We propose introducing an extended Hubbard Hamiltonian derived via the ab initio downfolding method, which was originally formulated for periodic materials, toward efficient quantum computing of molecular electronic structure calculations. By utilizing this method, the first-principles Hamiltonian of chemical systems can be coarse-grained by eliminating the electronic degrees of freedom in higher energy space and reducing the number of terms of electron repulsion integral from O ( N 4 ) to O ( N 2 ). Our approach is validated numerically on the vertical excitation energies and excitation characters of ethylene, butadiene, and hexatriene. The dynamical electron correlation is incorporated within the framework of the constrained random phase approximation in advance of quantum computations, and the constructed models capture the trend of experimental and high-level quantum chemical calculation results. As expected, the L1-norm of the fermion-to-qubit mapped model Hamiltonians is significantly lower than that of conventional ab initio Hamiltonians, suggesting improved scalability of quantum computing. Those numerical outcomes and the results of the simulation of excited-state sampling demonstrate that the ab initio extended Hubbard Hamiltonian may hold significant potential for quantum chemical calculations using quantum computers. Environmental pollution caused by heavy metals are problems worldwide. In particular, pollution and poisoning by lead ions (Pb2+) continue to be common and serious problems. Hence, there is a need for a widely usable method to easily detect Pb2+ from solutions containing organic materials from environmental water such as seas, ponds, etc. Here, we established a system to easily detect Pb2+ from such mixture solutions using Pb2+ binding peptide immobilized beads (peptidyl beads) and ultraviolet (UV) absorption measurements. This method could detect Pb2+ at low concentrations equivalent to inductively coupled plasmon-atomic emission spectroscopy (ICP-AES). Using the detected values to create a calibration curve, it was found that there was a positive correlation between the concentration of Pb2+ and absorbance, which also made it possible to quantify sub-µM Pb2+ in the solutions. Furthermore, Pb2+ was detected and quantified under mixed conditions of environmental water such as seas, rivers, and ponds. Objective: Pain is subjective, and self-reporting pain might be challenging. Studies conducted to detect pain using biological signals and real-time self-reports pain are limited. We evaluated the feasibility of collecting pain data on healthy females' menstrual pain and conducted preliminary analysis. Results: Five healthy adult females participated. They wore two wristwatch devices (Silmee and Fitbit) and a Holter ECG (electrocardiogram) during menstruation to record the pain intensity and timing. Subsequently, we analyzed the correlation between heart and pulse rates and assessed pre- and post-pain biometric differences. We collected sixty pain records from five participants. The correlation coefficients between heart rate and pulse rate ranged from 0.79 to 0.95 with Holter ECG vs. Fitbit and 0.32 to 0.74 with Holter ECG vs. Silmee. Analysis revealed significant changes in motion frequency post-pain (p = 0.04). For abdominal pain with a numerical rating scale score of ≥ 4 (n = 13), motion frequency (p < 0.001) and pulse rate (p = 0.02) showed significant differences post-pain compared to baseline values. Healthy females could wear the wristwatch device in daily life and report pain in real time. Only a few studies have explored whether high-dose-rate interstitial brachytherapy (HDR-ISBT) can be indicated as a palliative/symptomatic treatment. We present the good results of palliative treatment using HDR-ISBT combined with external beam radiotherapy (ERT) in a patient of base of tongue cancer (cT4aN1M0). The patient was an 81-year-old male who complained of local pain. He had a previous irradiation history for head and neck cancer receiving ERT with systemic chemotherapy and radical surgery 15 years ago. Since it might be difficult for him to receive radical radiation doses using ERT alone, palliative ERT of relatively lower doses of 37.5 Gy in 15 fractions was selected. One month after ERT, HDR-ISBT was implemented as a booster. Considering the burden on physical condition, single-fraction HDR-ISBT was selected. We employed a new technique in which we did not penetrate the ventral surface of the tongue to reduce the risk of infection and bleeding. The planning-aim dose was 9.5 Gy. The dose that covered 90% of the clinical target volume was 9.6 Gy. The treatment ended without any problems. Acute complications were not observed. The tumor size decreased, and local pain disappeared at post-treatment day 84. No late complications were observed. Two years and 8 months after the treatment, the patient is alive without any obvious recurrence. Additional single-fraction HDR-ISBT boost may be a useful modality as a palliative/symptomatic intent. Accidental bending of the snare sheath occasionally occurs during cold snare polypectomy (CSP). We aimed to demonstrate whether snare bending reduces resection ability and, if it does, what causes this reduction. Using currently available CSP snares and prototype snares, we investigated changes in the resection ability of bent snares as well as the stiffness of their sheaths and wire spindles. Furthermore, to evaluate the performance of the bent snares, we measured the completion rate of CSP for 5-mm-diameter pseudo-lesions on the porcine rectal mucosa. Snare bending significantly decreased resection ability and wire spindle stiffness. Compared with bending the snares once, bending the snares five times further decreased the resection ability and wire spindle stiffness. A strong correlation was observed between wire spindle stiffness and resection ability after snare bending (correlation coefficient = 0.82; P < 0.001). Additionally, snare bending reduced the completion rate of CSP. Bending a CSP snare causes decreased wire spindle stiffness, thereby reducing its resection ability and potentially lowering the resection completion rate. During fertilization, a series of reactions between the eggs and spermatozoa proceed predominantly in a species-specific manner. The molecules mediating these species-specific reactions remain unknown except in a few organisms. In this study, we focused on two species belonging to the phylum Nemertea, Kulikovia alborostrata and K. fulva, and explored molecules involved in species-specific interactions between gametes. Orthologs of molecules known to be involved in species-specific reactions were not expressed in the ovaries of these two species. In contrast, we identified six novel proteins, named NeZPL1–NeZPL6, containing