Abstract During the Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) expedition, the Balloon-bornE moduLar Utility for profilinG the lower Atmosphere (BELUGA) was deployed from an ice floe drifting in the Fram Strait from 29 June to 27 July 2020. The BELUGA observations aimed to characterize the cloudy Arctic atmospheric boundary layer above the sea ice using a modular setup of five instrument packages. The in situ measurements included atmospheric thermodynamic and dynamic state parameters (air temperature, humidity, pressure, and three-dimensional wind), broadband solar and terrestrial irradiance, aerosol particle microphysical properties, and cloud particle images. In total, 66 profile observations were collected during 33 balloon flights from the surface to maximum altitudes of 0.3 to 1.5 km. The profiles feature a high vertical resolution of 0.01 m to 1 m, including measurements below, inside, and above frequently occurring low-level clouds. This publication describes the balloon operations, instruments, and the obtained data set. We invite the scientific community for joint analysis and model application of the freely available data on PANGAEA.

Lewis Fry Richardson proposed his famous picture of turbulent flows in 1922, where the kinetic energy is transferred from large-scale to small-scale structures until the viscosity converts it into heat. This cascade idea, also known as the forward energy cascade, is now widely accepted and is treated as the cornerstone of not only turbulent modeling, but also global circulation models of the ocean and atmosphere. In this work, the Filter-Space-Technique is applied to the oceanic flow field provided by the CMEMS reanalysis model to quantify the scale-to-scale energy flux. A rich dynamical pattern associated with different scales is observed. More precisely, either positive or negative fluxes are observed, indicating the direction of the energy cascade, where the energy is transferred from large-scale structures to small-scale ones or vice versa. High-intensity energy exchange is found mainly in the Western Boundary Current Systems and Equatorial Counter Currents. For the latter case, a wavelike pattern is observed on the westward travel. Moreover, strong seasonal variation is evident for some scales and regions. These results confirm the existence of forward and inverse cascades and rich regional dynamics.

In a fully developed sea, the significant wave height (Hs) and wind speed (U10) are conventionally related to a pure quadratic equation. This relation is often violated, since in the field the measured local Hs is often contaminated by the swell, which is propagated from distant places. Therefore, a swell partition is required before the establishment of the wind-wave relation. The Spectra Energy Partition (SEP) is regarded as the best way to isolate the swell and the wind wave relation: it is based on the identification of a separation frequency in the ocean wave spectrum. However, for most field observations, the wave spectra information is unavailable, and thus the SEP is inapplicable. This work proposes a probability-based algorithm to identify the averaged swell without knowing the wave spectrum a priori. The local wind-wave relation is established by either a linear or an energy-conserved decomposition. We also find that the local wind-wave relation is a power-law when the wind speed U10 is above 4 m/s. The proposed method is first validated by applying the SEP method to buoy collected wave spectra data. The global pattern of the swell and the local wind waves is retrieved by applying the proposed method to a 17-year wind and wave database from the JASON satellite. Strong seasonal and spatial variations are obtained. Finally, a prediction model based on the empirical wind-wave relation is shown to successfully retrieve the wave field when the wind field is available.

Primary immune regulatory disorders (PIRD) represent a group of disorders characterized by immune dysregulation, presenting with a wide range of clinical disease, including autoimmunity, autoinflammation, or lymphoproliferation. Autosomal dominant germline gain-of-function (GOF) variants in STAT3 result in a PIRD with a broad clinical spectrum. Studies in patients have documented a decreased frequency of FOXP3+ Tregs and an increased frequency of Th17 cells in some patients with active disease. However, the mechanisms of disease pathogenesis in STAT3 GOF syndrome remain largely unknown, and treatment is challenging. We developed a knock-in mouse model harboring a de novo pathogenic human STAT3 variant (p.G421R) and found these mice developed T cell dysregulation, lymphoproliferation, and CD4+ Th1 cell skewing. Surprisingly, Treg numbers, phenotype, and function remained largely intact; however, mice had a selective deficiency in the generation of iTregs. In parallel, we performed single-cell RNA-Seq on T cells from STAT3 GOF patients. We demonstrate only minor changes in the Treg transcriptional signature and an expanded, effector CD8+ T cell population. Together, these findings suggest that Tregs are not the primary driver of disease and highlight the importance of preclinical models in the study of disease mechanisms in rare PIRD.

In fully developed turbulence, there is a flux of energy from large to small scales in the inertial range until the dissipation at small scales. It is associated with irreversibility, i.e., a breaking of the time reversal symmetry. Such turbulent flows are characterized by scaling properties, and we consider here how irreversibility depends on the scale. Indicators of time-reversal symmetry for time series are tested involving triple correlations in a non-symmetric way. These indicators are built so that they are zero for a time-reversal symmetric time series, and a departure from zero is an indicator of irreversibility. We study these indicators applied to two fully developed turbulence time series, from flume tank and wind tunnel databases. It is found that irreversibility occurs in the inertial range and has scaling properties with slopes close to one. A maximum value is found around the injection scale. This confirms that the irreversibility is associated with the turbulent cascade in the inertial range and shows that the irreversibility is maximal at the injection scale, the largest scale of the turbulent cascade.

