Copper ions (Cu 2+ ) pose significant risks to both human health and the environment as they tend to accumulate in soil and water. To address this issue, an innovative method using biomass-derived fluorescent carbon dots (D-CDs) synthesized via a hydrothermal process, with xylan serving as the carbon source was developed. D-CDs solution exhibited remarkable sensitivity and selectivity as a fluorescence sensor for Cu 2+ , boasting a low detection threshold of 0.64 μM. In order to facilitate real-time monitoring of Cu 2+ , solid-state fluorescent nanofiber membrane (NFD-CDs) through electrospinning was engineered. Additionally, D-CDs demonstrated successful Cu 2+ detection in various real water samples, including those sourced from Xuanwu Lake, the Yangtze River, tap water, and bottled water, with accurate recovery rates observed. As a result, this research introduces a dual-mode analytical system for onsite detection of Cu 2+ in real scenarios. Chemoresistance is the primary reason for poor prognosis in patients with pancreatic cancer (PC). Recent studies have indicated that ferroptosis may improve chemoresistance, but the underlying mechanisms remain unclear. In this study, significant upregulation of heat shock protein 90α (Hsp90α) expression is detected in the peripheral blood and tissue samples of patients with chemoresistant PC. Further studies reveal that Hsp90α promotes the proliferation, migration, and invasion of a chemoresistant pancreatic cell line (Panc-1-gem) by suppressing ferroptosis. Hsp90α competitively binds to Kelch-like ECH-associated protein 1 (Keap1), liberating nuclear factor erythroid 2-related factor 2 (Nrf2) from Keap1 sequestration. Nrf2 subsequently translocates into the nucleus and activates the glutathione peroxidase 4 (GPX4) pathway, thereby suppressing ferroptosis. This process further worsens the chemoresistance of PC cells. This study provides valuable insight into potential molecular targets to overcome chemoresistance in PC. It sheds light on the intricate mechanisms linking Hsp90α and ferroptosis to chemoresistance in PC and provides a theoretical foundation for the development of novel therapeutic strategies.

High invasive capacity and acquired tyrosine kinase inhibitors (TKI) resistance of kidney renal clear cell carcinoma (KIRC) cells remain obstacles to prolonging the survival time of patients with advanced KIRC. In the present study, we reported that sine oculis homeobox 1 (SIX1) was upregulated in sunitinib-resistant KIRC cells and metastatic KIRC tissues. Subsequently, we found that SIX1 mediated metastasis and sunitinib resistance via Focal adhesion (FA) signaling, and knockdown of SIX1 enhanced the antitumor efficiency of sunitinib in KIRC. Mechanistically, Integrin subunit beta 1 (ITGB1), an upstream gene of FA signaling, was a direct transcriptional target of SIX1. In addition, we showed that DExH-box helicase 9 (DHX9) was an important mediator for SIX1-induced ITGB1 transcription, and silencing the subunits of SIX1/DHX9 complex significantly reduced transcription of ITGB1. Downregulation of SIX1 attenuated nuclear translocation of DHX9 and abrogated the binding of DHX9 to ITGB1 promoter. Collectively, our results unveiled a new signal axis SIX1/ITGB1/FAK in KIRC and identified a novel therapeutic strategy for metastatic KIRC patients., (© 2024. Objective: To explore the clinical value of metagenomic next-generation sequencing (mNGS) in diagnosis and treatment of periprosthetic joint infection (PJI) after total knee arthroplasty (TKA)., Methods: Between April 2020 and March 2023, 10 patients with PJI after TKA were admitted. There were 3 males and 7 females with an average age of 69.9 years (range, 44-83 years). Infection occurred after 8-35 months of TKA (mean, 19.5 months). The duration of infection ranged from 16 to 128 days (mean, 37 days). The preoperative erythrocyte sedimentation rate (ESR) was 15-85 mm/1 h (mean, 50.2 mm/1 h). The C reactive protein (CRP) was 4.4-410.0 mg/L (mean, 192.8 mg/L). The white blood cell counting was (3.4-23.8)×10 9 /L (mean, 12.3×10 9 /L). The absolute value of neutrophils was (1.1-22.5)×10 9 /L (mean, 9.2×10 9 /L). After admission, the joint fluid was extracted for bacterial culture method and mNGS test, and sensitive antibiotics were chosen according to the results of the test, and the infection was controlled in combination with surgery., Results: Seven cases (70%) were detected as positive by bacterial culture method, and 7 types of pathogenic bacteria were detected; the most common pathogenic bacterium was Streptococcus lactis arrestans . Ten cases (100%) were detected as positive by mNGS test, and 11 types of pathogenic bacteria were detected; the most common pathogenic bacterium was Propionibacterium acnes . The difference in the positive rate between the two methods was significant ( P =0.211). Three of the 7 patients who were positive for both the bacterial culture method and the mNGS test had the same results for the type of pathogenic bacteria, with a compliance rate of 42.86% (3/7). The testing time (from sample delivery to results) was (4.95±2.14) days for bacterial culture method and (1.60±0.52) days for mNGS test, and the difference was significant ( t =4.810, P <0.001). The corresponding sensitive antibiotic treatment was chosen according to the results of bacterial culture method and mNGS test. At 3 days after the one-stage operation, the CRP was 6.8-48.2 mg/L (mean, 23.6 mg/L); the ESR was 17-53 mm/1 h (mean, 35.5 mm/1 h); the white blood cell counting was (4.5-8.1)×10 9 /L (mean, 6.1×10 9 /L); the absolute value of neutrophils was (2.3-5.7)×10 9 /L (mean, 4.1×10 9 /L). All patients were followed up 12-39 months (mean, 23.5 months). One case had recurrence of infection at 6 months after operation, and the remaining 9 cases showed no signs of infection, with an infection control rate of 90%., Conclusion: Compared with bacterial culture method, mNGS test can more rapidly and accurately detect pathogenic bacteria for PJI after TKA, which is important for guiding antibiotics combined with surgical treatment of PJI.

Herein, we report a transition-metal-free C-H bond sulfoximidation protocol of sulfonyl hydrazones with hypervalent iodine(III) reagents. A library of novel N -sulfaneylidenehydrazonamides was constructed via chemoselective C-N bond formation reactions at aldehyde C-H bonds of sulfonyl hydrazones in the presence of a base. The reaction demonstrated broad substrate group diversity under exceedingly mild reaction conditions, and excellent yields were achieved at room temperature.

SnSe 2 with high theoretical capacity has been identified as an emerging anode candidate for lithium-ion batteries (LIBs) and sodium-ion batteries (SIBs). However, the rate performance and cycling performance of this material in practical applications are still limited by unavoidable volume expansion and low conductivity. In this work, we designed and synthesized nitrogen-doped carbon-coated SnSe 2 /C-N composites using 2-aminoterephthalic acid (C 8 H 7 NO 4 ) as a nitrogen-containing compound for modification by hydrothermal and vacuum calcination methods to achieve efficient utilization of active sites and optimization of the electronic structure. The carbon skeleton inherited from the Sn-MOF precursor can effectively improve the electronic conduction properties of SnSe 2 . N-doping in the Sn-MOF can increase the positive and negative electrostatic potential energy regions on the molecular surface to further improve the electrical conductivity, and effectively reduce the binding energy with Li + /Na + which was determined by Density Functional Theory (DFT) methods. In addition, the N-doped carbon skeleton also introduces a larger space for Li + /Na + intercalation and enhances the mechanical properties. In particular, the post-synthetically modified MOF-derived SnSe 2 /C-N materials exhibit excellent cyclability, with a reversible capacity of 695 mA h g -1 for LIBs and 259 mA h g -1 for SIBs after 100 cycles at 100 mA g -1 .

