(1 → 4)-β-D-Xylans are the second most abundant plant biopolymers on Earth after cellulose. Although their structures have been extensively studied, and industrial applications have been found for them and their derivatives, they are still investigated due to the diversity of their structures and uses. In this work, hemicellulose fractions obtained previously with 1 M KOH from two species of woody bamboos, Phyllostachys aurea and Guadua chacoensis, were purified, and the structures of the glucuronoarabinoxylans (GAX) were studied by chemical and spectroscopic methods. In both cases, major amounts of α-L-arabinofuranose residues were linked to C3 of the xylose units of the backbone, and also α-D-glucuronic acid residues and their 4-O-methyl-derivatives were detected in minor quantities, linked to C2 of some xylose residues. Methylation analysis of the carboxyl-reduced derivative from GAX from P. aurea indicated the presence of terminal and 5-linked arabinofuranose units. NMR spectroscopy showed the presence of disaccharidic side chains of 5-O-α-l-arabinofuranosyl-L-arabinofuranose for the GAX from P. aurea, while for those of G. chacoensis, only single side chains were found. Water-extracted arabinoxylans (WEAXs) of different varieties and structures have important effects on wheat end products. However, the functional performances of WEAXs, particularly relating to prebiotic potential, are not yet clear. The present study compared the structural features, physicochemical properties, and prebiotic potential of WEAXs from three wheat varieties, which were used as special wheat varieties to make steamed buns, bread flour, and noodles. The results showed that WEAX-1, WEAX-2, and WEAX-3, derived from Jinqiang wheat, American red hard spring wheat, and Australian white wheat, respectively, had different structural properties, gelation properties, and prebiotic potential. WEAX-3 had a low arabinose to xylose (A/X) ratio (0.49), high ferulic acid content (2300 μg/g), and excellent gelation capacity. WEAX-2 had a high A/X ratio (0.62), low ferulic acid content (1300 μg/g), and poor gelation capacity. When fermented with human feces, WEAX-3 significantly increased the numbers of bifidobacteria and lactobacilli and increased the production of short-chain fatty acids (SCFAs), while WEAX-2 had weaker effects on the number of beneficial bacteria and SCFAs production (P < 0.05). The physicochemical properties and prebiotic potential of WEAXs depended strongly on their structural properties. WEAX with a low A/X ratio and a high ferulic acid content showed excellent gelation property and a strong prebiotic potential.

Insoluble fermentable cell wall matrix fibers have been shown to support beneficial butyrogenic gut Clostridia, but have restricted use in food products. Here, a soluble fiber matrix was developed that exhibited a similar effect. A low arabinose/xylose ratio corn bran arabinoxylan (CAX) was extracted with two concentrations of sodium hydroxide, 0.25 M and 1.5 M, to give soluble polymers with relatively low (L) and high (H) residual levels of bound ferulic acid (FA) (CAX-LFA and CAX-HFA). After laccase treatment to make diferulate crosslinks, soluble matrices were formed with average 3.5 to 4.5 mer. In vitro human fecal fermentation of CAX-LFA, CAX-HFA, soluble crosslinked ∼3.5 mer CAX-LFA (SCCAX-LFA), and ∼4.5 mer SCCAX-HFA revealed that the SCCAX matrices had somewhat slower fermentation properties by measuring the gas production, total short chain fatty acids, and carbohydrate disappearance, with a higher butyrate proportion in SCCAX-HFA. 16S rRNA gene sequencing showed that SCCAX-HFA promoted OTUs associated with butyrate production including unassigned Ruminococcaceae, unassigned Blautia, Fecalibacterium prausnitzii, and unassigned Clostridium. Thus, when the physical form of an individual soluble polysaccharide was changed to a soluble crosslinked matrix, in vitro fermentation was shifted to Clostridial butyrate producers. This study shows that the physical form of the fiber influences the competition for substrate among the gut bacteria. Crosslinking of soluble fibers may be a strategy for developing soluble matrices with good physical functionalities for beverages and other foods to improve gut health.

