Grape processing generates large amounts of by-products, including seeds rich in hydrophilic and lipophilic antioxidants. This study demonstrates, for the first time, that subjecting grape seeds to a single ultrasound-assisted extraction (UAE) with aqueous ethanolic solutions yields both flavan-3-ols and tocochromanols in the final extract. Notably, the water content in ethanol significantly influences the extractability of tocochromanols more than flavan-3-ols. Solid-to-solvent ratios of 1:50 to 1:2 were tested for both analytical and industrial applications. A sustainable analytical approach for recovering flavan-3-ols and tocochromanols using 60% and 96.4% ethanol extractions was validated and employed to profile nineteen genotypes of lesser-studied interspecific grape crosses (Vitis spp.). Different genotypes showed a wide range of concentrations of tocopherols (1.6-6.3 mg/100 g), tocotrienols (1.0-17.4 mg/100 g), and flavan-3-ols (861-9994 mg/100 g). This indicated that the genetic background and maturity of the plant material are crucial factors from an industrial perspective due to the initial concentration of bioactive compounds. Introduction: The safety and quality of many medicinally important herbs are compromised since farmers and small organizations are involved in the cultivation, aggregation, and primary processing of these herbs. Such organizations often lack adequate quality control facilities. To improve the safety and quality of herbal products, simple, rapid, and affordable quality control systems are required., Objectives: The aim of this study was to assess the suitability of microwave oven-drying for moisture content (MC) determination and sample preparation of herbs in small organizations., Methods: Microwave oven-drying (720 W) and convective oven-drying at 105°C for MC determination were compared. The effects of three different drying methods (microwave oven-drying, low-temperature convective drying, and freeze-drying) on in vitro antioxidant and polyphenol oxidase (PPO) activity were determined, similarity analysis was conducted using HPLC signature spectra, and validation was performed with LC-MS focusing on one herb., Results: Microwave oven-drying at 720 W significantly reduced the drying time (from hours to minutes), whereas the spatial variation of temperature in convective ovens set at 105°C can cause about 10% underestimation of MC. Microwave oven-drying showed similar macro-properties like freeze-drying and higher extractability (10%-20%) and in vitro antioxidant capacity (33%-66%) and lower PPO activity compared to low-temperature convective drying. HPLC signature spectra revealed strong similarity of soluble components between freeze-dried and microwave oven-dried herbs. LC-MS analysis demonstrated more common compounds between freeze-dried and microwave oven-dried Centella asiatica extracts, whereas convective tray-dried samples had fewer compounds common with samples obtained by freeze-drying or microwave oven-drying., Conclusions: Microwave oven-drying is rapid (tens of min) and shows small batch-to-batch variation compared to oven-drying at 105°C. This study investigated the effectiveness of various surfactants at different concentrations in removing high concentrations of polycyclic aromatic hydrocarbons (PAHs) from soil with high mineral content, focusing on the impact of surfactant treatment on the mobility of the residual PAHs in soil. The results revealed that the cationic surfactant (CTMAB) inhibited removal of PAHs in the whole tested concentration range of 0.1-8 g/L. In contrast, the non-ionic and anionic surfactants (Triton X-100 and SDBS) significantly enhanced removal of PAHs as their amendment concentrations reached 2 g/L and above. Triton X-100 exhibited steadily increased efficacy with increasing amendment concentrations and maintained favorable solubilization capability when continuously amended, making it the preferable choice for remediating PAHs-contaminated soil. Surfactant and water washing processes altered soil physicochemical properties by removing some clay minerals (e.g., faujasite) and organic matter that can bind or sequester PAHs, potentially increasing their extractability and bioavailability in the washed soil, thereby posing higher ecological risks compared to the original one. Although soil washing decreased retention of the remaining PAHs in soil, it did not significantly impact PAHs release from soil by flowing water. Amylase trypsin inhibitors (ATIs) play an important role in wheat allergies and potentially in non-coeliac wheat sensitivity. Food processing could be important to mitigate the pathogenic properties of ATIs, e.g., by denaturation, glycation, enzymatic hydrolysis, cross-linking, and oxidation and reduction. These modifications also impact the solubility and extractability. The complex solubility behaviour of ATI isoforms (water and salt soluble, but also chloroform-methanol soluble, solubility depending on the redox state) becomes even more complex upon processing due to denaturation and (bio)chemical modifications. This significantly hinders the feasibility of quantitative extraction. Moreover, changes in biofunctionality may occur during the process of extraction, and the changes in ATI due to food processing will be more difficult to assess. Heat treatment decreases the extractability of ATIs with water, NaCl, and other buffer extracts, and binding of IgE from wheat-allergic persons to ATIs as observed with Western blotting is decreased or absent. IgE binding is reduced with the total extract in chaotropic and reducing agents. However, it can be increased when the proteins are hydrolyzed by proteases. Fermentation involving certain species of Fructolactobacilli (FLB), followed by baking, decreases the amount of ATIs and IgE binding to ATIs. In yeast-fermented bread, the amount of ATIs decreased in a similar manner, but IgE binding was more prominent, indicating that there was a modification of ATIs that affected the epitope recognition. When isolated ATIs are ingested with high ATI degrading FLB, the immune response in mice is less elevated in vivo , when compared with ATI without high ATI degrading FLB. The pathogenic effects on the skin of dogs and one wheat-allergic child are also decreased when soluble proteins or isolated ATIs are reduced with the thioredoxin/thioredoxin reductase NADPH system. Glycation on the other hand has been shown to potentiate the allergenic properties of ATIs as evidenced by the large increase in IgE binding. The impact of food processing on the pathogenic properties of ATIs is hardly studied in vivo in humans. There seem to be opportunities to mitigate the pathogenic properties in vitro , but potentiation of pathogenic properties is also frequently observed. The risk assessment of heavy metals (HMs) in sewage sludge (SS) is essential before land application. Six HMs in nineteen SS collected in the Yangtze River Delta were analyzed to assess risks to environment, ecosystem, and human health. HMs concentrations were ranked in the order of Zn > Cu > Cr > Ni > Pb > Cd, with Cu, Zn, and Ni in a total of 16% of samples exceeding the legal standard. Zn showed greatest extractability according to EDTA-extractable concentrations. HMs in 16% of SS samples posed heavy contamination to the environment with Zn as the major pollutant. HMs in 26% of samples posed ecological risk to the ecosystem and Cd was the highest risky HM. The probabilistic health risk assessment revealed that HMs posed carcinogenic risks to all populations, but non-carcinogenic risks only to children. Background: The wine sector is constantly evolving, in order to adapt to consumer tastes. The organoleptic characteristics in wines are the main factors to obtain quality wines. Proanthocyanidins (PAs) are responsible in an important way for positive aspects in quality wines, such as body and color stability in red wines, but they are also responsible for sensory characteristics that can be negative for their quality when found in excessive concentrations. One strategy to improve grapevines and wines is to obtain new varieties, so our research institute has been selecting some of them from direct crosses between Monastrell and other considered premium varieties such as Cabernet Sauvignon and Syrah., Results: A quantitative analysis in grapes, seeds and wines was carried out during three consecutive seasons (2018, 2019 and 2020) in order to characterize PAs composition and concentration in the following new varieties: MC80 (Monastrell × Cabernet Sauvignon), MC98, MC4, MC18 and MS10 (Monastrell × Syrah). Other aspect to study was the extraction capacity of the different new varieties of PAs during maceration process into must/wine., Conclusion: In general, the results showed higher concentrations in PAs in most crosses for the three seasons studied compared to Monastrell variety. Hibiscus sabdariffa is a tropical plant with red calyxes whose anthocyanins, phenols, and antioxidant activity make it attractive to consumers both from a nutritional and medicinal standpoint. Its seasonality, perishability, and anthocyanin instability, led to the setup of stabilization methods comprising drying and powdering. However, its properties can often be altered during these stabilization processes. Treatments such as dehumidified-air-drying, infrared drying, and oven-drying, and their combination showed better quality preservation. Moreover, powder production enables superior biomolecule extractability which can be linked to a higher bioaccessibility. However, the required temperatures for powder production increase the bioactive molecules degradation leading to their antioxidant activity loss. To overcome this issue, ambient or cryogenic grinding could be an excellent method to improve the biomolecule bioavailability and accessibility if the processing steps are well mastered. To be sure to benefit from the final nutritional quality of the powder, such as the antioxidant activity of biomolecules, powders have to offer excellent reconstitutability which is linked to powder physicochemical properties and the reconstitution media. Typically, the finest powder granulometry and using an agitated low-temperature reconstitution media allow for improving anthocyanin extractability and stability. In this review, the relevant physicochemical and processing parameters influencing plant powder features from processing transformation to reconstitution will be presented with a focus on bioactive molecules and antioxidant activity preservation.

