Abstract: We present a variability, color and morphology based classifier, designed to identify transients, persistently variable, and non-variable sources, from the Zwicky Transient Facility (ZTF) Data Release 11 (DR11) light curves of extended and point sources. The main motivation to develop this model was to identify active galactic nuclei (AGN) at different redshift ranges to be observed by the 4MOST ChANGES project. Still, it serves as a more general time-domain astronomy study. The model uses nine colors computed from CatWISE and PS1, a morphology score from PS1, and 61 single-band variability features computed from the ZTF DR11 g and r light curves. We trained two versions of the model, one for each ZTF band. We used a hierarchical local classifier per parent node approach, where each node was composed of a balanced random forest model. We adopted a 17-class taxonomy, including non-variable stars and galaxies, three transient classes, five classes of stochastic variables, and seven classes of periodic variables. The macro averaged precision, recall and F1-score are 0.61, 0.75, and 0.62 for the g-band model, and 0.60, 0.74, and 0.61, for the r-band model. When grouping the four AGN classes into one single class, its precision, recall, and F1-score are 1.00, 0.95, and 0.97, respectively, for both the g and r bands. We applied the model to all the sources in the ZTF/4MOST overlapping sky, avoiding ZTF fields covering the Galactic bulge, including 86,576,577 light curves in the g-band and 140,409,824 in the r-band. Only 0.73\% of the g-band light curves and 2.62\% of the r-band light curves were classified as stochastic, periodic, or transient with high probability ($P_{init}\geq0.9$). We found that, in general, more reliable results are obtained when using the g-band model. Using the latter, we identified 384,242 AGN candidates, 287,156 of which have $P_{init}\geq0.9$., Comment: Accepted for publication in Astronomy & Astrophysics. Abstract shortened for arXiv. 2. Pulmonary hypertension in congenital diaphragmatic hernia: ANTENATAL PREDICTION AND IMPACT ON NEONATAL MORTALITY

Abstract: OBJECTIVE: To determine the prevalence of pulmonary hypertension(PAH) in left-sided congenital diaphragmatic hernia(CDH); how we could predict it; and how PAH contributed to the model for mortality prediction.; STUDY DESIGN: Retrospective analysis in three European centers. The primary outcome was the presence of PAH on postnatal day(d)1,7, and at discharge. Studied predictors of PAH were: observed/expected-lung/head-ratio(o/e LHR), liver-herniation, FETO, and gestational age(GA) at delivery. The combined effect of pre-and postnatal variables on mortality was modeled by Cox regression.; RESULTS: Of the 197 neonates, 56(28.4%) died. At d1, 67.5%(133/197) had PAH and 61.9% (101/163) by d7. Overall, 6.4% (9/141) had PAH at discharge.At d1, o/eLHR(OR 0.96) and FETO(OR 2.99) independently correlated to PAH(AUC:0.74). At d7, PAH significantly correlated only with the use of FETO (OR 3.9;AUC:0.65). None were significant for PAH at discharge.Combining the occurrence of PAH with antenatal biomarkers improved mortality prediction(p=0.02), in a model including o/eLHR(HR:0.94), FETO(HR:0.35), liver herniation(HR:16.78), and PAH(HR:15.95).; CONCLUSIONS: Antenatal prediction of PAH was only moderate. The postnatal occurrence of PAH further increases the risk of death. Whereas this may be used to counsel parents in the postnatal period, our study demonstrates there is a need to find more accurate antenatal predictors for PAH. This article is protected by copyright. Any further distribution of this work must maintain attribution to the author(s) and the title of the work, journal citation and DOI., We provide an updated assessment of the power of the Cherenkov Telescope Array (CTA) to search for thermally produced dark matter at the TeV scale, via the associated gamma-ray signal from pair-annihilating dark matter particles in the region around the Galactic centre. We find that CTA will open a new window of discovery potential, significantly extending the range of robustly testable models given a standard cuspy profile of the dark matter density distribution. Importantly, even for a cored profile, the projected sensitivity of CTA will be sufficient to probe various well-motivated models of thermally produced dark matter at the TeV scale. This is due to CTA's unprecedented sensitivity, angular and energy resolutions, and the planned observational strategy. The survey of the inner Galaxy will cover a much larger region than corresponding previous observational campaigns with imaging atmospheric Cherenkov telescopes. CTA will map with unprecedented precision the large-scale diffuse emission in high-energy gamma rays, constituting a background for dark matter searches for which we adopt state-of-the-art models based on current data. 4. Neuroprotection is improved by watertightness of fetal spina bifida repair in fetal lamb

