Coffee intake is increasingly recognized as a life-style factor associated with the preservation of health, but there is still a debate on the relative effects of caffeinated and decaffeinated coffee. We now tested how the regular drinking of caffeinated and decaffeinated coffee for 3 weeks impacted on the behavior of male and female adult mice. Males drinking caffeinated coffee displayed statistically significant lower weight gain, increased sensorimotor coordination, greater motivation in the splash test, more struggling in the forced swimming test, faster onset of nest building, more marble burying and greater sociability. Females drinking caffeinated coffee displayed statistically significant increased hierarchy fighting, greater self-care and motivation in the splash test and faster onset of nest building. A post-hoc two-way ANOVA revealed sex-differences in the effects of caffeinated coffee (p values for interaction between the effect of caffeinated coffee and sex) on the hierarchy in the tube test (p = 0.044; dominance), in the time socializing (p = 0.044) and in the latency to grooming (p = 0.048; selfcare), but not in the marble burying test (p = 0.089). Intake of decaffeinated coffee was devoid of effects in males and females. Since caffeine targets adenosine receptors, we verified that caffeinated but not decaffeinated coffee intake increased the density of adenosine A1 receptors (A1R) and increased A1R-mediated tonic inhibition of synaptic transmission in the dorsolateral striatum and ventral but not dorsal hippocampus, the effects being more evident in the ventral hippocampus of females and striatum of males. In contrast, caffeinated and decaffeinated coffee both ameliorated the antioxidant status in the frontal cortex. It is concluded that caffeinated coffee increases A1R-mediated inhibition in mood-related areas bolstering wellbeing of both males and females, with increased sociability in males and hierarchy struggling and self-care in females. Cisplatin (CP) is a chemotherapy drug widely prescribed to treat various neoplasms. Although fundamental for the therapeutic action of the drug, its cytotoxic mechanisms trigger adverse effects in several tissues, such as the kidney, liver, and heart, which limit its clinical use. In this sense, studies point to an essential role of damage to nuclear and mitochondrial DNA associated with oxidative stress, inflammation, and apoptosis in the pathophysiology of tissue injuries. Due to the limitation of effective preventive and therapeutic measures against CP-induced toxicity, new strategies with potential cytoprotective effects have been studied. Therefore, this article is timely in reviewing the characteristics and main molecular mechanisms common to renal, hepatic, and cardiac toxicity previously described, in addition to addressing the main validated strategies for the current management of these adverse events in clinical practice. We also handle the main promising antioxidant substances recently presented in the literature to encourage the development of new research that consolidates their potential preventive and therapeutic effects against CP-induced cytotoxicity. Background: Despite 18 years since health surveillance regulations were promulgated in Brazil to govern Long-Term Care Institutions for Older Adults (LTCIs), many institutions fail to comply with the Differentiated Regime for Public Procurement (Resolution No. 502/2021) due to structural and operational conditions. This study aimed to investigate Brazilian LTCI managers' understanding of challenges that significantly impact institutional operation and gather suggestions for enhancing RDC No. 502/21. Methods: A cross-sectional, exploratory, and qualitative study was conducted, involving 90 managers or technical supervisors from Brazilian LTCIs. Data were collected using a self-administered Google Forms instrument and analyzed through Thematic Analysis based on the Organizing for Quality (OQ) framework. Results: The most impactful challenges for LTCIs were healthcare, financing, human resources, relationship with oversight bodies, and family members. Discussion: Proposed improvements for RDC No. 502/21 included enhanced professional training, infrastructure revision, increased financial support from the state, realistic oversight/regulations, and tailored monitoring approaches. Conclusion: LTCIs in Brazil face numerous challenges, and the suggested improvements aim to adapt regulations to institutional realities. Synopsis: This is a systematic review and meta-analysis comparing surgical excision with percutaneous ultrasound-guided vacuum-assisted excision (USVAE) for the treatment of benign phyllodes tumor (PT) using local recurrence (LR) as the endpoint. Objective: To determine the frequency of local recurrence (LR) of benign phyllodes tumor (PT) after ultrasound-guided vacuum-assisted excision (USVAE) compared to the frequency of LR after surgical excision. Method: A systematic review and meta-analysis [following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) standard] was conducted by comparing LR in women older than 18 years treated for benign PT by US-VAE compared with local surgical excision with at least 12 months of follow-up. Studies were retrieved from PubMed, Scopus, Web of Science, and Embase. The pooled effect measure used was the odds ratio (OR) of recurrence. Results: Five comparative prospective or retrospective observational studies published between January 1, 1992, and January 10, 2022, comparing surgical excision with percutaneous US-VAE for LR of benign PT met the selection criteria. Four were retrospective observational cohorts, and one was a prospective observational cohort. A