Composting can turn organic waste into a valuable soil amendment that can improve physical, chemical, and biological soil quality. Compost amendments can also contribute to the remediation of areas anthropogenically degraded by metals. However, it is well known that compost, particularly self-produced compost, can show enrichment in metals. An experimental study was conducted to examine the short- and long-term distribution and the mobility of metals in soils amended with a self-produced compost when it was added alone or in combination with different doses of a natural zeolite to soil. The aim was also to study the interest of managing moderately metal-contaminated kitchen garden soils by assessing the chemical extractability, phytoavailability, and oral bioaccessibility of metals. When zeolite was added to compost alone, it had the tendency to better reduce extractability of Cd and Zn at 25%, and those of Pb at 15%. When the self-produced compost alone or in co-application with zeolite at these doses was applied to soils, the results showed (1) a decrease of NH 4 NO 3 -extractable Zn; (2) a reduction of Pb environmental availability, but not Pb bioaccessibility, and (3) an increase of ryegrass biomass. Nevertheless, the risk posed by the self-produced compost was minimal when applied at the proper rate (0.6% w/w). In the selected experimental conditions, the study recommends that self-produced compost be mixed with 15% zeolite to maximize vegetal biomass and minimize environmental risk. Organic waste comprises a large amount of hydrocarbon containing organic substances, which is regarded as a potential resource rather than simply a waste. A field experiment was conducted in a poly-metallic mining area to investigate the potential of organic waste to facilitate the soil remediation process. Different organic wastes and a commonly used commercial fertilizer were added to heavy metal contaminated soil, which was under phytoremediation using the As hyperaccumulator Pteris vittata. The influence of diverse fertilizer regimes on the biomass of P. vittata and heavy metal removal by P. vittata, was investigated. The soil properties were analyzed after the application of phytoremediation with or without the addition of organic wastes. Results indicated that sewage sludge compost is an appropriate amendment to improve the phytoremediation efficiency. Compared to the control, the application of sewage sludge compost significantly reduced the extractability of As in soil by 26.8 %, and increased the removal of As and Pb by 26.9 % and 186.5 %, respectively. The highest removal of As and Pb reached 33 and 34 kg/ha, respectively. The sewage sludge compost-strengthened phytoremediation improved soil quality. And the diversity and richness of the bacterial community were improved, as represented by the increase in Shannon and Chao index. The establishment of operation protocols for olive oil (OO) extraction at non-industrial scale is crucial for research purposes. Thus, the present study proposes a simple and cost-effective method for OO extraction at the laboratory scale (LS) level. To validate the proposed methodology, industrial OO extraction (IS) was performed in parallel, using the same cultivars ‘Galega vulgar’ (GV), ‘Cobrançosa’ (COB) and ‘Arbequina’ (ARB) collected from the same orchards, within the same period. Obtained results showed highest extractability for COB and ARB, of about 53%, while GAL showed 50%. All produced OO showed values lower than the regulated limits for the physicochemical parameters (acidity, K232, K268 and ΔK), classifying them as extra virgin OO (EVOO). Highest total phenolic content was observed for COB, with no significant differences (p-value > 0.05) between extraction methods. Regarding fatty acid composition, oleic acid (C18:1) showed the lowest percentage for ARB, with about 66% and 68%, for LS and IS, respectively, and the highest for GV with about 72% for both LS and IS. Furthermore, all samples from both extraction methods were compared to the European Community Regulation, with fatty acid composition within the regulated levels for EVOO. This work showed promising results regarding extraction yields and OO extractability, as well as its quality parameters.

The proportion of seed tannins in wine is higher than that of skin tannins during normal winemaking practices. The interaction between grape skin cell wall and seed tannin could alter seed tannin extraction and wine tannin profiles during winemaking. However, the influence of grape skin cell wall ripeness in the interaction is still unclear. The purpose of this work was to study the influence of grape skins cell wall ripeness on seed tannins extraction and the final wine tannin profiles during alcoholic fermentation. The percentage of seed tannins extraction increases according to the skin cell wall maturity, and the higher the mDP of tannins, the larger the difference of tannins extraction. During alcoholic fermentation, seed tannins are adsorbed to the skin cell wall and desorb gradually into wine. For the final wine, cell wall maturity was positively correlated with tannins concentration and negatively correlated with G%. In this study, we evaluated the extractability of three curcuminoids (curcumin, demethoxycurcumin, and bisdemethoxycurcumin) from turmeric powder in several solvents using high-performance liquid chromatography (HPLC) with the diode-array detection method. These solvents include water, milk (homogenized, 2% reduced fat, low fat, fat free, soy, almond, coconut, and milkadamia), and aqueous ethanols (0%, 4%, 10%, 20%, 30%, 40%, 50%, and 100%). Ambient water was able to extract only 0.55 mg/g of curcuminoids, whereas warm water extracted more than four-fold higher amounts (2.42 mg/g). Almond, coconut, and milkadamia milk were able to extract only small amounts of curcuminoids at ambient temperatures (0.01-0.07 mg/g). The extractability of curcuminoids in these milk types did not improve, even in warm conditions (0.08-0.37 mg/g). Whereas dairy and soy milk extracted 6.76-9.75 mg/g of curcuminoids under ambient conditions, their extractability increased significantly in warm conditions by 30-100% higher (11.7-14.9 mg/g). The solubility of curcuminoids also varied remarkably in different proportions of aqueous-alcohol mixtures. With 4% ethanol, only 1.7 mg/g of curcuminoids were extracted, and the amounts improved with the increase in ethanol content up to 50% (32.2 mg/g), while 100% ethanol extracted a similar amount as 50% ethanol (34.2 mg/g). This study suggests that the extractability of curcuminoids from turmeric will be dependent on the type of diets consumed with the turmeric supplements.