a ZP module. Among these, we found that NeZPL6 is located on the surface of an unfertilized egg and is suggested to be involved in its interaction with spermatozoa. Furthermore, we found an indel of three amino acids in the EGF-like domain of NeZPL6, which possibly confers species specificity to this interaction. PURPOSE: Inherited cancer susceptibility is often not suspected in the absence of a significant cancer family history. Pathogenic germline variants in pancreatic cancer are well-studied, and routine genetic testing is recommended in the guidelines. However, data on rare periampullary cancers other than pancreatic cancer are insufficient. We compared the prevalence of germline susceptibility variants in patients with pancreatic cancer and nonpancreatic periampullary cancers. MATERIALS AND METHODS: Six hundred and eight patients who had undergone pancreaticoduodenal resection at a tertiary referral hospital were studied, including 213 with pancreatic ductal adenocarcinoma, 172 with ampullary cancer, 154 with distal common bile duct cancer, and 69 with duodenal adenocarcinoma. Twenty cancer susceptibility and candidate susceptibility genes were sequenced, and variant interpretation was assessed by interrogating ClinVar and PubMed. RESULTS: Pathogenic or likely pathogenic, moderate- to high-penetrant germline variants were identified in 46 patients (7.7%), including a similar percentage of patients with pancreatic (8.5%) and nonpancreatic periampullary cancer (7.1%). Low-penetrant variants were identified in an additional 11 patients (1.8%). Eighty-nine percent of the moderate- to high-penetrant variants involved the major cancer susceptibility genes BRCA2 , ATM , BRCA1 , CDKN2A , MSH2 / MLH1 , and PALB2 ; the remaining 11% involved other cancer susceptibility genes such as BRIP1 , BAP1 , and MSH6. Almost all pathogenic variant carriers had a family history of cancer. CONCLUSION: Patients with pancreatic and nonpancreatic periampullary cancer have a similar prevalence of pathogenic cancer susceptibility variants. We investigated the field-dependent Néel relaxation time of magnetic nanoparticles (MNPs) in an AC excitation field. Specifically, a fundamental component of the magnetization for immobilized MNPs was studied by numerical simulation while changing the frequency f and amplitude Hac of the field. From the simulation results, we clarified the Hac dependence of the effective Néel relaxation time τN,e and obtained an empirical expression for τN,e(Hac) for the first time. The expression was obtained for the cases when the angle of the easy axis of magnetization in MNPs is fixed and randomly distributed. Using the Hac dependencies of τN,e and the previously reported Brownian relaxation time τB,e, we showed that the behavior of suspended MNPs changes from Brownian-dominant to Néel-dominant when Hac increases, even when the MNP parameters are fixed, and we obtained an expression for the boundary field between them. Furthermore, we classified several types of responses for the suspended MNPs in the AC field using the magnitude relationship among τN,e(Hac), τB,e(Hac), and 1/(2πf). Finally, we experimentally verified the classification, and reasonable agreement was observed between the experiment and analysis. In this research, we conducted two studies to validate a multidimensional scale of fan engagement behavior. In Study 1, we generated survey items through a systematic review of the relevant literature, collected data from fans of professional baseball (n = 319) and soccer (n = 301), and provided evidence for the construct and concurrent validity of the scale composed of six dimensions. In Study 2, we reassessed construct validity in professional baseball (n = 582) and found that fan engagement behavior was represented by the proposed six dimensions with a final list of 21 items. Further, our predictive analysis throughout a season showed that fan engagement behavior fully mediated the relationship between predictor (team identification and awareness of fan engagement initiatives) and outcome variables (media viewing frequency, attendance frequency, and flourishing). Illumination stress (IS) and negative bias under illumination stress (NBIS) cause considerable device instability in thin-film transistors based on amorphous In–Ga–Zn–O (a-IGZO). Models using in-gap states are suggested to explain device instability. Therefore, to provide reliably their density of states (DOS), this study investigated the valence band, conduction band, and in-gap states of an a-IGZO thin film. The DOS of in-gap states was directly determined in a dynamic range of six orders of magnitude through constant final state yield spectroscopy (CFS-YS) using low-energy and low-flux photons. Furthermore, light irradiation irreversibly induced extra in-gap states near the Fermi level and shifted the Fermi level to the vacuum level side, which should be related to the device instability due to IS and NBIS. Hard x-ray photoemission spectroscopy and ultraviolet photoemission spectroscopy using synchrotron radiation observed the large DOS of in-gap states near the Fermi level as in previous works. Here, we reveal that they are not intrinsic electronic states of undamaged a-IGZO, but induced by the intense measurement light of synchrotron radiation. This study demonstrates that CFS-YS is useful for determining the reliable DOS of the in-gap states for samples that are sensitive to light irradiation. The absorption spectrum measured through photothermal deflection spectroscopy is interpreted based on DOS directly determined via photoemission spectroscopies. This indicates that the line shape in the energy region below the region assigned to the Urbach tail in previous works actually roughly reflects the DOS of occupied in-gap states. We derive asymptotic properties of penalized estimators for singular models for which identifiability may break and the true parameter values can lie on the boundary of the parameter space. Selection consistency of the estimators is also validated. The problem that the true values lie on the boundary is solved by our previous results applicable to singular models, besides, penalized estimation and non-ergodic statistics. To overcome non-identifiability, we consider a suitable penalty such as the non-convex Bridge and the adaptive Lasso that stabilize the asymptotic behavior of the estimator and shrink inactive parameters. Then the estimator converges to one of the most parsimonious values among all the true values. The oracle property can also be obtained even if likelihood ratio tests for model selection are labor intensive due to singularity of models. The purpose of this article is to develop a general parametric estimation theory that allows the derivation of the limit distribution of