total of 778 women were followed up. Of them, 439 (56.4%) underwent local surgical excision, and 339 (43.6%) patients had US-VAE. The median age of patients in the five studies ranged from 33.7 to 39 years; the median size ranged from 1.5 cm to 3.0 cm, and the median follow-up ranged from 12 months to 46.6 months. The needle gauge ranged from 7G to 11G. LR rates were not statically significant between US-VAE and surgical excision (41 of 339 versus 34 of 439; OR 1.3; p = 0.29). Conclusion: This meta-analysis suggests that using US-VAE for the removal of benign PT does not increase local regional recurrence and is a safe minimally invasive therapeutic option. This study aimed to test the reproducibility of the 3-min all-out effort applied using shuttle running and compare its values to aerobic parameters. On the first day, 14 futsal players underwent an exhaustive test to determine the maximal incremental speed (MIS) and anaerobic threshold (AnT). On the second day, the participants performed the 3-min all-out effort (n=14), which was repeated after 48 h (third day) to test its reproducibility (n=11). Peak oxygen consumption (V̇ O2PEAK) and peak blood lactate concentrations ([La-]) were determined from 3-min all-out efforts performed through a 20-m shuttle run on the official court. The distance covered, mean speed, and critical speed (CS) during the 3-min all-out presented direct relationships with aerobic parameters determined through the incremental test (r>0.62). The distance covered above CS (D') presented a direct relationship with peak lactate concentrations induced by a 3-min all-out effort (r=0.81). Despite the acceptable levels of reproducibility observed for most of the 3-min all-out variables, the minimal detectable change for D' was high (72%). Our results demonstrated the potential use of mean speed to evaluate aerobic fitness. However, the applicability of the 3-min all-out shuttle run test to monitor training adaptations should be avoided, at least in nonexperienced athletes. [ABSTRACT FROM AUTHOR]

Introduction and purpose: Basilar artery occlusion (BAO) is still one of the most devastating neurological conditions associated with high morbidity and mortality. In the present study, we aimed to assess the role of posterior circulation collaterals as predictors of outcome in the BASICS trial and to compare two grading systems (BATMAN score and PC-CS) in terms of prognostic value. Methods: We performed a sub-analysis of the BASICS trial. Baseline clinical and imaging variables were analyzed. For the imaging analysis, baseline CT and CTA were analyzed by a central core lab. Only those patients with good or moderate quality of baseline CTA and with confirmed BAO were included. Multivariable binary logistic regression analysis was used to test the independent association of clinical and imaging characteristics with a favorable outcome at 3 months (defined as a modified Rankin Score of =3). ROC curve analysis was used to assess and compare accuracy between the two collateral grading systems. Results: The mean age was 67.0 (±12.5) years, 196 (65.3%) patients were males and the median NIHSS was 21.5 (IQR 11-35). Median NCCT pc-ASPECTS was 10 (IQR10-10) and median collateral scores for BATMAN and PC-CS were 8 (IQR 7-9) and 7 (IQR 6-8) respectively. Collateral scores were associated with favorable outcome at 3 months for both BATMAN and PC-CS but only with a modest accuracy on ROC curve analysis (AUC 0.62, 95% CI [0.55-0.69] and 0.67, 95% CI [0.60-0.74] respectively). Age (OR 0.97, 95% CI [0.95-1.00]), NIHSS (OR 0.91, 95% CI [0.89-0.94]) and collateral score (PC-CS - OR 1.2495% CI [1.02-1.51]) were independently associated with clinical outcome. Conclusion: The two collateral grading systems presented modest prognostic accuracy. Only the PC-CS was independently associated with a favorable outcome at 3 months. [ABSTRACT FROM AUTHOR]

Spermatozoa and the male reproductive system (MRS) present great variability, featuring morphological characters that could be used in the taxonomy of different insect groups. Despite their importance, such data are scarce for mosquitoes, and their potential application in taxonomy has been neglected. In this study, we describe the morphology of the MRS and spermatozoa of the mosquito Lutzia bigoti Bellardi, 1862. The MRS consists of a pair of testes, deferent ducts, seminal vesicles, accessory glands, and an ejaculatory duct. The testes have only a single well-compacted follicle. The deferent ducts originate at the base of each testis, with the first portion resembling a goblet, and present an enlargement that forms the seminal vesicles in the posterior region. Subsequently, the ducts merge with the respective ducts of the accessory glands and open into the ejaculatory duct. The spermatozoa of L. bigoti are characterized as filiform and long, measuring around 220 µm, providing a potential distinguishing feature of this species. We hypothesize that the spiral organization of the testes may confer advantages in spermatozoa production. Similar to other mosquitoes, the MRS organization plan of L. bigoti conforms to type B. In addition, we also suggest that Culicinae species possibly share juxtaposed vesicles. This work paved the way for future analyses exploring of reproductive biology of Lutzia, including ultrastructure examinations and taxonomic investigations in the Culicidae family. According to The World Alzheimer Report 2023 by Alzheimer's Disease International (ADI) estimates that 33 to 38.5 million people worldwide suffer from Alzheimer's Disease (AD). A crucial hallmark associated with this disease is associated with the deficiency of the brain neurotransmitter acetylcholine, due to an