Ricinus communis is an important oilseed crop worldwide and is also considered one of the best potential plants for salt‐affected soil improvement in northeast China. However, little is known about photosynthesis and carbohydrate metabolism in this plant, nor the distribution of carbohydrates in cotyledons and roots under salinity stress.In the present study, seedling growth, gas exchange parameters (PN, E, gs and Ci), carbohydrate (fructose, sucrose, glucose, soluble sugar and starch) metabolism and related enzymes and genes were measured in Ricinus plants.Under salt stress, PN of cotyledons decreased significantly (P < 0.05), resulting in weak photosynthetic capacity. Furthermore, salt stress increased sucrose and glucose content in cotyledons, but decreased soluble sugar and starch content. However, sucrose increased and starch decreased in roots. This may be correlated with the increasing sugar metabolism under salinity, including notable changes in sugar‐related enzyme activities (SPS, SuSy, α‐amylase and β‐amylase) and gene expression of RcINV, RcSUS, RcAmY, RcBAM and RcGBE1.The results suggest that salinity reduces photosynthesis of cotyledons, alters carbohydrate allocation between cotyledons and roots and also promotes starch utilization in cotyledons and starch biosynthesis in roots, leading to a functional imbalance between cotyledons and roots. Understanding how plants adjust their requirements for different N forms can help elucidate plant coexistence strategies in N‐limited desert ecosystems. To understand the mechanisms involved, we investigated whether two desert herbs can directly absorb dissolved organic nitrogen (N) and tested whether the patterns changed over different growth stages.Two dominant herbaceous species, Astragalus arpilobus and Arnebia decumbens, from the southern edge of the Gurbantunggut desert, China, were selected. Short‐term (24 h) 15N‐labelled tracer (15N‐NO3, 15N‐NH4, 2‐13C‐15N‐Glycine) treatments were conducted at two soil depths (0–5 cm and 5–15 cm) in the season of rapid growth (June) and in the peak biomass season (July). Enrichment in 13C and 15N was assessed in the two species receiving glycine.The ratio 13C:15N was 0.21–1.39 at the 24‐h harvest, suggesting that approximately 10.5–69.5% of glycine had been absorbed. The amount of absorbed 15N was significantly affected by species, month, soil depth and N form. The two species absorbed most 15N from the 0–5 cm soil layer, and the absorption rate in July was higher than that in June. The absorption of 15N‐NO3 and 15N‐NH4 was significantly higher than that of 2‐13C‐15N‐Glycine.The results indicate that these herbs could use amino acids in the N‐deficient desert ecosystem. Growth and functioning of Sphagnum mosses are closely linked to water level and chemistry. Sphagnum mosses occur in wet, generally acidic conditions, and when buffered, alkaline water is known to negatively impact Sphagnum. The effects of time, dose and species‐specific responses of buffered, alkaline water on Sphagnum are largely unknown.We investigated the effects of bicarbonate and calcium on the survival, growth and physiological functioning of seven Sphagnum species occurring in contrasting environments, from raised bogs to (rich) fens. Mosses were submerged in different concentrations of bicarbonate and calcium solutions for 10 weeks under climate‐controlled circumstances.After 2 weeks, all species exposed to the high bicarbonate treatment (2.0 mM) showed severe potassium leakage and swift discoloration. In contrast, species showed differential responses to the intermediate bicarbonate treatment (0.8 mM), some with a later onset of potassium leakage. S. squarrosum, S. teres & S. contortum generally persisted the longest, with all species dying after 6 to 10 weeks. Calcium alone, in contrast, negatively affected S. squarrosum, S. teres & S. contortum, causing discoloration and potassium leakage.Our study shows enrichment with bicarbonate, but not calcium, is detrimental for most Sphagnum species tested. A mechanistic model was developed that is consistent with dose and duration dependence and the species specificity. Future conservation and restoration measures for Sphagnum‐dominated habitats and Sphagnum farming (cultivation, production and harvest of Sphagnum moss biomass) should limit flooding with bicarbonate‐rich waters while investigating new management options, like acidifying surface waters to lower bicarbonate levels. Persistence and colonization by tree species in an environment following a fire depends on the effects on seed germination and seedling development. We used seeds of Kielmeyera coriacea and Qualea parviflora as a model to test the effects of high temperatures on germination and initial development of tree seedlings.We exposed the seeds to heat flow (70, 100, 130, 150 or 170 °C) for 2 or 5 min and compared the germination with that of unheated seeds (control). Seedlings were then harvested after 3, 7 or 15 days to evaluate aerial and root mass, root:shoot ratio, presence of cotyledon opening, true leaves, and secondary roots.We found no effect on germination for seeds exposed to temperatures ≥150 °C. However, germination was significantly reduced for seeds exposed to 100 °C for both 2 and 5 min. The mass of 15‐day‐old K. coriacea seedlings was smaller when seeds were heated at 70 °C for 5 min or at temperatures higher or equal to 100 °C. Qualea parviflora seedlings did not show any difference in mass, but there were marginal differences in the presence of roots and the opening of cotyledons. Kielmeyera coriacea seedlings allocated biomass faster than Q. parviflora.High temperatures affect both quantity and quality of germinable seeds, as well as biomass allocation during initial seedling development. These factors may explain the decrease in seedlings observed after fire, suggesting a bottleneck effect that influences population dynamics and species persistence in systems with frequent fires. Abscisic acid (ABA), gibberellins (GA), auxins, cytokinins (CK) and ethylene (ET) are the "classical" phytohormones, while more recent discoveries are jasmonates (JA), salicylic acid (SA), polyamines (PAs), brassinosteroids (BR) and strigolactones. Some molecules (such as phytohormones, kinases and special organic or inorganic components) have been demonstrated as central hubs for gene regulation, signalling or metabolism in plant stress responses. However, it is very difficult to accurately distinguish the roles of AMF through direct or indirect pathways, since plant stress responses are commonly followed by concurrent changes in plant growth and nutrient availability. Waterlogging and salinity impair crop growth and productivity worldwide, with their combined effects being larger than the additive effects of the two stresses separately. Here, a common forage tetraploid Lotus corniculatus (cv. San Gabriel) and a diploid L. corniculatus accession, collected from a coastal area with high frequency of waterlogging–saline stress events, were evaluated for tolerance to waterlogging, salinity and these two stresses combined. We hypothesize that, due to its environmental niche, the diploid accession would show better adaptation to combined waterlogging–saline stress compared to the tetraploid L. corniculatus.Plants were evaluated under control conditions, waterlogging, salinity and a combined waterlogging–saline treatment for 33 days. Shoot and root growth were assessed, together with chlorophyll fluorescence and gas exchange measurements. Results showed that salinity and waterlogging effects were more severe for the tetraploid accession, with a larger effect being observed under the combined stress condition.Concentrations of Na+, Cl− and K+ were measured in apical and basal leaves, and in roots. A larger accumulation of Na+ and Cl− was observed under both saline and combined stress treatments for the tetraploid L. corniculatus, for which ion toxicity effects were evident.The expression of CLC gene, coding for a Cl− transporter, was only increased in diploid L. corniculatus plants in response to the combined stress condition, suggesting that ion compartmentalization mechanisms were induced in this accession. Within an Open Science project, research was carried out to describe to the public of the Ghirardi Botanic Garden (BS, Lombardy, Italy) the invisible features of plants. This work is dedicated to Scutellaria altissima L. (Lamiaceae).Micromorphological, histochemical and phytochemical investigations were conducted