Because climate change will likely have major impacts in low-lying coastal areas, our ability for evaluating predictable consequences of coastal hazards in the future already represents a key issue in coastal risk reduction. This paper presents examples of the application of a method aimed at assessing the future position of the shoreline and at mapping areas that will be at risk of flooding by 2065 with future sea-level rise at two coastal localities of northern France where properties are already threatened by erosion or flooding. Our results show that erosion is the main coastal hazard at one of the two study sites where a large part of a housing estate may disappear by 2065 if no action is taken for limiting coastal erosion. Although marine flooding represents the primary risk at the second study site, coastal erosion will increase the risk of flooding of properties in the future due to the decrease in width of the protective coastal dunes. Comparison of our results with the coastal hazard maps published by the French government shows significant differences that can be explained by the fact that the risk of erosion was not considered in the Coastal Risk Protection Plans (PPRL) of the two municipalities considered in this study, which raises the question of the reliability of areas at risk defined without taking into account future shoreline evolution trends.

Andean glaciers have melted rapidly since the 1960s. While some melting is likely due to anthropogenic climate change driven by increasing greenhouse gases, deposition of light-absorbing particles such as black carbon (BC) may also play a role. We hypothesize that BC from fires in the Amazon Basin and elsewhere may be deposited on Andean glaciers, reducing the surface albedo and inducing further melting. Here we investigate the role of BC deposition on albedo changes in the Andes for 2014–2019 by combining atmospheric chemistry modeling with observations of BC in snow or ice at four mountain sites in Peru (Quelccaya, Huascarán, Yanapaccha, and Shallap) and at one site in Bolivia (Illimani). We find that annual mean ice BC concentrations simulated by the chemical transport model GEOS-Chem for 2014–2019 are roughly consistent with those observed at the site with the longest record, Huascarán, with overestimates of 15%–40%. Smoke from fires account for 20%–70% of total wet and dry deposition fluxes, depending on the site. The rest of BC deposited comes from fossil fuel combustion. Using a snow albedo model, we find that the annual mean radiative forcing from the deposition of smoke BC alone on snow ranges from +0.1 to +3.2 W m ^−2 under clear-sky conditions, with corresponding average albedo reductions of 0.04%–1.1%. These ranges are dependent on site and snow grain size. This result implies a potentially significant climate impact of biomass burning in the Amazon on radiative forcing in the Andes.

Abstract Background The γ-hexalactone is a flavoring agent for alcoholic beverages, teas, breads, dairy products, coffees, buttery products among others. It presents low molecular weight and exhibits sweet fruity aroma with nuances of nuts. As far as we know, both literature and government regulations have gaps regarding the safe use of the γ-hexalactone. In this context, the main objective of this work was to evaluate the effects of γ-hexalactone through in silico and in vitro approaches. Methods The in silico analysis was performed through four free online platforms (admetSAR, Osiris Property Explorer®, pkCSM platform and PreADMET) and consisted of comparative structural analysis with substances present in databases. The computational prediction was performed in the sense of complement and guide the in vitro tests. Regarding in vitro investigations, screening of cytotoxicity (assessed by cell proliferation and viability parameters) in lymphocytes exposed to γ-hexalactone for 72 h were carried out previously to determine non-cytotoxic concentrations. Following this screening, concentrations of 5.15, 0.515, and 0.0515 μM were selected for the study of the respective potentials: genotoxic (assessed by DNA comet assay), chromosomal mutation (analysis of micronucleus frequency) and immunomodulatory (cytokine quantification using ELISA immunoassay). The results of in vitro assays were compared by one-way analysis of variance (ANOVA), followed by Bonferroni’s post hoc test, conducted by statistic software. Results The platform PreADMET pointed out that γ-hexalactone is potentially mutagenic and carcinogenic. The comet assay data corroborate with these results demonstrating that γ-hexalactone at 5.15 μM caused lymphocytes DNA damage. In relation to cytokine secretion, the results indicate that lymphocytes were activated by γ-hexalactone at non-cytotoxic concentrations, involving an increase in the IL-1 levels in all tested concentrations, ranging from approximately 56 to 93%. The γ-hexalactone only at 5.15 μM induced increase in the levels of IL-6 (~ 60%), TNF-α (~ 68%) and IFN-γ (~ 29%), but decreased IL-10 (~ 46%) in comparison with the negative control (p

A new instrument for the quantification of light absorption by particles collected on filters has been developed to address long standing environmental questions about light-absorbing particles in air, water, and on snow and ice. The Light Absorption Heating Method (LAHM) uses temperature changes when filters are exposed to light to quantify absorption. Through the use of calibration standards, the observed temperature response of unknown materials can be related to the absorption cross section of the substance collected on the filter. Here, we present a detailed description of the instrument and calibration. The results of the calibration tests using a common surrogate for black carbon, Fullerene soot, show that the instrument provides stable results even when exposed to adverse laboratory conditions, and that there is little drift in the instrument over longer periods of time. Calibration studies using Fullerene soot suspended in water, airborne propane soot, as well as atmospheric particulates show consistent results for absorption cross section when using accepted values for the mass absorption cross section of the soot and when compared to results from a 3-wavelength photoacoustic instrument. While filter sampling cannot provide the time resolution of other instrumentation, the LAHM instrument fills a niche where time averaging is reasonable and high-cost instrumentation is not available. The optimal range of absorption cross sections for LAHM is from 0.1 to 5.0 cm2 (~1.0–50.0 µg soot) for 25 mm filters and 0.4 to 20 cm2 (4.0–200.0 µg soot) for 47 mm filters, with reduced sensitivity to higher values.