affected acetylcholinesterase (AChE) activity. Marine organisms synthesize several classes of compounds, some of which exhibit significant AChE inhibition, such as petrosamine, a coloured pyridoacridine alkaloid. The aim of this work was to characterize the activity of petrosamine isolated for the first time from a Brazilian marine sponge, using two neurotoxicity models with aluminium chloride, as exposure to aluminium is associated with the development of neurodegenerative diseases. The in vitro model was based in a neuroblastoma cell line and the in vivo model exploited the potential of zebrafish (Danio rerio) embryos in mimicking hallmarks of AD. To our knowledge, this is the first report on petrosamine's activity over these parameters, either in vitro or in vivo, in order to characterize its full potential for tackling neurotoxicity. Tissue-resident memory T cells (TRM cells) are vital for the promotion of barrier immunity. The lung, a tissue constantly exposed to foreign pathogenic or nonpathogenic antigens, is not devoid of these cells. Lung TRM cells have been considered major players in either the protection against respiratory viral infections or the pathogenesis of lung allergies. Establishment of lung TRM cells rely on intrinsic and extrinsic factors. Among the extrinsic regulators of lung TRM cells, the magnitude of the impact of factors such as the route of antigen entry or the antigen natural tropism for the lung is not entirely clear. In this perspective, we provide a summary of the literature covering this subject and present some preliminary results on this potential dichotomy between antigen location versus antigen type. Background: Pressure injuries (PIs) are a major problem for healthcare providers, impacting both care costs and patients' quality of life, although they are predominately preventable. These injuries are especially present in Intensive Care Units (ICUs) as a result of the severity of the clinical conditions of patients in this unit. Aim: To develop a simplified version of the Braden scale by removing two of the most subjective subscores—Nutrition and Sensory Perception—in an attempt to reduce the chance of errors by the nursing team during the application of the scale. Study Design: A cross‐sectional study was conducted on data collected from patients admitted to the ICU of a private Brazilian tertiary hospital. The resulting data consisted of 5194 patients, 6353 hospital admissions, and 6974 ICU stays. The overall prevalence of PI was 1.09%. Results: The T‐test showed that both the Braden and the simplified Braden scores were significantly different between patients with and without PI (p <.001). Patients who developed PIs scored lower than those who did not. The area under the Receiver Operating Characteristic curve of the Braden Scale was 74.21% (95% CI: 68.61%–79.8%) and of the simplified scale was 72.54% (95% CI: 66.87%–78.22%). The Positive Predictive Value of the Braden Scale was 3.17% when interpolated at the same sensitivity as the simplified scale (47.37%), which achieved 3.26%. Conclusions: By removing two of the six subscores of the Braden scale we propose a new tool for identifying patients at risk of developing PI in a more objective and fast way. Our results show that classification performance had little negative impact. Background: Noncontrast CT (NCCT) is used to evaluate for intracerebral hemorrhage (ICH) and ischemia in acute ischemic stroke (AIS). Large vessel occlusions (LVOs) are a major cause of AIS, but challenging to detect on NCCT. Aims: The purpose of this study is to evaluate an AI software called RAPID NCCT Stroke (RAPID, iSchemaView, Menlo Park, CA) for ICH and LVO detection compared to expert readers. Methods: In this IRB approved retrospective, multicenter study, stand-alone performance of the software was assessed based on the consensus of 3 neuroradiologists and sensitivity and specificity were determined. The platform's performance was then compared to interpretation by readers comprised of eight general radiologists (GR) and three neuroradiologists (NR) in detecting ICH and hyperdense vessel sign (HVS) indicating LVO. Results: A total of 244 cases were included. Of the 244, 115 were LVOs and 26 were ICHs. One hundred three cases did not have LVO nor ICH. Stand-alone performance of the software demonstrated sensitivities and specificities of 96.2 and 99.5% for ICH and 63.5 and 95.1% for LVO detection. Compared to all 11 readers and eight GR readers only respectively, the software demonstrated superiority, achieving significantly higher sensitivities (63.5% versus 43.6%, p < 0.0001 and 63.5% versus 40.9%, p = 0.001). Conclusion: The RAPID NCCT Stroke platform demonstrates superior performance to radiologists for detecting LVO from a NCCT. Use of this software platform could lead to earlier LVO detection and expedited transfer of these patients to a thrombectomy capable center. [ABSTRACT FROM AUTHOR]