Developing high-sensitivity TEA sensors has extremely important significance for human health. Design of three-dimensional (3D) nanostructures assembled from one-dimensional nanomaterials can effectively improve sensing performance. In this work, a nest-like structure assembled by Cr-doped MoO 3 (Cr-MoO 3 ) nanorods with relatively higher specific surface area was prepared. In order to improve the sensing performance, Cr-MoO 3 skeleton was combined with ZnSe nanospheres of different mass ratios as sensing materials (ZnSe/Cr-MoO 3 ), and the successful construction of the heterojunction structure was supported by various spectroscopies and charge density calculation. The prepared composite with an optimal moiety ratio showed very high response values of 371 and 1301 for 10 ppm and 50 ppm for TEA at 200 °C, respectively. Simultaneously, the composite sensor also exhibited a low detection limit (1.7 ppb). The improvement of the sensing performance of ZnSe/Cr-MoO 3 was attributed to the formation of oxygen vacancies induced by Cr doping, the 3D nest-like structure provided an efficient network for charge transport/collection and the n-n heterojunctions between Cr-MoO 3 nanorods and ZnSe nanospheres. Renal ischaemia-reperfusion injury (RIRI) is a primary cause of acute kidney damage, occurring frequently in situations like renal transplantation, yet the underlying mechanisms were not fully understood. Sentrin-specific protease 1 (SENP1) is an important member of the SENP family, which is widely involved in various diseases. However, the role of SENP1 in RIRI has been unclear. In our study, we discovered that SENP1 was involved in RIRI and reduced renal cell apoptosis and oxidative stress at elevated levels. Further mechanistic studies showed that hypoxia-inducible factor-1α (HIF-1α) was identified as a substrate of SENP1. Furthermore, SENP1 deSUMOylated HIF-1α, which reduced the degradation of HIF-1α, and exerted a renoprotective function. In addition, the protective function was lost after application of the HIF-1α specific inhibitor KC7F2. Briefly, our results fully demonstrated that SENP1 reduced the degradation of HIF-1α and attenuated oxidative stress and apoptosis in RIRI by regulating the deSUMOylation of HIF-1α, suggesting that SENP1 may serve as a potential therapeutic target for the treatment of RIRI., (© 2024 The Author(s). Background: Tumor mutation burden (TMB) and VHL mutation play a crucial role in the management of patients with clear cell renal cell carcinoma (ccRCC), such as guiding adjuvant chemotherapy and improving clinical outcomes. However, the time-consuming and expensive high-throughput sequencing methods severely limit their clinical applicability. Predicting intratumoral heterogeneity poses significant challenges in biology and clinical settings. Our aimed to develop a self-supervised attention-based multiple instance learning (SSL-ABMIL) model to predict TMB and VHL mutation status from hematoxylin and eosin-stained histopathological images., Methods: We obtained whole slide images (WSIs) and somatic mutation data of 350 ccRCC patients from The Cancer Genome Atlas for developing SSL-ABMIL model. In parallel, 163 ccRCC patients from Clinical Proteomic Tumor Analysis Consortium cohort was used as independent external validation set. We systematically compared three different models (Wang-ABMIL, Ciga-ABMIL, and ImageNet-MIL) for their ability to predict TMB and VHL alterations., Results: We first identified two groups of populations with high- and low-TMB (cut-off point = 0.9). In two independent cohorts, the Wang-ABMIL model achieved the highest performance with decent generalization performance (AUROC = 0.83 ± 0.02 and 0.8 ± 0.04 in predicting TMB and VHL, respectively). Attention heatmaps revealed that the Wang-ABMIL model paid the highest attention to tumor regions in high-TMB patients, while in VHL mutation prediction, non-tumor regions were also assigned high attention, particularly the stromal regions infiltrated by lymphocytes., Conclusions: Our results indicated that SSL-ABMIL can effectively extract histological features for predicting TMB and VHL mutation, demonstrating promising results in linking tumor morphology and molecular biology., (© 2024 The Author(s). Hyperlipidemic pancreatitis (HP) is an inflammatory injury of the pancreas triggered by elevated serum triglyceride (TG) levels. The mechanistic target of rapamycin (mTOR) signaling pathway plays a crucial role in regulating lipid homeostasis and inflammation. This study aimed to investigate whether the activity of mTOR complex 2 (mTORC2) affects the progression of HP and its underlying mechanisms. In vivo, a high-fat diet and retrograde administration of sodium taurocholate were employed to establish the HP models in rats, with pancreatic tissue pathology evaluated. The expression of Rictor and peroxisome proliferator-activator receptor (PPAR) was examined. The serum levels of TG, fatty acid metabolites, inflammatory and lipid metabolism-related factors were determined. In vitro, pancreatic acinar cells (PACs) were exposed to palmitic acid and cholecystokinin-8. PAC apoptosis, pyroptosis, and ferroptosis were assessed. In the HP models, rats and PACs exhibited upregulated Rictor and downregulated PPARα, and Rictor knockdown promoted PPARα expression. In vivo, Rictor knockdown decreased the serum levels of TG, α-amylase, total cholesterol, low-density lipoprotein cholesterol, lactate dehydrogenase, and inflammatory factors, while increasing high-density lipoprotein cholesterol levels. Rictor knockdown increased ACOX1 and CPT1α and decreased SREBP-1, CD36, SCD1, ACLY, and ACACA. Rictor knockdown reduced damage to pancreatic tissue structure. In vitro, Rictor knockdown inhibited PAC apoptosis, pyroptosis, and ferroptosis. Treatment with the PPARα antagonist GW6471 abolished the beneficial effects of Rictor knockdown. Rictor/mTORC2 deficiency reduces serum TG levels, maintains lipid homeostasis, and suppresses inflammation by inhibiting PPARα expression. Integrin β6 (ITGB6) is upregulated in multiple tumor types and elevated ITGB6 levels have been detected in patients with chronic pancreatitis. However, the role of ITGB6 in pancreatic fibrosis and cancer remains to be elucidated. In the present study, ITGB6 expression was assessed using western blotting and qRT-PCR. Besides, cell proliferation, cycling, migration, and invasion were evaluated using CCK-8, flow cytometry, wound healing, and transwell assays, respectively. The expression of fibrosis and JAK2/STAT3 signaling markers was detected by western blotting and immunofluorescence analysis. Moreover, nude mice were subcutaneously injected with co-cultured cell suspensions to establish an in vivo model. The results showed that ITGB6 was highly expressed in pancreatic cancer tissues and TGF-β-induced pancreatic stellate cells (PSCs). Inhibition of ITGB6 expression in PSCs resulted in clear inhibition of activated PSC proliferation, migration, and fibrogenesis. Additionally, reduced ITGB6 expression inhibits the JAK2/STAT3 signaling pathway. Interestingly, activators of the JAK2/STAT3 signaling pathway reversed the effects of ITGB6 disruption on PSCs. Activated PSCs notably promoted the proliferation, invasion, and migration of pancreatic cancer cells in a co-culture assay. In contrast, activated PSCs with