1. Next-Generation Proteomics of Brain Extracellular Vesicles in Schizophrenia Provide New Clues on the Altered Molecular Connectome

Abstract: Extracellular vesicles (EVs) are tiny membranous structures that mediate intercellular communication. The role(s) of these vesicles have been widely investigated in the context of neurological diseases; however, their potential implications in the neuropathology subjacent to human psychiatric disorders remain mostly unknown. Here, by using next-generation discovery-driven proteomics, we investigate the potential role(s) of brain EVs (bEVs) in schizophrenia (SZ) by analyzing these vesicles from the three post-mortem anatomical brain regions: the prefrontal cortex (PFC), hippocampus (HC), and caudate (CAU). The results obtained indicate that bEVs from SZ-affected brains contain region-specific proteins that are associated with abnormal GABAergic and glutamatergic transmission. Similarly, these vesicles from the analyzed regions were implicated in synaptic decay, abnormal brain immunity, neuron structural imbalances, and impaired cell homeostasis. Our findings also provide evidence, for the first time, that networks of molecular exchange (involving the PFC, HC, and CAU) are potentially active and mediated by EVs in non-diseased brains. Additionally, these bEV-mediated networks seem to have become partially reversed and largely disrupted in the brains of subjects affected by SZ. Taken as a whole, these results open the door to the uncovering of new biological markers and therapeutic targets, based on the compositions of bEVs, for the benefit of patients affected by SZ and related psychotic disorders.

Abstract: This chapter provides an overview of the major social scientific strands of theorizing about consumption. We distinguish (1) the individualist tradition prevalent in social-psychological and economic approaches to consumption, but also in the more analytical realms of sociology, (2) the socio-cultural approach to consumption, strongly influenced by philosophical and sociological accounts of modern, contemporary society, and finally (3) practice theories, which have become the most widespread theoretical approach in the sociological analysis of consumption. For each theoretical perspective, we discuss its treatment of three central aspects of consumption (acquisition, use, and disposal) and its performance as a scientific theory (e.g., function, clarity, testability). It is clear from our discussion that individualist theories adhere strongest to a classical idea of explanation and empirical testability, whereas the other two strands of theorizing often lack in analytical rigor and empirical generalizability. Socio-cultural theories provide formidable accounts of how needs have changed historically, which is a precondition for explaining acquisition. Individualist theories have studied in detail the act of purchasing (acquisition). Theories of practice are particularly well suited to theorize the actual use of products, that is, the particular ways in which goods are appropriated. However, individualist theories neglect the formation of needs and the use of products. Socio-cultural theories undertheorize the actual act of purchasing. Practice theories have largely neglected the disposal and to a certain degree also the acquisition of consumer goods. Therefore, for each theory exists uncharted territory to be tackled in the future.

3. A historical note on the complexity of scheduling problems

Abstract: In 1972 E.M. Livshits and V.I. Rublinetsky published a paper in Russian, in which they presented linear-time reductions of the partition problem to a number of scheduling problems. Unaware of complexity theory, they argued that, since partition is not known to have a simple algorithm, one cannot expect to find simple algorithms for these scheduling problems either. Their work did not go completely unnoticed, but it received little recognition. We describe the approach and review the results.

Abstract: Aims: Vegetation‐plot records provide information on the presence and cover or abundance of plants co‐occurring in the same community. Vegetation‐plot data are spread across research groups, environmental agencies and biodiversity research centers and, thus, are rarely accessible at continental or global scales. Here we present the sPlot database, which collates vegetation plots worldwide to allow for the exploration of global patterns in taxonomic, functional and phylogenetic diversity at the plant community level. Results: sPlot version 2.1 contains records from 1,121,244 vegetation plots, which comprise 23,586,216 records of plant species and their relative cover or abundance in plots collected worldwide between 1885 and 2015. We complemented the information for each plot by retrieving climate and soil conditions and the biogeographic context (e.g., biomes) from external sources, and by calculating community‐weighted means and variances of traits using gap‐filled data from the global plant trait database TRY. Moreover, we created a phylogenetic tree for 50,167 out of the 54,519 species identified in the plots. We present the first maps of global patterns of community richness and community‐weighted means of key traits. Conclusions: The availability of vegetation plot data in sPlot offers new avenues for vegetation analysis at the global scale.

Abstract: Aims: Understanding fine-grain diversity patterns across large spatial extents is fundamental for macroecological research and biodiversity conservation. Using the GrassPlot database, we provide benchmarks of fine-grain richness values of Palaearctic open habitats for vascular plants, bryophytes, lichens and complete vegetation (i.e., the sum of the former three groups). Location: Palaearctic biogeographic realm. Methods: We used 126,524 plots of eight standard grain sizes from the GrassPlot database: 0.0001, 0.001, 0.01, 0.1, 1, 10, 100 and 1,000 m² and calculated the mean richness and standard deviations, as well as maximum, minimum, median, and first and third quartiles for each combination of grain size, taxonomic group, biome, region, vegetation type and phytosociological class. Results: Patterns of plant diversity in vegetation types and biomes differ across grain sizes and taxonomic groups. Overall, secondary (mostly semi-natural) grasslands and natural grasslands are the richest vegetation type. The open-access file ”GrassPlot Diversity Benchmarks” and the web tool “GrassPlot Diversity Explorer” are now available online (https://edgg.org/databases/GrasslandDiversityExplorer) and provide more insights into species richness patterns in the Palaearctic open habitats. Conclusions: The GrassPlot Diversity Benchmarks provide high-quality data on species richness in open habitat types across the Palaearctic. These benchmark data can be used in vegetation ecology, macroecology, biodiversity conservation and data quality checking. While the amount of data in the underlying GrassPlot database and their spatial coverage are smaller than in other extensive vegetation-plot databases, species recordings in GrassPlot are on average more complete, making it a valuable complementary data source in macroecology.