ABSTRACT COVID-19 started in China on December 31, 2019 and has since been the subject of several studies in medical field. There is not much evidence about the pregnancy and puerperium with the susceptibility of coronavirus infection (COVID-19). The present work reports the laboratory and radiological aspects of a puerperium patient. A positive test for severe acute respiratory syndrome-coronavirus 2 (SARS-CoV-2) generally confirms the diagnosis of COVID-19, although false-positive and false-negative tests are possible. The test considered the gold standard for the diagnosis of coronavirus infection (COVID-19) is from in vivo or post-mortem polymerase chain reaction (PCR) sample. The purpose of the present case report is to demonstrate the case of a puerperium patient with an unfavorable clinical evolution, giving special attention to laboratory tests that were practically unchanged.

Reduced function of the serotonin transporter (SERT) is associated with increased susceptibility to anxiety and depression and with type-2 diabetes, which is especially true in older women. Preference for a “Western diet” (WD), enriched with saturated fat, cholesterol, and sugars, may aggravate these conditions. In previous studies, decreased glucose tolerance, central and peripheral inflammation, dyslipidemia, emotional, cognitive, and social abnormalities were reported in WD-fed young female mice. We investigated the metabolic, molecular, and behavioral changes associated with a 3-week-long dietary regime of either the WD or control diet in 12-month-old female mice with three different Sert genotypes: homozygous (Slc6a4) gene knockout (Sert−/−: KO), heterozygous (Sert+/−: HET), or wild-type mice (Sert+/+: WT). In the WT-WD and KO-WD groups, but not in HET-WD-fed mice, most of changes induced by the WD paralleled those found in the younger mice, including brain overexpression of inflammatory marker Toll-like receptor 4 (Tlr4) and impaired hippocampus-dependent performance in the marble test. However, the 12-month-old female mice became obese. Control diet KO mice exhibited impaired hippocampal-dependent behaviors, increased brain expression of the serotonin receptors Htr2c and Htr1b, as well as increased Tlr4 and mitochondrial regulator, peroxisome proliferator-activated receptor gamma-coactivator-1a (Ppargc1a). Paradoxically, these, and other changes, were reversed in KO-WD mutants, suggesting a complex interplay between Sert deficiency and metabolic factors as well as potential compensatory molecular mechanisms that might be disrupted by the WD exposure. Most, but not all, of the changes in gene expression in the brain and liver of KO mice were not exhibited by the HET mice fed with either diet. Some of the WD-induced changes were similar in the KO-WD and HET-WD-fed mice, but the latter displayed a “rescued” phenotype in terms of diet-induced abnormalities in glucose tolerance, neuroinflammation, and hippocampus-dependent performance. Thus, complete versus partial Sert inactivation in aged mice results in distinct metabolic, molecular, and behavioral consequences in response to the WD. Our findings show that Sert+/− mice are resilient to certain environmental challenges and support the concept of heterosis as evolutionary adaptive mechanism.

Barred macrotidal beaches are affected by continuous horizontal displacements of different hydrodynamic zones associated with wave transformation (shoaling, breaker and surf zones) due to significant tide-induced water level changes. A series of wave and current meters, complemented by a video imagery system, were deployed on a barred beach of northern France during a 6-day experiment in order to characterize the spatial and temporal variability of wave-induced processes across the beach. Wave and current spectral analyses and analyses of cross-shore current direction and asymmetry resulted in the identification of distinct hydrodynamic processes, including the development of infragravity waves and offshore-directed flows in the breaker and surf zones. Our results revealed a high spatial variability in the hydrodynamic processes across the beach, related to the bar-trough topography, as well as significant variations in the directions and intensity of cross-shore currents at fixed locations due to the horizontal translation of the different hydrodynamic zones resulting from continuous changes in water level due to tides.

Abstract Quark nuggets are theoretical objects composed of approximately equal numbers of up, down, and strange quarks and are also called strangelets and nuclearites. They have been proposed as a candidate for dark matter, which constitutes ~85% of the universe’s mass and which has been a mystery for decades. Previous efforts to detect quark nuggets assumed that the nuclear-density core interacts directly with the surrounding matter so the stopping power is minimal. Tatsumi found that quark nuggets could well exist as a ferromagnetic liquid with a ~1012-T magnetic field. We find that the magnetic field produces a magnetopause with surrounding plasma, as the earth’s magnetic field produces a magnetopause with the solar wind, and substantially increases their energy deposition rate in matter. We use the magnetopause model to compute the energy deposition as a function of quark-nugget mass and to analyze testing the quark-nugget hypothesis for dark matter by observations in air, water, and land. We conclude the water option is most promising.

