OBJECTIVES: The American Academy of Pediatrics recommends that children and adolescents with Down syndrome receive anticipatory guidance regarding development and behavior. However, few tools provide specific guidance on developmental norms for children with Down syndrome. Our objective was to estimate age ranges at which children and adolescents with Down syndrome achieve developmental milestones to facilitate developmental screening by pediatric practitioners. METHODS: We used standardized questionnaires to obtain information from clinicians and caregivers of children with Down syndrome who received care at the Boston Children's Hospital Down Syndrome Program between March 2018 and March 2023. Data included information from 2599 visits for 842 individuals with Down syndrome ages 2 months to 24 years. We used mixed-effects logistic regressions to predict the probability of achieving 25 specific developmental milestones with 15%, 30%, 45%, 60%, 75%, and 90% probability as a function of age. We further stratified results by individuals' sex. RESULTS: We present age norms for our study's population of people with Down syndrome for key milestones in academic, adaptive, language, and motor domains by calculating the ages at which milestone achievement was 75% probable. We then compare these norms to published norms for the general population. During the early 2000s, the U.S. government began to partner with local law enforcement agencies for assistance with the enforcement of immigration laws. Participation in the voluntary 287(g) program by law enforcement agencies was an important early iteration of this new strategy. Although relatively few agencies adopted 287(g), its implementation was a critical turning point in the nation's approach to immigration enforcement and has been linked to a wide range of negative consequences for Latinos, including higher levels of violent victimization, and decreases in police trust. This study advances knowledge about the factors associated with 287(g) program adoption during the late 2000s. We examined the role of agency ethnoracial diversity and fiscal capacity and several theoretically relevant indicators of the broader county context. Based on a sample of 880 law enforcement agencies located in 233 counties, rare event logistic regression models indicate that the strongest predictors of 287(g) adoption were the share of county population estimated to be undocumented immigrants, the degree of support for the Republican Presidential candidate, and the relative unemployment of Hispanic and non-Hispanic White residents. The reaction products formed in the SO2 – ROH – (HOCH2)3CNH2 (where R – CH3, C2H5, n-C3H7, i-C3H7, n-C4H9 and n-C5H11) systems were isolated and identified as: binary salt – ammonium O-methylsulfite [(HOCH2)3CNH3]+ [CH3OSO2]− (1); 1/1 charge transfer complex between diethyl sulfite (2) and tris(hydroxymethyl)aminomethane by S-N binding; polymorhps of zwitterionic internal salt – O-sulfite tris(hydroxymethyl)methylammonium (at R – n-C4H9 and n-C5H11); and its solvates with n-propanol 1/1 (3) and isopropanol 2/1. Sunflower syndrome is a rare photosensitive pediatric epilepsy characterized by stereotyped hand-waving in response to bright lights. These stereotyped movements with maintained awareness can be mistaken for a movement disorder. This study assessed neurology providers' diagnostic reasoning, evaluation, and treatment of Sunflower syndrome. A 32-question anonymized electronic survey, including a clinical vignette and video of hand-waving in sunlight, was distributed to child neurology providers to assess (1) initial diagnosis and evaluation based on clinical information, (2) updated diagnosis and management after electroencephalography (EEG), and (3) prior experience with Sunflower syndrome. Among 277 viewed surveys, 211 respondents provided information about initial diagnosis and evaluation, 200 about updated diagnosis, 191 about management, and 189 about prior clinical experience. Most providers (135, 64%) suspected seizure, whereas fewer suspected movement disorders (29, 14%) or were unsure of the diagnosis (37, 22%). EEG was recommended by 180 (85%). After EEG, 189 (95%) diagnosed epilepsy, 111 of whom specifically diagnosed Sunflower syndrome. The majority (149, 78%) recommended antiseizure medications (ASMs) and sun avoidance (181, 95%). Only 103 (55%) had managed Sunflower syndrome. Epileptologists and those with prior clinical experience were more likely to suspect a seizure, order an EEG, and offer ASMs than those without prior experience. Although many providers had not managed Sunflower syndrome, the majority recognized this presentation as concerning for epilepsy. Epilepsy training and prior clinical experience are associated with improved recognition and appropriate treatment. Down syndrome (DS) is a complex condition associated with multiple medical, developmental, and behavioral concerns. A