BACKGROUND: People with forward head posture (FHP) and rounded shoulder posture (RSP) may experience shifts in scapular position and alterations in the strength of the muscles surrounding the scapula. Therefore, a reliable scapula position and scapula muscle strength test are required to identified scapula problem and provided appropriate intervention programs. OBJECTIVE: This study aimed to determine the reliability of scapular position measurements obtained using a modified digital inclinometer and palpation meter, as well as muscle strength using a hand-held dynamometer (HHD) in individuals with FHP and RSP. METHODS: Thirty healthy subjects underwent scapular position tests (upward rotation, anterior tilt, depression, and protraction) and strength tests of periscapular muscles (serratus anterior, upper trapezius, middle trapezius, and lower trapezius) on both the dominant and non-dominant sides. The reliability of the measurements was confirmed by computing the intraclass correlation coefficient (ICC), standard error of measurement (SEM), and minimal clinically important difference (MCID). Test and re-tests were conducted over a period of one week and actual values for each measurement were also obtained and assessed. RESULTS: The ICC ranged from 0.802 to 0.935 for the scapular position tests and from 0.808 to 0.949 for muscle strength tests on the dominant and non-dominant sides. For scapular position tests, SEM values ranged from 0.3 cm to 0.6 cm (protraction and depression), from 1.0∘ to 1.7∘ (upward rotation and anterior tilt) and MCID values ranged from 0.7 cm to 1.6 cm (protraction and depression), from 2.8∘ to 4.7∘ (upward rotation and anterior tilt). Regarding muscle strength tests, SEM and MCID values were in the range 14.4N–27.3N and 39.9N–75.7N, respectively. CONCLUSIONS: Scapular position and periscapular muscle strength can be reproducibly measured in healthy subjects individuals with FHP and RSP. BACKGROUD: The control of hip and ankle joint movement is important for patients with dynamic knee valgus (DKV), but few studies have used non-elastic taping (NET) to adjust alignment of the hip and ankle joints during overhead squat (OHS) simultaneously in patients with DKV. OBJECTIVE: We investigated changes in lower extremity joint kinematics and dynamic balance after the application of NET to the hip and ankle joints during OHS. METHODS: A total of 30 DKV patients participated in this study. We determined the kinematics of the hip, knee, and ankle joints, and scores on the lower quarter Y-balance test (YBT-LQ) during OHS under three conditions (non-taping, NET on hip and ankle, and sham taping). RESULTS: Hip internal rotation, knee valgus, and the ankle dorsiflexion angle were significantly lower with NET than with non- or sham taping. The hip flexion angle and scores on the YBT-LQ were significantly greater with NET than with non- or sham taping. CONCLUSIONS: The application of NET while performing a OHS is a useful method for correcting lower extremity alignment in patients with DKV, however, application of NET on the ankle should be considered. Aims/hypothesis: Non-alcoholic fatty liver disease (NAFLD) associated with type 2 diabetes may more easily progress towards severe forms of non-alcoholic steatohepatitis (NASH) and cirrhosis. Although the Wnt effector transcription factor 7-like 2 (TCF7L2) is closely associated with type 2 diabetes risk, the role of TCF7L2 in NAFLD development remains unclear. Here, we investigated how changes in TCF7L2 expression in the liver affects hepatic lipid metabolism based on the major risk factors of NAFLD development. Methods: Tcf7l2 was selectively ablated in the liver of C57BL/6N mice by inducing the albumin (Alb) promoter to recombine Tcf7l2 alleles floxed at exon 5 (liver-specific Tcf7l2-knockout [KO] mice: Alb-Cre;Tcf7l2f/f). Alb-Cre;Tcf7l2f/f and their wild-type (Tcf7l2f/f) littermates were fed a high-fat diet (HFD) or a high-carbohydrate diet (HCD) for 22 weeks to reproduce NAFLD/NASH. Mice were refed a standard chow diet or an HCD to stimulate de novo lipogenesis (DNL) or fed an HFD to provide exogenous fatty acids. We analysed glucose and insulin sensitivity, metabolic respiration, mRNA expression profiles, hepatic triglyceride (TG), hepatic DNL, selected hepatic metabolites, selected plasma metabolites and liver histology. Results: Alb-Cre;Tcf7l2f/f essentially exhibited increased lipogenic genes, but there were no changes in hepatic lipid content in mice fed a normal chow diet. However, following 22 weeks of diet-induced NAFLD/NASH conditions, liver steatosis was exacerbated owing to preferential metabolism of carbohydrate over fat. Indeed, hepatic Tcf7l2 deficiency enhanced liver lipid content in a manner that was dependent on the duration and amount of exposure to carbohydrates, owing to cell-autonomous increases in hepatic DNL. Mechanistically, TCF7L2 regulated the transcriptional activity of Mlxipl (also known as ChREBP) by modulating O-GlcNAcylation and protein content of carbohydrate response element binding protein (ChREBP), and targeted Srebf1 (also called SREBP1) via miRNA (miR)-33-5p in hepatocytes. Eventually, restoring TCF7L2 expression at the physiological level in the liver of Alb-Cre;Tcf7l2f/f mice alleviated liver steatosis without altering body composition under both acute and chronic HCD conditions. Conclusions/interpretation: In mice, loss of hepatic Tcf7l2 contributes to liver steatosis by inducing preferential metabolism of carbohydrates via DNL activation. Therefore, TCF7L2 could be a promising regulator of the