low ITGB6 expression failed to significantly affect the malignancy of pancreatic cancer cells. Moreover, in vivo results showed that interference with ITGB6 inhibited the activation of PSCs and promoted the development of pancreatic cancer. Silencing ITGB6 inhibited the proliferation, migration, and fibrosis-like effects of activated PSCs and indirectly inhibited the metastasis and malignant process of pancreatic cancer by inhibiting the JAK2/STAT3 signaling pathway. Designing portable electrochemical sensors combined with highly efficient glucose oxidation electrodes offers a significant opportunity for convenient glucose detection. In this report, we present the design and preparation of platinum deposited Ni/NiFe 2 O 4 /Carbon composite (Pt/Ni/NiFe 2 O 4 /C) derived from Ni/Fe metal-organic frameworks (MOFs) followed by Pt deposition. Energy-dispersive X-ray spectroscopy (EDS), X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD), and electron microscopy (EM) were utilized to analyze the crystal structure, morphology, and chemical composition of the resulting materials. The glucose sensing capabilities of the optimal Pt/Ni/NiFe 2 O 4 /C-3 were assessed using amperometry methods on a smartphone-based portable device. Acting as a nonenzymatic glucose sensor, the Pt/Ni/NiFe 2 O 4 /C-3 electrode demonstrated notable sensitivity and a low limit of detection for glucose. The portable sensor exhibits high sensitivities of 131.88 μM mM cm -2 at low glucose concentration (3-500 μM) and 29.52 μA mM cm -2 at high glucose concentration (700-4000 μM), achieving a low detection limit of 1.1 μM (S/N = 3). The sensor also demonstrates enhanced selectivity and stability for detecting glucose. Furthermore, the portable sensor exhibits a clear step-ampere response in the detection of serum samples with satisfactory recovery ranging from 99.30 to 101.32%. Diabetic retinopathy disease contains lesions (e.g., exudates, hemorrhages, and microaneurysms) that are minute to the naked eye. Determining the lesions at pixel level poses a challenge as each pixel does not reflect any semantic entities. Furthermore, the computational cost of inspecting each pixel is expensive because the number of pixels is high even at low resolution. In this work, we propose a hybrid image processing method. Simple Linear Iterative Clustering with Gaussian Filter (SLIC-G) for the purpose of overcoming pixel constraints. The SLIC-G image processing method is divided into two stages: (1) simple linear iterative clustering superpixel segmentation and (2) Gaussian smoothing operation. In such a way, a large number of new transformed datasets are generated and then used for model training. Finally, two performance evaluation metrics that are suitable for imbalanced diabetic retinopathy datasets were used to validate the effectiveness of the proposed SLIC-G. The results indicate that, in comparison to prior published works' results, the proposed SLIC-G shows better performance on image classification of class imbalanced diabetic retinopathy datasets. This research reveals the importance of image processing and how it influences the performance of deep learning networks. The proposed SLIC-G enhances pre-trained network performance by eliminating the local redundancy of an image, which preserves local structures, but avoids over-segmented, noisy clips. It closes the research gap by introducing the use of superpixel segmentation and Gaussian smoothing operation as image processing methods in diabetic retinopathy-related tasks., (© 2024. Epidermal growth factor receptor 2 ( HER2 ) has been widely recognized as one of the targets for bladder cancer immunotherapy. The key to implementing personalized treatment for bladder cancer patients lies in achieving rapid and accurate diagnosis. To tackle this challenge, we have pioneered the application of deep learning techniques to predict HER2 expression status from H&E-stained pathological images of bladder cancer, bypassing the need for intricate IHC staining or high-throughput sequencing methods. Our model, when subjected to rigorous testing within the cohort from the People's Hospital of Wuhan University, which encompasses 106 cases, has exhibited commendable performance on both the validation and test datasets. Specifically, the validation set yielded an AUC of 0.92, an accuracy of 0.86, a sensitivity of 0.87, a specificity of 0.83, and an F1 score of 86.7%. The corresponding metrics for the test set were 0.88 for AUC, 0.67 for accuracy, 0.56 for sensitivity, 0.75 for specificity, and 77.8% for F1 score. Additionally, in a direct comparison with pathologists, our model demonstrated statistically superior performance, with a p -value less than 0.05, highlighting its potential as a powerful diagnostic tool.

The commercial application of lithium-sulfur batteries is primarily impeded by the constant shuttling of soluble polysulfides and sluggish redox kinetics. Nowadays, the discovery of the heterojunction, which combines materials with diverse properties, offers a new perspective for overcoming these obstacles. Herein, a functional coating separator for the lithium-sulfur battery is designed using a MnO 2 -ZnS p-n heterojunction with a spontaneous built-in electric field (BIEF). The MnO 2 nanowire provides suitable adsorption capacity for polysulfides, while the abundant reactive sites brought by ZnS ensure efficient conversion. Moreover, the BIEF significantly facilitates the migration of electrons and polysulfides at the MnO 2 -ZnS interface, enabling a smooth "adsorption-diffusion-conversion" reaction mechanism. By serving as both the adsorption module and catalytic sites, this BIEF allows batteries utilizing separators modified with MnO 2 -ZnS heterojunction to achieve an impressive initial capacity of 1511.1 mAh g -1 at 0.1C and maintain a capacity decay rate of merely 0.048% per cycle at 2.0C after 1000 cycles. Background: Due to the unfavourable neck-shoulder muscle loads caused by poor posture, the people who use the laptop for a long time may face the risk of neck and shoulder injuries., Objective: The purpose of this study investigates the impact of the screen height on the muscle activation of head flexion, neck and shoulder, and the cervical spine torque to provide the favorite screen height for laptop user., Methods: Twelve healthy young participants completed a15-minute task of the reading at the four different screen heights. sEMG signals of the splenius capitis (SC) and upper trapezius (UT) were measured and calculated the root mean square (RMS) and mean power frequency (MPF) to determine muscle fatigue. The different height of laptop users was simulated and the forces on the spine of users at different screen heights were analyzed by Jack., Results: Adjusting the height of the laptop screen can effectively reduce head flexion and muscle activity of SC and UT, and has a positive effect on reducing fatigue of SC, but has no significant effect on UT., Conclusions: Adjusting the height of the laptop screen can delay the occurrence of SC muscle fatigue to a certain extent. The joint analysis of sEMG spectrum and amplitude reports that the screen heights of D15 and D45 have the highest and the lowest frequency of fatigue, respectively. At the same time, the moment of spineT1/T2 and spineL4/L5 decrease with the increase of screen height.