prospective, longitudinal clinical database was integrated into a specialty Down Syndrome Program, with the goals of better understanding the incidence, course, and impact of co-occurring medical, neurodevelopmental, and mental health conditions in DS. We describe the process of developing the database, including a systematic approach to data collection and database infrastructure, and report on feasibility, challenges, and solutions of initial implementation. Between March 2018 and November 2021, data from 842 patients (ages 4.8 months to 26 years) was collected. Challenges included caregiver form completion as well as time and personnel required for successful implementation. With full integration into clinical visit flow, the database proved to be feasible. The database enables identification of patterns of development and health throughout the lifespan and it facilitates future data sharing and collaborative research to advance care. Cardiovascular homeostasis is maintained, in part, by neural signals arising from arterial baroreceptors that apprise the brain of blood volume and pressure. Here, we test whether neurons within the nodose ganglia that express angiotensin type-1a receptors (referred to as NGAT1aR) serve as baroreceptors that differentially influence blood pressure (BP) in male and female mice. Using Agtr1a-Cre mice and Cre-dependent AAVs to direct tdTomato to NGAT1aR, neuroanatomical studies revealed that NGAT1aR receive input from the aortic arch, project to the caudal nucleus of the solitary tract (NTS), and synthesize mechanosensitive ion channels, Piezo1/2. To evaluate the functionality of NGAT1aR, we directed the fluorescent calcium indicator (GCaMP6s) or the light-sensitive channelrhodopsin-2 (ChR2) to Agtr1a-containing neurons. Two-photon intravital imaging in Agtr1a-GCaMP6s mice revealed that NGAT1aR couple their firing to elevated BP, induced by phenylephrine (i.v.). Furthermore, optical excitation of NGAT1aR at their soma or axon terminals within the caudal NTS of Agtr1a-ChR2 mice elicited robust frequency-dependent decreases in BP and heart rate, indicating that NGAT1aR are sufficient to elicit appropriate compensatory responses to vascular mechanosensation. Optical excitation also elicited hypotensive and bradycardic responses in ChR2-expressing mice that were subjected to deoxycorticosterone acetate (DOCA)-salt hypertension; however, the duration of these effects was altered, suggestive of hypertension-induced impairment of the baroreflex. Similarly, increased GCaMP6s fluorescence observed after administration of phenylephrine was delayed in mice subjected to DOCA-salt or chronic delivery of angiotensin II. Reproductive biomechanics, a broad and developing field, contains fluid mechanics problems at every stage. In particular, the human female reproductive system is a complex and dynamic fluid-structure system. Until recently, the majority of this research focused on the early moments of reproduction—namely, the transport phenomena that dominate fertilization and implantation. However, in the past two decades, fluid mechanics has begun to be used in investigations of all aspects surrounding pregnancy, labor, and delivery. In this review, we discuss human reproduction starting with the nonpregnant uterus through implantation, delivery, and lactation and breastfeeding. We cover physiologic changes from the transformation of the uterus and the cervical remodeling process to the development of the placenta and the beginning of lactation. Sports analytics—broadly defined as the pursuit of improvement in athletic performance through the analysis of data—has expanded its footprint both in the professional sports industry and in academia over the past 30 years. In this article, we connect four big ideas that are common across multiple sports: the expected value of a game state, win probability, measures of team strength, and the use of sports betting market data. For each, we explore both the shared similarities and individual idiosyncracies of analytical approaches in each sport. While our focus is on the concepts underlying each type of analysis, any implementation necessarily involves statistical methodologies, computational tools, and data sources. Where appropriate, we outline how data, models, tools, and knowledge of the sport combine to generate actionable insights. We also describe opportunities to share analytical work, but omit an in‐depth discussion of individual player evaluation as beyond our scope. This article should serve as a useful overview for anyone becoming interested in the study of sports analytics. Several circular economy indicators have been proposed at the company level, but their applicability in industrial practice is limited. It is therefore necessary to assess the usefulness of existing metrics and refine their use through practical applications across different types of companies and industries. This