Purpose: To study the positivity rate of conjunctival realtime polymerase chain reaction (RT-PCR) testing for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Design: Systematic review and diagnostic accuracy meta-analysis. Methods: MEDLINE and EMBASE were queried using medical subject headings terms. Diagnostic accuracy meta-analyses and forest plots were obtained using the RevMan software. Results: After deduplication, appraisal of abstract titles and full-text analysis of 1441 articles, 42 articles with 3351 COVID-19 patients were included in this review. Of these, 412 conjunctival swabs/Schirmer paper strips tested positive for SARS-CoV-2 by RT-PCR. The pooled sensitivity of the RT-PCR tests across the 24 studies with laboratory-confirmed COVID-19 patients was 10.3%. Conclusions: Only 1 in 10 RT-PCR tests performed on conjunctival swabs were positive for SARS-CoV-2. Although this suggests that SARS-CoV-2 is likely present and detectable in the conjunctiva, this detection method has low diagnostic potential. [ABSTRACT FROM AUTHOR]

Vegetative propagation of potatoes makes it possible for potato viruses to be transmitted through tubers. Potato virus A (PVA) is one of these viruses, which belongs to the Potyvirus genus in the Potyviridae family. Potato tuber yield can be reduced by 30–40% by PVA alone. Losses can be further exacerbated by potato virus X and/or potato virus Y infection. PVA is transmitted primarily by several species of aphids in non-persistent manner. With the aim of resolving this problem, we developed one-step reverse transcription-recombinase polymerase amplification (RT-RPA), a highly sensitive and cost-effective method for detecting PVA in both potato tubers and leaves. Detection and amplification are performed using isothermal conditions in this method. There was good amplification of the coat protein gene in PVA with all three primers tested. To conduct this study, a primer set that can amplify specific 185 base pair (bp) product was selected. PVA detection was optimized by 30-min amplification reactions, which showed no cross-reactivity with other potato viruses. A simple heating block or water bath was used to amplify PVA product using RT-RPA at a temperature range of 38–42 °C. In comparison to conventional reverse transcription-polymerase chain reaction (RT-PCR), the newly developed RT-RPA protocol exhibited high sensitivity for both potato leaves and tuber tissues. Using cellular paper-based simple RNA extraction procedure, the virus was detected in leaf samples as efficiently as purified total RNA. We also found that combining LiCl-based RNA precipitation with cellular paper discs allowed us to successfully optimize RNA extraction for one-step RT-RPA for detecting PVA in tubers. Tests using this simplified one-step RT-RPA method were successfully applied to 300 samples of both leaves and tubers from various potato cultivars. In our knowledge, this is the first report of an RT-RPA assay utilizing simple RNA obtained from either cellular disc paper or LiCl coupled with cellular disc paper to detect PVA. As a result, this method was equally sensitive and specific for detecting PVA in potatoes. The developed RT-RPA assay is more versatile, durable, and do not require highly purified RNA templates, thus providing an effective alternative to RT-PCR assays for screening of germplasm, certifying planting materials, breeding for virus resistance, and real-time monitoring of PVA. Background: Reverse transcription polymerase chain reaction (RT-PCR) is the main technique used to identify COVID-19 from respiratory samples. It has been suggested in several articles that chest CTs could offer a possible alternate diagnostic tool for COVID-19; however, no professional medical body recommends using chest CTs as an early COVID-19 detection modality. This literature review examines the use of CT scans as a diagnostic tool for COVID-19. Method: A comprehensive search of research works published in peer-reviewed journals was carried out utilizing precisely stated criteria. The search was limited to English-language publications, and studies of COVID-19-positive patients diagnosed using both chest CT scans and RT-PCR tests were sought. For this review, four databases were consulted: these were the Cochrane and ScienceDirect catalogs, and the CINAHL and Medline databases made available by EBSCOhost. Findings: In total, 285 possibly pertinent studies were found during an initial search. After applying inclusion and exclusion criteria, six studies remained for analysis. According to the included studies, chest CT scans were shown to have a 44 to 98% sensitivity and 25 to 96% specificity in terms of COVID-19 diagnosis. However, methodological limitations were identified in all studies included in this review. Conclusion: RT-PCR is still the suggested first-line diagnostic technique for COVID-19; while chest CT is adequate for use in symptomatic patients, it is not a sufficiently robust diagnostic tool for the primary screening of COVID-19. Background: Newcastle disease significantly impacts the global poultry industry and is prevalent in many African countries, including Ethiopia. The objective of this research is to determine the humoral immune response to Newcastle Disease Virus (NDV), identify the circulating NDV genotype, and evaluate the correlation between the diagnostic tests used in backyard chickens in the Jimma Zone, southwest Ethiopia. Methods: A total of 90 swab and blood samples were purposively collected from symptomatic backyard chicken in the period between February and April 2022. Samples were collected from Jimma town, Seqa Chekorsa and Tiro Afeta districts of Jimma zone. Enzyme linked immunosorbent assay (ELISA) was conducted and seropositivity was determined from the collected serum samples. From the swab samples, total RNA was extracted and the viral genomic material was detected by amplifying the Fusion gene by Reverse transcription polymerase chain reaction (RT-PCR). The interconnection between ELISA and RT-PCR was also analyzed. Further, positive swab samples were nucleotide sequenced and genotyped. Results: Of the 90 serum samples, 62 (68.8%) were seropositive. From the 90 swab samples, 14 (15.5%) were RT-PCR positive. No statistically significant association between risk factors (breed, age, sex) and virus exposure was observed by RT-PCR (P = 0.41, 0.44, 0.67) or ELISA (P = 0.85, 0.19, 0.11). However, local breeds, young, and male birds were at higher risk according to RT-PCR results, while young and female birds were more likely to be seropositive. The antibody titer study showed that RT-PCR positive birds produced less than half the mean antibodies of negative birds (x̄=854 vs. 1885) and positive birds produced similar amount of antibody (σ = 626). Local (x̄=1978), adult (x̄=2558), and female (x̄=2620) birds had higher mean antibody titers than their counterparts. The agreement between RT-PCR and ELISA in identifying positive samples was minimal (k = 0.05). Nucleotide-sequenced isolates were nearly identical (99.7%) to each other and identified as velogenic based on the F-gene cleavage site (RRQKRF) with genotype VII.1.1. Conclusion: NDV was circulating in the study area, infecting birds with low antibody titers. Background/Objectives: Conventional live oral poliovirus vaccines (OPVs) effectively prevent poliomyelitis. These vaccines are derived from three attenuated Sabin strains of poliovirus, which can revert within the first week of replication to a neurovirulent phenotype, leading to sporadic cases of vaccine-associated paralytic poliomyelitis (VAPP) among vaccinees and their contacts. A novel OPV2 vaccine (nOPV2) with enhanced genetic stability was developed recently; type 1 and type 3 nOPV strains were engineered using the nOPV2 genome as a backbone by replacing the capsid precursor polyprotein (P1) with that of Sabin strains type 1 and type 3, respectively. The nOPV vaccines have a high degree of sequence homology with the parental Sabin 2 genome, and some manufacturing facilities produce and store both Sabin OPV and nOPV. Therefore, detecting Sabin virus contaminations in nOPV lots is crucial. Methods: This study describes the development of pan quantitative reverse transcription polymerase chain reaction (panRT-PCR) and multiplex one-step RT-PCR (mosRT-PCR) assays for the straightforward detection and identification of contaminating Sabin viruses when present in significantly higher amounts of nOPV strains. Results: The two assays exhibit high specificity, reproducibility, and