Lipocaline (LCNe) sind eine Familie von Proteinen, die eine Vielzahl an Funktionen erfüllen. Zahlreiche Allergene gehören zu dieser Proteinfamilie. Obwohl LCNe eine geringe Sequenzhomologie aufweisen, ist ihre Tertiärstruktur hoch konserviert. Ein antiparalleles β-barrel formt eine intramolekulare Tasche, welche das Binden von hydrophoben Molekülen wie Fettsäuren, Vitamin-Metaboliten und Steroidhormonen ermöglicht. Abhängig von der Beladung dieser Tasche können LCNe die Th2 Immunantwort modulieren. Ein Mitglied der LCN-Proteinfamilie ist das Kuhmilchallergen Bos d 5. Auch das Birkenpollenallergen Bet v 1, welches zur Familie der PR-10 Proteine gehört, wurde kürzlich aufgrund struktureller Ähnlichkeiten und seiner Fähigkeit Liganden zu binden als LCN-ähnliches Protein identifiziert. Wir untersuchten, ob Bos d 5 und Bet v 1, wenn sie den aktiven Vitamin A Metaboliten Retinolsäure (RA) gebunden haben, eine andere Immunantwort auslösen als die unbeladenen apo-Allergene. Mononukleäre Zellen des peripheren Blutes (PBMCs) wurden mit der leeren (apo-Protein) und der RA-beladenen Form (holo-Protein) der entsprechenden Proteine stimuliert und die Expression von Aktivierungsmarkern, Zytokinproduktion und Zellteilungsrate wurde gemessen. Die Stabilität der Protein-Ligand-Komplexverbindung wurde mittels eines endolysosomalen Verdauungs-Assays analysiert. Holo-Bos d 5 (+RA) unterdrückte die Expression des Aktivierungsmarkers HLA-DR auf CD4+ T Zellen signifikant und verringerte die IFN-γ und IL-10 Sekretion von stimulierten PBMCs. Eine Fluoreszenzfärbung mit CFSE zeigte eine signifikant geringere Zellteilungsrate von CD4+ und CD8+ T-Zellen nach Stimulation mit holo-Bos d 5. Das Binden von RA an das hyperallergene Bet v 1a führte zu einer signifkanten Reduktion der IFN-γ, IL-10 und IL-13 Produktio in PBMCs von Birkenpollen-Allergikern, während apo-Bet v 1a eine hohe Produktion von Th1/Th2 Zytokinen induzierte. Vorläufige Daten lassen darauf schließen, dass der immunsuppressive Effekt von holo–Bet v 1a im Zusammenhang mit einer höheren enzymatischen Stabilität gegenüber dem endolysosomalen Enzym Cathepsin S stehen könnte. Aus diesen Ergebnissen schließen wir, dass durch die Bindung von RA an Bos d 5 und Bet v 1 die Immunogenität dieser Allergene reduziert wird, wodurch zwischen Allergie und Toleranz entschieden werden könnte. Lipocalins (LCNs) are a functionally diverse family of proteins. Numerous allergens belong to this protein family. LCNs share a low sequence homology but they have a highly conserved core structure featuring an antiparallel ß-barrel that defines an intramolecular pocket. This pocket enables the binding of hydrophobic molecules such as fatty acids, vitamin metabolites and steroid hormones. Depending on their ligand load LCNs are able to modulate Th2 responses. One member of the LCN-protein family is the major cow’s milk allergen Bos d 5. The major birch pollen allergen Bet v 1, from the PR-10 family of proteins, has been recently identified as a LCN-like protein due to structural similarities and its ability to bind small ligands. We now investigated whether Bos d 5 and Bet v 1, when loaded with the active vitamin A metabolite retinoic acid (RA), would elicit different immune responses than the empty allergens. For this purpose we stimulated human PBMCs with the empty (apo-protein) or RA-loaded form (holo-protein) of the respective allergens and measured expression of activation markers, cytokine production and proliferative response. Stability of the allergen-ligand complex was analysed using an endolysosomal digestion assay. Holo-Bos d 5 (+ RA) significantly suppressed the expression of activation marker HLA-DR on CD4+ T cells and decreased IFN-γ and IL-10 secretion from stimulated human PBMCs. Fluorescence staining with CFSE revealed a significantly lower proliferative response of CD4+ and CD8+ T cells in holoBos d 5 treated groups. Loading of RA to hyperallergenic Bet v 1a led to a significant reduction