Lignocellulosic biomass has great potential for use as a carbon source for the production of second-generation biofuels by solventogenic bacteria. Here we describe the production of butanol by a newly discovered wild-type Clostridium species strain G117 with xylan as the sole carbon source for fermentation. Strain G117 produced 0.86 ± 0.07 g/L butanol and 53.4 ± 0.05 mL hydrogen directly from 60 g/L xylan provided that had undergone no prior enzymatic hydrolysis. After process optimization, the amount of butanol produced from xylan was increased to 1.24 ± 0.37 g/L. In contrast to traditional acetone-butanol-ethanol (ABE) solventogenic fermentation, xylan supported fermentation in strain G117 and negligible amount of acetone was produced. The expression of genes normally associated with acetone production (adc and ctfB2) were down-regulated compared to xylose fed cultures. This lack of acetone production may greatly simplify downstream separation process. Moreover, higher amount of butanol (2.94 g/L) was produced from 16.99 g/L xylo-oligosaccharides, suggesting a major role for strain G117 in butanol production from xylan and its oligosaccharides. The unique ability of strain G117 to produce a considerable amount of butanol directly from xylan without producing undesirable fermentation byproducts opens the door to the possibility of cost-effective biofuels production in a single step. Dietary fibre of quinoa and amaranth was analysed for its insoluble and soluble fibre content, composition, and structure. Total dietary fibre content was 10% for quinoa and 11% for amaranth. For both pseudocereals, 78% of its dietary fibre was insoluble. Insoluble fibre (IDF) from quinoa and amaranth was mainly composed of galacturonic acid, arabinose, galactose, xylose and glucose. Linkage analysis indicated that IDF was composed of homogalacturonans and rhamnogalacturonan-I with arabinan side-chains (∼55-60%), as well as highly branched xyloglucans (∼30%) and cellulose. For both pseudocereals, 22% of total dietary fibre was soluble; a higher proportion than that found in wheat and maize (∼15%). The soluble fibre (SDF) was composed of glucose, galacturonic acid and arabinose; for amaranth, xylose was also a major constituent. Background: Arabinoxylans (AXs) are partially water-extractable polymers that cause problems of viscosity and filterability during beer brewing. This study investigated the effects of Kolbach index (KI) on water-extractable AXs (WEAXs)., Results: KI had positive correlations with extract (r = 0.845) and free amino nitrogen (FAN) (r = 0.958). The contents of malt WEAXs (WEAXsm), total AXs and wort AXs (WAXs) were 10.8-11.6 g kg⁻¹, 57.5-60.8 g kg⁻¹ and 137.5-140.5 mg per 100 mL respectively and the corresponding arabinose/xylose (A/X) ratios were 0.52-0.56, 0.55-0.62 and 0.48-0.50. The molecular weight (Mw) ranged from 4.92 × 10⁴ to 6.93 × 10⁴ Da for WEAXsm and from 1.24 × 10⁴ to 2.90 × 10⁴ Da for WAXs. KI had negative correlations with the average degree of polymerization (avDP) of malt (r = -0.877) and wort (r = -0.978). Wort viscosity showed a positive correlation with the Mw of WEAXsm (Mwm) (r = 0.821)., Conclusion: Increasing KI can promote the degradation of the xylan backbone, while it cannot affect the branching of the polymer chain and the content and Mw of WEAXs., (© 2014 Society of Chemical Industry.)

Two-stage hydrothermal processing was employed to obtain feruloylated arabinoxylooligosaccharides (AXOS) from wheat bran. First, wheat bran in water (10% w/w solids) was heated to 130 degrees C, releasing 36.3% of total solids, 70.3% of starch, and 6.06% of pentose sugars. Wheat bran was then heated to 170-220 degrees C. Heating to 200 and 210 degrees C released the most AXOS (70% of the insoluble arabinoxylan) and esterified ferulate (30% of the initial ferulic acid). Treatment of 200 degrees C retained a higher proportion of high molecular weight (>1,338) compounds than 210 degrees C and was the preferred treatment temperature because autohydrolysate liquors contained lower concentrations of many contaminants. Purification of this autohydrolysate liquor with ethyl acetate extraction, vacuum concentration, and ion exchange resulted in a product containing 32.0% AXOS and 4.77% esterified ferulate, accompanied by 36.0% other oligosaccharides and free sugars, with an antioxidant activity of 29.7 micromol Trolox equivalents/g dry matter.

