Abstract

Single-Cell Sequencing (SCS) technology plays an important role in the field of Mesenchymal Stem Cells (MSCs) research. This paper comprehensively describes the application of SCS technology in the field of MSCs research, including (1) SCS enables more precise MSCs characterization and biomarker definition. (2) SCS reveals the prevalent gene expression heterogeneity among different subclusters within MSCs, which contributes to a more comprehensive understanding of MSCs function and diversity in developmental, regenerative, and pathological contexts. (3) SCS provides insights into the dynamic transcriptional changes experienced by MSCs during differentiation and the complex web of important signaling pathways and regulatory factors controlling key processes within MSCs, including proliferation, differentiation and regulation, and interactions mechanisms. (4) The analytical methods underpinning SCS data are rapidly evolving and converging with the field of histological research to systematically deconstruct the functions and mechanisms of MSCs. This review provides new perspectives for unraveling the biological properties, heterogeneity, differentiation potential, biological functions, and clinical potential of MSCs at the single-cell level. Abstract

Introduction: Stem cells are definite units competent of both self-renewal and differentiation into numerous cells. Embryonic and postnatal tissues are the major sources of stem cells. The aim of the study was to assess the knowledge and level of information about stem cells research and therapeutic potential among Medical and Dental postgraduate students of Sumandeep Vidyapeeth, Vadodara. Materials and Methods: The Medical and Dental postgraduate students of Sumandeep Vidyapeeth, Vadodara, were selected. The questionnaire form was taken to them by the principal investigator personally and explained about the study and asked to fill the questionnaire after obtaining consent. The collected questionnaire was excluded if more than 30% of questions were not filled. Results: A total of 435 medical and dental postgraduate students of Sumandeep Vidyapeeth, Vadodara, were included in the study. 109 dental postgraduates (26.58%), and 301 were medical postgraduates. All participants had a basic knowledge of stem cells (100%), but only a few had specific knowledge (3.90%). Only a few agreed to human embryo research and considered it ethical (0.73%). All agreed to conserve their child's umbilical cord for the child's therapeutic purposes. Conclusion: We can conclude that the knowledge and level of information about stem cells exhibited by most of the participants were minimal. However, all participants expressed a genuine desire to enhance their knowledge regarding stem cells. There is a need for Continuing Medical Education/ Continuing Dental Education (CME/CDE) workshops as well as conferences on this subject to upgrade the knowledge of participants and the community as a whole. The human body possesses an endogenous regeneration system based on stem cells, which may be found in practically every tissue type. They are classified as embryonic stem cells (ESCs) or nonembryonic stem cells (NESCs). Despite its enormous promise, the use of ESCs is presently limited because of ethical and scientific issues. Stem cells have the potential to improve healthcare by using and boosting the body's inherent regenerative capabilities. Although the stem cells offer an enormous promise for tissue regeneration and repair, much more about their biology, administration, and safety must be studied before they may be employed therapeutically. Stem cells and their derivatives will have enormous medical promise in the future. A new study on mesenchymal stem cells from dental tissues has been conducted in Mexico City. The research found that these dental tissue mesenchymal stem cells (DT-MSCs) have similar immunosuppressive effects to bone marrow-derived mesenchymal stem cells (BM-MSCs), but express more CD39 and generate more adenosine (ADO) from ATP. The concentrations of ADO produced by DT-MSCs inhibited the proliferation of CD3+ T cells and promoted the generation of Tregs and Th17+CD39+ lymphocytes. BACKGROUND: Meniscus cartilage injury is a most common injury of knee joint. In the past decade, stem cell based therapies have been widely applied, and the related studies have been increased rapidly. Pluripotent stem cells have been shown to exist in the meniscus, which may play a key role in the healing of meniscus. OBJECTIVE: To determine the source and culture conditions of stem cells related to tissue-engineered meniscus, and to understand the current development status of tissue- engineered meniscus. METHODS: PubMed, Embase, EBSCO, Cochrane library, and Clinical library databases were retrieved for relevant articles published between 2009 and 2019. The articles concerning tissue-engineered meniscus and selection of seed cells were selected, and finally 64 eligible articles were included for analysis. RESULTS AND CONCLUSION: Stem cells have been extensively used as seed cells in tissue-engineered meniscus due to its high proliferation ability and multipotent property. These cells can effectively repair the meniscus in animals, but the clinical outcomes still remain unclear. Therefore, there is a certain distance from basic research to clinical application. Abstract