of IFN-γ, IL-10 and IL-13 levels in PBMCs of birch pollen (BP)-allergic donors, while empty apo-Bet v 1a induced high Th1/Th2 cytokine production. Preliminary data indicate that the immunosuppressive effect of holo-Bet v 1a could be associated with a higher enzymatic stability to the endolysosomal enzyme cathepsin S. Hintergrund: Das Immunsystem besteht aus einem hochkomplexen Netzwerk, das die Reaktion auf endogene Stoffe und Fremdstoffe kontinuierlich reguliert. Eine Überempfindlichkeit des Immunsystems kann einerseits zu allergischen Krankheiten führen, andererseits fördert ein zu tolerantes Immunsystem die Krebsentwicklung. In dieser Arbeit untersuchten wir potentielle hyperreaktive Mechanismen verursacht durch das Milchallergen Bos d 5 auf der einen Seite und andererseits Acrolein, das immunosuprimierend wirkt. Methoden: Während in in vitro-Versuchen aus menschlichem Blutzellen M1 und M2 Makrophagen generiert und mit Bos d 5 stimuliert wurden, wurden die Auswirkungen von Acrolein direkt auf menschliche Blutzellen untersucht. Die Markophagen wurden anhand ihrer Oberflächenrezeptoren mittels Durchflusszytometrie phenotypisiert und die sekretierten IL-6 und IL-10 Zytokine mittels ELISA bestimmt. Die Wirkung von Acrolein an Blutzellen wurde mittels FACS-Analyse von immunesuppressiven CD4CD25Foxp3 positive Zellen bestimmt. Desweiteren wurde die Aktivierung des Aryl-Hyrocarbonrezeptors durch Acrolein unter Verwendung der rekombinaten Reporterzelllinie AZ-AhR untersucht. Ergebnisse: Das Milchallergen Bos d 5 verursachte eine signifikant erhöhte CD14 sowie eine signifikant verminderte CD163 Expression in M2, aber nicht M1 polarisierten Makrophagen. Dies wurden durch erhöhte Sekretion von IL-6 und IL-10 begleitet. Acrolein begünstigte die Entstehung von suppressiven T-Zellen in vitro, die durch die Zugabe von den AHR-Antagonist Resveratrol teilweise unterbunden werden konnte. Auch unter Verwendung der Reporterzelllinie AZ-AHR konnte Acrolein konzentrationsabhängig den Aryl-Hydrocarbon-Rezeptor aktivieren und dessen Aktivierung durch Zugabe von Resveratrol verhindert werden. Zusammenfassung: Das Milchallergen Bos d 5 war imstande M2 Makrophagen zu aktivieren, die die Sekretion von IL-6 und IL-10 zur Folge hatte. Im Gegensatz dazu aktivierte Acrolein den Aryl-Hydrocarbon-Rezeptor und förderte damit die Bildung von immunsuppressiven Zellen. Background: The immune system consists of a highly intricate network that regulates the continuous response to environmental and endogenous compounds. Hyperreactivity may lead to allergic diseases, whereas a hyporeactive immune system promote cancer development. Here we investigated potential hyperreactive mechanisms mediated by milk allergen Bos d 5 on the one hand and on the other acrolein that mediates hyporeactivity by immunosuppression. Methods: In in vitro experiments, Bos d 5 was incubated to human blood-derived macrophages polarized towards M1 or M2 for 16 h, before assessing the cytokine content of the supernatant and analyzing surface receptors of macrophages by flow cytometry. Similarly, PBMCs were incubated with acrolein and controls and assessed for CD4+CD25+Foxp3+ expression by FACS. Furthermore, Aryl hydrocarbon receptor (AhR) activation by acrolein was assessed using the reporter cell line AZ-AhR. Results: Bos d 5 incubation to macrophages experiments resulted in a significant upregulation of CD14 and downregulation of CD163 in M2 macrophages, but not in M1 macrophages after 16h. This was accompanied by a significant increase of IL-6 and IL-10. Acrolein promoted the generation of CD4+CD25+Foxp3+ cells, which was partly reversed by the addition of the AhR-antagonist resveratrol. Moreover, using the gene reporter cell line AZ-AhR, acrolein concentration-dependently activated AhR, which was blocked by addition of the AhR antagonist resveratrol. Conclusion: The major milk allergen Bos d 5 was able to activate M2 macrophages leading to increased secretion of IL-6 and IL-10.