estimators in non-regular models where the true parameter value may lie on the boundary of the parameter space or where even identifiability fails. For that, we propose a more general local approximation of the parameter space (at the true value) than previous studies. This estimation theory is comprehensive in that it can handle penalized estimation as well as quasi-maximum likelihood estimation (in the ergodic or non-ergodic statistics) under such non-regular models. In penalized estimation, depending on the boundary constraint, even the concave Bridge estimator does not necessarily give selection consistency. Therefore, we describe some sufficient condition for selection consistency, precisely evaluating the balance between the boundary constraint and the form of the penalty. A dentinogenic ghost cell tumor (DGCT) is a rare benign odontogenic tumor that commonly shows characteristics of solid proliferation and has a relatively high risk of recurrence after surgical treatment. We herein report a case of a central DGCT that occurred in the maxilla and resulted in bone expansion. This study highlights new imaging findings (particularly magnetic resonance imaging) along with histopathological observations. In addition, we conducted a review of the existing literature on this rare tumor. A 37-year-old man developed swelling around the right cheek. A benign odontogenic tumor such as ameloblastoma was suspected based on the imaging examination findings (including bone expansion and the internal characteristics of the tumor) on panoramic imaging, computed tomography, and magnetic resonance imaging. The lesion was surgically excised from the right maxilla. Postoperative histopathological examination led to a definitive diagnosis of central DGCT. The tumor comprised epithelial neoplastic islands, resembling ameloblastoma, inside tight fibroconnective tissue; masses of ghost cells and formation of dentin were also observed. We had suspected that the minute high-density region around the molars on the imaging examinations represented alveolar bone change; however, it represented dentin formation. This led to difficulty diagnosing the lesion. Although DGCT may present characteristic findings on imaging examinations, its occurrence is infrequent, and in some cases, the findings may include the presence or absence of an impacted tooth without obvious calcification. The present case suggests that we should consider the possibility of an odontogenic tumor with calcification when high-density structures are observed inside the lesion. [ABSTRACT FROM AUTHOR]

Chronic fibrotic tissue disrupts various organ functions. Despite significant advances in therapies, mortality and morbidity due to heart failure remain high, resulting in poor quality of life. Beyond the cardiomyocyte-centric view of heart failure, it is now accepted that alterations in the interstitial extracellular matrix (ECM) also play a major role in the development of heart failure. Here, we show that protein kinase N (PKN) is expressed in cardiac fibroblasts. Furthermore, PKN mediates the conversion of fibroblasts into myofibroblasts, which plays a central role in secreting large amounts of ECM proteins via p38 phosphorylation signaling. Fibroblast-specific deletion of PKN led to a reduction of myocardial fibrotic changes and cardiac dysfunction in mice models of ischemia-reperfusion or heart failure with preserved ejection fraction. Our results indicate that PKN is a therapeutic target for cardiac fibrosis in heart failure. Despite significant advances in therapies, mortality and morbidity resulting from heart failure remain high. Background: Numerous epidemiological studies have shown that hypertriglyceridemia is a significant risk factor for cardiovascular diseases (CVD). However, large clinical studies on triglyceride‐lowering therapy have yielded inconsistent results. In the current review, we reassess the importance of triglyceride‐lowering therapy in preventing CVD based on previous literature and the recently published findings of the PROMINENT trial. Methods: This narrative review is based on literature and public documents published up to November 2023. Results: Meta‐analyses of trials on peroxisome proliferator‐activated receptor α agonists and triglyceride‐lowering therapy, including the PROMINENT trial, have indicated that triglyceride‐lowering therapy can reduce CVD events. Mendelian randomization studies have also indicated that triglyceride is indeed a true risk factor for coronary artery disease, leaving no doubt about its relationship to CVD. Meanwhile, the negative results from the PROMINENT trial were likely due to the insufficient triglyceride‐lowering effect, slight increases in low‐density lipoprotein cholesterol and apolipoprotein B, and the inclusion of mostly high‐intensity statin users as target patients. It is unlikely that adverse events counteracted the effectiveness of pemafibrate on outcomes. Additionally, pemafibrate has shown positive effects on non‐alcoholic fatty liver disease and peripheral artery disease. Conclusion: Although the PROMINENT trial did not demonstrate the significance of pemafibrate as a triglyceride‐lowering therapy in a specific population, it does not necessarily negate the potential benefits of treating hypertriglyceridemia in reducing CVD events. It is necessary to explore appropriate populations that could benefit from this therapy, utilize data from the PROMINENT trial and other databases, and validate findings in real‐world settings. Background: The cancer genome contains several driver mutations. However, in some cases, no known drivers have been identified; these remaining areas of unmet needs, leading to limited progress in cancer therapy. Whole-genome sequencing (WGS) can identify non-coding alterations associated with the disease. Consequently, exploration of non-coding regions using WGS and other omics data such as ChIP-sequencing (ChIP-seq) to discern novel alterations and mechanisms related to tumorigenesis have been attractive these days. Methods: Integrated multi-omics analyses, including WGS, ChIP-seq, DNA methylation, and RNA-sequencing (RNA-seq), were conducted on samples from patients with non-clinically actionable genetic alterations (non-CAGAs) in lung adenocarcinoma (LUAD). Second-level cluster analysis was performed to reinforce the correlations associated with patient survival, as identified by RNA-seq. Subsequent differential gene expression analysis was performed to identify potential druggable targets. Results: Differences in H3K27ac marks in non-CAGAs LUAD were found and confirmed by analyzing RNA-seq data, in which mastermind-like transcriptional coactivator 2 (MAML2) was suppressed. The down-regulated genes whose expression was correlated to MAML2 expression were associated