on the vegetative and reproductive organs to correlate the structures involved in the emission of substances and their unique productivity. This work reports volatile organic compound (VOC) profiles of leaves and flowers and the composition of essential oil (EO) obtained from aerial parts of plants cultivated in Italy that have never been described before.Three morphotypes of glandular trichomes were observed: peltate, short‐stalked capitate and long‐stalked capitate. Peltate trichomes were the main producers of terpenes, short‐stalked capitates of polysaccharides and long‐stalked capitates of terpenes and polyphenols. The leaf VOC profile showed heterogeneous composition, with non‐terpene derivatives as the major chemical class (71.04%), while monoterpene hydrocarbons represented almost the totality of the flower (99.73%). The leaf presented a higher number of total (37 versus 11) and exclusive (33 versus 7) compounds. (Z)‐3‐Hexenol acetate was most abundant in the leaf and (E)‐β‐ocimene in the flower. Four common compounds were detected: β‐pinene, β‐caryophyllene, γ‐muurolene and germacrene‐D. The EO contaied 21 compounds, dominated by β‐caryophyllene, linalool and hexahydrofarnesyl acetone.This research allowed us to correlate morphotypes of the secretory structures with the production of secondary metabolites, with the aim of providing the public of the Ghirardi Botanic Garden with a dedicated iconographic approach, which accounts for olfactory perception linked to S. altissima. Nutrient resorption, one of the most important strategies for plant nutrient conservation, is significantly affected by soil fertility. However, the effects of experimentally altered soil fertility on plant N and P resorption are poorly understood.The potential nutrient resorption response mechanisms of the dominant species Leymus chinensis to six N addition levels (0, 2.5, 5, 10, 20 and 40 g·N·m−2·year−1), two P addition levels (0 and 10 g P·m−2·year−1) and their interactions were studied after 3 years of treatments in a temperate meadow steppe.In both green leaves and culms, N and P addition significantly increased N and P concentrations, respectively. Nitrogen addition led to a decrease in the N resorption efficiency (NRE) of both leaves and culms. Within each N treatment, P addition decreased the P resorption efficiency (PRE) of both leaves and culms and the NRE of leaves, except in the N2.5 treatment. Both NRE and PRE in leaves were higher than those in culms under N and P addition conditions. The nutrient concentrations and resorption efficiency were significantly correlated with the soil nutrient availability.Our results suggest that plants rely more on nutrient absorption from the soil, reducing the proportion of elements obtained through nutrient resorption in nutrient‐rich environments. Plants are known to respond to warming temperatures. Few studies, however, have included the temperature experienced by the parent plant in the experimental design, in spite of the importance of this factor for population dynamics.We investigated the phenological and growth responses of seedlings of two key temperate tree species (Fagus sylvatica and Quercus robur) to spatiotemporal temperature variation during the reproductive period (parental generation) and experimental warming of the offspring. To this end, we sampled oak and beech seedlings of different ages (1–5 years) from isolated mother trees and planted the seedlings in a common garden.Warming of the seedlings advanced bud burst in both species. In oak seedlings, higher temperatures experienced by mother trees during the reproductive period delayed bud burst in control conditions, but advanced bud burst in heated seedlings. In beech seedlings, bud burst timing advanced both with increasing temperatures during the reproductive period of the parents and with experimental warming of the seedlings. Relative diameter growth was enhanced in control oak seedlings but decreased with warming when the mother plant experienced higher temperatures during the reproductive period.Overall, oak displayed more plastic responses to temperatures than beech. Several Cerrado tree species have traits and structures that protect from fires. The effectiveness of a trait depends on the fire regime, especially the frequency. We used Vochysia elliptica, a common Cerrado tree, as a model to test whether different fire frequencies alter crown architecture and flower, fruit and seed production.We analysed the effect of fire on the production of inflorescences, fruits and seeds, as well as seed germination and tree architecture of 20 trees in each of three plots of a long‐term ecological experiment managed with different fire regimes: burned every 2 years (B), burned every 4 years (Q) in mid‐dry season and an area protected from fire (C).We found a large negative effect of fire frequency on crown architecture and on flower and fruit production. Trees in C and Q had significantly more main branches and a larger crown area than trees in B. At its peak, a tree in C was expected to produce 2.4 times more inflorescences than Q, and 15.5 times more than B, with similar magnitudes for fruits. Sixty per cent of trees in B and 10% in Q produced no fruits.The differences in architecture might explain the reduction in sexual reproduction due to a smaller physical space to produce flowers at the branch apices. Resource limitation due to plant investment to replace burned vegetative parts may also decrease sexual reproduction. Orchids are a classic angiosperm model for understanding biotic pollination. We studied orchid species within two species‐rich herbaceous communities that are known to have either hymenopteran or dipteran insects as the dominant pollinators, in order to understand how flower colour relates to pollinator visual systems.We analysed features of the floral reflectance spectra that are significant to pollinator visual systems and used models of dipteran and hymenopteran colour vision to characterise the chromatic signals used by fly‐pollinated and bee‐pollinated orchid species.In contrast to bee‐pollinated flowers, fly‐pollinated flowers had distinctive points of rapid reflectance change at long wavelengths and a complete absence of such spectral features at short wavelengths. Fly‐pollinated flowers also had significantly more restricted loci than bee‐pollinated flowers in colour space models of fly and bee vision alike.Globally, bee‐pollinated flowers are known to have distinctive, consistent colour signals. Our findings of different signals for fly pollination is consistent with pollinator‐mediated selection on orchid species that results from the distinctive features of fly visual systems. Nutations of plant organs are significantly affected by the circatidal modulation in the gravitational force exerted by the Moon and Sun (lunisolar tidal acceleration, Etide). In a previous study on nutational rotations of stem apices, we observed abrupt alterations in their direction and irregularities of the recorded trajectories. Such transitions have not yet been analysed in detail.Peppermint plants were continuously recorded with time‐lapse photography and aligned with contemporaneous time courses of the Etide estimates. Each nutational stem tip movement path was assigned to one of two groups, depending on its geometry, as: (i) regular elliptical movements and (ii) irregular movements (with a random type of trajectory). Analyses of the correlation between the plant nutation trajectory parameters and Etide, as well as of the trajectory geometry of the individual plants were performed.The trajectory geometry of young mint stem apices was related to the velocity of the apex rotation and significantly affected by the gravitational force estimated from the Etide. A low velocity of nutational movement, associated with the random character of the trajectory, usually occurred simultaneously with local minima or maxima of Etide. As the mint plant ages, the transitions in the stem tip trajectory were limited; no correspondence with Etide dynamics was observed.The results indicate that the plant tip geometry path transitions with respect to the changing gradient of lunisolar tidal acceleration could be interpreted as manifestation of a continuous accommodation of the shoot apical part to the state of minimum energy dissipation. Based on the elemental composition of major biochemical molecules associated with different biological functions, the 'growth rate hypothesis' proposed that organisms