Despite the implementation of lifesaving newborn screening programs and a galactose-restricted diet, many patients with classic galactosemia develop long-term debilitating neurological deficits and primary ovarian insufficiency. Previously, we showed that the administration of human GALT mRNA predominantly expressed in the GalT gene-trapped mouse liver augmented the expression of hepatic GALT activity, which decreased not only galactose-1 phosphate (gal-1P) in the liver but also peripheral tissues. Since each peripheral tissue requires distinct methods to examine the biomarker and/or GALT effect, this highlights the necessity for alternative strategies to evaluate the overall impact of therapies. In this study, we established that whole-body galactose oxidation (WBGO) as a robust, noninvasive, and specific method to assess the in vivo pharmacokinetic and pharmacodynamic parameters of two experimental gene-based therapies that aimed to restore GALT activity in a mouse model of galactosemia. Although our results illustrated the long-lasting efficacy of AAVrh10-mediated GALT gene transfer, we found that GALT mRNA therapy that targets the liver predominantly is sufficient to sustain WBGO. The latter could have important implications in the design of novel targeted therapy to ensure optimal efficacy and safety.

Goal: This study aims to explore how healthcare professionals perceive home-based pediatric cancer care provided in French.Methodology: A qualitative descriptive study was conducted using semi-directed individual interviews of 22 healthcare professionals. A thematic analysis of the transcribed interviews was carried out independently by two members of the research team.Findings: Pediatric cancer care is readily available in French in Quebec, but access to French-language services in Ontario is limited. The possible causes and effects of this lack of access and potential solutions are discussed in this paper.Conclusion: The perceptions compiled in this study should be taken into account to help provide quality home-based pediatric cancer care in French.Keywords: healthcare delivery, oncology, pediatric, home-based care, French language

But : Cette étude vise à comprendre les perceptions des professionnels de la santé quant aux soins oncologiques pédiatriques offerts à domicile en français.Méthodologie : Une étude descriptive qualitative a été menée à l’aide d’entrevues individuelles semi-dirigées faites auprès de 22 professionnels de la santé. Une analyse thématique des transcriptions a été menée de façon indépendante par deux membres.Résultats : Les soins oncologiques pédiatriques sont offerts en français surtout au Québec, alors qu’il y a un manque d’accès en Ontario. Les causes et les effets possibles de ce manque d’accès ainsi que les solutions sont discutés dans le présent article.Conclusion : Les perceptions recueillies dans la présente étude devraient être prises en compte pour s’assurer d’offrir des soins oncologiques pédiatriques à domicile de qualité en français.Mots-clés : prestation des soins de santé, oncologie, pédiatrie, soins à domicile, francophonie

The assessment of the air quality is a major concern to the current time. The monitoring and maintenance of air quality necessarily pass by detecting and estimating the overall air pollution. The use of lichens must be an assessmenttool to be studied. In our work we were interested about the toxicity of lead on the various parameters of stress in the lichen Xanthoria parietina. For this purpose, lichen thalli have been incubated at lead concentrations of 0, 0.5, 1.0, 5.0 and 10.0 mM, for time scale of 0, 24, 48 and 96 hours. The obtained results showed that lead has an action on the various studied parameters, and the intensity of oxidative stress observed in lichens thalli depends on the concentration, and time of exposure. Lead induced a decrease in chlorophyll and protein contents, and an increase in the contents of catalase, hydrogen peroxide and reduced glutathione. Furthermore, the results also showed that high concentrations of lead caused total destruction of reduced glutathione.

Vibrational circular dichroism (VCD) spectroscopy can be useful for determining the absolute configuration of chiral molecules, as long as the signal intensities are high enough. Here, the authors establish the absolute configurations of two large chiral porphyrin cages and, notably, discover that host-guest binding enhances their VCD intensities.

Objective(s): Bill duct ligation (BDL) is a representative model of biliary cholestasis in animals. Curcumin has a protective effect on the liver; however, its underlying mechanisms are not completely known. This study explored the hepatoprotective activity of curcumin on hepatic damage via measuring the expression of sirtuin3 (SIRT3), AMP-activated protein kinase (AMPK), carnitine palmitoyltransferase 1A (CPT-1A), isocitrate dehydrogenase2 (IDH2) and manganese superoxide dismutase (MnSOD) as well as the level of serum lipid profile in the BDL fibrotic rat model. Materials and Methods: The study consisted of four groups (n=8 for each of Wistar rats): sham group, sham+curcumin (sham+Cur) group (received curcumin 100 mg/kg/day), BDL+Cur group, and BDL group. Transcription levels of SIRT3, AMPK, CPT-1A, IDH2, MnSOD and protein expression level of SIRT3 were measured by real-time PCR and Western blotting, respectively. Results: It was identified that SIRT3, AMPK, CPT-1A, IDH2 and MnSOD expression significantly decreased in BDL rats compared to sham rats; however, in the curcumin treatment of BDL rats, the expression of these factors increased significantly compared to BDL (P-value

Oleander (Nerium oleander) is an ornamental plant common in tropical and sub-tropical regions that is becoming increasingly widespread, even in temperate regions. Oleander poisoning may occur in animals and humans. The main active components contained in the plant are cardiac glycosides belonging to the class of cardenolides that are toxic to many species, from human to insects. This work describes a case of oleander poisoning that occurred on a small cattle farm and resulted in the fatality of all six resident animals. Furthermore, the investigation of the poisonous agent is described, with particular focus on the characterization of the oleandrin toxin that was recovered from the forage and rumen contents. The innovation of this study is the first description of the detection and quantification of the oleandrin toxin by liquid chromatography-high resolution mass spectrometry (LC-HRMS) in rumen.
