The northeastern China cold vortex (NCCV) is the main weather system affecting Northeast China. Based on the precipitation products from the dual-frequency precipitation radar (DPR) onboard the Global Precipitation Measurement core observatory (GPM) satellite, the precipitation structures and microphysical properties for different rain types in 6432 NCCVs from 2014 to 2019 were studied using dynamic composite analysis. Our results show that the precipitation in NCCVs is dominated by stratiform precipitation. Regions with high stratiform and convective precipitation frequency have a comma shape. The growth mechanism of precipitation particles changes at ~4 km in altitude, the lower particles grow through collision (more pronounced in convective precipitation), and the upper hydrometeors grow through the Bergeron process. Additionally, the precipitation structures and microphysical properties exhibit great regional variations in NCCVs. The rainfall for all rain types is the strongest in the southeast region within an NCCV, mainly characterized by higher near-surface droplet concentration, while precipitation events occur more frequently in the southeast region for all rain types. There are active rimming growth processes above the melting layer for convective precipitation in the western region of an NCCV. In the southeast region of an NCCV, the collision growth of droplets in both types of precipitation is the most obvious.

The isothermal crystallization properties of polypropylene/graphite nanosheet (PP/GN) nanocomposites under supercritical N2 were systematically studied by a self-made in situ high-pressure microscope system. The results showed that the GN caused irregular lamellar crystals to form within the spherulites due to its effect on heterogeneous nucleation. It was found that the grain growth rate exhibits a decreasing and then increasing trend with the enhancement of N2 pressure. Using the secondary nucleation model, the secondary nucleation rate for spherulites of PP/GN nanocomposites was investigated from an energy perspective. The increase in free energy introduced by the desorbed N2 is the essential reason for the increase in the secondary nucleation rate. The results from the secondary nucleation model were consistent with those acquired through isothermal crystallization experiments, suggesting that the model can accurately predict the grain growth rate of PP/GN nanocomposites under supercritical N2 conditions. Furthermore, these nanocomposites demonstrated good foam behavior under supercritical N2.

Aim. To develop a set of infectious disease emergency response competencies specific to frontline nurses in China. Background. Nurses play an important role in the infectious disease emergency response. Competency-based training is the cornerstone of the professionalization of disaster rescue, including the infectious disease emergency response. Accordingly, reaching a consensus on a set of core competencies is essential. However, information regarding the competencies needed for nurses in the infectious disease emergency response is limited. Methods. A literature review and in-depth expert interviews were conducted to establish a draft of competencies, which consisted of 53 items, including 3 first-level index items, 12 second-level index items, and 38 third-level index items. Eighteen experts with the knowledge of infectious disease management and experience with infectious disease emergency rescue from different regions in China were recruited for Delphi consultation. The statistical characteristics of precipitation microphysics in lightning clouds are not yet fully understood, as a result of the limitations of traditional observational methods. Using the latest observations from the dual-frequency radar and microwave imager onboard the Global Precipitation Mission (GPM) and ground-based lightning observations, the precipitation microphysics of precipitation features with and without lightning (LPFs and NLPFs) was investigated across four typical regions of China in summer during the time period of 2014–2021. The statistical results show that the LPFs are characterized by smaller concentration and larger mass-weighted mean diameter (Dm) for rain and ice hydrometeors than those of NLPFs. Below the melting layer, the radar reflectivity (Ze) for both the LPFs and NLPFs generally decreases toward the surface, indicating the evaporation or strong break-up of rain hydrometeors. Above the melting layer, the Ze values mainly increase as the altitudes decrease for both LPFs and NLPFs, indicating the rimming, aggregation, or deposition processes. However, the change in slope is much smaller for the LPFs than for the NLPFs, which suggests a more uniform distribution of large ice hydrometeors at high altitudes, probably as a result of the stronger updrafts within the LPFs. The microphysical structures of the LPFs show great regional differences among the four regions of China, which is characterized by the low concentration of large-sized rain hydrometeors over Northeast China, and a high concentration of small-sized rain hydrometeors near the surface over the Yangtze-Huaihe River basin.