This study investigated the potential of ball milling as a dry process for in situ production of arabinoxylan oligosaccharides (AXOS) in arabinoxylan (AX)-rich wheat and rye bran. An extensive lab-scale ball mill treatment (120 h, 50% jar volume capacity) increased the wheat bran water-extractable arabinoxylan (WE-AX) level from 4% (untreated bran) to 61% of the wheat bran AX. Extractable AX fragments had an arabinose/xylose ratio (A/X ratio) of 0.72 and a molecular mass (MM) of 15 kDa. Ball milling of rye bran gave rise to similar results, with the A/X ratio of the extractable AX being considerably lower (0.51). Optimization of the ball mill treatment by varying the degree of filling of the milling jar permitted us to obtain equally high WE-AX levels (>70%) in wheat and rye bran within a 24 h ball-milling period. Ball milling at optimal conditions (24 h, 16% jar volume capacity) yielded wheat bran AXOS, with an A/X ratio of 0.65 and a MM of 6 kDa. Ball milling (24 h, 50% jar volume capacity) of pericarp-enriched wheat bran increased the WE-AX level from 1% (untreated pericarp) to 63%. The extractable material had a high A/X ratio (0.97) and a low MM (5 kDa). Fluorescence microscopy revealed that the extensive ball mill treatment led to the almost complete disappearance of discernible tissue structures in the ball-milled material, indicating bran particle size reductions down to the nanoscale level. It further visualized the aggregation of the treated material. These results show that AXOS can be produced in situ from wheat or rye bran in a single-stage dry milling process, rendering a wet extraction step redundant. The higher A/X ratio of the obtained AXOS than of enzymically produced wheat bran-derived AXOS offers perspectives for the production of a wide range of AXOS structures. Moreover, ball milling makes upgrading of the low-value pericarp layer feasible.

Rye arabinoxylan, with an initial arabinose to xylose (Ara/Xyl) ratio of 0.50, was enzymatically modified with alpha-L-arabinofuranosidase. Different enzyme dosages were used to prepare arabinoxylan samples with a gradient of arabinose content varying from Ara/Xyl ratio 0.50 to 0.20. The degree of polymerization of the arabinoxylans was not affected by the enzymatic treatment, as detected with SEC-MALLS. Arabinoxylan samples with an Ara/Xyl ratio of 0.30 and below agglomerated in a water solution as seen by changes in light scattering. All samples, however, formed cohesive films upon drying, without addition of external plasticizers. The film from untreated arabinoxylan was completely amorphous; whereas films of the enzyme-treated arabinoxylans were semicrystalline with an increasing degree of crystallinity with decreasing arabinose content as determined by WAXS. Oxygen permeability measurements of the films showed that decreased arabinose content also resulted in lower oxygen permeability of the films. All films were strong and relatively stiff, but showed variations in strain at break. The moderately debranched film with an Ara/Xyl ratio of 0.37 had highest strain at break among all the films tested, yet was stiff and strong. This material also exhibited yielding and had stress/strain behavior similar to synthetic semicrystalline polymers, with a tendency to strain-induced crystallization. Such a combination of mechanical properties combined with oxygen barrier properties is very attractive for packaging applications.

The impact of varying levels of endoxylanase activity in wheat flour on arabinoxylan (AX) in mixed and rested dough was studied using eight industrially milled wheat flour fractions with varying endoxylanase activity levels. Analysis of the levels of reducing end xylose (RX) and solubilized AX (S-AX) formed during mixing and resting and their correlation with the endoxylanase activity in the flour milling fractions showed that solubilization of AX during the mixing phase is mainly due to mechanical forces, while solubilization of AX during resting is caused by endoxylanase activity. Moreover, solubilization of AX during the dough resting phase is more outspoken than that during the mixing phase. Besides endoxylanase activity, there were significant xylosidase and arabinofuranosidase activities during the dough resting phase. The results indicate that wheat flour-associated endoxylanases can alter part of the AX in dough, thereby changing their functionality in bread making and potentially affecting dough and end product properties.

Samples of Arundo donax were subjected to isothermal autohydrolysis, defined by temperature, 150-195 degrees C; time, 0-15 h; and liquor to solid ratio, 8 g/g. The effect of the operational variables on the yield and composition of both liquid and solid phases obtained after the treatments has been studied. The oligomer concentration and composition have been determined. In the conditions leading to maximum oligomers concentration (defined by dimensionlees time theta=1) it can be produced up to 17.7 g oligomers/100 g raw material and four acetyl groups/10 xylose monomers. These oligomers are the mean of 50% of nonvolatile compounds. In these conditions, cellulose is almost quantitatively retained in the solid phase, whereas lignin is solubilized at 9%.