Converging evidence suggests a role of serotonin (5-hydroxytryptamine, 5-HT) and tryptophan hydroxylase 2 (TPH2), the rate-limiting enzyme of 5-HT synthesis in the brain, in modulating long-term, neurobiological effects of early-life adversity. Here, we aimed at further elucidating the molecular mechanisms underlying this interaction, and its consequences for socio-emotional behaviors, with a focus on anxiety and social interaction. In this study, adult, male Tph2 null mutant (Tph2-/-) and heterozygous (Tph2+/-) mice, and their wildtype littermates (Tph2+/+) were exposed to neonatal, maternal separation (MS) and screened for behavioral changes, followed by genome-wide RNA expression and DNA methylation profiling. In Tph2-/- mice, brain 5-HT deficiency profoundly affected socio-emotional behaviors, i.e., decreased avoidance of the aversive open arms in the elevated plus-maze (EPM) as well as decreased prosocial and increased rule breaking behavior in the resident-intruder test when compared to their wildtype littermates. Tph2+/- mice showed an ambiguous profile with context-dependent, behavioral responses. In the EPM they showed similar avoidance of the open arm but decreased prosocial and increased rule breaking behavior in the resident-intruder test when compared to their wildtype littermates. Notably, MS effects on behavior were subtle and depended on the Tph2 genotype, in particular increasing the observed avoidance of EPM open arms in wildtype and Tph2+/- mice when compared to their Tph2-/- littermates. On the genomic level, the interaction of Tph2 genotype with MS differentially affected the expression of numerous genes, of which a subset showed an overlap with DNA methylation profiles at corresponding loci. Remarkably, changes in methylation nearby and expression of the gene encoding cholecystokinin, which were inversely correlated to each other, were associated with variations in anxiety-related phenotypes. In conclusion, next to various behavioral alterations, we identified gene expression and DNA methylation profiles to be associated with TPH2 inactivation and its interaction with MS, suggesting a gene-by-environment interaction-dependent, modulatory function of brain 5-HT availability.

Brain serotonin (5-hydroxytryptamine, 5-HT) system dysfunction is implicated in exaggerated fear responses triggering various anxiety-, stress-, and trauma-related disorders. However, the underlying mechanisms are not well understood. Here, we investigated the impact of constitutively inactivated 5-HT synthesis on context-dependent fear learning and extinction using tryptophan hydroxylase 2 (Tph2) knockout mice. Fear conditioning and context-dependent fear memory extinction paradigms were combined with c-Fos imaging and electrophysiological recordings in the dorsal hippocampus (dHip). Tph2 mutant mice, completely devoid of 5-HT synthesis in brain, displayed accelerated fear memory formation and increased locomotor responses to foot shock. Furthermore, recall of context-dependent fear memory was increased. The behavioral responses were associated with increased c-Fos expression in the dHip and resistance to foot shock-induced impairment of hippocampal long-term potentiation (LTP). In conclusion, increased context-dependent fear memory resulting from brain 5-HT deficiency involves dysfunction of the hippocampal circuitry controlling contextual representation of fear-related behavioral responses.

Magnetized quark nuggets (MQNs) are a recently proposed dark-matter candidate consistent with the Standard Model and with Tatsumi’s theory of quark-nugget cores in magnetars. Previous publications have covered their formation in the early universe, aggregation into a broad mass distribution before they can decay by the weak force, interaction with normal matter through their magnetopause, and a first observation consistent MQNs: a nearly tangential impact limiting their surface-magnetic-field parameter Bo from Tatsumi’s ~1012+/−1 T to 1.65 × 1012 T +/− 21%. The MQN mass distribution and interaction cross section strongly depend on Bo. Their magnetopause is much larger than their geometric dimensions and can cause sufficient energy deposition to form non-meteorite craters, which are reported approximately annually. We report computer simulations of the MQN energy deposition in water-saturated peat, soft sediments, and granite, and report the results from excavating such a crater. Five points of agreement between observations and hydrodynamic simulations of an MQN impact support this second observation being consistent with MQN dark matter and suggest a method for qualifying additional MQN events. The results also redundantly constrain Bo to ≥ 4 × 1011 T.

The presence of light-absorbing particles, including black carbon in glaciers leads to a reduction in albedo (light reflection), leading to further melting of snow ice, increased amount of shortwave solar radiation and leads to the glacier. The objective has been to determine the variation in the temporal space of black carbon, the amount of light-absorbing particles and the decrease of albedo on the surface of the Ampay glacier. 10 snow samples were selected at various locations on the glacier during 2017. The light absorption heating method has been applied to measure light-absorbing particles, a technique that measures the temperature increase of the particle charge in a filter applying visible light that estimates the amount of light energy. The results show in terms of effective black carbon, the highest of 65,224 nanograms of black carbon per gram of water in the month of October and the minimum value of 20,941 nanograms of black carbon per gram of water in the month of February, typically associated with the rain. The energy absorbed by the light-absorbing particles in the Ampay glacier, the highest corresponds to the month of November with 8,952.92 J s/m2 and lowest in February with 2,747.26 J s/m2. In April, the amount of snow melted due to light-absorbing particles has been approximately 13.57 kg/m2. Melting has increased considerably in other months with the largest melting, with a value of 26.65 kg/m2, almost 7.0 kW/m2 of snow turned into water in the month of November. It is concluded that the technique of the Light absorption heating method is adequate because it is optimal for the achievement of the research objectives, it is economical, effective and has allowed quantifying light-absorbing particles in snow.