study assesses the usability and usefulness of the Overall Circularity Effectiveness Index. The index was applied in three different companies, such as rubber, steel, and plastic companies. Then, the usability and usefulness of the index were analyzed through a qualitative assessment considering nine criteria based on the literature. The results showed the index demonstrated to be adaptable to different business, the data collection and index calculation were simple and not too time-consuming. However, the index's usefulness can be improved concerning gaps identification and user experience. Research Summary: Our understanding of how immigration enforcement impacts crime has been informed exclusively by data from police crime statistics. This study complements existing research by using longitudinal multilevel data from the National Crime Victimization Survey for 2005–2014 to simultaneously assess the impact of the three predominant immigration policies that have been implemented in local communities. The results indicate that the activation of Secure Communities and 287(g) task force agreements significantly increased violent victimization risk among Latinos, whereas they showed no evident impact on victimization risk among non‐Latino Whites and Blacks. The activation of 287(g) jail enforcement agreements and anti‐detainer policies had no significant impact on violent victimization risk during the period. Policy Implications: Contrary to their stated purpose of enhancing public safety, our results show that the Secure Communities program and 287(g) task force agreements did not reduce crime, but instead eroded security in U.S. communities by increasing the likelihood that Latinos experienced violent victimization. These results support the Federal government's ending of 287(g) task force agreements and its more recent move to end the Secure Communities program. Today the challenge in paternity testing is to provide an accurate noninvasive assay that can be performed early during pregnancy. This requires the use of novel analytical methods capable of detecting the low fraction of circulating fetal DNA in maternal blood. We previously showed that forensic compound markers such as deletion/insertion polymorphisms-short tandem repeats (DIP-STR) can efficiently resolve complex mixed biological evidence including the target analysis of paternally inherited fetal alleles. In this study, we describe for the first time the validation of this type of markers in the first trimester of pregnancies, in addition to defining the statistical framework to evaluate paternity. To do so, we studied 47 DIP-STRs in 87 cases, with blood samples collected throughout gestation starting from the seven weeks of amenorrhea. Fetal DNA detection in the first trimester shows a false negative rate as low as 6%. The combined paternity index (CPI) results indicate that seven markers with fully informative genotypes are sufficient to determine the paternity. This study demonstrates that DIP-STR markers can be used from early pregnancy and that a small set of markers (about 40) is sufficient to address the question of paternity. To assess differences in willingness to vaccinate children against COVID-19, and factors that may be associated with increased acceptance, among US caregivers of various racial and ethnic identities who presented with their child to the Emergency Department (ED) after emergency use authorization of vaccines for children ages 5–11. A multicenter, cross-sectional survey of caregivers presenting to 11 pediatric EDs in the United States in November-December 2021. Caregivers were asked about their identified race and ethnicity and if they planned to vaccinate their child. We collected demographic data and inquired about caregiver concerns related to COVID-19. We compared responses by race/ethnicity. Multivariable logistic regression models served to determine factors that were independently associated with increased vaccine acceptance overall and among racial/ethnic groups. Among 1916 caregivers responding, 54.67% planned to vaccinate their child against COVID-19. Large differences in acceptance were noted by race/ethnicity, with highest acceptance among Asian caregivers (61.1%) and those who did not specify a listed racial identity (61.1%); caregivers identifying as Black (44.7%) or Multi-racial (44.4%) had lower acceptance rates. Factors associated with intent to vaccinate differed by racial/ethnic group, and included caregiver COVID-19 vaccine receipt (all groups), caregiver concerns about COVID-19 (White caregivers), and having a trusted primary provider (Black caregivers). Caregiver intent to vaccinate children against COVID-19 varied by race/ethnicity, but race/ethnicity did not independently account for these differences. While it is well known from numerous epidemiologic investigations that social determinants (socioeconomic, environmental, and psychosocial