sensitivity to detect 0.0001% and 0.00001% of Sabin viruses in nOPV, respectively. Additionally, an analysis of 12 trivalent nOPV formulation lots using both methods confirmed that the nOPV lots were free from Sabin virus contamination. MicroRNAs (miRNAs) are typically non-coding RNAs of 18–26 nucleotides (nts) that are produced endogenously and regulated post-transcriptionally through degradation or translational repression. Since miRNAs are evolutionarily conserved, their preservation is essential for important regulatory functions in plant development, growth, and responses to environmental stress. Sorghum bicolor (sbi) is a valuable food and fodder crop which is grown worldwide. A range of sbi miRNAs were identified so far as being connected to plant development and stress responses. Herein, we employed a variety of bioinformatics tools for miRNA profiling in sbi and a PCR-based platform for the validation of these miRNAs. In total, 74 new conserved sbi miRNAs from 52 miRNA families have been predicted. Using the psRNA Target method, 10613 different protein targets of these predicted miRNAs have been attained. These targets include 54 GO-terms which have substantial targets in the biological, molecular, and cellular processes. We particularly found that the sbi-miR1861c and sbi-miR5050 are involved to regulate sulphur compound biosynthetic process, while the significant spliceosomal complex is regulated by sbi-miR815b and sbi-miR7768b. Also, we report that the pre-ribosome, electron transport chain, cell communication, cellular respiration, protein localization, and photosynthesis are controlled by sbi-miR2907b, sbi-miR530, sbi-miR7749, sbi-miR1858a, sbi-mi7729a, and sbi-miR417, respectively. Grapevine rupestris stem pitting-associated virus (GRSPaV; Foveavirus rupestris, Betaflexiviridae) is the most widespread grapevine virus in most viticulture regions, and this virus has high genetic variability. Genetic diversity of GRSPaV in Algeria was examined, based on the complete capsid protein (CP) gene. Similarity proportions between the CP sequences varied from 76% to 99%. The complete coding sequence of Algerian GRSPaV isolate ALG99 (8,646 nt) was determined using highthroughput sequencing and bioinformatic analyses. Algerian GRSPaV isolates clustered into four groups, with most sequences in groups III and IV. This research is the first to determine the genetic variability of GRSPaV in Algeria. Background: Allium cepa is one of the major vegetable crops cultivated in Saudi Arabia. This economically important crop faces several biotic stresses which adversely affect the quality and quantity of its production. Several virus groups (potyviruses, carlaviruses, tospoviruses, and allexiviruses) have been reported infecting Allium crops. Methods: During the growing season of 2021-2022, a total of 81 onion samples exhibiting virus-like symptoms were collected from two different geographical regions in Saudi Arabia. The serological technique (ELISA) was used to detect the important allexiviruses and potyviruses. RT-PCR amplification of partial genome sequence was done using degenerate primers for allexiviruses and potyviruses and the phylogenetic trees were constructed using different bioinformatic tools. Results: The results obtained from ELISA tests showed that 26% and 32% of onion samples were positive with both Garlic Virus A (GarV-A) and Onion yellow dwarf virus (OYDV) respectively. RT-PCR amplification and sequencing results showed that two allexiviruses, GarV-A, garlic virus D (GarV-D), and one Potyvirus (OYDV) were detected in both regions. Sequence data were deposited in the GenBank database with accession numbers, OQ397545, OQ397546 for GarV-A, OQ397547 for GarV-D, and OQ397548, OQ397549 for OYDV, sequentially. Phylogenetic tree analysis of these virus isolates showed making clades with closely related isolates of their respective viruses. Pairwise nucleotide sequence identity showed their similarity with GarVA, GarV-D, and OYDV isolates reported earlier in the GenBank. Conclusion: To the best of our knowledge, these two distinct allexiviruses (GarV-A, GarV-D) and one Potyvirus (OYDV) were isolated for the first time from an onion crop in Saudi Arabia. Sweet potato (Ipomoea batatas L. Lam.) is a major source of food in many parts of Ethiopia. In recent years, viral diseases have become the main threat to sweet potato production in Ethiopia. Previous virus survey studies carried out from 1986 to 2020 reported eight viruses infecting sweet potato in Ethiopia. Consequently, obtaining and multiplying virus-free planting materials have been difficult for farmers and commercial multipliers. This study was conducted to detect viruses infecting the five sweet potato varieties used as source plants and compare the virus elimination efficiency between meristem cultures from untreated and heat-treated mother plants and production of virus-free sweet-potato-planting materials. Seven common viruses were tested for, using grafting to Ipomoea setosa, enzyme-linked immunosorbent assay (ELISA) and reverse-transcription polymerase chain reaction (RT–PCR) before and after elimination procedures as screening and confirmatory methods. The sweet potato feathery mottle virus (SPFMV) elimination efficiencies of meristem cultures from untreated (grown at 25 ± 1 °C) and heat-treated (grown at 39 ± 1 °C) potted plants of sweet potato varieties were evaluated and compared. Sweet potato feathery mottle virus (SPFMV) was detected in 12 of the 15 source plants tested. Triple infections of SPFMV, sweet potato chlorotic stunt virus (SPCSV), and sweet potato virus C (SPVC) were detected in one of the fifteen plants. This study reports the detection of SPVC for the first time in sweet potato plants from Ethiopia. The cutting of meristems from heat-treated plants further increased the percentage of virus-free plantlets by ca 10% to ca 16%, depending on the plant variety. Elimination efficiency also seemed to vary among varieties: the greatest difference was observed for 'Tola', and the least difference was observed for 'Guntute'. Introduction: SARS-CoV-2, seasonal influenza, and respiratory syncytial virus (RSV) are major causes of acute respiratory infections in all age groups and responsible for an enormous socio-economic burden. The recently coined term 'tripledemic' describes co-circulation of these three viruses, a novel epidemiological paradigm that poses profound public health implications. Areas covered: Real-time reverse transcription polymerase chain reaction (RT-PCR) is now considered the reference method for the diagnosis of SARS-CoV-2, influenza, and RSV infections. Syndromic-based multiplex RT-PCR panels that simultaneously detect several respiratory viruses have become increasingly common. This review explores available molecular diagnostics (MDx) platforms for the diagnosis of SARS-CoV-2, influenza, and RSV in the same biological sample. Within some limitations of the published validation and diagnostic accuracy studies, both laboratory-based and point-of-care multiplex panels proved highly performant in identifying SARS-CoV-2, influenza A, influenza B, and RSV. Improved operational efficiency and faster turnaround times make these assays potentially cost-effective or even cost-saving. Expert opinion: The adoption of multiplex MDx assays for the contemporary detection of SARS-CoV-2, influenza, RSV, and other respiratory pathogens will likely increase in the next few years. NTRK gene fusions have been detected in more than 25 types of tumors and their prevalence is approximately 0.3% in solid tumors. This low prevalence makes identifying patients who could benefit from TRK inhibitors a considerable challenge. Furthermore, while numerous papers on the evaluation of NTRK fusion genes are available, not all countries have guidelines that are suitable for their setting, as is the case with Latin America. Therefore, a group of oncologists and pathologists from several countries in Latin America (Argentina, Chile, Ecuador, Mexico, Peru and Uruguay) met to discuss and reach consensus on how to identify patients with NTRK gene fusions in solid tumors. Potatoes are developed vegetatively from tubers, and therefore potato virus transmission is always a possibility. The potato leafroll virus (PLRV) is a highly devastating virus of the genus Polerovirus and family Luteoviridae and is regarded as the second-most destructive virus after Potato virus Y. Multiple species of aphids are responsible for the persistent and non-propagating transmission of PLRV. Due