NAFLD associated with high-carbohydrate diets and diabetes since TCF7L2 deficiency may lead to development of NAFLD by promoting utilisation of excess glucose pools through activating DNL. Context: A winged scapula (WS) is associated with faulty posture caused by weakness of the serratus anterior (SA), which mainly acts as a scapular stabilizer muscle. It is important to accurately assess and train the SA muscle with a focus on scapula stabilizers during musculoskeletal rehabilitation of individuals with a WS. Objective: The authors examined muscle activity in the SA and pectoralis major (PM), upper trapezius (UT), and anterior deltoid (AD) as well as shoulder protraction strength during isometric shoulder protraction in individuals with and without a WS. Design: Cross-sectional study. Setting: A clinical biomechanics laboratory. Participants: In total, 27 males with no shoulder, neck, or upper-extremity pain participated. Main Outcome Measures: Isometric shoulder protraction strength was collected and surface electromyography used to measure the activity of the SA, PM, UT, and AD muscles and selective SA activity ratio to other shoulder muscles. Results: Electromyography activity of the SA muscle and shoulder protraction strength were significantly lower in individuals with a WS compared with the non-WS group (P <.05). In contrast, PM muscle activity and the PM-to-SA, UT-to-SA, and AD-to-SA ratios were significantly greater in individuals with a WS than in individuals without winging (P <.05). Conclusions: Isometric shoulder protraction for measuring SA strength in individuals with a WS should focus on isolated muscle activity of the SA, and SA strengthening exercises are important for individuals with a WS. Prominin-1, a lipid raft protein, is required for maintaining cancer stem cell properties in hepatocarcinoma cell lines, but its physiological roles in the liver have not been well studied. Here, we investigate the role of Prominin-1 in lipid rafts during liver regeneration and show that expression of Prominin-1 increases after 2/3 partial hepatectomy or CCl4 injection. Hepatocyte proliferation and liver regeneration are attenuated in liver-specific Prominin-1 knockout mice compared to wild-type mice. Detailed mechanistic studies reveal that Prominin-1 interacts with the interleukin-6 signal transducer glycoprotein 130, confining it to lipid rafts so that STAT3 signaling by IL-6 is effectively activated. The overexpression of the glycosylphosphatidylinsositol-anchored first extracellular domain of Prominin-1, which is the domain that binds to GP130, rescued the proliferation of hepatocytes and liver regeneration in liver-specific Prominin-1 knockout mice. In summary, Prominin-1 is upregulated in hepatocytes during liver regeneration where it recruits GP130 into lipid rafts and activates the IL6-GP130-STAT3 axis, suggesting that Prominin-1 might be a promising target for therapeutic applications in liver transplantation. A single-stage isolated ac–ac converter without the commutation problem is proposed. The proposed converter has the step-up/down function dependence on the phase shift control which provides zero-voltage switching zero-voltage switching (ZVS) for all switches in rated condition. Also, the commutation paths of the secondary side switches are provided for any switching case by adding a single capacitor; originally, a filter capacitor is used for the switching mode power supply input, and in this case, this filter capacitor also solves the primary-side commutation problem. The ZVS and solving the commutation problem increase the power efficiency and reliability. The proposed converter achieves high power quality with a simple control algorithm. A 1 kW/ 220 Vrms prototype is implemented to verify the theoretical analysis and performance of the proposed converter. The California energy commission efficiency is 97.3%, the total harmonic distortion (THD) of the input current is 2.45% at full load, and the THD of the output voltage is 1.07% at full load. BACKGROUND: The abductor hallucis (AbH) is a key muscle in the stability of the foot by supporting the medial longitudinal arch (MLA). Individuals with flat foot show a flattening of the MLA with a decreased selective AbH activity relative to the other foot extrinsic muscles during functional movement. OBJECTIVE: To examine the selective muscle activity of the abductor hallucis (AbH) during single-leg mini-squat (SMS) in subjects with flat foot and normal individuals. METHODS: Twenty-four healthy adults (13 men and 11 women) with flat or normal feet participated in this study. All subjects performed single minisquat (SMS), and data were collected using sEMG from the AbH, TA, and GCM during SMS. RESULTS: An inter-gender comparison of the EMG data revealed no difference. The EMG activity of the AbH in individuals with flat foot was significantly lower than that of individuals with normal feet. Moreover, the AbH/TA ratio in subjects with flat foot, , was significantly lower than that in subjects with normal feet. CONCLUSION: In a clinical setting, clinicians should focus on strengthening exercises of the AbH muscle due to reduced activation of the AbH muscle in individuals with flat foot. Prominin‐1 (Prom1) is a major cell surface marker of cancer stem cells, but its physiological functions in the liver have not been elucidated. We analyzed the levels of mRNA transcripts in serum‐starved primary WT (Prom1+/+) and KO (Prom1−/−) mouse hepatocytes using RNA sequencing (RNA‐seq) data, and found that CREB target genes were downregulated. This