With the advancements in utilizing Artificial Intelligence (AI) in embedded safety-critical systems based on Field-Programmable Gate Arrays (FPGAs), assuring that these systems meet their safety requirements is of paramount importance for their revenue service. Based on this context, this paper has two main objectives. The first of them is to present the Safety ArtISt method, developed by the authors to guide the lifecycle of AI-based safety-critical systems, and emphasize its FPGA-oriented tasks and recommended practice towards safety assurance. The second one is to illustrate the application of Safety ArtISt with an FPGA-based braking control system for autonomous vehicles relying on explainable AI generated with High-Level Synthesis. The results indicate that Safety ArtISt played four main roles in the safety lifecycle of AI-based systems for FPGAs. Firstly, it provided guidance in identifying the safety-critical role of activities such as sensitivity analyses for numeric representation and FPGA dimensioning to achieve safety. Furthermore, it allowed building qualitative and quantitative safety arguments from analyses and physical experimentation with actual FPGAs. It also allowed the early detection of safety issues—thus reducing project costs—and, ultimately, it uncovered relevant challenges not discussed in detail when designing safety-critical, explainable AI for FPGAs. Working conditions of musicians are marked by several specific characteristics that differ the occupation from most others in the labor market. For instance, non-pecuniary aspects of the profession may have a greater influence on the individuals who pursue musical activities than in most other occupations. Moreover, musicians are more susceptible to uncertainties in the short run and regarding the duration of their careers. Besides, informality and underemployment rates tend to be higher in the music industry when compared to the rest of the labor market. This paper aims to better understand musician's labor market participation using primary data. We conducted an online survey between February and December 2020, gathering data about working conditions, in particular job satisfaction and turnover, with all the musicians from Belo Horizonte who were registered and not were registered in musician's association. Belo Horizonte is known as a unique city and a highly heterogeneous environment for the development of the music industry. Thus, using work satisfaction indicators and Kaplan–Meier survival model, controlled by personal and work-position variables, this study highlights that most musicians were neither satisfied nor dissatisfied with their work. However, satisfaction was positively correlated with the following characteristics: being protected by social security, having a music career as main occupation, and holding a degree in music. These traits were also important for musicians to remain in the occupation throughout 2020, an atypical period due to the COVID-19 pandemic. [ABSTRACT FROM AUTHOR]

The intracerebroventricular (icv) injection of amyloid peptides (Aβ) models Alzheimer's disease (AD) in mice, as typified by the onset within 15 days of deficits of memory and of hippocampal long-term potentiation (LTP) that are prevented by the blockade of adenosine A2A receptors (A2AR). Since A2AR overfunction is sufficient to trigger memory deficits, we tested if A2AR were upregulated in hippocampal synapses before the onset of memory deficits to support the hypothesis that A2AR overfunction could be a trigger of AD. Six to eight days after Aβ-icv injection, mice displayed no alterations of hippocampal dependent memory; however, they presented an increased excitability of hippocampal synapses, a slight increase in LTP magnitude in Schaffer fiber-CA1 pyramid synapses and an increased density of A2AR in hippocampal synapses. A2AR blockade with SCH58261 (50 nM) normalized excitability and LTP in hippocampal slices from mice sacrificed 7–8 days after Aβ-icv injection. Fifteen days after Aβ-icv injection, mice displayed evident deficits of hippocampal-dependent memory deterioration, with reduced hippocampal CA1 LTP but no hyperexcitability and a sustained increase in synaptic A2AR, which blockade restored LTP magnitude. This shows that the upregulation of synaptic A2AR precedes the onset of deterioration of memory and of hippocampal synaptic plasticity, supporting the hypothesis that the overfunction of synaptic A2AR could be a trigger of memory deterioration in AD. Simple Summary: Chagas disease is a serious infection caused by an intracellular parasite and transmitted primarily through an infected insect. Despite available treatments, the disease continues to cause countless deaths around the world due to the ineffectiveness of drugs. Medicinal plants have been used as an alternative and effective treatment against various diseases. In this work, we verified the antioxidant properties of the extract and fractions obtained from the plant species Terminalia catappa and its action against the parasite responsible for Chagas disease, Trypanosoma cruzi. Initially, we observed that the ethyl acetate and aqueous fraction demonstrated antioxidant activity. In addition, the ethyl acetate fraction showed the best inhibitory activity against all cellular forms of the parasite (epimastigotes, trypomastigotes and intracellular amastigotes), and did not present toxicity to host cells. We also observed that the ethyl acetate fraction induced several morphological changes to the parasite, such as cytoplasmic disruption, cell disorganization, morphological variation and loss of integrity. In this sense, we conclude that the ethyl acetate fraction obtained from T. catappa leaves can be an effective alternative in the treatment and control of Chagas disease. Chagas disease is a severe infectious and parasitic disease caused by the protozoan Trypanosoma cruzi and considered a public health problem. Chemotherapeutics are still the main means of control and treatment of the disease, however with some limitations. As an alternative treatment, plants have been pointed out due to their proven pharmacological properties. Many studies carried out with Terminalia catappa have shown several biological activities, but its effect against T. cruzi is still unknown. The objective of this work is to evaluate the therapeutic potential of extracts and fractions obtained from T. catappa on the parasite T. cruzi, in addition to analyzing its antioxidant