factors exposed to over the life-course) can dramatically impact cardiovascular health, the molecular mechanisms by which social determinants lead to poor cardiometabolic outcomes are not well understood. This review comprehensively summarizes a variety of current topics surrounding the biological effects of adverse social determinants (i.e., the biology of adversity), linking translational and laboratory studies with epidemiologic findings. With a strong focus on the biological effects of chronic stress, we highlight an array of studies on molecular and immunological signaling in the context of social determinants of health (SDoH). The main topics covered include biomarkers of sympathetic nervous system and hypothalamic-pituitary-adrenal axis activation, and the role of inflammation in the biology of adversity focusing on glucocorticoid resistance and key inflammatory cytokines linked to psychosocial and environmental stressors (PSES). We then further discuss the effect of SDoH on immune cell distribution and characterization by subset, receptor expression, and function. Lastly, we describe epigenetic regulation of the chronic stress response and effects of SDoH on telomere length and aging. Ultimately, we highlight critical knowledge gaps for future research as we strive to develop more targeted interventions that account for SDoH to improve cardiometabolic health for at-risk, vulnerable populations. Much of the political rhetoric that facilitated mass incarceration was predicated on the promise of reducing fear among the public. Yet, it remains unclear whether the large increases in imprisonment experienced in many areas made residents feel less afraid. We examine this issue by integrating geographic data on imprisonment with individual-level data on fear from the General Social Survey (GSS). We find that people from states and counties with greater "cumulative imprisonment" rates were no less afraid than their counterparts from areas that imprisoned many fewer people. These findings hold for the public overall and for non-Latino whites and members of the working and middle classes, who frequently were target audiences for political rhetoric linking mass incarceration era policies to fear reduction. Our study supports growing calls to decouple crime and criminal justice policy from politics and electoral cycles, and to develop evidence-based punishment approaches organized around transparent normative principles. Extant research has provided support for the micro‐level predictions of rational choice models of crime. Yet, a central feature of the rational choice perspective in the broader social sciences—that it is multilevel in focus, situating individuals within broader community social structures—has been neglected within criminology. In this article, we discuss and test a model that links community structural characteristics to several individual expectations and preferences relevant to crime. Using data from the Pathways to Desistance study, we find that objective levels of neighborhood concentrated disadvantage influence individuals' perceptions of, and preferences for, the risks, costs, and rewards associated with offending indirectly by affecting perceived disorder and perceived opportunities for legitimate avenues of success within one's neighborhood. Objective  In obstetric emergencies, care coordination is critical in achieving a "decision-to-delivery" time of 30 minutes. Reliable communication is essential to optimize coordinated care of mother and baby. Clinical mobility (CM) platforms have been shown to improve communication during medical emergencies; however, their impact on improving decision-to-delivery times has not been shown. This study aimed to determine the impact of a new CM platform on decision-to-delivery time. Study Design  A multidisciplinary team designed a CM platform that employs a wall-mounted panel paired with mobile technology to alert all relevant clinical staff. This new platform uses in-room preprogrammed messages that alert predetermined responders matching the emergency. For example, the "STAT section" button summons doctors (obstetrics, anesthesiology, and neonatology), obstetric nurses, and newborn resuscitation staff via smartphones. Impact of this platform was assessed with process and outcome data: cord artery pH, 5-minute Apgar's score; and decision for cesarean section to time of: skin incision, uterine incision, and delivery. This pre- and postimplementation study (October–September 2018 vs. January–December 2019) centered on the opening of our new Obstetric Care Unit. Data were analyzed with Chi-square and Mann–Whitney U -test. Results  Emergent cesarean delivery was performed in 172 women pre- and 124 postimplementation of the new CM platform. In postimplementation, we observed a 7.4-minute reduction in time from decision-to-delivery (26 pre- vs. 18.6 minutes postimplementation, p  = 0.001). Delivery