The use of fungicides in the postharvest treatment of mangoes has been widespread due to the incidence of pathogens, but awareness of the health risks arising from their use has increased, driving the search for more sustainable treatments. This study aimed to evaluate the activity of antifungal treatment of seven essential oils (EO) against four fungi that cause postharvest diseases in mangoes and define the minimum inhibitory concentration (MIC) and chemical composition, analyzed by gas chromatography (GC-MS). The results showed that the EOs of oregano, rosemary pepper, cinnamon bark, and clove inhibited 100% of the mycelial growth of the studied pathogens, with MIC ranging from 250 to 2000 μL.L -1 . The main compound found in oregano was carvacrol (69.1%); in rosemary and pepper oil, it was thymol (77.2%); cinnamaldehyde (85.1%) was the main constituent of cinnamon bark, and the eugenol (84.84%) in cloves. When evaluating the antifungal activity of these compounds, thymol and carvacrol showed greater inhibitory activity against fungi. Therefore, this study showed the great potential of oregano, clove, rosemary pepper, and cinnamon bark essential oil as alternative treatments to synthetic fungicides in controlling postharvest diseases in mangoes., (© 2024. Objective: Considering that glucose is an important component of seminal plasma and is a cryoprotectant at high concentrations, the aim of this study was to investigate the possible association of glucose levels in fresh semen with the sperm survival and motility rates following cryopreservation., Methods: This was a prospective study including 149 men undergoing semen analysis due to male and/or female infertility. The seminal samples were analyzed according to the World Health Organization standards and glucose concentrations were measured using a dipstick glucometer. Samples were cryopreserved with Test Yolk Buffer-Gentamicine freezing medium under liquid nitrogen for an average of 120 days. The frozen aliquots were thawed at 37°C for 10 minutes and analyzed using the same methods and protocols used pre-freezing., Results: Glucose levels ranged from 14 to 99 mg/dL and were similar in individuals with normal (n=100) vs. abnormal (n=49) semen analysis. The rates of sperm recovery (total, alive or motile sperm) in the cryopreserved samples did not change among samples with different glucose levels (p>0.05, Kruskal-Wallis ANOVA and Spearman's correlation coefficient)., Conclusions: There appears to be no association between glucose levels in human semen samples and their resistance to cryopreservation.

The huge biological diversity of the Brazilian Cerrado is an important source of economically interesting microbial agents. The phylum Actinobacteria plays an important role in nutrient cycling, potentially improving their availability to plants. In this study, we isolated an actinobacteria (strain 3AS4) from wheat rhizospheres of crops cultivated in the Cerrado biome. Strain 3AS4 was identified as belonging to the genus Streptomyces and had phosphorus mobilization ability, mineralizing approximately 410 μg ml -1 from phytate, 300 μg ml -1 from calcium phosphate, and 200 μg ml -1 from rock phosphate. The analysis of the actinobacteria crude extract by spectrometric techniques revealed the presence of gluconic and 2-ketogluconic acid, and a greenhouse experiment was carried out to evaluate its plant growth promotion activity in soybean. Soil in its natural condition (with no phosphorus addition), 40 kg ha -1 rock phosphate from Bayovar (RP) added to soil, and triple super phosphate (SPT) added to soil were used. Significant differences in plant height were observed at 6 weeks when the plants were inoculated with the 3AS4 strain. The growth of inoculated plants in natural condition was promoted in 17% compared with the RP and SPT non-inoculated conditions, suggesting that inoculation can enable plants to grow with lower chemical P fertilizers. In the plants that were inoculated with the 3AS4 strain in the RP condition, the plant height increased by approximately 80% and the shoot:root ratio was approximately 30% higher compared to control conditions (non-inoculated plants in natural conditions). 3AS4 has P-solubilizing potential and can be exploited as an inoculant for soybean cultivation. Intensive fish cultivation has a high incidence of infection, which is often controlled by administering antibiotics. Florfenicol (FF) is one of the two antimicrobial drugs permitted for aquaculture in Brazil. Due to their intensive use, potentially harmful effects on aquatic organisms are of great concern. In this sense, we investigated whether the presence of FF in cultivation water could change the health parameters of Nile tilapia. For this, we evaluated hemoglobin, hematocrit, mean corpuscular hemoglobin (MCHC) concentration, mean corpuscular volume (MCV), total plasma protein (TPP), number of circulating red blood cells and leukocytes, as lipid peroxidation levels, catalase activity and glutathione S-transferase activity of fish exposed to 11.72 mg L -1 of FF in water for 48 h. The fish were divided into two groups: Nile tilapia in water with FF or without FF (control). Exposure to FF in cultivation water for a short period didn't change the hematological variables analyzed, but caused changes in liver ROS (Reactive oxygen species) markers of the Nile tilapia, which was revealed by lipid peroxidation levels, catalase activity, and glutathione S-transferase. The 48h exposure period was enough to induce oxidative stress in hepatocytes, causing cellular oxidative damage. Therefore, the antibiotic florfenicol may cause toxicity to organisms and aquatic ecosystems, even at a sublethal concentrations near 1/100 LC50-48h for fish species., Competing Interests: The authors declare no conflict of interest., (© 2020 The Author(s).)

