Abstract

International audience; We determined the frequencies of clinically relevant blood group alleles in 300 blood donors from Mali. Multiplex test based on xMAP technology was used to investigate six blood group systems (RH, KEL, MNS, FY, JK, DO, HPA) and complementary analysis were conducted for MNS and RH systems. Polymorphisms that affect the specificity of molecular tests leading to discrepant genotype results are discussed. Antigen expressions were predicted showing that 50% of donors expressed at least one traditional low prevalence antigen, and 11.6% lacked the ability to express at least one high prevalence antigen compatible with Dob-, HPA1a-, S-s-U-, Jsb-, RH:-31 and/or RH:-34 phenotypes.

Abstract

The Surface Ocean CO2 Atlas (SOCAT) is a synthesis of quality-controlled fCO2 (fugacity of carbon dioxide) values for the global surface oceans and coastal seas with regular updates. Version 3 of SOCAT has 14.7 million fCO2 values from 3646 data sets covering the years 1957 to 2014. This latest version has an additional 4.6 million fCO2 values relative to version 2 and extends the record from 2011 to 2014. Version 3 also significantly increases the data availability for 2005 to 2013. SOCAT has an average of approximately 1.2 million surface water fCO2 values per year for the years 2006 to 2012. Quality and documentation of the data has improved. A new feature is the data set quality control (QC) flag of E for data from alternative sensors and platforms. The accuracy of surface water fCO2 has been defined for all data set QC flags. Automated range checking has been carried out for all data sets during their upload into SOCAT. The upgrade of the interactive Data Set Viewer (previously known as the Cruise Data Viewer) allows better interrogation of the SOCAT data collection and rapid creation of high-quality figures for scientific presentations. Automated data upload has been launched for version 4 and will enable more frequent SOCAT releases in the future. High-profile scientific applications of SOCAT include quantification of the ocean sink for atmospheric carbon dioxide and its long-term variation, detection of ocean acidification, as well as evaluation of coupled-climate and ocean-only biogeochemical models. Users of SOCAT data products are urged to acknowledge the contribution of data providers, as stated in the SOCAT Fair Data Use Statement. This ESSD (Earth System Science Data) "living data" publication documents the methods and data sets used for the assembly of this new version of the SOCAT data collection and compares these with those used for earlier versions of the data collection (Pfeil et al., 2013; Sabine et al., 2013; Bakker et al., 2014). Abstract

International audience; Bolidomonas is a genus of picoplanktonic flagellated algae that is closely related to diatoms. Triparma laevis, a species belonging to the Parmales, which are small cells with a siliceous covering, has been shown to form a monophyletic group with Bolidomonas. We isolated several novel strains of Bolidophyceae that have permitted further exploration of the diversity of this group using nuclear, plastidial and mitochondrial genes. The resulting phylogenetic data led us to formally emend the taxonomy of this group to include the Parmales within the Bolidophyceae, to combine Bolidomonas within Triparma and to define a novel species, Triparma eleuthera sp. nov. The global distribution of Bolidophyceae was then assessed using environmental sequences available in public databases, as well as a large 18S rRNA V9 metabarcode data set from the Tara Oceans expedition. Bolidophyceans appear ubiquitous throughout the sampled oceans but always constitute a minor component of the phytoplankton community, corresponding to at most ~ 4% of the metabarcodes from photosynthetic groups (excluding dinoflagellates). They are ~ 10 times more abundant in the small size fraction (0.8– 5 μm) than in larger size fractions. T. eleuthera sp. nov. constitutes the most abundant and most widespread operational taxonomic unit (OTU) followed by T. pacifica, T. mediterranea and the T. laevis clade. The T. mediterranea OTU is characteristic of Mediterranean Sea surface waters and the T. laevis clade OTU is most prevalent in colder waters, in particular off Antarctica.

Abstract

International audience; In this study, we identified 9 polymorphic compound microsatellite markers in the toxic dinoflagellate Alexandrium catenella, which was isolated from Thau lagoon (France, Mediterranean Sea), using the compound microsatellite marker technique. These new microsatellites were characterized by screening DNA templates from 43 A. catenella clonal strains, which were collected from a seawater sample from Inokushi Bay (Oita Prefecture, Japan). These loci provide one class of highly variable genetic marker: the number of alleles ranged from 3 to 8, and the estimate of gene diversity varied between 0.285 and 0.762. We also analyzed the same 43 DNA samples using microsatellite markers previously identified for A. catenella, comparing the PCR amplification success, the number of alleles and gene diversity. These three parameters were not significantly different, showing that the compound microsatellite markers have the same potential to reveal A. catenella genetic structure. This simple and efficient method reduces the costs for developing SSR markers and for analyzing the genetic structure of populations, therefore, suggesting the effectiveness of applying this method to other species.

