Objectives: This study aimed to evaluate the relationship between mobile phone usage and miRNA-574-5p and miRNA-30C-5p levels in patients diagnosed with differentiated thyroid cancer (DTC). Methods: Fifty patients diagnosed with DTC and 50 healthy volunteers were included in the study. miRNA-574-5p and miRNA-30C-5p gene expression levels in the blood of all subjects were analyzed by real time-polymerase chain reaction, and a questionnaire including various questions was administered to both groups. Results: Although there was a 7.60-fold increase in miRNA-30C-5p gene expression levels in the patient group compared with the control group, it was not found to be statistically significant. Considering the miRNA-574-5p gene expression levels, although there was a 2.96-fold increase in the patient group compared with the control group, no significant relationship was found. In our study, 85% of our patients were using mobile phones with internet access, whereas 98% of our healthy volunteers were using mobile phones (p<0.05). While 53.5% of the patients had their mobile phones with them while they were sleeping, this rate was 83.7% in healthy volunteers (p<0.05). However, 93.9% of the healthy volunteers did not have a Wi-Fi device in their bedrooms, and this rate was 75% in the patient group (p<0.05). Conclusion: Although miRNA-30C-5p and miRNA-574-5p gene expression levels were higher in patients than in healthy volunteers, the differences were not statistically significant. Although there was no significant difference in miRNA levels, we believe that due to the higher rate of Wi-Fi device presence in bedrooms in patients compared with healthy volunteers, the effects of electromagnetic radiation on the thyroid can be reduced by paying attention to this simple change. Objective: We aimed to analyzed the association between gastric cancer and polymorphism of XPO5 gene. Method: The polymorphism in the XPO5 gene (rs11544382) was determined in 120 individuals (60 gastric cancer patients; 60 healthy controls) using Real-Time PCR method. Results: The comparison of gastric cancer patients and controls revealed a statistically significant relationship for alcoholic drink consumption (p<0.05). The relationship between XPO5 gene (rs11544382) polymorphism and gastric cancer was statistically not significant. There was no a statistically significant relationship between mutant (GG) genotype with both wild type (AA) and heterozygous (AG) polymorphic genotypes when evaluated in XPO5 polymorphism gastric cancer patients and control groups (X2:0.12, p=0.729). The heterozygous (AG) was dominant in gastric cancers patients and control subjects, 93.3 and 91.7% respectively. Conclusions: This study provides information about allele and genotype frequency distribution of XPO5 gene polymorphism (rs11544382) in Turkish At the same time, AG genotype was found to be dominant in gastric cancer patients and their controls. Objective: Symptomatic anastomotic leakage observed after colorectal surgery is one of the major complications. One of the factors affecting the anastomosis healing is the type of suture material used. This study aims to investigate the effects of the suture material polydioxanone (PDS, Ethicon), which is late absorbable and has a monofilament structure, and the suture material polyglactin 910 (Vicryl, Ethicon), which is absorbable and has a multifilament structure, on the healing of colonic anastomosis, and to compare the traditional forms of these sutures with their antibacterial effective triclosan-impregnated forms (PDS Plus, Ethicon and Vicryl Plus, Ethicon). Materials and Methods: The rats were divided into four equal groups consisting of 10 subjects each: Group I: Vicryl; Group II: Vicryl Plus; Group III: PDS; and Group IV: PDS Plus. The presence of wound infection, whether the integrity of the abdominal wall was maintained, intra-abdominal adhesion scoring, the presence of intra-abdominal abscess, and whether the macroscopic integrity of anastomosis was maintained were evaluated. Results: According to the results of this experimental study, while the highest ABP and hydroxyproline levels were observed in the PDS Plus group, the lowest values were observed in the Vicryl group. However, the difference was not statistically significant. Conclusion: Slowly absorbable and monofilament PDS suture material causes less tissue reaction and inflammatory response compared to the Vicryl suture material that is absorbable in the colonic anastomosis line and multifilament. Objective: Gastric cancer is a multifunctional disease. Emotional stress, physiological and neuroendocrine changes in cancer patients can lead