with patient prognosis. WGS analysis revealed somatic mutations associated with the H3K27ac marks in the MAML2 region and high levels of DNA methylation in MAML2 were observed in tumor samples. The second-level cluster analysis enabled patient stratification and subsequent analyses identified potential therapeutic target genes and treatment options. Conclusions: We overcome the persistent challenges of identifying alterations or driver mutations in coding regions related to tumorigenesis through a novel approach combining multi-omics data with clinical information to reveal the molecular mechanisms underlying non-CAGAs LUAD, stratify patients to improve patient prognosis, and identify potential therapeutic targets. Background: Brain metastasis (BrM) is prevalent among patients with NSCLC, and surgical resection of BrM constitutes a promising treatment strategy for local management and histopathological diagnosis, although it is offered for a select group of patients. Limited information exists concerning the improvement in performance status (PS) following BrM resection or the outcomes stratified by subsequent systemic therapy. Methods: We conducted a retrospective single-center cohort study including NSCLC patients with surgically resected BrM and focused on the improvement in PS and subsequent therapy after BrM resection. Results: 71 patients were included, and the median overall survival was 18.3 months (95% confidence interval [95% CI]: 8.7, not reached). Patients with NSCLC who underwent surgical resection of BrM showed significant improvement in PS (18% and 39% showed ECOG PS of 0–1, before and after BrM resection, respectively [p = 0.006]), and patients with PS improvement were younger than those with PS unimprovement (median, 62 years versus 66 years; p = 0.041). Regarding subsequent systemic therapy after BrM resection, 21 patients (30%) received cytotoxic chemotherapy, 14 patients (20%) received tyrosine kinase inhibitors (TKIs), 3 patients (4%) received immune checkpoint inhibitors (ICIs), and 21 patients (30%) received no subsequent therapy. The survival outcomes of patients stratified by subsequent systemic treatments suggested the tendency that those who received TKI or ICI showed better survival outcomes, although a small number of patients hindered statistical comparisons. Conclusions: We describe the outcomes of patients with NSCLC who underwent surgical resection of BrM, revealing that younger patients were more likely to anticipate improvement in PS, and patients who received TKI or ICI after BrM resection tended to exhibit a more preferable prognosis. The functional maturation of the pituitary gland requires adequate cell differentiation and vascular network formation. Although spatiotemporal signaling and transcription factors are known to govern pituitary development, the involvement of primary cilia, nonmoving hair-like organelles, remains unclear. In this study, we uncovered the contribution of primary cilia to cell-type determination and vascular network formation during pituitary development. Homozygous knockout mice lacking a ciliary kinase, Dyrk2 −/−, exhibit abnormalities in ciliary structure and pituitary hypoplasia, accompanied by varying degrees of failure in differentiation among all types of hormone-producing cells in the anterior lobe. Aberrations in cell differentiation in Dyrk2 −/− mice arise from a decrease in the expression of crucial transcription factors, Lhx4 , Lhx3 , and Prop1 , resulting from the inactivity of Hedgehog (Hh) signaling during the early stages of development. Furthermore, the loss of Dyrk2 results in vascular system abnormalities during the middle to late stages of development. Mechanistically, transcriptome analyses revealed the downregulation of vitronectin-integrin αvβ3-VEGFR2 signaling, essential for orchestrating vascular development. Collectively, our findings demonstrate that primary cilia play a pivotal role as critical regulators of cell survival, cell determination, and angiogenesis during pituitary gland development through the activation of Hh signaling. Background: Delivering cancer treatment to elderly patients with dementia is often challenging. We describe performing palliative surface mold brachytherapy (SMBT) in an elderly patient with advanced dementia for pain control using music therapy to assist with agitation. Case Description: The patient was a 97-year-old Japanese woman with advanced dementia. Exudate was observed from her tumor, and she complained of Grade 2 severity pain using Support team assessment schedule (STAS), especially when undergoing would dressings. Given her advanced dementia, she was not considered a candidate for radical surgery or external beam radiotherapy. We instead treated her with high-dose-rate (HDR) SMBT. Due to her advanced dementia associated with agitation, she could not maintain her position. She was able to remain calm while listening to traditional Japanese enka music, which enables our team to complete her radiation without using anesthetics or sedating analgesics. Her localized pain severity decreased ≤21 days and the exudate fluid disappeared ≤63 days after HDR-SMBT. Her tumor was locally controlled until her death from intercurrent disease 1 year after HDR-SMBT. Discussion: Single fraction palliative HDR-SMBT was useful for successful treatment of skin cancer in an elderly patient. Traditional Japanese music helped reduce her agitation to complete HDR-SMBT. Aim: To investigate metabolic risk factors (RFs) that accumulated over 20 years related to left ventricular mass index (LVMI), relative wall thickness (RWT) and LV remodelling patterns in participants with versus without early‐onset type 2 diabetes (T2D) or prediabetes (pre‐D). Methods: A total of 287 early‐onset T2D/pre‐D individuals versus 565 sociodemographic‐matched euglycaemic individuals were selected from the Coronary Artery Risk Development in Young Adults (CARDIA) study, years 0‐25. We used the area under the growth curve (AUC) derived from quadratic random‐effects models of four or more repeated measures of RFs (fasting glucose [FG], insulin, triglycerides [TG], low‐density lipoprotein cholesterol, high‐density lipoprotein cholesterol (HDL‐c), total cholesterol (total‐c), blood pressure and body mass index) to estimate the cumulative burden, and their associations with LV outcomes. Results: One standard deviation greater AUC of log (TG) (per 0.48) and HDL‐c (per 13.5 mg/dL) were associated with RWT (β 0.21 and −0.2) in the early‐onset T2D/pre‐D group, but not in the euglycaemia group (β 0.01 and 0.05, P interactions.02 and.03). In both the early‐onset T2D/pre‐D and euglycaemia groups, greater AUCs of log (FG) (per 0.17) and log (insulin) (per 0.43) were associated with higher RWT (β ranges 0.12‐0.24). Greater AUCs of