with a higher growth rate would be coupled to lower C:N, especially lower C:P and N:P ratios. However, the applicability of the growth rate hypothesis for plants is unclear, especially for shrubs growing under different water supply.We performed an experiment with eight soil moisture levels (soil water content: 4%, 6%, 8%, 13%, 18%, 23%, 26% and 28%) to evaluate the effects of water availability on leaf C:N:P stoichiometry in the shrub Zygophyllum xanthoxylum.We found that leaves grew slowly and favored accumulation of P over C and N under both high and low water supply. Thus, leaf C:P and N:P ratios were unimodally related to soil water content, in parallel with individual leaf area and mass. As a result, there were significant positive correlations between leaf C:P and N:P with leaf growth (u).Our result that slower‐growing leaves had lower C:P and N:P ratios does not support the growth rate hypothesis, which predicted a negative association of N:P ratio with growth rate, but it is consistent with recent theoretical derivations of growth–stoichiometry relations in plants, where N:P ratio is predicted to increase with increasing growth for very low growth rates, suggesting leaf growth limitation by C and N rather than P for drought and water saturation. Rice is the most important staple food for half of the world's population, but also accounts for about 10% of all anthropogenic CH4 emissions. In spite of a wealth of information on the mechanistic basis and the importance of the rice plant in mediating these emissions, the significance of root exudation for CH4 emissions and the processes that determine root exudation are not well understood. Root exudates derive from photosynthate allocated to the root and subjected to root anabolic and catabolic processes. Key processes in roots that determine the extent of root exudation and, hence, CH4 emission from rice agriculture, include (i) deviation of metabolites from root anabolic and catabolic pathways facilitating root exudation, but also (ii) xylem loading and transport of potential root exudates for reallocation to the leaves, and (iii) xylem loading of sucrose in roots for its transport into reproductive organs, both suppressing root exudation. These processes are modulated by plant development and metabolic requirements resulting from different functions of root exudation. Amino acids represent an important component in the diet of the Venus flytrap ( Dionaea muscipula), and supply plants with much needed nitrogen resources upon capture of insect prey. Little is known about the significance of prey-derived carbon backbones of amino acids for the success of Dionaea's carnivorous life-style., The present study aimed at characterizing the metabolic fate of 15N and 13C in amino acids acquired from double-labeled insect powder. We tracked changes in plant amino acid pools and their δ13C- and δ15N-signatures over a period of five weeks after feeding, as affected by contrasting feeding intensity and tissue type ( i.e., fed and non-fed traps and attached petioles of Dionaea)., Isotope signatures ( i.e., δ13C and δ15N) of plant amino acid pools were strongly correlated, explaining 60% of observed variation. Residual variation was related to contrasting effects of tissue type, feeding intensity and elapsed time since feeding. Synthesis of nitrogen-rich transport compounds ( i.e., amides) during peak time of prey digestion increased 15N- relative to 13C- abundances in amino acid pools. After completion of prey digestion, 13C in amino acid pools was progressively exchanged for newly fixed 12C. The latter process was most evident for non-fed traps and attached petioles of plants that had received ample insect powder., We argue that prey-derived amino acids contribute to respiratory energy gain and loss of 13CO2 during conversion into transport compounds ( i.e., 2 days after feeding), and that amino-nitrogen helps boost photosynthetic carbon gain later on ( i.e., 5 weeks after feeding). Naturalists Fritz and Hermann Müller hypothesised that heteranthery often leads to a division of labour into 'feeding' and 'pollinating' stamens; the latter often being as long as the pistil so as to promote successful pollination on the bees' back. In many buzz-pollinated species of Senna, however, the so-called pollinating stamens are short and not level with the stigma, raising the question of how pollen is shed on the bees' back. Here we explore a mechanism called 'ricochet pollination'. We test whether division of labour is achieved through the interaction between short lower stamens and strongly concave 'deflector petals'., We studied the arrangement and morphology of the floral organs involved in the ricochet pollination, functioning of the flowers through artificial sonication and observed the interactions between bees and flowers in the field., The middle stamens are adapted to eject pollen downwards, which can be readily collected on the bee mid legs. Most of the pollen is ejected towards the deflector petal(s). Pollen from this set of stamens is more likely to contribute to pollination. The pollen grains seem to ricochet multiple times against the deflector petals to eventually reach the bee's back., The pollen ricochet mechanism promotes a division of labour by involving additional floral organs, such as petals, reinforcing the Müllers' division-of-labour hypothesis. Orbital movement of the Moon generates a system of gravitational fields that periodically alter the gravitational force on Earth. This lunar tidal acceleration (Etide) is known to act as an external environmental factor affecting many growth and developmental phenomena in plants. Our study focused on the lunar tidal influence on stem elongation growth, nutations and leaf movements of peppermint., Plants were continuously recorded with time-lapse photography under constant illumination as well in constant illumination following 5 days of alternating dark-light cycles. Time courses of shoot movements were correlated with contemporaneous time courses of the Etide estimates. Optical microscopy and SEM were used in anatomical studies., All plant shoot movements were synchronised with changes in the lunisolar acceleration. Using a periodogram, wavelet analysis and local correlation index, a convergence was found between the rhythms of lunisolar acceleration and the rhythms of shoot growth. Also observed were cyclical changes in the direction of rotation of stem apices when gravitational dynamics were at their greatest. After contrasting dark-light cycle experiments, nutational rhythms converged to an identical phase relationship with the Etide and almost immediately their renewed movements commenced. Amplitudes of leaf movements decreased during leaf growth up to the stage when the leaf was fully developed; the periodicity of leaf movements correlated with the Etide rhythms., For the fist time, it was documented that lunisolar acceleration is an independent rhythmic environmental signal capable of influencing the dynamics of plant stem elongation. Forests store the largest terrestrial pools of carbon (C), helping to stabilize the global climate system, yet are threatened by climate change (CC) and associated air pollution (AP, highlighting ozone (O3) and nitrogen oxides (NOx)). We adopt the perspective that CC-AP drivers and physiological impacts are universal, resulting in consistent stress responses of forest ecosystems across zonobiomes. Evidence supporting this viewpoint is presented from the literature on ecosystem gross/net primary productivity and water cycling. Responses to CC-AP are compared across evergreen/deciduous foliage types, discussing implications of nutrition and resource turnover at tree and ecosystem scales. The availability of data is extremely uneven across zonobiomes, yet unifying patterns of ecosystem response are discernable. Ecosystem warming results in trade-offs between respiration and biomass production, affecting high elevation forestsmore than in the lowland tropics and low-elevation temperate zone. Resilience to drought is modulated by tree size and species richness. Elevated O3 tends to counteract stimulation by elevated carbon dioxide (CO2). Biotic stress and genomic structure ultimately determine ecosystem responsiveness. Aggrading early-rather than mature late-successional communities respond to CO2 enhancement, whereas O3 affects North American and Eurasian tree species consistently under free-air fumigation. Insect herbivory is exacerbated by CC-AP in