Aims: It is evaluated the effectiveness of the combined action of two highly thermostable enzymes for the hydrolysis of xylans at high temperature in order to produce D-xylose., Methods and Results: Xylans from different sources were hydrolyzed at high degree at 70 degrees C by co-action of a xylanase from the thermophilic bacterium Anoxybacillus flavithermus BC and the novel beta-xylosidase/alpha-arabinosidase from the hyperthermophilic crenarchaeon Sulfolobus solfataricus Oalpha. Beechwood xylan was the best substrate among the xylans tested giving, by incubation only with xylanase, 32.8 % hydrolysis after 4 h. The addition of the beta-xylosidase/alpha-arabinosidase significantly improved the rate of hydrolysis, yielding 63.6% conversion after 4 h incubation, and the main sugar identified was xylose., Conclusions: This study demonstrates that a significant degree of xylan degradation was reached at high temperature by co-action of the two enzymes. Xylose was obtained as a final product in considerable yield., Significance and Impact of the Study: Although the xylan represents the second most abundant polysaccharide in nature, it still doesn't have significant utilization for the difficulties encountered in its hydrolysis. Its successful hydrolysis to xylose in only one stage process could make of it a cheap sugar source and could have an enormous economic potential for the conversion of plant biomass into fuels and chemicals.

Cryptococcus adeliensis was initially described as a psycrophilic species containing a single strain CBS 8351(T) isolated from decayed algae in Terre Adelie (Antartida). Later, a second strain of this species was isolated from an immunosuppressed patient affected by leukaemia in Germany and recently several strains from this species have been found in human patients and pigeon droppings of the same country. In this study, we isolated from sheep droppings in Spain a xylanolytic strain named LEVX01 that was phenotypically related to the strain CBS 8351(T) and showed a 100% similarity in the D1/D2 domain and 5.8S-ITS region sequences with respect to the remaining described strains of C. adeliensis. These findings suggest that this species has a wide geographical distribution and that the animal faeces are a common habitat for C. adeliensis. The chemotaxonomic analyses showed the absence of detectable amounts of xylose in the cell walls of the strains LEVX01 and CBS8351(T) in contrast to other Cryptococcus species. Interestingly, the ultrastructural study showed the presence of fimbriae in these two strains that could be involved in the attachment to the host cells and, as occurs in Candida albicans, they could also be a pathogenicity factor for the man.

The polysaccharide isolated from the gum exudate of palm Scheelea phalerata (SPN) was water-insoluble and composed of Fuc, Ara, Xyl, and uronic acid moieties in a 5:34:54:7 molar ratio: 12% of phenolics were also present. A soluble polysaccharide (SPNa) was obtained after alkaline treatment, which contained Fuc, Ara, Xyl and uronic acid in a 7:44:42:7 molar ratio, with only 2% phenolics. SPNa had an M(W) approximately 1.04 x 10(5) g mol(-1) and was almost monodisperse (M(W)/M(N) : 1.25 +/-0.22). It had a branched structure with side chains of 2-O-substituted Xylp (approximately 8%) and 3-O-substituted Araf (12%) units, and a large proportion of nonreducing end-units of Araf (15%), Fucp (10%), Xylp (4%), and Arap (6%). The (1 --> 4)-linked beta-Xylp main-chain units were 3-O- (9%), 2-O- (13%), and 2,3-di-O- (13%) substituted. Its (13)C NMR spectrum contained at least 9 C-1 signals, those at delta 108.6 and 107.7 arising from alpha-Araf units. Others were present at delta 175.4 from C-6 of alpha-GlcpA and delta 15.6 from C-6 of Fucp units. The main chain of SPNa was confirmed by analysis of a Smith-degraded polysaccharide (SPDS): methylation analysis provided a 2,3-Me(2)-Xyl (65%) derivative and its (13)C NMR spectrum showed five main signals typical of a (1 --> 4)-linked beta-Xylp units. Methylation analysis of a carboxy-reduced polysaccharide (SPN-CR) revealed a 2,3,4,6-Me(4)-Glc derivative (4%) arising from nonreducing end-units of GlcpA. Alpha-GlcpA-(1 --> 2)-alphabeta-Xy1p and alpha-GlcpA-(1 --> 2)-beta-Xylp-(1 --> 4)-alphabeta-Xylp were obtained via partial acid hydrolysis of SPN, showing the structure of side-chain substituents on O-2 of the main-chain units.

High-throughput screening requires simple assays that give reliable quantitative results. A microplate assay was developed for reducing sugar analysis that uses a 2,2'-bicinchoninic-based protein reagent. Endo-1,4-beta-D-xylanase activity against oat spelt xylan was detected at activities of 0.002 to 0.011 IU ml(-1). The assay is linear for sugar concentrations from 0 to 86 microg ml(-1) and can also be used to assay protein concentrations (0 to 143 microg ml(-1)) on the same plate. A variety of temperatures and pH conditions can be used and, after incubation, the assay requires only one detection reagent and one heating step.