Abstract: Motivation: Assessing biodiversity status and trends in plant communities is critical for understanding, quantifying and predicting the effects of global change on ecosystems. Vegetation plots record the occurrence or abundance of all plant species co-occurring within delimited local areas. This allows species absences to be inferred, information seldom provided by existing global plant datasets. Although many vegetation plots have been recorded, most are not available to the global research community. A recent initiative, called ‘sPlot’, compiled the first global vegetation plot database, and continues to grow and curate it. The sPlot database, however, is extremely unbalanced spatially and environmentally, and is not open-access. Here, we address both these issues by (a) resampling the vegetation plots using several environmental variables as sampling strata and (b) securing permission from data holders of 105 local-to-regional datasets to openly release data. We thus present sPlotOpen, the largest open-access dataset of vegetation plots ever released. sPlotOpen can be used to explore global diversity at the plant community level, as ground truth data in remote sensing applications, or as a baseline for biodiversity monitoring. Main types of variable contained: Vegetation plots (n = 95,104) recording cover or abundance of naturally co-occurring vascular plant species within delimited areas. sPlotOpen contains three partially overlapping resampled datasets (c. 50,000 plots each), to be used as replicates in global analyses. Abstract: Aim: Alpine ecosystems differ in area, macroenvironment and biogeographical history across the Earth, but the relationship between these factors and plant species richness is still unexplored. Here, we assess the global patterns of plant species richness in alpine ecosystems and their association with environmental, geographical and historical factors at regional and community scales. Location: Global. Time period: Data collected between 1923 and 2019. Major taxa studied: Vascular plants. Methods: We used a dataset representative of global alpine vegetation, consisting of 8,928 plots sampled within 26 ecoregions and six biogeographical realms, to estimate regional richness using sample-based rarefaction and extrapolation. Then, we evaluated latitudinal patterns of regional and community richness with generalized additive models. Using environmental, geographical and historical predictors from global raster layers, we modelled regional and community richness in a mixed-effect modelling framework. Results: The latitudinal pattern of regional richness peaked around the equator and at mid-latitudes, in response to current and past alpine area, isolation and the variation in soil pH among regions. At the community level, species richness peaked at mid-latitudes of the Northern Hemisphere, despite a considerable within-region variation. Community richness was related to macroclimate and historical predictors, with strong effects of other spatially structured factors. Main conclusions: In contrast to the well-known latitudinal diversity gradient, the alpine plant species richness of some temperate regions in Eurasia was comparable to that of hyperdiverse tropical ecosystems, such as the páramo. The species richness of these putative hotspot regions is explained mainly by the extent of alpine area and their glacial history, whereas community richness depends on local environmental factors. Abstract: Background: Pediatric trials to add missing data for evidence-based pharmacotherapy are still scarce. A tailored training concept appears to be a promising tool to cope with critical and complex situations before enrolling the very first patient and subsequently to ensure high-quality study conduct. The aim was to facilitate study success by optimizing the preparedness of the study staff shift. Method: An interdisciplinary faculty developed a simulation training focusing on the communication within the informed consent procedure and the conduct of the complex pharmacokinetic/pharmacodynamic (PK/PD) sampling within a simulation facility. Scenarios were video-debriefed by an audio-video system and manikins with artificial blood simulating patients were used. The training was evaluated by participants' self-assessment before and during trial recruitment. Results: The simulation training identified different optimization potentials for improved informed consent process and study conduct. It facilitated the reduction of avoidable errors, especially in the early phase of a clinical study. The knowledge gained through the intervention was used to train the study teams, improve the team composition and optimize the on-ward setting for the FP-7 funded “LENA” project (grant agreement no. 602295). Self-perceived ability to communicate core elements of the trial as well as its correct performance of sample preparation increased significantly (mean, 95% CI, p ≤ 0.0001) from 3 (2.5–3.5) to four points (4.0–4.5), and from 2 (1.5–2.5) to five points (4.0–5.0). Conclusion: An innovative training concept to optimize the informed consent process and study conduct was successfully developed and enabled high-quality conduct of the pediatric trials as of the very first patient visit.