6. VERTICAL FLUX OF ICE ALGAL CELLS DURING THE ICE MELTING AND BREAKING PERIODS IN SAROMA KO LAGOON, HOKKAIDO, JAPAN (18th Symposium on Polar Biology)

Abstract

The vertical flux of ice algae was determined with a multiple sediment trap at 4 m depth at the central station in Saroma Ko lagoon, Hokkaido, Japan. Collection was done in 5-day intervals from March 1 to April 24, 1994. The sea ice started breaking at the end of that period, from April 10 to 14,1994 and disappeared completely from the lagoon on April 17, 1994. The average fluxes of CHLa, PHEO, POC, PON, and BioSi during the melting season of sea ice were 20±6.5 mg CHLa, 2.0±1.0 mg PHEO, 630±190 mg C, 110±39 mg N, and 490±170 mg Si m^ d^, respectively. The average % loss rates, determined by the ratio of the average vertical flux to the average ice algal production estimated from the standing stock and the growth rate, was calculated as 100% d^ for CHLa during the ice melting period. Once the sea ice started breaking and disappeared, they increased by at least 1.5 fold. A relatively high increase was observed for PHEO (2.5 fold) and BioSi (2.3 fold) compared to the lowest increase of CHLa (1.5 fold). This implies that either the vertical flux of CHLa has been depressed due to unfavorable physical conditions, such as low salinity produced by a combination of melting sea ice and runoff of fresh water and exposure to high light provided by a disturbed position of broken sea ice, or the vertical flux of BioSi has been accelerated through physical breakage of ice algal cells by heterotrophic grazing and possibly resuspension of PHEO-rich water induced by wind forced mixing, evidenced by a significant increase of PHEO under the ice free water. Wind-forced resuspension should be considered in any study of vertical flux of ice algae when sea ice disappears in shallow water.

Abstract

The contributions of heterotrophic and autotrophic prey to the diet of Euphausia pacifica were examined in coastal waters off northeastern Japan in May and October 1999. The autotrophic contribution was estimated from gut pigment contents, and the heterotrophic contribution from the volume of each taxonomic group in the stomach. Our observations suggest that heterotrophic prey, especially copepods, were more important than autotrophic prey in both seasons. The contributions of heterotrophic prey items in adult E. pacifica ranged from 57-79% in May and from 27-93% of the total ingested carbon in October. The contribution of copepods was largest, ranging from 54-77% in May and from 27-93% in October. We discuss the contribution of athecate ciliates, another possible heterotrophic prey of importance, to the diet of E. pacifica.

Abstract

The respiration rate of adult female Calanus hyperboreus was determined in order to assess physiological activity and its relationship to food availability during spring in the North Water Polynya, northern Baffin Bay. The respiration rate increased hyperbolically with increasing ambient chlorophyll α (Chl. α) concentration regardless of the reproductive status. The increase of respiration with Chl. α concentration may be caused by activated feeding behavior. This suggests that all adult females during spring were physiologically active individuals. Possible advantages of the rapid response to food concentration in adult female copepods are discussed.

Abstract

Experiments were conducted during January 1995 in the Southern Ocean to examine the impact of enhanced solar ultraviolet-B radiation (UVBR) on the biosynthesis and composition of organic compounds in Antarctic marine phytoplankton. Our results revealed distinct changes in the biochemical composition of phytoplankton attributable to UVBR exposure. Fatty acid concentrations increased in the presence of UVBR, mainly on account of a large increase in the content of saturated fatty acids within the cells. On the other hand, polyunsaturated fatty acids declined in cells exposed to UVBR. Amino acid concentrations were higher in the UVBR exposed samples, attributable largely to a UVBR induced increase in cellular concentrations of glutamic' acid (glutamic acid+glutamine) and aspartic acid. Monosaccharide constituents of cellular storage and structural carbohydrates, however, showed a decline in the cells exposed to UVBR. Except for the decline in structural monosaccharides, these changes in the patterns of organic compounds observed in Antarctic phytoplankton were remarkably similar but, greater in magnitude in comparison to those observed in temperate phytoplankton exposed to UVBR.