Abstract: OBJECTIVES: the MOMS randomized trial has demonstrated that prenatal spina bifida aperta (SBA) repair via open approach is safe and effective for both mother and fetus, yet half of the infants have incomplete reversal of hindbrain herniation and no improvement in neuromotor function. One contributing factor may be the incompleteness of the neurosurgical repair causing persistent in utero leakage of cerebrospinal fluid (CSF) and exposure of the spinal cord to the amniotic fluid until birth. We aim to investigate the neurostructural and neurofunctional efficacy of the watertightness of prenatal SBA repair. METHODS: a superiority study was conducted in the validated SBA fetal lamb model and powered (n=7 per group). Outcomes of lambs undergoing watertight or non-watertight multilayer repairs through an open approach were compared to those in unrepaired SBA lambs at delivery (term=145 days). At ~75-day fetal lambs underwent standardized induction of lumbar SBA. At ~100 days, they were assigned to one of the three groups and subsequently to either watertight or non-watertight layered repair group based on an intraoperative watertightness test using subcutaneous fluoresceine injection. Finally, at 1-2 postnatal days, we assessed reversal of hindbrain herniation on Magnetic Resonance Imaging (MRI) as primary outcome. Secondary proxies of neuroprotection were CSF leakage at the repair site; hindlimb motor function based on joint movement score, locomotor grade and Motor Evoked Potentials (MEP); neuroprotection score encompassing live birth, hindbrain herniation reversal, absent CSF leakage and joint score =9/15; and brain and spinal cord histology and immunohistochemistry. As the watertightness test is not clinically usable, we developed a potential surrogate intraoperative quality score and assessed its relationship with improved outcomes. This four-point scoring system is based on visual assessment of the quality of the skin repair (suture inter-run distance =3mm, absence of tear and ischemia). RESULTS: Compared to unrepaired lambs, watertight repair was neuroprotective in 5/7 lambs (neuroprotection score of 4/4), as evidenced by a 100% reversal of hindbrain herniation on MRI; lower CSF leakage rate (14%); better hindlimb motor function evidenced by higher joint movement score, locomotor grade, and MEP area-under-the-curve and peak-to-peak amplitude; higher neuronal density in the hippocampus and corpus callosum; and higher reactive astrogliosis at the SBA lesion epicenter. Conversely, non-watertight SBA repair did not achieve the same level of neuroprotection (1/7) due to non-significant 86% hindbrain herniation reversal, no motor function improvement, high CSF leakage (43%), low brain neuron count in both regions and low spinal astroglial cell area at the epicenter. Like watertight layered repair, a high quality score (=2/3) was associated with improved outcomes yet watertightness test and quality score could not be used interchangeably due to results discrepancies. CONCLUSIONS: A watertight layered fetal SBA repair improves brain and spinal cord structure and function in the fetal lamb model. This translational research has important clinical implication and neurosurgical technique that achieves watertightness should be adopted in all fetal centers to improve neuroprotection. 5. Antenatal management of congenital diaphragmatic hernia: What's next ?

Abstract: Congenital diaphragmatic hernia can be diagnosed in the prenatal period and its severity can be measured by fetal imaging. There is now level I evidence that, in selected cases, Fetoscopic Endoluminal Tracheal Occlusion with a balloon increases survival to discharge from the neonatal unit as well as the risk for prematurity. Both effects are dependent on the time point of tracheal occlusion. Fetoscopic Endoluminal Tracheal Occlusion may also lead to iatrogenic death when the balloon cannot be timely retrieved. The implementation of the findings from our clinical studies, may also vary based on local conditions. These may be different in terms of available skill set, access to fetal therapy, as well as outcome based on local neonatal management. We encourage prior benchmarking of local outcomes with optimal postnatal management, based on large enough numbers and using identical criteria as in the recent trials. We propose to work further on prenatal prediction methods, and the improvement of fetal intervention. In this manuscript, we describe a research agenda from a fetal medicine perspective. This research should be in parallel with innovation in neonatal and pediatric (surgical) management of this condition.

6. Fetoscopic insufflation of heated-humidified carbon dioxide during simulated spina bifida repair is safe under controlled anesthesia in the fetal lamb

Abstract: Objective To assess the safety of Partial-Amniotic-Insufflation-of-heated-humidified-CO2 (hPACI) during fetoscopic spina bifida repair (fSB-repair). Method A simulated fSB-repair through an exteriorized uterus under hPACI was performed in 100-day fetal lambs (term = 145 days) under a laboratory anesthesia protocol (n = 5; group 1) which is known to induce maternal-fetal acidosis and hypercapnia. Since these may not occur clinically, we applied a clinical anesthesia protocol (n = 5; group 2), keeping maternal parameters within physiological conditions, that is, controlled maternal arterial carbon dioxide (CO2) pressure (pCO(2) = 30 mmHg), blood pressure (>= 67 mmHg), and temperature (37.1-39.8 degrees C). Our superiority study used fetal pH as the primary outcome. Results Compared to group 1, controlled anesthesia normalized fetal pH (7.23 +/- 0.02 vs. 7.36 +/- 0.02, p < 0.001), pCO(2) (70.0 +/- 9.1 vs. 43.0 +/- 1.0 mmHg, p = 0.011) and bicarbonate (27.8 +/- 1.1 vs. 24.0 +/- 0.9 mmol/L, p = 0.071) at baseline. Abstract: The astrophysical journal 924(2), 95 (2022). doi:10.3847/1538-4357/ac32bd, Flat-spectrum radio quasars (FSRQs) are the most luminous blazars at GeV energies but only rarely emit detectable fluxes of TeV gamma rays, typically during bright GeV flares. We explore the gamma-ray variability and spectral characteristics of three FSRQs that have been observed at GeV and TeV energies by Fermi-LAT and VERITAS, making use of almost 100 hr of VERITAS observations spread over 10 yr: 3C 279, PKS 1222+216, and Ton 599. We explain the GeV flux distributions of the sources in terms of a model derived from a stochastic differential equation describing fluctuations in the magnetic field in the accretion disk and estimate the timescales of magnetic flux accumulation and stochastic instabilities in their accretion disks. We identify distinct flares using a procedure based on Bayesian blocks and analyze their daily and subdaily variability and gamma-ray energy spectra. Using observations from VERITAS, as well as Fermi, Swift, and the Steward Observatory, we model the broadband spectral energy distributions of PKS 1222+216 and Ton 599 during very high energy (VHE)��detected flares in 2014 and 2017, respectively, strongly constraining the jet Doppler factors and gamma-ray emission region locations during these events. Abstract: PurposeThe COVID-19 pandemic has resulted in a global health crisis of unparalleled magnitude. The direct risk to the health of children is low. However, disease containment measures have society-wide impacts. This study explored the pandemic experiences of parents of children with oesophageal atresia/tracheo-oesophageal fistula in the UK.DesignAn online forum was conducted using a private group on Facebook, in collaboration with a patient support group. Thematic analysis was used to identify key themes.ResultsThe online forum ran between 7th November-18th December 2020 with 109 participants. Themes related to healthcare and non-healthcare impacts. Parents experienced changes and limitations to healthcare access, anxiety regarding health risks, “collateral” damage to well-being because of isolation and an impact on finances and employment. Parents described a transition from worry about direct health risks to concern about the impact of isolation on socialisation and development. A process of risk-benefit analysis led some to transition to a more “normal life”, while others continued to isolate. Benefits to their child’s health from isolation, positive experiences with remote healthcare and a gradual easing of anxiety were also identified.Implications and relevanceThis study highlights the wide-ranging impact of the COVID-19 pandemic on children and their families. Although focussed on oesophageal atresia/tracheo-oesophageal fistula, the emerging themes will be relevant to many children with complex, chronic health conditions. There are implications for healthcare delivery, including telehealth, during and after the pandemic period. Accurate and consistent messaging is required. Third sector organisations are ideally positioned support this.