initial observation led us to determine that Prom1 deficiency inhibited cAMP response element‐binding protein (CREB) activation and gluconeogenesis, but not cyclic AMP (cAMP) accumulation, in glucagon‐, epinephrine‐, or forskolin‐treated liver tissues and primary hepatocytes, and mitigated glucagon‐induced hyperglycemia. Because Prom1 interacted with radixin, Prom1 deficiency prevented radixin from localizing to the plasma membrane. Moreover, systemic adenoviral knockdown of radixin inhibited CREB activation and gluconeogenesis in glucagon‐treated liver tissues and primary hepatocytes, and mitigated glucagon‐elicited hyperglycemia. Based on these results, we conclude that Prom1 regulates hepatic PKA signaling via radixin functioning as an A kinase‐anchored protein (AKAP). Van der Waals (vdW) layered materials are promising channel materials for next‐generation field‐effect transistors (FETs). However, in vdW layered‐material FETs, the Schottky barrier and tunnel barrier at the vdW interfaces significantly reduce the electron injection efficiencies at electrical contacts, thereby limiting the development of high‐speed and low‐power FETs. This study demonstrates that intercalated lithium (Li) ions at vdW interfaces lower the Schottky and tunnel barriers at the electrical contacts of multilayer tungsten diselenide FETs owing to the low potential energy of Li and the mild electron doping from intercalation, thus decreasing the resistance at the vdW interfaces and enhancing the current flow and field‐effect mobility. Compared with before‐Li intercalation, the multilayer WSe2 planar FET demonstrates ≈1000 times higher current (IDS = 17.8 μA) at VGS = 50 V, ≈110 times higher maximum field‐effect mobility (μFE = 97.67 cm2 V−1 s−1), and ≈1000 times higher on/off current ratio (on/off ratio = ≈107). Furthermore, in multilayer WSe2 vertical FETs with vdW heterostructures having a structural strength of approximately the same as the current density, the intercalated Li ions at the graphene/WSe2 vdW interfaces additionally improves the current density and the vertical mobility by ≈20 times (1.00 A cm−2) and 10 times (2.52 × 10−4 cm2 V−1 s−1), respectively. The feasibility of pretreatment involving soaking in aqueous ammonia (SAA) for the anaerobic digestion (AD) of eight different types of agricultural biomass of Ugandan origin was investigated. Moderate pretreatment temperatures of 60 and 90 °C were employed, and the NH3 concentration, solid-to-liquid ratio, and pretreatment time were fixed at 15.0% (w/w), 1:6, and 6 h, respectively. The delignification efficiencies of the SAA pretreatment ranged from 51.1 to 76.6%, and the maximum value was observed for maize bran pretreated at 90 °C. Biochemical methane potential experiments proved that the breaking of the complex bonds of lignin made fermentable sugars easily accessible to microorganisms. In all cases, the SAA pretreatment enhanced the methane potential of the eight types of Ugandan biomass compared with its untreated counterparts. The pretreated maize bran exhibited the highest methane yield of 291.5 mL CH4/g COD, which is 83.1% of the theoretical conversion. BACKGROUND: The abdominal draw-in maneuver (ADIM) has been suggested to increase trunk stabilizing muscle activity; however, no study has identified the effects of the ADIM on trunk muscles and trunk movements during lifting and lowering (L & L) tasks. OBJECTIVE: We examined the effects of the ADIM on transversus abdominis (TrA) thickness during standing, the activity of the TrA, internal oblique (IO) and multifidus (MF), and the kinematics of the lumbo-pelvic-hip complex during L & L tasks. METHODS: Fifteen male participants practiced expirations without and with the ADIM using ultrasound biofeedback. After training for each condition, the participants performed expirations with and without the ADIM in the standing position and during L & L tasks. TrA thickness during the standing position and muscular activities (EMG) of the bilateral TrA/IO and MF, and the peak angle of the lumbo-pelvic-hip complex in the sagittal plane during L & L tasks were measured. RESULTS: TrA thickness during standing was increased with expiration with ADIM compared to that without the ADIM (p < 0.05). When performing expiration with the ADIM, increased activity of the TrA/IO and MF was observed by EMG, as well as peak pelvic anterior tilt and hip flexion with decreased peak lumbar flexion during L & L tasks (p < 0.05). Objective: To evaluate the effectiveness of poloxamer‐based thermo‐sensitive sol–gel instillation, after transurethral resection of the prostate (TURP), for preventing urethral stricture. Patients and Methods: In all, 198 patients underwent TURP for benign prostatic hyperplasia. Recruited patients were randomly divided into two groups: groups A and B. Patients in Group A (100 patients, experimental group) received poloxamer‐based thermo‐sensitive sol–gel instillation and patients in the Group B (98 patients, control group) received lubricant instillation after TURP. Each patient was evaluated at 4 (V1), 12 (V2), and 24 weeks (V3) after TURP. The effectiveness of poloxamer‐based thermo‐sensitive sol–gel instillation was evaluated based on the International Prostate Symptom Score (IPSS), IPSS‐Quality of Life (QoL), Overactive bladder questionnaire (OAB‐q), maximum urinary flow rate (Qmax), post‐void residual urine volume (PVR), and cystoscopy. Results: Amongst the initial 198 participants, 80 patients in Group A and 83 in Group B completed the study. There were no significant differences in IPSS‐QoL