Abstract: Aim: To evaluate the impact of left ventricular ejection fraction (LVEF) on 3-year outcomes in patients with left main coronary artery disease (LMCAD) undergoing percutaneous coronary intervention (PCI) or coronary artery bypass grafting (CABG) in the EXCEL trial. Methods and results: The EXCEL trial randomized patients with LMCAD to PCI with everolimus-eluting stents (n = 948) or CABG (n = 957). Among 1804 patients with known baseline LVEF, 74 (4.1%) had LVEF <40% [heart failure with reduced ejection fraction (HFrEF)], 152 (8.4%) LVEF 40–49% [heart failure with mid-range ejection fraction (HFmrEF)] and 1578 (87.5%) LVEF ≥50% (heart failure with preserved ejection fraction). Patients with HFrEF vs. HFmrEF vs. preserved LVEF experienced a longer postoperative hospital stay (9.0 vs. 7.0 vs. 6.0 days, P = 0.02) with greater peri-procedural complications after CABG, while hospital stay after PCI was unaffected by LVEF (1.5 vs. 2.0 vs. 1.0 days, P = 0.20). The composite primary endpoint of death, stroke, or myocardial infarction at 3 years was 29.3% (PCI) vs. 27.6% (CABG) in patients with HFrEF, 16.2% vs. 15.0% in patients with HFmrEF, and 14.5% vs. 14.6% in those with preserved LVEF, respectively (Pinteraction = 0.90). Smoothing spline analysis demonstrated that the 3-year risk of all-cause death increased when LVEF decreased, both in patients undergoing CABG and PCI. Conclusion: In the EXCEL trial, the composite rate of death, stroke or myocardial infarction at 3 years was significantly higher in patients with HFrEF compared with HFmrEF or preserved LVEF, driven by an increased rate of all-cause death. No significant differences after PCI vs. CABG were observed among patients with HFrEF, HFmrEF and preserved LVEF. Longer-term follow-up could provide important insights on differences in clinical outcomes that might emerge over time. Clinical Trial Registration: ClinicalTrials.gov ID NCT01205776.

Abstract: One of the most widely conserved hallmarks of aging is a decline in functional capabilities. Mobility loss is particularly burdensome due to its association with negative health outcomes, loss of independence and disability, and the heavy impact on quality of life. Recently, a new condition, physical frailty and sarcopenia, has been proposed to define a critical stage in the disabling cascade. Physical frailty and sarcopenia are characterized by weakness, slowness, and reduced muscle mass, yet with preserved ability to move independently. One of the strategies that have shown some benefits in combatting mobility loss and its consequences for older adults is physical activity. Here, we describe the opportunities and challenges for the development of physical activity interventions in people with physical frailty and sarcopenia. The aim of this article is to review age-related physio(patho)logical changes that impact mobility in old age and to provide recommendations and procedures in accordance with the available literature.

Abstract: Aim: A fundamental question in macroecology centres around understanding the relationship between species’ local abundance and their distribution in geographical and climatic space (i.e. the multi‐dimensional climatic space or climatic niche). Here, we tested three macroecological hypotheses that link local abundance to the following range properties: (a) the abundance–range size relationship, (b) the abundance–range centre relationship and (c) the abundance–suitability relationship. Location: Europe. Taxon: Vascular plants. Methods: Distribution range maps were extracted from the Chorological Database Halle to derive information on the range and niche sizes of 517 European vascular plant species. To estimate local abundance, we assessed samples from 744,513 vegetation plots in the European Vegetation Archive, where local species’ abundance is available as plant cover per plot. We then calculated the ‘centrality’, that is, the distance between the location of the abundance observation and each species’ range centre in geographical and climatic space. The climatic suitability of plot locations was estimated using coarse‐grain species distribution models (SDMs). The relationships between centrality or climatic suitability with abundance was tested using linear models and quantile regression. We summarized the overall trend across species’ regression slopes from linear models and quantile regression using a meta‐analytical approach. Results: We did not detect any positive relationships between a species’ mean local abundance and the size of its geographical range or climatic niche. Contrasting yet significant correlations were detected between abundance and centrality or climatic suitability among species. Abstract: When deciding to purchase a particular car, for instance, most consumers will consider the price and technical equipment of various alternatives. But apart from this, consumers might also reflect on their design and/or eco-compatibility. Put differently, consumption decisions are not only a function of economic or functional value, but also of aesthetic and ethical considerations. They are influenced by evaluations as to which products are beautiful and good, or ugly and bad. While sociology has paid increasing attention to the aesthetic and ethical underpinnings of consumption and lifestyles in recent decades, for the most part, research has either dealt with their formal-aesthetic or the ethical-moral aspects, without much cross-fertilization. In this chapter, we argue, in contrast, that lifestyles are structured by both aesthetic and ethical orientations. The impact and overlap of each of these two orientations, however, is likely to vary between different domains of consumption. Therefore, we theoretically analyze the connections between ethical and aesthetic orientations, consumption, and lifestyles. We locate the debate on aesthetics and ethics in the two classical sociological accounts of consumption, put forward by Max Weber and Pierre Bourdieu. Further, we discuss recent empirical findings on the aesthetic and ethical underpinnings of consumption and lifestyles. Finally, we provide a quantitative analysis of the relative importance of aesthetic and ethical orientations for a broad range of consumption behaviors, including cultural consumption, media consumption, food, sports, and housing. We find that the majority of these behaviors are collectively shaped by aesthetic and ethical orientations, although aesthetic orientations are the dominant structuring factor of consumption.

