In this work, a model for anisotropic interactions between proteins and cellular membranes is proposed for large-scale continuum simulations. The framework of the model is based on dynamic density functional theory, which provides a formalism to describe the lipid densities within the membrane as continuum fields while still maintaining the fidelity of the underlying molecular interactions. Within this framework, we extend recent results to include the anisotropic effects of protein–lipid interactions. As applications, we consider two membrane proteins of biological interest: a RAS–RAF complex tethered to the membrane and a membrane embedded G protein-coupled receptor. A strong qualitative and quantitative agreement is found between the numerical results and the corresponding molecular dynamics simulations. Combining the scope of continuum level simulations with the details from molecular level particle simulations enables research into protein–membrane behaviors at a more biologically relevant scale, which crucially can also be accessed via experiment. Seafood is an important source for meeting future global nutrient demands. However, it also contributes disproportionately to the radiological ingestion dose of more than five billion world consumers– up to ∼70%–80% of the total-foods dose in some countries. Although numerous studies report seafood doses in specific populations, there is still no comprehensive evaluation answering basic questions such as "what is the ingestion dose to the average global seafood consumer?" Analysis of 238 worldwide seafood dose estimates suggests that typical adult consumers receive from 0.13 to 0.21 mSv, with a likely best estimate of 0.15 mSv per annual seafood intake. Those consuming large amounts of seafood, particularly bivalves, may experience ingestion doses exceeding 1 mSv per annual intake, surpassing other routine background dose sources. The published studies suggest that doses of 3 mSv or greater are surpassed in about 150 million adult seafood consumers worldwide. Almost all this dose comes from the natural radionuclides that are prevalent in marine systems–especially 210Po. While trace levels of anthropogenic radionuclides are ubiquitous in seafoods (e.g.,137Cs and 239Pu), the added dose from these is typically orders of magnitude lower. Even following the large-scale releases from the Fukushima accident, with food safety controls in place, the additional dose to consumers in Japan was small relative to routine dose from natural background radionuclides. However, the worldwide seafood dose estimates span seven orders of magnitude, indicating a need for an assessment that integrates global seafood radionuclide data as well as incorporating changes in seafood consumption and production patterns. The thymus is a primary lymphoid organ critical for the development of mature T cells from hematopoietic progenitors. A highly structured organ, the thymus contains distinct regions, precise cytoarchitecture, and molecular signals tightly regulating thymopoiesis. Although the above are well-understood, the structural and functional implications of thymic innervation are largely neglected. In general, neural regulation has become increasingly identified as a critical component of immune cell development and function. The central nervous system (CNS) in the brain coordinates these immunological responses both by direct innervation through peripheral nerves and by neuroendocrine signaling. Yet how these signals, particularly direct neural innervation, may regulate the thymus biology is unclear and understudied. In this review, we highlight historical and current data demonstrating direct neural input to the thymus and assess current evidence of the neural regulation of thymopoiesis. We further discuss the current knowledge gaps and summarize recent advances in techniques that could be used to study how nerves regulate the thymic microenvironment. [ABSTRACT FROM AUTHOR]

Antarctic microbial mats, with their significant biodiversity and key role in biogeochemical cycling, were the focus of our study. We employed a metagenomic approach to analyze 14 microbial mats from meltwater streams of western Antarctica, covering the Maritime, Peninsula, and Dry Valleys regions. Our findings revealed that the taxonomic compositional level of the microbial mat communities is characterized by similar bacterial groups, with diatoms being the main distinguishing factor between the rapidly warming Maritime Antarctica and the other mats. Bacteria were found to be the predominant component of all microbial mats (>90%), followed by Eukarya (>3%), Archaea (<1%), and Viruses (<0.1%). The average abundance of the main phyla composing Antarctic microbial mats included Bacteroidota (35%), Pseudomonadota (29%), Cyanobacteriota (19%), Verrucomicrobiota (3%), Bacillariophyta (2%), Planctomycetota (2%), Acidobacteriota (2%), Actinomycetota (2%), Bacillota (1%), and Chloroflexota (1%). We also identified some microeukaryotes that could play essential roles in the functioning of Antarctic microbial mats. Background: Minimally invasive liver surgery (MILS) is superior to open surgery when considering decreased blood loss, fewer complications, shorter hospital stay, and similar or improved oncologic outcomes. However, operative limitations in laparoscopic hepatectomy have curved its applicability and momentum of complex minimally invasive liver surgery. Transitioning to robotic hepatectomy may bridge this complexity gap. Methods: Retrospective cohort study conducted on comparable hepatectomies (open, laparoscopic, robotic) for benign or malignant diseases at Weill Cornell by three surgeons from 2017 to 2023. Case volume and Iwate difficulty scoring were examined over time by surgical approach. Outcome associations (operative time, estimated blood loss, length of stay, 90-day complications, open conversion, and resection margin) were analyzed using generalized estimating equations to account for the hierarchical data structure of different surgeons and controlled for clinical covariates. Results: Among 353 hepatectomies, 112 were open (OH), 107 were laparoscopic (LH), and 134 were robotic (RH). OH patients were more likely to have malignant pathology (83% vs. LH 69%, RH 57%) and less likely to have cirrhosis (6% vs. LH 6%, RH 14%). OH and RH had similar case complexity (Median Iwate: OH 7 vs. RH 7). After adjustments, LH and RH had 39% and 43% shorter median lengths of stay, respectively, and 89% and 62% lower odds of complications compared to OH. RH had 87% lower odds of conversion to OH compared to LH. The odds of R0 resection were similar between LH, RH, and OH. These results remained consistent in high difficulty cases (Iwate 7–12). Over the study period, RH usage increased from 36 to 68%, while LH decreased from 39 to 9%. By 2023, RH was predominantly used over OH (74% vs.26%). Conclusion: The transition from laparoscopic to all-robotic approach resulted in increased case volume and complexity in MILS, largely improving perioperative outcomes in hepatectomy. Background: The COVID-19 pandemic not only disproportionately impacted the health of immigrants in the US, but led to financial disparities as well. However, few studies to date capture the experience of asylum-seeking and refugee populations. Methods: We