Background Infection is one of the most common causes of death in patients with hematological malignancies during chemotherapy. Due to its special location, local warmth and humidity, repeated pollution with stool and urine, and characteristically wrinkled anatomical structure within which bacteria can hide, the perianal becomes a site with a high incidence of infection. Such infection also has a high recurrence rate and high mortality, increasing the economic burden of patients, delaying the time of treatment and reducing the quality of life. In severe cases, sepsis occurs and endangers the patient’s life. Previous studies have confirmed the effectiveness of povidone iodine (PI) in the prevention of perianal infection in patients with hematological malignancies during chemotherapy, but these reports have not documented in detail the adverse events associated with sitz bathing and the lack of randomized controlled trials of different concentrations of dilute povidone iodine sitz bathing. Therefore, the evidence is insufficient. Hence, the objective of this paper is to determine whether a povidone iodine diluent sitz bath can reduce the incidence of perianal infection compared with conventional perianal cleaning care and to observe the incidence of perianal infection, the severity of perianal infection, and the complications related to the sitz bath among groups treated with different concentrations of povidone iodine diluent, especially in high-risk patients prone to perianal infection, to screen for the optimal concentration. Methods The trial is designed as a single-center, parallel, randomized, controlled and intervention trial with four parallel groups, and a primary endpoint of perianal infection occurred after this hospitalization chemotherapy. Randomization will be performed as simple randomization with a 1:1:1:1 allocation. This study received full ethics committee approval. The first patient was enrolled on May 1, 2021. A total of 268 patients with hematological malignancies undergoing chemotherapy who have risk factors for perianal infection will be enrolled with informed consent and randomly allocated to one of the four arms receiving (1) perianal cleaning care (control group D), (2) 1:100 PI diluted sitz bath (intervention group A), (3) 1:200 PI diluted sitz bath (intervention group B), and (4) 1:300 PI diluted sitz bath (intervention group C). The primary endpoint of the trial was the incidence of perianal infection. The secondary endpoints of the study will be the results of anal swab bacterial culture, the severity of perianal infection, the incidence of perianal adverse events (dryness, peeling, pigmentation, burning sensation), and pain scores. The length of hospitalization in days and hospitalization expenses will be recorded. Safety will be assessed with consideration of all adverse and severe adverse events related to the study treatment. Discussion We hypothesized that patients with hematological malignancies during chemotherapy would benefit from a povidone iodine diluted sitz bath. This study will provide evidence-based recommendations for clinicians and nurses. Trial registration Chinese Clinical Trial Registry (registration ID: ChiCTR2000041073). Protein-stabilized gold nanoclusters (Prot-Au NCs) have been widely used in biosensing and cell imaging owing to their excellent optical properties and low biotoxicity. However, several Prot-Au NCs reported in the literature do not retain the biological role of the protein, which greatly limits their ability to directly detect biomarkers. This study demonstrated for the first time the successful synthesis of dual-function avidin-stabilized gold nanoclusters (Av–Au NCs) using a one-pot method. The resulting Av–Au NCs exhibited intense blue and red emissions under 374 nm excitation. Furthermore, the Av–Au NCs retained the native functionality of avidin to bind to biotin. When DNA strands modified with biotin at both ends (i.e., linker chains) were mixed with Av–Au NCs, large polymers were formed, indicating that Av–Au NCs could achieve fluorescence signal amplification by interacting with biotin. Taking advantage of the aforementioned properties, we constructed a novel enzyme-free fluorescent biosensor based on the Av–Au NCs-biotin system to detect DNA. The designed fluorescent biosensor could detect target DNA down to 0.043 nM, with a wide line range from 0.2 nM to 20 µM. Thus, these dual-functional Av–Au NCs were shown to be an excellent fluorescent material for biosensing. Graphic abstract