Grape cell wall polysaccharides influence the extraction of phenolic compounds during winemaking and consequently polyphenol concentrations in the final wine. During ripening, both compound groups undergo pronounced structural and compositional changes, resulting in a dynamic change of extractability. Grape cell wall polysaccharides from different ripe grapes were added to fermentations of Cabernet Sauvignon and Pinot noir grapes. Polyphenol-polysaccharide interactions affected the concentrations of tannins and monomeric flavanols in the wines depending on the maturity of the added polysaccharides. With higher polysaccharide maturity, the effects became more pronounced. Polysaccharides protected monomeric flavanols and tannin in Pinot noir, thereby increasing the concentrations, but they precipitated or masked these compounds in Cabernet Sauvignon. The added polysaccharides affected the concentrations in anthocyanins and polymeric pigments much less compared to the ripening status of the grapes. It was concluded that structural changes of polysaccharides during ripening affect the extraction of tannins and monomeric flavanols the most.

The presence of engineered nanoparticles (ENPs) in soil gradually increases, among others due to the nano-agrochemicals application. So far, the co-existence of different ENPs in soil is poorly examined. Here, the metal extractability and toxicity of soils spiked (300 mg kg -1 ) singly and jointly with Zn- and Cu-based ENPs or metal salts were tested. The samples were aged for 1 and 90 days. The predicting available metal component of ENPs concentrations were determined by different methods including soil pore water collection and batch extractions with H 2 O, CaCl 2 or DTPA. Survival and reproduction of Folsomia candida were also evaluated. The combined effect of ENPs on the extractability of metals was mainly found with DTPA characterized by the highest leaching capacity among the used extractants. In fresh soil, the mixtures of ENPs differentiated only DTPA-extractable Cu level, while aging resulted in changes in both Zn and Cu concentrations leached by CaCl 2 or DTPA. However, the character of the combined effect was an ENPs- and soil type-dependent, whereas the mixtures of metal salts mostly provided higher Zn and Cu recovery than the individual compounds. The pattern of co-toxicity of metal-oxide ENPs was also time-dependent: the antagonistic and synergistic effect was observed in the samples after 1 and 90 days, respectively. However, the toxicity was weakly related with extractable concentrations in both single and joint treatment of metal compounds. The co-existence of engineered nanoparticles (ENPs) in the environment is an emerging issue remaining poorly investigated. The present study aimed at analyzing the fate of binary mixtures of CuO and ZnO ENPs in a soil-plant system. The ENPs were singly or jointly dosed into soil at 300 mg kg -1 and aged for 7 and 30 days. To evaluate nano-specific effects, individual and combined treatments of metal salts were also applied. Interactions between ENPs and soil-grown barley Hordeum vulgare were determined in terms of biomass, plant mineral composition as well as expression of genes regulating metal homeostasis (ZIP1,3,6,8,10,14, RAN1, PAA1,2, MTP1, COPT5) and detoxification (MT1-3). The bioavailability of Zn and Cu in bulk soil and in the rooting zone was determined using the 0.01 mol L -1 CaCl 2 extraction. After combined treatment of ENPs, the extractable concentrations of Cu and Zn were lower than upon individual exposure in bulk soil. The opposite tendency was noted for metal salts. Genes related to metal uptake (ZIP) and cellular compartment (PAA2, RAN1) were mostly up-regulated by single rather than combined application of ENPs. The objective of this study is to assess the effects of installation and operation of a high-power ultrasound machine (HPU) for the treatment of olive paste by using ultrasound technology in order to evaluate the best way installation and the best definition of the operating conditions of the machine. The study was conducted installing in an industrial olive oil mill a continuous processing ultrasound machine, which used a frequency of 20 kHz able to work at 3200 kg h -1 as feed capacity. Checking of performance has been carried out by the assessment of the different operating and process conditions, assessing in particular the impact of the ultrasound treatment before and after the malaxation phase on performance indicators of the continuous olive oil plant (plant extractability, olive paste rheological characteristic) and on selected chemical properties of the olive oil extracted (quality parameters, antioxidant content, and volatile profiles). In the tested conditions, high-power ultrasound treatment did not produce significant effect on the legal parameters (free acidity, peroxide index and spectrophotometric indexes), while a significant increase in the content of phenolic compounds was generally observed; higher enhancements were more evident when the high-power ultrasound treatment was carried out before the malaxation phase., (Copyright © 2021 The Author(s). The bioavailability and bioaccumulation of metal-based engineered nanoparticles (ENPs) in soils need to be evaluated in environmentally relevant scenarios. The aim of this study was an analysis of potentially available metal-component ENPs (nano-ZnO and nano-CuO) in soils. Earthworms (Eisenia fetida) were used to examine the bioaccumulation potential of ENPs. Micro-particles (micro-ZnO and micro-CuO) and metal salts (ZnCl 2 and CuCl 2 ) were used to evaluate the nano-effect and the activity of dissolved ions, respectively. Zn- and Cu-compounds were added to sandy loam and silt loam at a concentration of 10 mg kg -1 . The bioavailable fractions of metals were extracted from soil using H 2 O, MgCl 2 with CH 3 COONa or EDTA. EDTA was the most effective extractant of Zn and Cu (10.06-11.65 mg Zn kg -1 and 2.69-3.52 mg Cu kg -1 ), whereas the H 2 O-extractable metal concentration was at the lowest level (1.98-2.12 mg Zn kg -1 and 0.54-0.82 Cu mg kg -1 ). The bioavailable metal concentrations were significantly higher in silt loam than sandy loam soil, which was related to the higher pH value of silt. There were no significant differences between the Zn content in the earthworms incubated in the two soils, which may confirm the auto-regulation of the Zn content by earthworms. However, the bioaccumulation of Cu was strongly correlated with the extractable Cu concentrations. The juvenile earthworms accumulated Cu and Zn more than adults. Sandy soils in Florida are vulnerable to toxic metal pollution, and it is necessary to identify desirable amendments for the remediation of metal contaminated soils. Sorption and incubation experiments were conducted to compare the effectiveness of dolomite phosphate rock (DPR), humic acid activated dolomite phosphate rock (ADPR) and biochar (BC) in immobilizing Cd 2+ and Pb 2+ in two representative agricultural soils in south Florida (Alfisol-Riviera and Spodosol -Ankona series). The results showed that the soils had a low sorption capacity for metals with maximum sorption of 0.767-3.30 mg/g. Application of amendments increased the maximum sorption by 4.2-4.8 times for Pb 2+ and 1.5-2.2 times for Cd 2+ in Alfisol soil, and 7.1-7.9 times for Pb 2+ and 1.7-3.1 times for Cd 2+ in Spodosol soil. ADPR was the most effective amendment for increasing the soil's sorption capacity for Cd 2+ and Pb 2+ . 