analyzed semi-structured interviews conducted in fall 2021 with immigrants (primarily asylum-seekers and refugees from countries in Central and East Africa) and key informants, and conducted focus group discussions in spring 2023 with employees of a non-profit supporting the financial needs of asylum-seekers and refugees in Portland, ME. Results: We identified several ways in which the COVID-19 pandemic negatively impacted the financial stability of asylum-seekers and refugees, including: job loss, work permit backlog, lack of sick leave benefits, childcare expenses and related job strain, rising cost of basic goods, issues of housing availability/affordability, and healthcare expenses. Conclusion: Policy changes that address structural factors and support financial stability among asylum-seekers and refugees and other low-income populations could help mitigate the impact of future public health crises and ultimately improve health equity. Finding scattered resources is a challenge for marine predators, their search strategies shaping in turn their response to global changes. Three types of search strategies are generally considered: random opportunistic search, reliance on conspicuous social cues or long-term individual fidelity to predictably productive areas. More complex strategies, namely intermediate, composite and/or those varying across time and environmental conditions are likely to prevail but are rarely explored. We investigated memory strategies in Cape gannets, a marine predator heavily relying on social cues, and closely related to the northern gannet which shows long-term individual foraging fidelity. Repeat GPS tracks within seasons revealed that two different individuals leaving the colony more than 3 days apart showed random chances to forage in a similar direction, whereas within-individuals, birds persisted in a same direction for longer timescales, reaching random levels only after 10 days, or 9 consecutive trips. This strategy of transient individual preferences within a breeding season was observed in most individuals every year. These results suggest a consistent and complex search strategy mixing social cues and private information updated every few days. The high temperature acoustic loss and elastic moduli of the Ni-based superalloys RR1000, Waspaloy, and Allvac® 718Plus™ were investigated using resonant ultrasound spectroscopy. An exponential increase in acoustic loss was seen in all alloys, which was viscoelastic in character with an activation energy of ∼ 200–400 kJ.mol−1, similar to that of the self-diffusion of Ni. The magnitude of the loss was found to be proportional to d−0.85, where d is the grain size. Deviations from the high temperature background were seen in Waspaloy and 718Plus around their γ′ solvus temperatures, attributed to increased diffusion during γ′ dissolution. Determining migratory connectivity is essential to understanding population dynamics and managing and conserving migratory species. Gallinago delicata (Wilson's Snipe) is a widely hunted wetland bird distributed throughout North America and parts of South America with little known regarding migratory strategy, connectivity, or population trends. Here, we used stable hydrogen isotope analysis of feathers to identify breeding areas and determine the degree of migratory mixing of 323 G. delicata harvested in Florida during the nonbreeding season. Our results demonstrate a high degree of migratory mixing between breeding and nonbreeding seasons and provide evidence that breeding birds from throughout Canada and the northwestern United States migrate to Florida. We also found evidence of long-distance migration across both latitudinal and longitudinal bands, with 104 individuals traveling from breeding grounds in western Canada or Alaska to nonbreeding habitat in Florida; the farthest-traveling individual likely migrated at least 4,270 km. Our findings highlight the crucial role of active and coordinated monitoring and management of the widely harvested G. delicata , which as long-distance migrants are disproportionately vulnerable to global environmental change. Objective: This study measured the rate of college student mental health concerns and mental health service utilization. The roles of mental health and seeking treatment regarding anticipated enrollment were explored. Methods: One thousand eight hundred thirty-one randomly selected students participated in this online survey. Results: Most students reported clinically significant symptoms (88.3%), and many sought treatment in the past year (28.8%). Most students had favorable attitudes toward telemental health. Barriers to seeking care included a preference for self-management of symptoms (68.8%) and limited time (43.3%). Mental health was the most commonly reported reason for anticipating reduced enrollment (ps <.001), and these individuals were more likely to seek treatment. Conclusions: Past and anticipated use of mental health treatment likely exceeds on-campus capacity. Student mental health and retention are linked, and treatment may support retention. Purpose: To explore whether continuous positive airway pressure (CPAP) adherence among adult patients with obstructive sleep apnea (OSA) and posttraumatic stress disorder (PTSD) affects the PTSD Checklist (PCL), a validated tool that measures severity of PTSD symptoms. Method: Studies focusing on PTSD and OSA were searched in PubMed and CINAHL databases. Inclusion and exclusion criteria were applied, decreasing the article yield to nine. All articles chosen focused on the adult population and were conducted in North America. Results: The literature review yielded eight observational cohort studies and one descriptive study. Seven studies were conducted with Veterans and all studies used the PCL tool. Three themes were identified: (a) Trauma and OSA, (b) OSA and PTSD Symptoms, and (c) CPAP Therapy and PTSD Symptoms. An area of further study is assessing how CPAP adherence might affect improvement in PTSD symptoms and how adherence can be improved. Conclusion: OSA is correlated with more severe PTSD symptoms, but CPAP use is associated with improvement in PTSD symptoms. The literature reviewed found PCL scores improved up to 10 points within 3 months of CPAP use. These findings highlight the importance of collaboration between mental health and sleep medicine professionals and opportunity for further study in this area. Mental health has been shown to impact rheumatoid arthritis (RA) outcomes and is associated with self-management behaviors. The extent to which mental health impacts outcomes via different self-management behaviours has not been thoroughly investigated. Adult RA patients who were starting a new medication or dosage were recruited to a prospective cohort with follow-ups at 3 and 12-months covering clinical and patient-reported outcomes. The longitudinal relationships between mental health, self-management behaviors (diet, physical activity, sleep, smoking, alcohol, and medication nonadherence), disease outcome, and function were assessed. Self-management behaviors were considered mediators of mental health at baseline on outcomes at 3 and 12 months. Depression did not worsen the odds of EULAR response for the total PHQ at 3 months (OR = 0.96, p = 0.36) or 12 months (OR = 0.99, p = 0.99) nor