Necrotic enteritis (NE) is a potentially fatal poultry disease that causes enormous economic losses in the poultry industry worldwide. The study aimed to evaluate the effects of dietary organic yeast-derived selenium (Se) on immune protection against experimental necrotic enteritis (NE) in commercial broilers. Chickens were fed basal diets supplemented with different Se levels (0.25, 0.50, and 1.00 Se mg/kg). To induce NE, Clostridium perfringens (C. perfringens) was orally administered at 14 days of age post hatch. The results showed that birds fed 0.25 Se mg/kg exhibited significantly increased body weight gain compared with the non-supplemented/infected birds. There were no significant differences in gut lesions between the Se-supplemented groups and the non-supplemented group. The antibody levels against α-toxin and NetB toxin increased with the increase between 0.25 Se mg/kg and 0.50 Se mg/kg. Hyperuricemia is prevalent globally and potentially linked to environmental pollution. As a typical persistent organic pollutant, phenanthrene (Phe) poses threats to human health through biomagnification. Although studies have reported Phe-induced toxicities to multiple organs, its impact on uric acid (UA) metabolism remains unclear. In this study, data mining on NHANES 2001-2016 indicated a positive correlation between Phe exposure and the occurrence of hyperuricemia in population. Subsequently, adolescent Balb/c male mice were orally exposed to Phe at a dosage of 10 mg/kg bw every second day for 7 weeks, resulting in dysfunction of intestinal UA excretion and disruption of the intestinal barrier. Utilizing intestinal organoids, 16S rRNA sequencing of gut microbiota, and targeted metabolomic analysis, we further revealed that an imbalance in bile acid metabolism derived from gut microbiota might mediate the intestinal barrier damage. Additionally, the tea extract theabrownin (TB) effectively improved Phe-induced hyperuricemia and intestinal dysfunction at a dose of 320 mg/kg bw per day. In conclusion, this study demonstrates that Phe exposure is positively associated with hyperuricemia and intestinal damage, which provides new insights into the toxic effects induced by Phe. Improving the proton conductivity (σ) of proton exchange membranes at low temperatures is very important for expanding their application areas. Here, sulfonated poly ether ether ketone (SPEEK) membranes were prepared with different sulfonation degrees, and its maximum ion exchange capacity is 3.15 mmol/g for 10 h at 60 °C. Highly sulfonated SPEEK membrane exhibits ultra-high water uptake and excellent proton conductivity of 0.074 S/cm at -25 °C due to its abundant -SO 3 H. Nevertheless, its high swelling ratio and low mechanical strength are not conducive to the practical application of the membrane. Luckily, by employing the chelation of Cu 2+ with -SO 3 - on the SPEEK chain, Cu 2+ -coordinated SPEEK membranes were prepared, and they not only retain high -SO 3 H content but also possess robust mechanical properties and good dimensional stability compared to pristine SPEEK membrane. Meanwhile, the σ of the SPEEK-Cu membrane reaches 0.054 S/cm at -25 °C, and its fuel cell maximum power (W max ) reaches 0.42 W/cm 2 at -10 °C, demonstrating superior low-temperature performance in comparison to other reported materials. Particularly, water states in the prepared membranes are quantified by low-temperature differential scanning calorimetry. Because much more water bound to the plentiful -SO 3 H and Cu 2+ inside the membrane endows it with anti-freezing performance, the decay of the σ and the W max for the SPEEK-Cu membrane is retarded at sub-zero temperatures. Polysaccharides from a medicinal fungus Ganoderma sinense represent important and adjunctive therapeutic agents for treating various diseases, including leucopenia and hematopoietic injury. However, the synthetic accessibility to long, branched, and complicated carbohydrates chains from Ganoderma sinense polysaccharides remains a challenging task in chemical synthesis. Here, we report the modular chemical synthesis of nona-decasaccharide motif from Ganoderma sinense polysaccharide GSPB70-S with diverse biological activities for the first time through one-pot stereoselective glycosylation strategy on the basis of glycosyl ortho -(1-phenyvinyl)benzoates, which not only sped up carbohydrates synthesis but also reduced chemical waste and avoided aglycones transfer issues inherent to one-pot glycosylation on the basis of thioglycosides. The synthetic route also highlights the following key steps: (1) preactivation-based one-pot glycosylation for highly stereoselective constructions of several 1,2- cis -glycosidic linkages, including three α-d-GlcN-(1 → 4) linkages and one α-d-Gal-(1 → 4) bond via the reagent N -methyl- N -phenylformamide modulation; (2) orthogonal one-pot assembly of 1,2- trans -glycosidic linkages in various linear and branched glycans fragments by strategic combinations of glycosyl N -phenyltrifluoroacetimidates, glycosyl ortho -alkynylbenzoates, and glycosyl ortho -(1-phenyvinyl)benzoates; and (3) the final [1 × 4 + 15] Yu glycosylation for efficient assembly of nona-decasaccharide target. Additionally, shorter sequences of 4-mer, 5-mer, and 6-mer are also prepared for structure-activity relationship biological studies. The present work shows that this one-pot stereoselective glycosylation strategy can offer a reliable and effective means to streamline chemical synthesis of long, branched, and complex carbohydrates with many 1,2- cis -glycosidic bonds.

Bladder urothelial carcinoma (BLCA) presents a persistent challenge in clinical management. Despite recent advancements demonstrating the BLCA efficacy of immune checkpoint inhibitors (ICI) in BLCA patients, there remains a critical need to identify and expand the subset of individuals who benefit from this treatment. Mitochondria, as pivotal regulators of various cell death pathways in eukaryotic cells, exert significant influence over tumor cell fate and survival. In this study, our objective was to investigate biomarkers centered around mitochondrial function and cell death mechanisms to facilitate prognostic prediction and guide therapeutic decision-making in BLCA. Utilizing ssGSEA and LASSO regression, we developed a prognostic signature termed mitochondrial function and cell death (mtPCD). Subsequently, we evaluated the associations between mtPCD score and diverse clinical outcomes, including prognosis, functional pathway enrichment, immune cell infiltration, immunotherapy response analysis and drug sensitivity, within high- and low-risk subgroups. Additionally, we employed single-cell level functional assays, RT-qPCR, and immunohistochemistry to validate the differential expression of genes comprising the mtPCD signature. The mtPCD signature comprises a panel of 10 highly influential genes, strongly correlated with survival outcomes in BLCA patients and exhibiting robust predictive capabilities. Importantly, individuals classified as high-risk according to mtPCD score displayed a subdued overall immune response, characterized by diminished immunotherapeutic efficacy. In summary, our findings highlight the development of a novel prognostic signature, which not only holds promise as a biomarker for BLCA prognosis but also offers insights into the immune landscape of BLCA. Currently, there is a lack of relevant research on the efficacy difference between SHD combined with IBG and PVIBGT in the treatment of osteonecrosis of the femoral head(ONFH). Firstly, this study intends to compare the effectiveness of surgical hip dislocation combined with impacting bone grafts (SHD-IBG) and pedicled vascularised iliac bone graft transfer (PVIBGT) in treating ONFH. And the study investigates patients who suffered from hip preservation failures from both groups to better comprehend failure reasons. 