The process of freezing/thawing causes structural and functional damage to sperm samples, which can be mitigated by seminal plasma proteins. This study investigated the proposition that seminal protein measurements using a urinary dipstick prior to freezing could help predict the post-thaw recovery of live spermatozoa. This was a prospective study including 149 men undergoing semen analysis due to male and/or female infertility. The seminal samples were analysed according to World Health Organisation standards and protein concentrations were measured using a commercially available urinary dipstick following quantitative validation. The median live sperm recovery rates were 79%, 81% and 94%, respectively, in samples with protein concentrations of ≤1.0 g/L (+/++), 3.0 g/L (+++) and ≥20.0 g/L (++++) measured in fresh specimen dipstick analysis ( p  < 0.05) indicating that the probability of recovering at least 50% of frozen spermatozoa increased progressively with higher protein concentrations in the fresh sample (chi-square for linear association = 7.17. p  = 0.007). In conclusion, fresh seminal protein concentration levels assessed with a dipstick test correlate with the proportion of live spermatozoa recovered from cryopreserved samples. This simple, low-cost test may add prognostic information to baseline semen analysis prior to sperm banking.

Sulfamethazine (SMZ) is one of the most commonly used sulfonamide compounds in fish farming, and its physiological effects on fish are unknown. SMZ was administered to juvenile Nile tilapia (Oreochromis niloticus) at a dose level of 422 mg kg(-1) body weight, for a period of 11 days, via medicated feed. Fish were divided into two groups, the control group (CG) and the group fed with SMZ in feed. The administration of SMZ did not alter the erythrograms and leukograms of the Nile tilapia. The SMZ-fed group showed the same hepatic lipid peroxidation (LPO) concentration as the CG. Nonetheless, the oral administration of SMZ raised the hepatic catalase (CAT) and glutathione S-transferase (GST) activities, the increase probably being sufficient to prevent hepatic LPO production. The oral administration of SMZ affects the hepatic GST and CAT activities of Nile tilapia.

At least 52 % of the planted rice area in Rio Grande do Sul, a major rice producing state in Brazil, employs Clearfield(®) production system, corresponding to 580,000 ha of cultivated area. To grow rice with Clearfield(®) technology, producers combine imazethapyr and imazapic herbicides. However, these herbicides leave residual activity in soil; consequently, the repeated application of imazethapyr and imazapic on Brazilian Clearfield(®) rice fields has increased these herbicides persistence in treated soils. In this study, a method has been developed for removal and quantification of imazethapyr and imazapic residues in soil through ultrasonic assisted extraction using methanol-phosphoric acid aqueous solution (pH 2.0). The detected response was linear for both herbicides within the range of 0.25-5 ng mL(-1) with correlations coefficients >0.99. The quantification limit was limit of quantification 0.2 µg Kg(-1) for both pesticides. The good recovery rate from all pesticides, which ranges between 70 % and 120 %, demonstrates the method's validity.