for the categorical PHQ at 3 months (OR = 0.64, p = 0.34) or 12 months (OR = 0.67, p = 0.44). Anxiety also did not worsen the odds of EULAR response for the total GAD at 3 months (OR = 0.98, p = 0.76) or 12 months (OR1.04, 0.53) nor for the categorical GAD at 3 months (OR = 0.99, p = 0.99) or 12 months (OR = 0.94, p = 0.75). However, depression was associated with the DAS-28 at 3 months (b = 0.22, p = 0.04). Among the self-management behaviors, insomnia was found to be a significant mediator between depression and the WSAS (b = 0.08, p = 0.03) as well as anxiety and the WSAS (b = 0.07, p = 0.03). Alcohol was also a significant mediator between depression and the DAS-28 (b = 0.21, p = 0.04). Mental health was associated with worse quality of life and disease outcomes, but not EULAR response. The extended endoscopic endonasal approach (EEEA) provides direct access to the tumor with minimal manipulation of the perforating arteries from the circle of Willis, avoiding brain retraction. EEEA allows a clear view of the hypothalamic wall and tumor interface, preventing hypothalamic injury. Background: Seriously ill older adults are admitted for post-acute care in skilled nursing facilities (SNFs) for curative, rehabilitative treatments, yet experience high rates of re-hospitalization, and death. The primary palliative care in post-acute care (PPC-PAC) intervention is an evidence-based approach designed to help people with serious illness align treatment plans with goals of care, optimize quality of life, and improve satisfaction with their care. Objectives: To conduct a preliminary study and evaluate the feasibility of implementing the PPC-PAC intervention in the post-acute care SNF setting. Design: Two-group, multisite feasibility pilot pragmatic clinical trial with a non-equivalent design. Measurements: Primary outcome measures—eligibility, enrollment, and data collection rates; consultation satisfaction; and fidelity. Effectiveness outcome measure—quality of life using the Palliative Outcomes Scale version 2. Results: Close to 70% of those who were eligible and approached by the study team (45/65) enrolled in the trial throughout 12 SNFs in the Northeast and Mid-Atlantic United States. Thirty-five were enrolled from intervention sites; 10 were enrolled from control sites (usual care). Most participants (80%) expressed general satisfaction with PPC-PAC, and 90% of clinicians implemented the PPC-PAC intervention as intended. At 21 days follow-up, there were no significant differences in effectiveness outcomes. Conclusion: Implementation of the PPC-PAC intervention proved to be feasible and acceptable among older adults and clinicians. Future research should focus on testing the effectiveness of PPC-PAC and explore strategies for optimal intervention implementation and SNF staff engagement in the post-acute care setting. This study assessed the morbidity risk for respiratory (RDs) and cardiovascular diseases (CVDs) from the exposure to black carbon (BC), p-SO42−, and p-NO3− measured in PM2.5 collected from Jeddah. The 24 h PM2.5 sampling was done at three sites for 6 weeks quarterly from 8 April 2013 to 18 February 2014, with concurrent hospital data collection. Though the data were collected more than a decade ago, the results are still applicable to the current middle east region and beyond. The relative risk (RR) for CVDs and RDs was computed in a single pollutant GLM time-series model using moving averages (MAs) and presented with 95% confidence intervals (CI). Overall, the morbidity risk was highest in outpatients. The risk of RDs was highest in younger age groups with age group 0–14 years being the most vulnerable, while the risk of CVDs was highest in age groups above 30 years. BC: The risk of RDs was highest in outpatient females of 0–14 years at all MAs; 1.483 (1.035, 2.123) to 2.501 (1.522, 4.112), and males of 31–45 years; 1.758 (1.134, 2.725) to 1.895 (1.142, 3.145) at MAs 5–7. The risk of CVDs was highest in age group 61–75 years but with a marginal statistical significance. p-SO42−: The risk of RDs was highest in males of 31–45 years; 1.173 (1.066, 1.291) to 1.256 (1.126, 1.400) at MAs_3–7. The risk of CVDs was also highest in age group 31–45 years; 1.078 (1.004, 1.157) to 1.090 (1.010, 1.177), with higher risk estimates in males; 1.084 (0.986, 1.191) to 1.120 (1.013, 1.239) at MAs 4–7. p-NO3−: No significant risk was observed for RD morbidity. The overall risk of CVDs was significant in outpatients with an age group of 61–75 years at all MAs; 1.204 (1.014, 1.429) to 1.316 (1.096, 1.579); females, 1.221 (0.987, 1.511) to 1.428 (1.123, 1.816); and males, 1.205 (1.022, 1.421) to 1.258 (1.052, 1.505). This is the first study to assess the cardiopulmonary risk from BC, p-SO42−, and p-NO3− exposure in Jeddah. A high morbidity risk was observed in all age groups, accentuating the effects of elevated air pollution in Saudi Arabia and beyond. BBT-059 is a long-acting PEGylated interleukin-11 analog that has been shown to have hematopoiesis-promoting and anti-apoptotic attributes, and is being studied as a radiation countermeasure for the hematopoietic acute radiation syndrome (H-ARS). This potential countermeasure has been demonstrated to enhance survival in irradiated mice. To investigate the toxicity and safety profile of this agent, 14 nonhuman primates (NHPs, rhesus macaques) were administered two different doses of BBT-059 subcutaneously 24 h after 4 Gy total-body irradiation and were monitored for the next 60 days postirradiation. Blood samples were investigated for the pharmacokinetics and pharmacodynamics of this agent and its effects on complete blood counts, cytokines, vital signs, and metabolomics. No adverse health effects were observed in either treatment group. Radiation-induced metabolomic dysregulation was observed in both treatment groups, and BBT-059 afforded some short-term radiomitigation. A few pathways were commonly dysregulated by radiation exposure including steroid hormone biosynthesis pathways, fatty acid activation, and glycerophospholipid metabolism. Notably, radiation-induced dysregulation to the linoleate metabolism pathway was significantly mitigated by either dose of BBT-059. In brief, this study suggests that BBT-059 has a good safety profile in irradiated NHPs and that its development as a medical countermeasure for U.S. Food and Drug Administration approval for human use should be continued. Rationale: Both the paraventricular nucleus of the thalamus (PVT) and the neuropeptide, pituitary adenylate cyclase-activating polypeptide (PACAP), are thought to be involved in food intake. Importantly, PACAP is expressed in cells of the PVT. Objectives: To determine if PACAP in cells of the PVT might mediate some of the involvement of the PVT with palatable food intake. Methods: In male and female C57BL/6 J mice and PACAP-Cre transgenic mice on a C57BL/6 J background, limited access to Milk Chocolate Ensure Plus® was used to establish a model of binge-type eating. Next, using quantitative real-time PCR, gene expression of PACAP in the PVT was measured in relation to this binge-type eating. Finally, using chemogenetics in PACAP-Cre transgenic mice, the effect of activation of PVT PACAP+ cells on binge-type eating was determined. Results: Males and females both engaged in binge-type eating