The design of solid electrolytes with a wide electrochemical stability window and high Li-ion conductivity is a prerequisite for the realization of all solid-state Li batteries, which promises to enable extraordinary levels of safety for the battery system and may potentially revolutionize the energy storage field. Among all the promising inorganic solid electrolytes, Li4MS4 (M = Si, Ge, and Sn) with a crystal structure of Pnma symmetry have recently been recommended due to their good air stability and ionic conductivity. Here, we employ ab initio simulations to conduct a systematic investigation of the electrochemical stability and Li conductivity of Li4MS4 compounds. Our computation results reveal that the edge-sharing and face-sharing characteristics of LiS4 and LiS6 polyhedra not only facilitate the formation of a percolating Li diffusion network but would severely destabilize the crystal structure as well, thus resulting in a rather narrow electrochemical window. Although the stronger M-S bonds manifested in Li4SiS4 can benefit the overall stability, unfortunately, it also contributes to a more rugged energy landscape that inhibits Li diffusion. Li4SnS4 with a less densely packed lattice exhibits a substantially lower energy for Li ions to be accommodated at interstitial sites, which is a trigger for high Li conductivity in the bulk material, reaching 11 mS/cm at room temperature. The fast Li diffusion occurs through the concerted migration of multiple Li ions at lattice and interstitial sites. These findings open up new possibilities for the rational design of Li4MS4 (M = Si, Ge, and Sn) solid electrolytes for next-generation Li batteries.

This study aimed to investigate compassion satisfaction (CS) and compassion fatigue (CF) in haematology nurses and their associated factors.A cross-sectional survey.The survey was conducted on 336 haematology nurses from 21 hospitals in Sichuan, China. The CS and CF were assessed by the Professional Quality of Life Scale version 5. The CF was determined by burnout and secondary traumatic stress.Haematology nurses in China had moderate levels of CS and moderate-to-low CF. Better nursing competence of teaching/consultation and communication/coordination and the percentage of critically ill patients60% predicted higher CS. The permanent nurse, better nursing competence of communication/coordination and specialized clinical practice predicted less burnout, while working40 hr per week or more nurse-patient conflict events predicted more burnout. In addition, working40 hr per week, more nurse-patient conflict events and having the need of psychological support predicted higher secondary traumatic stress.

In the summers of 1998 and 1999, Chinese and Japanese scientists cooperatively conducted the first large-scale energy and water cycle experiment (WCRP/GEWEX/GAME/HUBEX: World Climate Research Program/Global Energy and Water Cycle Experiment/Asian Monsoon Experiment/Huaihe River Basin Energy and Water Cycle Experiment) in the Huaihe River basin, Anhui Province of China. The main objective of this field experiment (HUBEX) was to investigate the multiple-scale structure characteristics, life cycles, and genesis and development mechanisms of the Meiyu system in East Asia as well as the cause of related flooding disasters. It was a joint China–Japan cooperative meteorological and hydrological observation experiment. On the basis of intensive observations, scientists from the two countries conducted follow-up investigations through collating and compiling data and performing scientific analysis during the following five years. It can be concluded that the HUBEX project has yielded comprehensive and remarkable achievements. This paper introduces the major scientific results derived from this field experiment and the ensuing investigations, and reassesses their merits and shortages for the purpose of providing useful experience and proposing new research targets as well as prospects for the initiation of a new joint scientific Meiyu experiment in the middle and lower Yangtze River basin.

Dendritic growth is a major obstacle for the development of potassium metal anodes in K-ion batteries, which causes low coulombic efficiency, poor cycling stability and safety hazards. Herein, we report a 3D SnO2-coated conductive porous carbon nanofiber (PCNF) framework (PCNF@SnO2) as a host for K metal anodes. The SnO2 coating layer has a higher binding energy with K than with carbon and makes the PCNF framework highly potassiphilic, which regulates uniform K nucleation and deposition and suppresses dendritic growth. In addition, the interconnected 3D structure of the PCNF framework provides a large amount of void space and a robust structure to accommodate a large amount of K (87% @ 15 mg cm−2) and alleviate the stress/strain caused by volume expansion during plating/stripping. The PCNF@SnO2–K composite anode demonstrates an ultralong cycling stability of 1700 h at 1 mA cm−2, which is superior to those of other K metal anodes and even comparable with those of Li metal anodes reported in the literature. More importantly, enhanced performance is also achieved in a full cell using the composite K metal anode. Our results highlight a promising strategy for the development of stable and safe K metal anodes in KIBs.

