Purpose/Objective: Recognising the urgency to safeguard the planet and foster resilience against climate-related health challenges, this study aligns with the 2030 Agenda and explores local initiatives, such as those by the local action group (LAG). Design/Methodology/Approach: This analysis identified the strengths, weaknesses, opportunities, and threats faced by the Tirreno-Eolie LAG, providing a solid foundation for developing the Local Development Plan (LDP). Moreover, an analytic hierarchy process (AHP) analysis was employed to yield a prioritised ranking of the outlined LDP strategies based on the stakeholder input. Findings: The analysis of the LDP for LAG revealed a comprehensive set of actions designed to promote sustainable development at the local level: 1. urban redevelopment projects, 2. preservation of historical buildings, 3. environmental awareness initiatives in Sicily, 4. biological diversity programmes and education, 5. promoting circular economy principles, 6. implementing sustainable corporate governance, 7. developing information systems for control and security, 8. establishing partnerships with local associations, 9. leveraging European initiatives, 10. deployment of newer and greener technologies, 11. redevelopment of infrastructure. Significance/Implications/Conclusions: By strategically implementing the LDP's recommendations, LAG can contribute to building a more sustainable, resilient, and culturally vibrant Sicily. Limitations: Firstly, the available information on the LAG Tirreno-Eolie may be limited. Secondly, the complex and ever-changing nature of social and environmental systems can constrain the objective of the research. Future Research: Future research may investigate the effectiveness of formulated actions and outcomes over time, conduct comparative analyses with regions implementing similar sustainability initiatives, and investigate the scalability and adaptability of the LDP framework to diverse contexts. Background: Cangrelor provides rapid platelet inhibition, making it a potential option for out-of-hospital cardiac arrest (OHCA) survivors undergoing percutaneous coronary intervention (PCI). However, clinical data on its use after OHCA are limited. This study investigates in-hospital outcomes of cangrelor use in this population. Methods: We conducted a prospective, observational study involving OHCA patients from the Lombardia CARe Registry (January 2015–December 2022) who underwent PCI in seven centers in Northern Italy. Propensity score (PS) matching compared patients who received cangrelor to those who did not. Logistic regression tested associations between cangrelor and discharge outcomes. Results: Of 612 OHCA patients admitted, 414 (67.4%) underwent PCI with known antithrombotic therapy, of whom 34 (8.2%) received cangrelor. Radial access was more common in the cangrelor group, which also had a higher troponin peak and a final TIMI flow grade of 3. Survival at discharge was 82.4% in the cangrelor group, compared to 65.3% in the no-cangrelor group (p = 0.043). Univariable logistic regression showed that cangrelor use was associated with higher survival at discharge (OR 2.5; 95% CI: 1.1–6.1, p = 0.049). After multiple PS matchings, cangrelor remained associated with better survival (OR 2.07; 95% CI: 1.16–2.98). Major bleeding rates were higher in the cangrelor group, even after adjusting for baseline bleeding risk (OR: 7.0; 95% CI: 2.9–17.0; p < 0.001). Conclusions: In OHCA patients undergoing PCI, cangrelor use was linked to improved in-hospital survival but higher major bleeding, suggesting a potential net clinical benefit. [ABSTRACT FROM AUTHOR]

Klebsiella pneumoniae represent a common invasive infection etiological agent, whose potential carbapenem-resistance and hypermucoviscosity complicate the patient's management. Infection development often derives from gastrointestinal colonization; thus, it is fundamental to monitor asymptomatic K. pneumoniae colonization through surveillance protocols, especially for intensive care and immunocompromised patients. We described a six-month routine screening protocol from the Policlinico of Catania (Italy), while blood samples were collected from the same patients only in cases of a systemic infection suspicion. All the patients who had dissemination episodes were furtherly investigated through next-generation sequencing, analyzing both colonizing and disseminating strains. This study documents emerging invasive sequence types such as ST101, ST307, and ST395, mainly revealing blaNDM or blaKPC genes, along with siderophores and hyperproduction capsule markers as virulence factors. Most of the detected factors are presumably related to a specific plasmid content, which are extremely varied and rich. In conclusion, active surveillance through sequencing is essential to enhance awareness of local epidemiology within high-risk multi-drug resistance areas. A random sequencing analysis on the most warning microorganisms could enhance sequence typing (ST) awareness within specific settings, allowing for better prevention control strategies on their eventual persistence or diffusion. In recent years, the invasive Atlantic blue crab (Callinectes sapidus) has increased its spread throughout the Mediterranean Sea, threatening native biodiversity and local economies. This study aimed to valorize C. sapidus sampled in Sicily by utilizing its exoskeleton as a source of chitosan, astaxanthin, and bio-phenolic compounds. These biomolecules were evaluated for their reducing, radical scavenging, and antitumor activity. The ferric ion reducing antioxidant power (FRAP) and the free radical scavenging activity against radical 2,2-Diphenyl-1-picrylhydrazyl (DPPH) were significantly higher for chitosan (3.16 ± 0.10 mg AAE/g and 8.1 ± 0.10 µmol TE/g). No significant differences were observed among the tested biomolecules in their activity in scavenging the radical 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS). Both bio-phenolic compounds and astaxanthin exhibited dose-dependent cytotoxicity on CaCo-2 (IC50 = 12.47 and 18 µg/mL) and HepG2 (IC50 = 10.25 and 1.26 µg/mL) cell lines, while only bio-phenols showed no cytotoxic effect on differentiated CaCo-2 cells up to 20 µg/mL. These findings highlight the value of blue crab by-products in supporting a circular economy, offering a sustainable approach to managing this invasive species while providing bioactive compounds with promising medical and nutraceutical applications. Background: The inclusion of sustainable protein sources in poultry feed has become essential for improving animal welfare in livestock production. Black soldier fly larvae are a promising solution due to their high protein content and sustainable production. However, most research has focused on fast-growing poultry breeds, while the effects on native breeds, such as the Bianca di Saluzzo, are less explored. This study aimed to evaluate the impact of BSFL supplementation in the diet of slow-growing chickens, with a focus on growth, intestinal health and meat quality for final consumers. Results: The study demonstrated that Black soldier fly larvae supplementation, both in dehydrated and live form, improved growth performance, with an increase in final weights compared to the control group. No significant differences were found in feed conversion ratio, liver and spleen weight or histomorphometry between groups. Black soldier fly larvae supplementation did not negatively affect immune function or liver metabolism. Higher production propionic acid was detected in the black soldier fly larvae fed groups compared to the control, suggesting an effect on volatile fatty acid production. Gut microbiota analysis showed an