This requires interdisciplinary collaboration and very often collaboration across a variety of research organizations. This study highlights the key topics, purposes, and approaches of socio‐economic analysis within the CGIAR related to cropping. Although each CGIAR center has a different mission, all CGIAR centers share a common strategy of striving towards a world free of hunger, poverty, and environmental degradation. This means research is mostly focused towards resource‐constrained smallholder farmers. The review covers global modeling efforts using the IMPACT model to farm household bio‐economic models for assessing the potential impact of new technologies on farming systems and livelihoods. Although the CGIAR addresses all aspects of food systems, the focus of this review is on crop commodities and the economic analysis linked to crop‐growth model results. This study, while not a comprehensive review, provides insights into the richness of the socio‐economic modeling endeavors within the CGIAR. The study highlights the need for interdisciplinary approaches to address the challenges this type of modeling faces.

Abstract: Background: In all societies, the burden and cost of allergic and chronic respiratory diseases are increasing rapidly. Most economies are struggling to deliver modern health care effectively. There is a need to support the transformation of the health care system into integrated care with organizational health literacy. Main body: As an example for chronic disease care, MASK (Mobile Airways Sentinel NetworK), a new project of the ARIA (Allergic Rhinitis and its Impact on Asthma) initiative, and POLLAR (Impact of Air POLLution on Asthma and Rhi-nitis, EIT Health), in collaboration with professional and patient organizations in the field of allergy and airway diseases, are proposing real-life ICPs centred around the patient with rhinitis, and using mHealth to monitor environmental exposure. Three aspects of care pathways are being developed: (i) Patient participation, health literacy and self-care through technology-assisted "patient activation", (ii) Implementation of care pathways by pharmacists and (iii) Next-generation guidelines assessing the recommendations of GRADE guidelines in rhinitis and asthma using real-world evidence (RWE) obtained through mobile technology. The EU and global political agendas are of great importance in supporting the digital transformation of health and care, and MASK has been recognized by DG Santé as a Good Practice in the field of digitally-enabled, integrated, person-centred care. Conclusion: In 20 years, ARIA has considerably evolved from the first multimorbidity guideline in respiratory diseases to the digital transformation of health and care with a strong political involvement.

Abstract: Due to actual global concerns for farm animal welfare and an also prominent issue of climate change effect, this study was conducted in order to examine the expression of dairy cows’ health parameters in two main seasons (winter and summer). Thirteen health parameters classified into two main groups, injuries and diseases, were selected and analysed according to their relevance for welfare provision by Welfare Quality® Assessment Protocol for Dairy Cows (2009). According to the final calculation score (0100 points) parameters were descriptively categorized as unacceptable, acceptable, enhanced or excellent in terms of welfare provision. Data were collected through clinical scoring and veterinary reports available on a total of 16 dairy farms (N=4.833 cows). The temperature ranges for winter and summer season in a given year were -4.2 to 7.2˚C and 19.3 to 27.4 ˚C respectively. Obtained welfare scores for injuries (41/100 points) and diseases (59/100 points) showed that cows in both seasons were more prone to injuries than to diseases. Estimated welfare condition for injuries was categorized as acceptable, and for diseases as enhanced, indicating no severe risk for welfare was determined. No significant differences for examined parameters were found between seasons with exception of somewhat higher lameness frequency in the winter season (39% vs. 32%). Frequencies of diseases were, in majority below alarm thresholds except for mastitis (2.6%) and conjunctivitis (6.0%) in the summer season.

Abstract: Objective: Alcohol-related pancreatitis is associated with a disproportionately large number of hospitalisations among GI disorders. Despite its clinical importance, genetic susceptibility to alcoholic chronic pancreatitis (CP) is poorly characterised. To identify risk genes for alcoholic CP and to evaluate their relevance in non-alcoholic CP, we performed a genome-wide association study and functional characterisation of a new pancreatitis locus. Design: 1959 European alcoholic CP patients and population-based controls from the KORA, LIFE and INCIPE studies (n=4708) as well as chronic alcoholics from the GESGA consortium (n=1332) were screened with Illumina technology. For replication, three European cohorts comprising 1650 patients with non-alcoholic CP and 6695 controls originating from the same countries were used. Results: We replicated previously reported risk loci CLDN2-MORC4, CTRC, PRSS1-PRSS2 and SPINK1 in alcoholic CP patients. We identified CTRB1-CTRB2 (chymotrypsin B1 and B2) as a new risk locus with lead single-nucleotide polymorphism (SNP) rs8055167 (OR 1.35, 95% CI 1.23 to 1.6). We found that a 16.6 kb inversion in the CTRB1-CTRB2 locus was in linkage disequilibrium with the CP-associated SNPs and was best tagged by rs8048956. The association was replicated in three independent European non-alcoholic CP cohorts of 1650 patients and 6695 controls (OR 1.62, 95% CI 1.42 to 1.86). The inversion changes the expression ratio of the CTRB1 and CTRB2 isoforms and thereby affects protective trypsinogen degradation and ultimately pancreatitis risk. Conclusion: An inversion in the CTRB1-CTRB2 locus modifies risk for alcoholic and non-alcoholic CP indicating that common pathomechanisms are involved in these inflammatory disorders.