to the activation of the hypothalamic-pituitary-adrenal axis and the release of hormone dependent hormones such as catecholamine. In particular, it has been reported that catecholamine induction directly affects the biological behavior of tumor cells via β2-Adrenergic receptor (β2-AR) mediated signaling. In this study, it was aimed to investigate polymorphism of β2-AR gene (rs1042717) in gastric cancer patients. Method: Polymorphism in the β2-AR gene (rs1042717) was determined by Real-Time PCR method in 60 gastric cancer patients and 50 healthy controls. The results were evaluated using logistic regression and Chi-square (χ2) test. Results: The comparison of gastric cancer patients and controls determined a statistically significant relationship for alcoholic drink consumption (p<0,05). There was a statistically significant relationship between β2-AR (rs1042717) polymorphism and stomach cancer (p <0.05). There was a statistically significant relationship between mutant (AA) genotype with wild type (GG) and heterozygous (AG) polymorphic genotypes when evaluated in β2-AR polymorphism gastric cancer patients and control group (χ2: 19,38, p: 0.001). Similarly, there was a statistically significant correlation between heterozygous (AG) with wild type (GG) and mutant (AA) polymorphic genotypes in gastric cancer (χ2: 14, 27, p: 0,001). Conclusions: In this study, it was found that the AG genotype is predominant in gastric cancer patients and controls, and that the AA genotype has a protective effect against gastric cancer. The increasing prevalence of antibiotic-resistant bacterial infections has led to the search for new approaches. This study aimed to evaluate the effects of carvacrol and N-acetyl cysteine, both individually and in combination, on the planktonic cells and biofilm formations of Staphylococcus aureus , including methicillin-resistant and methicillin-sensitive strains. Additionally, the study sought to perform cytotoxicity tests and chemical characterization to further understand the properties and potential applications of these substances. A total of 19 S. aureus strains were included in the study. Minimum inhibitory concentration and minimum bactericidal concentration were determined by assays. Synergy analysis tests were carried out. Cytotoxicity tests were conducted on the fibroblast cell line. Characterization test was performed. While Minimum inhibitory concentration and minimum bactericidal concentration values for carvacrol varied between 250 and 500 μg/ml, these values were in the range of 32–64 mg/ml for N-acetyl cysteine. Biofilm formation activities were identified. A total of eight strains, including six clinical and two standard strains with the highest biofilm-forming ability, were selected for combination studies. The combination of Carvacrol and N-acetyl cysteine exhibited synergistic and partially synergistic effects on the tested planktonic and biofilm strains, and these effects were dose-dependent. Carvacrol was found to be the most active drug at the end of 24, 48, and 72 h. Regarding the synergistic effect of N-acetyl cysteine + carvacrol, it was revealed to exhibit higher activity than N-acetyl cysteine and lower activity than carvacrol. The combination of carvacrol and N-acetyl cysteine demonstrated synergistic and partially synergistic effects against both planktonic and biofilm forms of Staphylococcus aureus. These results suggest potential for novel approaches in managing orthopedic infections, warranting further research to explore their therapeutic applications. • The CAR and NAC combination inhibits MRSA biofilm formation. • The NAC + CAR combination balances efficacy and safety for infections. • The study provides insights into the antimicrobial mechanisms of NAC and CAR through detailed chemical analysis. Objectives: This study aims to compare the serum calcitonin gene-related peptide (CGRP) and CGRP receptor protein levels between patients with fibromyalgia syndrome (FM) and healthy control subjects. Patients and methods: The study included 88 patients (7 males, 81 females; mean age 44.5±9.1 years; range, 20 to 72 years) newly-diagnosed with FM according to the 2010 American College of Rheumatology criteria and 88 healthy volunteers (6 males, 82 females; mean age 43.0±6.1 years; range, 20 to 57 years). Venous blood samples were collected from both groups for the measurement of the levels of serum CGRP and CGRP receptor proteins (receptor component protein [RCP], receptor activity modifying protein 1 [RAMP 1] and calcitonin receptor-like receptor [CLR]). Results: A comparison of the serum CGRP, CLR and RCP levels of the FM and control groups revealed a