9. A numerical study of long-term multiwavelength blazar variability

Abstract: Decade-long monitoring of blazars at optical and infrared (OIR) wavelengths with the Small and Moderate Aperture Research Telescope System (SMARTS) in Chile and in gamma-rays with the Fermi -Large Area Telescope (LAT) has enabled the systematic study of their multiwavelength long-term variability. In this work, we investigate, from a theoretical perspective, the long-term variability properties of blazar emission by introducing an observationally motivated time-dependence to four main parameters of the one-zone leptonic model: injection luminosity of relativistic electrons, strength of magnetic field, Doppler factor, and external photon field luminosity. For the first time, we use both the probability density function and the power spectral density of the 10-yr-long Fermi-LAT light curves to create variation patterns for the model parameters. Using as test beds two bright blazars from the SMARTS sample (PKS 2155-304 and 3C 273), we compute 10-yr-long OIR, X-ray, and. -ray model light curves for different varying parameters. We compare the findings of our theoretical investigation with multiwavelength observations using various measures of variability. While no single-varying parameter simulation can explain all multiwavelength variability properties, changes in the electron luminosity and external radiation field in PKS 2155-304 and 3C 273, respectively, can account for most of them. Our results motivate future time-dependent studies with coupling between two or more physical parameters to describe the multiwavelength long-term blazar variability.

10. Validation of a high-fidelity training model for fetoscopic spina bifida surgery

Abstract: Open fetal surgery for spina bifida (SB) is safe and effective yet invasive. The growing interest in fetoscopic SB repair (fSB-repair) prompts the need for appropriate training. We aimed to develop and validate a high-fidelity training model for fSB-repair. fSB-repair was simulated in the abdominal cavity and on the stomach of adult rabbits. Laparoscopic fetal surgeons served either as novices (n = 2) or experts (n = 3) based on their experience. Technical performance was evaluated using competency Cumulative Sum (CUSUM) analysis and the group splitting method. Main outcome measure for CUSUM competency was a composite binary outcome for surgical success, i.e. watertight repair, operation time = 180 min and Objective-Structured-Assessment-of-Technical-Skills (OSATS) score = 18/25. Construct validity was first confirmed since competency levels of novices and experts during their six first cases using both methods were significantly different. Criterion validity was also established as 33 consecutive procedures were needed for novices to reach competency using learning curve CUSUM, which is a number comparable to that of clinical fSB-repair. Finally, we surveyed expert fetal surgeons worldwide to assess face and content validity. Respondents (26/49; 53%) confirmed it with = 71% of scores for overall realism = 4/7 and usefulness = 3/5. We propose to use our high-fidelity model to determine and shorten the learning curve of laparoscopic fetal surgeons and retain operative skills.

Abstract: © 2019 authors. We present an experimental setup for monochromatic propagation-based x-ray phase-contrast imaging based on a conventional rotating-copper-anode source, capable of an integrated flux up to 108 photons/s at 8 keV. In our study, the system is characterized in terms of spatial coherence, resolution, contrast sensitivity, and stability. Its quantitativeness is demonstrated by comparing theoretical predictions with experimental data on simple wire phantoms both in planar and computerized-tomography-scan geometries. Application to two biological samples of medical interest shows the potential for bioimaging on the millimeter scale with spatial resolution of the order of 10 μm and contrast resolution below 1%. Abstract: Polarization is a critical parameter for understanding jet flows, as their radio to optical emission is produced by synchrotron radiation, which is naturally polarized, with the inferred magnetic field direction indicating the magnetic field direction in the emission region. Polarization has proven essential in characterizing the physics of FR I jets, where it has helped us map out their magnetic field and energetic structure and the relationship of this structure to the high-energy emission and particle acceleration. To date, high-quality HST polarimetry has been analyzed for just one FR II jet, that of PKS 1136-135. To rectify this, we have obtained new HST polarimetry observations of three key FR II jets - 3C 273, PKS 0637-752, and 1150+497. These new observations allow for the determination of the magnetic field structure and confirmation of which emission mechanisms are operating to create the observed optical to X-ray emission, and will allow us to greatly advance modeling efforts for these jets and nail down their kinetic power, a key parameter for understanding quasars and their cosmological effects.