This study reports on the origin of small-scale ice crystal complexity and its influence on the angular light scattering properties of cirrus clouds. Cloud simulation experiments were conducted at the AIDA (Aerosol Interactions and Dynamics in the Atmosphere) cloud chamber of the Karlsruhe Institute of Technology (KIT). A new experimental procedure was applied to grow and sublimate ice particles at defined super- and subsaturated ice conditions and for temperatures in the −40 to −60 °C range. The experiments were performed for ice clouds generated via homogeneous and heterogeneous initial nucleation. Small-scale ice crystal complexity was deduced from measurements of spatially resolved single particle light scattering patterns by the latest version of the Small Ice Detector (SID-3). It was found that a high crystal complexity dominates the microphysics of the simulated clouds and the degree of this complexity is dependent on the available water vapor during the crystal growth. Indications were found that the small-scale crystal complexity is influenced by unfrozen H2SO4 / H2O residuals in the case of homogeneous initial ice nucleation. Angular light scattering functions of the simulated ice clouds were measured by the two currently available airborne polar nephelometers: the polar nephelometer (PN) probe of Laboratoire de Métérologie et Physique (LaMP) and the Particle Habit Imaging and Polar Scattering (PHIPS-HALO) probe of KIT. The measured scattering functions are featureless and flat in the side and backward scattering directions. It was found that these functions have a rather low sensitivity to the small-scale crystal complexity for ice clouds that were grown under typical atmospheric conditions. These results have implications for the microphysical properties of cirrus clouds and for the radiative transfer through these clouds.

Sea breeze (SB) phenomena may strongly influence air quality and lead to important effects on human health. In order to study the impact of SB dynamics on the properties and toxicity of aerosols, an atmospheric mobile unit was deployed during a field campaign performed in an urbanized and industrialized coastal area in Northern France. This unit combines aerosol samplers, two scanning lidars (Doppler and elastic) and an air-liquid interface (ALI, Vitrocell®) in vitro cell exposure device. Our study highlights that after the passage of an SB front, the top of the atmospheric boundary layer collapses as the thermal internal boundary layer (TIBL) develops, which leads to high aerosol extinction coefficient values (>0.4 km−1) and an increase of PM2.5 and NOx concentrations in the SB current. The number-size distribution of particles indicates a high proportion of fine particles (with diameter below 500 nm), while the volume-size distribution shows a major mode of coarse particles centered on 2−3 µm. Individual particle analyses performed by cryo-transmission scanning electron microscopy (cryo-TSEM)-EDX highlights that submicronic particles contained a high fraction of secondary compounds, which may result from nucleation and/or condensation of condensable species (vapors or gaseous species after photo-oxidation). Secondary aerosol (SA) formation can be enhanced in some areas, by the interaction between the SB flow and the upper continental air mass, particularly due to the effect of both turbulence and temperature/humidity gradients between these two contrasting air masses. Potential areas of SA formation are located near the ground, during the SB front passage and in the vicinity of the SB current top. During the sea breeze event, an increase in the oxidative stress and inflammation processes in exposed lung cells, compared to the unexposed cells, can also be seen. In some instances, short singularity periods are observed during SB, corresponding to a double flow structure. It consists of two adjacent SB currents that induce an important increase of the TIBL top, improving the pollutants dispersion. This is associated with a substantial decrease of aerosol mass concentrations.

Rats intracerebroventricularily (icv) treated with streptozotocin (STZ), shown to generate an insulin resistant brain state, were used as an animal model for the sporadic form of Alzheimer’s disease (sAD). Previously, we showed in an in vivo study that 3 months after STZ icv treatment hippocampal adult neurogenesis (AN) is impaired. In the present study, we examined the effects of STZ on isolated adult hippocampal neural stem cells (NSCs) using an in vitro approach. We revealed that 2.5 mM STZ inhibits the proliferation of NSCs as indicated by reduced number and size of neurospheres as well as by less BrdU-immunoreactive NSCs. Double immunofluorescence stainings of NSCs already being triggered to start with their differentiation showed that STZ primarily impairs the generation of new neurons, but not of astrocytes. For revealing mechanisms possibly involved in mediating STZ effects we analyzed expression levels of insulin/glucose system-related molecules such as the glucose transporter (GLUT) 1 and 3, the insulin receptor (IR) and the insulin-like growth factor (IGF) 1 receptor. Applying quantitative Real time-PCR (qRT-PCR) and immunofluorescence stainings we showed that STZ exerts its strongest effects on GLUT3 expression, as GLUT3 mRNA levels were found to be reduced in NSCs, and less GLUT3-immunoreactive NSCs as well as differentiating cells were detected after STZ treatment. These findings suggest that cultured NSCs are a good model for developing new strategies to treat nerve cell loss in AD and other degenerative disorders.