statistically significant difference (p=0.001, p=0.005, p=0.001, respectively). The difference between the groups in respect of the serum RAMP 1 levels was not statistically significant (p=0.107). Conclusion: The serum CGRP, CLR and RCP levels were found to be higher in the FM patients, but no difference was determined between the FM patients and the healthy control group in respect of the RAMP 1 level. Superoxide dismutases (SOD) are a class of enzymes that catalyze the dismutation of superoxide into oxygen and hydrogen peroxide [1]. They play an important role in antioxidant mechanism in nearly all cells exposed to oxygen. Three isoforms of SOD are expressed in humans, encoded by different genes. SOD1 (CuZn-SOD) is mainly located in the cytoplasm, SOD2 in the mitochondria and SOD3 is extracellular. SOD1 accounts for ~ 85% of the total cellular SOD activity of most mammalian cells [2]. Genetic polymorphisms in SOD have been implicated in proneness to cancer and other diseases. These potentially significant genetic variants related to oxidative stress have already been studied extensively, including single nucleotide polymorphisms (SNP) -251A/G (rs2070424) of the SOD1 gene[3,4]. In the present study, the distribution of SOD1 gene -251A/G polymorphisms in a Turkish population including 494 (females: 278, 56,3% and males: 216, 43,7%) unrelated healthy individuals was examined using a PCRRFLP method. All genotypes and alleles were in Hardy-Weinberg equilibrium (X2=0,107, p>0.05). The mean age of the study population was 38,4±16,6 years (males, 39,8±17,1; females, 37,3±16,1). The frequency distributions of A and G alleles were found to be respectively 0,93 and 0,07. The observed genotype frequencies of SOD1 were 86.2, 13.4 and 0.4 % for AA, AG and GG, respectively. Süperoksit dismutazlar (SOD), süperoksidin oksijen ve hidrojen perokside dismutasyonunu katalizleyen bir enzin sınıfıdır [1]. Bu enzimler oksijene maruz kalan hemen hemen tüm hücrelerde antioksidan mekanizmada önemli rol oynarlar. İnsanlarda farklı genlerce kodlanan üç SOD izoformu eksprese edilmektedir. SOD1 (CuZn-SOD) ağırlıkla sitoplazmada bulunurken, SOD2 mitokondride ve SOD3 ise ekstraselüler de lokalizedir. SOD1, birçok memeli hücresinde toplam hücresel SOD aktivitesinin ~ 85%' ini oluşturmaktadır [2]. SOD genetic polimorfizmi, kanser ve diğer hastalıklara yatkınlıkla ilişkilendirilmiştir. Tek nükleotid polimorfizmi içeren (SNP) -251A/G (rs2070424) SOD1 gen, oksidatif stresle ilgili bu potansiyel olarak önemli genetic variantlar kapsamlı olarak çalışılmıştır [3,4]. Objective: Lung cancer is one of the most common cancers in the world. It is known that angiogenesis plays a role in the development and metastasis of lung cancer. Azomethine derivatives known as Schiff bases have many biological activities. In this study, we aimed to determine the anticancer activity of the newly synthesized azomethine derivative compound B-47/2 on lung cancer and to determine the effect of this component on vascular endothelial growth factor B (VEGFB) gene expression. Material and Method: Compound B-47/2 was synthesized for the first time. B-47/2 compound was applied to lung cancer cell line (A549) at varying concentrations (1-100 µg/mL) and its anticancer activity was found after 24, 48 and 72 hours incubations using the 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide (MTT) method. The half maximal inhibitory concentration (IC50) dose of B-47/2 was applied to the cells and ribonucleic acid (RNA) isolation followed by complementary deoxyribonucleic acid (cDNA) synthesis was performed. Then, reverse transcription-polymerase chain reaction (RT-PCR) method was used to determine the expression level of VEGFB gene. Results: As a result, it was determined that the B-47/2 compound applied to the A-549 cell line showed the highest cytotoxic activity after 72 hours of incubation. In addition, it was determined that the B-47/2 compound decreased the expression of the VEGFB gene. Discussion: There are studies in which the anticancer activity of azomethine derivatives has been observed. The topic of synthesizing new drugs to prevent cancer is popular. Objective: Cancer arises as a result of the failure of the mechanisms controlling normal division in a group of cells. It is known that some new synthesis compounds intended for use in cancer treatment have anti-fungal, anti-bacterial, anti-carcinogenic effects. In this study, it was aimed to apply the newly synthesized B-108 compound to the A-549 cell line and then to investigate the effect of this compound on the ERCC1 gene expression profile. Materials and Methods: Firstly, compound B-108 was synthesized