30 patients (34 hips) with ARCO stage IIIA femoral head necrosis were selected between January 2012 and July 2022. They were divided into group A(SHD-IBG) and group B (PVIBGT) according to different surgical methods. Firstly, compared the 1-year effect between SHD-IBG and PVIBGT at 1 year postoperatively; Secondly, assessed the medium and long-term efficacy of SHD-IBG hip preservation treatment; Lastly, based on study of the femoral head removed from patients with hip preservation failure in the two groups, the reasons for the failure of hip preservation were comprehensively analyzed in the two groups. Group A: 11 males (13 hips), 4 females (4 hips);Group B: 9 males (11 hips), 6 females (6 hips).Firstly, the average Harris scores of the two groups at 1 year after surgery: preoperative: 70.7, 1 year after surgery: 78.9 in group A; preoperative: 69.5, 1 year after surgery: 81.5 in group B. The differences were statistically significant (P < 0.05).Compared to the preoperative period, quantitative analysis by DCE-MRI showed an increase in perfusion in the necroticarea and an improvement in hyperperfusion in the repair-responsive area one year after the surgery. Secondly, in group A, the hip preservation rate was 88.2% at 2.5-11 (average of 77 months) years of follow-up, and the mean Harris score at the last follow-up was 73.2.Semi-quantitative analysis of postoperative DCE-MRI showed that the perfusion curves of necrotic and repaired areas were similar to those of the normal area. This suggests the instability within the femoral head had been effectively improved, and the perfusion had partially recovered. Thirdly, according to Micro-CT and pathologica studies of patients with hip preservation failure in these two groups, all these patients' femoral head was significantly collapsed and deformed. Their trabeculae was thin and partially disorganized, with fractures in the subchondral bone and separation of the cartilage from the subchondral bone. The necrotic areas had sparse trabeculae, disorganized arrangement, loss of continuity, and disappearance of cells in the trabecular traps. The necrotic area was covered with fibrous tissue, and partial restoration was observed in the repair area. Mechanical finite element analysis showed that the maximum equivalent force was observed in the weight- bearing area and the cortical bone surrounding the shaft of femurand. The result of DCE-MRI showed that the repair reaction area exhibited abnormal hyperperfusion. In this study, the efficacy of SHD-IBG and PVIBGT was compared at 1 year after operation, and the long-term follow-up of SHD-IBG was 2.5-11 (mean 77 months) years, combined with DCE-MRI results, we found that the short-term effect of PVIBGT was more significant than that of SHD-IBG. Esketamine is a widely used intravenous general anesthetic. However, its safety, particularly its effects on the heart, is not fully understood. In this study, we investigated the effects of esketamine exposure on zebrafish embryonic heart development. Zebrafish embryos were exposed to esketamine at concentrations of 1, 10, and 100 mg/L from 48 h post-fertilization (hpf) to 72 hpf. We found that after exposure, zebrafish embryos had an increased hatching rate, decreased heart rate, stroke volume, and cardiac output. When we exposed transgenic zebrafish of the Tg( cmlc2:EGFP ) strain to esketamine, we observed ventricular dilation and thickening of atrial walls in developing embryos. Additionally, we further discovered the abnormal expression of genes associated with cardiac development, including nkx2.5 , gata4 , tbx5 , and myh6 , calcium signaling pathways, namely ryr2a , ryr2b , atp2a2a , atp2a2b , slc8a3 , slc8a4a , and cacna1aa , as well as an increase in acetylcholine concentration. In conclusion, our findings suggest that esketamine may impair zebrafish larvae's cardiac development and function by affecting acetylcholine concentration, resulting in weakened cardiac neural regulation and subsequent effects on cardiac function. The insights garnered from this research advocate for a comprehensive safety assessment of esketamine in clinical applications.

Background: Substances that can efficiently enhance skin penetration while exerting no adverse effect are useful for drug and cosmetics formulation., Objective: To investigate the safety and enhance skin penetration efficacy of Putocrin®, a combination containing 2% isosorbide dimethyl ether, 1% pentanediol, and 0.5% inositol., Methods: An in vitro keratinocyte cell assay using 3-(4,5-dimethylthiazolyl-2)-2,5 diphenyltetrazolium bromide (MTT), and an in vitro EpiKutis® skin study adopted hematoxylin and eosin staining, immunostaining, and liquid chromatography-mass spectrometry (LC-MS) analysis were carried out to investigate the safety of Putocrin®. A pigskin-Franz cell system experiment applied high-performance liquid chromatography (HPLC) to compare the skin penetration efficiency of fluorescein isothiocyanate (Fitc)-labeled tranexamic acid with or without the assistance of Putocrin®. The safety and efficacy of Putocrin® was further evaluated on zebrafish embryos., Results: The MTT assay showed that Putocrin® at concentration ≤2.5% did not significantly affect cell viability. The in vitro EpiKutis® skin study revealed that 2.5% Putocrin® did not affect skin morphology, filaggrin content, ceramide/protein, or fatty acid/protein ratios, but significantly increased loricrin content by 86.00% (p < 0.001). The pigskin-Franz cell penetration experiment demonstrated that Fitc-labeled tranexamic acid could barely penetrate the skin (with penetration rate of 1.121%), while Putocrin® significantly enhanced the penetration rate up to 83.983%, which was close to unlabeled tranexamic acid (90.013%). The zebrafish embryo study showed that 2.5% Putocrin® did not exert observable toxicity and obviously assisted the skin penetration of Fitc-labeled tranexamic acid into fish embryos. Graph Convolutional Networks (GCN) have shown outstanding performance in skeleton-based behavior recognition. However, their opacity hampers further development. Researches on the explainability of deep learning have provided solutions to this issue, with Class Activation Map (CAM) algorithms being a class of explainable methods. However, existing CAM algorithms applies to GCN often independently compute the contribution of individual nodes, overlooking the interactions between nodes in the skeleton. Therefore, we propose a game theory based class activation map for GCN (GT-CAM). Firstly, GT-CAM integrates Shapley values with gradient weights to calculate node importance, producing an activation map that highlights the critical role of nodes in decision-making. It also reveals the cooperative dynamics between nodes or local subgraphs for a more comprehensive explanation. Secondly, to reduce the computational burden of Shapley values, we propose a method for calculating Shapley values of node coalitions. Lastly, to evaluate the rationality of coalition partitioning, we propose a rationality evaluation method based on bipartite game interaction and cooperative game theory. Additionally, we introduce an efficient calculation method for the coalition rationality coefficient based on the Monte Carlo method. Experimental results demonstrate that GT-CAM outperforms other competitive interpretation methods in visualization and quantitative analysis.

Sodium-ion batteries (SIBs), as the next-generation high-performance electrochemical energy storage devices, have attracted widespread attention due to their cost-effectiveness and wide geographical distribution of sodium. As a crucial component of the structure of SIBs, the anode material plays a crucial role in determining its electrochemical performance. Significantly, metal phosphide exhibits remarkable application prospects as an anode material for SIBs because of its low redox potential and high theoretical capacity. However, due to volume expansion limitations and other factors, the rate and cycling performance of metal phosphides have gradually declined. To address these challenges, various viable solutions have been explored. In this paper, the recent research progress of metal phosphide materials for SIBs is systematically reviewed, including the synthesis strategy of metal phosphide, the storage mechanism of sodium ions, and the application of metal phosphide in electrochemical aspects. The multispecies biofilm is a naturally occurring and dominant lifestyle of bacteria in nature, including in rhizosphere soil, although the current understanding of it is limited. Here, we provide an approach to rapidly establish synergistic multispecies biofilm communities. The first step is to extract cells from rhizosphere soil using the differential centrifugation method. Afterward, these soil cells are inoculated into the culture medium to form pellicle biofilm. After 36 h of incubation, the bacterial composition of the biofilm and the solution underneath are determined using the 16S rRNA gene amplicon sequencing method. Meanwhile, high-throughput bacterial isolation from pellicle biofilm is conducted using the limiting dilution method. Then, the top 5 bacterial taxa are selected with the highest abundance in the 16S rRNA gene amplicon sequencing data (pellicle biofilm samples) for further use in constructing multispecies biofilm communities. All combinations of the 5 bacterial taxa were quickly established using a 24-well plate, selected for the strongest biofilm formation ability by the crystal violet staining assay, and quantified by qPCR. Finally, the most robust synthetic bacterial multispecies biofilm communities were obtained through the methods above. This methodology provides informative guidance for conducting research on rhizosphere multispecies biofilm and identifying representative communities for studying the principles governing interactions among these species.