Abstract: The possible association of the blazar TXS 0506+056 with a high-energy neutrino detected by IceCube holds the tantalizing potential to answer three astrophysical questions: 1. Where do high-energy neutrinos originate? 2. Where are cosmic rays produced and accelerated? 3. What radiation mechanisms produce the high-energy {\gamma}-rays in blazars? The MeV gamma-ray band holds the key to these questions, because it is an excellent proxy for photo-hadronic processes in blazar jets, which also produce neutrino counterparts. Variability in MeV gamma-rays sheds light on the physical conditions and mechanisms that take place in the particle acceleration sites in blazar jets. In addition, hadronic blazar models also predict a high level of polarization fraction in the MeV band, which can unambiguously distinguish the radiation mechanism. Future MeV missions with a large field of view, high sensitivity, and polarization capabilities will play a central role in multi-messenger astronomy, since pointed, high-resolution telescopes will follow neutrino alerts only when triggered by an all-sky instrument., Comment: White paper submitted to the Astro2020 Decadal Survey

Abstract: The constantly improving sensitivity of ground-based and space-borne observatories has made possible the detection of high-energy emission (X-rays and gamma-rays) from several thousands of extragalactic sources. Enormous progress has been made in measuring the continuum flux enabling us to perform imaging, spectral and timing studies. An important remaining challenge for high-energy astronomy is measuring polarization. The capability to measure polarization is being realized currently at X-ray energies (e.g. with IXPE), and sensitive gamma-ray telescopes capable of measuring polarization, such as AMEGO, AdEPT, e-ASTROGAM, etc., are being developed. These future gamma-ray telescopes will probe the radiation mechanisms and magnetic fields of relativistic jets from active galactic nuclei at spatial scales much smaller than the angular resolution achieved with continuum observations of the instrument. In this white paper, we discuss the scientific potentials of high-energy polarimetry, especially gamma-ray polarimetry, including the theoretical implications, and observational technology advances being made. Abstract: Contenuto della presente memoria è la descrizione della metodologia adottata per la fase di implementazione della rete di monitoraggio nell’area pilota di Milano ai fini della realizzazione delle mappe dinamiche previste dal progetto LIFE 13 ENV/IT/001254 Dynamap. L’area pilota coincide con il Municipio 9 del Comune di Milano, porzione dell’agglomerato urbano caratterizzata da infrastrutture stradali e numerose altre attività acusticamente rilevanti che rendono la procedura per la scelta dei punti di installazione dei dispositivi di monitoraggio estremamente complessa. Lo scopo del presente lavoro è quello di mostrare i criteri adottati nella procedura di scelta dei siti. Il criterio principale si basa sugli esiti dello studio svolto dall’Università Bicocca nell’ambito delle azioni preparatorie relativo all’analisi statistica dei flussi di traffico per la caratterizzazione del reticolo stradale dell’area pilota. Abstract: Long gap oesophageal atresia presents a surgical challenge as there is insufficient length of the oesophagus to restore continuity. Oesophageal replacement is generally achieved using a conduit, taken from the stomach, jejunum or colon. Preferences of approach vary between and within surgical centres. Specific to colonic interposition, the continued growth and dilation of the interposed segment may lead to redundancy. Revision surgery in these cases is challenging and has been sparsely described in adult patients. We present two patients who had colonic interposition for long gap oesophageal atresia in infancy and who then underwent successful revision surgery in their fifth decade.

Abstract: Several acquired or congenital pathological conditions can affect skeletal muscle leading to volumetric muscle loss (VML), i.e., an irreversible loss of muscle mass and function. Decellularized tissues are natural scaffolds derived from tissues or organs, in which the cellular and nuclear contents are eliminated, but the tridimensional (3D) structure and composition of the extracellular matrix (ECM) are preserved. Such scaffolds retain biological activity, are biocompatible and do not show immune rejection upon allogeneic or xenogeneic transplantation. An increase number of reports suggest that decellularized tissues/organs are promising candidates for clinical application in patients affected by VML. Here we explore the different strategies used to generate decellularized matrix and their therapeutic outcome when applied to treat VML conditions, both in patients and in animal models. The wide variety of VML models, source of tissue and methods of decellularization have led to discrepant results. Abstract: e-ASTROGAM (‘enhanced ASTROGAM’) is a breakthrough Observatory space mission, with a detector composed by a Silicon tracker, a calorimeter, and an anticoincidence system, dedicated to the study of the non-thermal Universe in the photon energy range from 0.3 MeV to 3 GeV – the lower energy limit can be pushed to energies as low as 150 keV for the tracker, and to 30 keV for calorimetric detection. The mission is based on an advanced space-proven detector technology, with unprecedented sensitivity, angular and energy resolution, combined with polarimetric capability. Thanks to its performance in the MeV–GeV domain, substantially improving its predecessors, e-ASTROGAM will open a new window on the non-thermal Universe, making pioneering observations of the most powerful Galactic and extragalactic sources, elucidating the nature of their relativistic outflows and their effects on the surroundings. With a line sensitivity in the MeV energy range one to two orders of magnitude better than previous generation instruments, e-ASTROGAM will determine the origin of key isotopes fundamental for the understanding of supernova explosion and the chemical evolution of our Galaxy. The mission will provide unique data of significant interest to a broad astronomical community, complementary to powerful observatories such as LIGO-Virgo-GEO600-KAGRA, SKA, ALMA, E-ELT, TMT, LSST, JWST, Athena, CTA, IceCube, KM3NeT, and LISA.