increase in beneficial bacteria, such as Faecalibacterium, in the live larvae group. Furthermore, the meat fatty acid profile and atherogenicity and thrombogenicity indices did not undergo significant changes, implying a low potential risk to consumers' cardiovascular health. Conclusions: Black soldier fly larvae supplementation in the diet of Bianca di Saluzzo improved growth without compromising animal health or meat quality. Moreover, the increase in beneficial volatile fatty acids and the modulation of gut microbiota suggest a positive impact on gut health. Finally, the absence of negative effects on meat lipid profiles confirms nutritional safety for consumers, making black soldier fly larvae a valid alternative in poultry feed. These results offer new perspectives for the use of Black soldier fly larvae in the nutrition of local breeds, contributing to sustainability in line with the One Health approach. Background: Leishmaniasis, caused by Leishmania protozoan parasites transmitted by Phlebotomine sand flies, is a significant public health concern in the Mediterranean basin. Effective monitoring of Leishmania-infected sand flies requires standardized tools for comparing their distribution and infection prevalence. Consistent quantitative real-time PCR (qPCR) parameters and efficient DNA extraction protocols are crucial for reliable results over time and across regions. However, the absence of standardized technical recommendations for Leishmania DNA detection hinders effective surveillance. This study aimed to compare different DNA extraction protocols and conduct a qPCR-based External Quality Assessment (EQA) through a multicenter study involving nine reference laboratories, with a focus on optimizing Leishmania DNA detection in sand fly. Methodology/Principal findings: EQA samples consisted of Leishmania infantum and L. major species, at concentrations ranging from 101 to 104 parasites/mL. All but one center detected all concentrations, demonstrating strong diagnostic proficiency. The ability to detect low concentrations highlighted the robustness of the qPCR assay used, though variations in Cq values indicated differences in sensitivity related to technical capabilities or DNA extraction kit performance. A comparative analysis of seven DNA extraction methods identified the EZ1 DSP Virus Kit and QIAamp DNA mini-kit as the most efficient, supporting their use in standardized protocols. The study also assessed the effects of lyophilization and shipment conditions, showing no significant compromise in Leishmania detection despite slight variations in Cq values. Experimentally infected sand flies were included to simulate field conditions, and all centers successfully detected positive samples with varying Cq values, probably reflecting differences in infection load. Conclusion and significance: This study emphasizes the importance of standardized DNA extraction protocols and continuous quality assurance for accurate Leishmania DNA detection. The results highlight the superior performance of certain extraction kits and the need for ongoing technical training, essential for reliable leishmaniasis surveillance, particularly in field settings with low infection densities. The use of soybean meal in animal feed is crucial for nutrition but has a significant environmental impact, especially related to deforestation and biodiversity loss. With the increasing demand for sustainable agricultural practices, it becomes essential to explore alternatives to soybean meal. This study evaluated the effect of a soy-free diet, replaced with fava beans and pea protein, on an Italian slow-growing chicken breed: the Bianca di Saluzzo. The results indicate that the experimental diet did not compromise the growth, final weight or intestinal health of the animals, demonstrating a good tolerance and adaptability of the breed to these alternative ingredients. Furthermore, the analysis of the intestinal microbiota showed a positive impact, with an increase in organic acids such as succinic and citric, which could improve intestinal health and metabolic efficiency. Some changes in liver enzyme levels were observed, such as the increase in glutamate-oxaloacetate transaminase, an enzyme involved in amino acid metabolism. This increase could indicate a higher efficiency in protein metabolism, suggesting that the diet based on alternative ingredients could support an improvement in liver metabolism. Although this aspect deserves further investigation, it could represent a positive effect of the diet on liver function and overall health of the animals. Further studies are needed to fully understand the long-term implications of these dietary modifications, particularly in relation to the gut microbiota and metabolism. Septate uterus is one of the most common uterine malformations. Recent studies suggest that uterine septa may negatively affect fertility. In cases of recurrent pregnancy loss (RPL) or infertility, hysteroscopic metroplasty has been considered the primary treatment for septate uterus. This systematic review and meta-analysis aims to evaluate whether intraoperative ultrasound monitoring may improve the efficacy of hysteroscopic metroplasty compared to other types of intraoperative monitoring or to unguided resections. An electronic database search was performed to identify articles published until June 15, 2023. Five studies (two randomized clinical trials, two prospective studies and one retrospective cohort study) fulfilled the inclusion criteria. The primary outcome was the rate of residual septum > 10 mm after hysteroscopic metroplasty in the ultrasound (US) monitoring group compared to the rate of residual septum using other types of intraoperative monitoring/no monitoring (control group). The secondary outcomes were any residual septa, surgical time, complications, uterine perforations and reproductive outcomes. Intraoperative ultrasound for uterine septum resection significantly reduced the rate of residual septum > 10 mm and the rate of any residual septa compared to the control group. There was no statistically significant difference in the procedure time between women undergoing intraoperative US monitoring versus the control group. A trend toward reduction of surgical complications was observed in the intraoperative US group compared to the control group. In conclusion, intraoperative ultrasound during metroplasty may reduce the rate of the residual septum with no surgical time differences. Within the range of composite laminates for structural applications, sandwich laminates are a special category intended for applications characterized by high flexural stresses. As it is well known from the technical literature, structural sandwich laminates have a simple configuration consisting of two skins of very strong material, to which the flexural strength is delegated, between which an inner layer (core) of light material with sufficient shear strength is interposed. As an example, a sandwich configuration widely used in civil, naval, and mechanical engineering is that obtained with fiberglass skins and a core of various materials, such as polyurethane foam or another lightweight material, depending on the application. Increasingly stringent regulations aimed at protecting the environment by reducing harmful emissions of carbon dioxide and carbon monoxide have directed recent research towards the development of new composites and new sandwiches characterized by low environmental impact. Among the various green composite solutions proposed in the literature, a very promising category is that of high-performance biocomposites, which use bio-based matrices reinforced by fiber reinforcements. This approach can also be used