The secretion of renin from the juxtraglomerular (JG) cells is inhibited by calcium (Ca2+). An increase in intracellular Ca2+ concentration of JG cells inhibits renin secretion by inhibiting the primary signal for the secretory pathway. By contrast, inhibitors of Ca2+ release such as protein kinase and phosphatases block the inhibition of renin secretion. Thus, it is concluded that inhibitors specific for myosin light chain kinase blocks Ca2+-induced inhibition of renin secretion.

Calcium (Ca2+)-induced inhibition of renin secretion may be elicited by osmotic swelling of renin secretion granules and through the effects of Ca2+-calmodulin interaction on the myosin light chain kinase (MLCK). Herein, it is shown that inhibition of renin secretion by MLCK acts synergistically with osmotic swelling of renin secretory granules. The pathway is regulated by contractile actomyosin interaction.