Drug-resistance is a major problem preventing a cure in patients with multiple myeloma (MM). Previously, we demonstrated that activated-leukocyte-cell-adhesion-molecule (ALCAM) is a prognostic factor in MM and inhibits EGF/EGFR-initiated MM clonogenicity. In this study, we further showed that the ALCAM-EGF/EGFR axis regulated the MM side population (SP)-mediated drug-resistance. ALCAM-knockdown MM cells displayed an enhanced ratio of SP cells in the presence of bone marrow stromal cells (BMSCs) or with the supplement of recombinant EGF. SP MM cells were resistant to chemotherapeutics melphalan or bortezomib. Drug treatment stimulated SP-genesis. Mechanistically, EGFR, primed with EGF, activated the hedgehog pathway and promoted the SP ratio; meanwhile, ALCAM inhibited EGFR downstream pro-MM cell signaling. Further, SP MM cells exhibited an increased number of mitochondria compared to the main population. Interference of the mitochondria function strongly inhibited SP-genesis. Animal studies showed that combination therapy with both an anti-MM agent and EGFR inhibitor gefitinib achieved prolonged MM-bearing mice survival. Hence, our work identifies ALCAM as a novel negative regulator of MM drug-resistance, and EGFR inhibitors may be used to improve MM therapeutic efficacy.

IntroductionTo explore the diagnostic value of combining superb microvascular imaging (SMI), shear-wave elastography (SWE), and Breast Imaging Reporting and Data System (BI-RADS) to distinguish different molecular subtypes of invasive ductal carcinoma (IDC).Material and methodsA total of 239 surgically confirmed IDC masses in 201 patients underwent conventional ultrasound, SMI, and SWE examination, the information such as echo pattern, posterior features, margins, SMI pixels, and hardness of the masses was recorded. According to the St. Gallen standard, breast masses were classified as Luminal A, Luminal B, HER2 overexpression, and triple-negative subtype. We further explored the differences between different molecular subtypes of IDC.ResultsLuminal A subtype had the following characteristics: low histologic grade, posterior acoustic shadowing (p= 0.019), spiculated margins (pConclusionsOur study demonstrated the significant differences and trends among the IDC four subtypes by the combined application of SMI, SWE, and BI-RADS lexicon, which are of great significance for early diagnosis, selection of treatment methods, and evaluation of prognosis of IDC.

Olfactory dysfunction is an early pre-motor symptom of Parkinson’s disease (PD) but the neural mechanisms underlying this dysfunction remain largely unknown. Aggregation of α-synuclein is observed in the olfactory bulb (OB) during the early stages of PD, indicating a relationship between α-synuclein pathology and hyposmia. Here we investigate whether and how α-synuclein aggregates modulate neural activity in the OB at the single-cell and synaptic levels. We induced α-synuclein aggregation specifically in the OB via overexpression of double-mutant human α-synuclein by an adeno-associated viral (AAV) vector. We found that α-synuclein aggregation in the OB decreased the ability of mice to detect odors and to perceive attractive odors. The spontaneous activity and odor-evoked firing rates of single mitral/tufted cells (M/Ts) were increased by α-synuclein aggregates with the amplitude of odor-evoked high-gamma oscillations increased. Furthermore, the decreased activity in granule cells (GCs) and impaired inhibitory synaptic function were responsible for the observed hyperactivity of M/Ts induced by α-synuclein aggregates. These results provide direct evidences of the role of α-synuclein aggregates on PD-related olfactory dysfunction and reveal the neural circuit mechanisms by which olfaction is modulated by α-synuclein pathology.

Background Notch signaling mediates the committed induced differentiation of ear sensory cells and promotes the formation of a precise arrangement of mosaics between hair cells and supporting cells. Embryonic stem cells (ESCs) are pluripotent stem cells which have the potential to differentiate into cell lines through three germ layers. Therefore, it is necessary to study the effects of regulating Notch receptors and ligand expression on the in vitro differentiation equilibrium of hair cells and supporting cells from ESCs.Methods and Results The temporal ex-pression pattern of Notch ligands and receptors during in vitro hair cell-like cell differentia-tion from human embryonic stem cells (hESCs) was detected by quantitative reverse transcription-polymerase chain reaction (qRT-PCR). Subsequently, pAJ-U6-shRNA-CMV-Puro/GFP recombinant lentiviral vectors, encoding short hairpin RNAs, were used to silence JAG-1, JAG-2, and DLL-1, according to the temporal expression pattern of Notch ligands. Then the effect of each ligand on the in vitro differentiation of hair cells was examined by RT-PCR, immunofluorescence, and scanning electron microscopy (SEM).Conclusions Results showed that JAG-1 played an important role in regulating hESC differentiation to otic progenitors. The individual deletion of JAG-2 or DLL-1 had no significant effect on the differentiation of hair cell-like cells. Although the simultaneous inhibition of both DLL-1 and JAG-2 could increase the number of hair cell-like cells, it decreased the number of supporting cells.