Abstract: Previous studies have shown an increased risk for mental health problems in children born to both younger and older parents compared to children of average-aged parents. We previously used a novel design to reveal a latent mechanism of genetic association between schizophrenia and age at first birth in women (AFB). Abstract: Background: Despite the well-established benefits of secondary cardiovascular prevention, the importance of concurrent medical therapy in clinical trials of coronary revascularization is often overlooked. Objectives: The goal of this study was to assess compliance with guideline-directed medical therapy (GDMT) in clinical trials and its potential impact on the comparison between percutaneous coronary intervention (PCI) and coronary artery bypass grafting (CABG). Methods: The Cochrane Central Register of Controlled Trials and MEDLINE were searched from 2005 to August 2017. Clinical trial registries and reference lists of relevant studies were also searched. Randomized controlled trials comparing PCI with drug-eluting stents versus CABG and reporting medical therapy after revascularization were included. The study outcome was compliance with GDMT, defined as the following: 1) any antiplatelet agent plus beta-blocker plus statin (GDMT1); and 2) any antiplatelet agent plus beta-blocker plus statin plus angiotensin-converting enzyme inhibitor/angiotensin receptor blocker (GDMT2). Data collection and analysis were performed according to the methodological recommendations of The Cochrane Collaboration. Results: From a total of 439 references, 5 trials were included based on our inclusion and exclusion criteria. Overall, compliance with GDMT1 was low and decreased over time from 67% at 1 year to 53% at 5 years. Compliance with GDMT2 was even lower and decreased from 40% at 1 year to 38% at 5 years. Compliance with both GDMT1 and GDMT2 was higher in PCI than in CABG at all time points. Meta-regression suggested an association between lower use of GDMT1 and adverse clinical outcomes in PCI versus CABG at 5 years. Conclusions: Compliance with GDMT in contemporary clinical trials remains suboptimal and is significantly lower after CABG than after PCI, which may influence the comparison of clinical trial endpoints between those study groups.

Abstract: Plant functional traits directly affect ecosystem functions. At the species level, trait combinations depend on trade-offs representing different ecological strategies, but at the community level trait combinations are expected to be decoupled from these trade-offs because different strategies can facilitate co-existence within communities. A key question is to what extent community-level trait composition is globally filtered and how well it is related to global versus local environmental drivers. Here, we perform a global, plot-level analysis of trait–environment relationships, using a database with more than 1.1 million vegetation plots and 26,632 plant species with trait information. Although we found a strong filtering of 17 functional traits, similar climate and soil conditions support communities differing greatly in mean trait values. The two main community trait axes that capture half of the global trait variation (plant stature and resource acquisitiveness) reflect the trade-offs at the species level but are weakly associated with climate and soil conditions at the global scale. Similarly, within-plot trait variation does not vary systematically with macro-environment. Our results indicate that, at fine spatial grain, macro-environmental drivers are much less important for functional trait composition than has been assumed from floristic analyses restricted to co-occurrence in large grid cells. Instead, trait combinations seem to be predominantly filtered by local-scale factors such as disturbance, fine-scale soil conditions, niche partitioning and biotic interactions.

Abstract: Altered energy metabolism is a cancer hallmark as malignant cells tailor their metabolic pathways to meet their energy requirements. Glucose and glutamine are the major nutrients that fuel cellular metabolism, and the pathways utilizing these nutrients are often altered in cancer. Here, we show that the long ncRNA CCAT2, located at the 8q24 amplicon on cancer risk-associated rs6983267 SNP, regulates cancer metabolism in vitro and in vivo in an allele-specific manner by binding the Cleavage Factor I (CFIm) complex with distinct affinities for the two subunits (CFIm25 and CFIm68). The CCAT2 interaction with the CFIm complex fine-tunes the alternative splicing of Glutaminase (GLS) by selecting the poly(A) site in intron 14 of the precursor mRNA. These findings uncover a complex, allele-specific regulatory mechanism of cancer metabolism orchestrated by the two alleles of a long ncRNA. Redis et al. report that the two alleles of the lncRNA, CCAT2, induce distinct metabolic phenotypes. By interacting with the CFIm complex with allele-specific affinities, CCAT2 regulates the alternative splicing of GLS, resulting in the preferential expression of the more aggressive splice isoform.