Abstract: Journal of high energy astrophysics 19, 1 - 106 (2018). doi:10.1016/j.jheap.2018.07.001, e-ASTROGAM (‘enhanced ASTROGAM’) is a breakthrough Observatory space mission, with a detector composed by a Silicon tracker, a calorimeter, and an anticoincidence system, dedicated to the study of the non-thermal Universe in the photon energy range from 0.3 MeV to 3 GeV – the lower energy limit can be pushed to energies as low as 150 keV for the tracker, and to 30 keV for calorimetric detection. The mission is based on an advanced space-proven detector technology, with unprecedented sensitivity, angular and energy resolution, combined with polarimetric capability. Thanks to its performance in the MeV–GeV domain, substantially improving its predecessors, e-ASTROGAM will open a new window on the non-thermal Universe, making pioneering observations of the most powerful Galactic and extragalactic sources, elucidating the nature of their relativistic outflows and their effects on the surroundings. With a line sensitivity in the MeV energy range one to two orders of magnitude better than previous generation instruments, e-ASTROGAM will determine the origin of key isotopes fundamental for the understanding of supernova explosion and the chemical evolution of our Galaxy. The mission will provide unique data of significant interest to a broad astronomical community, complementary to powerful observatories such as LIGO-Virgo-GEO600-KAGRA, SKA, ALMA, E-ELT, TMT, LSST, JWST, Athena, CTA, IceCube, KM3NeT, and LISA., Published by Elsevier, Amsterdam [u.a.]

Abstract: © 2017. The American Astronomical Society. All rights reserved. High-resolution X-ray spectroscopy with Hitomi was expected to resolve the origin of the faint unidentified E ≈ 3.5 keV emission line reported in several low-resolution studies of various massive systems, such as galaxies and clusters, including the Perseus cluster. We have analyzed the Hitomi first-light observation of the Perseus cluster. The emission line expected for Perseus based on the XMM-Newton signal from the large cluster sample under the dark matter decay scenario is too faint to be detectable in the Hitomi data. However, the previously reported 3.5 keV flux from Perseus was anomalously high compared to the sample-based prediction. We find no unidentified line at the reported high flux level. Taking into account the XMM measurement uncertainties for this region, the inconsistency with Hitomi is at a 99% significance for a broad dark matter line and at 99.7% for a narrow line from the gas. We do not find anomalously high fluxes of the nearby faint K line or the Ar satellite line that were proposed as explanations for the earlier 3.5 keV detections. We do find a hint of a broad excess near the energies of high-n transitions of S xvi (E ≃ 3.44 keV rest-frame) - a possible signature of charge exchange in the molecular nebula and another proposed explanation for the unidentified line. While its energy is consistent with XMM pn detections, it is unlikely to explain the MOS signal. A confirmation of this interesting feature has to wait for a more sensitive observation with a future calorimeter experiment.

Abstract: The development of human liver scaffolds retaining their 3-dimensional structure and extra-cellular matrix (ECM) composition is essential for the advancement of liver tissue engineering. We report the design and validation of a new methodology for the rapid and accurate production of human acellular liver tissue cubes (ALTCs) using normal liver tissue unsuitable for transplantation. The application of high shear stress is a key methodological determinant accelerating the process of tissue decellularization while maintaining ECM protein composition, 3D-architecture and physico-chemical properties of the native tissue. ALTCs were engineered with human parenchymal and non-parenchymal liver cell lines (HepG2 and LX2 cells, respectively), human umbilical vein endothelial cells (HUVEC), as well as primary human hepatocytes and hepatic stellate cells. Both parenchymal and non-parenchymal liver cells grown in ALTCs exhibited markedly different gene expression when compared to standard 2D cell cultures. Remarkably, HUVEC cells naturally migrated in the ECM scaffold and spontaneously repopulated the lining of decellularized vessels. The metabolic function and protein synthesis of engineered liver scaffolds with human primary hepatocytes reseeded under dynamic conditions were maintained. These results provide a solid basis for the establishment of effective protocols aimed at recreating human liver tissue in vitro.

Abstract: Oesophageal tissue engineering is a therapeutic alternative when oesophageal replacement is required. Decellularised scaffolds are ideal as they are derived from tissue-specific extracellular matrix and are non-immunogenic. However, appropriate preservation may significantly affect scaffold behaviour. Here we aim to prove that an effective method for short- and long-term preservation can be applied to tissue engineered products allowing their translation to clinical application. Rabbit oesophagi were decellularised using the detergent-enzymatic treatment (DET), a combination of deionised water, sodium deoxycholate and DNase-I. Samples were stored in phosphate-buffered saline solution at 4°C (4°C) or slow cooled in medium with 10% Me2SO at -1°C/min followed by storage in liquid nitrogen (SCM). Structural and functional analyses were performed prior to and after 2 and 4 weeks and 3 and 6 months of storage under each condition. Efficient decellularisation was achieved after 2 cycles of DET as determined with histology and DNA quantification, with preservation of the ECM. Only the SCM method, commonly used for cell storage, maintained the architecture and biomechanical properties of the scaffold up to 6 months. On the contrary, 4°C method was effective for short-term storage but led to a progressive distortion and degradation of the tissue architecture at the following time points. Efficient storage allows a timely use of decellularised oesophagi, essential for clinical translation. Here we describe that slow cooling with cryoprotectant solution in liquid nitrogen vapour leads to reliable long-term storage of decellularised oesophageal scaffolds for tissue engineering purposes.

