in control conditions (37.8 degrees C, 56% relative humidity; RH) or exposed to thermal manipulation (TM; 12 h/d, 39.5 degrees C, 65% RH) from d 7 to 16 of embryogenesis. This study was planned in a pedigree design to identify possible heritable characters for further selection of broiler chickens to improve thermotolerance. Thermal manipulation did not affect hatchability but resulted in lower Tb at hatching and until d 28 post-hatch, with associated changes in plasma thyroid hormone concentrations. At d 34, chickens were exposed to a moderate heat challenge (5 h, 32 degrees C). Greater O-2 saturation and reduced CO2 partial pressure were observed (P < 0.05) in the venous blood of TM than in that of control chickens, suggesting long-term respiratory adaptation. At slaughter age, TM chickens were 1.4% lighter and exhibited 8% less relative abdominal fat pad than controls. Breast muscle yield was enhanced by TM, especially in females, but without significant change in breast meat characteristics (pH, color, drip loss). Plasma glucose/insulin balance was affected (P < 0.05) by thermal treatments. The heat challenge increased the heterophil/lymphocyte ratio in controls (P < 0.05) but not in TM birds, possibly reflecting a lower stress status in TM chickens. Interestingly, broiler chickens had moderate heritability estimates for the plasma triiodothyronine/thyroxine concentration ratio at d 28 and comb temperature during the heat challenge on d 34 (h(2) > 0.17). In conclusion, TM of the embryo modified the physiology of broilers in the long term as a possible adaptation for heat tolerance, without affecting breast meat quality. This study highlights the value of 2 new heritable characters involved in thermoregulation for further broiler selection.

Abstract

Chantier qualité GA; International audience; A polymorphism in the promoter of the β,β-carotene 15,15'-monooxygenase 1 (BCMO1) gene recently was identified in an experimental cross between 2 chicken lines divergently selected on growth rate and found to be associated with variations in the yellow color of the breast meat. In this study, the effects of the polymorphism on several aspects of carotenoid metabolism were evaluated in chickens sharing the same genetic background except for their genotype at the BCMO1 locus. We confirmed that BCMO1 mRNA abundance varied (P < 0.001) between the 2 homozygous genotypes (GG < AA) and in the pectoralis major muscle. By contrast, BCMO1 mRNA expression was not affected (P > 0.05) by the polymorphism in the duodenum, liver, or sartorius muscle. The breast meat of GG chickens was more (P < 0.001) yellow and richer in lutein (P < 0.01) and zeaxanthin (P < 0.05) compared to that of AA chickens whereas these variables did not differ (P > 0.05) in the other tissues tested. The GG were also characterized by reduced (P < 0.01) plasma lutein and zeaxanthin concentrations than AA without affecting plasma and tissue content of fat-soluble vitamins A and E. As lutein and zeaxanthin are usually not considered as substrates of the BCMO1 enzyme, the impact of BCMO1 polymorphism on the activity of other genes involved in carotenoid transport (SCARB1 and CD36 encoding the scavenger receptor class B type I and the cluster determinant 36, respectively) and metabolism (BCDO2 encoding β,β-carotene 9',10'-dioxygenase 2) was evaluated. The BCMO1 polymorphism did not affect mRNA abundance of BCDO2, SCARB1, or CD36, regardless of tissue considered. Taken together, these results indicated that a genetic variant of BCMO1 specifically changes lutein and zeaxanthin content in the chicken plasma and breast muscle without impairing vitamin A and E metabolism.