Partially depolymerized, water-soluble hemicelluloses were solubilized from flax shive employing hydrothermal microwave treatment and thereafter subjected to ion-exchange chromatography, enzymatic purification and/or size-exclusion chromatography (SEC). The oligo- and polysaccharide fractions thus obtained were characterized with respect to molar mass, molar mass distribution, degree of polymerization (DP) and degree of substitution with acetyl moieties (DSAc) by employing SEC in combination with MALDI-TOF mass spectrometry. The major portion of the water-soluble flax hemicellulose consisted of an O-acetyl-4-O-methylglucuronoxylan exhibiting a DPp value (i.e., peak-average DP) of 28. When the DSAc for this O-acetyl-4-O-methylglucuronoxylan was calculated on the basis of the MALDI-MS spectra obtained without and following deacetylation, a value of 0.7 was obtained. In addition, an O-acetyl-glucomannan (DPp=9, DS=0.4) and minor quantities of small neutral O-acetyl-xylooligosaccharides were also isolated from the mixture of water-soluble hemicelluloses released from the flax shive by microwave treatment.

Structural features of the acidic, highly substituted glycanoxylan (LCP; 87% yield) from the gum exudate of the palm, Livistona chinensis, family Arecaceae, were determined. It had [alpha]D -30 degrees, Mw 1.9x10(5) and a polydispersity ratio Mw/Mn of approximately 1.0. Acid hydrolysis gave rise to Rha, Fuc, Ara, Xyl, and Gal, in a 1:6:46:44:3 molar ratio, and 12% of uronic acid was present. LCP had a highly branched structure with side-chains containing nonreducing end-units (% values are approximate) of Araf (15%), Fucp (4%), Xylp (7%), GlcpA, and 4-Me-GlcpA, and internal 2-O- (5%) and 3-O-substituted Araf (8%), and 2-O-substituted Xylp (14%) units. The (1-->4)-linked beta-Xylp main-chain units of LCP were substituted at O-3 (4%), O-2 (17%), and O-2,3 (16%). Partial acid hydrolysis gave 4-Me-alpha-GlcpA-(1-->2)-[beta-Xylp-(1-->4)](0-2)-Xyl, identified by showing that the uronic acids were single-unit side-chain substituents on O-2. Milder hydrolysis conditions removed from O-3 other side-chains containing Fucp and Araf nonreducing end-units and internal Arap, and 2-O- and 3-O-substituted Araf units. Carboxyl-reduced LCP contained 4-O-methylglucose and glucose in a 3.2:1 molar ratio, arising from GlcpA and 4-OMe-GlcpA nonreducing end-units, respectively. The gum contained small amounts of free alpha-Fucp-(1-->2)-Ara, which corresponds to structures in the polysaccharide. Free myo- and D- or L-chiro-inositol were present in a 9:1 ratio.

The water extractable (WE) arabinoxylans from two rye flours differing in baking quality were studied following sequential extraction with water at 4, 40, and 100 degrees C. Ammonium sulfate fractionation of the resulting WE fractions and subsequent analysis revealed substantial differences in the structure of the isolated materials. Furthermore, it allowed us to identify the factors contributing to arabinoxylan water extractability. Our results provide compelling evidence for the existence of separate polymers in rye WE arabinoxylans with different substitution degrees, ranging from quantitatively dominating, lowly substituted populations (arabinose to xylose ratio, Ara/Xyl approximately 0.5) to comparatively less abundant, highly substituted analogues (Ara/Xyl approximately 1.3). Generally, arabinoxylan water extractability was governed by the relative proportion of lowly and highly branched structures. A gradually increasing proportion of highly substituted populations was observed from cold to hot WE fractions. This was associated with the lower proportion of monosubstituted xylopyranosyl residues in the backbone, the higher proportion of disubstituted xylopyranosyl residues, and the higher level of substitution with feruloyl residues. Notable differences in the ratio of phenolic compounds to arabinose residues were observed between corresponding polymers isolated from rye flours of high and low baking quality, whereas the differences in their molecular weights were much less pronounced.