to intrinsic tuber damage (net necrosis), the yield and quality are drastically diminished. PLRV is mostly found in phloem cells and in extremely low amounts. Therefore, we have attempted to detect PLRV in both potato tuber and leaves using a highly sensitive, reliable and cheap method of one-step reverse transcription-recombinase polymerase amplification (RT-RPA). In this study, an isothermal amplification and detection approach was used for efficient results. Out of the three tested primer sets, one efficiently amplified a 153-bp product based on the coat protein gene. In the present study, there was no cross-reactivity with other potato viruses and the optimal amplification reaction time was thirty minutes. The products of RT-RPA were amplified at a temperature between 38 and 42 °C using a simple heating block/water bath. The present developed protocol of one-step RT-RPA was reported to be highly sensitive for both leaves and tuber tissues equally in comparison to the conventional reverse transcription-polymerase chain reaction (RT-PCR) method. By using template RNA extracted employing a cellular disc paper-based extraction procedure, the method was not only simplified but it detected the virus as effectively as purified total RNA. The simplified one-step RT-RPA test was proven to be successful by detecting PLRV in 129 samples of various potato cultivars (each consisting of leaves and tubers). According to our knowledge, this is the first report of a one-step RT-RPA performed using simple RNA extracted from cellular disc paper that is equally sensitive and specific for detecting PLRV in potatoes. In terms of versatility, durability and the freedom of a highly purified RNA template, the one-step RT-RPA assay exceeds the RT-PCR assay, making it an effective alternative for the certification of planting materials, breeding for virus resistance and disease monitoring. To evaluate effectivity results among Nasopharyngeal, Throat, Saliva, and Nasal Swab Sample Media for Detection of SARS-Cov-2 virus using RT-PCR. SARS-CoV-2 is a coronavirus microorganism found in humans. A known viral infection causes the covid-19 disease to Severe Acute Respiratory Syndrome Coronavirus 2 (SARSCoV-2). Covid-19 has confused the public because of the different places where the samples were taken. Sampling was taken from the Nasopharynx, Throat, Saliva, and nasal Swab. This study used mini-review journals from several leading search engine journals such as PubMed, Elsevier, Jama Network, BMJ, Cochrane, Wiley, medRXiv, Lancet, and others, as well as from government websites such as WHO selected between 2020 and 2021 in the English language. Each sampling place has its advantages and disadvantages. Any place that is used as the gold standard is the nasal swab and nasopharyngeal. This paper attempts to compare the efficacy of four sample media to find the best method for detecting the SARS-CoV-2 virus. It is hoped that repeating this paper can make us aware of every method that we can use to detect the SARS-CoV-2 virus and reduce the spread of this virus, which is increasingly widespread. Overuse of artificial chemical fertilizers could be detrimental to the environment. Utilizing beneficial microorganisms as biofertilizers is a sustainable technique that promotes soil health, crop yield, and ecosystem preservation. Curcuma longa L. is utilized as a medication since it has its antibacterial, anti-microbial, and anti-tumor characteristics, which reduce inflammation and hasten wound healing. The effect of E. cloacae strain MSR1, which is common in the roots of alfalfa grown in the Al-Ahsaa region, on C. longa plants is being investigated. C. longa rhizomes were planted under greenhouse conditions after being submerged in a solution of E. cloacae strain MSR1 (OD 500) or water treatment as a control for 12 hours. After 240 days of growing, ten randomly selected plants from each treatment were collected, and the vegetative growth and yield metrics were assessed. To investigate how E. cloacae influences C. longa production and chemical composition (photosynthetic pigment, nitrogen, phosphorus, potassium, and curcuminoid), measurements were conducted as well as genes diketide-CoA and curcumin synthases genes. Our research showed that C. longa's growth and yield were favorably impacted by E. cloacae. Significant increases in the related plants' chlorophyll a,b, carotenoid, nitrogen, phosphorus, and potassium levels were likewise a reflection of the enhanced effects shown in the growth and yield parameters. Treatment with E. cloacae raised the curcuminoid's three sub-components' compositions to varying degrees: bisdemethoxycurcumin, demethoxycurcumin, and curcumin. Comparing E. cloacae treated plants to the control, high expression levels of the genes diketide-CoA and curcumin synthase-1, -2, and 3 were also found. The purpose of this study was to determine the impact of feeding time on NPY/AgRP protein and mRNA expression in the brain during prenatal and postnatal periods. Feeding at different times of the day during pregnancy may have lasting effects on the hypothalamic circuitry in offspring energy homeostasis. In the experimental design, adult female hamsters were randomly assigned to receive three different feeding conditions: ad libitum, night-time feeding and day-time feeding groups. After the lactation period, feeding regimens were continued for the offspring born from mothers who were given nutritional regimens, until they were 30 days old. RT-PCR for NPY/AgRP mRNA expression and ELISA analysis for protein levels were performed on hypothalamus tissues of hamsters at 10, 20 and 30 days of postnatal stage. There was no difference between the groups in terms of the daily measured food consumption of the offspring's. Body weights were significantly decreased in both night-time and day-time feeding groups compared to the ad libitum group (p<0.05). The highest increase in mRNA expression of NPY/AgRP was seen in the samples taken at 10. day of lactation in the night-time and day-time feeding groups of the offspring treated with three different feeding regimens. When the feeding regimes were compared, it was observed that the protein and mRNA expression of both NPY and AGRP increased the most in the offspring groups in which the feeding was restricted night -time only. Early stages of development have shown that maternal factors have significantly affected the offspring NPY and AgRP mRNA expression and protein levels. Titanium and its alloys are commonly used in dentistry for implants due to their strength, lightweight nature, durability, corrosion resistance, and biocompatibility. These implants can osseointegrate after surface treatments such as SLA, plasma-spray, and nanotubes, providing a stable foundation for prostheses. However, Candida albicans, an opportunistic fungal pathogen, can threaten the success of titanium dental implants, causing oral infections in vulnerable individuals. A dual novel silane blend of 3-acryloxypropyltrimethoxysilane (ACPS) and bis-1,2-(triethoxysilyl)ethane (BTSE) has been shown to improve the shear bond strength in resin cement bonds with titanium and ceramics. This study evaluated the effects of Candida albicans colonization on blended silane-coated SLA-Ti surfaces compared to non-coated SLA-Ti (positive control) and flat titanium (negative control). Candida albicans biofilms were cultured on all surfaces, and it was found that silane-coated SLA-Ti had significantly lower CFU counts than non-coated SLA-Ti. However, no significant differences were observed in the RT-PCR results. In conclusion, a combination of 1.0 vol% ACPS and 0.3 vol% BTSE shows promise as a silane coupling agent with potential antifungal properties for inhibiting Candida albicans proliferation. Recurrence of thrombotic events during aspirin therapy is known as aspirin resistance (AR). This study aimed to investigate the rate of AR, the factors influencing AR in patients with acute ischemic stroke under regular aspirin use, and the relationship between AR and ABCB1 (MDR-1) C3435T (rs1045642) polymorphism. Throughout this multicenter prospective study, 174 patients with acute ischemic stroke who had been prescribed aspirin for at least one month due to the risk of vascular disease, along with 106 healthy volunteers, were included as part of the study group. The results of our study indicate that AR was detected in 21.3% of the patient group. According to the results of an analysis of the polymorphism of the ABCB1 C3435T in patients with AR compared to