with Ensure, although females engaged in this behavior to a greater degree than males. While females also had a higher baseline level of PVT PACAP mRNA than males, only males showed an increase in levels of PACAP after a history of exposure to Ensure, and only males showed a reduction in levels of PACAP immediately prior to a binge session. Conversely, activation of PACAP+ cells in the PVT reduced binge-type eating of Ensure, specifically in male mice. Conclusions: The present findings indicate that PVT PACAP+ cells influence and are influenced by binge-type eating. Purpose: Parent-Child Interaction Therapy (PCIT) is an empirically supported behavioral parenting program for disruptive behavior and has been shown to also be effective for children with autism spectrum disorder (ASD). Telehealth delivery of PCIT (Tele-PCIT) is also supported, but no trials have focused on children with ASD. The purpose of the study was to examine the initial efficacy of a time limited version of Tele-PCIT within an ASD sample. Methods: Participants included parents of 20 children (ages 2–6) with ASD who received 10 sessions of Tele-PCIT. Parents reported on their parenting stress, parenting practices, and child behavior. A play observation was coded for parent use of treatment skills and for child compliance. Results: 80% of participants completed treatment (n = 16) and results revealed significant improvements across parenting and child outcomes. Parents reported decreases in parenting stress from pre-to-post-treatment, which were maintained at a 3-month follow-up, along with decreases in negative parenting practices (i.e., Laxness and Overactivity) from pre-to-post treatment that were also maintained at follow-up. Significant increases in parent use of positive parenting skills (i.e., "Do" Skills) during child-led play and decreases in negative parenting skills (i.e., "Don't" Skills) were observed. Results also revealed significant reductions in parent rated child externalizing behavior problems from pre-to-post-treatment that were also maintained at follow-up. Conclusions: Results of the current study support the initial efficacy of Tele-PCIT for treating disruptive behavior in young children with ASD. Egg donation is a procedure that is powerfully advertised as a beneficial experience with limited mention of the associated risks. Egg donor recruitment advertisements target young and financially insecure women and can serve as a catalyst for interest in egg donation. In the absence of explicit egg donation advertisement regulations and without counterbalancing information from other sources, potential donors may not be able to recognize how advertisements can be misleading. In this paper, we enumerate two types of misleading information in egg donor recruitment advertisements that can influence the decision to donate that might not exist otherwise: (1) the focus on financial benefits and (2) the minimization of health risks. We draw upon existing Federal Drug Administration and Federal Trade Commission regulations that govern other types of medical advertisements and demonstrate their suitability to govern egg donor recruitment advertisements. We conclude that similar regulations could address the regulatory gap in which egg donation advertisements are situated by ensuring that information is balanced and not overly persuasive, thus better protecting the autonomy of egg donors. Study Selection: Two investigators independently identified studies on risk classification and treatment choice after GC testing for patients with localized PCa considering first-line treatment. Data Extraction: Relevant data extracted by 1 researcher and overread by a second. Risk of bias (ROB) was assessed in duplicate. Data Synthesis: Ten studies reported risk reclassification after GC testing. In low ROB observational studies, very low- or low-risk patients with PCa were more likely to have their risk levels classified as the same or lower (GPS, 100% to 88.1%; Decipher, 87.2% to 82.9%; Prolaris, 76.9%). However, 1 randomized trial found that GC testing with GPS reclassified 34.5% of very low-risk and 29.4% of low-risk patients to a higher risk category. Twelve observational studies indicated that treatment decisions after GC testing either remained unchanged or slightly favored active surveillance. In contrast, analyses from a single randomized trial found fewer choices for active surveillance after GPS testing. Limitations: Heterogeneity in screening patterns, risk-determination cutoffs, pathology, and clinical practices. Studies on treatment choice were moderate to high ROB. Conclusion: Although GC tests do not consistently influence risk classification or treatment decisions, the differences observed between observational and randomized studies highlight a need for well-designed trials to explore the role of GC tests in patients with newly diagnosed PCa considering first-line treatment. Globally, youth mental health services are evolving, with Australia's headspace services presented as a leading exemplar. headspace services were designed as enhanced primary care-based entities and were expected to collaborate with local acute, and specialist clinical and psychosocial services. The lack of large-scale health services trials necessitates understanding their impact through systematic monitoring and evaluation. This paper compares data from differing sources that describe the demographic and clinical features, and functional outcomes, of young people presenting to various headspace services. headspace National reports that care is provided largely to youth with transient distress, minimal clinical disorders, suicidality or comorbidities and limited functional impairment. Almost 50% of clients are reported to have no significant psychological symptoms or risk factors, and less than 30% to have a clinical disorder. Consequently, 36% receive only a single session of care and the median number of clinical sessions provided is three. By contrast, empirically derived estimates, utilising data from an academic centre and its affiliated centres, other independent agencies and more refined analyses of headspace national data variously portray 50–60% of youth as presenting with at least moderate clinical complexity (including at least 20% with high complexity), and with another 27% requiring active clinical intervention. Together, these data suggest approximately 75% of attendees require substantive clinical care. Clinical outcomes data from all sources indicate limited impacts on functional outcomes, with less than a third achieving significant improvement. These data support the original intent of headspace services to focus on equitable access to multidimensional and clinical assessment, evidence-based early interventions for early stages of major anxiety, mood or psychotic disorders. As demand for youth services continues to rise, there is an urgent need to reconfigure our national youth service networks to address the unmet clinical and psychosocial needs of youth presenting in the early stages of major mental disorders. Self-directed educator professional learning is commonplace, and such activities increasingly span multiple digital spaces and formats, and blur boundaries between online and offline. In this exploratory research, we analyze the case of