Background Oral mucositis (OM) is a common adverse effect in hematological cancer patients who have received hematopoietic stem cell transplantation (HSCT). In this study, we compared mouthwash with nystatin, aloe vera or plain water in OM prevention.Design A retrospective chart review was conducted in participants. The incidence, severity and duration of OM, duration of oral pain, and the use of antalgesics were recorded.Setting/participants Hematological cancer patients who had received HSCT from January 2014 to December 2020 in West China Hospital were included in this study.Results A total of 150 patients were included in our retrospective analysis. The nystatin group had the highest incidence (100.0%) of OM, followed by the aloe vera group (44.8%), the plain water group had the lowest incidence of OM (20.0%), and the nystatin group had the highest severity and the longest duration of oral mucositis. The incidence, severity and duration of pain in the nystatin group were significantly higher than the aloe vera group and plain water group, and the use of analgesic drugs in the nystatin group was also significantly higher than the other two groups (PConclusions In our hospital setting, plain water mouthwash achieves great effect in OM prevention in hematological cancer patients who have received HSCT.

Background Papillary thyroid carcinoma (PTC) is the most common histological type of thyroid malignancy that tends to metastasize to cervical lymph nodes. In the present study, we aimed to investigate which clinicopathologic and ultrasound features of PTC are associated with clinical lymph node metastasis (LNM) and numbers of pathological LNM. Methods From January 2016 to December 2018, we identified a cohort of patients with PTC who underwent cervical ultrasonography and were diagnosed through operation and pathology. Clinical N1(cN1) and > 5 pathologic N1(pN1) were considered in the postoperative stratification to have an intermediate risk according to the 2015 ATA guidelines. Clinicopathological and ultrasound features in PTC patients were performed in accordance with the independent risk factors of cN1 and > 5pN1 respectively by using the univariate and multivariate analyses. Results We collected 748 PTC patients in the final inclusion criteria. There were 688 cN0 cases and 60 cN1 cases. From the analyses, primary tumor size > 2 cm, capsule contact, extrathyroidal extensions (ETE) and central LNM remained independent risk factors for cN1 in PTC patients. In the 748 PTC patients, 707 cases had ≤ 5 pN1, and 41 cases had > 5 pN1. Multifocality, primary tumor size > 2 cm, capsule contact and ETE are significant independent risk factors for > 5 pN1. Conclusions We concluded that multifocality, primary tumor size > 2 cm, capsule contact, ETE and central LNM were independent risk factors for the intermediate risk stratification in patients with PTC. Ultrasonography is a good technique for the preoperative lymph node staging of PTC and is helpful for detecting LNM.

The change of extreme precipitation with temperature has regional characteristics in the context of global warming. In this study, radiosonde data, co-located rain gauge (RG) observations, and Tropical Rainfall Measuring Mission (TRMM) precipitation radar (PR) products are used to explore the relationship between extreme precipitation intensity and near-surface temperature in Middle–East China (MEC) and the eastern Tibetan Plateau (TP) during 1998–2012. The results show that extreme precipitation intensity increases with increasing temperature at an approximate Clausius–Clapeyron (C–C) rate (i.e., water vapor increases by 7% as temperature increases by 1°C based on the C–C equation) in MEC and TP, but the rate of increase is larger in TP than in MEC. This is probably because TP (MEC) is featured with deep convective (stratiform) precipitation, which releases more (less) latent heat and strengthens the convection intensity on a shorter (longer) timescale. It is also found that when temperature is higher than 25°C (15°C) in MEC (TP), the extreme precipitation intensity decreases with rise of temperature, suggesting that the precipitation intensity does not always increase with warming. In this case, the limited atmospheric humidity and precipitable water could be the primary factors for the decrease in extreme precipitation intensity at higher temperatures.