systolic blood pressure (per 10 mmHg) and diastolic blood pressure (per 7.3 mmHg) were associated with higher RWT and LVMI, irrespective of glycaemic status (β ranges 0.17‐0.28). Cumulative TG (odds ratio 3.4, 95% confidence interval: 1.8‐6.3), HDL‐c (0.23, 0.09‐0.59), total‐c (1.9, 1.1‐3.1) and FG (2.2, 1.25‐3.9) were statistically associated with concentric hypertrophy in the T2D/pre‐D group only. Conclusions: Sustained hyperglycaemia and hyperinsulinaemia are associated with RWT, and those individuals with early T2D/pre‐D are potentially at greater risk because of their higher levels of glucose and insulin. Simple Summary: Mucosal malignant melanoma is a type of head and neck cancer with a high mortality rate. Although favorable local control has been reported when using carbon ion radiotherapy (CIRT) to treat mucosal malignant melanoma of the head and neck, the prognosis remains unsatisfactory because of the tendency for early distant metastasis. In recent years, favorable outcomes have been reported for the use of immune checkpoint inhibitors (ICIs) as adjuvant therapy for cutaneous malignant melanoma, indicating their potential applicability to mucosal malignant melanoma. In addition to achieving favorable local control with CIRT, the control of distant metastasis using ICIs is expected to prolong survival. The findings of this analysis indicate that the use of ICIs as adjuvant therapy can improve prognosis following CIRT, offering a new treatment option. The development of new treatment strategies to improve the prognosis of mucosal malignant melanoma of the head and neck (MMHN) after carbon ion radiotherapy (CIRT) is essential because of the risk of distant metastases. Therefore, our objective was to evaluate the outcomes of immune checkpoint inhibitor (ICI) treatment to justify its inclusion in the regimen after CIRT. Thirty-four patients who received CIRT as an initial treatment were included in the analysis and stratified into three groups: those who did not receive ICIs (Group A), those who received ICIs after recurrence or metastasis (Group B), and those who received ICIs as adjuvant therapy after CIRT (Group C). In total, 62% of the patients (n = 21) received ICIs. The 2-year local control and overall survival (OS) rates for all patients were 90.0% and 66.8%, respectively. The 2-year OS rates for patients in Groups A, B, and C were 50.8%, 66.7%, and 100%, respectively. No significant differences were observed between Groups A and B (p = 0.192) and Groups B and C (p = 0.112). However, a significant difference was confirmed between Groups A and C (p = 0.017). Context: Nordic hamstring strength (NH strength) and single leg bridge test (SLBT) scores are used to predict the risk of hamstring strain injury. Although NH strength and SLBT scores may be related, the relationship between NH strength and SLBT score remains unknown. Objectives: This study investigated the relationship between NH strength and SLBT scores in university soccer players. Design: Cross-sectional study. Setting: Research laboratory. Participants: 38 male university soccer players. Main Outcome Measures: NH strength and SLBT scores. Interventions: A participant was instructed to lean forward gradually at the slowest possible speed from a kneeling posture with the knee joint flexed 90° for the NH strength measurement. Participants in the SLBT crossed their arms over their chests, pushed down from their heels, and lifted their hips off the ground as many times as they could until they failed. We investigated the relationship between NH strength and SLBT scores in the left and right sides using Spearman rank correlation coefficient. Additionally, we calculated the percentage of left–right asymmetry in NH strength and SLBT scores and investigated the relationship between these variables using Pearson correlation coefficient. Results: There were no significant correlations between NH strength and SLBT scores in the right (rs =.239, P =.16) and left (rs =.311, P =.065) legs. Furthermore, there was no significant relationship between NH strength and SLBT between-limb asymmetry (r =.073, P =.671). Conclusions: NH strength and SLBT scores could be different indexes, indicating either maximal muscle strength or muscle endurance. We investigated the association between the cross-sectional area (CSA) of the gluteus medius muscle (GMM) and activities of daily living in patients with hip fractures. This retrospective cohort study comprised 111 patients aged ≥65 years who underwent hip fracture rehabilitation. The CSA of the GMM was measured using computed tomography scans in the early stages of hospitalization. The group with decreased CSA of the GMM had a median GMI ≤17 cm2/m2 for male patients and ≤16 cm2/m2 for female patients. Patients in the group with decreased CSA of the GMM had lower functional independence measure gains than those in the control group. After adjusting for confounders, we found that decreased CSA of the GMM was significantly associated with lower functional independence measure gains (β: −0.432, p <.001). Mixing Gibbs energy and phase equilibria of aqueous solutions of polyglycine were studied theoretically by means of polymer reference interaction site model integral equation theory combined with the Gibbs–Duhem method. In addition to the ordinary liquid–liquid phase separation between dilute and concentrated solutions, the theoretical calculation predicted the coexistence of two coacervate phases, namely, the lower- and higher-density coacervates. The relative thermodynamic stabilities of these two phases change with the polymerization degree of polyglycine. The higher-density coacervate phase was rapidly stabilized by increasing the polymer length, and the lower-density phase became metastable at large polymers. The hydrogen bonds between the peptide chains were strengthened, and water was thermodynamically destabilized in the higher-density coacervate. This study evaluated the relationship between the muscle mass of the gluteus medius (GM) and skeletal muscle mass index (SMI) measured in patients with hip fractures. In this study, 141 patients with hip fractures were divided into those with high or low SMI. The GM index (GMI) was calculated by dividing the GM by the square of the height in meters. The correlation between GMI and SMI was subsequently analyzed, and cutoff values for determining the loss of skeletal muscle mass were calculated using the receiver operating characteristic curve. GMI and SMI showed a positive correlation for both sexes (male: r =.890, female: r =.626, p <.001). The GMI cutoff values were 19.460 cm2/m2 for males and 17.850 cm2/m2 for females. Two Dion–Jacobson (DJ) double-layered perovskites CsNdNb2O7 and RbNdNb2O7 exhibit hybrid improper ferroelectricity, in which spontaneous