Abstract: Pharmacogenomics aims to correlate inter-individual differences of drug efficacy and/or toxicity with the underlying genetic composition, particularly in genes encoding for protein factors and enzymes involved in drug metabolism and transport. In several European populations, particularly in countries with lower income, information related to the prevalence of pharmacogenomic biomarkers is incomplete or lacking. Here, we have implemented the microattribution approach to assess the pharmacogenomic biomarkers allelic spectrum in 18 European populations, mostly from developing European countries, by analyzing 1,931 pharmacogenomics biomarkers in 231 genes. Our data show significant interpopulation pharmacogenomic biomarker allele frequency differences, particularly in 7 clinically actionable pharmacogenomic biomarkers in 7 European populations, affecting drug efficacy and/or toxicity of 51 medication treatment modalities. These data also reflect on the differences observed in the prevalence of high-risk genotypes in these populations, as far as common markers in the CYP2C9, CYP2C19, CYP3A5, VKORC1, SLCO1B1 and TPMT pharmacogenes are concerned. Also, our data demonstrate notable differences in predicted genotype-based warfarin dosing among these populations. Our findings can be exploited not only to develop guidelines for medical prioritization, but most importantly to facilitate integration of pharmacogenomics and to support pre-emptive pharmacogenomic testing. This may subsequently contribute towards significant cost-savings in the overall healthcare expenditure in the participating countries, where pharmacogenomics implementation proves to be cost-effective.

27. Estimation of nonadditive genetic influences on standard lactation production (305 days) in upgrading of Serbian-Fleckvieh to Holstein-Friesian cattle breed

Abstract: The purpose of this paper is to estimate the effects of systematic factors, primarily the degree of crossing, on the standard lactation lengths of 305 days in 6 different genotypes of cows (F1, R1, R2, R3, R4, R5) obtained by the upgrading of the Serbian Fleckvieh (SF) to the Holstein-Friesian (HF) breed of cattle. The systematic effects included in the general linear model (GLM) exerted a significant (P<0.05) influence on the milk yield, as well as the fat yield and content. During the upgrading process, statistically significant (P<0.05) differences were recorded between the traits of the genotypes examined. With an increase in the share of HF genes, the milk yield increased from 5020 kg (F1) to 5801 kg (R5) and the fat yield from 176.2 kg to 201.6 kg, whereas the fat content simultaneously decreased from 3.55 % (F1) to 3.49 % (R5). The F1 cows had the highest heterosis realized (hR) regarding the milk yield (185.8 kg), while the R1 and R2 cows displayed negative heterosis effects (-21.0 and -205.7 kg) and negative recombination effects (rI). The highest value of hR (regarding the milk fat yield) was recorded in the F1 cows (6.07 kg) in comparison with the R2 cows, which had the negative hR (-3.79 kg), whereas the negative rI was recorded in the R1 and R2 genotypes. The positive rI (regarding the milk fat content) was recorded in the R1, R2 and R3 genotypes accompanied by the positive hR., Cilj ovog rada bio je ispitati procjenu sistematskih faktora, prvenstveno stupnja križanja na standardnu laktacijsku proizvodnju od 305 dana kod 6 genotipova krava (F1, R1, R2, R3, R4 i R5) dobivenih pretapanjem domaćeg šarenog goveda (SF) s holštajn-frizijskom (HF) pasminom goveda. Sistematski utjecaji uključeni u GLM model imali su značajan utjecaj (P<0,05) na količinu mlijeka, mliječne masti i postotak mliječne masti. Između dobivenih genotipova tijekom procesa pretapanja, zabilježene su statistički značajne razlike (P<0,05) u promatranim osobinama. S povećanjem udjela HF gena, povećavala se i količina mlijeka od 5020 kg kod F1 generacije do 5801 kg kod R5 generacije, mliječne masti od 176,2 do 201,6 kg, dok je istovremeno došlo do pada postotka mliječne masti (od 3,55 % kod F1 do ,49 % kod R5). Krave F1 generacije su imale najveći realizirani heterozis (hR) za količinu mlijeka od 185,8 kg, dok su krave genotipa R1 i R2 imale negativan realizirani heterozis (-21,0 odnosno -205,70 kg) pa samim tim i negativne rekombinacije (rI). Najveća realizirana vrijednost heterozisa (hR) za količinu mliječne masti je kod krava F1 generacije (6,07 kg), u odnosu na krave R2 generacije koje su imale negativan realizirani heterozis (-3,79 kg), a negativne rekombinacije su zabilježene kod krava R1 i R2 genotipa. Kod krava genotipa R1, R2 i R3 ustanovljene su pozitivne rekombinacije (rI) za postotak mliječne masti, uslijed čega su i realizirani heterozisi (hR) bili pozitivni.