the #CharlasEducativas, a dynamic professional learning ecosystem that began in 2020 and is based in Spain. We describe the platforms, modalities, and activities that comprised the #CharlasEducativas from 2020–2023, and how these different elements combine to create a multiplatform learning ecosystem. Relying upon multiple data sources, we also analyze the topics and content associated with various components of this unique ecosystem, and share participant perceptions of the #CharlasEducativas. Although the ecosystem was first developed relying mostly on YouTube and X/Twitter, the #CharlasEducativas have evolved over time to include additional platforms, and even in-person events, with different spaces functioning in overlapping and distinct ways. These spaces have been employed in synchronous and asynchronous ways, using text, images, voice, and visuals to discuss and share information on a wide array of education topics. Many participants reported perceiving the #CharlasEducativas as a space of learning and community building, and credited this learning and community with sparking reflection upon and changes in their own teaching practices. We discuss how the #CharlasEducativas reflect opportunities and challenges of contemporary educator professional learning in the context of ubiquitous social media platforms. Background: Preclinical studies and anecdotal case reports support the potential therapeutic benefit of low-dose oral ketamine as a treatment of clinical symptoms in Rett syndrome (RTT); however, no controlled studies have been conducted in RTT to evaluate safety, tolerability and efficacy. Design: This was a sequentially initiated, dose-escalating cohort, placebo-controlled, double blind, randomized sequence, cross-over study of oral ketamine in 6–12-year-old girls with RTT to evaluate short-term safety and tolerability and explore efficacy. Methods: Participants were randomized to either five days treatment with oral ketamine or matched placebo, followed by a nine-day wash-out period and then crossed-over to the opposite treatment. Ketamine was dosed twice daily at 0.75 mg/kg/dose (Cohort 1) or 1.5 mg/kg/dose (Cohort 2). An independent safety monitoring committee evaluated safety and approved proceeding to the next dose cohort. Caregivers, participants, outcome assessors, and study staff except pharmacists were blinded to allocation. The primary endpoint was safety and tolerability. Exploratory efficacy endpoints included change in clinician- and caregiver-rated measures of RTT features, brain activity on electroencephalography, and wearable biosensors to measure respiration, heart rate, sleep, and activity. Results: Twenty-three participants enrolled (11 in Cohort 1, 12 in Cohort 2) from 3/12/2019–11/22/2021. One participant was excluded from analysis due to not meeting inclusion criteria on blinded review prior to analysis. One participant was withdrawn from the study due to an adverse event (vomiting) after the first dose of ketamine. Although planned for four dose cohorts, the trial was stopped after Cohort 2 due to enrollment challenges associated with the COVID-19 pandemic. Ketamine was safe and tolerated in both cohorts, with 1 related treatment emergent adverse event of vomiting. No difference was observed in efficacy between ketamine and placebo. Electroencephalography showed the expected increase in high frequency power with ketamine. Conclusions: Short-term, low-dose oral ketamine was safe and well tolerated in girls with RTT. No clinical efficacy of ketamine in treating symptoms of RTT was observed with 5 days of treatment, despite electroencephalography evidence of ketamine target engagement during the first dose. The Sc2.0 global consortium to design and construct a synthetic genome based on the Saccharomyces cerevisiae genome commenced in 2006, comprising 16 synthetic chromosomes and a new-to-nature tRNA neochromosome. In this paper we describe assembly and debugging of the 902,994-bp synthetic Saccharomyces cerevisiae chromosome synXVI of the Sc2.0 project. Application of the CRISPR D-BUGS protocol identified defective loci, which were modified to improve sporulation and recover wild-type like growth when grown on glycerol as a sole carbon source when grown at 37˚C. LoxPsym sites inserted downstream of dubious open reading frames impacted the 5' UTR of genes required for optimal growth and were identified as a systematic cause of defective growth. Based on lessons learned from analysis of Sc2.0 defects and synXVI, an in-silico redesign of the synXVI chromosome was performed, which can be used as a blueprint for future synthetic yeast genome designs. The in-silico redesign of synXVI includes reduced PCR tag frequency, modified chunk and megachunk termini, and adjustments to allocation of loxPsym sites and TAA stop codons to dubious ORFs. This redesign provides a roadmap into applications of Sc2.0 strategies in non-yeast organisms. The Sc2.0 project involved synthesis and debugging of 16 chromosomes, and a tRNA neochromosome. We provide new evidence on marital status and family outcomes of transgender and other gender minority populations in the United States using more than fifteen thousand non-cisgender adults in the Census Bureau’s 2021–2023 Household Pulse, the only nationally representative survey in the United States with in- formation on sex at birth and current gender. These data indicate that non-cisgender individuals—those whose sex at birth does not align with their current gender—are significantly less likely to be married and more likely to have a spouse who has passed away than their cisgender counterparts. Non-cisgender indi- viduals are also significantly less likely than otherwise similar cisgender women to have children in the household. Alzheimer's disease (AD) is the most prevalent neurodegenerative disorder. Antiamyloid antibody treatments modestly slow disease progression in mild dementia due to AD. Emerging evidence shows that homeostatic dysregulation of the brain immune system, especially that orchestrated by microglia, plays an important role in disease onset and progression. Thus, a major question is how to modulate the phenotype and function of microglia to treat AD. Xenon (Xe) gas is a noble gas used in human patients as an anesthetic and a neuroprotectant used for treating brain injuries. Xe penetrates the blood-brain barrier, which could make it an effective therapeutic. To assess the effect of Xe on microglia and AD pathology, we designed a custom Xe inhalation chamber and treated several mouse models of AD with Xe gas. Xe treatment induced mouse microglia to adopt an intermediate activation state that we have termed pre–neurodegenerative microglia (pre-MGnD). This microglial phenotypic transition was observed in mouse models of acute neurodegeneration and amyloidosis (APP/PS1 and 5xFAD mice) and tauopathy (P301S mice). This microglial state enhanced amyloid plaque compaction and reduced dystrophic neurites in the APP/PS1 and 5xFAD mouse models. Moreover, Xe inhalation reduced brain atrophy and neuroinflammation and improved nest-building behavior in P301S mice. Mechanistically, Xe inhalation induced homeostatic brain microglia toward a pre-MGnD state through IFN-γ signaling that maintained the microglial phagocytic response in APP/PS1 and 5xFAD mice while