Marine coastal processes are highly variable over different space scales and timescales. In this paper we analyse the intermittency properties of particle size distribution (PSD) recorded every second using a LISST instrument (Laser In-Situ Scattering and Transmissometry). The particle concentrations have been recorded over 32 size classes from 2.5 to 500 μm, at 1 Hz resolution. Such information is used to estimate at each time step the hyperbolic slope of the particle size distribution, and to consider its dynamics. Shannon entropy, as an indicator of the randomness, is estimated at each time step and its dynamics is analysed. Furthermore, particles are separated into four classes according to their size, and the intermittent properties of these classes are considered. The empirical mode decomposition (EMD) is used, associated with arbitrary-order Hilbert spectral analysis (AHSA), in order to retrieve scaling multifractal moment functions, for scales from 10 s to 8 min. The intermittent properties of two other indicators of particle concentration are also considered in the same range of scales: the total volume concentration Cvol-total and the particulate beam attenuation coefficient cp(670). Both show quite similar intermittent dynamics and are characterised by the same exponents. Globally we find here negative Hurst exponents (meaning the small scales show larger fluctuation than large scales) for each time series considered, and nonlinear moment functions.

The Taylor power law (or temporal fluctuation scaling), is a scaling relationship of the form σ ~ (P)λ where !! is the standard deviation and hPi the mean value of a sample of a time series has been observed for power output data sampled at 5 min and 1 s and from five wind farms and a single wind turbine, located at different places. Furthermore, an analogy with the turbulence field is performed, consequently allowing the establishment of a scaling relationship between the turbulent production IP and the mean value (P).

Glaciers in the tropical Andes have been rapidly losing mass since the 1970s. In addition to the documented increase in temperature, increases in light-absorbing particles deposited on glaciers could be contributing to the observed glacier loss. Here we report on measurements of light-absorbing particles sampled from glaciers during three surveys in the Cordillera Blanca Mountains in Peru. During three research expeditions in the dry seasons (May–August) of 2011, 2012 and 2013, 240 snow samples were collected from 15 mountain peaks over altitudes ranging from 4800 to nearly 6800 m. Several mountains were sampled each of the 3 years and some mountains were sampled multiple times during the same year. Collected snow samples were melted and filtered in the field then later analyzed using the Light Absorption Heating Method (LAHM), a new technique that measures the ability of particles on filters to absorb visible light. LAHM results have been calibrated using filters with known amounts of fullerene soot, a common industrial surrogate for black carbon (BC). As sample filters often contain dust in addition to BC, results are presented in terms of effective black carbon (eBC). During the 2013 survey, snow samples were collected and kept frozen for analysis with a Single Particle Soot Photometer (SP2). Calculated eBC mass from the LAHM analysis and the SP2 refractory black carbon (rBC) results were well correlated (r2 = 0.92). These results indicate that a substantial portion of the light-absorbing particles in the more polluted regions were likely BC. The 3 years of data show that glaciers in the Cordillera Blanca Mountains close to human population centers have substantially higher levels of eBC (as high as 70 ng g−1) than remote glaciers (as low as 2.0 ng g−1 eBC), indicating that population centers can influence local glaciers by sourcing BC.

We consider here wind speed time series and the aggregate output wind power from a wind farm. We study their scaling statistics in the framework of fully developed turbulence and Kolmogorov's theory. We estimate their Fourier power spectra and consider their scaling properties in the physical space. We show that the atmospheric wind speed and the aggregate power output from a wind farm are intermittent and multifractal over a wide range of scales. The coupling between simultaneous data of the wind speed and aggregate power output is investigated through a joint multifractal description using the generalized correlation functions (GCFs). This multiscaling test is compatible with a linear relation between the wind speed and the aggregate power output fluctuations for timescales T ⩾ 103 s ≃ 15 min.

Wind and wave effects on tidal current structure and turbulence throughout the water column are examined using an upward-looking acoustic Doppler current profiler (ADCP). The instrument has been deployed on the seafloor of 18-m mean depth, off the north-eastern French coast in the eastern English Channel, over 12 tidal cycles, and covered the period of the transition from mean spring to neap tide, and forcing regimes varied from calm to moderate storm conditions. During storms, we observed gusty winds with magnitudes reaching 15 m s−1 and wave heights reaching up to 1.3 m. Analysis of velocity spectra revealed a noticeable contribution of wind-induced waves to spectral structure of velocity fluctuations within the subsurface layer. Near the surface, stormy winds and waves produced a significant intensification of velocity fluctuations, particularly when the sustained wind blew against the ebb tide flow. As during wavy periods, the variance-derived Reynolds stress estimates might include a wave-induced contamination, we applied the Variance Fit method to obtain unbiased stresses and other turbulent quantities. Over calm periods, the turbulent quantities usually decreased with height above the seabed. The stresses were found to vary regularly with the predominantly semidiurnal tidal flow. The along-shore stress being generally greater during the flood flow (~2.7 Pa) than during the ebb flow (~−0.6 Pa). The turbulent kinetic energy production rate, P, and eddy viscosity, Az, followed a nearly regular cycle with close to a quarter-diurnal period. As for the stresses, near the seabed, we found the maximum values of estimated quantities of P and Az to be 0.1 Wm−3 and 0.5 m2 s−1, respectively, during the flood flow. Over the storm periods, we found the highest unbiased stress values (~−2.6 Pa) during ebb when tidal currents were opposite to the southwesterly winds while, during the flood, the surface stresses slightly exceeded those estimated for a calm period. A comparison of obtained results gives a good agreement with those of other researchers working on direct measurements of turbulence in tidal flows.