activity. T. catappa ethyl acetate fraction were produced and submitted the chemical characterization by Liquid Chromatography Coupled to Mass Spectrometry (LC-MS). From all T. catappa extracts and fractions evaluated, the ethyl acetate and the aqueous fraction displayed the best antioxidant activity by the 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging method (IC50 of 7.77 ± 1.61 and 5.26 ± 1.26 µg/mL respectively), and by ferric ion reducing (FRAP) method (687.61 ± 0.26 and 1009.32 ± 0.13 µM of Trolox equivalent/mg extract, respectively). The ethyl acetate fraction showed remarkable T. cruzi inhibitory activity with IC50 of 8.86 ± 1.13, 24.91 ± 1.15 and 85.01 ± 1.21 µg/mL against epimastigotes, trypomastigotes and intracellular amastigotes, respectively, and showed no cytotoxicity for Vero cells (CC50 > 1000 µg/mL). The treatment of epimastigotes with the ethyl acetate fraction led to drastic ultrastructural changes such as the loss of cytoplasm organelles, cell disorganization, nucleus damage and the loss of integrity of the parasite. This effect could be due to secondary compounds present in this extract, such as luteolin, kaempferol, quercetin, ellagic acid and derivatives. The ethyl acetate fraction obtained from T. catappa leaves can be an effective alternative in the treatment and control of Chagas disease, and material for further investigations. Simple Summary: Meeting the growing animal food demand while reducing greenhouse gas emissions (GGEs) remains one of the challenges for global livestock industries. Enteric methane emission (EME) is the main GGE source in dairy cattle systems. Therefore, evaluating the drivers of EME intensity (per animal product) and developing more accurate predictive models of EME may contribute to dairy cattle sustainability. In this study, we built a large and intercontinental experimental dataset to: (1) explain the effect of EME yield (g methane/kg diet intake) and feed conversion (kg diet intake/kg milk yield) on EME intensity (g methane/kg milk yield); (2) develop models from predicting EME (g/cow/day); and (3) compare the proposed models with 43 external models. Increasing the milk yield reduced EME intensity, and this effect was more due to the enhancement in feed conversion than EME yield. Our models predicted methane emissions better than most external models, with the exception of only two other models which had similar adequacy. Our findings confirm that the improvement in feed conversion should be prioritized for reducing methane emissions in dairy cattle systems. Enteric methane emission is the main source of greenhouse gas contribution from dairy cattle. Therefore, it is essential to evaluate drivers and develop more accurate predictive models for such emissions. In this study, we built a large and intercontinental experimental dataset to: (1) explain the effect of enteric methane emission yield (g methane/kg diet intake) and feed conversion (kg diet intake/kg milk yield) on enteric methane emission intensity (g methane/kg milk yield); (2) develop six models for predicting enteric methane emissions (g/cow/day) using animal, diet, and dry matter intake as inputs; and to (3) compare these 6 models with 43 models from the literature. Feed conversion contributed more to enteric methane emission (EME) intensity than EME yield. Increasing the milk yield reduced EME intensity, due more to feed conversion enhancement rather than EME yield. Improved productivity of dairy cows reduces emission intensity by enhancing feed conversion. Artisanal and small-scale gold mining (ASGM) is the largest source of anthropogenic Hg emissions on the planet. In addition, Hg-contaminated tailings are often reprocessed with sodium cyanide (NaCN) to extract the residual gold remaining in the material. This leads to the formation of mercury cyanide (Hg(CN)2) complexes, which are often discharged in untreated form into local drainages, leading to large amounts of free cyanide being released. However, data on mercury-cyanide interactions are scarce. In this study, we investigated the impact of cyanide and Hg bioavailability in zebrafish when added as Hg(CN)2. Different concentrations of Hg(CN)2 and NaCN were used, leading to an LC50 of 0.53 mg.L−1 for NaCN and 0.16 mg.L−1 for Hg(CN)2. Analyzing free cyanide concentrations in aquarium water, >40% dissociation was observed for NaCN and about 5% for Hg(CN)2. The accumulation of total Hg (THg) in the brain, gills, muscle and kidney was quantified. All fish exposed to Hg(CN)2 had higher THg levels than their controls and kidney was the tissue with higher Hg(CN)2 accumulation. Histological effects on the kidney and gills of both cyanides in D. rerio tissues were investigated, suggesting renal alterations in fish exposed to Hg(CN)2 and showing hyperplasia in the gills of animals exposed to NaCN and Hg(CN)2. Alzheimer's disease (AD), which predominantly affects women, involves at its onset a metabolic deregulation associated with a synaptic failure. Here, we performed a behavioral, neurophysiological and neurochemical characterization of 9-month-old female APPswe/PS1dE9 (APP/PS1) mice as a model of early AD. These animals showed learning and memory deficits in the Morris water maze, increased thigmotaxis and anxiety-like behavior and showed signs of fear generalization. Long-term potentiation (LTP) was decreased in the prefrontal cortex (PFC), but not in the CA1 hippocampus or amygdala. This was associated with a decreased density of sirtuin-1 in cerebrocortical synaptosomes and a decreased density of sirtuin-1 and sestrin-2 in total cerebrocortical extracts, without alterations of sirtuin-3 levels or of synaptic markers (syntaxin, synaptophysin, SNAP25, PSD95). However, activation of sirtuin-1 did not affect or recover PFC-LTP deficit in APP/PS1 female mice; instead, inhibition of sirtuin-1 increased PFC-LTP magnitude. It is concluded that mood and memory dysfunction in 9-month-old female APP/PS1 mice is associated with a parallel decrease in synaptic plasticity and in synaptic sirtuin-1 levels in the prefrontal cortex, although sirtiun1 activation failed to restore abnormal cortical plasticity. To determine survival and prognostic factors in children and adolescents living with HIV (CLWH) in Rio de Janeiro (RJ), Brazil, who developed lymphomas, we performed a retrospective and observational study of vertically infected CLWH aged from 0 to 20 incomplete years during1995 to 2018 at five reference centers for cancer and HIV/AIDS treatment. Of the 25 lymphomas, 19 were AIDS-defining malignancies (ADM) and 6 were non-AIDS-defining malignancies (NADM). The 5-year overall survival (OS) and 5-year event-free survival (EFS) probabilities were both 32.00% (95% CI = 13.72–50.23%), and the 5-year disease-free survival (DFS) probability was 53.30% (95% CI = 28.02–78.58%). In the multivariate Cox regression analysis, performance status 4 (PS 4) was considered a poor prognostic factor for OS (HR 4.85, 95% CI = 1.81–12.97, p = 0.002) and EFS (HR 4.95, 95% CI = 1.84–13.34, p = 0.002). For the DFS, higher CD4+ T-cell counts were considered a better prognostic factor (HR 0.86, 95% CI = 0.76–0.97, p = 0.017) in the multivariate Cox regression analysis. Extracellular ATP can be a danger signal, but its role in striatal circuits afflicted in Parkinson's disease (PD) is unclear and was now investigated. ATP was particularly released at high stimulation intensities from purified striatal nerve terminals of mice, which were endowed with different ATP-P2 receptors (P2R), although P2R antagonists did not alter corticostriatal transmission or plasticity. Instead, ATP was extracellularly catabolized into adenosine through CD73 to activate adenosine A2A receptors (A2AR) modulating corticostriatal long-term potentiation (LTP) in mice. In the presymptomatic phase of a 6-hydroxydopamine rat model of PD, ATP release from striatal nerve terminals was increased and was responsible for a greater impact of CD73 and A2AR on corticostriatal LTP. These observations identify increased ATP release and ATP-derived formation of extracellular adenosine bolstering A2AR activation as a key pathway responsible for abnormal synaptic plasticity in circuits involved in the onset of PD motor symptoms. The translation of these findings to humans prompts extending the use of A2AR antagonists from only co-adjuvants of motor control in Parkinsonian patients to neuroprotective drugs delaying the onset of motor symptoms. The main purpose of this paper is to analyze the efficiency of Clean Development Mechanism (CDM) projects implemented worldwide and understand their main characteristics, seeking to support investment decisions and expand the scientific understanding of the financial and environmental impact of these projects. To achieve this goal, a sample of 2352 CDM projects was selected from a United Nations Framework Convention on Climate Change (UNFCCC) database and analyzed first with a two‐stage data envelopment analysis (DEA) model that allowed the evaluation of the financial return and environmental efficiency of these projects. DEA results provide an efficiency ranking that was then analyzed with a classification tree (CHAID algorithm), revealing some main characteristics of the projects with higher efficiencies, like their sizes, locations, and type of CDM. CDMs are projects that demand a significant quantity of resources and effort to produce the expected outcomes, so it is crucial for public managers and investors to know the project profiles that generate the best financial and environmental results. This work aimed to develop chitosan/gelatin scaffolds loaded with ethanolic extract of Jatropha mollissima (EEJM) to evaluate the influence of its content on the properties of these structures. The scaffolds were prepared by freeze-drying, with different EEJM contents (0–10% (w/w)) and crosslinked with genipin (0.5% (w/w)). The EEJM were characterized through High Performance Liquid Chromatography coupled to a Diode Array Detector (HPLC-DAD), and the determination of three secondary metabolites contents was accomplished. The physical, chemical and biological properties of the scaffolds were investigated. From the HPLC-DAD, six main substances were evidenced, and from the quantification of the total concentration, the condensed tannins were the highest (431.68 ± 33.43 mg·g−1). Spectroscopy showed good mixing between the scaffolds' components. Adding and increasing the EEJM content did not significantly influence the properties of swelling and porosity, but did affect the biodegradation and average pore size. The enzymatic biodegradation test showed a maximum weight loss of 42.89 within 28 days and reinforced the efficiency of genipin in crosslinking chitosan-based materials. The addition of the extract promoted the average pore sizes at a range of 138.44–227.67 µm, which is compatible with those reported for skin regeneration. All of the scaffolds proved to be biocompatible for L929 cells, supporting their potential application as skin tissue engineering materials. [ABSTRACT FROM AUTHOR]