in our study. Afterwards, this synthesized molecule was applied in eight different concentrations (1-100 μg/ml) in A-549 cell line and 3-(4,5- dimethylthiazol-2-yl)-2,5-yl for 24 hours, 48 hours and 72 hours. Anticancer activities were determined using diphenyltetrazolium bromide (MTT) method. Expression level of DNA repair gene (ERCC1) was determined using RT-PCR method. Results: As a result, it was determined that the molecule applied to the A-549 cell line showed the highest activity after 72 hours of incubation. It was observed that the ERCC1 gene expression of the molecule applied on lung cancer was lower than the control group. Background: Lumbar plexus blockade (LPB) combined with sciatic nerve block (SNB) is frequently used for lower extremity surgery. Perioperative nerve injury is a rarely encountered complication of peripheral nerve blocks (PNB). Case Report: Here we report a 44-year-old male patient who developed a partial femoral nerve injury (FNI) following a LPB which was performed before the surgery of a patellar fracture. The clinical and electroneuromyographic findings of the patient were recovered almost completely within the following six months. Conclusion: The presented case demonstrated a FNI despite the absence of any pain or paresthesia sensation, with the disappearance of motor response under 0.3 mA of neurostimulation in the experienced hands. Background: The objective of our study was to measure and compare the elution characteristics of teicoplanin from poly(methyl methacrylate) PMMA beads with those of poly(glycolide-co-lactide) PGLA-added beads. Methods: The study included two groups of PMMA + teicoplanin beads. PMMA was added to teicoplanin in Group 1 and PMMA + PGLA was added to teicoplanin in Group 2. A total of 16 beads of 1 cm3 were created for each group. Samples were added individually to tubes containing 3 ml of phosphate-buffered saline (PBS). Antibiotic elution was measured by measuring absorbance values of 1-ml samples taken at regular intervals using a UV–Vis spectrophotometer and cumulative percentages of drug release were calculated. In addition, the spectra of teicoplanin were identified using a FTIR spectrophotometer in a wavelength range of 400–4000 cm−1. Results: Drug elution in the PBS medium was measured and compared for Groups 1 and 2. The cumulative percentage of drug release from the PGLA-added beads (Group 2) was significantly higher (p = 0.01). The molecular structure of teicoplanin was also confirmed using FTIR. Conclusion: The in vitro results showed that the addition of biodegradable PGLA into bone cement functions as a water-soluble porogen which allows for significant increases in the elution of teicoplanin from cement. This increase in elution suggests that the PGLA would allow for further fluid contact and exchange with the previously entrapped drug. These results may have important clinical applications. Objectives: This study aims to compare the beta-2 adrenergic receptor (ADRB2) gene polymorphisms of patients with fibromyalgia syndrome (FMS) with those of healthy control subjects, and to investigate the possible relationship between symptoms of FMS and polymorphisms of the ADRB2 gene. Patients and methods: The study included 170 females (mean age 47.8±10.3 years; range, 21 to 75 years) diagnosed with FMS according to the 2010 American College of Rheumatology criteria and 170 healthy females (mean age 47.2±8.8 years; range, 20 to 72 years) as the control group. Several clinical symptoms of the participants related to FMS were questioned and recorded. The visual analog scale (VAS) and Fibromyalgia Impact Questionnaire (FIQ) scores of the fibromyalgia group were recorded. In both groups, the ADRB2 (rs1042717) single-nucleotide polymorphism was detected by way of a real-time polymerase chain reaction. The wild-type (Guanine/Guanine), the mutant type (Adenine/Adenine) and heterozygous type (Adenine/Guanine) were detected. The sample power was calculated considering the minor allele frequency. Results: The comparison of the ADRB2 gene polymorphism between patients with FMS and the control subjects showed that the groups were similar in terms of ADBR2 gene polymorphism and genotype (p>0.05). There was no significant difference in terms of genotype when the ADRB2 gene polymorphisms in patients with FMS were compared in terms of clinical symptoms, VAS and FIQ scores (p>0.05). Conclusion: Beta-2 adrenergic receptor (rs1042717) gene polymorphisms and genotype distribution are no different between patients with FMS and healthy individuals. ADRB2 gene polymorphisms in patients with FMS have no effect on clinical symptoms and VAS and FIQ scores.