The Antarctic endophytic fungus (strain ITA1-CCMA 952) was isolated from the moss Schistidium antarctici found in Admiralty Bay, King George Island, Antarctica. Strain ITA1-CCMA 952 was assigned to the specie Mortierella alpina by phylogenetic analysis based on 18S rRNA gene sequences. This strain produces high levels of polyunsaturated fatty acids (PUFAs), including y-(gamma) linolenic acid and arachidonic acid, which when combined represents 48.3% of the total fatty acid content. Fungal extracts demonstrated strong antioxidant activity with the EC50 value of 48.7 μg mL(-1) and also a strong antibacterial activity, mainly against the following bacteria: Escherichia coli, with a MIC of 26.9 μg mL(-1) and Pseudomonas aeruginosa and Enterococcus faecalis, both with a MIC of 107 μg mL(-1). A GC-MS analysis of the chloroform fraction obtained from the crude extract revealed the presence of potential antimicrobials (Pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro-3-(2-methylpropyl) and Pyrrolo[1,2-a]pyrazine-1,4-dione, hexahydro-3-(phenylmethyl)) as the major compounds. Therefore, the M. alpina strain ITA1-CCMA 952 is a promising fungus for the biotechnological production of antibiotics, antioxidant substances and PUFAs. This study highlights the need for more research in extreme environments, such as Antarctica.

Conyza canadensis (L.) Cronquist syn. (horseweed) is a problematic and invasive weed with reported allelopathic properties. To identify the phytotoxic constituents of the aerial parts, a systematic bioactivity-guided fractionation of the dichloromethane extract was performed. Three active enyne derivatives, (2Z,8Z)-matricaria acid methyl ester, (4Z,8Z)-matricaria lactone, and (4Z)-lachnophyllum lactone, were identified. The lactones inhibited growth of the monocot Agrostis stolonifera (bentgrass) and the dicot Lactuca sativa (lettuce) at 1 mg mL(-1), while the (2Z,8Z)-matricaria acid methyl ester was less active. In a dose-response screening of the lactones for growth inhibitory activity against Lemna paucicostata , (4Z)-lachnophyllum lactone was the most active with an IC50 of 104 μM, while the (4Z,8Z)-matricaria lactone was less active (IC50 of 220 μM). In a fungal direct bioautography assay, the two lactones at 10 and 100 μg/spot inhibited growth of the plant pathogenic fungi Colletotrichum acutatum , Colletotrichum fragariae , and Colletotrichum gloeosporioides . In a dose-response screening of the lactones against six different plant pathogenic fungi, (4Z,8Z)-matricaria lactone was more active than the commercial fungicide azoxystrobin on Col. acutatum , Col. fragariae , and Col. gloeosporioides at 30 μM and about as active as the commercial fungicide captan against Col. gloeosporioides , while (4Z)-lachnophyllum lactone was less active.

A simple method using LC/MS/MS was developed and validated to determine residues of malachite green (MG) and leucomalachite green (LMG) in fish fillet. A modified QuEChERS (Quick, Easy, Cheap, Effective, Rugged, and Safe) technique was used to perform the sample preparation. The optimal extraction and cleanup conditions were established using an experimental design. The validation parameters obtained to determine both MG and LMG complied with the requirements established by regulatory agencies for the presence of such substances in fish, which establish that the method must attain, at least, a minimum required performance limit of 2 ng/g. The accuracy values ranged between 95 and 107%, and the precision values were lower than 11.2%. The method was used in the analysis of tilapia samples (n = 20) commercialized in Campinas, SP, Brazil. None of the samples presented detectable levels of MG or LMG residues.

This study presents a model to simulate the organic substance concentrations in corn grains assuming that the substances in soil solution are absorbed via the transpiration stream by plants growing in soils fertilized with sewage sludge (SS). The model was applied and validated using soil and corn grain samples from a long-term field experiment with six successive yearly applications of SS to the soil. The following polycyclic aromatic hydrocarbons (PAHs) were simulated and evaluated in soil and grain samples: acenaphthene, acenaphthylene, anthracene, benz(a)anthracene, benz(a)pyrene, benz(b)fluoranthene, benz(g,h,i)perylene, benz(k)fluoranthene, chrysene, dibenz(a,h)anthracene, fluoranthene, fluorene, indeno(1,2,3-c,d)pyrene, naphthalene, phenanthrene and pyrene. The PAH bioconcentration factors (BCF) in corn grains ranged from 1.57 to 10.97 L kg(-1). Polycyclic aromatic hydrocarbons with low soil distribution coefficients and high values of transpiration stream concentration factors (TSCF) are more likely to be absorbed by corn plants and accumulated in grains. It was possible to estimate and observe that highly lipophilic PAH molecules (heavy PAHs) show lower accumulative potential in corn grains than the less lipophilic ones (light PAHs). Sewage sludges containing significant concentrations of light PAHs with two, three or four benzene rings should be avoided as fertilizers in alimentary field crops., (Copyright 2010 Elsevier B.V. Several different Advanced Oxidation Processes (AOPs) including ozonation at pH 6.5 and 10, photolysis and heterogeneous photocatalysis using TiO(2) as semiconductor and dissolved oxygen as electron acceptor were applied to study the degradation of glyphosate (N-phosphonomethyl glycine) in water. The degree of glyphosate degradation, the reactions kinetic and the formation of the major metabolite, aminomethyl phosphonic acid (AMPA), were evaluated. Ozonation at pH 10 resulted in the maximum mineralization of glyphosate. It was observed that under the experimental conditions used in this study the degradation of glyphosate followed the first-order kinetics. The half-life obtained for glyphosate degradation in the O(3)/pH 10 process was 1.8 minutes.