those with aspirin sensitivity, patients with AR possessed more heterozygous (CT) and homozygous genotypes (TT) than those with aspirin sensitivity (p = 0.001). Based on multivariate logistic regression analysis of factors affecting AR in acute ischemic stroke patients, hypertension (OR: 5.679; 95% CI: 1.144–28.19; p = 0.034), heterozygous (CT) genotype (OR: 2.557; 95% CI: 1.126–5.807; p = 0.025), increased platelet values (OR: 1.005; 95% CI: 1.001–1.009; p = 0.029), and CRP/albumin values (OR: 1.547; 95% CI: 1.005–2.382; p = 0.047) were found to be associated with a greater risk of AR. The presence of heterozygous (CT) genotype in the ABCB1 C3435T gene region in the Turkish population is associated with an increased risk of AR. Cantú syndrome (CS), a multisystem disease with a complex cardiovascular phenotype, is caused by gain-of-function (GoF) variants in the Kir6.1/SUR2 subunits of ATP-sensitive potassium (KATP) channels and is characterized by low systemic vascular resistance, as well as tortuous, dilated, vessels, and decreased pulse-wave velocity. Thus, CS vascular dysfunction is multifactorial, with both hypomyotonic and hyperelastic components. To dissect whether such complexities arise cell autonomously within vascular smooth muscle cells (VSMCs) or as secondary responses to the pathophysiological milieu, we assessed electrical properties and gene expression in human induced pluripotent stem cell-derived VSMCs (hiPSC-VSMCs), differentiated from control and CS patient-derived hiPSCs, and in native mouse control and CS VSMCs. Whole-cell voltage clamp of isolated aortic and mesenteric arterial VSMCs isolated from wild-type (WT) and Kir6.1[V65M] (CS) mice revealed no clear differences in voltage-gated K+ (Kv) or Ca2+ currents. Kv and Ca2+ currents were also not different between validated hiPSC-VSMCs differentiated from control and CS patient-derived hiPSCs. While pinacidil-sensitive KATP currents in control hiPSC-VSMCs were similar to those in WT mouse VSMCs, they were considerably larger in CS hiPSC-VSMCs. Under current-clamp conditions, CS hiPSC-VSMCs were also hyperpolarized, consistent with increased basal K conductance and providing an explanation for decreased tone and decreased vascular resistance in CS. Increased compliance was observed in isolated CS mouse aortae and was associated with increased elastin mRNA expression. This was consistent with higher levels of elastin mRNA in CS hiPSC-VSMCs and suggesting that the hyperelastic component of CS vasculopathy is a cell-autonomous consequence of vascular KATP GoF. The results show that hiPSC-VSMCs reiterate expression of the same major ion currents as primary VSMCs, validating the use of these cells to study vascular disease. Results in hiPSC-VSMCs derived from CS patient cells suggest that both the hypomyotonic and hyperelastic components of CS vasculopathy are cell-autonomous phenomena driven by KATP overactivity within VSMCs. Overuse of artificial chemical fertilizers could be detrimental to the environment. Utilizing beneficial microorganisms as biofertilizers is a sustainable technique that promotes soil health, crop yield, and ecosystem preservation. Curcuma longa L. is utilized as a medication since it has its antibacterial, anti-microbial, and anti-tumor characteristics, which reduce inflammation and hasten wound healing. The effect of E. cloacae strain MSR1, which is common in the roots of alfalfa grown in the Al-Ahsaa region, on C. longa plants is being investigated. C. longa rhizomes were planted under greenhouse conditions after being submerged in a solution of E. cloacae strain MSR1 (OD 500) or water treatment as a control for 12 hours. After 240 days of growing, ten randomly selected plants from each treatment were collected, and the vegetative growth and yield metrics were assessed. To investigate how E. cloacae influences C. longa production and chemical composition (photosynthetic pigment, nitrogen, phosphorus, potassium, and curcuminoid), measurements were conducted as well as genes diketide-CoA and curcumin synthases genes. Our research showed that C. longa's growth and yield were favorably impacted by E. cloacae. Significant increases in the related plants' chlorophyll a,b, carotenoid, nitrogen, phosphorus, and potassium levels were likewise a reflection of the enhanced effects shown in the growth and yield parameters. Treatment with E. cloacae raised the curcuminoid's three sub-components' compositions to varying degrees: bisdemethoxycurcumin, demethoxycurcumin, and curcumin. Comparing E. cloacae treated plants to the control, high expression levels of the genes diketide-CoA and curcumin synthase-1, -2, and 3 were also found. The treatment of E. cloacae is a good biostimulant candidate for boosting growth and yield as well as raising the medicinal qualities of C. longa, according to the overall results. Albinism is a genetically heterogeneous disease in which 21 genes are known so far. Its inheritance mode is autosomal recessive except for one X-linked form. The molecular analysis of exonic sequences of these genes allows for about a 70% diagnostic rate. About half (15%) of the unsolved cases are heterozygous for one pathogenic or probably pathogenic variant. Assuming that the missing variant may be located in non-coding regions, we performed sequencing for 122 such heterozygous patients of either the whole genome (27 patients) or our NGS panel (95 patients) that includes, in addition to all exons of the 21 genes, the introns and flanking sequences of five genes, TYR, OCA2, SLC45A2, GPR143 and HPS1. Rare variants (MAF < 0.01) in trans to the first variant were tested by RT-PCR and/or minigene assay. Of the 14 variants tested, nine caused either exon skipping or the inclusion of a pseudoexon, allowing for the diagnosis of 11 patients. This represents 9.8% (12/122) supplementary diagnosis for formerly unsolved patients and 75% (12/16) of those in whom the candidate variant was in trans to the first variant. Of note, one missense variant was demonstrated to cause skipping of the exon in which it is located, thus shedding new light on its pathogenic mechanism. Summary. Sixty-five samples of grapevine from commercial vineyards in the Rasina district of Serbia were tested for the presence of grapevine Pinot gris virus (GPGV), using the reverse transcription-polymerase chain reaction. Fourteen samples of the grapevine varieties ‘Red Globe’, ‘Victoria’ and ‘Preobraženje’ were infected with GPGV. All the infected plants showed symptoms of leaf chlorotic mottling, puckering, and deformation, stunting, and reduced yields. The coding regions of the movement and coat protein (MP/CP) and a region of the RNA-dependent RNA polymerase (RdRP) domains of eight virus isolates were sequenced. Phylogenetic analyses of these genom- ic regions showed high nucleotide similarity among the Serbian GPGV isolates. This study is the first to describe genetic diversity of GPGV isolates in Serbia. Summary. Grapevine Pinot gris virus (GPGV) is a recently identified trichovirus infecting grapevines. Despite wide distribution, there is limited available information on epidemiology, transmission, and associated symptoms of grapevine leaf mottling and deformation. Occurrence and genetic diversity of GPGV variants were surveyed in an organically cultivated Hungarian vineyard that was planted between 1996 and 2014. Sequence analysis demonstrated the widespread presence and high variability of GPGV, and according to phylogenetic analyses, the Hungarian virus isolates were clas- sified into three groups. Most of the identified variants clustered with the representa- tive asymptomatic isolates, but all isolates from one grapevine cultivar grouped with representative isolates of clade B. The COVID-19 pandemic has spread worldwide for over two years. It was considered a significant threat to global health due to its transmissibility and high pathogenicity. The standard test for COVID-19, namely, reverse transcription polymerase chain reaction (RT–PCR), is somehow inaccurate and might have a high false-negative rate (FNR). As a result, an infected person with a negative test result may unknowingly continue to spread the virus, especially if they are infected with an undiscovered COVID-19 strain. Thus, a more accurate diagnostic technique is required. In this study, we propose 3Cs, which is a capsule neural network (CapsNet) used to classify computed tomography (CT) images as novel coronavirus pneumonia (NCP), common pneumonia (CP), or normal lungs. Using 6123 CT images of healthy