biome-specific ways. Rhizosphere responses reflect similar stand-level nutritional dynamics across zonobiomes, but are modulated by differences in tree-soil nutrient cycling between deciduous and evergreen systems, and natural versus anthropogenic nitrogen (N) oversupply. The hypothesis of consistency of forest responses to interacting CC-AP is supported by currently available data, establishing the precedent for a global network of long-term coordinated research sites across zonobiomes to simultaneously advance both bottom-up (e.g., mechanistic) and top-down (systems-level) understanding. This global, synthetic approach is needed because high biological plasticity and physiographic variation across individual ecosystems currently limit development of predictive models of forest responses to CC-AP. Integrated research on C and nutrient cycling, O3-vegetation interactions and water relations must target mechanisms' ecosystem responsiveness. Worldwide case studies must be subject to biostatistical exploration to elucidate overarching response patterns and synthesize the resulting empirical data through advanced modelling, in order to provide regionally coherent, yet globally integrated information in support of internationally coordinated decision-making and policy development. With the projected increase in drought duration and intensity in future, small water bodies, and especially the terrestrial-aquatic interfaces, will be subjected to longer dry periods with desiccation of the sediment. Drought effects on the plant-sediment microorganism carbon continuum may disrupt the tight linkage between plants and microbes which governs sediment carbon and nutrient cycling, thus having a potential negative impact on carbon sequestration of small freshwater ecosystems. However, research on drought effects on the plant-sediment carbon transfer in aquatic ecosystems is scarce. We therefore exposed two emergent aquatic macrophytes, Phragmites australis and Typha latifolia, to a month-long summer drought in a mesocosm experiment., We followed the fate of carbon from leaves to sediment microbial communities with 13 CO2 pulse labelling and microbial phospholipid-derived fatty acid ( PLFA) analysis., We found that drought reduced the total amount of carbon allocated to stem tissues but did not delay the transport. We also observed an increase in accumulation of 13C-labelled sugars in roots and found a reduced incorporation of 13C into the PLFAs of sediment microorganisms., Drought induced a switch in plant carbon allocation priorities, where stems received less new assimilates leading to reduced starch reserves whilst roots were prioritised with new assimilates, suggesting their use for osmoregulation. There were indications that the reduced carbon transfer from roots to microorganisms was due to the reduction of microbial activity via direct drought effects rather than to a decrease in root exudation or exudate availability. We studied biotically pollinated angiosperms on Macquarie Island, a remote site in the Southern Ocean with a predominately or exclusively dipteran pollinator fauna, in an effort to understand how flower colour affects community assembly. We compared a distinctive group of cream-green Macquarie Island flowers to the flora of likely source pools of immigrants and to a continental flora from a high latitude in the northern hemisphere. We used both dipteran and hymenopteran colour models and phylogenetically informed analyses to explore the chromatic component of community assembly. The species with cream-green flowers are very restricted in colour space models of both fly vision and bee vision and represent a distinct group that plays a very minor role in other communities. It is unlikely that such a community could form through random immigration from continental source pools. Our findings suggest that fly pollination has imposed a strong ecological filter on Macquarie Island, favouring floral colours that are rare in continental floras. This is one of the strongest demonstrations that plant-pollinator interactions play an important role in plant community assembly. The two-pore cation channel TPC1 operates as a dimeric channel in animal and plant endomembranes. Each subunit consists of two homologous Shaker-like halves, with 12 transmembrane domains in total (S1-S6, S7-S12). In plants, TPC1 channels reside in the vacuolar membrane, and upon voltage stimulation, give rise to the well-known slow-activating SV currents., Here, we combined bioinformatics, structure modelling, site-directed mutagenesis, and in planta patch clamp studies to elucidate the molecular mechanisms of voltage-dependent channel gating in TPC1 in its native plant background., Structure-function analysis of the Arabidopsis TPC1 channel in planta confirmed that helix S10 operates as the major voltage-sensing site, with Glu450 and Glu478 identified as possible ion-pair partners for voltage-sensing Arg537. The contribution of helix S4 to voltage sensing was found to be negligible. Several conserved negative residues on the luminal site contribute to calcium binding, stabilizing the closed channel., During evolution of plant TPC1s from two separate Shaker-like domains, the voltage-sensing function in the N-terminal Shaker-unit (S1-S4) vanished. Bamboos are one of the fastest growing plants on Earth, and are widely considered to have high ability to capture and sequester atmospheric carbon, and consequently to mitigate climate change. We tested this hypothesis by measuring carbon dioxide ( CO2) emissions from bamboo culms and comparing them with their biomass sequestration potential. We analysed diurnal effluxes from Bambusa vulgaris culm surface and gas mixtures inside hollow sections of various bamboos using gas chromatography. Corresponding variations in gas pressure inside the bamboo section and culm surface temperature were measured. SEM micrographs of rhizome and bud portions of bamboo culms were also recorded. We found very high CO2 effluxes from culm surface, nodes and buds of bamboos. Positive gas pressure and very high concentrations of CO2 were observed inside hollow sections of bamboos. The CO2 effluxes observed from bamboos were very high compared to their carbon sequestration potential. Our measurements suggest that bamboos are net emitters of CO2 during their lifespan. Prescribed burning is an important management tool in many parts of the world. While natural fires generally occur during the driest and warmest period of the year, prescribed burning is often timed out-of-season, when there is higher soil moisture and lower biomass combustibility. However, fire season may influence seedling recruitment after fire, e.g. through the effect of seed hydration status on fire tolerance. In non-fire-prone temperate regions, anthropogenic fire may occur exclusively in periods outside the growing season with higher soil moisture, which may have negative consequences on seedling recruitment. Fire tolerance of moist and dry seeds of 16 temperate European herbaceous species belonging to four families was assessed using heat treatment of 100 °C for 5 min and subsequent germination trials. Moist seeds of Asteraceae, Poaceae and Brassicaceae had a predominantly negative reaction to the heat treatment, while those of Fabaceae tolerated it or germination was even enhanced. The reaction of dry seeds was completely different, with positive responses in three species of the Fabaceae and fire tolerance in species of other families. Our results point out that hydration status may significantly influence the post-fire germination of seeds. Dry seeds were found to tolerate high heat, while moist seeds were harmed in more than half of the species. This implies that if prescribed burning is applied in temperate grasslands of Europe, it should be timed to dry periods of the dormant season in order to protect seeds from negative effects of fire. Fungal endophytes have been documented in almost all terrestrial plant groups. Although the endophyte infection syndrome in agronomic cultivars is well studied, relatively little work addresses questions of spatial ecology and fire effects on epichloae endophyte infection in native grasses, and none, to our knowledge, in sub- Saharan Africa. We sampled seven populations of the native Festuca costata Nees along the spline of the Drakensberg range in South Africa at several spatial scales, including both recently burned and unburned stands. We tested epichloae presence and prevalence with