to develop green sandwiches for structural applications, consisting of biocomposite skins and cores made by low-environmental impact or renewable materials. In order to make a contribution to this field, a structural sandwich consisting of high-performance sisal–epoxy biocomposite skins and an innovative renewable core made of balsa wood laminates with appropriate lay-ups has been developed and then properly characterized in this work. Through a systematic theoretical–experimental analysis of three distinct core configurations, the unidirectional natural core, the cross-ply type, and the angle-ply type, it has been shown how the use of natural balsa gives rise to inefficient sandwiches, whereas performance optimization is fully achieved by considering the angle-ply core type [±45/90]. Finally, the subsequent comparison with literature data of similar sandwiches has shown how the optimal configuration proposed can be advantageously used to replace synthetic glass–resin sandwiches widely used in various industrial sectors (mechanical engineering, shipbuilding, etc.) and in civil engineering. This study evaluates, for the first time, the reducing capacity, radical scavenger activity, and in vitro antitumor and anti-inflammatory effects of chitosan, astaxanthin, and bio-phenols extracted from the exoskeleton of Sicilian Procambarus clarkii, the most widespread species of invasive crayfish in the Mediterranean region. Among the extracted compounds, astaxanthin exhibited the highest antioxidant activity in all assays. Chitosan and polyphenols demonstrated reducing and radical scavenging activity; chitosan showed significant ferric ion reducing capacity in the FRAP test, while bio-phenolic compounds displayed notable radical scavenging activity in the DPPH and ABTS assays. Both astaxanthin and polyphenols showed dose-dependent cytotoxicity on two different cancer cell lines, with IC50 values of 1.45 µg/mL (phenolic extract) and 4.28 µg/mL (astaxanthin extract) for HepG2 cells and 2.45 µg/mL (phenolic extract) and 4.57 µg/mL (astaxanthin extract) for CaCo-2 cells. The bio-phenolic extract also showed potential anti-inflammatory effects in vitro by inhibiting nitric oxide production in inflamed RAW 264.7 macrophages, reducing the treated/control NO ratio to 77% and 74% at concentrations of 1.25 and 1.5 μg/mL, respectively. These results suggest that P. clarkii exoskeletons could be a valuable source of bioactive molecules for biomedical, pharmaceutical, and nutraceutical application while contributing to the sustainable management of this invasive species. A growing number of studies in both the scientific and the philosophical literature have drawn on a Bayesian predictive processing framework to account for the formation of delusions. The key here is that delusions form because of disrupted prediction error signalling. Parrott's recent critique argues that the framework is incomplete in two respects: (i) it leaves unclear why delusional hypotheses are selected over none at all, or over more plausible alternatives; and (ii) it leaves unclear how exactly it is that delusional hypotheses are generated in the first place. In this paper, we defend the framework's explanatory power by way of showing how it can go a long way in helping disentangle these concerns. We take up each of them in turn. As for the first, our argument turns on two claims: (a) believing nothing, although possible in principle, may not be a viable procedure in the circumstances; and (b) alternative hypotheses may be rendered unavailable for selection, due to aberrant prediction errors. Introduction: Roller skating shares biomechanical similarities with other sports, but specific studies on speed skaters are limited. Injuries, particularly to the groin, are frequent and related to acute and chronic muscle stress. Technology, particularly surface electromyography, can now be used to monitor performance and prevent injuries, especially those caused by muscular asymmetries. Such studies can be used to enhance training and for educational purposes. Materials and Methods: This pilot study was conducted on three subjects: two cadet-athletes and a novice, compared with the performance model of an elite athlete. Surface electromyography and kinematic analysis monitored the lower limb muscles during the propulsion and recovery phases of skating. Electrodes were placed on specific muscles, and triaxial accelerometers were used to detect kinematic differences and asymmetries. The results: Cadet 1 was closest to the elite athlete's performance model compared to Cadet 2, especially in kinematics and muscle efficiency. However, both cadets showed electromyographic differences compared to the elite athlete, with uneven muscle co-activations. The novice exhibited more oscillations and earlier propulsion compared to the elite athlete. Discussion: Using electromyography and kinematic analysis made it possible to identify differences between elite athletes, cadets, and novices. Foodborne diseases result from the consumption of foods contaminated with pathogens or their toxins and represent a serious public health problem worldwide. This study aimed to assess the presence of Rotavirus (RoV), Adenovirus (AdV), Norovirus (NoV), Hepatitis A and Hepatitis E viruses (HAV and HEV, respectively), Toxoplasma gondii, Coxiella burnetii and Leptospira spp. across various food matrices in Sicily. The analysis concerned 504 samples, including mussels, farmed meat, game meat, vegetables and bulk milk. Following appropriate pre-treatment, acid nucleic extraction was carried out and amplification of pathogen nucleic acids was carried out by molecular methods. The mussels tested positive for NoVs (3/51, 5.9%) and farm meat resulted positive for T. gondii (1/34, 2.9%). The game offal samples tested positive for HEV, which was detected in 17 out of 222 samples (7.7%), and T. gondii (18/318, 5.7%) and Leptospira spp. (2/318, 0.6%). The milk samples tested positive for C. burnetii (15/85, 17.6%), T. gondii (2/85, 2.4%) and Leptospira spp. (1/85, 1.2%). This study highlights the variability in the risk of contamination of different food matrices, confirming the importance of vigilance in the consumption of potentially contaminated food products. We introduce and solve a 2 α -order fractal differential equation with constant coefficients across different scenarios. We propose a uniqueness theorem for 2 α -order fractal linear differential equations. We define the solution space as a vector space with non-integer orders. Context. Current and future large surveys will produce unprecedented amounts of data. Realistic simulations have become essential for the design and development of these surveys, as well as for the interpretation of the results. Aims. We present MAMBO, a flexible and efficient workflow to build empirical galaxy and active galactic nucleus (AGN) mock catalogues that reproduce the physical and observational properties of these sources. Methods. We started with simulated dark matter (DM) haloes, to preserve the link with the cosmic web, and we populated them with galaxies and AGN using abundance matching techniques. We followed an empirical methodology, using stellar mass functions, host galaxy AGN mass functions, and AGN accretion rate distribution functions studied at different redshifts to assign, among other properties, stellar masses, the fraction of quenched galaxies, or the AGN activity (demography, obscuration, multiwavelength emission, etc.). Results. As a proof test, we applied the method to a Millennium DM lightcone of 3.14 deg2 up to a redshift of z = 10 and down to stellar masses of ℳ ≳ 1075 M⊙. We show that the AGN population from the mock lightcone presented here