patients' lungs and those of patients with CP and NCP, the 3Cs method achieved an accuracy of around 98% and an FNR of about 2%, demonstrating CapNet's ability to extract features from CT images that distinguish between healthy and infected lungs. This research confirmed that using CapsNet to detect COVID-19 from CT images results in a lower FNR compared to RT–PCR. Abaca is an indigenous fiber crop in the Philippines and is an important source of raw fiber worldwide. Banana bract mosaic virus (BBrMV) causes the abaca bract mosaic disease which results in decreased fiber yield and quality. Out of the 261 abaca samples tested, 175 tested positive via ELISA using raw antiserum against the recombinant BBrMV coat protein (CP). To further validate the antisera produced, a total of 48 samples were screened for BBrMV CP fragment via RT-PCR. Out of the 26 positives for RT-PCR, 22 were cloned and sequenced to validate the RT-PCR amplified fragments. Nucleotide analysis showed a high percentage identity (97–98%) towards BBrMV CP Pacific and Indian isolates while phylogenetic analysis revealed clustering of BBrMV CP Philippine abaca isolates with the Pacific group. The COVID-19 pandemic necessitated sensitive, fast, and inexpensive testing for the virus in 2020 prior to the widespread availability of vaccines. Early testing efforts were limited by bottlenecks on reagents, low-throughput testing options, and the slow return of test results. In this paper, we detail the testing pipeline we established at the University of Wisconsin-Madison for rapid, inexpensive, and sensitive surveillance testing for SARS-CoV-2, and we highlight the strengths of the platform that would allow it to be applied to other disease surveillance projects, SARS-CoV-2 variant testing, or future pandemics. Diverse clinical and laboratory features of multisystem inflammatory syndrome (MIS‐C) have been reported in the literature. Despite the worldwide distribution, systemic studies regarding the laboratory results do not exist. Therefore, we aimed to perform this systematic review and meta‐analysis to evaluate the serological, immunological, and cardiac parameters of the MIS‐C associated with SARS‐CoV‐2 infection. We searched the PubMed, Scopus, and Web of Science databases using specific keywords for any papers published in English since the disease onset and the first report until July 19, 2020. The inclusion criteria were children <21 years diagnosed with MIS‐C without any limitation on defining criteria. Forty‐eight studies were included in the final analysis, with a total population size of 3543 children with MIS‐C. The median age of the included patients was 8.3 (6.7–9) years. The pooled prevalence of male patients was 59% (95% CI: 56%–61%) and 62% (95% CI: 55%–69%) were admitted in ICU. The pooled prevalence of positive SARS‐CoV‐2 RT‐PCR, SARS‐CoV‐2 IgM, and SARS‐CoV‐2 IgG antibody tests was 33% (95% CI: 27%–40%), 39% (95% CI: 22%–58%) and 81% (95% CI: 76%–86%), respectively. The positivity rate of the inflammatory markers was as follows: CRP (96%, 95% CI: 90%–100%), d‐dimer (87%, 95% CI: 81%–93%), ESR (81%, 95% CI: 74%–87%), procalcitonin (88%, 95% CI: 76%–97%), ferritin (79%, 95% CI: 69%–87%), and fibrinogen (77%, 95% CI: 70%–84%). The pooled prevalence of elevated brain natriuretic peptide (BNP) level, pro‐BNP, and troponin were found in 60% (95% CI: 44%–75%), 87% (95% CI: 75%–96%), and 55% (95% CI: 45%–64%), respectively. The majority of patients had positive SARS‐CoV‐2 IgG test. Nearly one‐third of the cases showed negative RT‐PCR results. Cardiac and inflammatory markers were elevated in the majority of cases. It has been validated beyond doubt that High-Resolution Computed Tomography (HRCT) chest and to some extent chest radiographs have a role in corona virus disease-19 (COVID-19). Much less is known about the role of lung ultrasonography (LUS) in COVID-19. In this paper, our main purpose was to gauge the relationship between LUS and chest HRCT in reverse transcriptase polymerase chain reaction (RT–PCR) documented cases of COVID-19, as well as in those with high suspicion of COVID-19 with negative RT–PCR. It was a prospective study carried out at our tertiary care hospital, namely, SKIMS Soura. The total number of patients in this study were 152 (200 patients were selected out of which only 152 had undergone both LUS and chest HRCT). The patients were subjected to both LUS and chest HRCT. The radiologist who performed LUS was blinded to clinical findings and HRCT was evaluated by a radiologist with about a decade of experience. The LUS findings compatible with the disease were subpleural consolidations, B-lines and irregular pleural lines. Findings that were compatible with COVID-19 on chest HRCT were bibasilar, subpleural predominant ground glass opacities, crazy paving and consolidations. COVID-19-positive patients were taken up for chest HRCT for disease severity stratification and were also subjected to LUS. On HRCT chest, the imaging abnormalities compatible with COVID-19 were evident in 110 individuals (72.37%), and on Lung Ultrasound they were observed in 120 individuals (78.95%). Imaging of COVID-19 patients assessed by both LUS and HRCT chest,, showed a positive correlation (p < 0.0001). The study revealed a sensitivity of 88%, a specificity of 76.62%, a positive predictive value of 78.57% and a negative predictive value of 86.76%. None of the individuals with a diagnosis of COVID-19 on HRCT were missed on LUS. An excellent correlation was derived between the LUS score and CT total severity score (p < 0.0001 with a kappa of 0.431). HIV-HBV coinfected patients have higher rates of liver-related morbidity, hospitalizations, and mortality compared to HBV or HIV mono-infected ones. Clinical studies have shown an accelerated progression of liver fibrosis and an increased incidence of HCC, resulting from the combined action of HBV replication, immune-mediated hepatocytolysis, and HIV-induced immunosuppression and immunosenescence. Antiviral therapy based on dually active antiretrovirals is highly efficient, but late initiation, global disparities in accessibility, suboptimal regimens, and adherence issues may limit its impact on the development of end-stage liver disease. In this paper, we review the mechanisms of liver injuries in HIV-HBV coinfected patients and the novel biomarkers that can be used for treatment monitoring in HIV-HBV coinfected persons: markers that assess viral suppression, markers for liver fibrosis evaluation, and predictors of oncogenesis. Background: The SARS-CoV-2 pandemic has resulted in an unprecedented level of worldwide testing for epidemiologic and diagnostic purposes, and due to the extreme need for tests, the gold-standard Reverse Transcription Polymerase Chain Reaction (RT-PCR) testing capacity has been unable to meet the overall worldwide testing demand. Consequently, although the current literature has shown the sensitivity of rapid antigen tests (RATs) to be inferior to RT-PCR, RATs have been implemented on a large scale without solid data on performance. Objective: This study will compare analytical and clinical sensitivities and specificities of 50 lateral flow-- or laboratory-based RATs and 3 strand invasion--based amplification (SIBA)-RT-PCR tests from 30 manufacturers to RT-PCR testing of samples obtained from the deep oropharynx. In addition, the study will compare sensitivities and specificities of the included RATs as well as RT-PCR on clinical samples obtained from the deep oropharynx, the anterior nasal cavity, saliva, the deep nasopharynx, and expired air to RT-PCR on deep oropharyngeal samples. Methods: In the prospective part of the study, 200 individuals found SARS-CoV-2 positive and 200 individuals found SARS-CoV-2 negative by routine RT-PCR testing will be retested with each RAT, applying RT-PCR as the reference method. In the retrospective part of the study, 304 deep oropharyngeal cavity swabs divided into 4 groups based on RT-PCR quantification cycle (Cq) levels will be tested with each RAT. Results: The results will be reported in several papers with different aims. The first paper will report retrospective (analytical sensitivity, overall and stratified into different Cq range groups) and prospective (clinical sensitivity) data for RATs, with RT-PCR as the reference method. The second paper will report results for RAT based on anatomical sampling location. The third paper will compare different anatomical sampling locations by RT-PCR testing. The fourth paper will focus on RATs that rely on