immunoblot assays, PCR and genetic sequencing. We found epichloae endophytes were present and prevalent (38-98% infection rates depending on location). Variation in infection rates occurred primarily among locations, but also among bunches. There was little evidence that endophyte infection rates varied with fire. Novel evidence of epichloae infection of a native Festuca in South Africa opens the door to several new research questions, from the phylogenetic relationship between epichloae of sub- Saharan Africa and other continents to the ecological advantages or disadvantages that endophytes confer upon their hosts, especially in a fire-prone ecosystem vulnerable to global environmental change. Living plant neighbours, but also their dead aboveground remains ( i.e. litter), may individually exert negative or positive effects on plant recruitment. Although living plants and litter co-occur in most ecosystems, few studies have addressed their combined effects, and conclusions are ambivalent. Therefore, we examined the response in terms of seedling emergence and growth of herbaceous grassland and forest species to different litter types and amounts and the presence of competitors. We conducted a pot experiment testing the effects of litter type (grass, oak), litter amount (low, medium, high) and interspecific competition (presence or absence of four Festuca arundinacea individuals) on seedling emergence and biomass of four congeneric pairs of hemicryptophytes from two habitat types (woodland, grassland). Interactions between litter and competition were weak. Litter presence increased competitor biomass. It also had positive effects on seedling emergence at low litter amounts and negative effects at high litter amounts, while competition had no effect on seedling emergence. Seedling biomass was negatively affected by the presence of competitors, and this effect was stronger in combination with high amounts of litter. Litter affected seedling emergence while competition determined the biomass of the emerged individuals, both affecting early stages of seedling recruitment. High litter accumulation also reduced seedling biomass, but this effect seemed to be additive to competitor effects. This suggests that live and dead plant mass can affect species recruitment in natural systems, but the mechanisms by which they operate and their timing differ. [ABSTRACT FROM AUTHOR]

Rising temperatures and more frequent and severe climatic extremes as a consequence of climate change are expected to affect growth and distribution of tree species that are adapted to current local conditions. Species distribution models predict a considerable loss of habitats for Pinus sylvestris. These models do not consider possible intraspecific differences in response to drought and warming that could buffer those impacts. We tested 10 European provenances of P. sylvestris, from the southwestern to the central European part of the species distribution, for their response to warming and to drought using a factorial design. In this common-garden experiment the air surrounding plants was heated directly to prevent excessive soil heating, and drought manipulation, using a rain-out shelter, permitted almost natural radiation, including high light stress. Plant responses were assessed as changes in phenology, growth increment and biomass allocation. Seedlings of P. sylvestris revealed a plastic response to drought by increased taproot length and root-shoot ratios. Strongest phenotypic plasticity of root growth was found for southwestern provenances, indicating a specific drought adaptation at the cost of overall low growth of aboveground structures even under non-drought conditions. Warming had a minor effect on growth but advanced phenological development and had a contrasting effect on bud biomass and diameter increment, depending on water availability. The intraspecific variation of P. sylvestris provenances could buffer climate change impacts, although additional factors such as the adaptation to other climatic extremes have to be considered before assisted migration could become a management option. Turions play an important role in the survival strategy of the duckweed Spirodela polyrhiza. Therefore, factors influencing the formation of these survival organs were studied. Phosphate deficiency is the main natural factor inducing turion formation and the specific turion yield ( SY), i.e. the number of turions formed per frond, varied widely for five different clones from different climate zones. The concentrations of phosphate and nitrate in the nutrient media were investigated at the onset of turion formation, with SY ranging from 0.22 to 5.9. Tissue P and N content was also investigated in vegetative fronds at the onset of turion formation and in newly formed turions. The clones were selected to test possible correlations between SY and threshold nutrient concentration for turion formation. Only one correlation, between SY and external phosphate concentration, was significant: clones with high SY started turion formation at higher external phosphate concentrations. Turion formation is thus mainly induced by the external phosphate concentration, below a defined, clone-dependent threshold. We propose the following mechanism: a switch of the developmental programme of frond primordia from vegetative frond to turion formation at a higher phosphate threshold saves more phosphate for turion formation instead of using it for a further vegetative growth. However, the period of growth preceding turion formation does not depend on this threshold value, but rather on the growth rate of the vegetative fronds, which actually produce the phosphate shortage by taking it up from the surrounding medium. Lemnaceae, commonly called duckweeds, comprise a diverse group of floating aquatic plants that have previously been classified into 37 species based on morphological and physiological criteria. In addition to their unique evolutionary position among angiosperms and their applications in biomonitoring, the potential of duckweeds as a novel sustainable crop for fuel and feed has recently increased interest in the study of their biodiversity and systematics. However, due to their small size and abbreviated structure, accurate typing of duckweeds based on morphology can be challenging. In the past decade, attempts to employ molecular barcoding techniques for species assignment have produced promising results; however, they have yet to be codified into a simple and quantitative protocol. A study that compiles and compares the barcode sequences within all known species of this family would help to establish the fidelity and limits of this DNA-based approach. In this work, we compared the level of conservation between over 100 strains of duckweed for two intergenic barcode sequences derived from the plastid genome. By using over 300 sequences publicly available in the NCBI database, we determined the utility of each of these two barcodes for duckweed species identification. Through sequencing of these barcodes from additional accessions, 30 of the 37 known species of duckweed could be identified with varying levels of confidence using this approach. From our analyses using this reference dataset, we also confirmed two instances where mis-assignment of species has likely occurred. Measuring biomarkers from plant tissue samples is challenging and expensive when the desire is to integrate transcriptomics, fluxomics, metabolomics, lipidomics, proteomics, physiomics and phenomics. We present a computational biology method where only the transcriptome needs to be measured and is used to derive a set of parameters for deterministic kinetic models of metabolic pathways. The technology is called Transcriptome-To-Metabolome™ ( TTM™) biosimulations, currently under commercial development, but available for non-commercial use by researchers. The simulated results on metabolites of 30 primary and secondary metabolic pathways in rice ( Oryza sativa) were used as the biomarkers to predict whether the transcriptome was from a plant that had been under drought conditions. The rice transcriptomes were accessed from public archives and each individual plant was simulated. This unique quality of the TTM™ technology allows standard analyses on biomarker assessments, i.e. sensitivity, specificity, positive and negative predictive values, accuracy, receiver operator characteristics ( ROC) curve and area under the ROC curve ( AUC). Two validation methods were also