Xylo-oligosaccharides with degrees of polymerisation 5-13, formed by partial acid hydrolysis from an extract representative of olive pulp glucuronoxylans (GX), were analysed by electrospray ionisation mass spectrometry (ESI-MS), both in positive and negative modes. The positive spectrum showed the presence of xylo-oligosaccharides in the mass range between m/z 500 and 1500 corresponding to singly [M+Na](+) charged ions of neutral (Xyl(7-9)) and acidic xylo-oligosaccharides (Xyl(5-9)MeGlcA), and doubly [M+2Na](2+) charged ions of Xyl(9-13) and Xyl(7-11)MeGlcA. Ammonium adducts [M+NH(4)](+) were also observed for Xyl(5-9)MeGlcA. The negative spectra showed the contribution of ions in the mass range between m/z 600 and 1400, ascribed to the deprotonated molecules [M-H](-) of Xyl(3-9)MeGlcA. Tandem mass spectrometry (MS/MS) of the major ions observed in the MS spectra was performed. The MS/MS spectra of the [M+Na](+) adducts showed the loss of MeGlcA residues as the major fragmentation pathway and glycosidic fragment ions of Xyl(n) and Xyl(n)MeGlcA structures. The MS/MS spectra of the [M+NH(4)](+) adducts suggests the occurrence of isomers of Xyl(5-9)MeGlcA oligosaccharides with the MeGlcA residue at the reducing end and at the non-reducing end of the molecules, although other structural isomers can also occur. Both glycosidic bond and cross-ring cleavages in the MS/MS spectra of the [M-H](-) ion suggest the occurrence of Xyl(3-9)MeGlcA with the substituting group at the reducing end position of the xylose backbone, as the main fragmentation ions. The results obtained by ESI-MS/MS, both in positive and negative modes, of Xyl(7-13)- and Xyl(5-11)MeGlcA, allow to identify fragmentation patterns of the structural isomers with MeGlcA linked to the terminal xylosyl residues of the oligosaccharides. The occurrence of these higher molecular weight oligosaccharides with a low substitution pattern allows to infer a scatter and random distribution of MeGlcA along the xylan backbone of olive pulp.

Oat spelt xylan was treated with water in a batch reactor at temperaturesof 180 and 200 degrees C. Ion-moderated partition (IMP) chromatography was thenapplied to separate oligomers in solution according to their molecular size. Calibration of the IMP measurements based on peak height was found toquantify dissolved monomer and oligomer yields well. Oligomer concentrationsin the liquid hydrolysate were also determined from the difference inxylose monomer concentrations measured by high-performance liquid chromatographybefore and after posthydrolysis of dissolved xylooligosaccharidesto xylose. Delayed formation and then rapid disappearance of oligomersfrom DP10 to DP2 was observed by IMP, and total oligomer yields measuredby IMP and posthydrolysis were very similar at these times. However, whileIMP detected virtually no oligomers initially, posthydrolysis measurementsgave significant amounts of soluble oligomers at these times, indicating thatoligomers with chain lengths >10 were in solution but not detectable by theIMP system used.

It has been proposed that plant cell-wall polysaccharides are subject in vivo to non-enzymic scission mediated by hydroxyl radicals (-*OH). In the present study, xyloglucan was subjected in vitro to partial, non-enzymic scission by treatment with ascorbate plus H(2)O(2), which together generate -*OH. The partially degraded xyloglucan appeared to contain ester bonds within the backbone, as indicated by an irreversible decrease in viscosity upon alkaline hydrolysis. Aldehyde and/or ketone groups were also introduced into the polysaccharide by -*OH-attack, as indicated by staining with aniline hydrogen-phthalate and by reaction with NaB(3)H(4). The introduction of ester and oxo groups supports the proposed sequence of reactions: (a) -*OH-mediated H-abstraction to produce a carbon-centred carbohydrate radical; (b) reaction of the latter with O(2); and (c) elimination of a hydroperoxyl radical (HO(2)*-). When the partially degraded xyloglucan was reduced with NaB(3)H(4) followed by acid hydrolysis, several 3H-aldoses were detected ([3H]galactose, [3H]xylose, [3H]glucose, [3H]ribose and probably [3H]mannose), in addition to unidentified 3H-products (probably including anhydroaldoses). 3H-Alditols were undetectable, showing that few or no conventional reducing termini were introduced. Digestion of the NaB(3)H(4)-reduced, partially degraded xyloglucan with Driselase released 25 times more [3H]Xyl-alpha-(1-->6)-Glc than Xyl-alpha-(1-->6)-[3H]Glc, suggesting that the xylose side-chains of the xyloglucan had been more heavily attacked by -*OH than the glucose residues of the backbone. The radioactive xyloglucan was readily digested by cellulase, yielding 3H-products in the hepta- to nonasaccharide range. A fingerprinting strategy for identifying -*OH-attacked xyloglucan in plant cell walls is proposed.