Abstract: The purpose of this paper is to estimate the effects of systematic factors, primarily the degree of crossing, on the standard lactation lengths of 305 days in 6 different genotypes of cows (F1, R1, R2, R3, R4, R5) obtained by the upgrading of the Serbian Fleckvieh (SF) to the Holstein-Friesian (HF) breed of cattle. The systematic effects included in the general linear model (GLM) exerted a significant (P<0.05) influence on the milk yield, as well as the fat yield and content. During the upgrading process, statistically significant (P<0.05) differences were recorded between the traits of the genotypes examined. With an increase in the share of HF genes, the milk yield increased from 5020 kg (F1) to 5801 kg (R5) and the fat yield from 176.2 kg to 201.6 kg, whereas the fat content simultaneously decreased from 3.55 % (F1) to 3.49 % (R5). The F1 cows had the highest heterosis realized (hR) regarding the milk yield (185.8 kg), while the R1 and R2 cows displayed negative heterosis effects (-21.0 and -205.7 kg) and negative recombination effects (rI). The highest value of hR (regarding the milk fat yield) was recorded in the F1 cows (6.07 kg) in comparison with the R2 cows, which had the negative hR (-3.79 kg), whereas the negative rI was recorded in the R1 and R2 genotypes. The positive rI (regarding the milk fat content) was recorded in the R1, R2 and R3 genotypes accompanied by the positive hR., Cilj ovog rada bio je ispitati procjenu sistematskih faktora, prvenstveno stupnja križanja na standardnu laktacijsku proizvodnju od 305 dana kod 6 genotipova krava (F1, R1, R2, R3, R4 i R5) dobivenih pretapanjem domaćeg šarenog goveda (SF) s holštajn-frizijskom (HF) pasminom goveda. Sistematski utjecaji uključeni u GLM model imali su značajan utjecaj (P<0,05) na količinu mlijeka, mliječne masti i postotak mliječne masti. Između dobivenih genotipova tijekom procesa pretapanja, zabilježene su statistički značajne razlike (P<0,05) u promatranim osobinama. S povećanjem udjela HF gena, povećavala se i količina mlijeka od 5020 kg kod F1 generacije do 5801 kg kod R5 generacije, mliječne masti od 176,2 do 201,6 kg, dok je istovremeno došlo do pada postotka mliječne masti (od 3,55 % kod F1 do ,49 % kod R5). Krave F1 generacije su imale najveći realizirani heterozis (hR) za količinu mlijeka od 185,8 kg, dok su krave genotipa R1 i R2 imale negativan realizirani heterozis (-21,0 odnosno -205,70 kg) pa samim tim i negativne rekombinacije (rI). Najveća realizirana vrijednost heterozisa (hR) za količinu mliječne masti je kod krava F1 generacije (6,07 kg), u odnosu na krave R2 generacije koje su imale negativan realizirani heterozis (-3,79 kg), a negativne rekombinacije su zabilježene kod krava R1 i R2 genotipa. Kod krava genotipa R1, R2 i R3 ustanovljene su pozitivne rekombinacije (rI) za postotak mliječne masti, uslijed čega su i realizirani heterozisi (hR) bili pozitivni.

Abstract: PURPOSE. To evaluate the validity of a novel fully automated three-dimensional (3D) method capable of segmenting the choroid from two different optical coherence tomography scanners: swept-source OCT (SS-OCT) and spectral-domain OCT (SD-OCT). METHODS. One hundred eight subjects were imaged using SS-OCT and SD-OCT. A 3D method was used to segment the choroid and quantify the choroidal thickness along each A-scan. The segmented choroidal posterior boundary was evaluated by comparing to manual segmentation. Differences were assessed to test the agreement between segmentation results of the same subject. Choroidal thickness was defined as the Euclidian distance between Bruch’s membrane and the choroidal posterior boundary, and reproducibility was analyzed using automatically and manually determined choroidal thicknesses. RESULTS. For SS-OCT, the average choroidal thickness of the entire 6-by 6-mm2 macular region was 219.5 lm (95% confidence interval [CI], 204.9–234.2 µm), and for SD-OCT it was 209.5 µm (95% CI, 197.9–221.0 µm). The agreement between automated and manual segmentations was high: Average relative difference was less than 5 lm, and average absolute difference was less than 15 µm. Reproducibility of choroidal thickness between repeated SS-OCT scans was high (coefficient of variation [CV] of 3.3%, intraclass correlation coefficient [ICC] of 0.98), and differences between SS-OCT and SD-OCT results were small (CV of 11.0%, ICC of 0.73). CONCLUSIONS. We have developed a fully automated 3D method for segmenting the choroid and quantifying choroidal thickness along each A-scan. The method yielded high validity. Our method can be used reliably to study local choroidal changes and may improve the diagnosis and management of patients with ocular diseases in which the choroid is affected.

Abstract: The large 2014/2015 Ebola virus outbreak in West Africa points out the urgent need to develop new preventive and therapeutic approaches that are effective against Ebola viruses and can be rapidly utilized. Recently, a simple theoretical criterion for the virtual screening of molecular libraries for candidate inhibitors of Ebola virus infection was proposed. Using this method the 'drug space' was screened and 267 approved and 382 experimental drugs as candidates for treatment of the Ebola virus disease (EVD) have been selected. Detailed analysis of these drugs revealed the non-steroidal anti-inflammatory drug ibuprofen as an inexpensive, widely accessible and minimally toxic candidate for prevention and treatment of EVD. Furthermore, the molecular mechanism underlying this possible protective effect of ibuprofen against EVD is suggested in this article.