and OAB‐q between the groups. However, Qmax was significantly different between groups A and B, at a mean (SD) of 18.92 (9.98) vs 15.58 (9.24) mL/s (P = 0.028) at 24 weeks after TURP. On cystoscopic examination, urethral stricture after TURP was found in two of the 80 patients in Group A and 10 of 83 in Group B (P = 0.023). Coastal areas are favorable for the establishment of industrial facilities from an economic perspective and provide valuable space for tourism and leisure activities. However, these areas have faced significant problems since the late 20th century due to indiscriminate development and accelerated erosion caused by global warming. To prevent erosion, various coastal structures such as submerged breakwaters, detached breakwaters and groins are used for coastal restoration projects. However, the coastal structures used in coastal restoration projects might cause secondary damage. This study performed a movable-bed hydraulic experiment using a gravity drainage method that installs drainage layers with different permeability in the beach to naturally drain groundwater into the sea, thus reducing coastal erosion while minimizing the risk of secondary damage. The push-off mechanism to generate forward movement in skating has been analyzed by using high-speed cameras and specially designed skates because it is closely related to skater performance. However, using high-speed cameras for such an investigation, it is hard to measure the three-dimensional push-off force, and a skate with strain gauges is difficult to implement in the real competitions. In this study, we provided a new method to evaluate the three-dimensional push-off angle in short-track speed skating based on motion analysis using a wearable motion analysis system with inertial measurement unit sensors to avoid using a special skate or specific equipment insert into the skate for measurement of push-off force. The estimated push-off angle based on motion analysis data was very close to that based on push-off force with a small root mean square difference less than 6% when using the lateral marker in the left leg and the medial marker in the right leg regardless of skating phase. These results indicated that the push-off angle estimation based on motion analysis data using a wearable motion capture system of inertial measurement unit sensors could be acceptable for realistic situations. The proposed method was shown to be feasible during short-track speed skating. Background: Aortic valve calcification quantification using cardiac computed tomography (CCT) is a reliable marker for aortic stenosis (AS) in patients with bicuspid aortic valve (BAV) disease.Purpose: To determine the association of Agatston aortic valve calcium score (AVCS) with morphological and hemodynamic characteristics of BAV and define cut-off AVCS for optimizing the grade of AS in patients with bicuspid AS.Material and Methods: This study included 161 BAV patients with AS regardless of aortic regurgitation who underwent transthoracic echocardiography and CCT. BAVs were classified according to orientation of cusps and presence of raphe. Associations of AVCS with characteristics of BAV morphology and functional variables were determined by linear regression analysis. Area under the receiver operating characteristic curve (AUC) was used to determine the cut-off AVCS greater than which the diagnosis of severe AS was optimized.Results: AVCS was significantly different according to sex ( P < 0.001), AS severity ( P < 0.001), type of valvular dysfunction ( P = 0.011), and orientation of cusps ( P = 0.028). Multiple linear regression showed that AVCS was significantly associated with sex (estimate = -0.583, P < 0.001) and AS severity (estimate = 0.817, P < 0.001). AVCS was a predictor for severe AS with AUC of 0.80 in both women ( P = 0.002) and men ( P < 0.001). Its cut-off value was 1423 Agatston unit (AU) in women and 2573 AU in men.Conclusions: In patients with bicuspid AS, AVCS was significantly higher in men and those with severe AS. We aimed at enhancing interaction in the polyimide/sepiolite hybrid films with the loading of sepiolite in the polyimide matrix ranged from 0 to 8 wt% via acid activating and polydopamine doping. First, sepiolite was treated with a 3M HCl solution under reflux conditions. Then the acid‐activated sepiolite was coated by mussel‐inspired polydopamine under alkaline conditions. The coated polydopamine was found to successfully enhance dispersibility of sepiolite in the polyimide matrix. The modification of the sepiolite in the hybrid films was investigated by Fourier transform infrared spectroscopy, wide‐angle X‐ray diffraction, field emission scanning electron microscopy, thermogravimetric analysis, and transmission electron microscope. This study proposes a bidirectional grid-connected single-power-conversion converter with low-input battery voltage. The proposed bidirectional converter consists of a bidirectional dc–dc converter and an unfolding bridge, and the power conversion stage only corresponds to a bidirectional dc–dc converter. The bidirectional dc-dc converter can perform bidirectional power conversion between the low input battery voltage and a rectified sine wave due to its step-up/down voltage regulation functions. The unfolding bridge unfolds the rectified sine wave into the grid voltage and provides a current path to the grid. The study also proposes a control algorithm to regulate the grid current through a single power-processing stage. The control algorithm is comprised of a feed-forward nominal voltage