Durum wheat (Triticum durum Desf.) semolina water-extractable arabinoxylan (TWEAX) (yield 0.28%, arabinose-to-xylose ratio (A/X) 0. 62) was fractionated by a stepwise increase in ethanol concentration (up to 65%). The A/X ratios of the resulting fractions varied between 0.42 and 0.80. With increasing ethanol concentrations, increasing A/X ratios went hand in hand with a relative increase of low molecular weight compounds, indicating that high molecular weight compounds with a low A/X ratio are preferentially precipitated from alcohol/water mixtures. (1)H NMR showed that, whereas in TWEAX the levels of unsubstituted xyloses (X(0)), monosubstituted (X(1)), and disubstituted (X(2)) xyloses were 63.1%, 11.8%, and 25.1%, respectively, fractions that precipitated with increasing ethanol concentrations had decreasing levels of X(0). Simultaneously, the level of X(1) decreased equally until it leveled of at ca. 10%. Concomitantly, the level of X(2) increased. The levels of X(0), X(1), and X(2) varied between 69.7% and 53.4%, 18.2% and 10.7%, and 12.2% and 35.9%, respectively.

Jojoba seed xyloglucan was shown to be a convenient source of biologically active xyloglucan oligosaccharides that contain both L- and D-galactosyl residues [E. Zablackis et al., Science, 272 (1996) 1808-1810]. Oligosaccharides were isolated by liquid chromatography of the mixture of oligosaccharides generated by treating jojoba seed xyloglucan with a beta-(1-->4)-endoglucanase. The purified oligosaccharides were reduced with NaBH4, converting them to oligoglycosyl alditol derivatives that were structurally characterized by a combination of mass spectrometry and 2-dimensional NMR spectroscopy. This analysis established that jojoba xyloglucan oligosaccharides contain the novel side-chain [alpha-L-Gal p-(1-->2)-beta-D-Galp-(1-->2)-alpha-D-Xyl p-(1-->6)-], which is structurally homologous to the fucose-containing side-chain [alpha-L-Fucp-(1-->2)-beta-D-Galp-(1-->2)-alpha-D-Xyl p-(1-->6)-] found in other biologically active xyloglucan oligosaccharides.

Several structural features, most notably the presence of alpha-L-Araf-(1-->2)-alpha-D-Xylp side chains, distinguish the arabinoxyloglucans (AXGs) produced by solanaceous plants from the xyloglucans produced by other dicotyledonous plants. However, previous studies did not establish the exact order of attachment of the various side chains along the backbone of these AXGs. Therefore, oligosaccharide subunits of the AXGs secreted by suspension-cultured tobacco and tomato cells were generated by treatment of the isolated AXGs with a fungal endo-beta-(1-->4)-D-glucanase (EG). The oligosaccharides were reduced with sodium borohydride to the corresponding oligoglycosyl alditol derivatives and purified by a combination of gel-permeation chromatography, reversed-phase HPLC, and HPAE chromatography. The isolated oligoglycosyl alditols were chemically characterized by NMR spectroscopy, matrix-assisted laser-desorption/ionization time-of-flight mass spectrometry (MALDITOFMS), fast-atom bombardment mass spectrometry (FABMS), FABMS/MS, and glycosyl-linkage analysis. The results confirmed that the AXGs from these species are composed of a (1-->4)-linked beta-D-Glcp backbone substituted at O-6 with various side chains. Both tobacco and tomato AXG contain alpha-D-Xylp and alpha-L-Araf-(1-->2)-alpha-D-Xylp side chains. However, oligosaccharide fragments of tomato AXG were also shown to contain beta-D-Galp-(1-->2)-alpha-D-Xylp and beta-Araf-(1-->3)-alpha-L-Araf-(1-->2)-alpha-D-Xylp side chains that are not present in the tobacco AXG. This is the first report of beta-Araf residues in a xyloglucan. The primary structures of 20 oligosaccharides generated by EG-treatment of tobacco AXG were determined. The generation of such a large number of oligosaccharides is due in part to the presence of O-acetyl substituents at O-6 of many of the backbone beta-D-Glcp residues of tobacco AXG. The presence of either an O-acetyl or a glycosidic substituent at O-6 of a beta-D-Glc p residue in the AXG backbone protects the glycosidic bond of this residue from cleavage by the EG. Removal of the O-acetyl substituents prior to EG-treatment of the AXG-results in oligosacharide fragments that are smaller than those produced by EG-treatment of the O-acetylated AXG. Therefore, analysis of the complex mixture of oligosaccharides obtained by EG treatment of native tobacco AXGs provides information regarding the distribution of AXG side chains that would be lost if the AXG is de-O-acetylated prior to EG-treatment. Furthermore, the large library of oligosaccharide fragments generated by this approach revealed additional correlations between the structural features of AXGs and diagnosis chemical shift effects in their 1H NMR spectra.