Objective: In this paper, a single-hand-operated hepatic pedicle clamp was introduced, and its application value in laparoscopic liver tumor resection was preliminarily discussed., Methods: The clinical data of 67 patients who underwent laparoscopic liver tumor resection at the First Affiliated Hospital of Wannan Medical College from March 2019 to October 2023 were retrospectively analyzed. The Pringle maneuver was performed with a hepatic pedicle clamp during the operation. The preoperative, intraoperative and postoperative clinical data were observed and recorded., Results: Sixty-seven patients had a median block number, block time, intraoperative blood loss, and postoperative length of hospital stay of 4, 55 min, 400 ml, and 7 days, respectively. The average operation time was 304.9±118.4 min, the time required for each block was 3.2±2.4 s, and the time required for each removed block was 2.6±0.7 s. None of the patients developed portal vein thrombosis or hepatic artery aneurysm formation., Conclusion: The hepatic pedicle clamping clamp is simple to use in laparoscopic hepatectomy, optimizes the operation process, and has a reliable blocking effect. Background: Accurate estimation of the glomerular filtration rate (GFR) is clinically crucial for determining the status of obstruction, developing treatment strategies, and predicting prognosis in obstructive nephropathy (ON). We aimed to develop a deep learning-based system, named UroAngel, for non-invasive and convenient prediction of single-kidney function level., Methods: We retrospectively collected computed tomography urography (CTU) images and emission computed tomography diagnostic reports of 520 ON patients. A 3D U-Net model was used to segment the renal parenchyma, and a logistic regression multi-classification model was used to predict renal function level. We compared the predictive performance of UroAngel with the Modification of Diet in Renal Disease (MDRD), Chronic Kidney Disease Epidemiology Collaboration (CKD-EPI) equations, and two expert radiologists in an additional 40 ON patients to validate clinical effectiveness., Results: UroAngel based on 3D U-Net convolutional neural network could segment the renal cortex accurately, with a Dice similarity coefficient of 0.861. Using the segmented renal cortex to predict renal function stage had high performance with an accuracy of 0.918, outperforming MDRD and CKD-EPI and two radiologists., Conclusions: We proposed an automated 3D U-Net-based analysis system for direct prediction of single-kidney function stage from CTU images. Autoimmune hepatitis (AIH) is a severe immune-mediated inflammatory liver disease that currently lacks feasible drug treatment methods. Our study aimed to evaluate the protective effect of succinic acid against AIH and provide a reliable method for the clinical treatment of AIH. We performed an in vivo study of the effects of succinic acid on concanavalin A (ConA)-induced liver injury in mice. We examined liver transaminase levels, performed hematoxylin and eosin (HE) staining, and observed apoptotic phenotypes in mice. We performed flow cytometry to detect changes in the number of neutrophils and monocytes, and used liposomes to eliminate the liver Kupffer cells and evaluate their role. We performed bioinformatics analysis, reverse transcription-quantitative polymerase chain reaction (RT-qPCR), and western blotting to detect mitochondrial apoptosis-induced changes in proteins from the B-cell lymphoma 2(Bcl-2) family. Succinic acid ameliorated ConA-induced AIH in a concentration-dependent manner, as reflected in the survival curve. HE and TUNEL staining and terminal deoxynucleotidyl transferase dUTP nick end labeling revealed decreased alanine transaminase and aspartate aminotransferase levels, and reduced liver inflammation and apoptosis. RT-qPCR and enzyme-linked immunosorbent assay revealed that succinic acid significantly reduced liver pro-inflammatory cytokine levels. Flow cytometry revealed significantly decreased levels of liver neutrophils. Moreover, the protective effect of succinic acid disappeared after the Kupffer cells were eliminated, confirming their important role in the effect. Bioinformatics analysis, RT-qPCR, and western blotting showed that succinic acid-induced changes in proteins from the Bcl-2 family involved mitochondrial apoptosis, indicating the molecular mechanism underlying the protective effect of succinic acid. Succinic acid ameliorated ConA-induced liver injury by regulating immune balance, inhibiting pro-inflammatory factors, and promoting anti-apoptotic proteins in the liver. Growth hormone inducible transmembrane protein (GHITM), one member of Bax inhibitory protein-like family, has been rarely studied, and the clinical importance and biological functions of GHITM in kidney renal clear cell carcinoma (KIRC) still remain unknown. In the present study, we found that GHITM was downregulated in KIRC. Aberrant GHITM downregulation related to clinicopathological feature and unfavourable prognosis of KIRC patients. GHITM overexpression inhibited KIRC cell proliferation, migration and invasion in vitro and in vivo. Mechanistically, GHITM overexpression could induce the downregulation of Notch1, which acts as an oncogene in KIRC. Overexpression of Notch1 effectively rescued the inhibitory effect induced by GHITM upregulation. More importantly, GHITM could regulate PD-L1 protein abundance and ectopic overexpression of GHITM enhanced the antitumour efficiency of PD-1 blockade in KIRC, which provided new insights into antitumour therapy. Furthermore, we also showed that YY1 could decrease GHITM level via binding to its promoter. Taken together, our study revealed that GHITM was a promising therapeutic target for KIRC, which could modulate malignant phenotype and sensitivity to PD-1 blockade of renal cancer cells via Notch signalling pathway., (© 2024 The Authors. The radioresistant signature of colorectal cancer (CRC) hampers the clinical utility of radiotherapy. Here, we find that fecal microbiota transplantation (FMT) potentiates the tumoricidal effects of radiation and degrades the intertwined adverse events in azoxymethane (AOM)/dextran sodium sulfate (DSS)-induced CRC mice. FMT cumulates Roseburia intestinalis (R. intestinalis) in the gastrointestinal tract. Oral gavage of R. intestinalis assembles at the CRC site and synthetizes butyrate, sensitizing CRC to radiation and alleviating intestinal toxicity in primary and CRC hepatic metastasis mouse models. R. intestinalis-derived butyrate activates OR51E1, a G-protein-coupled receptor overexpressing in patients with rectal cancer, facilitating radiogenic autophagy in CRC cells. OR51E1 shows a positive correlation with RALB in clinical rectal cancer tissues and CRC mouse model. Blockage of OR51E1/RALB signaling restrains butyrate-elicited autophagy in irradiated CRC cells. In mammals, many retrotransposons are de-repressed during zygotic genome activation (ZGA). However, their functions in early development remain elusive largely due to the challenge to simultaneously manipulate thousands of retrotransposon insertions in embryos. Here, we applied CRISPR interference (CRISPRi) to perturb the long terminal repeat (LTR) MT2&#95;Mm, a well-known ZGA and totipotency marker that exists in ∼2,667 insertions throughout the mouse genome. CRISPRi robustly perturbed 2,485 (∼93%) MT2&#95;Mm insertions and 1,090 (∼55%) insertions of the closely related MT2C&#95;Mm in 2-cell