central laboratory testing. Tests from 4 different manufacturers will be compared for analytical performance data on retrospective deep oropharyngeal swab samples. The fifth paper will report the results of 4 RATs applied both as professional use and as self-tests. The last paper will report the results from 2 breath tests in the study. A comparison of sensitivity and specificity between RATs will be conducted using the McNemar test for paired samples and the chi-squared test for unpaired samples. Comparison of the positive predictive value (PPV) and negative predictive value (NPV) between RATs will be performed by the bootstrap test, and 95% CIs for sensitivity, specificity, PPV, and NPV will be calculated as bootstrap CIs. Conclusions: The study will compare the sensitivities of a large number of RATs for SARS-CoV-2 to with those of RT-PCR and will address whether lateral flow--based RATs differ significantly from laboratory-based RATs. Determining the safety of a newly developed experimental product is a crucial condition for its medical use, especially for clinical trials. In this regard, four hydrogel-type formulations were manufactured, all of which were based on carbomer (Blank-CP940) and encapsulated with caffeine (CAF-CP940), phosphorus derivatives (phenyl phosphinic (CAF-S1-CP940) and 2-carboxyethyl phenyl phosphinic acids (CAF-S2-CP940)). The main aim of this research was to provide a comprehensive outline of the biosafety profile of the above-mentioned hydrogels. The complex in vitro screening (cell viability, cytotoxicity, morphological changes in response to exposure, and changes in nuclei morphology) on two types of healthy skin cell lines (HaCaT—human keratinocytes and JB6 Cl 41-5a—murine epidermal cells) exhibited a good biosafety profile when both cell lines were treated for 24 h with 150 μg/mL of each hydrogel. A comprehensive analysis of the hydrogel's impact on the genetic profile of HaCaT cells sustains the in vitro experiments. The biosafety profile was completed with the in vivo and in ovo assays. The outcome revealed that the developed hydrogels exerted good biocompatibility after topical application on BALB/c nude mice's skin. It also revealed a lack of toxicity after exposure to the hen's chicken embryo. Even among vaccinated rabbits, the virus that causes rabbit viral hemorrhagic disease (RHDV) has been increasing in Egypt recently. So, the current study screened the emergent RHDV of vaccinated and unvaccinated domestic rabbits in some Egyptian provinces between 2022 and 2023. 40 pooled Samples were collected from different rabbit flocks with age range of 2 months up to 4 months with high mortality rates, clinical symptoms, and post-mortem lesions related to RHDV. Hemagglutination (HA) test, reverse transcriptase-polymerase chain reaction (RT-PCR) targeting the partial VP60 and sequencing and phylogenetic analysis were conducted for genotyping of RHDV strains. The results revealed that 16 out of 40 cases that were positive for RHDV and were hemagglutinin-positive, with titer ranging from 2³ to 214 Randomly six isolates were selected for genetic sequence and phylogenetic analysis which revealed their clustering with RHDV2 strains which were detected for the first time in Upper Egypt and submitted on GenBank as (OQ925947 Assiut .vac1 to OQ925952Assiut .vac6). The nucleotide sequence identities of the six isolates were 98.8-100 % compared to each other. The recently isolated strains had nucleotide difference 23, 1 % when compared to commonly vaccinal strain (JQ995154Giza 2006). The inoculated rabbits expressed RHDV-typical signs and postmortem findings and Ninety percent (90 %) mortality rate was recorded within3-6 days post-infection. Microscopic examination revealed that presence of acute necrotizing hepatitis, congestion and widespread hemorrhages in all internal organs; liver, lungs, spleen, kidneys and brain. A cross-sectional study has been undertaken in El-Minia province to survey an outbreak of foot-and-mouth disease (FMD) in cattle and identify the associated risk factors. From November 2021 to October 2022, researchers collected 100 blood samples, ten tongue epithelium specimens, and saliva swabs from animals of various ages and sexes in different locations. Additionally, the animals were examined for FMD signs. The overall seroprevalence was tested via SNT, which was 67%. Tissue samples and swabs were isolated and examined using conventional RT-PCR for detection of the FMD serotype, revealing serotype A. Statistically, no significant variation (P > 0.05) was observed in the prevalence of FMD in relation to age, sex, season, or locality. The findings of this study indicated that the recent FMD outbreak has been caused by serotype A in the studied locations and that FMD is widespread in cattle populations in El-Minia province. These findings underline the necessity for immediate attention and future research to detect FMDV strains spreading in the region that aid in executing effective prevention strategies. The study was designed to investigate the effect of curcumin, known for its antidiabetic properties, on the immunohistochemical localization and gene expression of glucagon-like peptide-1 receptor (GLP-1R) in the liver tissues of experimental diabetic rats using reverse transcription polymerase chain reaction (RT-PCR). For this, 24 Sprague–Dawley rats were divided into four groups—control, sham, diabetic, and diabetic + curcumin groups. The control group received no treatment, and 50 mg/kg streptozotocin was administered to the rats in the diabetic and diabetic + curcumin groups received 50 mg/kg streptozotocin. Once diabetes had been established, 100 mg/kg of curcumin was administered intraperitoneally to rats in the diabetic + curcumin group for a period of 21 days. Thesham group was administeredintraperitoneal ethanol and isotonic sodium chloride solution. At the ends of the experiment,tissues were subjected to histological and immunohistochemical examination to ascertain the localization of GLP-1R. Additionally, RT-PCR was employed to determine the levels of GLP-1R gene expression.The histological examinations revealed that the tissue samples from the control and sham groups exhibited a normal histological structure. In contrasr, the diabetic group displayed a range of degenerative changes, including enlargement of the sinusoidal wall enlargement and vacuolization of the hepatocytes. Furthermore, these degenerative findings were mitigated in the diabetic + curcumin group. In the immunohistochemical examinations, the majority of hepatocytes surrounding the vena centralis, as well as some endothelial, and some Kupffer cells,exhibited positively for GLP-1R. The diabetic group exhibited reduced immunoreactivity, while the diabetic + curcumin group demonstrated elevated immunoreactivity compared to the diabetes group. With regard to the molecular analysis, the mean expression level was observed to be higher in the diabetes + curcumin group. However, no significant difference in GLP-1R gene expression was identified between the groups. In conclusion, the administration of curcumin was observed to enhance GLP-1R expression in the liver of the rats with diabetes. Given that GLP-1R is a targets for diabetes treatment, curcumin can be used as a viable therapeutic agent for treating diabetes and alleviating its complications. [ABSTRACT FROM AUTHOR]