compensator and a repetitive control scheme. The feed-forward nominal voltage compensator presets the operating point to lighten the burden of the grid current control, and the repetitive controller provides precise control of the grid current. Thus, the proposed bidirectional grid-connected single-power-conversion converter results in high power quality and high efficiency. This paper proposes a high-efficiency two-stage three-level grid-connected photovoltaic (PV) inverter. The proposed two-stage inverter comprises a three-level step-up converter and a three-level inverter. The three-level step-up converter not only improves the power-conversion efficiency by lowering the voltage stress but also guarantees the balancing of the dc-link capacitor voltages using a simple control algorithm; it also enables the proposed inverter to satisfy the VDE 0126-1-1 standard of leakage current. The three-level inverter minimizes the overall power losses with zero reverse-recovery loss. Furthermore, it reduces harmonic distortion, the voltage ratings of the semiconductor device, and the electromagnetic interference by using a three-level circuit configuration; it also enables the use of small and low-cost filters. To control the grid current effectively, we have used a feed-forward nominal voltage compensator with a mode selector; this compensator improves the control environment by presetting the operating point. The proposed high-efficiency two-stage three-level grid-connected PV inverter overcomes the low efficiency problem of conventional two-stage inverters, and it provides high-power quality with maximum efficiency of 97.4%. Summary The regulation of M1/M2 polarization in liver macrophages is closely associated with the progression of nonalcoholic steatohepatitis (NASH); however, the mechanism involved in this process remains unclear. Here, we describe the orphan nuclear receptor retinoic-acid-related orphan receptor α (RORα) as a key regulator of M1/M2 polarization in hepatic residential Kupffer cells (KCs) and infiltrated monocyte-derived macrophages. RORα enhanced M2 polarization in KCs by inducing the kruppel-like factor 4. M2 polarization was defective in KCs and bone-marrow-derived macrophages of the myeloid-specific RORα null mice, and these mice were susceptible to HFD-induced NASH. We found that IL-10 played an important role in connecting the function of M2 KCs to lipid accumulation and apoptosis in hepatocytes. Importantly, M2 polarization was controlled by a RORα activator, JC1-40, which improved symptoms of NASH. This study proposes a monitoring chart for anomaly detection of non-linear process signals generated by semiconductor manufacturing processes. In these manufacturing processes, fault detection and classification (FDC) and statistical process control (SPC) have been established as fundamental techniques to improve production efficiency and yield. Non-linear process signals are collected through automatic sensing during each operation cycle of a manufacturing process. As these cyclic signals non-linearly vary on the process state, the usage of the prevalent SPC chart is limited. Therefore, we propose a more efficient monitoring chart considering non-linear and time-variant characteristics. Using the principal curve, a non-linear smoothing algorithm, we construct a time-variant centerline that represents the standard pattern of the process. Then, control limits are calculated with time-variant variances over the course of the process. To evaluate performance, the proposed method was applied to industrial data for chemical vapor deposition (CVD), a semiconductor manufacturing process. A high-efficiency bridgeless three-level power factor correction (PFC) rectifier is proposed. The circuit configuration of the proposed rectifier consists of four metal oxide semiconductor field-effect transistor (MOSFET) switches, and the reverse recovery problems of the switches are eliminated. Also, the proposed rectifier with three voltage levels reduces the power losses, harmonic components, voltage ratings, and electromagnetic interference. To control the grid current and the output voltage effectively, a feed-forward nominal voltage compensator with the mode selector is developed; by presetting the operating point of the grid voltage, this compensator improves the control environment. Thus, the proposed three-level PFC rectifier with developed control algorithm provides high power quality and high efficiency of 99.05%. Objective: This study investigated the morphological and functional characteristics of the aortic valve and the left ventricular (LV) systolic functional parameters and myocardial mass related to the severity of myocardial fibrosis (MF) in patients with severe aortic valve stenosis (AS). Materials and Methods: We retrospectively enrolled 81 patients (48 men; mean age: 59±12 years) with severe AS who underwent transthoracic echocardiography (TTE), cardiac computed tomography (CCT), and cardiovascular magnetic resonance (CMR) within 1 month and subsequent aortic valve surgery. Degree of MF was determined on delayed contrast-enhanced CMR with visual sub-segmental analysis-based quantification and was classified into three groups (no, mild, and severe) for identifying the differences in LV function and characteristics of the aortic valve. One-way ANOVA, Chi-square test or Fisher’s exact test were used to compare variables of the three groups. Univariate multinomial logistic regression analysis was performed to determine the association between the severity of MF and variables on imaging