embryos. Remarkably, such perturbation caused downregulation of hundreds of ZGA genes and embryonic arrest mostly at the morula stage. The objective of this study was to investigate the potential associations between serum iron levels, dietary iron intake, and iron supplementation, and the prevalence of metabolic syndrome (MetS) in adolescents A cross-sectional analysis was conducted, utilizing data from adolescents participating in the 2003-2018 cycle of the National Health and Nutrition Examination Survey (NHANES). Odds ratios (ORs) and their corresponding 95% confidence intervals (CIs) pertaining to serum iron, dietary iron, and iron supplementation were derived through multivariate logistic regression models. Additionally, a restricted cubic spline (RCS) regression model was applied to explore the nonlinear relationship between dietary iron and serum iron concerning MetS. The study encompassed 4858 American adolescents aged 12 to 19, among whom 413 (8.5%) manifested MetS. The study cohort exhibited an average age of 15.52 years, comprising 2551 males (52.51%) and 2307 females (47.49%). Relative to individuals in the lowest serum iron quartile, those in the highest quartile for serum iron (OR = 0.33, 95% CI 0.21-0.50), the highest quartile for dietary iron (OR = 0.53, 95% CI 0.32-0.89), and those utilizing iron supplements (OR = 0.61, 95% CI 0.37-0.99) evinced a diminished prevalence of MetS, even post adjustment for potential confounding variables. A non-linear relationship was discerned between serum iron and MetS, exhibiting a statistically significant negative correlation when serum iron concentrations exceeded the inflection point (serum iron = 8.66 µmol/L, P for nonlinear < 0.001). This investigation reveals that higher levels of serum iron, increased dietary iron intake, and the use of iron supplements are linked to a lower prevalence of MetS in US adolescents. Chronic postsurgical pain may have a substantial impact on patient's quality of life, and has highly heterogenous presentation amongst sufferers. We aimed to explore the risk factors relating to chronic pain and the related miRNA phenotypes in patients with lung adenocarcinoma after video-assisted thoracoscopic lobectomy to identify potential biomarkers. Our prospective study involved a total of 289 patients with early invasive adenocarcinoma undergoing thoracoscopic lobotomy and a follow-up period of 3 months after surgery. Blood was collected the day before surgery for miRNA detection and patient information including operation duration, duration of continuous drainage of the chest, leukocyte count before and after operation, and postoperative pain scores were recorded. Using clinical and biochemical information for each patient, the risk factors for chronic postsurgical pain and related miRNA phenotypes were screened. We found that chronic postsurgical pain was associated with higher body mass index; greater preoperative history of chronic pain; longer postoperative drainage tube retention duration; higher numerical rating scale scores one, two, and three days after surgery; and changes in miRNA expression, namely lower expression of miRNA 146a-3p and higher expression of miRNA 550a-3p and miRNA 3613-3p in peripheral blood (p < 0.05). Of these factors, patient body mass index, preoperative history of chronic pain, average numerical rating scale score after operation, and preoperative peripheral blood miRNA 550a-3P expression were independent risk factors for the development of chronic postsurgical pain. With the advent of lithium-ion batteries (LIBs), the selection and application of electrode materials have been the subject of much discussion and study. Among them, graphite has been widely investigated for use as electrode materials in LIBs due to its abundant resources, low cost, safety and electrochemical diversity. While it is commonly recognized that conventional graphite materials utilized for commercial purposes have a limited theoretical capacity, there has been a steady emergence of new and improved carbonaceous materials for use as anodes in light of the progressive development of LIBs. In this paper, the latest research progress of various carbon materials in LIBs is systematically and comprehensively reviewed. Firstly, the rocking chair charging and discharging mechanism of LIBs is briefly introduced in this paper, using graphite anodes as an example. After that, the general categories of carbonaceous materials are highlighted, and the recent research on the recent progress of various carbonaceous materials (graphite-based, amorphous carbon-based, and nanocarbon-based) used in LIB anodes is presented separately based on the classification of the structural morphology, emphasizing the influence of the morphology and structure of carbon-based materials on the electrochemical performance of the batteries. Finally, the current challenges of carbonaceous materials in LIB applications and the future development of other novel carbonaceous materials are envisioned.

All vertebrates organisms produce erythroferrone, a secretory hormone with structure-related functions during iron homeostasis. However, limited knowledge exists regarding the effect of this hormone on the occurrence and progression of cancer. To systematically and comprehensively identify the diverse implications of Erythroferrone (ERFE) in various malignant tumors, we conducted an in-depth analysis of multiple datasets, including the expression levels of oncogenes and target proteins, biological functions, and molecular characteristics. This analysis aimed to assess the diagnostic and prognostic value of ERFE in pan-cancer. Our findings revealed a significant elevation in ERFE expression across 20 distinct cancer types, with notable increases in gastrointestinal cancers. Utilizing the Cytoscape and STRING databases, we identified 35 ERFE-targeted binding proteins. Survival prognosis studies, particularly gastrointestinal cancers indicated by Colon adenocarcinoma (COAD), demonstrated a poor prognosis in patients with high ERFE expression (p < 0.001), consistently observed across various clinical subgroups. Furthermore, the ROC curve underscored the high predictive ability of EFRE for gastrointestinal cancer (AUC >0.9). Understanding the roles and interactions of ERFE in biological processes can also be aided by examining the genes co-expressed with ERFE in the coat and ranking the top 50 positive and negative genes. In the correlation analysis between the ERFE gene and different immune cells in COAD, we discovered that the expression of ERFE was positively correlated with Th1 cells, cytotoxic cells, and activated DC (aDC) abundance, and negatively correlated with Tcm (T central memory) abundance (P < 0.001). in summary, ERFE emerges as strongly associated with various malignant cancers, positioning it as a prospective biological target for cancer treatment. An oxidative cascade [4 + 2] radical cycloaddition/dehydroaromatization reaction of aryl alkenes to access α-aryl substituted naphthalenes under redox-neutral conditions was achieved. This reaction was found to require the addition of [Cu] catalyst along with stoichiometric concentrations of NFSI as a trigger of radical series of steps. Norbornene (NBE), rather than the conventional oxidant, manifested optimal performances as a H-acceptor in this procedure. The results herein might shed encouraging insight into the transition-metal-catalyzed dehydrogenative C-H activation protocols.