0.01 M CaCl 2 extractable metals in the contaminated soils were significantly decreased by all the amendments, especially ADPR, which reduced extractable Cd 2+ and Pb 2+ by 87.2 and 76.0% in Alfisol and 91.3 and 76.3% in Spodosol soil as compared to control. The amounts of extractable Cd 2+ and Pb 2+ were negatively correlated with soil pH and available P, indicating that the change of soil characteristics by amendments was the dominant mechanism for enhanced immobilization of metals in the contaminated soils. The interaction between multi-walled carbon nanotubes (MWCNTs) and soil heavy metals was rarely studied. With the convenience of detecting multiple metal elements by ICP-AES, this paper examined the potential effectiveness of MWCNTs on extractability of antimony (Sb) and cadmium (Cd) in contaminated soil. Three-step sequential extraction procedure, toxicity characteristic leaching procedure, bioaccessibility and CaCl 2 single extraction were employed to evaluate Sb and Cd speciations and their extractabilities. According to our results, only at low Sb content level of 100 mg/kg, antimony bioavailability reduced with MWCNTs addition of 0.3% and 0.9% by 22.97% and 20.74%, respectively, which might due to the increase of adsorption point, nevertheless, the excess Sb(OH) 6 - -extractable Sb and Cd, no significant, continuous, regular extractability pattern were found. Overall, MWCNTs were selective on extractability of soil heavy metals due to mechanisms of physical adsorption. The study focused on two grass species Holcus lanatus and Agrostis capillaris abundant in the sites of former As mining and processing in the Sudetes. Arsenic uptake from soils was examined to assess a risk associated with its accumulation in grass shoots and to check its dependence on soil fertilization. The research involved a field study and greenhouse experiment. In the field study, soil and plant samples were collected from 33 sites with 72-98,400 mg/kg total soil As. Arsenic uptake by grasses differed widely. Both species indicated a strategy typical for eliminators, although As concentrations in more than 50% of the shoot samples exceeded 4 mg/kg, a maximum permissible value for fodder. In the greenhouse experiment, commercial cultivars of both species were grown in five soils containing 394-19,600 mg/kg, untreated and fertilized. All seedlings died in the soil with highest total As, and considerable phytotoxicity was observed in other soils, particularly in nonfertilized ones. Fertilization resulted in the improvement of plant growth and reduction of As uptake except for Agrostis capillaris fertilized with manure. Further research should focus on identifying tolerant genotypes growing in extremely enriched sites and analysis of factors that will efficiently reduce As phytoaccumulation.

The extraction of seed and skin tannins in wine has been investigated at three different grape maturity stages. For that, the tannins content and composition of seeds and skins at three different maturity stages were characterized. After that, an original approach of nanovinification was conducted. At each maturity stages, three winemaking modalities have been produced: (i) a control modality, (ii) a seed modality made of exclusively with seed and (iii) a skin modality made of exclusively with skins. The aim of this work is to describe and explain the seed tannins kinetics release in wine but also the impact of grape maturity on seed tannins extractability. The uptake of As by various plants growing in highly enriched sites was examined in order to identify potential As accumulators and to assess the risk associated with As presence in plant shoots. Representative samples of 13 plant species, together with soil samples, were collected from various sites affected by historical As mining: mine and slag dumps, tailings and contaminated soils with As concentrations in a range 72-193,000 mg/kg. Potentially and actually soluble As forms, extracted with 0.43 M HNO 3 and, 1M NH 4 NO 3 were examined in relation to As concentrations in plant roots and shoots. The latter differed strongly among the species and within them and were in the ranges 2.3-9400 mg/kg and 0.5-509 mg/kg, respectively. The majority (over 66%) of plant samples had As shoot concentrations above 4 mg/kg, an upper safe limit for animal fodder. The uptake of As by plants correlated well with total and extractable soil As, though As concentrations in plants could not be predicted based on soil parameters. Equisetum spp. and C. epigejos indicated a particularly strong accumulation of As in shoots, while A. capillaris , and H. lanatus showed a limited As root-to-shoot transfer, apparently associated with species-related tolerance to As., Competing Interests: The authors declare no conflict of interest.

Dose-response (toxicity) relationships explain how the response changes with exposure doses. However, the results of tests or observations are commonly based on total concentrations of contaminants in environments, not the exposure dose that causes toxicity. In the present study, the copper (Cu) phytotoxicity to barley root elongation was studied in 17 representative Chinese soils. Also, the EDTA-extractable Cu concentration was used to evaluate the extractability of Cu in soils. The results showed that the concentrations of EDTA-extractable Cu accounted for 89.6-91.2 % of total added Cu in soils and that soil pH, organic carbon content (OC) and cation exchange capacity (CEC) could explain over 85 % of the variance in Cu phytotoxicity thresholds based on EDTA-extractable concentration. The integrated relationship of EDTA-extractable Cu doses, toxicity, and soil properties was firstly derived quantitatively. The EDTA-extractable Cu plus soil properties can explain more than 90 % of the variance in the toxicity response of barley root elongation. The increasing release of metallic nanoparticles (NPs) or their sulfidized forms into soils have raised concerns about their potential risks to soil ecosystems. Hence, there is a need for novel strategies to remediate metallic NPs pollution in soils. In this study, to explore the feasibility of using earthworm Eisenia fetida to manage soils contaminated with metallic NPs, we simultaneously investigated the chronic soil toxicities of ZnO NPs and ZnS NPs to E. fetida, and the effects of E. fetida on Zn extractability in soils amended with ZnO NPs and ZnS NPs. After a 28 d exposure, survival rate and weight loss of earthworms were not impacted by either ZnO NPs or ZnS NPs at a concentration of 400 mg Zn per kg soil. Further, while ZnO NPs activated earthworm antioxidative system, ZnS NPs resulted in significant alleviation of oxidative damage in earthworm. The presence of earthworms significantly decreased the bioavailability of Zn in ZnO NPs contaminated soil, whereas significantly increased the bioavailability of Zn in ZnS NPs contaminated soil. Higher soil pH and electrical conductivity (EC) were suspected to result in higher extractability and bioavailability of benzo[a]pyrene (B[a]P) in soils. In this study, we investigated the influence of pH, EC and ageing on the extractability of B[a]P in two contracting soils (varied largely in soil texture, clay mineralogy and organic carbon content) over 4 months. Dilute sodium hydroxide (0.2 mol L -1 ) and sodium chloride (0.1 mol L -1 ) solutions were used to adjust soil pH and EC either separately or simultaneously. Extractability of B[a]P in these soils was monitored using a mild solvent extraction using butanol (BuOH, end-over-end shake over 24 h), and an exhaustive mix-solvent extraction using dichloromethane/acetone (DCM/Ace, v:v = 1:1) facilitated by sonication and a subsequent NaOH saponification method following the DCM/Ace extraction. Results showed that increased pH and/or EC significantly increased the B[a]P extractability in the sandy soil (GIA). Variance analysis of contribution of pH and/or EC modification and ageing time on changes in B[a]P extractability indicated that in GIA > 55% and over 25% of the changes in B[a]P extractability was attributed to increased pH&EC and pH only respectively. While ageing resulted in >85% of the change in B[a]P extractability in the clayey soil (BDA), following by increased pH&EC (contribution < 15%). Large amount of non-extractable residue (NER) were formed over the ageing period, up to 95% and 79% in GIA/BDA and its modified soils, respectively. Significant correlations were observed between B[a]P BuOH extractability and the exhaustive sequential extraction using DCM/Ace followed by NaOH saponification for all soils (p < 0.001). The effects of temperature and ethanol concentration on the kinetics of anthocyanin adsorption and desorption interactions with five cell wall materials (CWM) of different composition were investigated. Using temperatures of 15 °C and 30 °C and model wine with ethanol concentrations of 0% and 15% ( v / v ) over 120 min, the adsorption and desorption rates of five anthocyanin-glucosides were recorded in triplicate. Small-scale experiments were conducted using a benchtop incubator to mimic a single berry fermentation. Results indicate that more than 90% of the adsorption occurs within the first 60 min of the addition of anthocyanins to CWM. However, desorption appears to occur much faster, with maximum desorption being reached after 30 min. The extent of both adsorption and desorption was clearly dependent not only on temperature and ethanol concentration but also on the CWM composition.