The advancement in human stem cell research has promised a viable alternative treatment for a range of 'incurable diseases' such as neurological diseases. To date, several studies have documented substantial evidences on the therapeutic properties of stem cells in promoting repair in different diseases including common neurological disorders i.e. ischaemic stroke and spinal cord injury. However, the progress of stem cell research has been surrounded by ethical issues which largely due to the usage of human embryos as one of the sources. These embryonic stem cells which originally derived from human embryo of aborted foetus or already existing human embryonic stem cells (hESCs) lines, has sparked an intense moral and religious argument among people of various faith, including Muslim community. From the therapeutic point of view, amongst the currently available stem cells, hESCs show the greatest potential for the broadest range of cell replacement therapies and are regarded as the most commercially viable. This review focuses on the major ethical issues, particularly to Muslim community, related to human embryonic stem cells research with special emphasis on the moral status of the embryo and the beginning of life according to the Islamic ethics and rulings. In this paper, we also discuss some ethical positions towards embryonic stem cell research in the Islamic world, including official regulations existing in some Muslim countries. We examine the justification and the necessity on the usage of hESCs following the newly discovered Induced Pluripotent Stem Cells (IPSCs) in the laboratory. In practice, stem-cell and embryo research have encountered little resistance in Arab and Islamic countries, and it is already in progress in Egypt, Iran, Turkey, Malaysia, and elsewhere. The Islamic scientific community is largely free of religious inhibitions that jeopardize attempts of scientists in Christian-oriented countries, for example, to engage in embryo and stem-cell research. The reason lies with the nature of Islamic medical ethics. According to this ethical system, obtaining knowledge is a human's duty, and an act of faith. Also, science is revealed by God to humans to serve humankind, and human life must be saved by all possible means. According to Islamic theology, "humanity"/the soul, is inspired into the fetus only at 120 days of pregnancy. Therefore, the use of fetuses younger than 120 days in research, and especially if sick or disabled and with a short life expectancy, if born alive at all, is not prohibited, provided that certain stipulations are followed. The use of superfluous zygotes in research is also encouraged, instead of their simply being destroyed. The use of fetal bovine serum hinders obtaining reproducible experimental results and should also be removed in hormone and growth factor studies. In particular hormones found in FBS act globally on cancer cell physiology and influence transcriptome and metabolome. The aim of our study was to develop a renal carcinoma serum free culture model optimized for (embryonal) renal cells in order to select the best study model for downstream auto-, para- or endocrine research. Secondary aim was to verify renal carcinoma stem cell culture for this application. In the study, we have cultured renal cell carcinoma primary tumour cell line (786-0) as well as human kidney cancer stem cells in standard 2D monolayer cultures in Roswell Park Memorial Institute Medium or Dulbecco’s Modified Eagle’s Medium and Complete Human Kidney Cancer Stem Cell Medium, respectively. Serum-free, animal-component free Human Embryonic Kidney 293 media were tested. Our results revealed that xeno-free embryonal renal cells optimized culture media provide a useful tool in RCC cancer biology research and at the same time enable effective growth of RCC. Abstract

Microfabrication and microfluidics contribute to the research of cellular functions of cells and their interaction with their environment. Previously, it has been shown that microfluidics can contribute to the isolation, selection, characterization and migration of cells. This review aims to provide stem cell researchers with a toolkit of microtechnology (mT) instruments for elucidating complex stem cells functions which are challenging to decipher with traditional assays and animal models. These microdevices are able to investigate about the differentiation and niche interaction, stem cells transcriptomics, therapeutic functions and the capture of their secreted microvesicles. In conclusion, microtechnology will allow a more realistic assessment of stem cells properties, driving and accelerating the translation of regenerative medicine approaches to the clinic. Researchers from the Institute of Genetics and Animal Biotechnology of the Polish Academy of Sciences have conducted a study on the role of E-cadherin in the specification of embryonic stem cells (ESCs) in the embryo environment. The study aimed to determine the location of ESCs lacking E-cadherin in the blastocyst, specifically in the trophoblast (TE), epiblast (Epi), or primitive endoderm (PrE) cell lines. The researchers found that these cells tended to locate in the TE and less frequently in the Epi, but never in the PrE. Abstract

The concept of "stemness" incorporates the molecular mechanisms that regulate the unlimited self-regenerative potential typical of undifferentiated primitive cells. These cells possess the unique ability to navigate the cell cycle, transitioning in and out of the quiescent G0 phase, and hold the capacity to generate diverse cell phenotypes. Stem cells, as undifferentiated precursors endow with extraordinary regenerative capabilities, exhibit a heterogeneous and tissue-specific distribution throughout the human body. The identification and characterization of distinct stem cell populations across various tissues have revolutionized our understanding of tissue homeostasis and regeneration. From the hematopoietic to the nervous and musculoskeletal systems, the presence of tissue-specific stem cells underlines the complex adaptability of multicellular organisms. Recent investigations have revealed a diverse cohort of non-hematopoietic stem cells (non-HSC), primarily within bone marrow and other stromal tissue, alongside established hematopoietic stem cells (HSC). Among these non-HSC, a rare subset exhibits pluripotent characteristics. In vitro and in vivo studies have demonstrated the remarkable differentiation potential of these putative stem cells, known by various names including multipotent adult progenitor cells (MAPC), marrow-isolated adult multilineage inducible cells (MIAMI), small blood stem cells (SBSC), very small embryonic-like stem cells (VSELs), and multilineage differentiating stress enduring cells (MUSE). The diverse nomenclatures assigned to these primitive stem cell populations may arise from different origins or varied experimental methodologies. This review aims to present a comprehensive comparison of various subpopulations of multipotent/pluripotent stem cells derived from stromal tissues. By analysing isolation techniques and surface marker expression associated with these populations, we aim to delineate the similarities and distinctions among stromal tissue-derived stem cells. Understanding the nuances of these tissue-specific stem cells is critical for unlocking their therapeutic potential and advancing regenerative medicine. The future of stem cells research should prioritize the standardization of methodologies and collaborative investigations in shared laboratory environments. Abstract

Lung cancer is the most common cancer worldwide, accounting for 1.8 million new cases and 1.6 million deaths in 2012. Non-small cell lung cancer (NSCLC), which is one of two types of lung cancer, accounts for 85-90% of all lung cancers. Despite advances in therapy, lung cancer still remains a leading cause of death. Cancer relapse and dissemination after treatment indicates the existence of a niche of cancer cells that are not fully eradicated by current therapies. These chemoresistant populations of cancer cells are called cancer stem cells (CSCs) because they possess the self-renewal and differentiation capabilities similar to those of normal stem cells. Targeting the niche of CSCs in combination with chemotherapy might provide a promising strategy to eradicate these cells.