Arabinoxylans consisting of more than 90% of arabinose and xylose were extracted from water-insoluble cell wall material of dehusked barley grain, and degraded with a purified endo-(1-->4)-beta-D-xylanase from Aspergillus awamori. Twelve of the oligosaccharide fragments released were isolated by a combination of size-exclusion and anion-exchange chromatography, and their structures were determined by 1H NMR spectroscopy. The oligosaccharides identified consisted of (1-->4)-linked beta-D-xylopyranosyl residues, some of which were substituted at O-2, at O-3, or at both O-2 and O-3, with alpha-L-arabinofuranosyl groups. Two new structures, a tetrasaccharide and a pentasaccharide, both containing terminal 2-O-arabinofuranosylxylopyranose as a structural element are described.

Alkali-extractable wheat-flour arabinoxylan, treated with endo-(1-->4)-beta-D-xylanase III from Aspergillus awamori CMI 142717, was fractionated by Bio-Gel P-2 size exclusion chromatography at 60 degrees C. Column fractions, corresponding to oligosaccharides with degrees of polymerisation from 5 to 10, were collected, and subfractionated by high performance anion-exchange chromatography on CarboPac PA-1. The structures of the oligosaccharides thus obtained were elucidated by 1H NMR spectroscopy, showing chains of (1-->4)-linked beta-D-xylopyranosyl residues differently substituted at O-3 and/or O-2,3 with alpha-L-arabinofuranosyl groups. The structures were different from those obtained with endo-(1-->4)-beta-D-xylanase I of the same xylanolytic enzyme system.

A water-soluble arabinoxylan (D-xylose and L-arabinose in the molar ratio 1.0:3.4) was isolated from the mucilaginous bark of Litsea glutinosa (Lauraceae). The results of methylation analysis, partial hydrolysis, and 1H- and 13C-n.m.r. spectroscopy indicated a backbone of (1----4)-linked beta-D-xylopranosyl residues substituted at both positions 2 and 3 with side chains composed of either single or (1----3)-linked arabinofuranosyl residues. Both alpha-L- and beta-L-arabinofuranosyl residues were present. It is possible that side chains composed of two beta-L-arabinofuranosyl residues are attached mainly at O-2 of some xylosyl residues.

alpha-Linked D-xylosyl side chains represent the typical feature common to all xyloglucans not shared by other cell wall polysaccharides. Since no easily available alpha-D-xyloxidase is known, advantage was taken of the conformational and configurational homologies between alpha-D-xylopyranose and alpha-D-glucopyranose to make an alpha-D-glucosidase-gold complex which was able to recognize alpha-D-xylosyl terminal residues of xyloglucans. This marker was used together with alpha-L-fucosidase gold complex for the double labeling on two different structural features of the same macromolecule in plant primary cell wall.

Proteins from the culture filtrates of Thermoascus aurantiacus grown on paper were found to hydrolyze larchwood xylan completely to form xylose and 4-O-methyl-alpha-D-glucuronic acid. Partial hydrolysis of xylan by a xylanase purified from the culture filtrates resulted in the formation of neutral xylooligosaccharides of dp from 2 to 6 and acidic xylooligosaccharides of dp from 5 to 8. Each of these acidic sugars contained a single molecule of 4-O-methyl-alpha-D-glucuronic acid as a branch. Extensive hydrolysis of these oligosaccharides or xylan by xylanase led to the isolation of xylose, xylobiose, and an aldotetrauronic acid as terminal products. The structure of the aldotetrauronic acid was established by NMR as (2(2)-O-alpha-D,4-O-methyl-alpha-D-glucurono)-xylotriose. A beta-glucosidase, also purified from the culture filtrates, hydrolyzed xylan and the neutral or the acidic xylooligosaccharides from the nonreducing end to release only xylose. Neither xylanase nor beta-glucosidase hydrolyzed the beta-(1----4) linkage between the xylose carrying the branch and the adjacent xylose residue on each side.