polarization is induced by coupling of two types of nonpolar oxygen octahedral rotations (OORs). Their crystal structures adopt common polar distortion modes, although they exhibit distinct OOR patterns. In this study, complete solid-solution Cs1−xRbxNdNb2O7 ceramics (x = 0, 0.25, 0.5, 0.75, and 1) have been synthesized by conventional solid-state reaction methods, and the temperature-composition phase diagram has been established by various methods including variable-temperature synchrotron X-ray diffractometry, differential scanning calorimetry, optical second harmonic generation, and dielectric permittivity measurements. The powder X-ray diffractometry reveals that Cs-rich compounds (x ≤ 0.25) crystalize into polar P21am structures with a−a−c+/a−a−c+ OORs at room temperature, while Rb-rich compounds (x ≥ 0.5) adopt polar I2cm structures with a−a−c+/−(a−a−c+) OORs. All the solid solutions exhibit ferroelectricity at room temperature. Upon heating, successive phase transitions involving the loss of polar distortions and OORs have been observed. The phase transition temperatures of the solid solutions with intermediate compositions are lower than those predicted based on Vegard's law, indicating structural frustration effects due to the random arrangement of two alkali metal ions. The epithelial and mesenchymal features of colorectal adenocarcinoma (CRAC) cell lines were compared in two-dimensional (2D) and three-dimensional (3D) cultures. In 2D cultures, the three CRAC cell lines exhibited epithelial characteristics with high E-cadherin and low vimentin levels, whereas two exhibited mesenchymal traits with opposite expression patterns. In 3D cultures using low-attachment plates, mesenchymal cells from 2D cultures showed reduced vimentin mRNA levels. Morphologically, the five CRAC cell lines appeared similarly shaped in 2D culture but formed different structures in 3D culture. Epithelial DLD-1 and mesenchymal COLO-320 cells produced large granular spheres, whereas epithelial HCT-15 cells formed small solid spheres. Tubular structures were observed in epithelial CACO-2 and mesenchymal SW480 spheres. Desmosome-like structures developed in epithelial CRAC cells, whereas entosis was observed in CACO-2, HCT-15, and SW480 cells. The Ki-67-positive proliferating cell count varied in 2D and 3D cultures of epithelial cells but remained high and unchanged in mesenchymal cells. These findings suggest that while CRAC cells display distinct epithelial and mesenchymal properties in 2D cultures, they form diverse 3D structures, irrespective of these traits. Viruses that infect cyanobacteria are an integral part of aquatic food webs, influencing nutrient cycling and ecosystem health. However, the significance of virus host range, replication efficiency, and host compatibility on cyanobacterial dynamics, growth, and toxicity remains poorly understood. In this study, we examined the effects of cyanophage additions on the dynamics and activity of optimal, sub-optimal, and non-permissive cyanobacterial hosts in cultures of Microcystis aeruginosa and Raphidiopsis raciborskii. Our findings reveal that cross-infectivity can substantially reduce the proliferative success of the cyanophage under conditions of high-density of sub-optimal hosts which suggests phage dispersal limitation as a result of shared infections, in turn impairing their top-down control over the host community. Furthermore, we found that cyanophage addition triggers host strain-specific responses in photosynthetic performance, population size and toxin production, even among non-permissive hosts. These non-lytic effects suggest indirect impacts on co-existing cyanobacteria, increasing the overall complexity and variance in many ecologically relevant cyanobacterial traits. The high variability in responses observed with a limited subset of cyanophage-cyanobacteria combinations not only highlights the intricate role of viral infections in microbial ecosystems but also underscores the significant challenges in predicting the composition, toxicity, and dynamics of cyanobacterial blooms. Neutron imaging is a nondestructive and noninvasive inspection technique with a wide range of potential applications. However, the fundamentals of this technique still need to be improved, one of which involves achieving micrometer scale or even better resolution, which is a challenging task. Recently, a high-resolution neutron imaging device based on fine-grained nuclear emulsions was developed. Although these detectors demonstrate exceptionally high resolutions, they have several limitations. Furthermore, these detectors require an additional chemical development process and are thus not reusable. To overcome these limitations, we investigated whether neutron imaging devices based on fluorescent nuclear track detectors were suitable for high-resolution neutron imaging. Fluorescent nuclear track detectors are reusable solid-state detectors that do not require additional chemical processing. A novel technique combining neutron imaging based on fluorescent nuclear track detectors with a neutron converter layer formed using B C was developed with unprecedented resolution. The neutron imaging of a gadolinium-based grating with a periodic structure of 9 m was performed using the proposed fluorescent nuclear track detector-based neutron imaging device, and the grating structure was successfully resolved. Background: Japan has one of the lowest COVID-19 death rates, while the annual suicide rate in 2020 has risen for the first time since 2007. This study aimed to identify high-risk populations and assess the impact of medical visits on suicide trends post-COVID-19 pandemic in Japan. Method: This quasi-experimental study analyzed a population-based database from Hyogo Prefecture (2012–2022). Interrupted time-series analyses identified level and trend changes in monthly suicide rates per 1 million population during the exposure period (2020–2022) versus the control period (2012–2019). Regular visits to primary care and psychiatry stratified analysis. Results: 2181 cases were analyzed, with two-thirds male and a median age of 54. Primary care physicians and psychiatric history were present in 69% and 40% of patients. The study found significant level changes in suicide rates overall (4.14, 95% CI: 1.70, 6.58) among individuals without regular primary care physician visits (2.83, 95% CI: 1.35, 4.32) and without psychiatric visits (2.85, 95% CI: 0.56, 5.14). In contrast, no significant changes were observed in individuals with regular primary care (0.99, 95% CI: -0.78, 2.76) or regular psychiatric visits (0.59, 95% CI: -0.98, 2.16). The trend changes were not significant in any of the groups. Conclusion: This study suggests that a history of attending a medical institution may have prevented