reproduces with good accuracy various observables, such as state-of-the-art luminosity functions in the X-ray up to z~7 and in the ultraviolet up to z~5, optical/near-infrared colour-colour diagrams, and narrow emission line diagnostic diagrams. Finally, we demonstrate how this catalogue can be used to make useful predictions for large surveys. Using Euclid as a case example, we compute, among other forecasts, the expected surface densities of galaxies and AGN detectable in the Euclid HE band. We find that Euclid might observe (on HE only) about 107 and 8 × 107 type 1 and 2 AGN, respectively, and 2 × 109 galaxies at the end of its 14 679 deg2 Wide survey, in good agreement with other published forecasts. Introduction: Several studies have investigated the specific neural correlates of trans people, highlighting mixed results. This study aimed to compare the presence of specific functional connectivity and differences in cognitive profile and hormone levels in trans men diagnosed with gender dysphoria (GD), and a homogeneous group of cisgender men and cisgender women. Methods: A total of 42 participants (19 trans men, 11 cisgender men, and 12 cisgender women) underwent a resting state fMRI and were measured for blood levels of testosterone, estradiol, and progesterone. A neuropsychological battery evaluated executive functions, attention, visual-perceptual ability, verbal fluency, manual preference, and general intelligence. Results: Trans men showed weaker functional connectivity in the precentral gyrus, subcallosal cortex, paracingulate gyrus, temporal pole, and cingulate gyrus than cisgender men (p < 0.01). Trans men performed worse than cisgender men in verbal and visuospatial working memory but similarly to cisgender women (p < 0.05). In trans men, functional connectivity of the precentral gyrus correlated positively with testosterone (r = 0.459, p = 0.064) and negatively with estradiol (r = −0.654, p = 0.004) and progesterone blood levels (r = −0.475, p = 0.054). The cluster involving the subcallosal cortex showed a positive correlation with testosterone (r = 0.718, p = 0.001), and a negative correlation with estradiol (r = −0.602, p = 0.011). The functional connectivity from a cluster involving the paracingulate gyrus showed a positive correlation with testosterone (r = 0.592, p = 0.012). Conclusions: This study highlights the importance of overpassing the binary model by underlining the presence of neural pathways that could represent the peculiarity of the neural profile of people with GD. Since its introduction in 1987, dual-energy X-ray absorptiometry (DXA) has revolutionized bone assessment, becoming the gold standard for measuring bone mineral density (BMD). Its low radiation exposure and high accuracy have made it indispensable in diagnosing osteoporosis, aligning with World Health Organization criteria. However, DXA evolution extends beyond BMD measurement, with emerging tools like the Trabecular Bone Score (TBS) and the DXA-based Bone Strain Index (BSI). TBS provides insights into trabecular bone architecture, enhancing the prediction of fracture risk. Despite limitations like body mass index correlation, TBS aids in evaluating patients with conditions such as diabetes and glucocorticoid exposure. BSI, introduced in 2019, evaluates bone strength using finite element analysis, complementing BMD and TBS by assessing bone fatigue. Advancements in DXA-based tools extend to Hip Structural Analysis and three-dimensional DXA software, offering valuable insights into hip fracture risk. Moreover, DXA serves beyond bone assessment, aiding in abdominal aortic calcification assessment, enhancing cardiovascular risk stratification. Aim: To analyze the prevalence of anxiety and depression in a large cohort of adults with autoimmune diabetes, identifying sex-driven associated factors. Materials and methods: In this cross-sectional study, we enrolled 553 consecutive adults with Type 1 diabetes mellitus or latent autoimmune diabetes in adults who came to the Division of Endocrinology of the S.Orsola-Malpighi Polyclinic, Bologna (Italy), to receive their second dose of SARS-CoV-2 vaccine. We administered the questionnaires: Hospital Anxiety and Depression Scale, Diabetes Distress Scale, Diabetes-related Quality of Life, Diabetes Treatment Satisfaction Questionnaire. We collected clinical and biochemical data and 14 days glucose metrics in patients with sensor use > 70% in a time span of ± 4 months from the questionnaires' administration. We excluded 119 patients from our analyses with missing data (final cohort n = 434: 79% of those enrolled). Results: Anxiety and depression prevalence was respectively 30.4% and 10.8%. According to the multivariate analysis, higher diabete-related emotional burden, lower treatment satisfaction, but not physician-related distress, were risk factors for anxiety and depression; female sex was associated with anxiety (OR 0.51, 95% 0.31–0.81; p = 0.005); in women, depression was associated with increasing age (males vs. females OR 0.96 per 1 year increase, 95% CI 0.92–1.00; p = 0.036), whilst in men with HbA1c (OR 1.08 per 1 mmol/mol increase, 95% CI 1.03–1.13; p = 0.002). Conclusion: Nearly 1/3 of patients with autoimmune diabetes suffers from anxiety and 1/10 from depression. Antimicrobials, such as antibiotics or antivirals are medications employed to prevent and treat infectious diseases in humans, animals, and plants. Antimicrobial Resistance occurs when bacteria, viruses, and parasites no longer respond to these medicines. This resistance renders antibiotics and other antimicrobial drugs ineffective, making infections challenging or impossible to treat. This escalation in drug resistance heightens the risk of disease spread, severe illness, disability, and mortality. With datasets now containing hundreds or even thousands of pathogen genomes, machine learning techniques are on the rise for predicting antibiotic resistance in pathogens, prediction based on gene content and genome composition. In recent decades, in order to replace traditional synthetic polymer composites, engineering research has focused on the development of new alternatives such as green biocomposites constituted by an eco-sustainable matrix reinforced by natural fibers. Such innovative biocomposites are divided into two different typologies: random short fiber biocomposites characterized by low mechanical strength, used for non-structural applications such as covering panels, etc., and high-performance biocomposites reinforced by long fibers that can be used for semi-structural and structural applications by replacing traditional materials such as metal (carbon steel and aluminum) or synthetic composites such as fiberglass. The present research work focuses on the high-performance biocomposites reinforced by optimized sisal fibers. In detail, in order to contribute to the extension of their application under fatigue loading, a systematic experimental fatigue test campaign has been accomplished by considering four different lay-up configurations (unidirectional, cross-ply, angle-ply and quasi-isotropic) with volume fraction Vf = 70%. The results analysis found that such laminates exhibit good fatigue performance, with fatigue ratios close to 0.5 for unidirectional and angle-ply (±7.5°) laminates. However, by passing from isotropic to unidirectional lay-up, the fatigue strength increases significantly by about four times; higher increases are revealed in terms of fatigue life. In terms of damage, it has been observed that, thanks to the high quality of the proposed laminates, in any case, the fatigue failure involves the fiber failure, although secondary debonding and delamination can occur, especially in