We consider here the dynamics of two polychaete populations based on a 20 yr temporal benthic survey of two muddy fine sand communities in the Bay of Morlaix, Western English Channel. These populations display high temporal variability, which is analyzed here using scaling approaches. We find that population densities have heavy tailed probability density functions. We analyze the dynamics of relative species abundance in two different communities of polychaetes by estimating in a novel way a "mean square drift" coefficient which characterizes their fluctuations in relative abundance over time. We show the usefulness of using new tools to approach and model such highly variable population dynamics in marine ecosystems.

We consider here pH and temperature fluctuations in marine waters, recorded at fixed points using high resolution automatic devices. We analyze time series coming from 4 monitoring stations located along French coast: one station is situated in the coastal area off Boulogne-sur-mer (Eastern English Channel) and 3 stations in the Bay of Seine. All these pH time series reveal large fluctuations at all scales similar to turbulent temperature fluctuations. We compare the pH and temperature time series through Fourier spectral analysis methods: spectra, compensated spectra, cospectra. We find good scaling properties of pH fluctuations, with power spectral slopes close to 1.5 for marine stations and 1.2 for the estuarine station. These analyses show that pH fluctuations in marine waters are strongly influenced by turbulent hydrodynamical transport, and may be considered as a turbulent active scalar.

Satellite remote sensing observations allow the ocean surface to be sampled synoptically over large spatio-temporal scales. The images provided from visible and thermal infrared satellite observations are widely used in physical, biological, and ecological oceanography. The present work proposes a method to understand the multi-scaling properties of satellite products such as the Chlorophyll-a (Chl-a), and the Sea Surface Temperature (SST), rarely studied. The specific objectives of this study are to show how the small scale heterogeneities of satellite images can be characterised using tools borrowed from the fields of turbulence. For that purpose, we show how the structure function, which is classically used in the frame of scaling time series analysis, can be used also in 2D. The main advantage of this method is that it can be applied to process images which have missing data. Based on both simulated and real images, we demonstrate that coarse-graining (CG) of a gradient modulus transform of the original image does not provide correct scaling exponents. We show, using a fractional Brownian simulation in 2D, that the structure function (SF) can be used with randomly sampled couple of points, and verify that 1 million of couple of points provides enough statistics.

Extensive diagnostic and scientific investigations are often restricted by limited availability of material. Therefore, methods like multiplex PCR strategies are needed to conserve as much sample as possible. Unfortunately, the establishment of such procedures poses several difficulties. Here we describe the advantages of a new enzyme, AmpliTaq Gold™ DNA Polymerase, in multiplex and time-release PCR. The application of this thermostable recombinant Taq DNA polymerase allows the specific amplification of DNA/cDNA targets with very high sensitivity. With our protocol, the specific amplification of 13 different cDNAs of cytokines and cytokine receptors can be realized in three multiplex PCRs (IL-2Rα, IL-2/15Rβ, γc-chain, IL-4 and IL-4Rα; IL-10, IL-15 and IL-15Rα; and IL-2, IFNγ, IL-7, IL-7Rα and IL-9Rα). The novel application of AmpliTaq Gold DNA Polymerase in a time-release PCR protocol allows specific amplification of target DNA/cDNA when only limited amounts of material are available or only low-copy-number DNA/cDNA is suspected. No IL-9 cDNA can be detected in peripheral blood mononuclear cells (PBMC) in the absence of any stimulation, thus it was difficult to amplify this target with routine PCR protocols. Here we demonstrate the reliable and reproducible amplification of IL-9 cDNA in the Hodgkin’s lymphoma cell line KM-H2, in PBMC and in stimulated PBMC. Results with AmpliTaq Gold DNA Polymerase were more sensitive and specific compared with AmpliTaq® DNA Polymerase, with and without manual hot-start procedure.

Taphonomic study of marine vertebrate remains has traditionally focused on single skeletons, lagerstätten, or bonebed genesis with few attempts to document environmental gradients in preservation. As such, establishment of a concrete taphonomic model for shallow marine vertebrate assemblages is lacking. The Neogene Purisima Formation of Northern California, a richly fossiliferous unit recording nearshore to offshore depositional settings, offers a unique opportunity to examine preservational trends across these settings.Lithofacies analysis was conducted to place vertebrate fossils within a hydrodynamic and depositional environmental context. Taphonomic data including abrasion, fragmentation, phosphatization, articulation, polish, and biogenic bone modification were recorded for over 1000 vertebrate fossils of sharks, bony fish, birds, pinnipeds, odontocetes, mysticetes, sirenians, and land mammals. These data were used to compare both preservation of multiple taxa within a single lithofacies and preservation of individual taxa across lithofacies to document environmental gradients in preservation. Differential preservation between taxa indicates strong preservational bias within the Purisima Formation. Varying levels of abrasion, fragmentation, phosphatization, and articulation are strongly correlative with physical processes of sediment transport and sedimentation rate. Preservational characteristics were used to delineate four taphofacies corresponding to inner, middle, and outer shelf settings, and bonebeds. Application of sequence stratigraphic methods shows that bonebeds mark major stratigraphic discontinuities, while packages of rock between discontinuities consistently exhibit onshore-offshore changes in taphofacies.Changes in vertebrate preservation and bonebed character between lithofacies closely correspond to onshore-offshore changes in depositional setting, indicating that the dominant control of preservation is exerted by physical processes. The strong physical control on marine vertebrate preservation and preservational bias within the Purisima Formation has implications for paleoecologic and paleobiologic studies of marine vertebrates. Evidence of preservational bias among marine vertebrates suggests that careful consideration of taphonomic overprint must be undertaken before meaningful paleoecologic interpretations of shallow marine vertebrates is attempted.