The bioactive compounds from Actinomycetes have antibacterial, anticancer, antioxidant, and antifungal properties. The antioxidant activity of ethyl acetate extract of Bacillus licheniformis was determined by carrying out several assays. Preparation of a bioactive feed encapsulated with sodium alginate and chitosan was carried out for treating the bacterial infection in Macrobrachium rosenbergii. An experimental trial with three different groups was conducted to determine the effect of crude extract of B. licheniformis against pathogenic bacteria, Aeromonas hydrophila in freshwater prawn, M. rosenbergii. The first group of prawns was given commercial diet. The second group of prawns challenged with A. hydrophila was given commercial feed. The third group of A. hydrophila challenged prawns was given the encapsulated B. licheniformis feed. The survival percentage of prawns was decreased in group 2 compared to the prawns of group 1 and group 3. The antioxidant properties were found to be significantly increased in case of group 3 prawns. Real time Reverse transcription - Polymerase chain reaction (RT-PCR) study of three different groups with three immune genes - Superoxide dismutase (SOD), Crustin and Prophenoloxidase was carried out and antioxidant enzymatic activity, and immunological parameters were recorded. Group 1 was designated as control, injected with saline. Group 2 was designated as infected, challenged with A. hydrophila and group 3 as treated, challenged with A. hydrophila and injected with crude extract of B. licheniformis. The results revealed that the SOD and Prophenoloxidase of gills and hemolymph showed highest gene expression in the case of treated group compared to the infected and control group. The data indicated that B. licheniformis showed enhanced antioxidant activities, immune response, disease resistance, and gene expression in M. rosenbergii challenged with A. hydrophila. As the SARS‐CoV‐2 virus spread throughout the world, millions of positive cases of COVID‐19 were registered and, even though there are millions of people already vaccinated against SARS‐CoV‐2, a large part of the global population remains vulnerable to contracting the virus. Massive nasopharyngeal sample collection in Puerto Rico at the beginning of the pandemic was limited by the scarcity of trained personnel and testing sites. To increase SARS‐CoV‐2 molecular testing availability, we evaluated the diagnostic accuracy of self‐collected nasal, saliva, and urine samples using the TaqPath reverse transcription polymerase chain reaction (RT‐PCR) COVID‐19 kit to detect SARS‐CoV‐2. We also created a colorimetric loop‐mediated isothermal amplification (LAMP) laboratory developed test (LDT) to detect SARS‐CoV‐2, as another strategy to increase the availability of molecular testing in community‐based laboratories. Automated RNA extraction was performed in the KingFisher Flex instrument, followed by PCR quantification of SARS‐CoV‐2 on the 7500 Fast Dx RT‐PCR using the TaqPath RT‐PCR COVID‐19 molecular test. Data was interpreted by the COVID‐19 Interpretive Software from Applied Biosystems and statistically analyzed with Cohen's kappa coefficient (k). Cohen's kappa coefficient (k) for paired nasal and saliva samples showed moderate agreement (0.52). Saliva samples exhibited a higher viral load. We also observed 90% concordance between LifeGene‐Biomarks' SARS‐CoV‐2 Rapid Colorimetric LAMP LDT and the TaqPath RT‐PCR COVID‐19 test. Our results suggest that self‐collected saliva is superior to nasal and urine samples for COVID‐19 testing. The results also suggest that the colorimetric LAMP LDT is a rapid alternative to RT‐PCR tests for the detection of SARS‐CoV‐2. This test can be easily implemented in clinics, hospitals, the workplace, and at home; optimizing the surveillance and collection process, which helps mitigate global public health and socioeconomic upheaval caused by airborne pandemics. In November 2021, an investigation was conducted into an outbreak of abortion, stillbirth, and the birth of calves with congenital abnormalities (arthrogryposis and hydranencephaly) at a dairy farm in Dasht-e-Mughan city, Ardabil province. A total of 70 cows experienced these issues. To determine the cause of the outbreak, post-mortem brain tissue samples were collected from two calves affected by hydranencephaly, which occurred shortly after their birth. Polymerase chain reaction (PCR) testing was conducted for multiple viruses, including bovine viral diarrhea (BVD), border disease, Akabane, Schmallenberg, and bluetongue viruses (BTVs). The samples were positive only for Akabane virus. Serum samples were collected from a group of 60 cattle, consisting of 45 adult cows and 15 younger calves aged between 8 to 10 months. These samples were analyzed to detect the presence of antibodies against the Akabane and Schmallenberg viruses. Both of these viruses are known to be responsible for causing abortion, stillbirth, and congenital abnormalities in calves. Among 45 cows that tested by competitive enzyme-linked immunosorbent assay (cELISA), 26.66% and 33.33% exhibited antibodies against Akabane and Schmallenberg viruses, respectively. Notably, 20.00% of cows co-exhibited antibodies for both viruses. Despite PCR evidence implicating Akabane virus as the principal etiology of clinical signs observed in the affected herd, the high coseropositivity to Schmallenberg virus, warrants a thorough investigation into potential viral interactions. Further research is required to determine the source of the virus and their transmission routes. The versatile potential of bacteriophage MS2-derived virus-like particles (VLPs) in medical biotechnology has been extensively studied during the last 30 years. Since the first reports showing that MS2 VLPs can be produced at high yield and relatively easily engineered, numerous applications have been proposed. Particular effort has been spent in developing MS2 VLPs as protective capsules and delivery platforms for diverse molecules, such as chemical compounds, proteins and nucleic acids. Among these, two are particularly noteworthy: as scaffolds displaying heterologous epitopes for vaccine development and as capsids for encapsulation of foreign RNA. In this review, we summarize the progress in developing MS2 VLPs for these two areas. Plain language summary Hollow, nanosized protein particles have many potential uses. If they can be appropriately engineered, they may for example be able to carry therapeutic cargoes to diseased cells or be used as a vaccine where appropriate antigens are mounted on their external surface. Many viruses offer a ready-made protein particle, the capsid, which can be made hollow by exclusion of the viral genetic material. MS2 is a virus that targets bacteria – a bacteriophage – which is well characterized and has been developed over many years for a number of applications. It has particular promise for development as a vaccine and for RNA delivery, both of which are reviewed here. Executive summary MS2 virus-like particles as scaffolds displaying antigenic peptides A number of virus-like particles (VLPs) are known to be effective immunogens and in some cases have been developed into clinically approved vaccines. MS2 is an icosahedral ssRNA bacteriophage around 26 nm in diameter. Immunogenic epitopes can be genetically engineered into the externally displayed AB loop of the capsid proteins. MS2 VLPs displaying such epitopes have been shown to be capable of eliciting an immune response in mice. Genetic fusion of identical coat protein monomers to form fused dimers stabilizes the resulting VLP but only one A/B loop of the resulting dimer can tolerate an insertion if stability is to be maintained. Proof-of-principle studies have been carried out for MS2 modified to act as vaccines against HPV, foot-and-mouth virus and Chlamydia trichomatis. The ability of MS2 VLPs to encapsidate their own encoding nucleic acid means that CPs with fused peptide can be used to 'biopan' a peptide library for binding to targets (e.g., antibodies) wherein the sequence encoding binding peptides can be easily recovered. MS2 VLPs as capsids for encapsulation of foreign RNA MS2 encapsidates its own RNA by binding to the 'pac site', a stem-loop structure. MS2 VLPs can encapsidate foreign RNA sequences by fusion to a pac site sequence. Fusion of target RNA to a copy of the pac site sequence with expression from the same transcript as the MS2 CP and the maturase avoids nonspecific RNA packing. The RNA is protected and stabilized and is known as armored RNA. Armored RNA can be used as internal control standards in real-time-PCR. Armored RNAs able to protect longer RNA sequences were developed through modification of expression vector strategy and pac site. Both in vivo and in vitro RNA packaging approaches have been developed. Foreign sRNAs and mRNA can be encapsidated within MS2 VLPs. Aim: To investigate the therapeutic potential of mebendazole (MBZ)-loaded nanostructured lipid carriers (NLCs). Methodology: NLC-MBZ was prepared and characterized to evaluate the in vitro and in vivo anticancer effects and the inhibitory effect on RanGTP and its potential as an antimetastatic treatment in vivo. Results: NLC-MBZ exhibited a size and charge of 155 ± 20 nm and -27 ± 0.5 mV, respectively, with 90.7% encapsulation. Free MBZ and NLC-MBZ had a 50% inhibitory concentration of 610 and 305 nM, respectively, against MDA-MB-231 cell lines. NLC-MBZ decreased tumor size, suppressed tumor lung metastases, and lowered the expression of CDC25A, SKP2, RbX1 and Cullin1 while boosting the Rb proteins. Conclusion: NLC-MBZ displayed antiangiogenic potential and resulted in a reduced rate of lung metastasis