suppressing the microglial proinflammatory phenotype in P301S mice. These results support the translation of Xe inhalation as an approach for treating AD. Editor's summary: Aberrant regulation of microglia is known to contribute to Alzheimer's disease (AD). Bacterial transcription activator-like effectors (TALEs) promote pathogenicity by activating host susceptibility (S) genes. To understand the pathogenicity and host adaptation of Xanthomonas citri pv. malvacearum (Xcm), we assemble the genome and the TALE repertoire of three recent Xcm Texas isolates. A newly evolved TALE, Tal7b, activates GhSWEET14a and GhSWEET14b, different from GhSWEET10 targeted by a TALE in an early Xcm isolate. Activation of GhSWEET14a and GhSWEET14b results in water-soaked lesions. Transcriptome profiling coupled with TALE-binding element prediction identify a pectin lyase gene as an additional Tal7b target, quantitatively contributing to Xcm virulence alongside GhSWEET14a/b. CRISPR-Cas9 gene editing supports the function of GhSWEETs in cotton bacterial blight and the promise of disrupting the TALE-binding site in S genes for disease management. Collectively, our findings elucidate the rapid evolution of TALEs in Xanthomonas field isolates and highlight the virulence mechanism wherein TALEs induce multiple S genes to promote pathogenicity. Newly evolved Xanthomonas citri pv. malvacearum isolates triggers recent bacterial blight outbreaks in cotton. Our ability to balance upright provides a stable platform to perform daily activities. Balance deficits associated with various clinical conditions may affect activities of daily living, highlighting the importance of quantifying standing balance in ecological environments. Although typically performed in laboratory settings, the growing availability of low-cost inertial measurement units (IMUs) allows the assessment of balance in the real world. However, it is unclear how many IMUs are required to adequately estimate linear displacements of the centre of mass (CoM) at stance widths associated with daily activities. While wearing IMUs on their head, sternum, back, right thigh, right shank, and left shank, 16 participants stood quietly on a force platform in narrow, hip-width, and shoulder-width stances, each for three two-minute trials. Using a multi-segment biomechanical model, we estimated CoM displacements from all possible combinations of the IMUs. We then calculated the correlation between the IMU- and force platform- CoM estimates to determine the minimal number of IMUs needed to estimate CoM sway. Four IMUs were necessary to accurately estimate anteroposterior (AP) and mediolateral (ML) CoM displacements across stance widths. Using IMUs on the back, right thigh, and both shanks, we found strong correlations between the IMU CoM estimation and the force platform CoM estimation in narrow stance (AP: r = 0.92±0.04, RMSE = 2.39±2.08 mm; ML: r = 0.97±0.02, RMSE = 1.16±0.77 mm), hip-width stance (AP: r = 0.93±0.04, RMSE = 2.00±1.18 mm; ML: r = 0.92±0.06, RMSE = 0.92±0.70 mm), and shoulder-width stance (AP: r = 0.93±0.03, RMSE = 1.95±1.66 mm; ML: r = 0.86±0.13, RMSE = 1.39±1.46 mm). These results indicate that IMUs can be used to estimate CoM displacements during quiet standing and that four IMUs are necessary to do so. Using an algorithm based on a simple biomechanical model, researchers and clinicians can estimate whole-body CoM displacements accurately during unperturbed quiet standing. Allogeneic haematopoietic stem cell transplantation (alloHSCT) is safe and effective for adolescents and adults with inborn errors of immunity (IEI) with severe disease manifestations of their disease. The haematopoietic cell transplantation comorbidity index (HCT-CI) score predicts transplant survival in non-malignant diseases, including IEIs. We hypothesised that immune dysregulation pre-transplant may also influence transplant outcomes. We calculated the pre-transplant immune dysregulation and disease activity score (IDDA v2.1) for 82 adolescent and adult IEI patients (aged ≥ 13 years). Three-year overall survival (OS) for the whole cohort was 90% (n = 82) with a median follow up of 44.7 months (range 8.4 to 225.8). Events were defined as acute graft-versus-host disease (GvHD) grades II or above, chronic GvHD of any grade, graft failure, or death from any cause. Three-year event free survival (EFS) for the whole cohort was 72%. In multivariable analysis the IDDA v2.1 score pre-transplant and HCT-CI score significantly impacted OS (hazard ratio 1.08, p = 0.028) and EFS (hazard ratio 1.04, p = 0.0005). Importantly, 35% of this cohort had a high IDDA v2.1 score (≥ 15) and low HCT-CI score (< 3) suggesting that the risks of alloHSCT may be underestimated in a proportion of patients with IEI if the HCT-CI score is used alone. These findings support the potential for improved outcomes following successful modulation of immune dysregulation pre-transplant. The IDDA v2.1 score has utility as an objective measurement of pre-transplant immune dysregulation providing additional information reagrding the risks and potential complications of alloHSCT in an individual IEI patient. Background: Evolutionary tradeoffs between life-history strategies are important in animal evolution. Because microbes can influence multiple aspects of host physiology, including growth rate and susceptibility to disease or stress, changes in animal-microbial symbioses have the potential to mediate life-history tradeoffs. Scleractinian corals provide a biodiverse, data-rich, and ecologically-relevant host system to explore this idea. Results: Using a comparative approach, we tested if coral microbiomes correlate with disease susceptibility across 425 million years of coral evolution by conducting a cross-species coral microbiome survey (the "Global Coral Microbiome Project") and combining the results with long-term global disease prevalence and coral trait data. Interpreting these data in their phylogenetic context, we show that microbial dominance predicts disease susceptibility, and traced this dominance-disease association to a single putatively beneficial symbiont genus, Endozoicomonas. Endozoicomonas relative abundance in coral tissue explained 30% of variation in disease susceptibility and 60% of variation in microbiome dominance across 40 coral genera, while also correlating strongly with high growth rates. Conclusions: These results demonstrate that the evolution of Endozoicomonas symbiosis in corals correlates with both disease prevalence and growth rate, and suggest a mediating role. Exploration of the mechanistic basis for these findings will be important for our understanding of how microbial symbioses influence animal life-history tradeoffs. [ABSTRACT FROM AUTHOR]