This study evaluated the relationship between cell wall breakdown, from Shiraz grapes harvested at three different ripeness levels and the colour and phenolics extracted during alcoholic fermentation into wines. Phenolic differences between the ripeness treatments were minimal after ¼ of the fermentation was completed. However, colour and phenolic content were significantly higher in finished wines made from 25°Brix grapes compared to those from 21°Brix and 23°Brix. Levels of grape cell wall polysaccharide deconstruction during fermentation was a determining correlative factor in relation to phenolic extractability. In this context, the de-pectination observed during ripening was found to enhance this deconstruction or "opening-up" of the grape pomace during fermentation, thus increasing the differential extraction of specific polyphenols, especially polymeric polyphenols, into the wines. Additionally, the degree of cell wall deconstruction seemed to play a role in the possible retention and extraction of specific grape proanthocyanidins, depending on their nature and polymer length., (Copyright © 2018. Two fish parvalbumin models were established to study relationships among matrix effect, extractability, and thermostability during in vitro immunodetection using two parvalbumin-specific monoclonal antibodies (3E1 and PARV19). Our results illustrated that matrix-induced thermal instability of parvalbumin was due mainly to physical (hydrophobic effect) and chemical (thiol-disulfide interchange) interactions. The addition of sodium dodecyl sulfate (SDS, surfactant), β-mercaptoethanol (reducing agent) or ethylenediaminetetraacetic acid (EDTA, metal chelator) during sample preparation could not only increase the extractability of parvalbumin but also enhanced its immunodetection. Our findings demonstrated excess EDTA completely chelated Ca 2+ in parvalbumin and rendered it undetectable using PARV19 (a Ca 2+ -dependent antibody). Overall, our resulted showed that matrix effect on in vitro analyte quantification cannot be underestimated. The aim of this study was to evaluate the use of drinking-water treatment residuals (DWTR) in the amendment of a soil affected by mining activities (Aljustrel mine, Portuguese sector of the Iberian Pyrite Belt), considering the effects on its chemical, biochemical and ecotoxicological characteristics. The DWTR had neutral characteristics (pH 6.7) and an organic matter (OM) content of 575 g kg -1 dry matter (DM), which makes them a potential amendment for the remediation of mine degraded soils, as they may correct soil acidity and reduce the extractable metal fraction. An incubation assay, with soil and DWTR, with or without lime, was carried out to test the doses to be used in the assisted-phytostabilization experiment. Based on the results obtained, the doses of DWTR used were the equivalent to 48, 96, and 144 t DM ha -1 , with and without lime application (CaCO 3 11 t DM ha -1 ). Agrostis tenuis Sibth was used as the test plant. Some amendments doses were able to improve soil characteristics (pH and OM content), to decrease metal extractability by 0.01 M CaCl 2 (especially for Cu and Zn), and to allow plant growth, that did not occur in the non-amended soil. Copper, Pb and Zn concentrations in the plant material were lower than the maximum tolerable level for cattle feed, used as an indicator of risk of entry of those metals into the human food chain. The simultaneous application of DWTR (96 and 144 t ha -1 ), with lime, allowed a reduction in the mine soil ecotoxicity, as evaluated by some lethal and sub-lethal bioassays, including luminescence inhibition of Vibrio fischeri, Daphnia magna acute immobilization test, mortality of Thamnocephalus platyurus, and 72-h growth inhibition of the green microalgae Pseudokirchneriella subcapitata. However, DWTR were unable to increase soil microbial activity, evaluated by dehydrogenase activity, an important soil-health indicator. Also, OM content and N Kjeldahl , concentrations increased slightly but remained low or very low (P and K extractable concentrations were not affected). In general, the bioassays highlighted a decrease in soil ecotoxicity with the presence of lime and DWTR (144 t DM ha -1 ). The Fukushima Daiichi nuclear power plant accident caused serious radiocesium ( 137 Cs) contamination in soils in a range of terrestrial ecosystems. It is well documented that the interaction of 137 Cs with soil constituents, particularly clay minerals, in surface soil layers exerts strong control on the behavior of this radionuclide in the environment; however, there is little understanding of how soil aggregation-the binding of soil particles together into aggregates-can affect the mobility and bioavailability of 137 Cs in soils. To explore this, soil samples were collected at seven sites under different land-use conditions in Fukushima and were separated into four aggregate-size fractions: clay-sized (<2 μm); silt-sized (2-20 μm); sand-sized (20-212 μm); and macroaggregates (212-2000 μm). The fractions were then analyzed for 137 Cs content and extractability and mineral composition. In forest soils, aggregate formation was significant, and 69%-83% of 137 Cs was associated with macroaggregates and sand-sized aggregates. In contrast, there was less aggregation in agricultural field soils, and approximately 80% of 137 Cs was in the clay- and silt-sized fractions. Across all sites, the 137 Cs extractability was higher in the sand-sized aggregate fractions than in the clay-sized fractions. Mineralogical analysis showed that, in most soils, clay minerals (vermiculite and kaolinite) were present even in the larger-sized aggregate fractions. These results demonstrate that larger-sized aggregates are a significant reservoir of potentially mobile and bioavailable 137 Cs in organic-rich (forest and orchard) soils. Near infrared hyperspectral data were collected for 200 Syrah and Tempranillo grape seed samples. Next, a sample selection was carried out and the phenolic content of these samples was determined. Then, quantitative (modified partial least square regressions) and qualitative (K-means and lineal discriminant analyses) chemometric tools were applied to obtain the best models for predicting the reference parameters. Quantitative models developed for the prediction of total phenolic and flavanolic contents have been successfully developed with standard errors of prediction (SEP) in external validation similar to those previously reported. For these parameters, SEPs were respectively, 11.23 mg g -1 of grape seed, expressed as gallic acid equivalents and 4.85 mg g -1 of grape seed, expressed as catechin equivalents. The application of these models to the whole sample set (selected and non-selected samples) has allowed knowing the distributions of total phenolic and flavanolic contents in this set. Moreover, a discriminant function has been calculated and applied to know the phenolic extractability level of the samples. On average, this discrimination function has allowed a 76.92% of samples correctly classified according their extractability level. In this way, the bases for the control of grape seeds phenolic state from their near infrared spectra have been stablished.