Abstract: International audience; e-ASTROGAM (‘enhanced ASTROGAM’) is a breakthrough Observatory space mission, with a detector composed by a Silicon tracker, a calorimeter, and an anticoincidence system, dedicated to the study of the non-thermal Universe in the photon energy range from 0.3 MeV to 3 GeV – the lower energy limit can be pushed to energies as low as 150 keV, albeit with rapidly degrading angular resolution, for the tracker, and to 30 keV for calorimetric detection. The mission is based on an advanced space-proven detector technology, with unprecedented sensitivity, angular and energy resolution, combined with polarimetric capability. Thanks to its performance in the MeV-GeV domain, substantially improving its predecessors, e-ASTROGAM will open a new window on the non-thermal Universe, making pioneering observations of the most powerful Galactic and extragalactic sources, elucidating the nature of their relativistic outflows and their effects on the surroundings. With a line sensitivity in the MeV energy range one to two orders of magnitude better than previous generation instruments, e-ASTROGAM will determine the origin of key isotopes fundamental for the understanding of supernova explosion and the chemical evolution of our Galaxy. The mission will provide unique data of significant interest to a broad astronomical community, complementary to powerful observatories such as LIGO-Virgo-GEO600-KAGRA, SKA, ALMA, E-ELT, TMT, LSST, JWST, Athena, CTA, IceCube, KM3NeT, and the promise of eLISA.

Abstract: Clusters of galaxies are the most massive gravitationally bound objects in the Universe and are still forming. They are thus important probes of cosmological parameters and many astrophysical processes. However, knowledge of the dynamics of the pervasive hot gas, the mass of which is much larger than the combined mass of all the stars in the cluster, is lacking. Such knowledge would enable insights into the injection of mechanical energy by the central supermassive black hole and the use of hydrostatic equilibrium for determining cluster masses. X-rays from the core of the Perseus cluster are emitted by the 50-million-kelvin diffuse hot plasma filling its gravitational potential well. The active galactic nucleus of the central galaxy NGC 1275 is pumping jetted energy into the surrounding intracluster medium, creating buoyant bubbles filled with relativistic plasma. These bubbles probably induce motions in the intracluster medium and heat the inner gas, preventing runaway radiative cooling - a process known as active galactic nucleus feedback. Here we report X-ray observations of the core of the Perseus cluster, which reveal a remarkably quiescent atmosphere in which the gas has a line-of-sight velocity dispersion of 164 ± 10 kilometres per second in the region 30-60 kiloparsecs from the central nucleus. A gradient in the line-of-sight velocity of 150 ± 70 kilometres per second is found across the 60-kiloparsec image of the cluster core. Turbulent pressure support in the gas is four per cent of the thermodynamic pressure, with large-scale shear at most doubling this estimate. Abstract: Hepatic tissue engineering using decellularized scaffolds is a potential therapeutic alternative to conventional transplantation. However, scaffolds are usually obtained using decellularization protocols that destroy the extracellular matrix (ECM) and hamper clinical translation. We aim to develop a decellularization technique that reliably maintains hepatic microarchitecture and ECM components. Isolated rat livers were decellularized by detergent-enzymatic technique with (EDTA-DET) or without EDTA (DET). Histology, DNA quantification and proteomics confirmed decellularization with further DNA reduction with the addition of EDTA. Quantification, histology, immunostaining, and proteomics demonstrated preservation of extracellular matrix components in both scaffolds with a higher amount of collagen and glycosaminoglycans in the EDTA-DET scaffold. Scanning electron microscopy and X-ray phase contrast imaging showed microarchitecture preservation, with EDTA-DET scaffolds more tightly packed. DET scaffold seeding with a hepatocellular cell line demonstrated complete repopulation in 14 days, with cells proliferating at that time. Decellularization using DET preserves microarchitecture and extracellular matrix components whilst allowing for cell growth for up to 14 days. Addition of EDTA creates a denser, more compact matrix. Transplantation of the scaffolds and scaling up of the methodology are the next steps for successful hepatic tissue engineering.

Abstract: Contains fulltext : 168576.pdf (Publisher’s version ) (Open Access) In 2010, the congenital diaphragmatic hernia (CDH) EURO Consortium published a standardized neonatal treatment protocol. Five years later, the number of participating centers has been raised from 13 to 22. In this article the relevant literature is updated, and consensus has been reached between the members of the CDH EURO Consortium. Key updated recommendations are: (1) planned delivery after a gestational age of 39 weeks in a high-volume tertiary center; (2) neuromuscular blocking agents to be avoided during initial treatment in the delivery room; (3) adapt treatment to reach a preductal saturation of between 80 and 95% and postductal saturation >70%; (4) target PaCO2 to be between 50 and 70 mm Hg; (5) conventional mechanical ventilation to be the optimal initial ventilation strategy, and (6) intravenous sildenafil to be considered in CDH patients with severe pulmonary hypertension. This article represents the current opinion of all consortium members in Europe for the optimal neonatal treatment of CDH.