Context Tumor-induced osteomalacia (TIO) is an ultra-rare, paraneoplastic syndrome caused by tumors that secrete fibroblast growth factor 23 (FGF23). Initial signs and musculoskeletal symptoms can be nonspecific and unrecognized, leading to long delays in diagnosis and treatment, and resulting in severe and progressive disability in patients with TIO. Objective This review aimed to identify published evidence on healthcare resource use in TIO to better understand the burden of the disease. Evidence acquisition A targeted literature review was conducted to identify publications reporting on disease characteristics and healthcare resource use associated with TIO. Evidence synthesis In total, 414 publications were included in the review, of which 376 were case reports. From the case reports, data on 621 patients were extracted. These patients had a mean (SD) age of 46.3 (15.8) years; 57.6% were male. Mean time from first symptoms to diagnosis of TIO was 4.6 (4.7) years and, in cases where imaging tests were reported, patients underwent a mean of 4.1 (2.7) procedures. Tumor resection was attempted in 81.0% of patients and successful in 67.0%. Fracture was reported in 49.3% of patients. Results from association analyses demonstrated that longer time to diagnosis was associated with poorer tumor resection outcomes and a higher probability of tumor recurrence. Unfavorable tumor resection outcomes were associated with greater use of pharmacologic treatment and a greater likelihood of orthopedic surgery. Conclusion TIO is associated with a substantial healthcare resource burden. Improvements in the diagnostic process could lead to better management of TIO, thereby benefiting patients and reducing that burden. [ABSTRACT FROM AUTHOR]

Assessment of Wilson disease is complicated, with neither ceruloplasmin, nor serum or urine copper, being reliable. Two new indices, accurate non-ceruloplasmin copper (ANCC) and relative ANCC were developed and applied to a cohort of 71 patients, as part of a Wilson Disease Registry Study. Elemental copper-protein speciation was developed for holo-ceruloplasmin quantitation using strong anion exchange chromatography coupled to triple quadrupole inductively coupled plasma mass spectrometry. The serum proteins were separated using gradient elution and measured at m/z 63 (63Cu+) and 48 (32S16O+) using oxygen reaction mode and Cu-EDTA as calibration standard. The ANCC was calculated by subtraction of the ceruloplasmin bound copper from the total serum copper and the RelANCC was the percentage of total copper present as the ANCC. The accuracy of the holo-ceruloplasmin measurement was established using two certified reference materials, giving a mean recovery of 94.2 %. Regression analysis between the sum of the copper containing species and total copper concentration in the patient samples was acceptable (slope=0.964, intercept=0, r=0.987) and a difference plot, gave a mean difference for copper of 0.38 μmol/L. Intra-day precision for holo-ceruloplasmin at serum copper concentrations of 0.48 and 3.20 μmol/L were 5.2 and 5.6 % CV and the intermediate precision at concentrations of 0.80 and 5.99 μmol/L were 6.4 and 6.4 % CV, respectively. The limit of detection (LOD) and lower limit of quantification (LLOQ) for holo-ceruloplasmin were 0.08 and 0.27 μmol/L as copper, respectively. Background: Obesity is a major risk factor for noncommunicable diseases and is associated with a reduced life expectancy of up to 20 years, as well as with other consequences such as unemployment and increased economic burden for society. It is a multifactorial disease, and physiopathology of obesity involves dysregulated calorie utilization and energy balance, disrupted homeostasis of appetite and satiety, lifestyle factors including sedentary lifestyle, lower socioeconomic status, genetic predisposition, epigenetics, and environmental factors. Some endocrine-disrupting chemicals (EDCs) have been proposed as "obesogens" that stimulate adipogenesis leading to obesity. In this review, definition of obesogens, their adverse effects, underlying mechanisms, and metabolic implications will be updated and discussed. Summary: Disruption of lipid homeostasis by EDCs involves multiple mechanisms including increase in the number and size of adipocytes, disruption of endocrine-regulated adiposity and metabolism, alteration of hypothalamic regulation of appetite, satiety, food preference and energy balance, and modification of insulin sensitivity in the liver, skeletal muscle, pancreas, gastrointestinal system, and the brain. At a cellular level, obesogens can exert their endocrine disruptive effects by interfering with peroxisome proliferator-activated receptors and steroid receptors. Human exposure to chemical obesogens mainly occurs by ingestion and, to some extent, by inhalation and dermal uptake, usually in an unconscious manner. Persistent pollutants are lipophilic features; thus, they bioaccumulate in adipose tissue. Key Messages: Although there are an increasing number of reports studying the effects of obesogens, their mechanisms of action remain to be elucidated. Purpose: This study aimed to identify clinician-perceived barriers and facilitators before the implementation of surface electromyography (sEMG) for swallowing management, implement sEMG biofeedback in swallowing rehabilitation sessions using implementation strategies, and investigate the perceived benefits and drawbacks after the implementation of the sEMG device from the perspectives of speech-language pathologists (SLPs). Method: An initial pre-implementation survey characterized the SLPs' practices in swallowing management regarding the use of biofeedback modalities as well as facilitators and barriers to the implementation of sEMG. In the implementation phase, six SLPs attended educational and training meetings, tested, and used sEMG with patients during their swallowing sessions. Finally, a postimplementation survey and focus group assessed the six SLPs' perceptions and experiences using sEMG and identified areas for improvement in the implementation process. Results: The majority of the 44 SLPs who answered the pre-implementation survey did not use sEMG in swallowing therapy. The most frequently reported barriers to implementation were reduced sEMG knowledge and lack of training (86.4%), lack of equipment access or availability (68.2%), and patient-related barriers (36.6%). The six SLPs who participated in the implementation phase used sEMG with 30 different patients, conducting a total of 105 sessions. They reported that it was easy to use sEMG during sessions and that sEMG helped them to teach swallowing maneuvers. Additionally, they stated that patients were motivated during sEMG sessions. Ongoing training and mentoring were some of the suggestions for implementation improvements. Conclusions: This study exemplifies the collaborative work between researchers and clinicians to facilitate the translation of technologies into clinical practice. An 8-y-old National Show Horse mare was presented for evaluation of pneumonia and laminitis. Harsh bronchovesicular sounds were auscultated throughout both lung fields, and the mare had signs of moderately painful laminitis. Thoracic ultrasonography revealed lung consolidation throughout the dorsal aspect of both lungs, and radiography revealed an extensive diffuse-to-patchy bronchointerstitial lung pattern. The mare's clinical condition rapidly deteriorated, and euthanasia was elected. On postmortem examination, the lungs, omentum, spleen, liver, adrenal glands, kidneys, and femur contained 0.5–2.5-cm, firm, tan nodules. Histologically, the lungs, spleen, liver, kidneys, adrenal glands, omentum, left eye, and femur were infiltrated by bundles and nests of pleomorphic polygonal-to-spindloid cells intermixed with frequent multinucleate cells. Lymphatic vessels in the affected