orthotropic and cross-ply lay-up. The comparison with classical synthetic composites and other similar biocomposite has shown that in terms of fatigue ratio, the examined biocomposites exhibit performance comparable with the biocomposites reinforced by the more expensive flax and with common fiberglass. Staphylococcus (S.) aureus is a pathogenic bacterium able to cause several diseases in humans and animals as well as foodborne intoxications. S. argenteus, being phenotypically and genotypically related to S. aureus, is part of the so-called S. aureus complex and recently recognized as an emerging pathogen able to cause, like S. aureus, several diseases both in humans and animals, and foodborne poisoning outbreaks. However, it has been reported that the widely used conventional PCR of Brakstad et al. [Journal of Clinical Microbiology, 30(7), 1654–1660, (1992)] targeting the thermostable nuclease gene may provide false-positive S. aureus, as it is able to amplify also S. argenteus. Here, we developed a novel two-step approach that, following the PCR of Brakstad et al. (1992), discriminates S. aureus from S. argenteus by a real-time PCR with high-resolution melting analysis (rt-PCR-HRM). In particular, targeting a polymorphic 137 bp region of the sodA gene, our developed rt-PCR-HRM method clearly discriminated S. aureus from S. argenteus, showing a remarkable difference in their amplification product melting temperatures (approximately 1.3 °C) as well as distinct melting curve shapes. The good sensitivity, reproducibility, user friendliness, and cost effectiveness of the developed method are advantageous attributes that will allow not only its easy employment to correctly identify misidentified isolates present in various collections of S. aureus, but also expand the still lacking knowledge on the prevalence and distribution of S. argenteus. Hidradenitis suppurativa (HS) is a chronic inflammatory skin disease characterized by recurrent and painful nodules and abscesses in intertriginous skin areas, which can progress to sinus tract formation, tissue destruction, and scarring. HS is highly debilitating and severely impairs the psychological well-being and quality of life of patients. The therapeutic approach to HS is based on medical therapy and surgery. First-line medical therapy includes topical antibiotics, systemic antibiotics, and biologics. Main surgical procedures include deroofing, local excision, and wide local excision. Despite the availability of multiple therapeutic options, the rates of disease recurrence and progression continue to be high. In recent years, the possibility of combining biologic therapy and surgery has raised considerable interest. In a clinical trial, the perioperative use of adalimumab has been associated with greater response rates and improved inflammatory load and pain, with no increased risk of postoperative infectious complications. However, several practical aspects of combined biologic therapy and surgery are poorly defined. In June 2022, nine Italian HS experts convened to address issues related to the integration of biologic therapy and surgery in clinical practice. To this purpose, the experts identified 10 areas of interest based on published evidence and personal experience: (1) patient profiling (diagnostic criteria, disease severity classification, assessment of response to treatment, patient-reported outcomes, comorbidities); (2) tailoring surgery to HS characteristics; (3) wide local excision; (4) presurgery biologic treatment; (5) concomitant biologic and surgical treatments; (6) pre- and postsurgery management; (7) antibiotic systemic therapy; (8) biologic therapy after radical surgery; (9) management of adverse events to biologics; and (10) management of postoperative infectious complications. Consensus between experts was reached using the Estimate-Talk-Estimate method (Delphi Method). The statements were subsequently presented to a panel of 27 HS experts from across Italy, and their agreement was assessed using the UCLA Appropriateness Method. Consider a time series that takes values in a general topological space, and suppose that its Small-ball probability is factorised into two terms that play the role of a surrogate density and a volume term. The latter allows us to study the complexity of the underlying process. In some cases, the volume term can be analytically specified in a parametric form as a function of a complexity index. This work presents the study of an estimator for such an index whenever the volume term is monomial. Weak consistency and asymptotic Gaussianity are shown under an appropriate dependence structure, providing theoretical support for the construction of confidence intervals. A Monte Carlo simulation is performed to evaluate the performance of the approach under various conditions. Finally, the method is applied to identify the complexity of two real data sets. The overall beneficial effect of live black soldier fly larvae (BSFL) on the welfare of broiler chickens, turkeys, and laying hens has already been discussed in the literature. However, scant information is available regarding the benefits of feeding live BSFL to medium‐growing chicken hybrids reared under organic/free‐range conditions, and whose welfare is frequently cited as being inadequate. The aim of this research was to advance our knowledge of this topic. To this end, 240 label naked neck birds (Hubbard JA57 hybrid) were assigned, at 21 days of age, to four experimental groups (6 replicates/treatment, 10 chickens/replicate), created according to sex (M/F) and the provision of a 10% live BSFL dietary supplementation (control males, control females, larvae males, and larvae females), and raised until 82 days of age. We performed behavioural observations, a tonic immobility test, and an avoidance distance (AD) test. We assessed feather damage and cleanliness, hock burn, footpad dermatitis, and skin lesion scores, and determined the concentration of excreta corticosterone metabolites (ECM) and the heterophile to lymphocyte heterophile/lymphocyte (H/L) ratio. The behavioural observations demonstrated increased physical and foraging activity (p < 0.05) in the live BSFL administered groups compared with C ones, providing valuable data on the explorative and recreational behaviour of this chicken genotype. The results also evidenced the usefulness of live BSFL as a fear reducer in females, as those receiving the BSFL supplement moved closer to the operator during the AD test (p < 0.01). No physical injuries or damage were observed on the birds, regardless of whether they received the BSFL supplementation or not. The ECM were unaffected by BSFL supplementation, while the H/L ratio was higher in the larvae groups than in the control ones (p = 0.050). In conclusion, live BSFL provision could constitute a powerful tool for improving life quality in medium‐growing chickens. A popular form of neoliberal feminism seeks to advance gender equality in leadership and beyond by encouraging women to be resilient as individuals. By locating career advancement as within an individual's control, recent research has shown that this focus subtly shifts the blame for gender inequality onto women and reduces support for needed structural changes to tackle gender discrimination. We extend research into neoliberal feminism by examining anticipated negative effects on women's protest motivation. Across four studies in the United Kingdom (total N = 1,168), undergraduate women students and employed women with university degrees in both the control and resilience conditions first read about gender inequality. Participants in the neoliberal feminist conditions then read messages promoting individual resilience