This study was undertaken to evaluate the degradation and mobility of the herbicide tebuthiuron (N-[5-(1,1-dimethylethyl)-1,3,4-thiadiazol-2-yl]-N,N'-dimethylurea) in soil under field conditions, and its potential for leaching and groundwater contamination. A watershed, Espraiado, located over a recharge area in Brazil, was chosen for soil and water studies. At Espraiado, water samples were collected from seven wells at intervals of three months from March 2004 to June 2006 and analyzed for tebuthiuron. Other samples were taken from city wells located outside of the recharge area. To assess the potential movement to the aquifer, tebuthiuron was also applied to trial plots at the recommended label rate of 1.0 kg/ha a.i. in May of 2004, with and without sugarcane coverage, on sandy soil. Soil samples were collected during the years of 2004 and 2005, at depths intervals of 20 cm from soil surface down to 120 cm and analyzed for tebuthiuron at zero, 3, 30, 60, 90, 120, 150, 180, 240, and 300 days after application. There was no clear effect of sugarcane coverage on the tebuthiuron degradation in soils, but it moved faster into the soil where there was no cover. After 180 days there were no measurable residues in the soil, and tebuthiuron was not found below 40 cm depth in any time. Tebuthiuron had a half-life of 20 days under those conditions. No tebuthiuron residue was found in ground water samples at any sampling time.

A method for the simultaneous determination of diuron and linuron pesticides in human urine was developed, using both solid-phase extraction (SPE) and high-performance liquid chromatography (HPLC) phases made in our own laboratory. These materials were prepared by sorption of polysiloxanes onto a silica surface, followed by immobilization. The HPLC columns were prepared from poly(methyloctylsiloxane), PMOS, immobilized onto silica with microwave radiation while the SPE cartridges where made with poly(methyloctadecylsiloxane), immobilized thermally. Method validation was performed for diuron and linuron for three fortification levels. The recoveries obtained were 85-103%, the inter- and intra-assay precisions were less than 1.6 and 1.8%, respectively. The limits of quantitation and detection for diuron were 2.4 and 8.0 microg/l and for linuron were 5.0 and 12 microg/l, respectively.

This work presents a fast, easy and effective procedure to obtain silica-based C18 type sorbents for use in solid-phase extraction (SPE). The solid-phase was made by depositing a polymer, poly(methyloctadecylsiloxane) (PMODS), on the silica support followed by immobilization using gamma irradiation. The method presents an application of green chemistry in the analytical laboratory, since there are no toxic residues after the synthesis. Water samples spiked with a mixture of pesticides (benomyl, tebuthiuron, diuron, simazine, atrazine and ametryn) were extracted in the off-line mode in order to evaluate the performance of this sorbent.

From the petrol extract of Lonchocarpus latifolius roots, 10 flavonoids were isolated. These included: 3,5-dimethoxy-2'',2''-dimethylpyrano-(5'',6'':8,7)-flavone, 3-methoxy-(2'',3'':7,8)-furanoflavanone, 3',4'-methylenedioxy-(2'',3'':7,8)-furanoflavanone, and (2,3-trans-3,4-trans)-3,4-dimethoxy-(2'',3'':7,8)-furanoflavan, as well as the previously known karanjachromene, karanjin, lanceolatin B, pongachromene, pongaglabrone and ponganpin. Only nine flavonoids could be quantified through HPLC analysis.