used, the holdout and 10-fold cross validations. Initially 17 metabolites were identified as candidate biomarkers based on either statistical significance on binary phenotype when compared with control samples or recognition from the literature. The top three biomarkers based on AUC were gibberellic acid 12 (0.89), trehalose (0.80) and sn1-palmitate-sn2-oleic-phosphatidylglycerol (0.70). Neither heat map analyses of transcriptomes nor all 300 metabolites clustered the stressed and control groups effectively. Plant growth in semi-arid ecosystems is usually severely limited by soil nutrient availability. Alleviation of these resource stresses by fertiliser application and aboveground litter input may affect plant internal nutrient cycling in such regions. We conducted a 4-year field experiment to investigate the effects of nitrogen (N) addition (10 g N·m−2·year−1) and plant litter manipulation on nutrient resorption of Leymus chinensis, the dominant native grass in a semi-arid grassland in northern China. Although N addition had no clear effects on N and phosphorus (P) resorption efficiencies in leaves and culms, N fertilisation generally decreased leaf N resorption proficiency by 54%, culm N resorption proficiency by 65%. Moreover, N fertilisation increased leaf P resorption proficiency by 13%, culm P resorption proficiency by 20%. Under ambient or enriched N conditions, litter addition reduced N and P resorption proficiencies in both leaves and culms. The response of P resorption proficiency to litter manipulation was more sensitive than N resorption proficiency: P resorption proficiency in leaves and culms decreased strongly with increasing litter amount under both ambient and enriched N conditions. In contrast, N resorption proficiency was not significantly affected by litter addition, except for leaf N resorption proficiency under ambient N conditions. Furthermore, although litter addition caused a general decrease of leaf and culm nutrient resorption efficiencies under both ambient and enriched N conditions, litter addition effects on nutrient resorption efficiency were much weaker than the effects of litter addition on nutrient resorption proficiency. Taken together, our results show that leaf and non-leaf organs of L. chinensis respond consistently to altered soil N availability. Our study confirms the strong effects of N addition on plant nutrient resorption processes and the potential role of aboveground litter, the most important natural fertiliser in terrestrial ecosystems, in influencing plant internal nutrient cycling. Auxin is a small molecule involved in most processes related to plant growth and development. Its effect usually depends on the distribution in tissues and the formation of concentration gradients. Until now there has been no tool for the direct tracking of auxin transport at the cellular and tissue level; therefore the majority of studies have been based on various indirect methods. However, due to their various restrictions, relatively little is known about the relationship between various pathways of auxin transport and specific developmental processes. We present a new research tool: fluorescently labelled auxin in the form of a conjugate with two different fluorescent tracers, FITC and RITC, which allows direct observation of auxin transport in plant tissues. Chemical analysis and biological tests have shown that our conjugates have auxin-like biological activity and transport; therefore they can be used in all experimental systems as an alternative to IAA. In addition, the conjugates are a universal tool that can be applied in studies of all plant groups and species. The conjugation procedure presented in this paper can be adapted to other fluorescent dyes, which are constantly being improved. In our opinion, the conjugates greatly expand the possibilities of research concerning the role of auxin and its transport in different developmental processes in plants. [ABSTRACT FROM AUTHOR]

Serpentine soils represent stressful habitats where plants have to cope with heavy metals, moisture limitation and low nutrient availability. We propose that facilitation is an important mechanism structuring plant communities under such stressful conditions. Facilitation has been shown to generate the spatial association of species, forming discrete vegetation patches of phylogenetically distant species. We measured these spatial and phylogenetic signatures left by facilitation in a serpentine plant community of central Cuba. Our results show that seedlings preferentially grow under plants of different species, and that adults are significantly aggregated into vegetation patches. In these patches, adults tend to co-occur with distant relatives, ultimately forming phylogenetically diverse neighbourhoods. We discuss possible mechanisms explaining how species adapted to serpentine areas may be acting as nurses, reducing the stressful conditions for the establishment of other species. Little is known about the effects of nutrient availability on the growth of Trifolium medium in alkaline soil. In 2010, a pot experiment (10 N, P and K fertiliser treatments) with seeding of T. medium into alkaline soil was performed and emergence of seedlings, survival, aboveground and belowground organs were studied. The positive effects of increased nutrient availability on seedling emergence ranged from 5% in the control to 17% in the high P treatment. The lowest mortality was in treatments with P and K supply and the highest in treatments with N supply, due to the sensitivity of young plants to high N availability. The highest values of most measured aboveground plant traits were recorded in treatments with simultaneous application of N, P and K. There were highly positive effects of P supply alone or in combination with N and K on the development of belowground organs. Taproot length ranged from 11.5 in high N to 40.2 cm in P treatment. There was a negative effect of N application on nodulation, especially in N treatments, where growth of T. medium was limited by insufficient P supply. The number of nodules per plant ranged from 0.8 to 4.5 in the high N and P treatments. As demonstrated in this study, T. medium is a potentially suitable legume for alkaline soils. It requires a relatively high P and K supply as well as moderate mineral N supply to achieve its maximum growth potential. [ABSTRACT FROM AUTHOR]

Gravity has shaped the evolution of life since its origin. However, experiments in the absence of this overriding force, necessary to precisely analyse its role, e.g. for growth, development, and orientation of plants and single cells, only became possible with the advent of spaceflight. Consequently, this research has been supported especially by space agencies around the world for decades, mainly for two reasons: first, to enable fundamental research on gravity perception and transduction during growth and development of plants; and second, to successfully grow plants under microgravity conditions with the goal of establishing a bioregenerative life support system providing oxygen and food for astronauts in long-term exploratory missions. For the second time, the International Space Life Sciences Working Group ( ISLSWG), comprised of space agencies with substantial life sciences programmes in the world, organised a workshop on plant biology research in space. The present contribution summarises the outcome of this workshop. In the first part, an analysis is undertaken, if and how the recommendations of the first workshop held in Bad Honnef, Germany, in 1996 have been implemented. A chapter summarising major scientific breakthroughs obtained in the last 15 years from plant research in space concludes this first part. In the second part, recommendations for future research in plant biology in space are put together that have been elaborated in the various discussion sessions during the workshop, as well as provided in written statements from the session chairs. The present paper clearly shows that plant biology in space has contributed significantly to progress in plant gravity perception, transduction and responses - processes also relevant for general plant biology, including agricultural aspects. In addition, the interplay between light and gravity effects has increasingly received attention. It also became evident that plants will play a major role as components of bioregenerative life support and energy systems that are necessary to complement physico-chemical systems in upcoming