within 30 minutes improved by 15.2% (p  = 0.018). Times to skin and uterine incision were also significantly reduced. The two groups had similar neonatal outcomes: birth weight, Apgar's score at 5 minutes, and cord artery pH did not differ, but the study was underpowered to compare these outcomes. Conclusion  This new CM platform significantly reduced decision-to-delivery time, in turn improving compliance with the "30-minute rule." All relevant personnel were contacted specifically, while avoiding overhead paging and other unnecessary messages. • 37% of caregivers opposed vaccine mandates at all school levels. • Only 20% of caregivers agreed with immediate vaccine mandates. • Vaccine safety was the most important concern regarding vaccine mandates. • 26% of caregivers would remove their child from school if vaccines became mandated. Caregiver attitudes toward mandating COVID-19 vaccines for their children are poorly understood. We aimed to determine caregiver acceptability of COVID-19 vaccine mandates for schools/daycares and assess if opposition to mandates would result in removal of children from the educational system. Perform a cross-sectional, anonymous survey of adult caregivers with children ≤ 18 years presenting to 21 pediatric emergency departments in the United States, Canada, Israel, and Switzerland, November 1st through December 31st, 2021. The primary outcome was caregiver acceptance rates for school vaccine mandates, and the secondary outcomes included factors associated with mandate acceptance and caregiver intention to remove the child from school. Of 4,393 completed surveys, 37% of caregivers were opposed to any school vaccine mandate. Caregiver acceptance was lowest for daycare settings (33%) and increased as the child's level of education increased, college (55%). 26% of caregivers report a high likelihood (score of 8–10 on 0–10 scale) to remove their child from school if the vaccine became mandatory. Child safety was caregivers' greatest concern over vaccine mandates. A multivariable model demonstrated intent to vaccinate their child for COVID-19 (OR = 8.9, 95% CI 7.3 to 10.8; P < 0.001) and prior COVID-19 vaccination for the caregiver (OR = 3.8, 95% CI 3.0 to 4.9; P < 0.001) had the greatest odds of increasing mandate acceptance for any school level. Many caregivers are resistant to COVID-19 vaccine mandates for schools, and acceptance varies with school level. We report on a 72-year-old patient who presented to the emergency department due to a proximal multiple-fragment humeral fracture on the left. Despite correct therapeutic approaches, there was no improvement in the left arm swelling. Further investigations showed a provoked arm vein thrombosis on the left with a post-humeral fracture on the left. • Perceived increase in vascular stretch decreases salt intake after water deprivation. • NTS AT1R+ neurons and afferents may integrate sensory and endocrine input. • NTS AT1R+neurons and afferents regulate salt intake in water-deprived mice. Angiotensin-II (Ang-II) production is driven by deviations in blood volume and osmolality, and serves the role of regulating blood pressure and fluid intake to maintain cardiovascular and hydromineral homeostasis. These actions are mediated by Ang-II acting on its type 1a receptor (AT1aR) within the central nervous system and periphery. Of relevance, AT1aR are expressed on sensory afferents responsible for conveying cardiovascular information to the nucleus of the solitary tract (NTS). We have previously determined that optical excitation of neurons and vagal afferents within the NTS that express AT1aR (referred to as NTSAT1aR) mimics the perception of increased vascular stretch and induces compensatory responses to restore blood pressure. Here, we test whether NTSAT1aR are also involved in the modulation of water and sodium intake. We directed the light-sensitive excitatory channelrhodopsin-2 (ChR2) or inhibitory halorhodopsin (Halo) to Agtr1a -containing neurons and measured water and sodium chloride (NaCl) intake in the presence and absence of optical stimulation within the NTS during various challenges to fluid homeostasis. Optical perturbation of NTSAT1aR modulates NaCl intake, such that excitation attenuates, whereas inhibition increases intake. This effect is only observed in the water-deprived condition, suggesting that NTSAT1aR are involved in the regulation of sodium intake during an imbalance in both the intracellular and extracellular fluid compartments. Furthermore, optical excitation of NTSAT1aR increases c-Fos expression within oxytocinergic neurons of the paraventricular nucleus of the hypothalamus (PVN), indicating that the regulation of sodium intake by NTSAT1aR may be mediated by oxytocin. Objective: The aim of this study was to determine whether selection of treatment for children with infantile spasms (IS) varies by race/ethnicity. Methods: The prospective US National