Background Emerging studies demonstrate that long noncoding RNAs (lncRNAs) play crucial roles in hepatocarcinogenesis through various mechanisms. LncRNA CCAT2 was a newly discovered lncRNA and amplified in several cancers. However, the mechanisms involved in function of CCAT2 in hepatocellular carcinoma (HCC) remain to be explored. Methods CCAT2 expressions in HCC tissues and cell lines were measured by RT-qPCR. MTS assay, colony formation assay, wound-healing assay and transwell assay were used to explore the biological functions of CCAT2 on HCC cells proliferation and metastasis. Experiments in vivo were carried out to confirm these effects. The underlying mechanisms were analyzed by western blot and dual-luciferase reporter assay. Results In this study, we found that CCAT2 were significantly elevated in HCC tissues and cell lines, and it promoted HCC cells proliferation and metastasis both in vitro and in vivo. Additionally, we identified that NDRG1 was a downstream target of CCAT2. Meanwhile, depletion of CCAT2 inhibited cellular proliferation and metastasis behaviors induced by NDRG1- overexpression. Analysis of mechanism underlying these effects revealed that CCAT2 increased the expression of NDRG1 by enhancing its promoter activity. Furthermore, the active region between CCAT2 and NDRG1 promoter was confirmed by dual-luciferase reporter assay. Conclusions All these observations demonstrate that CCAT2 acts as an oncogene by up-regulating NDRG1, which may have the potential to be used as a promising prognostic biomarker and therapeutic target for HCC.

Herein, a novel and pragmatic electrochemiluminescence (ECL) biosensing method was developed for ultrasensitive and specific detection of Group B Streptococci (GBS) by combining self-enhanced luminol complex functionalized CuMn-CeO2 (CuMn-CeO2-PEI-luminol) with MNAzyme-mediated target-recycling amplification. First, the efficient self-enhanced PEI-luminol luminophore was prepared by combining PEI co-reactant with luminol in one molecular, which shortened electron transfer distance and enhanced ECL signal. And CuMn-CeO2 was applied to load a large number of PEI-luminol and strengthen luminous efficiency of luminol by the high catalytic activity toward H2O2 oxidation. Then, target-driven MNAzyme system was used to realize the circulation of GBS nucleic acid sequence, producing plentiful triggers to initiate the hybridization reaction on the surface of electrode. The developed enzyme-free ECL biosensor showed ultra-sensitivity for target DNA detection with detection limits of 68 aM (synthetic DNA) and 5 × 102 CFU mL−1 (genomic DNA extracted from GBS strain). More importantly, this biosensor was successfully applied for detection of genomic DNA of GBS extracted from clinical vaginal/anal swabs as low as 320 copies. Thus, this proposed strategy might be an pragmatic ECL platform for ultrasensitive and specific detection of GBS in clinical vaginal/anal swabs.

Long non-coding RNAs (lncRNAs) have emerged as critical regulators in a variety of diseases, including many tumors, such as hepatocellular carcinoma (HCC). However, the function and mechanisms responsible for these molecules in HCC are not thoroughly understood. In our previous study, we found that LINC00052 was acted as a tumor suppressor in HCC. In this study, we performed transcription microarray analysis to investigate the target gene of LINC00052, and found that knockdown of LINC00052 significantly increased the expression of SRY-related HMG-box gene 9 (SOX9), which plays an oncogenic role in HCC. Moreover, luciferase reporter assay revealed that LINC00052 promoted miR-101-3p expression by enhancing its promoter activity. In addition, online database analysis tools and luciferase assays showed that miR-101-3p could target SOX9. Quantitative real-time polymerase chain reaction (qRT-PCR) demonstrated that miR-101-3p was downregulated in HCC tissues and HCC cell lines. And we found a positive relationship between LINC00052 and miR-101-3p, and a negative relationship between miR-101-3p and SOX9 in HCC tissues. Besides, miR-101-3p was involved in LINC00052 inhibits HCC cells proliferation and metastasis. At the molecular level, LINC00052 downgulated SOX9 to inhibit HCC cells proliferation and metastasis by interacting with miR-101-3p. It might be a potential application for HCC therapy.