long-term exploratory missions. In order to achieve major progress in the future, however, standardised experimental conditions and more advanced analytical tools, such as state-of-the-art onboard analysis, are required. The tripeptide glutathione ( GSH) is an important metabolite with a broad spectrum of functions, and its homeostasis is essential to maintain cellular redox poise and effective responses to stress in plants. In Arabidopsis GSH is synthesised in two successive enzymatic steps by γ-glutamylcysteine synthetase ( GSH1), localised exclusively in plastids, forming the pathway intermediate γ-glutamylcysteine (γ- EC), and then by glutathione synthetase ( GSH2), which is located in both plastids and cytosol. This suggests a mechanism for γ- EC export from the plastids and, because the majority of GSH2 transcripts (90%) encode the cytosolic isoform, it is speculated that the cytosol may be the main compartment for GSH biosynthesis. With the availability of knockout lethal mutants of GSH1 and GSH2 in Arabidopsis, we were able to manipulate the GSH biosynthetic pathway within cells through transgenic techniques. We successfully complemented the gsh1 and gsh2 null mutants with a cytosol-targeted bacterial Ec GSHA and plastid-targeted Arabidopsis GSH2 protein, respectively, to wild-type phenotypes. These transgenics were little affected under heavy metal (cadmium) or oxidative stress ( H2O2) when compared to the wild type. Collectively, our data show that redirecting GSH1 activity exclusively to the cytosol or restricting GSH biosynthesis to the plastids has no significant impact on development or stress resistance, suggesting efficient exchange of γ- EC and GSH between the plastid and cytosol compartments within cells. In this work, we analyse morpho-physiological modifications presented during the allomorphic growth of the aroid vine Rhodospatha oblongata Poepp throughout its ascent into the forest canopy. We test the hypothesis that morphological modifications in the root, shoot and leaf are followed by a gradual improvement of the xylem vascular system in order to increase water acquisition and transport as body size increases. The characterisation of these structural modifications was based on 30-35 specimens divided into six size classes. The dimensions of shoots, leaves and roots were quantified and qualified. The transition from the terrestrial to the epiphytic phase was followed by a simultaneous increase of leaf number and lamina area, together with increased length and diameter of the petiole. Furthermore, as the plant grows, the shoot internodes become shorter and thicker. However, occurrence of aerial roots is the most important characteristic in the ascending phase. In taller individuals, the increase in number of roots with wider xylem vessels guarantees an increased theoretical xylem hydraulic conductance for this growth phase. Along an acropetal direction of the same shoot, the diameter of xylem vessels increased, while the number of vessels per stele area decreased, in contrast with such allometric models as that of West, Brown and Enquist, showing that xylem vessel number and diameter taper in a reverse manner along the same direction. Such structural changes of R. oblongata allow improved foraging for light and water, facilitating the survival of bigger-sized plants of this vine in the canopy. [ABSTRACT FROM AUTHOR]

Starch pasting viscosity is an important quality trait in cassava ( Manihot esculenta Crantz) cultivars. The aim here was to identify loci and candidate genes associated with the starch pasting viscosity. Quantitative trait loci ( QTL) mapping for seven pasting viscosity parameters was carried out using 100 lines of an F1 mapping population from a cross between two cassava cultivars Huay Bong 60 and Hanatee. Starch samples were obtained from roots of cassava grown in 2008 and 2009 at Rayong, and in 2009 at Lop Buri province, Thailand. The traits showed continuous distribution among the F1 progeny with transgressive variation. Fifteen QTL were identified from mean trait data, with Logarithm of Odds ( LOD) values from 2.77-13.01 and phenotype variations explained ( PVE) from10.0-48.4%. In addition, 48 QTL were identified in separate environments. The LOD values ranged from 2.55-8.68 and explained 6.6-43.7% of phenotype variation. The loci were located on 19 linkage groups. The most important QTL for pasting temperature ( PT) ( q PT.1 LG1) from mean trait values showed largest effect with highest LOD value (13.01) and PVE (48.4%). The QTL co-localised with PT and pasting time ( PTi) loci that were identified in separate environments. Candidate genes were identified within the QTL peak regions. However, the major genes of interest, encoding the family of glycosyl or glucosyl transferases and hydrolases, were located at the periphery of QTL peaks. The loci identified could be effectively applied in breeding programmes to improve cassava starch quality. Facilitation, reduced competition or increased competition can arise in mixed stands and become essential to the performance of these stands when compared to pure stands. Facilitation and over-yielding are widely held to prevail on poor sites, whereas neutral interactions or competition, leading to under-yielding of mixed versus pure stands, can occur on fertile sites. While previous studies have focused on the spatial variation of mixing effects, we examine the temporal variation of facilitation and competition and its effect on growth. The study is based on tree ring measurement on cores from increment borings from 559 trees of Norway spruce ( Picea abies [ L.] Karst.), European beech ( Fagus sylvatica [ L.]) and sessile oak ( Quercus petraea ( Matt.) Liebl.) in southern Germany, half of which were in pure stands and half in adjacent mixed stands. Mean basal area growth indices were calculated from tree ring measurements for pure and mixed stands for every species and site. The temporal variation, with positive correlations between species-specific growth indices during periods of low growth and neutral or negative correlations during periods of high growth, is more distinct in mixed than in neighbouring pure stands. We provide evidence that years with low growth trigger over-yielding of trees in mixed as opposed to pure stands, while years with high growth lead to under-yielding. We discuss the relevance of the results in terms of advancing our understanding and modelling of mixed stands, extension of the stress gradient hypothesis, and the performance of mixed versus pure stands in the face of climate change. Phosphorus ( P) acquisition, cycling and use efficiency has been investigated intensively with herbaceous plants. It is known that local as well as systemic signalling contributes to the control of P acquisition. Woody plants are long-lived organisms that adapt their life cycle to the changing environment during their annual growth cycle. Little is known about P acquisition and P cycling in perennial plants, especially regarding storage and mobilisation, its control by systemic and environmental factors, and its interaction with the largely closed ecosystem-level P cycle. In the present-day O2-rich atmosphere, the photorespiratory pathway is essential for organisms performing oxygenic photosynthesis; i.e. cyanobacteria, algae and land plants. The presence of enzymes for the plant-like 2-phosphoglycolate cycle in cyanobacteria indicates that, together with oxygenic photosynthesis, genes for photorespiratory enzymes were endosymbiotically conveyed from ancient cyanobacteria to photosynthetic eukaryotes. The genome information for Cyanophora paradoxa, a member of the Glaucophyta representing the first branching group of primary endosymbionts, and for many other eukaryotic algae was used to shed light on the evolutionary relationship of photorespiratory enzymes among oxygenic phototrophs. For example, it became possible to analyse the phylogenies of 2-phosphoglycolate phosphatase, serine:glyoxylate aminotransferase and hydroxypyruvate reductase. Analysis of the Cyanophora genome provided clear evidence that some photorespiratory enzymes originally acquired from cyanobacteria were lost, e.g. glycerate 3-kinase, while others were replaced by the corresponding enzymes from the α-proteobacterial endosymbiont, e.g. serine:glyoxylate aminotransferase. Generally, our analysis supports the view that many C2 cycle enzymes in eukaryotic phototrophs were obtained from the cyanobacterial endosymbiont, but during