tissues were frequently distended with tumor emboli. Neoplastic cells were diffusely positive for vimentin, desmin, sarcomeric actin, myoblastic differentiation protein 1, and myogenin, supportive of the diagnosis of rhabdomyosarcoma (RMS), which is a rare neoplasm in horses. Cross-striations were not evident with H&E or phosphotungstic acid–hematoxylin stains. Iron deficiency (ID) is common during gestation and early infancy and can alter developmental trajectories with lasting consequences on cardiovascular health. Iron plays a critical role in systemic oxygen transport (via hemoglobin) and aerobic respiration (as a component of mitochondrial complexes). Perinatal ID has been shown to cause cardiac dysfunction in neonates, but the mechanisms underlying these changes have not been characterized. Here, we examined the effects of perinatal ID on cardiac mitochondrial function in rats in the early postnatal period. Female rats were fed an iron-restricted or iron-replete diet before and during pregnancy. Offspring hearts were collected postmortem for quantitative shotgun proteomic analysis [postnatal days (PD) 0 and 28] and mitochondrial function was assessed by high-resolution respirometry (at PD 0, 14, and 28). Markers of oxidative stress were measured by fluorescence microscopy and assessment of antioxidant gene expression profiles. Both male and female ID pups had reduced body weight and increased relative heart weights at all time points assessed, despite recovering from anemia by PD28. Proteomics analysis revealed dysregulation of mitochondrial proteins by ID, and these differences were most pronounced in males. In male hearts, ID increased mitochondrial content and decreased normalized mitochondrial respiration through the NADH-pathway, succinate-pathway, and fatty acid oxidation (FAO)-pathway. In conclusion, ID causes changes in cardiac mitochondrial function in neonates, which may reflect inadequate or maladaptive compensation during the transition from intrauterine to extrauterine life. Furthermore, the results presented herein, which were stratified by offspring sex, underscore the need for follow-up studies to directly assess differences in how male and female offspring cope with ID as a perinatal stressor. The North Atlantic Climate System Integrated Study (ACSIS) was a large multidisciplinary research programme funded by the UK's Natural Environment Research Council (NERC). ACSIS ran from 2016 to 2022 and brought together around 80 scientists from seven leading UK-based environmental research institutes to deliver major advances in the understanding of North Atlantic climate variability and extremes. Here, we present an overview of the data generated by the ACSIS programme. The datasets described cover the North Atlantic Ocean, the atmosphere above it (including its composition), and Arctic sea ice. Atmospheric composition datasets include measurements from seven aircraft campaigns (45 flights in total, 0–10 km altitude range) in the northeastern Atlantic (∼ 15–55° N, ∼ 40° W–5° E) made at intervals of 6 months to 2 years between February 2017 and May 2022. The flights measured chemical species (including greenhouse gases; ozone precursors; and volatile organic compounds – VOCs) and aerosols (organic aerosol – OA; SO4; NH4; NO3; and non-sea salt chloride – nss-Cl) (10.5285/6285564c34a246fc9ba5ce053d85e5e7, FAAM et al., 2024). Ground-based stations at the Cape Verde Atmospheric Observatory (CVAO), Penlee Point Atmospheric Observatory (PPAO), and Plymouth Marine Laboratory (PML) recorded ozone, ozone precursors, halocarbons, greenhouse gases (CO2 and methane), SO2, and photolysis rates (CVAO; http://catalogue.ceda.ac.uk/uuid/81693aad69409100b1b9a247b9ae75d5 , National Centre for Atmospheric Science et al., 2010); O3 and CH4 (PPAO, https://catalogue.ceda.ac.uk/uuid/8f1ff8ea77534e08b03983685990a9b0 (Plymouth Marine Laboratory and Yang, 2017); and aerosols (PML, 10.5285/e74491c96ef24df29a9342a3d57b5939, Smyth, 2024), respectively. Complementary model simulations of atmospheric composition were performed with the UK Earth System Model (UKESM1) for the period from 1982 to 2020 using Coupled Model Intercomparison Project Phase 6 (CMIP6) historical forcing up to 2014 and Shared Socioeconomic Pathways (SSP) scenario SSP3-7.0 from 2015 to 2020. Model temperature and winds were relaxed towards ERA reanalysis. Monthly mean model data for ozone, NO, NO2, CO, methane, stratospheric ozone tracers, and 30 regionally emitted tracers are available for download (https://data.ceda.ac.uk/badc/acsis/UKESM1-hindcasts , Abraham, 2024). An ensemble of atmosphere-forced global-ocean sea-ice simulations using the NEMO-CICE model was performed with horizontal resolutions of 1/4 and 1/12 ° covering the period from 1958 to 2020 using several different atmosphere-reanalysis-based surface forcing datasets, supplemented by additional global simulations and stand-alone sea-ice model simulations with advanced sea-ice physics using the CICE model (http://catalogue.ceda.ac.uk/uuid/770a885a8bc34d51ad71e87ef346d6a8 , Megann et al., 2021e). Generalist predator populations often surge during resource pulses but decline sharply afterwards and shift to alternative prey sources, which experience elevated predation rates. Understanding these numerical and functional responses of predators to resource pulses is especially important for effectively managing island ecosystems, where invasive mammals are generalist predators and vulnerable indigenous birds are alternative prey. We used DNA metabarcoding of stomach contents to test whether the proportion of black rats (Rattus rattus) consuming birds changed during and after a mast seeding year in a New Zealand beech forest. Contrary to prevailing assumptions that black rats in forests consume birds only occasionally and increase their consumption of birds once a resource pulse finishes, we found that the overall proportion of rats consuming birds was surprisingly high (19.4%), and that the proportion of rats consuming birds did not change significantly across the three years of a beech mast cycle. We detected 15 unique bird taxa in the rat stomachs and 40 unique plant taxa. The frequency of beech consumption declined across the three years, as predicted. We conclude that (1) forest birds are a common part of rat diet rather than an occasional opportunistic prey item, and (2) the impact of black rats on indigenous birds following seed pulses is predominantly driven by the numerical response (increased numbers of rats present) rather than a functional (prey switching) response. We link the 2010 Census microdata to the 2010-2020 American Community Surveys and Social Security Administration records to test patterns of ethnoracial identification change across this decade. After documenting substantial ethnoracial stability in some categories, we find substantial flows between many racial categories, more movement into Hispanic identification than movement out of the Hispanic category, foreign-born Hispanic multiracial respondents are 14 percentage points (40 percent) more likely to identify later as Hispanic White than their native-born counterparts, and foreign-born non-Hispanic multiracial respondents are 19 percentage points (90 percent) less likely to identify later as non-Hispanic White than native-born.
