One of the major outcomes of the SCOOP project dealing with trichothecenes was that for a more accurate assessment of the exposure of the European population, exposure data of specific population groups (like children and infants) are needed. In the present study analytical measurements in duplicate diets of young children are done in order to investigate the real exposure to trichothecenes. Based on the individual results, exposure calculations were made. For deoxynivalenol the TDI (1 g/kg bw) was exceeded by 9 children. For the combined intake of T-2 and HT-2 toxin the tTDI (0.06 g/kg bw) was exceeded by 9 children. One child exceeded the (t)TDI for both DON and combined HT-2 and T-2. The exposure to the other trichothecenes was very low (nivalenol) or almost absent. As the TDI concept is based on a life-time exposure, an incidental exposure above the TDI does not necessarily indicate that there is an appreciable risk for adverse health effects. However as one of the effects of deoxynivalenol is growth inhibition in experimental animals and young children are in a fast growing phase, exceeding of the TDI should be avoided.

The exposure to the other trichothecenes was very low (nivalenol) or almost absent. As the TDI concept is based on a life-time exposure, an incidental exposure above the TDI does not necessarily indicate that there is an appreciable risk for adverse health effects. However as one of the effects of deoxynivalenol is growth inhibition in experimental animals and young children are in a fast growing phase, exceeding of the TDI should be avoided.

RIVM rapport:Increased nitrate intake might affect the thyroid gland function in humans, as has been observed in animals. The reason is that the nitrate-ion (NO3-) inhibits the iodide (I-) transport into the thyroid gland because it shares the same transport mechanism. This inhibition could lead to a decrease in thyroid hormone (T4, T3) secretion, followed by an increase in the thyroid-stimulating hormone (TSH). In the end thyroid gland enlargement (goitre) could occur. Since there is only weak epidemiological evidence that thyroid gland enlargement may occur in humans, our aim was to investigate the effect of nitrate on the human thyroid gland function by means of a four-week nitrate exposure study. Once a day for 28 days ten volunteers received an oral solution of 15 mg sodium nitrate per kg body weight (three times the allowed daily intake, ADI) in 200 ml distilled water (nitrate group), and ten volunteers received 200 ml distilled water (control group). Both groups followed an iodine-restricted and low-nitrate diet; this was checked by measuring urinary iodide and plasma nitrate concentration. Before and after the 28-day exposure period the percentage (%) radioiodine (131I) uptake (RAIU) was measured 5 hours and 24 hours after 131I-capsule intake to investigate the competition of nitrate in the iodide transport. Before (nitrate) exposure and 2, 3 and 4 weeks after the start of the exposure period blood samples were taken to measure the hormones T4, T3, rT3, TSH and IGF I to investigate the thyroid gland function. Nitrate was found to have no effect on the hormone concentrations during the four-week nitrate exposure of three times the ADI of nitrate. The bioavailability study was preceded by a single , intravenous, ascending dose study to investigate the maximum nitrite dose which can safely be administered in vivo; i.e. inducing approximately 10 to 15% metHb. This study demonstrates that a sodium nitrite dose of 0.12 mmol NaNO2 per mmol Hb (between 290 and 370 mg) and inducing approximately 10.8% methemoglobin in the blood, is the maximum tolerated dose for adult volunteers.

Calculating the oral bioavailability of nitrite from the plasma nitrite concentration is hampered and requires a substantial nitrite dose to be administered to healthy volunteers. Voor het bepalen van NDELA in cosmetica is de GC-TEA methode ontwikkeld en gevalideerd. De terugwinning voor NDELA is gemiddeld 99% (N=4) en de bepaalbaarheidsgrens is 5 mug NDELA / kg cosmetica. De GC-TEA methode is gebruikt om in 48 cosmetische producten, waaronder shampoos, cremes, melk , conditioners en schuimen het NDELA gehalte te bepalen. This study demonstrates that, after an overnight fast, 90 to 95% of orally ingested sodium nitrite is absorbed from the gastro-intestinal tract. Nitrite rapidly disappeared from the plasma with an elimination half-life of approximately 30 minutes.

RIVM rapport:Het nitraat-gehalte in drinkwater en voedingsmiddelen dreigt steeds verder toe te nemen in West-Europese landen. De toxiciteit van nitraat is laag. In het eerste deel van het maag-darmkanaal wordt echter een deel van het nitraat omgezet in nitriet, dat meer toxisch is. Het nitriet dat gevormd wordt kan alleen aanleiding geven tot systemische toxiciteit als het wordt geabsorbeerd uit het maag-darmkanaal. De orale biobeschikbaarheid van natriumnitriet werd daarom onderzocht in gezonde proefpersonen. De studie had een open, 3-weg gekruisde en gerandomiseerde proefopzet. Aan 9 proefpersonen werden 2 enkelvoudige, orale doseringen en een enkelvoudige, intraveneuze dosering van natriumnitriet toegediend. Gedurende 24 uur na doseren werden, op regelmatige tijdstippen, plasma monsters verzameld voor nitraat/nitriet analyse. Deze studie toont aan dat, onder nuchtere omstandigheden 90-95% van het nitriet wordt geabsorbeerd uit het maag-darmkanaal. Nitriet wordt snel uit het plasma geklaard met een terminale halfwaarde-tijd van ongeveer 30 minuten., Nitrate levels in food products and drinking water tend to increase continuously in Western Countries. The toxicity of nitrate is low. However, in the proximal part of the intestinal tract, part of the nitrate is converted to nitrite, a more toxic compound. This nitrite can only induce systemic toxicity in the case it is absorbed from the gastro-intestinal tract. The oral bioavailability of sodium nitrite was, therefore, determined in healthy adult volunteers. The study was designed as a single dose, open, randomized, three way cross-over study in which 9 volunteers received 2 single oral doses of sodium nitrite and one intravenous sodium nitrite dose, with a wash out period of 7 days between the treatments. At timed intervals, plasma samples were collected until 24 hours post-dose and analysed for the nitrate and nitrite content. This study demonstrates that, after an overnight fast, 90 to 95% of orally ingested sodium nitrite is absorbed from the gastro-intestinal tract. Nitrite rapidly disappeared from the plasma with an elimination half-life of approximately 30 minutes.

RIVM rapport:In 1995 is op verzoek van de Inspectie Gezondheids Bescherming de N-nitrosamine problematiek in cosmetische producten in kaart gebracht. Voor het bepalen van NDELA in cosmetica is de GC-TEA methode ontwikkeld en gevalideerd. De terugwinning voor NDELA is gemiddeld 99% (N=4) en de bepaalbaarheidsgrens is 5 mug NDELA / kg cosmetica. De GC-TEA methode is gebruikt om in 48 cosmetische producten, waaronder shampoos, cremes, melk , conditioners en schuimen het NDELA gehalte te bepalen. Alle bepalingen werden in tweevoud uitgevoerd en voor elke 10 cosmetica monsters is een terugwinnings- en blanco-experiment uitgevoerd. De resultaten van deze kwaliteitsexperimenten voldeden aan de kwaliteitskenmerken van de ontwikkelde methode.Van de 48 onderzochte monsters cosmetica bevatten vier monsters meetbare hoeveelheden NDELA., In response to the 1995 review of the N-nitrosamine problem in cosmetics at the request of the Inspectorate of Health Protection (IGB), a GC-TEA method for determining NDELA in cosmetics has been developed and validated.