modalities. Results: Of 81 patients, 34 (42%) had MF (mild, n = 18; severe, n = 16). Aortic valve calcium volume score on CCT, aortic valve area, LV mass index, LV end-diastolic volume index on CMR, presence of mild aortic regurgitation (AR), transaortic mean pressure gradient, and peak velocity on TTE were significantly different among the three groups and were associated with severity of MF on a univariate multinomial logistic regression analysis. Aortic valve calcium grade was different (p = 0.008) among the three groups but not associated with severity of MF (p = 0.375). Conclusions: A multi-imaging approach shows that severe AS with MF is significantly associated with more severe calcific AS, higher LV end-diastolic volume, higher LV mass, and higher prevalence of mild AR. Trichoderma species are widely used for the commercial production of cellulolytic enzymes. In the present investigation, medium components were optimized using a central composite design and response surface methodology to produce endoglucanase (EG) from Trichoderma harzianum KUC1716. From the various medium components tested, cellulose, soy peptone, and thiamine HCl were selected as the optimal carbon, nitrogen, and vitamin sources, respectively. The highest EG (1.97 U/mL) production was obtained with 1.85% cellulose, 0.48% soy peptone, and 0.10% thiamine HCl. EG production in the optimized medium was 2.6 fold higher than in the unoptimized medium. In addition, the crude enzyme preparation from T. harzianum KUC1716 supplemented with β- glucosidase from Schizophyllum commune KUC9397 was used to hydrolyze various types of lignocellulosic materials and showed significant saccharification yields on all lignocellulosic materials, surpassing that of a commercial enzyme cocktail. It was verified that the crude enzyme preparation derived from T. harzianum KUC1716 could replace the commercial enzymes. Eight aminoacyl-tRNA synthetases (M, K, Q, D, R, I, EP and LARS) and three auxiliary proteins (AIMP1, 2 and 3) are known to form a multi-tRNA synthetase complex (MSC) in mammalian cells. We combined size exclusion chromatography (SEC) with reversed-phase liquid chromatography multiple reaction monitoring mass spectrometry (RPLC-MRM-MS) to characterize MSC components and free ARS proteins in human embryonic kidney (HEK 293T) cells. Crude cell extract and affinity-purified proteins were fractionated by SEC in non-denaturing state and ARSs were monitored in each fraction by MRM-MS. The eleven MSC components appeared mostly in earlier SEC fractions demonstrating their participation in complex formation. TARSL2 and AIMP2-DX2, despite their low abundance, were co-purified with KARS and detected in the SEC fractions, where MSC appeared. Moreover, other large complex-forming ARS proteins, such as VARS and FARS, were detected in earlier fractions. The MRM-MS results were further confirmed by western blot analysis. Our study demonstrates usefulness of combined SEC-MRM analysis for the characterization of protein complexes and in understanding the behavior of minor isoforms or variant proteins. Background: Mitral valve repair is favored over replacement due to superior outcomes. However, extensive infective endocarditis (IE) often has been considered unreconstructable. We retrospectively analyzed the mid-term outcomes of an individualized repair approach using a lifting annuloplasty strip.Methods: Between December 2007 and March 2014, 27 consecutive patients with acute single mitral valve IE (age 43 ± 16 years, 11 men) underwent lifting mitral annuloplasty (LMA) with a strip (Mitracon® strip, 28 mm in 4, 32 mm in 10, and 34 mm in 13). Blood culture was positive in 70 % (Streptococcus 10, Staphylococcus 4, HACEK 3, Enterococcus 1, Gram negative bacilli 1). One case (4 %) had a previously repaired mitral valve-the repair was redone. Via right thoracotomy (74 %) or median sternotomy (26 %), repair was performed by removal of vegetation and resection of infected tissue, the defect typically then being repaired using a bovine pericardial patch (81 %). Artificial chordae were formed in 5 patients. Nine (33 %) of them had posterior leaflet augmentation (PLA) to get sufficient coaptation height. Clinical and echocardiographic follow-up were performed.Results: Compared with preoperative ones, postoperative echocardiograms revealed decreases of left ventricular (LV) end-diastolic dimensions (57.2 ± 6.3 versus, 45.4 ± 6.2, or 44.8 ± 4.1 mm, all p < 0.01). The LV ejection fraction decreased immediately, but recovered (64.4 ± 9.6 % vs. 54.5 ± 9.8 %, or 65.2 ± 6.1 %, p = 0.002, p = 1.000, respectively). The latest follow-up echocardiograms (median 28 months) universally showed no or minimal regurgitation, with a preserved mitral valve opening area (2.27 ± 0.48 cm(2)). During the clinical follow-up (median, 54 months), one (3.7 %) death was observed (in-hospital, due to biliary sepsis and pneumonia). There was no reoperation or major cardiovascular event. The 5 year survival rate was 96.3 %.Conclusions: The repair technique of LMA and/or PLA in patients with IE achieved good structural and functional outcomes as well as an excellent 5 year survival rate. Multiple reaction monitoring (MRM) mass spectrometry has been successfully applied to monitor targeted proteins in biological specimens, raising the possibility that assays could be configured to measure all human proteins. We report the results of a pilot study designed to test the feasibility of a large-scale, international effort for MRM assay generation. We have configured, validated across three