Abstract: The Very Energetic Radiation Imaging Telescope Array System (VERITAS) is a ground-based array located at the Fred Lawrence Whipple Observatory in southern Arizona and is one of the world's most sensitive gamma-ray instruments at energies of 85 GeV to $>$30 TeV. VERITAS has a wide scientific reach that includes the study of extragalactic and Galactic objects as well as the search for astrophysical signatures of dark matter and the measurement of cosmic rays. Abstract: We present multi-epoch optical spectroscopy of seven southern Fermi-monitored blazars from 2008 - 2013 using the Small and Medium Aperture Research Telescope System (SMARTS), with supplemental spectroscopy and polarization data from the Steward Observatory. We find that the emission lines are much less variable than the continuum; 4 of 7 blazars had no detectable emission line variability over the 5 years. This is consistent with photoionization primarily by an accretion disk, allowing us to use the lines as a probe of disk activity. Comparing optical emission line flux with Fermi $\gamma$-ray flux and optical polarized flux, we investigate whether relativistic jet variability is related to the accretion flow. In general, we see no such dependence, suggesting the jet variability is likely caused by internal processes like turbulence or shock acceleration rather than a variable accretion rate. However, three sources showed statistically significant emission line flares in close temporal proximity to very large Fermi $\gamma$-ray flares. While we do not have sufficient emission line data to quantitatively assess their correlation with the $\gamma$-ray flux, it appears that in some cases, the jet might provide additional photoionizing flux to the broad line region, which implies some gamma-rays are produced within the broad line region, at least for these large flares., Comment: 15 pages, 15 figures, Accepted to the Astrophysical Journal

Abstract: La molteplicità delle infrastrutture presenti e dei relativi gestori rende la realizzazione della mappa acustica strategica di Milano estremamente complessa. Lo scopo del presente lavoro è quello di mostrare i criteri adottati in fase di strutturazione del modello acustico a partire dalle diverse tipologie di dati acquisiti, preliminarmente raccolti ed elaborati all’interno di un Sistema Informativo Territoriale. In particolare vengono illustrate le soluzioni tecniche intraprese sia nell’organizzazione delle informazioni all’interno del SIT, con particolare riferimento all’elaborazione dei dati di popolazione e dei dati riguardanti i flussi di traffico, sia nell’utilizzo delle stesse nel software di simulazione (caratterizzazione delle diverse sorgenti e dell’ambiente di propagazione e scelta dei parametri di calcolo). Nello sviluppo del progetto ci si è basati sulle linee guida contenute nella “Good Practice Guide for Strategic Noise Mapping” (WG-AEN).

Abstract: The broad energy range spanned by ASTRO-H instruments, from ~0.3 to 600 keV, with its high spectral resolution calorimeter and sensitive hard X-ray imaging, offers unique opportunities to study black holes and their environments. The ability to measure polarization is particularly novel, with potential sources including blazars, Galactic pulsars and X-ray binaries. In this White Paper, we present an overview of the synergistic instrumental capabilities and the improvements over prior missions. We also show how ASTRO-H fits into the multi-wavelength landscape. We present in more detail examples and simulations of key science ASTRO-H can achieve in a typical 100 ksec observation when data from all four instruments are combined. Specifically, we consider observations of black-hole source (Cyg X-1 and GRS 1915+105), blazars (Mrk 421 and Mrk 501), a quasar (3C 273), radio galaxies (Centaurus A and 3C 120), and active galaxies with a strong starburst (Circinus and NGC 4945). 42. The Contrasting Nature of Gamma-Ray/Optical Variability in the Blazar PKS 0208-512 During Successive Outbursts

Abstract: The Yale/SMARTS optical-near-IR monitoring program has followed the variations in emission of the Fermi-LAT monitored blazars in the southern sky with closely spaced observations since 2008. We report the discovery of an optical-near-IR (OIR) outburst with no accompanying gamma-rays in the blazar PKS 0208-512, one of the targets of this program. While the source undergoes three outbursts of 1 mag or more at OIR wavelengths lasting for longer than 3 months during 2008-2011, only interval 1 and 3 have corresponding bright phases in GeV energies lasting longer than 1 month. The OIR outburst during interval 2 is comparable in brightness and temporal extent to the OIR flares during intervals 1 and 3 which do have gamma-ray counterparts. Gamma-ray and OIR variability are very well-correlated in most cases in the Fermi blazars and the lack of correlation in this case is anomalous. By analyzing the gamma-ray, OIR, and supporting multi-wavelength variability data in details, we speculate that the location of the outburst in the jet during interval 2 was closer to the black hole where the jet is more compact and the magnetic field strength is higher, and the bulk Lorentz factor of the material in the jet is smaller. Abstract: We report on the discovery of high-energy (HE; E > 0.1 GeV) and very high energy (VHE; E > 100 GeV) gamma-ray emission from the high-frequency-peaked BL Lac object RBS 0413. VERITAS, a ground-based gamma-ray observatory, detected VHE gamma rays from RBS 0413 with a statistical significance of 5.5 standard deviations and a gamma-ray flux of (1.5 ± 0.6stat ± 0.7syst) x 10^-8 photons m^-2 s^-1 (~1% of the Crab Nebula flux) above 250 GeV. The observed spectrum can be described by a power law with a photon index of 3.18 ± 0.68stat ± 0.30syst. Contemporaneous observations with the Large Area Telescope (LAT) on the Fermi Gamma-ray Space Telescope detected HE gamma rays from RBS 0413 with a statistical significance of more than 9sigmas, a power-law photon index of 1.57 ± 0.12stat +0.11 -0.12sys, and a gamma-ray flux between 300 MeV and 300 GeV of (1.64 ± 0.43stat +0.31 -0.22sys) x 10^-5 photons m^-2 s^-1. We present the results from Fermi-LAT and VERITAS, including a spectral energy distribution modeling of the gamma-ray, quasi simultaneous X-ray (Swift-XRT), ultraviolet (Swift-UVOT), and R-band optical (MDM) data. We find that, if conditions close to equipartition are required, both the combined synchrotron self-Compton/external-Compton and the lepto-hadronic models are preferred over a pure synchrotron self-Compton model. Abstract: Pediatric patients with SCID constitute medical emergencies. In the absence of an HLA-identical hematopoietic stem cell (HSC) donor, mismatched related-donor transplantation (MMRDT) or unrelated-donor umbilical cord blood transplantation (UCBT) are valuable treatment options. To help transplantation centers choose the best treatment option, we retrospectively compared outcomes after 175 MMRDTs and 74 UCBTs in patients with SCID or Omenn syndrome. Median follow-up time was 83 months and 58 months for UCBT and MMRDT, respectively. Most UCB recipients received a myeloablative conditioning regimen; most MMRDT recipients did not. UCB recipients presented a higher frequency of complete donor chimerism (P = .04) and faster total lymphocyte count recovery (P = .04) without any statistically significance with the preparative regimen they received. The MMRDT and UCBT groups did not differ in terms of T-cell engraftment, CD4+ and CD3+ cell recoveries, while Ig replacement therapy was discontinued sooner after UCBT (adjusted P = .02). There was a trend toward a greater incidence of grades II-IV acute GVHD (P = .06) and more chronic GVHD (P = .03) after UCBT. The estimated 5-year overall survival rates were 62% ± 4% after MMRDT and 57% ± 6% after UCBT. For children with SCID and no HLA-identical sibling donor, both UCBT and MMRDT represent available HSC sources for transplantation with quite similar outcomes.