13. ESTIMATION OF VERTICAL DISTRIBUTION OF CHLOROPHYLL a OFF EAST HOKKAIDO BY GAUSSIAN CURVE FITTING (15th Symposium on Polar Biology)

Abstract

Sea truth data on the concentration of chlorophyll pigments were used to determine the empirical relationship between surface chlorophyll a concentration and parameters derived from a Gaussian curve for a possible development of a local algorithm of primary production in the western subarctic North Pacific Ocean. Data were obtained from the line between 39°N, 146°45'E and 42°50'N, 144°50'E from April 1990 to July 1991. One optical depth (1/k) can be estimated from the concentration of chlorophyll a (B_c) at the surface depth. Surface Bc can be estimated from B_c+B_p (concentration of pheopigments) in one optical depth. The background concentration of chlorophyll a (B_o), the depth of the subsurface chlorophyll maximum (z_m), and the total biomass above the background (h) which control the Gaussian curve can be determined from the surface Bc. Therefore, once surface Bc is obtained, the vertical profile of Bc is determined. However relative variability in the estimated values from the surface Bc can be high when the standing stock of chlorophyll a is lower than 300 mg Chl. a m^. This may suggest that the present model can be applied only to a relatively productive area. The standing crop of chlorophyll pigments in one optical depth, which can be estimated from the ocean color obtained from the satellite, would be useful to determine the standing crop of chlorophyll pigments in the euphotic layer.

14. FEEDING HABITS OF PACIFIC COD, GADUS MACROCEPHALUS, OFF EASTERN HOKKAIDO, NORTH JAPAN (14th Symposium on Polar Biology)

Abstract

We collected a total of 336 specimens of Pacific cod, Gadus macrocephalus during April, May and June 1989-1992 off the eastern coast of Hokkaido. Examination of their stomach contents revealed that fishes (mainly walleye pollock, Theragra chalcogrammd) and decapod crustaceans (mainly crangonid shrimps) were major diet. The cod showed an obvious body size related dietary shift; the importance of shrimps (Neocrangon communis and Argis lar) and myctophid fishes for ≤300 mm individuals switched abruptly to walleye pollock and other fishes (e.g. Sardinops melanostictus, Laemonema longipes, stichaeid and cottid fishes) and octopus, Paroctopus spp. for larger individuals. The cod showed an obvious feeding periodicity with a peak of feeding during 0600-1200 h. The comparison of prey size and prey type (benthos and nekton) for different size classes of predators indicated that the cod diet is determined by predator-prey size relationship.

15. EFFECT OF SHAPE OF SEDIMENT TRAP ON MEASUREMENT OF VERTICAL FLUX OF PARTICLES : PRELIMINARY RESULTS (14th Symposium on Polar Biology)

Abstract

Settling experiments were conducted to study sinking characteristics of phytoplankton cells within sediment traps. Simulated traps with three different shapes, i.e. cylindrical and conical shape with 75 and 60 degree angles, were employed for laboratory experiments. Navicula sp. as non-motile species and Tetraselmis sp. as motile one were employed for the experiment. Vertical flux of phytoplankton cells was determined continuously with a fluorometer. The present study indicated that conical shape traps caused underestimation of flux and the degree of underestimation was a function of wall angles. The underestimation was accelerated for Tetraselmis sp. The present results may suggest that a deployment of conical shape traps with collectors at the bottom would seriously underestimate the vertical flux of particles in the ocean.

16. Usage of seawater system for long-term study of phytoplankton in the boreal coastal water of Hokkaido, Japan

Weekly sampling was begun in October 1989 for 13 months to monitor the seasonal change of environmental characteristics and biological events in the boreal coastal water of Hokkaido, Japan. Water sample was taken from a seawater system for aquaculture studies. Concentration of chlorophyll a indicated clearly a seasonality. The low concentration was observed in winter, and spring bloom peaked on March 5 and lasted for eight weeks. The termination of productive season was a result of the change of water mass which occurred in May and June. During summer the concentration was lower than 1.0mg・m^. No fall bloom was observed. The present monitoring program using the existing seawater system will help to identify the succession of water mass, the seasonal change of chemical environments, and the timing and duration of spring bloom, which may be the important factor for maintenance of the coastal ecosystem including larval fish oroduction.