laboratories and made publicly available as a resource to the community 645 novel MRM assays representing 319 proteins expressed in human breast cancer. Assays were multiplexed in groups of >150 peptides and deployed to quantify endogenous analytes in a panel of breast cancer-related cell lines. The median assay precision was 5.4%, with high interlaboratory correlation (R2 > 0.96). Peptide measurements in breast cancer cell lines were able to discriminate among molecular subtypes and identify genome-driven changes in the cancer proteome. Background: Diabetes mellitus tends to have the greatest impact on the smaller vessels and contributes significantly to occlusive disease from the popliteal artery distally. Purpose: To evaluate the clinical outcomes after a balloon angioplasty with or without stent placement in diabetic patients with critical limb ischemia (CLI) by chronic total occlusion (CTO) limited to below-the-knee (BTK) arteries. Material and Methods: From August 2005, patients who presented CLI and CTO limited to the BTK arteries, and who underwent endovascular treatment, were included in this study. The primary endpoints evaluated were technical success, limb salvage, and primary patency. The secondary endpoints evaluated were 30-day access site, intervention site, and systemic complications. Patency and limb salvage were evaluated using the Kaplan-Meier method and compared using Fisher's exact test. Results: The BTK endovascular treatment (EVT) was performed on 64 limbs. Technical success rate was 93.8% and limb salvage rate was 90.6%. Three of four limbs with technical failure and three of 60 limbs with technical success underwent BTK amputation and the comparison of these rates were significantly different (75% vs. 5%, P = 0.002). Primary patency rates for the limbs were 75% and 59.1% at 6-month and 12-month follow-up, respectively. Minor complications disappeared through the follow-up periods and there was no 30-day complication or systemic adverse events for the treated vessel. Conclusion: Even though EVT for CLI in patients with diabetes and CTO in isolated BTK arteries does not have comparable primary patency, it can lead to a very high rate of limb salvage. This result can accentuate the importance of more blood flow to the foot by means of successful revascularization using EVT rather than long-term patency in CTO of isolated BTK arteries. This paper presents an asynchronous pipelined all-digital 10-b time-to-digital converter (TDC) with fine resolution, good linearity, and high throughput. Using a 1.5-b/stage pipeline architecture, an on-chip digital background calibration is implemented to correct residue subtraction error in the seven MSB stages. An asynchronous clocking scheme realizes pipeline operation for higher throughput. The TDC was implemented in standard 0.13-µm CMOS technology and has a maximum throughput of 300 MS/s and a resolution of 1.76 ps with a total conversion range of 1.8 ns. This paper describes the first implementation of the well-known cyclic ADC architecture into a time-to-digital converter. With an asynchronous clocking scheme, an all-digital 1.5b time-domain multiplying DAC (MDAC) is repetitively used for 8b conversion. The MDAC is based on a 2\times time amplifier with an offset-compensated gain calibration scheme. The proposed cyclic TDC, fabricated in a 0.13 \mu\m CMOS, shows a resolution of 1.25 ps with a total conversion range of \pm160 ps, the maximum operating frequency of 100 MHz, and a power consumption of 4.3 mW at 50 MHz. Background: With recent advances in post-operative care and surgical methods, the number of cardiovascular re-operations is increasing. We analyzed our institutional experience to evaluate the safety and efficacy of the approach methods for cardiac re-operations. Methods: Between September 2007 and December 2010, we performed 208 cardiac re-operations, defined as surgery which was not performed within a month from the previous operation or during the same hospitalization for the same disease and reviewed retrospectively. According to the surgical approach, we divided patients into two groups: median sternotomy group (S-group, n = 146), and thoracotomy group (T-group, n = 62). Results: There were no differences in sex or mean interval from the first surgery to re-operation between the two groups. Mean cardiopulmonary bypass, adhesion dissection time, bleeding control time, and operation time were significantly shorter in the T-group. The need for transfusion (p = 0.001) during adhesion dissection and the chest tube drainage (p < 0.001) were significantly lower in the T-group. There were 10 operative deaths in the S-group (6.8%) and 5 in the T-group (8.1%) (p = 0.757). Pneumonia was the most common cause of death in both groups. Post-operative bleeding did not result in death and there were no cases of wound infection in the T-group. Conclusions: Two approaches for repeated cardiac surgery were safe and effective in terms of mortality, wound infection, bleeding, operation time, adhesion dissecting time, and bleeding control time. We were able to obtain a good visual field and perform safe surgery by applying the thoracotomy method in selective patients for cardiovascular re-operation. [ABSTRACT FROM AUTHOR]