in vivo. The coordinated action of VEGF, IGF1/2 and H19 factors influences the development of endometriosis. The aim of this study was to analyze the expression level of these genes in patients with endometriosis. The study group consisted of 100 patients who were diagnosed with endometriosis on laparoscopic and pathological examination. The control group consisted of 100 patients who were found to be free of endometriosis during the surgical procedure and whose eutopic endometrium wasnormal on histopathological examination. These patients were operated on for uterine fibroids. Gene expression was determined by RT-PCR. The expression of the VEGF gene was significantly higher in the samples classified as clinical stage 1–2 compared to the control material (p < 0.05). There was also a statistically significant difference between the samples studied at clinical stages 1–2 and 3–4 (p < 0.01). The expression of the VEGF gene in the group classified as 1–2 was significantly higher. IGF1 gene expression was significantly lower both in the group of samples classified as clinical stages 1–2 and 3–4 compared to the control group (p < 0.05 in both cases). The expression of the H19 gene was significantly lower in the group of samples classified as clinical stage 3–4 compared to the control group (p < 0.01). Coronavirus disease 2019 (COVID-19) is a disease caused by the SARS-CoV-2 virus. Diagnosis and screening for COVID-19 are crucial in controlling the disease. RT-PCR is used for diagnosis. However, this test is high-risk, time-consuming, and expensive. There is a need for specific non-invasive biomarkers to detect COVID-19 rapidly. Volatile organic compounds (VOCs) produced by the human body can be used to depict metabolic conditions. A COVID-19 detection device is an electronic device designed to differentiate and detect odors. This study aims to assess the effectiveness of armpit sweat odor with a COVID-19 detection device to detect COVID-19. This study was an observational analytic study with a cross-sectional design conducted on COVID-19 and non-COVID-19 patients in the special isolation ward of Dr. Soetomo Hospital and the outpatient clinic of the Indonesian Navy Hospital during April 2021-December 2021. COVID-19 examination using armpit sweat odor with a COVID-19 detection device. The results of the COVID-19 detection device and RT-PCR were then compared using the McNemar test. The general characteristics of the 168 subjects (81 COVID-19 patients, 87 non-COVID-19 patients) showed that the most common gender was male with the most common comorbidities being hypertension and diabetes mellitus. The results of the diagnostic test showed sensitivity and specificity of 88.9% and 97.7%, respectively with accuracy of 93.45%. The McNemar test showed no significant difference with the RT-PCR results. The results of RT-PCR were not different from the results of armpit sweat odor using COVID-19 detection device. Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has become a "wicked evil" in this century due to its extended progression and huge human mortalities. Although the diagnosis of SARS-CoV-2 viral infection is made simple and practical by employing reverse transcription polymerase chain reaction (RT-PCR) investigation, the process is costly, complex, time-consuming, and requires experts for testing and the constraints of a laboratory. Therefore, these challenges have raised the paradigm of on-site portable biosensors on a single chip, which reduces human resources and enables remote access to minimize the overwhelming burden on the existing global healthcare sector. This article reviews the recent advancements in biosensors for long coronavirus disease (COVID) management using a multitude of devices, such as point-of-care biosensors and lab-on-chip biosensors. Furthermore, it details the shift in the paradigm of SARS-CoV-2-on-chip biosensors from the laboratory to on-site detection with intelligent and economical operation, representing near-future diagnostic technologies for public health emergency management. The Corona Virus was first started in the Wuhan city, China in December of 2019. It belongs to the Coronaviridae family, which can infect both animals and humans. The diagnosis of coronavirus disease-2019 (COVID-19) is typically detected by Serology, Genetic Real-Time reverse transcription–Polymerase Chain Reaction (RT-PCR), and Antigen testing. These testing methods have limitations like limited sensitivity, high cost, and long turn-around time. It is necessary to develop an automatic detection system for COVID-19 prediction. Chest X-ray is a lower-cost process in comparison to chest Computed tomography (CT). Deep learning is the best fruitful technique of machine learning, which provides useful investigation for learning and screening a large amount of chest X-ray images with COVID-19 and normal. There are many deep learning methods for prediction, but these methods have a few limitations like overfitting, misclassification, and false predictions for poor-quality chest X-rays. In order to overcome these limitations, the novel hybrid model called "Inception V3 with VGG16 (Visual Geometry Group)" is proposed for the prediction of COVID-19 using chest X-rays. It is a combination of two deep learning models, Inception V3 and VGG16 (IV3-VGG). To build the hybrid model, collected 243 images from the COVID-19 Radiography Database. Out of 243 X-rays, 121 are COVID-19 positive and 122 are normal images. The hybrid model is divided into two modules namely pre-processing and the IV3-VGG. In the dataset, some of the images with different sizes and different color intensities are identified and pre-processed. The second module i.e., IV3-VGG consists of four blocks. The first block is considered for VGG-16 and blocks 2 and 3 are considered for Inception V3 networks and final block 4 consists of four layers namely Avg pooling, dropout, fully connected, and Softmax layers. The experimental results show that the IV3-VGG model achieves the highest accuracy of 98% compared to the existing five prominent deep learning models such as Inception V3, VGG16, ResNet50, DenseNet121, and MobileNet. Aim: Osteoporosis is a systemic skeletal disorder characterized by reduced osteoblast differentiation, predominantly by overexpression of the Sost gene. A layer-by-layer approach enabled encapsulation of Sost siRNA to enhance the short half-life and poor transfection capacity of siRNA. Materials & methods: Polyethyleneimine and siRNA on chitosan-coated gold nanoparticles (PEI/siRNA/Cs–AuNPs) were engineered using chitosan-reduced gold nanoparticles. They were characterized by dynamic light scattering, scanning electron microscopy, transmission electron microscopy, Fourier transform infrared and gel-mobility assays. Detailed in vitro experiments, gene silencing and western blots were performed. Results: A total of 80% knockdown of the target sclerostin protein was observed by PEI/siRNA/Cs–AuNPs, q-PCR showed threefold downregulation of the Sost gene. Osteogenic markers RunX2 and Alp were significantly upregulated. Conclusion: We report a safe, biocompatible nanotherapeutic strategy to enhance siRNA protection and subsequent silencing to augment bone formation. [ABSTRACT FROM AUTHOR]

The hazel allergen Cor a 1 is a PR-10 protein, closely related to the major birch pollen allergen Bet v 1. Hazel allergies are caused by cross-reactive IgE antibodies originally directed against Bet v 1. Despite the importance of PR-10 proteins in allergy development, their function and localization in the plant remain largely elusive. Therefore, the presence of Cor a 1 mRNA and proteins was investigated in different tissues, i.e., the female flower, immature and mature nuts, catkins, and pollen. Four yet unknown Cor a 1 isoallergens, i.e., Cor a 1.0501–1.0801, and one new Cor a 1.03 variant were discovered and characterized. Depending on the isoallergen, the occurrence and level of mRNA expression varied in different tissues, suggesting different functions. Interestingly, Cor a 1.04 previously thought to be only present in nuts, was also detected in catkins and pollen. The corresponding Cor a 1 genes were expressed in Escherichia coli. The purified proteins were analysed by CD and NMR spectroscopy. Immunoblots and ELISAs to determine their allergenic potential showed that the new proteins reacted positively with sera from patients allergic to birch, hazel and elder pollen and were recognized as novel isoallergens/variants by the WHO/IUIS Allergen Nomenclature Sub-Committee.