CD4+ CD25+ Foxp3+ regulatory T (Treg) cells are essential to the balance between pro- and anti-inflammatory responses. There are two major subsets of Treg cells, natural Treg (nTreg) cells that develop in the thymus, and induced Treg (iTreg) cells that arise in the periphery from CD4+ Foxp3– conventional T (Tconv) cells and can be generated in vitro. Previous work has established that both subsets are required for immunological tolerance. Additionally, in vitro-derived iTreg cells can reestablish tolerance in situations where Treg cells are decreased or defective. This review will focus on iTreg cells, drawing comparisons to nTreg cells when possible. We discuss the molecular mechanisms of iTreg cell induction, both in vivo and in vitro, review the Foxp3-dependent and -independent transcriptional landscape of iTreg cells, and examine the proposed suppressive mechanisms utilized by each Treg cell subset. We also compare the T cell receptor repertoire of the Treg cell subsets, discuss inflammatory conditions where iTreg cells are generated or have been used for treatment, and address the issue of iTreg cell stability.

The purpose of this study was to evaluate whether spatial hippocampus-dependent learning is affected by the serotonergic system and stress. Therefore, 5-HTT knockout (-/-), heterozygous (+/-) and wildtype (+/+) mice were subjected to the Barnes maze (BM) and the Morris water maze (WM), the latter being discussed as more aversive. Additionally, immediate early gene (IEG) expression, hippocampal adult neurogenesis (aN), and blood plasma corticosterone were analyzed. While the performance of 5-HTT-/- mice in the BM was undistinguishable from both other genotypes, they performed worse in the WM. However, in the course of the repeated WM trials 5-HTT-/- mice advanced to wildtype level. The experience of a single trial of either the WM or the BM resulted in increased plasma corticosterone levels in all genotypes. After several trials 5-HTT-/- mice exhibited higher corticosterone concentrations compared with both other genotypes in both tests. Corticosterone levels were highest in 5-HTT-/- mice tested in the WM indicating greater aversiveness of the WM and a greater stress sensitivity of 5-HTT deficient mice. Quantitative immunohistochemistry in the hippocampus revealed increased cell counts positive for the IEG products cFos and Arc as well as for proliferation marker Ki67 and immature neuron marker NeuroD in 5-HTT-/- mice compared to 5-HTT+/+ mice, irrespective of the test. Most differences were found in the suprapyramidal blade of the dentate gyrus of the septal hippocampus. Ki67-immunohistochemistry revealed a genotype x environment interaction with 5-HTT genotype differences in naïve controls and WM experience exclusively yielding more Ki67-positive cells in 5-HTT+/+ mice. Moreover, in 5-HTT-/- mice we demonstrate that learning performance correlates with the extent of aN. Overall, higher baseline IEG expression and increased an in the hippocampus of 5-HTT-/- mice together with increased stress sensitivity may constitute the neurobiological correlate of raised alertness, possibly impeding optimal learning performance in the more stressful WM.

Brain serotonin (5-HT) is implicated in a wide range of functions from basic physiological mechanisms to complex behaviors, including neuropsychiatric conditions, as well as in developmental processes. Increasing evidence links 5-HT signaling alterations during development to emotional dysregulation and psychopathology in adult age. To further analyze the importance of brain 5-HT in somatic and brain development and function, and more specifically differentiation and specification of the serotonergic system itself, we generated a mouse model with brain-specific 5-HT deficiency resulting from a genetically driven constitutive inactivation of neuronal tryptophan hydroxylase-2 (Tph2). Tph2 inactivation (Tph2-/-) resulted in brain 5-HT deficiency leading to growth retardation and persistent leanness, whereas a sex- and age-dependent increase in body weight was observed in Tph2+/- mice. The conserved expression pattern of the 5-HT neuron-specific markers (except Tph2 and 5-HT) demonstrates that brain 5-HT synthesis is not a prerequisite for the proliferation, differentiation and survival of raphe neurons subjected to the developmental program of serotonergic specification. Furthermore, although these neurons are unable to synthesize 5-HT from the precursor tryptophan, they still display electrophysiological properties characteristic of 5-HT neurons. Moreover, 5-HT deficiency induces an up-regulation of 5-HT(1A) and 5-HT(1B) receptors across brain regions as well as a reduction of norepinephrine concentrations accompanied by a reduced number of noradrenergic neurons. Together, our results characterize developmental, neurochemical, neurobiological and electrophysiological consequences of brain-specific 5-HT deficiency, reveal a dual dose-dependent role of 5-HT in body weight regulation and show that differentiation of serotonergic neuron phenotype is independent from endogenous 5-HT synthesis.