the rapid increase in suicides during the early stages of the COVID-19 pandemic. The Japanese Global Observing SATellite for Greenhouse gases and Water cycle (GOSAT-GW) will be an Earth-observing satellite to conduct global observations of atmospheric carbon dioxide (CO2), methane (CH4), and nitrogen dioxide (NO2) simultaneously from a single platform. GOSAT-GW is the third satellite in the series of the currently operating Greenhouse gases Observing SATellite (GOSAT) and GOSAT-2. It will carry two sensors, the Total Anthropogenic and Natural emissions mapping SpectrOmeter-3 (TANSO-3) and the Advanced Microwave Scanning Radiometer 3 (AMSR3), with the latter dedicated to the observation of physical parameters related to the water cycle. TANSO-3 is a high-resolution grating spectrometer designed to measure reflected sunlight in the visible to short-wave infrared spectral ranges. It aims to retrieve the column-averaged dry-air mole fractions of CO2 and CH4 (denoted as XCO2 and XCH4, respectively), as well as the vertical column density of tropospheric NO2. The TANSO-3 sensor onboard GOSAT-GW will utilize the wavelength bands of 0.45, 0.76, and 1.61 µm for NO2, O2, and CO2 and CH4 retrievals, respectively. GOSAT-GW will fly in a sun-synchronous orbit with a local overpass time of approximately 13:30 and a 3-day ground-track repeat cycle. The TANSO-3 sensor has two observation modes in the push-broom operation: Wide Mode, which provides globally covered maps with a 10-km spatial resolution within 3 days, and Focus Mode, which provides snapshot maps over targeted areas with a high spatial resolution of 1–3 km. The objectives of the GOSAT-GW mission include (1) monitoring atmospheric global-mean concentrations of greenhouse gasses (GHGs), (2) verifying national anthropogenic GHG emissions inventories, and (3) detecting GHG emissions from large sources, such as megacities and power plants. A comprehensive validation exercise will be conducted to ensure that the sensor products' quality meets the required precision to achieve the above objectives. With a projected operational lifetime of seven years, GOSAT-GW will provide vital space-based constraints on both anthropogenic and natural GHG emissions. These measurements will contribute significantly to climate change mitigation efforts, particularly by supporting the Global Stocktake (GST) mechanism, a key element of the Paris Agreement. Purpose: This study aimed to validate the accuracy of the Active Style Pro HJA-750C (ASP) in measuring metabolic equivalents (METs) during walking and reaching tasks in individuals with subacute stroke using a respiratory gas analyzer as a reference. Methods: Twenty-three hospitalized patients with subacute stroke participated in this study. They performed sitting and standing reaching tasks, as well as walking while wearing a VO2 Master respiratory gas analyzer and ASP devices on both the paretic and non-paretic sides. The METs values recorded by the ASP were compared with those obtained using a VO2 Master respiratory gas analyzer. Pearson's correlation coefficients were calculated for each task, and Bland–Altman plots were used to assess the agreement between the two methods. Results: The ASP demonstrated good concurrent validity, with correlation coefficients of 0.71 and 0.74 for the sitting reaching task, 0.75 and 0.79 for the standing reaching task, and 0.83 and 0.85 for walking when the ASP was placed on the paretic and non-paretic sides, respectively. Bland–Altman analysis indicated no significant fixed or proportional errors. The ASP accurately measures METs whether worn on the affected or unaffected side of the waist. Conclusion: The ASP provides valid measurements of physical activity during walking and reaching tasks in patients with subacute stroke. Antibodies are extensively used in biomedical research, clinical fields, and disease treatment. However, to enhance the reproducibility and reliability of antibody-based experiments, it is crucial to have a detailed understanding of the antibody's target specificity and epitope. In this study, we developed a high-throughput and precise epitope analysis method, DECODE (Decoding Epitope Composition by Optimized-mRNA-display, Data analysis, and Expression sequencing). This method allowed identifying patterns of epitopes recognized by monoclonal or polyclonal antibodies at single amino acid resolution and predicted cross-reactivity against the entire protein database. By applying the obtained epitope information, it has become possible to develop a new 3D immunostaining method that increases the penetration of antibodies deep into tissues. Furthermore, to demonstrate the applicability of DECODE to more complex blood antibodies, we performed epitope analysis using serum antibodies from mice with experimental autoimmune encephalomyelitis (EAE). As a result, we were able to successfully identify an epitope that matched the sequence of the peptide inducing the disease model without relying on existing antigen information. These results demonstrate that DECODE can provide high-quality epitope information, improve the reproducibility of antibody-dependent experiments, diagnostics and therapeutics, and contribute to discover pathogenic epitopes from antibodies in the blood. Background: A new circulating biomarker superior to carbohydrate antigen 19–9 (CA19-9) is needed for diagnosing pancreatobiliary cancer (PBca). The aim of this study was to identify serum microRNA (miRNA) signatures comprising reproducible and disease-related miRNAs. Methods: This multicenter study involved patients with treatment-naïve PBca and healthy participants. The optimized serum processing conditions were evaluated using t-distributed stochastic neighbor embedding (t-SNE) visualization. Serum miRNA candidates for disease association were selected using weighted gene coexpression network analysis (WGCNA). A miRNA signature combining multiple serum miRNAs was tested in exploratory, validation, and independent validation sets. The synthesis and secretion of diagnostic miRNAs were evaluated using human pancreatic cancer cells. Results: In total, 284 (150 healthy and 134 PBca) of 827 serum samples were processed within 2 h of blood collection before freezing, distributed in the same area as that in the t-SNE map, and assigned to an exploratory set. The 193 optimized samples were assigned to either the validation (50 healthy, 47 PBca) or independent validation (50 healthy, 46 PBca) set. Index-1, a combination of five serum miRNAs (hsa-miR-1343-5p, hsa-miR-4632-5p, hsa-miR-4665-5p, hsa-miR-665, and hsa-miR-6803-5p) with disease association in WGCNA, showed a sensitivity and specificity of > 80% and an AUC outperforming that of CA19-9 in the exploratory, validation, and independent validation sets. The AUC of