Abstract: Origin of extragalactic gamma-ray background (EGRB) is still a matter of debate. EGRB can either have truly diffuse or unresolved discrete point sources origin. Majority of the Fermi and EGRET detected identified sources are blazar. So, they are expected to be a significant contributors to the EGRB. In order to estimate their contribution to the EGRB one needs to construct their luminosity functions. Here, we examine and construct the evolution and luminosity function of blazars using first LAT AGN catalog. We consider both pure luminosity and pure density evolution models for flat spectrum radio quasars (FSRQs). Abstract: The synergy between the Fermi-LAT and ground-based Cherenkov telescope arrays gives us the opportunity for the first time to characterize the high energy emission (100 MeV - 10 TeV) from more than 30 blazars. In this study we performed a Fermi-LAT spectral analysis for all TeV-detected blazars and combined it with archival TeV spectra. Our results for low synchrotron-peaked BL Lacs (LBL) show hints of absorption features in the GeV band that could be interpreted as internal opacity at the source. 48. Regenerative medicine as applied to solid organ transplantation: current status and future challenges

Abstract: In the last two decades, regenerative medicine has shown the potential for "bench-to-bedside" translational research in specific clinical settings. Progress made in cell and stem cell biology, material sciences and tissue engineering enabled researchers to develop cutting-edge technology which has lead to the creation of nonmodular tissue constructs such as skin, bladders, vessels and upper airways. In all cases, autologous cells were seeded on either artificial or natural supporting scaffolds. However, such constructs were implanted without the reconstruction of the vascular supply, and the nutrients and oxygen were supplied by diffusion from adjacent tissues. Engineering of modular organs (namely, organs organized in functioning units referred to as modules and requiring the reconstruction of the vascular supply) is more complex and challenging. Models of functioning hearts and livers have been engineered using "natural tissue" scaffolds and efforts are underway to produce kidneys, pancreata and small intestine. Creation of custom-made bioengineered organs, where the cellular component is exquisitely autologous and have an internal vascular network, will theoretically overcome the two major hurdles in transplantation, namely the shortage of organs and the toxicity deriving from lifelong immunosuppression. This review describes recent advances in the engineering of several key tissues and organs.

49. Similarity of the Optical-IR and Gamma-Ray Time Variability of Fermi Blazars

Abstract: We present the time variability properties of a sample of six blazars, AO 0235+164, 3C 273, 3C 279, PKS 1510-089, PKS 2155-304, and 3C 454.3, at optical-IR as well as gamma-ray energies. These observations were carried out as a part of the Yale/SMARTS program during 2008-2010 that has followed the variations in emission of the bright Fermi-LAT-monitored blazars in the southern sky with closely-spaced observations at BVRJK bands. We find the optical/IR time variability properties of these blazars to be remarkably similar to those at the gamma-ray energies. The power spectral density (PSD) functions of the R-band variability of all six blazars are fit well by simple power-law functions with negative slope such that there is higher amplitude variability on longer timescales. No clear break is identified in the PSD of any of the sources. The average slope of the PSD of R-band variability of these blazars is similar to what was found by the Fermi team for the gamma-ray variability of a larger sample of bright blazars. This is consistent with leptonic models where the optical-IR and gamma-ray emission is generated by the same population of electrons through synchrotron and inverse-Compton processes, respectively. The prominent flares present in the optical-IR as well as the gamma-ray light curves of these blazars are predominantly symmetric, i.e., have similar rise and decay timescales, indicating that the long-term variability is dominated by the crossing time of radiation or a disturbance through the emission region rather than by the acceleration or energy-loss timescales of the radiating electrons. In the blazar 3C 454.3, which has the highest-quality light curves, the location of a large gamma-ray outburst during 2009 December is consistent with being in the jet at ~18 pc from the central engine. Abstract: Ground-based gamma-ray astronomy has had a major breakthrough with the impressive results obtained using systems of imaging atmospheric Cherenkov telescopes. Ground-based gamma-ray astronomy has a huge potential in astrophysics, particle physics and cosmology. CTA is an international initiative to build the next generation instrument, with a factor of 5-10 improvement in sensitivity in the 100 GeV-10 TeV range and the extension to energies well below 100 GeV and above 100 TeV. CTA will consist of two arrays (one in the north, one in the south) for full sky coverage and will be operated as open observatory. The design of CTA is based on currently available technology. This document reports on the status and presents the major design concepts of CTA. © 2011 The Author(s).