as key to women's advancement (Study 1–3) or participated in activities designed to build their own resilience as individuals to help them advance (Study 4). In Studies 1, 3, and 4, participants in the neoliberal feminist conditions compared to the control had lower collective action intentions – a negative effect that was either indirect, via reduced perceptions of gender discrimination (Studies 1 and 4) and reduced anger over inequality (Study 1), or direct (Study 3). Together, these studies provided partial support for our hypothesis that neoliberal feminism can undermine women's protest motivation. Future research can help establish how contextual and other factors contribute to the strength of these effects and explore how feminists can better harness messages of resilience. To advance gender equality, our findings suggest that advocates should focus less on individualistic solutions and more on addressing structural barriers, laying the groundwork for effective protest action and social change. The Average Oracle, a simple and very fast covariance filtering method, is shown to yield superior Sharpe ratios than the current state-of-the-art (and complex) methods, Dynamic Conditional Covariance coupled to Non-Linear Shrinkage (DCC-NLS). We pit all the known variants of DCC-NLS (quadratic shrinkage, gross-leverage or turnover limitations, and factor-augmented NLS) against the Average Oracle in large-scale randomized experiments. We find generically that while some variants of DCC-NLS sometimes yield the lowest average realized volatility, albeit with a small improvement, their excessive gross leverage and investment concentration, and their 10-time larger turnover contribute to smaller average portfolio returns, which mechanically result in smaller realized Sharpe ratios than the Average Oracle. The Hyphomicrobiales (Rhizobiales) order contains soil bacteria with an irregular distribution of the Calvin–Benson–Bassham cycle (CBB). Key enzymes in the CBB cycle are ribulose 1,5-bisphosphate carboxylase/oxygenase (RuBisCO), whose large and small subunits are encoded in cbbL and cbbS, and phosphoribulokinase (PRK), encoded by cbbP. These genes are often found in cbb operons, regulated by the LysR-type regulator CbbR. In Bradyrhizobium, pertaining to this order and bearing photosynthetic and non-photosynthetic species, the number of cbbL and cbbS copies varies, for example: zero in B. manausense, one in B. diazoefficiens, two in B. japonicum, and three in Bradyrhizobium sp. BTAi. Few studies addressed the role of CBB in Bradyrhizobium spp. symbiosis with leguminous plants. To investigate the horizontal transfer of the cbb operon among Hyphomicrobiales, we compared phylogenetic trees for concatenated cbbL-cbbP-cbbR and housekeeping genes (atpD-gyrB-recA-rpoB-rpoD). The distribution was consistent, indicating no horizontal transfer of the cbb operon in Hyphomicrobiales. We constructed a ΔcbbLS mutant in B. diazoefficiens, which lost most of the coding sequence of cbbL and has a frameshift creating a stop codon at the N-terminus of cbbS. This mutant nodulated normally but had reduced competitiveness for nodulation and long-term adhesion to soybean (Glycine max (L.) Elite athletes in speed roller skates perceive skating to be a more demanding exercise for the groin when compared to other cyclic disciplines, increasing their risk of injury. The objective of this study was to monitor the kinematic and electromyographic parameters of roller speed skaters, linearly, on a treadmill, and to compare different skating speeds, one at 20 km/h and one at 32 km/h, at a 1° inclination. The acquisition was carried out by placing an inertial sensor at the level of the first sacral vertebra, and eight surface electromyographic probes on both lower limbs. The kinematic and electromyographic analysis on the treadmill showed that a higher speed requires more muscle activation, in terms of maximum and average values and co-activation, as it not only increases the intrinsic muscle demand in the district, but also the athlete's ability to coordinate the skating technique. The present study allows us to indicate not only how individual muscle districts are activated during skating on a surface different from the road, but also how different speeds affect the overall district load distributions concerning effective force, which is essential for the physiotherapist and kinesiologist for preventive and conditional purposes, while also considering possible variations in the skating technique in linear advancement. In this work we examined the contents of 14 polyphenols in white lupin (Lupinus albus L.) samples cultivated in Southern Italy by the optimisation and validation of a LC-HRMS method. The validation of the LC-HRMS method showed linearity results r2 > 0.989 and recovery values between 71 and 119% for a very wide range of concentrations. Ellagic acid was the most abundant polyphenol, with mean concentrations of 16271.86 ± 19798.53 μg/Kg, followed by apigenin (2749.51 ± 889.95 μg/Kg). A significant variability in ellagic acid contents was found between the areas of cultivation examined (p < 0.05). As far as we know, this work provides the first data on the polyphenols contents of white lupins cultivated in Italy. The comparison with other study confirms the role of the cultivation area for the determination of the polyphenol's contents. We adopt a Bayesian X-ray spectral approach to investigate the accretion properties of unobscured (⁠|$20\lt \log (N_{\rm H}/{\rm cm}^{-2}\lt 22$|⁠) and obscured (⁠|$22\lt \log (N_{\rm H}/{\rm cm}^{-2}\lt 24$|⁠) active galactic nuclei (AGNs) to shed light on the orientation versus evolution scenarios for the origin of the obscuring material. For a sample of 3882 X-ray-selected AGN from the Chandra COSMOS Legacy, AEGIS, and CDFS extragalactic surveys, we constrain their stellar masses, |$M_\star$|⁠ , intrinsic X-ray luminosities, |$L_{\rm X}$|⁠ , obscuring column densities, |$N_{\rm H}$|⁠ , and specific accretion rates |$\lambda \propto L_{\rm X}/M_\star$|⁠. By combining these observables within a Bayesian non-parametric approach, we infer, for the first time, the specific accretion rate distribution (SARD) of obscured and unobscured AGN to |$z\approx 3$|⁠ , i.e. the probability of a galaxy with mass |$M_\star$| at redshift z hosting an AGN with column density |$N_{\rm H}$| and specific accretion rate |$\lambda$|⁠. Our findings indicate that (1) both obscured and unobscured SARDs share similar shapes, shifting towards higher accretion rates with redshift, (2) unobscured SARDs exhibit a systematic offset towards higher |$\lambda$| compared to obscured SARD for all redshift intervals, (3) the obscured AGN fraction declines sharply at |$\log \lambda _{\rm break} \sim -2$| for |$z \lt 0.5$|⁠ , but shifts to higher |$\lambda$| values with increasing redshift, (4) the incidence of AGN within the theoretically unstable blow-out region of the |$\lambda -N_{\rm H}$| plane increases with redshift. These observations provide compelling evidence for AGN 'downsizing' and radiation-regulated nuclear-scale obscuration with an increasing host galaxy contribution towards higher redshifts. Digital polymerase chain reaction (dPCR) has emerged as a groundbreaking technology in molecular biology and diagnostics, offering exceptional precision and sensitivity in nucleic acid detection and quantification. This review highlights the core principles and transformative potential of dPCR, particularly in infectious disease diagnostics and environmental surveillance. Emphasizing its evolution from traditional PCR, dPCR provides accurate absolute quantification of target nucleic acids through advanced partitioning techniques. The