Index-1 was superior to that of CA19-9 (0.856 vs. 0.649, p = 0.038) for detecting T1 tumors. miR-665, a component of Index-1, was expressed in human pancreatic cancer cells, and its transfection inhibited cell growth. Conclusions: The serum miRNA signature Index-1 is useful for detecting PBca and could facilitate the early diagnosis of PBca. Histological chorioamnionitis (HCA) is a form of maternal immune activation (MIA) linked to an increased risk of neurodevelopmental disorders in offspring. Our previous study identified neurodevelopmental impairments in an MIA mouse model mimicking HCA. Thus, this study investigated the role of CD11c+ microglia, key contributors to myelination through IGF-1 production, in this pathology. In the mouse model, the CD11c+ microglial population was significantly lower in the MIA group than in the control group on postnatal day 3 (PN3d). Furthermore, myelination-related protein levels significantly decreased in the MIA group at PN8d. In humans, preterm infants with HCA exhibited higher IL-6 and IL-17A cord-serum levels and lower IGF-1 levels than those without HCA, followed by a higher incidence of delayed myelination on magnetic resonance imaging at the term-equivalent age. In silico analysis revealed that the transient induction of CD11c+ microglia during early development occurred similarly in mice and humans. Notably, a lack of high CD11c+ microglial population has been observed in children with neurodevelopmental disorders. This study reports impaired induction of CD11c+ microglia during postnatal development in a mouse model of MIA associated with delayed myelination. Coral disease outbreaks have increased in frequency and extent worldwide since the 1970s, coinciding with the rapid increase in ocean warming. Summer and winter temperature-based metrics have proven effective in predicting coral disease outbreaks in seasonal coral reef regions. However, their utility is unknown in non-seasonal coral reef areas. Here, a new methodology, independent of seasonal patterns, is developed for application in both seasonal and non-seasonal coral reef regions. Percentile-based metric thresholds were defined from seasonal equivalents in the Great Barrier Reef (GBR) and tested in seasonal and non-seasonal coral reef regions of the tropical Pacific Ocean. Between new and existing methodologies, median differences of 0.00 °C (thresholds) and 0.00 °C-weeks (metrics) for Hot Snap and Cold Snap; and 0.01 °C (threshold) and −0.17 °C-weeks (metric) for Winter Condition were observed among reef pixels of the GBR. The new methodology shows strong consistency with the existing tools used for seasonal regions (e.g., R2 = 0.811–0.903; GBR case studies). Comparisons of the new metrics with disease observations were constrained by the limited availability of disease data; however, the comparisons undertaken suggest predictive capability in non-seasonal regions. To establish robust correlations, further direct comparisons of the new metrics with disease data across various non-seasonal regions and timeframes are essential. Chemoradiotherapy (CRT) followed by durvalumab is standard for unresectable locally advanced non-small-cell lung cancer (LA-NSCLC). This study assesses how CRT alters the T-cell receptor (TCR) repertoire in CD8 + PD-1 + T-cells and its impact on clinical outcomes. This prospective study, conducted from November 2019 to May 2021 at three institutions in Japan, evaluated the diversity of TCR repertoire (DE50) in PD-1 + CD8 + T-cells and CD8 + T-cell phenotypes in peripheral blood before and after CRT. Forty patients treated with CRT were included. The diversity and usage of TCR beta variable chains (TRBV) and 14 junctional chains (TRBJ) were significantly and positively correlated before and after CRT. Regarding the DE50, the progression-free survival (PFS) of patients with DE50High before CRT was significantly greater than that of those with DE50Low (NR vs. NR months, HR 0.17, p = 0.01). Background: Perivascular epithelioid cell tumors (PEComas) rarely appear in the head and neck region. This case report describes two transcription factor E3 (TFE3)-rearranged PEComa cases, consisting of one in the orbit and one in the nasal cavity. Case presentation: Both cases demonstrated sheet-like or focal nested architecture and comprised epithelioid cells with abundant clear to eosinophilic cytoplasm and vascular stroma. The first case exhibited partial pleomorphism, a small necrosis area, and slightly increased mitosis and was classified as malignant. The second case demonstrated mild atypia and no mitosis or necrosis and was categorized as benign. The nasal tumor was initially considered a TFE3-rearranged renal cell carcinoma metastasis. However, a subsequent renal tumor biopsy revealed angiomyolipoma. The RNA sequence revealed ZC3H4::TFE3 and PRCC::TFE3 fusions in the first and second cases, respectively. Conclusion: The fusion partner gene ZC3H4 is uncommon, and this is the third reported PEComa case. The fusion partner gene PRCC is often reported in TFE3-rearranged renal cell carcinoma, and this PEComa case is the second reported in the head and neck region. Purpose: The impact of body-cavity depth on open (OLR) and laparoscopic liver resection (LLR) of segment 7 remains unclear. Therefore, we investigated the influence of body-cavity depth at the upper-right portion of the abdomen on LLR and OLR of segment 7. Methods: In total, 101 patients who underwent segment-7 liver resection over 2010–2023 were included. Body-cavity depth was measured from the abdominal-wall surface to the deepest site on the right side of the liver. Patients were categorized into shallow (< 18.4 cm) and deep (≥ 18.4 cm) populations based on median body-cavity depth. We compared surgical outcomes between OLR and LLR in shallow and deep populations after propensity-score adjustments. Results: In OLR and LLR groups, 27 and 22 patients in the shallow population, respectively, and 26 and 26 patients were included in the deep population, respectively, were included. The OLR group in the deep population had significantly greater blood loss than the corresponding LLR group (difference: 144 mL, 95% confidence interval (CI): [50, 238], P = 0.004). Other surgical outcomes, including operative time, were similar between groups. In the shallow population, the OLR group had significantly shorter operative time (difference: − 54 mL, 95% CI: [–101, − 6], P = 0.028) and similar blood loss than the LLR group. Conclusions: For segment-7 liver resection, LLR is likely favorable for patients with a deep body cavity, with similar operative time and lower blood loss compared to OLR. Body-cavity depth could be a useful indicator for determining the suitable surgical approach for segment-7 liver resection. [ABSTRACT FROM AUTHOR]