Adenosine receptors mainly control synaptic function, and excessive activation of adenosine receptors may worsen the onset of many neurological disorders. Accordingly, the regular intake of moderate doses of caffeine antagonizes adenosine receptors and affords robust neuroprotection. Although caffeine intake alters brain functional connectivity and multi-omics analyses indicate that caffeine intake modifies synaptic and metabolic processes, it is unclear how caffeine intake affects behavior, synaptic plasticity and its modulation by adenosine. We now report that male mice drinking caffeinated water (0.3 g/L) for 2 weeks were behaviorally indistinguishable (locomotion, mood, memory) from control mice (drinking water) and displayed superimposable synaptic plasticity (long-term potentiation) in different brain areas (hippocampus, prefrontal cortex, amygdala). Moreover, there was a general preservation of the efficiency of adenosine A1 and A2A receptors to control synaptic transmission and plasticity, although there was a tendency for lower levels of endogenous adenosine ensuring A1 receptor-mediated inhibition. In spite of similar behavioral and neurophysiological function, caffeine intake increased the energy charge and redox state of cortical synaptosomes. This increased metabolic competence likely involved a putative increase in the glycolytic rate in synapses and a prospective greater astrocyte–synapse lactate shuttling. It was concluded that caffeine intake does not trigger evident alterations of behavior or of synaptic plasticity but increases the metabolic competence of synapses, which might be related with the previously described better ability of animals consuming caffeine to cope with deleterious stimuli triggering brain dysfunction. In addition, it displays an acceptable safety profile, with most reported adverse reactions constituting rare occurrences. This study aimed to achieve bioactivity on the PEEK surface using piranha solution through a lower functionalization time. For this purpose, the functionalization occurred with piranha solution and 98% sulfuric acid in the proportions of 1:2, 1:1, and 2:1 at periods of 30, 60, and 90 s. The samples treated for longer times at higher concentrations registered the characteristic spectroscopy band associated with sulfonation. Additionally, both chemical treatments allowed the opening of the aromatic ring, increasing the number of functional groups available and making the surface more hydrophilic. The piranha solution treatments with higher concentrations and longer times promoted greater heterogeneity in the surface pores, which affected the roughness of untreated PEEK. Furthermore, the treatments induced calcium deposition on the surface during immersion in SBF fluid. In conclusion, the proposed chemical modifications using sulfuric acid SPEEK 90 and, especially, the piranha solution PEEK-PS 2:1-90, were demonstrated to be promising in promoting the rapid bioactivation of PEEK-based implants. The combined antiretroviral therapy (cART) changed the natural history of HIV infection. To determine the incidence and profile of these CLWH malignancies in Rio de Janeiro (RJ), Brazil, we conducted a retrospective and observational study of vertically infected CLWH, ranging from 0–20 incomplete years, from 1995 to 2018, at five reference centers. The study period was divided into three eras in accordance with the widespread use of cART in Brazil. 1306 patients were included. Of the 25 lymphomas found, 19 were AIDS-defining malignancies (ADM); 6 were non-AIDS-defining malignancies (NADM). The incidence rate (IR) of lymphoma developing was 1.70 per 1000 children-year (95% CI 1.09–2.50). ADM development IR decreased from 2.09–1.75–0.19 per 1000 children-year (p < 0.001) through cART eras. Cumulative Nelson–Aalen hazards of developing ADM over a 20-year period were 3.73% in the Early-cART era, 3.07% in the Mid-cART era, and 0.32% in the Late-cART era (p = 0.013). Summary: The use of mutation testing for mobile applications (apps for short) is still a challenge. Mobile apps are usually event‐driven and encompass graphical user interfaces (GUIs) and a complex execution environment. Then, they require mutant operators to describe specific apps faults, and the automation of the mutation process phases like execution and analysis of the mutants is not an easy task. To encourage research addressing such challenges, this paper presents results from a mapping study on mutation testing for mobile apps. Following a systematic plan, we found 16 primary studies that were analysed according to three aspects: (i) trends and statistics about the field; (ii) study characteristics such as focus, proposed operators and automated support for the mutation testing phases; and (iii) evaluation aspects. The great majority of studies (98%) have been published in the last 3 years. The most addressed language is Java, and Android is the only operating system considered. Mutant operators of GUI and configuration types are prevalent in a total of 138 operators found. Most studies implement a supporting tool, but few tools support mutant execution and analysis. The evaluation conducted by the studies includes apps mainly from the finance and utility domain. Nevertheless, there is a lack of benchmarks and more rigorous experiments. Future research should address other specific types of faults, languages, and operating systems. Interleukin-15 (IL-15) is often considered a central regulator of memory CD8+ T cells, based primarily on studies of recirculating subsets. However, recent work identified IL-15-independent CD8+ T cell memory populations, including tissue-resident memory CD8+ T cells (TRM) in some nonlymphoid tissues (NLTs). Whether this reflects the existence of IL-15-insensitive memory CD8+ T cells is unclear. We report that IL-15 complexes (IL-15c) stimulate rapid proliferation and expansion of both tissue-resident and circulating memory CD8+ T cell subsets across lymphoid and nonlymphoid tissues with varying magnitude by tissue