review addresses the significant impact of dPCR in sepsis diagnosis and management, showcasing its superior sensitivity and specificity in early pathogen detection and identification of drug-resistant genes. Despite its advantages, challenges such as optimization of experimental conditions, standardization of data analysis workflows, and high costs are discussed. Furthermore, we compare various commercially available dPCR platforms, detailing their features and applications in clinical and research settings. Additionally, the review explores dPCR's role in water microbiology, particularly in wastewater surveillance and monitoring of waterborne pathogens, underscoring its importance in public health protection. In conclusion, future prospects of dPCR, including methodological optimization, integration with innovative technologies, and expansion into new sectors like metagenomics, are explored. Cu2ZnSnSe4 (CZTSe) nanoparticles (NPs) were successfully synthesized via a solvothermal method. Their structural, compositional, morphological, optoelectronic, and electrochemical properties have been characterized by X-ray diffraction (XRD), Raman spectroscopy, X-ray photoelectron spectroscopy (XPS), Field-emission scanning electron microscopy (FE-SEM), transmission electron microscope (TEM), UV–vis absorption spectroscopy, and electrochemical impedance spectroscopy (EIS) techniques. Porosimetry and specific surface area in terms of the Brunauer–Emmett–Teller (BET) technique have also been studied. XRD indicates the formation of a polycrystalline kesterite CZTSe phase. Raman peaks at 173 and 190 cm−1 confirm the formation of a pure phase. TEM micrographs revealed the presence of nanoparticles with average sizes of ~90 nm. A BET surface area of 7 m2/g was determined. The CZTSe NPs showed a bandgap of 1.0 eV and a p-type semiconducting behavior. As a proof of concept, for the first time, the CZTSe NPs have been used as a visible-light-driven photocatalyst to Congo red (CR) azo dye degradation. The nanophotocatalyst material under simulated sunlight results in almost complete degradation (96%) of CR dye after 70 min, following a pseudo-second-order kinetic model (rate constant of 0.334 min−1). The aim of this study was to assess the nutraceutical qualities of extra virgin olive oil (EVOO) samples obtained from three Sicilian olive cultivars: Nocellara, Biancolilla, and Cerasuola. We also evidenced the relationship among biophenols, base parameters and panel test scores, and evaluated the stability of the biophenols in EVOO. The assessment also took into consideration variations in olive harvesting periods and the influence of four different milling methods. A statistical analysis of the collected data revealed that the cultivar and harvesting period were the primary factors influencing the bio-phenol content, while the milling methods employed did not significantly affect the levels of biophenols in the oils. The panel test results were also illuminating as they were strongly related to the cultivar and polyphenol content. Following the criteria outlined in EC Regulation 432/2012, we selected three samples, each representing one of the cultivars, which exhibited the highest bio-phenol content to evaluate the biophenol stability during a time span of 16 months. We report the identification of 64 new candidates of compact galaxies, potentially hosting faint quasars with bolometric luminosities of Lbol = 1043–1046 erg s−1, residing in the reionization epoch within the redshift range of 6 ≲ z ≲ 8. These candidates were selected by harnessing the rich multiband datasets provided by the emerging JWST-driven extragalactic surveys, focusing on COSMOS-Web, as well as JADES, UNCOVER, CEERS, and PRIMER. Our search strategy includes two stages: applying stringent photometric cuts to catalog-level data and detailed spectral energy distribution fitting. These techniques effectively isolate the quasar candidates while mitigating contamination from low-redshift interlopers, such as brown dwarfs and nearby galaxies. The selected candidates indicate physical traits compatible with low-luminosity active galactic nuclei, likely hosting ≈105–107M⊙ supermassive black holes (SMBHs) living in galaxies with stellar masses of ≈108–1010M⊙. The SMBHs selected in this study, on average, exhibit an elevated mass compared to their hosts, with the mass ratio distribution slightly higher than those of galaxies in the local Universe. As with other high-z studies, this is at least in part due to the selection method for these quasars. An extensive Monte Carlo analysis provides compelling evidence that heavy black hole seeds from the direct collapse scenario appear to be the preferred pathway to mature this specific subset of SMBHs by z ≈ 7. Notably, most of the selected candidates might have emerged from seeds with masses of ∼105M⊙, assuming a thin disk accretion with an average Eddington ratio of fEdd = 0.6 ± 0.3 and a radiative efficiency of ϵ = 0.2 ± 0.1. This work underscores the significance of further spectroscopic observations, as the quasar candidates presented here offer exceptional opportunities to delve into the nature of the earliest galaxies and SMBHs that formed during cosmic infancy. Respiratory syncytial virus (RSV) bronchiolitis remains a significant global health burden, particularly in newborns and infants during their first year of life. The quest for an effective preventive strategy against RSV has long been sought, and recent developments have shown promise in the form of nirsevimab, a monoclonal antibody specifically designed for RSV prophylaxis. Valle d'Aosta was the first Italian region to propose universal prophylaxis with nirsevimab for newborns and infants in their first epidemic season as early as 2023–2024. This study describes the effectiveness and safety of the universal prevention program of RSV bronchiolitis using the monoclonal antibody nirsevimab in children resident in Valle d'Aosta born during the 2023–2024 epidemic season. There were 556 neonates born from 1 May 2023 to 15 February 2024. The risk of hospitalization for RSV bronchiolitis in 2023–2024 was 3.2%, compared to 7% in the 2022–2023 epidemic season (p < 0.001). After the start of the prophylaxis campaign with nirsevimab, the risk of hospitalization was 8.3% in the sample of infants who did not adhere to the prophylaxis, while no child in the sample of those treated (p < 0.001) was hospitalized for bronchiolitis. Few mild transient side effects were reported. This study shows the efficacy and safety of universal prophylaxis with nirsevimab in neonates, making Valle d'Aosta the first Italian region to offer universal prophylaxis to newborns without risk factors for RSV complications. Objectives: Carbapenem-resistance is a challenging healthcare concern and require specific stewardship programs. Monitoring workflows include the identification from surveillance samples, such as rectal swabs. Although culture assays represent the gold standard, data report a significant effectiveness in detecting carbapenemases genes directly from rectal swabs. The aim of this study was to evaluate the REALQUALITY Carba-Screen kit (AB ANALITICA, Padova, Italy) in detecting carbapenemases genes directly from rectal swabs, also comparing its effectiveness to culture assays results. A next-generation sequencing (NGS) was performed to investigate the positive samples about resistance markers and sequence type (ST). Methods: A number of 136 rectal swabs were collected from the University Hospital Policlinico of Catania critical wards. The samples simultaneously underwent culture and molecular assays (REALQUALITY Carba-Screen kit). The molecular method included two-steps. The first step (1 