Organic/inorganic hybrid coating materials were synthesized using acrylate end-capped polyester, 1,6-hexanediolacrylate, tetraethoxysilane (TEOS), and 3-trimethoxysilylpropylmethacrylate (TMSPM). The hybrid materials were cast onto a polycarbonate (PC) substrate and cured by UV irradiation to give a hybrid film with covalent linkage between the inorganic and the organic networks. The coating layer was characterized by FT-IR and 29Si-NMR, and pencil hardness and oxygen permeation rate of coated films were investigated. The pencil hardness of all samples examined in this study was higher than 1H, whereas that of uncoated PC substrate was 6B. The hardness enhancement after coating may due to incorporation of organic acrylate resin. The oxygen permeability coefficient of the film coated with hybrid material on 3-aminopropyltriethoxysilane (APTEOS) pretreated polycarbonate substrate was 1.67×10−3 GPU, the lowest value in this work, whereas that of uncoated PC substrate was 8.07×10−3 GPU. The lower oxygen permeation rates of these films are attributed to the good adhesion between organic/inorganic hybrid coating layer and PC substrate and a dense structure induced by an increase of network density. In this paper, we develop a low-cost manufacturing technology for RF substrate and a high-performance process technology for integrated passive devices by electrochemically forming thick oxide on Si wafer and processing Cu thick metal and BCB. Porcine heart xenotransplantation is a potential treatment for patients with end-stage heart failure. To understand molecular mechanisms of graft rejection after heart transplantation, we transplanted a 31-day-old alpha-1,3-galactosyltransferase knockout (GTKO) porcine heart to a five-year-old cynomolgus monkey. Histological and transcriptome analyses were conducted on xenografted cardiac tissue at rejection (nine days after transplantation). The recipient monkey's blood parameters were analyzed on days −7, −3, 1, 4, and 7. Validation was conducted by quantitative real-time PCR (qPCR) with selected genes. A non-transplanted GTKO porcine heart from an age-matched litter was used as a control. The recipient monkey showed systemic inflammatory responses, and the rejected cardiac graft indicated myocardial infarction and cardiac fibrosis. The transplanted heart exhibited a total of 3748 differentially expressed genes compared to the non-transplanted heart transcriptome, with 2443 upregulated and 1305 downregulated genes. Key biological pathways involved at the terminal stage of graft rejection were cardiomyopathies, extracellular interactions, and ion channel activities. The results of qPCR evaluation were in agreement with the transcriptome data. Transcriptome analysis of porcine cardiac tissue at graft rejection reveals dysregulation of the key molecules and signaling pathways, which play relevant roles on structural and functional integrities of the heart. The present paper proposes a direct space vector modulation and novel balance algorithm for easy software application of three-level converters which operate in three-phase. In the case of the conventional space vector modulation, to get the on-state times of the switches, the dwell times of the three nearest stationary vectors, which are obtained after sector and region selection algorithms, should be rearranged. These processes, therefore, contain diverse conditional statements and complicated calculations such as inverse trigonometric functions and square roots. However, the burden of the software application of the proposed algorithm is greatly reduced by not using the sector selection algorithm, the region selection algorithm, and the on-state time allocation process as the proposed modulation can directly control the switch on-state time. In a three-level topology, it is required to balance top and bottom capacitor voltages because the DC-link voltage is composed of two capacitor voltages; the unbalanced voltage of each DC-link capacitor causes the overvoltage of the switching devices. Thus, the DC-link voltage balancing algorithm is proposed, and it is also very simple and effective without additional circuits because it controls the switch on-state time directly as well. The surface area and pore size distribution of Eucalyptus samples that were pretreated by different methods were determined by the Brunauer–Emmett–Teller (BET) technique. Three methods were applied to prepare cellulosic biomass samples for the BET measurements, air, freeze, and critical point drying (CPD). The air and freeze drying caused a severe collapse of the biomass pore structures, but the CPD effectively preserved the biomass morphology. The surface area of the CPD prepared Eucalyptus samples were determined to be 58–161 m2/g, whereas the air and freeze dried samples were 0.5–1.3 and 1.0–2.4 m2/g, respectively. The average pore diameter of the CPD prepared Eucalyptus samples were 61–70 Å. The CPD preserved the Eucalyptus sample morphology by replacing water with a non-polar solvent, CO2 fluid, which prevented hydrogen bond reformation in the cellulose. [ABSTRACT FROM AUTHOR]

In this study, the chemical pretreatment method using ethanol organosolv with hydrogen peroxide was investigated to improve enzymatic saccharification of corn stover. The pretreatment method using ethanol with hydrogen peroxide in a flow-through reaction was proposed to lower the reaction severity such as the pretreatment temperature. With the same reaction time, the pretreatment process using organosolv (30 wt.% ethanol) containing 1 wt.% hydrogen peroxide at 150 °C resulted in a similar conversion yield as the result of the alkali pretreatment method using 15 wt.% aqueous ammonia at 170 °C. When corn stover was pretreated with 30 wt.% ethanol solution containing 5 wt.% hydrogen peroxide, a glucose conversion yield of 69.7 wt.% and glucose production of 23.8 g were achieved.