Oat bran was fermented by filamentous fungi to improve the extractability of β-glucan. Box-Behnken experimental design and response surface methodology were used to obtain the maximum extractability of β-glucan. Inoculum volume, fermentation temperature, and time were evaluated as three variables. Under optimal fermentation conditions, the extractability of β-glucan in oat bran increased to 45.57 ± 1.82% and 51.10 ± 2.32% by Aspergillus niger and Rhizopus oryzae, respectively. The extractability was about 3-fold of that before fermentation (16.86 ± 0.76%). Fermentation of oat bran by Aspergillus niger and Rhizopus oryzae reduced the molecular weight of the extracted β-glucan from 6.74 × 10 5 to 2.84 × 10 5 and 2.20 × 10 5  Da, respectively. Correspondingly, the apparent viscosity of the extracted β-glucan decreased after fermentation. But the increased extractability compensated for the reduced molecular weight in the formation of viscous solution. The effects of 137 Cs deposit forms on its ageing in soil have not yet been reported. Soluble and Solid 137 Cs input forms were mixed with the mineral soils collected under Fukushima's coniferous and broadleaf forests, incubated under controlled laboratory, and examined the evolution of 137 Cs availability over time. Results show that the extracted 137 Cs fraction with water was less than 1% for the soluble input form and below detection limit for the solid input forms. Likewise, with an acetate reagent, the extracted 137 Cs fraction ranged from 46 to 56% for the soluble input and from 2 to 15% for the solid input, implying that the nature of the 137 Cs contamination strongly influences its extractability and mobility in soil. Although the degradation of organic materials was apparent, its impact on the 137 Cs extractability was found to be weak. Nevertheless, more Ac-available 137 Cs was obtained from broadleaf organic material mixes than the coniferous counterparts, suggesting that the lignified nature of latter tend to retain more 137 Cs. When extrapolated to a field context, more available 137 Cs fraction may be expected from wet-derived contaminated forest soils than contaminated via solid-derived inputs. Postharvest withering of grapes strongly affects the content and extractability of phenolic compounds in the production of sfursat, fortified and passito wines. This work evaluated the effectiveness of enzymes applied individually and/or in multi-enzyme blends, on the extraction of anthocyanins, oligomeric flavanols and polymeric flavanols from withered grape skins during simulated maceration. The study was performed on Vitis vinifera L. cv. Nebbiolo and Barbera because of their different skin phenolic profile and cell wall composition. Our findings highlight that the relationship between skin mechanical properties (berry skin break force and energy) and extraction yield of phenolic compounds is variety dependent. Significant correlations were found between the skin softening associated with cell wall degradation and the extraction of anthocyanins and flavanols in Nebbiolo, for which polygalacturonase, individually or in multi-enzyme blends, plays an important role. Background: Double maturation raisonnée (DMR) is a potential canopy measure that affects grape and wine composition. The aim of this work was to study for the first time the DMR impact on the physical, biochemical and sensorial characteristics of the berries and wines of Merlot, one of the world's fastest-expanding grapevine varieties., Results: DMR significantly increased the content of soluble solids (1.2-fold), free amino nitrogen (1.8-fold) and acidity in berries but decreased the weight of 100 berries on harvest (approx. 28%). Irrespective of the vintage, DMR-treated grapes had a significantly higher content of non-astringent tannins (0.73-0.78 mg L -1 ) and anthocyanin extractability (34.7-36.4%) but a lower index of astringency (31.2-33.7) when compared to the control. Consequently, the DMR wines achieved higher alcohol, total acidity and extract, hydroxycinnamic acids, flavanol and flavonol contents, whereas the content of anthocyanins was similar to that of the control. Sensorial evaluation showed that DMR wines were not rated higher and would not be appreciated more than control wines., Conclusion: Changes in berries during DMR altered the wine characteristics only in terms of primary metabolites. A reduced accumulation of phenolics, especially anthocyanin content, in the berry skin of DMR-treated grapes was not reflected in their presence in wines. Phenanthrene (Phe) and pentachlorophenol (PCP) are classified as persistent organic pollutants and represent serious concern for the environment as they are toxic and ubiquitous. Biochar based remediation is an emerging technology used in water and soil contamination. In this study we used poplar (BP) and conifer (BC) biochars to remediate water and soil contaminated by Phe and PCP. BP and BC were able to remove completely either Phe or PCP from contaminated water within one to three days. When biochar was confined in a porous membrane, BC and BP maintained their sorption efficiency for several remediation cycles. However, in these conditions BC allowed faster Phe removal. In soil remediation experiments, addition of two biochar rates, i.e. 2.5 and 5 mg g -1 , strongly reduced Phe extractability (up to 2.7% of the initially added Phe with the larger BC dose). This was similar to the behavior observed when compost was applied in order to verify the role of soil organic matter in the fate of both contaminants. PCP extractability was reduced only up to 75% (in average) in all samples including those with compost amendment. Recently the use of ozone as sanitizing agent has been proposed on winegrapes in order to control mycobiota after harvest. The aim of this work was to investigate possible indirect physico-chemical effects of ozone treatment on berry skin phenolic composition and extractability. Vitis vinifera L. cv Nebbiolo and Barbera, chosen for their different anthocyanin profiles, were post-harvest treated for 24 and 72h with gaseous ozone (30μL/L). Skin anthocyanin and flavanol extractability was assessed during maceration (6, 24, 48, 96, 168 and 240h) using a wine-like solution. In our experimental conditions, ozone did not affect significantly the final extraction yield of anthocyanins (TA), proanthocyanidins (PRO), and flavanols reactive to vanillin (FRV) in Barbera, although TA and FRV extractabilities were higher in control samples than in ozone-treated samples during the first stages of maceration. In Nebbiolo, the final TA extraction yield was positively influenced by the ozone treatment (68.6, 64.2, and 59.9% for 24h ozone-treated berries, 72h ozone-treated berries and control samples, respectively). Final PRO and FRV extractability also increased in both ozone-treated samples compared to the control (+8.6-9.1% for PRO and +7.3-11.7% for FRV). No significant differences were found among treatments for individual anthocyanins in both cultivars at the end of maceration. Therefore, the use of ozone as sanitizing agent in red varieties prior to winemaking process can be considered because it did not negatively affect the extractability of skin anthocyanins and flavanols., (Copyright © 2016 Elsevier Ltd. The hard-to-cook (HTC) defect in legumes is characterized by the inability of cotyledons to soften during the cooking process. Changes in the non-starch polysaccharides of common bean seed coat and cotyledon were studied before and after development of the HTC defect induced by storage at 35°C and 75% humidity for 8months. Distinct differences in the yields of alcohol insoluble residues, degree of methoxylation (DM), sugar