Every year, almost 1.15-2.41 million tons of plastic from terrestrial rivers undergo fragmentation under certain conditions and settle in the estuarine delta and shallow marine shelf areas, making this region a "sink" for land-based microplastics. Owing to its fast deposition rate, relatively soft sediment bed, and shallow water depth, the estuarine delta region is prone to liquefaction under high wind and wave conditions. This could potentially release deeply buried microplastic particles during the liquefaction process, posing further threats to marine ecology and human health. To investigate this phenomenon, laboratory experiments were conducted using a water tank to simulate wave-induced liquefaction of sediment beds. The results showed that under the influence of wave-induced liquefaction, 56.2 % of microplastic particles were released back into the sediment surface, with larger particles being released to a greater extent. The Sabatier principle suggests that an excessive adsorption of lithium polysulfides (LiPSs) by metal compounds may hinder their conversion in the absence of a conversion module. Therefore, it is imperative to establish a synergetic effect mechanism between "strong adsorption" and "rapid conversion" for LiPSs. To achieve this coexistence, a molybdenum-doped MnS/MnO@C porous structure is designed as a multifunctional coating on the polypropylene (PP) separator. The incorporation of MnS/MnO@C enhances the adsorption capacity towards LiPSs, while molybdenum facilitates subsequent conversion. Benefiting from the synergistic effect of each component and its large specific surface area, the cell with Mo-doped MnS/MnO@C coating achieves smooth adsorption-diffusion-conversion processes and exhibits an appreciable rate performance with outstanding cycling stability. Even when sulfur loading increases to 9.68 mg cm -2 , the modified battery delivers an excellent initial areal capacity of 11.69 mAh cm -2 and maintains 6.97 mAh cm -2 after 50 cycles at 0.1 C. Unsupervised representation learning (URL) that learns compact embeddings of high-dimensional data without supervision has achieved remarkable progress recently. However, the development of URLs for different requirements is independent, which limits the generalization of the algorithms, especially prohibitive as the number of tasks grows. For example, dimension reduction (DR) methods, t-SNE and UMAP, optimize pairwise data relationships by preserving the global geometric structure, while self-supervised learning, SimCLR and BYOL, focuses on mining the local statistics of instances under specific augmentations. To address this dilemma, we summarize and propose a unified similarity-based URL framework, GenURL, which can adapt to various URL tasks smoothly. In this article, we regard URL tasks as different implicit constraints on the data geometric structure that help to seek optimal low-dimensional representations that boil down to data structural modeling (DSM) and low-dimensional transformation (LDT). Specifically, DSM provides a structure-based submodule to describe the global structures, and LDT learns compact low-dimensional embeddings with given pretext tasks. Moreover, an objective function, general Kullback-Leibler (GKL) divergence, is proposed to connect DSM and LDT naturally. Comprehensive experiments demonstrate that GenURL achieves consistent state-of-the-art performance in self-supervised visual learning, unsupervised knowledge distillation (KD), graph embeddings (GEs), and DR.

Background: Bladder cancer (BCa) is a common malignancy in the urinary system. Necroptosis, a recently discovered form of programmed cell death, is closely associated with the development and progression of various types of tumors. Targeting necroptosis through anti-cancer strategies has shown potential as a therapy for cancer. We aimed to develop a necroptosis-related lncRNAs (NRlncRNAs) risk model that can predict the survival and tumor immunity of BCa patients., Methods: We analyzed sequencing data obtained from the TCGA database, and applied least absolute shrinkage and selection operator (LASSO) and Cox regression analysis to identify crucial NRlncRNAs for building a risk model. Using the risk score, we categorized patients into high- and low-risk groups, and assessed the accuracy with the area under the receiver operating characteristic (AUROC) and Kaplan-Meier curves. We performed the RT-qPCR to detect the expression differences of the genes based on the risk model., Results: We identified a total of 296 NRlncRNAs, and 6 of them were included in the prognostic model. The AUC values for 1-, 3-, and 5-year predictions were 0.675, 0.726 and 0.734, respectively. Our risk model demonstrated excellent predictive performance and served as an independent predictor with high predictive power. Additionally, we performed PCA, TMB, GSEA analyses, and evaluated immune cell infiltration, to reveal significant differences between the high- and low-risk groups in functional signaling pathways, immunological status, and mutation profiles. Finally, we assessed the chemotherapeutic response of several drugs. According to the RT-qPCR results, we found that four NRlncRNAs of the risk model were more highly expressed in BCa cell lines than human immortalized uroepithelial cell line and regulated the occurrence and progression of bladder cancer., Conclusion: We constructed a novel NRlncRNAs-associated risk model, which could predict the prognosis and immune response of BCa patients.

Renal fibrosis is considered to be the ultimate pathway for various chronic kidney disease, with a complex etiology and great therapeutic challenges. Tripartite motif-containing (TRIM) family proteins have been shown to be involved in fibrotic diseases, but whether TRIM39 plays a role in renal fibrosis remain unexplored. In this study, we investigated the role of TRIM39 in renal fibrosis and its molecular mechanism. TRIM39 expression was analyzed in patients' specimens, HK-2 cells and unilateral ureteral obstruction (UUO) mice were used for functional and mechanistic studies. We found an upregulated expression of TRIM39 in renal fibrosis human specimens and models. In addition, TRIM39 knockdown was found efficient for alleviating renal fibrosis in both UUO mice and HK-2 cells. Mechanistically, we demonstrated that TRIM39 interacted with PRDX3 directly and induced ubiquitination degradation of PRDX3 at K73 and K149 through the K48 chain, which resulted in ROS accumulation and increased inflammatory cytokine generation, and further aggravated renal fibrosis. Lithium metal batteries are deemed as an optimal candidate for the next generation of durable energy storage devices. However, the growth of lithium dendrite and significant volume expansion pose as obstacles that impede the application of lithium metal batteries. In this work, a functional copper current collector was designed by coating it with Co-doped ZnO (Co/ZnO) to enhance the lithiophilicity through local electric fields and built-in magnetic fields induced by the ferromagnetic material. The incorporation of Co not only induces a local electric field and thus accelerating electron transfer, but also imparts the ferromagnetic behavior to ZnO, resulting in an internal magnetic field to regulate the dynamic trajectory. Profiting from the above advantages, the symmetric cells have excellent cycle stability in 1 mA cm -2 and 1 mAh cm -2 , maintaining ultra-low voltage for over 2000 h. Background: Bladder cancer (BCa) is the most common malignancy of the urinary system. Inflammation is critical in the occurrence and development of BCa. The purpose of this study was to identify key genes and pathways of inflammatory bowel disease in BCa through text mining technology and bioinformatics technology and to explore potential therapeutic drugs for BCa., Methods: Genes associated with BCa and Crohn's disease (CD) were detected using the text mining tool GenClip3, and analyzed using Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG). A protein-protein interaction (PPI) network was constructed by STRING and visualized in Cytoscape, and modular analysis was performed using the Molecular Complex Detection plugin (MCODE). Finally, the genes clustered in the first two modules were selected as core genes, and the drug-gene interaction database was used to discover potential therapeutic drugs., Results: We identified 796 genes shared by "Bladder cancer" and "Crohn's disease" by text mining. Gene function enrichment analysis yielded 18 enriched GO terms and the 6 most relevant KEGG pathways. A PPI network with 758 nodes and 4014 edges was constructed, and 20 gene modules were obtained using MCODE. We selected the top two gene clusters as core candidate genes. We found that 3 out of 55 selected core genes could be targeted by 26 existing drugs., Conclusion: The results indicated that CXCL12, FGF2 and FSCN1 are potential key genes involved in CD with BCa. The pollution of heavy metals such as Cu 2+ is still serious and the discharge of sewage of Cu 2+ will cause damage to soil environment and human health. Herein, a biomass-based solid-state fluorescence detection platform (CPU-CDs) was developed as fluorescent sensor for detection Cu 2+ via fluorescence and colorimetric dual-model methods in real time. CPU-CDs was composed of xylan-derived CDs (U-CDs) and cotton cellulose paper, which exhibiting good reusability, non-toxicity, excellent fluorescence characteristics and high biocompatibility. Further, CPU-CDs displayed high effectiveness and sensitivity for Cu 2+ with the detection limit as low as 0.14 μM, which was well below U.S. EPA safety levels (20 μM). Practical application indicated that CPU-CDs could achieve precision response of Cu 2+ change in real environment water samples with good recovery range of 90 %-119 %.