and memory subset, in some sites correlating with differing levels of the IL-2Rß. This was conserved for memory CD8+ T cells recognizing distinct antigens and elicited by different pathogens. Following IL-15c-induced expansion, divided cells contracted to baseline numbers and only slowly returned to basal proliferation, suggesting a mechanism to transiently amplify memory populations. Through parabiosis, we showed that IL-15c drive local proliferation of TRM, with a degree of recruitment of circulating cells to some NLTs. Hence, irrespective of homeostatic IL-15 dependence, IL-15 sensitivity is a defining feature of memory CD8+ T cell populations, with therapeutic potential for expansion of TRM and other memory subsets in an antigen-agnostic and temporally controlled fashion. The objective of this study was to evaluate the performance, feed intake, carcass traits, and economic aspects of cattle fed different levels of supplementation during the growing phase (mineral supplementation—MS; low protein supplementation—PS, at 0.15% of body weight (BW); and a high protein-energy supplementation—PES at 0.40% of BW) and finished on the pasture with concentrate supplementation at 1.6% of BW. Ninety bulls were distributed in a completely randomized design and allocated into paddocks predominantly composed of Urochloa brizantha cv. Marandu grass. During the finishing phase, cattle were managed in paddocks of Urochloa decumbens grass. The average daily gain (ADG) and total weight gain during the growing phase were not different between animals fed PS (0.670 kg/day; 57.1 kg, respectively) and PES (0.730 kg/day; 62.4 kg, respectively), but both differed significantly from those supplemented with MS (0.540 kg/day; 45.1 kg). There was no difference between groups for average daily gain and total weight gain during the finishing phase (0.600 kg/day; 48.3 kg) and the same response was observed for carcass yield (55.18%, on average). However, bulls fed PES had higher hot carcass weight compared with those supplemented with MS and PS. Protein supplementation (0.15% of BW) and protein-energy supplementation (0.40% of BW) improved beef cattle performance during the growing phase. However, supplementation during the growing phase has a small impact on cattle performance in the finishing phase, although protein-energy supplementation at the growing phase improves the hot carcass weight and carcass traits during the finishing phase. The aim of this study was to document the impacts of an environmental education program with adolescents and older adults on attitudes about old age, and to investigate possible associations of these attitudes with age group, economic class, residing with grandparents, and grandparents' contribution to parenting. We investigated adolescents between 10 and 11, and between 14 and 15 years old divided into Experimental Group (EG), who participated in three intergenerational workshops at School, and Control Group (CG). The results indicated that younger adolescents had more positive attitudes than older ones and there was an association between a higher frequency of contact with grandparents and more positive attitudes in the Persona domain among 14- to 15-year-olds. There was no association between attitudes and the other variables investigated. At the end of the intervention, all EG adolescents showed more positive attitudes in the cognition domains, and older adolescents in the Agency domain. [ABSTRACT FROM AUTHOR]

Simple Summary: Lemongrass is an herb used in folk medicine for the treatment of hypertension, although its pharmacological potential has not yet been thoroughly studied. This paper provides the first comprehensive review on the anti-hypertensive potential of lemongrass, from in vitro to clinical studies. Even though the composition of lemongrass is dependent on its geographical origin, citral constitutes its major compound. Both citral and lemongrass display vasorelaxant activity ex vivo, promoting the secretion of endothelial vasodilators and the blockage of calcium channels in the vascular smooth muscle. Additionally, citral also displays a negative chronotrope effect. In animal models and in human subjects, lemongrass significantly decreases blood pressure, probably due to the combination of the above-mentioned effects together with diuretic activity. Future preclinical studies are necessary to identify other anti-hypertensive compounds/pathways, as well as to better characterize the safety profile of lemongrass. This paper reviews the anti-hypertensive effects of both lemongrass and its main compound citral in in vitro, ex vivo, preclinical and clinical studies. Lemongrass essential oil contains terpenes and their derivatives, whereas extracts contain different classes of polyphenols. Both citral and lemongrass display vasorelaxant activity ex vivo, acting by the promotion of endothelial nitric oxide/prostanoids secretion together with the blockage of calcium channels in the vascular smooth muscle. Citral also displays a negative chronotrope effect, probably due to a centrally mediated enhancement of parasympathetic activity. In both healthy and hypertensive animals, the acute administration of lemongrass results in a decrease in blood pressure, sometimes accompanied by a compensatory increase in heart rate. Similarly, in healthy and hypertensive human subjects, the consumption of lemongrass tea decreases blood pressure. Additionally, a weak/moderate diuretic activity has also been reported in animals and humans, although the mechanisms of action remain elusive. Future preclinical studies are necessary to identify other compounds with anti-hypertensive activity and additional pharmacological pathways.