composition, and molar mass distribution of non-starch polysaccharides were found between the seeds coat and cotyledons. The non-starch polysaccharide profiles, both for seed coats and cotyledons, significantly differed when comparing HTC and easy-to-cook (ETC) beans. In conclusion, differences in the structure, composition and extractability of non-starch polysaccharides between the ETC and HTC beans confirmed the significant role of pectin polysaccharides in interaction with divalent ions in the HTC development, which consequently affect their cooking behaviors., (Copyright © 2016 Elsevier Ltd. The rate of tannin extraction was studied in a vinification of red grapes and the results compared with another vinification made with white grapes fermented as for typical red wine, in the presence of skins and seeds. Even though the grapes presented a quite similar skin and seed tannin content, the differences in tannin concentration between both vinifications was very large, despite the fact that the only apparent difference between the phenolic composition of both wines was the anthocyanin content. This suggests that anthocyanins play an important role in tannin extractability, perhaps because they affect the extent of the tannin-cell wall interaction, a factor that largely controls the resulting quantity of tannins in wines. To confirm this observation, the effect of anthocyanins on the tannin extractability from grape seeds and skin and on the interaction between tannins and grape cell walls suspended in model solutions were studied. A significant fraction of pesticides sprayed on crops may be returned to soils via plant residues, but its fate has been little documented. The objective of this work was to study the fate of glyphosate associated to plants residues. Oilseed rape was used as model plant using two lines: a glyphosate-tolerant (GT) line and a non-GT one, considered as a crucifer weed. The effects of different fragmentation degrees and placements in soil of plant residues were tested. A control was set up by spraying glyphosate directly on the soil. The mineralization of glyphosate in soil was slower when incorporated into plant residues, and the amounts of extractable and non-extractable glyphosate residues increased. Glyphosate availability for mineralization increased when the size of plant residues decreased, and as the distribution of plant residues in soil was more homogeneous. After 80 days of soil incubation, extractable (14)C-residues mostly involved one metabolite of glyphosate (AMPA) but up to 2.6% of initial (14)C was still extracted from undecayed leaves as glyphosate. Thus, the trapping of herbicides in plant materials provided a protection against degradation, and crops residues returns may increase the persistence of glyphosate in soils. During the extraction, a portion of oil remains trapped inside the cells and its release requires the degradation of the walls and cell membranes, especially when the fruits have not reached a maximum maturity which is likely to cause an optimal embrittlement of the parietal structures and cell membrane. This can be done by specific enzymes necessary for the degradation of various cellular barriers. Three different enzyme treatments proteolytic, pectolytic or both are applied on the Moroccan Picholine olives from veraison to maturity of the fruit. The effect of these treatments is evaluated by olive oil diffusion, its phenolic content (PC) and cellular embrittlement determination of olives during ripening. The pectolytic activities lead to a significant increase in both the oil extractability (76 % at veraison and 14 % at maturity) and the PC (up to 50 % of gain compared to the control at veraison and 27 % at maturity). The proteolytic activities applied alone have no significant effect on the extractability and the polyphenols levels of oils. Furthermore, when these proteolytic activities are added in combination with the pectolytic activities, the oil extractability is doubled at veraison and its flowing up to 99 % at maturity that barely 84 % in the control in addition to a richness of polyphenols which can reach 84 % more compared to the control. This increase in polyphenols wealth is probably due to the degradation of cell walls, cellular and vacuolar membranes by enzyme activities releasing PCs that were previously associated with these structures in the drupe.

The lipid extractability of 14 microalgae species and strains was assessed using organic solvents (methanol and chloroform). The high variability detected indicated the potential for applying this parameter as an additional criterion for microalgae screening in industrial processes such as biofuel production from microalgae. Species without cell walls presented higher extractability than species with cell walls. Analysis of cell integrity by flow cytometry and staining with propidium iodide showed a significant correlation between higher resistance to the physical treatments of cell rupture by sonication and the lipid extractability of the microalgae. Changes in benzo[a]pyrene (B[a]P) extractability over 160 days ageing in four contrasting soils varying in organic matter content and clay mineralogy were investigated using dichloromethane: acetone 1:1 (DCM/Ace), 60 mM hydroxypropyl-β-cyclodextrin (HPCD) solution, 1-butanol (BuOH) and Milli-Q water. The B[a]P extractability by the four methods decreased with ageing and a first-order exponential model could be used to describe the kinetics of release. Correlation of the kinetic rate constant with major soil properties showed a significant effect of clay and sand contents and pore volume fraction (<6 nm) on sequestration of the desorbable fraction (by HPCD) and the water-extractable fraction. Analysis of (14)C-B[a]P in soils after ageing showed a limited loss of B[a]P via degradation. Fractionation of B[a]P pools associated with the soil matrix was analysed according to extractability of B[a]P by the different extraction methods. A summary of the different fractions is proposed for the illustration of the effect of ageing on different B[a]P-bound fractions in soils. Sample processing is often ignored during analytical method development and validation, but accurate results for real samples depend on all aspects of the analytical process. Also, validation is often conducted using only spiked samples, but extraction yields may be lower in incurred samples. In this study, different variables in extraction for incurred pesticides and environmental contaminants in fish were investigated. Among 207 analytes screened using low-pressure gas chromatography-tandem mass spectrometry, consisting of 150 pesticides, 15 polycyclic aromatic hydrocarbons (PAHs), 14 polychlorinated biphenyls (PCBs), 6 polybrominated diphenyl ethers (PBDEs), and 22 other flame retardants (FRs), 35 (16 pesticides, 9 PCBs, 5 PBDEs, and 5 PAHs) were identified for quantification in samples of salmon, croaker, and NIST Standard Reference Material 1947 (Lake Michigan Fish Tissue). Extraction efficiencies using different extraction devices (blending, vortexing, and vibrating) versus time, sample size, and sample/solvent ratio were determined. In comparison to blending results, use of a pulsed-vortexer for 1 min with 1/1 (g/mL) sample/acetonitrile ratio was generally sufficient to extract the incurred contaminants in the homogenized fish tissues. Conversely, extraction with a prototype vibration shaker often took 60 min to achieve 100% extraction efficiency. A main conclusion from this study is that accurate results for real samples can be obtained using batch extraction with a pulsed-vortexer in a simple and efficient method that achieves high sample throughput.