the subsequent evolution of algae and land plants multiple losses and replacements occurred, which resulted in a reticulate provenance of photorespiratory enzymes with different origins in different cellular compartments. Photorespiration is an essential prerequisite for all autotrophic organisms performing oxygenic photosynthesis. In contrast to the well-characterised enzymes accomplishing photorespiratory metabolism, current knowledge on the involved transport processes and the respective proteins is still quite limited. In this review, we focus on the status quo of translocators involved in photorespiratory metabolism. Although the transport of some of the photorespiratory intermediates could be characterised biochemically, using isolated organelles, the genes encoding these transporters have to date not been identified in most cases. Here, we describe the postulated transport processes, present information on established or hypothetical photorespiratory transporters, depict strategies on how to identify the transport proteins on the molecular level and, finally, discuss strategies for how to find the remaining candidates. Oxygenic photosynthesis would not be possible without photorespiration in the present day O2-rich atmosphere. It is now generally accepted that cyanobacteria-like prokaryotes first evolved oxygenic photosynthesis, which was later conveyed via endosymbiosis into a eukaryotic host, which then gave rise to the different groups of algae and streptophytes. For photosynthetic CO2 fixation, all these organisms use Rubis CO, which catalyses both the carboxylation and the oxygenation of ribulose 1,5-bisphosphate. One of the reaction products of the oxygenase reaction, 2-phosphoglycolate (2 PG), represents the starting point of the photorespiratory C2 cycle, which is considered largely responsible for recapturing organic carbon via conversion to the Calvin-Benson cycle ( CBC) intermediate 3-phosphoglycerate, thereby detoxifying critical intermediates. Here we discuss possible scenarios for the evolution of this process toward the well-defined 2 PG metabolism in extant plants. While the origin of the C2 cycle core enzymes can be clearly dated back towards the different endosymbiotic events, the evolutionary scenario that allowed the compartmentalised high flux photorespiratory cycle is uncertain, but probably occurred early during the algal radiation. The change in atmospheric CO2/ O2 ratios promoting the acquisition of different modes for inorganic carbon concentration mechanisms, as well as the evolutionary specialisation of peroxisomes, clearly had a dramatic impact on further aspects of land plant photorespiration. Shedding light on yet uncharacterised components of photorespiration, such as transport processes required for the function of this pathway, is a prerequisite for manipulating photorespiratory fluxes and hence for decreasing photorespiratory energy loss. The ability of forward genetic screens to identify missing links is apparently limited, as indicated by the fact that little progress has been made with this approach during the past decade. The availability of large amounts of gene expression data and the growing power of bioinformatics, paired with availability of computational resources, opens new avenues to discover proteins involved in transport of photorespiratory intermediates. Co-expression analysis is a tool that compares gene expression data under hundreds of different conditions, trying to find groups of genes that show similar expression patterns across many different conditions. Genes encoding proteins that are involved in the same process are expected to be simultaneously expressed in time and space. Thus, co-expression data can aid in the discovery of novel players in a pathway, such as the transport proteins required for facilitating the transfer of intermediates between compartments during photorespiration. The Arabidopsis mutant shm1-1 is defective in mitochondrial serine hydroxymethyltransferase 1 activity and displays a lethal photorespiratory phenotype at ambient CO2 concentration but grows normally at high CO2. After transferring high CO2-grown shm1-1 plants to ambient CO2, the younger leaves remain photosynthetically active while developed leaves display increased yellowing and decreased FV/ FM values. Metabolite analysis of plants transferred from high CO2 to ambient air indicates a massive light-dependent (photorespiratory) accumulation of glycine, 2-oxoglutarate (2 OG) and D-2-hydroxyglutarate ( D-2 HG). Amino acid markers of senescence accumulated in ambient air in wild-type and shm1-1 plants maintained in darkness and also build up in shm1-1 in the light. This, together with an enhanced transcription of the senescence marker SAG12 in shm1-1, suggests the initiation of senescence in shm1-1 under photorespiratory conditions. Mitochondrial D-2 HG dehydrogenase ( D-2 HGDH) converts D-2 HG into 2 OG. In vitro studies indicate that 2 OG exerts competitive inhibition on D-2 HGDH with a Ki of 1.96 m m. 2 OG is therefore a suitable candidate as inhibitor of the in vivo D-2HGDH activity, as 2 OG is produced and accumulates in mitochondria. Inhibition of the D-2 HGDH by 2 OG is likely a mechanism by which D-2HG accumulates in shm1-1, however it cannot be ruled out that D-2HG may also accumulate due to an active senescence programme that is initiated in these plants after transfer to photorespiratory conditions. Being intimately intertwined with ( C3) photosynthesis, photorespiration is an incredibly high flux-bearing pathway. Traditionally, the photorespiratory cycle was viewed as closed pathway to refill the Calvin- Benson cycle with organic carbon. However, given the network nature of metabolism, it hence follows that photorespiration will interact with many other pathways. In this article, we review current understanding of these interactions and attempt to define key priorities for future research, which will allow us greater fundamental comprehension of general metabolic and developmental consequences of perturbation of this crucial metabolic process. Sulphate uptake and its distribution within plants depend on the activity of different sulphate transporters ( SULTR). In long-living deciduous plants such as trees, seasonal changes of spatial patterns add another layer of complexity to the question of how the interplay of different transporters adjusts S distribution within the plant to environmental changes. Poplar is an excellent model to address this question because its S metabolism is already well characterized. In the present study, the importance of SULTRs for seasonal sulphate storage and mobilization was examined in the wood of poplar ( Populus tremula × P. alba) by analysing their gene expression in relation to sulphate contents in wood and xylem sap. According to these results, possible functions of the respective SULTRs for seasonal sulphate storage and mobilization in the wood are suggested. Together, the present results complement the previously published model for seasonal sulphate circulation between leaves and bark and provide information for future mechanistic modelling of whole tree sulphate fluxes. Carnivorous plants acquire most of their nutrients by capturing ants, insects and other arthropods through their leaf-evolved biological traps. So far, the best-known attractants in carnivorous prey traps are nectar, colour and olfactory cues. Here, fresh prey traps of 14 Nepenthes, five Sarracenia, five Drosera, two Pinguicula species/hybrids, Dionaea muscipula and Utricularia stellaris were scanned at UV 366 nm. Fluorescence emissions of major isolates of fresh Nepenthes khasiana pitcher peristomes were recorded at an excitation wavelength of 366 nm. N. khasiana field pitcher peristomes were masked by its slippery zone extract, and prey capture rates were compared with control pitchers. We found the existence of distinct blue fluorescence emissions at the capture spots of Nepenthes, Sarracenia and Dionaea prey traps at UV 366 nm. These alluring blue emissions gradually developed with the growth of the prey traps and diminished towards their death. On excitation at 366 nm, N. khasiana peristome 3:1 CHCl3-Me OH extract and its two major blue bands showed strong fluorescence emissions at 430-480 nm. Masking of blue emissions on peristomes drastically reduced prey capture in N. khasiana pitchers. We propose these molecular emissions as a critical factor attracting arthropods and other visitors to these carnivorous traps.