h and 6 min) rapidly excluded negative samples, while the second one (1 h and 6 min) included only positive samples for a resistance confirmation. All the positive culture samples underwent NGS analysis. Results: Statistical evaluations demonstrated high sensitivity (100%) and detection rates (92.6%) for the REALQUALITY Carba-Screen kit, which mostly correlated to the standard workflow. All the culture positive results matched the positive molecular results, which were mainly confirmed by the NGS resistome analysis. The identified ST appeared to be diversified and different from the clinically significative strains of the same setting, furnishing interesting epidemiological evidence. Conclusion: The molecular detection allowed a coordinate approach in a high-prevalence multi-drug-resistance area. The rapid identification with a multi-step procedure accelerated the infection control procedures, while the preliminary negative results reduced the overtreatment episodes. Structure and functions of S100 proteins are regulated by two distinct calcium binding EF hand motifs. In this work, we used solution‐state NMR spectroscopy to investigate the cooperativity between the two calcium binding sites and map the allosteric changes at the target binding site. To parse the contribution of the individual calcium binding events, variants of S100A12 were designed to selectively bind calcium to either the EF‐I (N63A) or EF‐II (E31A) loop, respectively. Detailed analysis of the backbone chemical shifts for wildtype protein and its mutants indicates that calcium binding to the canonical EF‐II loop is the principal trigger for the conformational switch between 'closed' apo to the 'open' Ca2+‐bound conformation of the protein. Elimination of binding in S100‐specific EF‐I loop has limited impact on the calcium binding affinity of the EF‐II loop and the concomitant structural rearrangement. In contrast, deletion of binding in the EF‐II loop significantly attenuates calcium affinity in the EF‐I loop and the structure adopts a 'closed' apo‐like conformation. Analysis of experimental amide nitrogen (15N) relaxation rates (R1, R2, and 15N–{1H} NOE) and molecular dynamics (MD) simulations demonstrate that the calcium bound state is relatively floppy with pico–nanosecond motions induced in functionally relevant domains responsible for target recognition such as the hinge domain and the C‐terminal residues. Experimental relaxation studies combined with MD simulations show that while calcium binding in the EF‐I loop alone does not induce significant motions in the polypeptide chain, EF‐I regulates fluctuations in the polypeptide in the presence of bound calcium in the EF‐II loop. These results offer novel insights into the dynamic regulation of target recognition by calcium binding and unravels the role of cooperativity between the two calcium binding events in S100A12. In this work, a colorimetric indicator based on gold nanoparticles (AuNP) and a biodegradable and eco-friendly polymer (sodium alginate, Alg.), was developed for the real-time detection of fish spoilage products. The AuNPs and the colorimetric indicator were characterized using UV-VIS, FTIR spectroscopies, TGA, DSC, XRD, TEM, and colorimetry. The UV-VIS spectrum and TEM showed the successful synthesis, the spherical shape, and the size of AuNPs. The results indicated color changes of the indicator in packaged fish on day 9 of storage at a refrigerated temperature (5 °C. While much progress has been made towards gender equality, diversity and inclusion in the workplace, education and society, recent years have also revealed continuing challenges that slow or halt this progress. To date, the majority of gender equality action has tended to approach gender equality from one side: being focused on the need to remove barriers for girls and women. We argue that this is only half the battle, and that a focus on men is MANdatory, highlighting three key areas: First, we review men's privileged status as being potentially threatened by progress in gender equality, and the effects of these threats for how men engage in gender-equality progress. Second, we highlight how men themselves are victims of restrictive gender roles, and the consequences of this for men's physical and mental health, and for their engagement at work and at home. Third, we review the role of men as allies in the fight for gender equality, and on the factors that impede and may aid in increasing men's involvement. Hole‐transporting layer‐free mesoporous carbon (mC) architectures represent a printable, low‐cost, and stable solution for the future commercialization of perovskite solar cells (PSCs). CsPbI3 perovskite is attracting attention for its inorganic structure, which yields higher structural stability compared to hybrid counterparts and allows reversibility of its photoactive phase. Here the photovoltaic performance of large‐area (144 mm2) devices infiltrated with CsPbI3:EuCl3 is systematically evaluated, using AVA‐MAPbI3 mC‐PSCs as a reference. Measured and simulated J–V curves acquired at various scan rates show a significantly reduced hysteresis for Eu‐doped CsPbI3 with respect to AVA‐MAPbI3 mC‐PSCs. The synergic comparison between experiments and simulations reveals the complex interplay between ionic and electronic charges in the two mC‐PSCs, supporting the argument that cation migration is suppressed in CsPbI3:EuCl3. This also agrees with steady‐state photoconversion efficiency measured and simulated at fixed bias, which is constant over time in CsPbI3:EuCl3, contrary to what occurs in AVA‐MAPbI3 where a decay arises from enhanced ionic migration. In addition, CsPbI3:EuCl3 mC‐PSCs maintain their initial efficiency up to 250 h at 55 °C under continuous illumination during maximum power point tracking measurements. Intracellular survival and immune evasion are typical features of staphylococcal infections. USA300 is a major clone of methicillin-resistant S. aureus (MRSA), a community- and hospital-acquired pathogen capable of disseminating throughout the body and evading the immune system. Carnosine is an endogenous dipeptide characterized by antioxidant and anti-inflammatory properties acting on the peripheral (macrophages) and tissue-resident (microglia) immune system. In this work, RAW 264.7 murine macrophages were infected with the USA300 ATCC BAA-1556 S. aureus strain and treated with 20 mM carnosine and/or 32 mg/L erythromycin. Stable small colony variant (SCV) formation on blood agar medium was obtained after 48 h of combined treatment. Whole genome sequencing of the BAA-1556 strain and its stable derivative SCVs when combining Illumina and nanopore technologies revealed three single nucleotide differences, including a nonsense mutation in the shikimate kinase gene aroK. Gene expression analysis showed a significant up-regulation of the uhpt and sdrE genes in the stable SCVs compared with the wild-type, likely involved in adaptation to the intracellular milieu. Extracellular vesicles (EVs) are lipid bilayers derived from cell membranes, released by both eukaryotic cells and bacteria into the extracellular environment. During production, EVs carry proteins, nucleic acids, and various compounds, which are then released. While Gram-positive bacteria were traditionally thought incapable of producing EVs due to their thick peptidoglycan cell walls, recent studies on membrane vesicles (MVs) in Gram-positive bacteria have revealed their significant role in bacterial physiology and disease progression. This review explores the current understanding of MVs in Gram-positive bacteria, including the characterization of their content and functions, as well as their interactions with host and bacterial cells.