Broccoli (Brassica oleracea L. var. Italica) is largely consumed all over the world and has a high economic importance. Likewise, broccoli contains high levels of glucosinolates, carotenoids and total phenols, which are related with the prevention of chronic diseases. The present project's objective was to evaluate the effect of industrial freezing on the stability of bioactive molecules in seven commercial broccoli cultivars (Tlaloc®, Endurance®, Florapack®, Domador®, Steel®, Iron Man® and Avenger®). In general, industrial freezing increased the extractability of total glucosinolates, whereas total phenols remained constant in most broccoli cultivars. Likewise, broccoli subjected to industrial freezing showed higher levels of total carotenoids (∼60-300% higher) as compared with fresh broccoli. Results suggest that bioactive compounds in frozen broccoli would be more bioavailable than in raw.

Oral bioavailability of benzo[a]pyrene (B[a]P) was studied in a swine model using eight spiked soil samples after incubation for 50 and/or 90 days. Silica sand was used as a reference material and the relative bioavailability (RB) of B[a]P in soils was calculated as the quotient of the area under the plasma B[a]P curve (AUC) for soil and AUC for the silica sand. Significantly reduced RB was observed in all study soils after 90 days ageing, ranging from 22.1±0.4% to 62.7±10.1%, except for one very sandy soil (sand content 87.6%) where RB was unchanged (108.1±8.0%). Apart from this, bioavailability decreased during ageing with the decrease (from day 50 to day 90) being only significant for a clayey soil containing expandable clay minerals. Statistical analyses of B[a]P RB at day 90 (eight soils) and soil properties showed no direct correlation between RB and specific soil properties such as total organic carbon (TOC) and clay content which were commonly linked to organic contaminant sequestration. However, strongly significant relationships (p<0.001) were found between RB and the fine particle associated carbon (FPAC) defined as (Silt+Clay)/TOC, and between RB and the soil mesopore (<6nm; p<0.001) fraction, after two samples with high pH and high EC being excluded from the analyses. The bioaccessibility estimated by four in vitro extraction methods: dichloromethane/acetone sonication (DCM/Ace), butanol vortex (BuOH), hydroxypropyl-β-cyclodextrin extraction (HPCD) and Milli Q water leaching methods at different sampling time (1 day, 50 days and 90 days after spiking) also showed a decreasing trend. Background: Polyphenols have a favourable antioxidant potential on human health, suggesting that their high content in apple is responsible for the beneficial effects of apple consumption. They are also linked to the quality of apple juices and ciders since they are predominantly responsible for astringency, bitterness and colour. Major phenolic compounds were quantified by liquid chromatography in fruits and juices from a cider apple progeny harvested for 3 years. The total content of procyanidins and their average degree of polymerisation (DPn) were also determined in fruits by phloroglucinolysis. Variability and extraction yield of these compounds were determined., Results: The variability observed in the progeny was representative of the variability observed in many cider apple varieties. Hydroxycinnamic acids were the most extractable group, with an average extraction yield of 67%, whereas flavonols and anthocyanins were the least., Conclusion: This study is the first to introduce variability and extraction yields of the main phenolic compounds in both fruits and juices of a cider apple progeny. Artificial soil is an important standard medium and reference material for soil ecotoxicity bioassays. Recent studies have documented the significant variability of their basic properties among different laboratories. Our study investigated (i) the variability of ten artificial soils from different laboratories by means of the fate, extractability and bioavailability of phenanthrene and lindane, and (ii) the relationships of these results to soil properties and ageing. Soils were spiked with (14)C-phenanthrene and (14)C-lindane, and the total residues, fractions extractable by hydroxypropyl-β-cyclodextrin, and the fractions of phenanthrene mineralizable by bacteria were determined after 1, 14, 28 and 56 days. Significant temporal changes in total residues and extractable and mineralizable fractions were observed for phenanthrene, resulting in large differences between soils after 56 days. Phenanthrene mineralization by indigenous peat microorganisms was suggested as the main driver of that, outweighing the effects of organic matter. Lindane total residues and extractability displayed much smaller changes over time and smaller differences between soils related to organic matter. Roughly estimated, the variability between the artificial soils was comparable to natural soils. The implications of such variability for the results of toxicity tests and risk assessment decisions should be identified. The main objective of this study was to determine the effects of long-term abiotic processes during aging of organic pollutants in soil on their microbial degradability and formation of non-extractable residues. The specific aims of our study were to investigate how the fate of p353-nonylphenol (NP) and phenanthrene (Phe) in soils might be affected by: (i) saturation of soil by cations with different valency (Na(+), Ca(2+) or Al(3+)), (ii) addition of organic substrate (wood flour) during incubation period and (iii) different soil moisture levels. This study showed positive effect of long-term aging of sterilized samples on respiration of re-inoculated samples. However, the lack of aging effects on the mineralization of NP and Phe indicates that slow sorption processes by diffusion into less bioaccessible domains were not relevant in studied soils. Similarly, the lower respiration and xenobiotic mineralization rates in the Na(+) and Al(3+) treated soils indicate that this is due to toxic effects on microbial activity and not due to xenobiotic accessibility. Instead, the formation of non-extractable residues was strongly promoted by biological activity, most likely through formation of more reactive metabolites. The addition of wood flour greatly stimulated microbial respiration and enhanced NP mineralization while inhibiting that of Phe. Along with negligible effect of water addition after 4 weeks of incubation on kinetics of soil respiration, the soil moisture effect on xenobiotics mineralization indicates that most probably the bioavailability of NP and Phe increased due to bridging role of water films in soil., (Copyright © 2013 Elsevier Ltd. Arabinoxylan (AX) is a major dietary fibre component found in a variety of cereals. Numerous health benefits of arabinoxylans have been reported to be associated with their solubility and molecular features. The current study reports the development of a functional bread using a combination of AX-enriched material (AEM) and optimal commercial endoxylanase. The total AX content of bread was increased to 8.2 g per 100 g available carbohydrates. The extractability of AX in breads with and without endoxylanase was determined. The results demonstrate that water-extractable AX (WE-AX) increased progressively through the bread making process. The application of endoxylanase also increased WE-AX content. The presence of 360 ppm of endoxylanase had positive effects on the bread characteristics in terms of bread volume and firmness by converting the water unextractable (WU)-AX to WE-AX. In addition, the molecular weight (Mw) distribution of the WE-AX of bread with and without endoxylanase was characterized by size-exclusion chromatography. The results show that as the portion of WE-AX increased, the amount of high Mw WE-AX (higher than 100 kDa) decreased, whereas the amount of low Mw WE-AX (lower than 100 kDa) increased from 33.2% to 44.2% through the baking process. The low Mw WE-AX further increased to 75.5% with the application of the optimal endoxylanase (360 ppm).