Abstract: Purpose. To determine, first, which regions of 3-D optical coherence tomography (OCT) volumes can be segmented completely in the majority of subjects and, second, the relationship between analyzed area and thickness measurement test-retest variability. Methods. Three-dimensional OCT volumes (6 × 6 mm) centered around the fovea and optic nerve head (ONH) of 925 Rotterdam Study participants were analyzed; 44 participants were scanned twice. Volumes were segmented into 10 layers, and we determined the area where all layers could be identified in at least 95% (macula) or 90% (ONH) of subjects. Macular volumes were divided in 2 × 2, 4 × 4, 6 × 6, 8 × 8, or 68 blocks. We placed two circles around the ONH; the ONH had to fit into the smaller circle, and the larger circle had to fit into the segmentable part of the volume. The area between the circles was divided in 3 to 12 segments. We determined the test-retest variability (coefficient of repeatability) of the retinal nerve fiber layer (RNFL) and ganglion cell layer (RGCL) thickness measurements as a function of size of blocks/segments. Results. Eighty-two percent of the macular volume could be segmented in at least 95% of subjects; for the ONH, this was 65% in at least 90%. The radii of the circles were 1.03 and 1.84 mm. Depending on the analyzed area, median test-retest variability ranged from 8% to 15% for macular RNFL, 11% to 22% for macular RGCL, 5% to 11% for the two together, and 18% to 22% for ONH RNFL. Conclusions. Test-retest variability hampers a detailed analysis of 3-D OCT data. Combined macular RNFL and RGCL thickness averaged over larger areas had the best test-retest variability.

33. Behavioral Finance and Agent-Based Artificial Markets

Abstract: Studying the behavior of market participants is important due to its potential impact on asset prices and the dynamics of financial markets. The idea of individual investors who are prone to biases in judgment and who use various heuristics, which might lead to anomalies on the market level, has been explored within the field of behavioral finance. In this dissertation, we analyze market-wise implications of investor behavior and their irrationalities by means of agent-based simulations of financial markets. The usefulness of agent-based artificial markets for studying the behavioral finance topics stems from their ability to relate the micro-level behavior of individual market participants (represented as agents) and the macro-level behavior of the market (artificial time-series). This micro-macro mapping of agent-based methodology is particularly useful for behavioral finance, because that link is often broken when using other methodological approaches. In this thesis, we study various biases commented in the behavioral finance literature and propose novel models for some of the behavioral phenomena. We provide mathematical definitions and computational implementations for overconfidence (miscalibration and better-than-average effect), investor sentiment (optimism and pessimism), biased self-attribution, loss aversion, and recency and primacy effects. The levels of these behavioral biases are related to the features of the market dynamics, such as the bubbles and crashes, and the excess volatility of the market price. The impact of behavioral biases on investor performance is also studied.

34. A Conceptual Model of Investor Behavior

Abstract: Based on a survey of behavioral finance literature, this paper presents a descriptive model of individual investor behavior in which investment decisions are seen as an iterative process of interactions between the investor and the investment environment. This investment process is influenced by a number of interdependent variables and driven by dual mental systems, the interplay of which contributes to boundedly rational behavior where investors use various heuristics and may exhibit behavioral biases. In the modeling tradition of cognitive science and intelligent systems, the investor is seen as a learning, adapting, and evolving entity that perceives the environment, processes information, acts upon it, and updates his or her internal states. This conceptual model can be used to build stylized representations of (classes of) individual investors, and further studied using the paradigm of agent-based artificial financial markets. By allowing us to implement individual investor behavior, to choose various market mechanisms, and to analyze the obtained asset prices, agent-based models can bridge the gap between the micro level of individual investor behavior and the macro level of aggregate market phenomena. It has been recognized, yet not fully explored, that these models could be used as a tool to generate or test various behavioral hypothesis.

35. Testing for smooth transition nonlinearity in adjustments of cointegrating systems

Abstract: This paper studies testing for the presence of smooth transition nonlinearity in adjustment parameters of the vector error correction model. We specify the generalized model with multiple cointegrating vectors and different transition functions across equations. Given that the nonlinear model is highly complex, this paper proposes an optimal LM test based only on estimation of the linear model. The null asymptotic distribution is derived using empirical process theory and since the transition parameters of the model cannot be identified under the null hypothesis bootstrap procedures are used to approximate the limit. Monte Carlo simulations indicate a good performance of the test.

Abstract: Frequency of INherited Disorders database (FINDbase) (http://www.findbase.org) is a relational database, derived from the ETHNOS software, recording frequencies of causative mutations leading to inherited disorders worldwide. Database records include the population and ethnic group, the disorder name and the related gene, accompanied by links to any corresponding locus-specific mutation database, to the respective Online Mendelian Inheritance in Man entries and the mutation together with its frequency in that population. The initial information is derived from the published literature, locus-specific databases and genetic disease consortia. FINDbase offers a user-friendly query interface, providing instant access to the list and frequencies of the different mutations. Query outputs can be either in a table or graphical format, accompanied by reference(s) on the data source. Registered users from three different groups, namely administrator, national coordinator and curator, are responsible for database curation and/or data entry/ correction online via a password-protected interface. Databaseaccess is free of charge and there are no registration requirements for data querying. FINDbase provides a simple, web-based system for population-based mutation data collection and retrieval and can serve not only as a valuable online tool for molecular genetic testing of inherited disorders but also as a non-profit model for sustainable database funding, in the form of a 'database-journal'.