Infantile Spasms Consortium database includes children with IS treated from 2012 to 2018. We examined the relationship between race/ethnicity and receipt of standard IS therapy (prednisolone, adrenocorticotropic hormone, vigabatrin), adjusting for demographic and clinical variables using logistic regression. Our primary outcome was treatment course, which considered therapy prescribed for the first and, when needed, the second IS treatment together. Results: Of 555 children, 324 (58%) were non‐Hispanic white, 55 (10%) non‐Hispanic Black, 24 (4%) non‐Hispanic Asian, 80 (14%) Hispanic, and 72 (13%) other/unknown. Most (398, 72%) received a standard treatment course. Insurance type, geographic location, history of prematurity, prior seizures, developmental delay or regression, abnormal head circumference, hypsarrhythmia, and IS etiologies were associated with standard therapy. In adjusted models, non‐Hispanic Black children had lower odds of receiving a standard treatment course compared with non‐Hispanic white children (odds ratio [OR], 0.42; 95% confidence interval [CI], 0.20–0.89; p = 0.02). Adjusted models also showed that children with public (vs. private) insurance had lower odds of receiving standard therapy for treatment 1 (OR, 0.42; CI, 0.21–0.84; p = 0.01). Interpretation: Non‐Hispanic Black children were more often treated with non‐standard IS therapies than non‐Hispanic white children. Likewise, children with public (vs. private) insurance were less likely to receive standard therapies. Investigating drivers of inequities, and understanding the impact of racism on treatment decisions, are critical next steps to improve care for patients with IS. Recent developments in critical information visualization have brought the field's attention to political, feminist, ethical, and rhetorical aspects of data visualization. However, less work has explored the interplay between design decisions and political ramifications—structures of authority, means of representation, etc. In this paper, we build upon these critical perspectives and highlight the political aspect of civic text visualization especially in the context of democratic decision‐making. Based on a critical analysis of survey papers about text visualization in general, followed by a review on the status quo of text visualization in civics, we argue that civic text visualization inherits an exclusively analytic framing. This framing leads to a series of issues and challenges in the fundamentally political context of civics, such as misinterpretation of data, missing minority voices, and excluding the public from decision making processes. To span this gap between political context and analytic framing, we provide a series of two‐pole conceptual dimensions, such as from singular user to multiple relationships, and from complexity to inclusivity of visualization design. For each dimension, we discuss how the tensions between these poles can help surface the political ramifications of design decisions in civic text visualization. These dimensions can thus help visualization researchers, designers, and practitioners attend more intentionally to these political aspects and inspire their design choices. Recent scholarship has examined changes in the geographic distribution of poor persons in America, but it remains unclear whether high- and low- poverty neighborhoods have become more, or less, spatially clustered over the past several decades. Additionally, while many have argued that growth in both high-poverty spatial clusters and high-low poverty spatial clusters could yield conditions that are conducive to increases in homicide, previous research has not considered that possibility. We contribute to knowledge by examining whether there have been important shifts in the spatial clustering of poverty in America between 1980 and 2010, and if so, whether those shifts were related to changes in homicide during the period. The descriptive results of our study reveal that there were notable changes in population exposure to both high- poverty and high-low poverty spatial clusters between 1980 and 2010. Fixed-effects negative binomial regression models yield limited support for the idea that changes in spatial inequality, as measured by the clustering of high- and low-poverty neighborhoods, are associated with changes in homicide rates. In contrast, the results indicate a significant positive association between changes in exposure to very high-poverty spatial clusters and homicide trends. Place of residence is one factor that may affect individual engagement and experiences in outdoor recreation. In this paper we assess four measures of place of residence and their associations with fears related to outdoor recreation. Data are from a statewide survey of Missouri households conducted in 2013 (n = 3,204). Logistic regression models are used to examine different measurements of place of residence and their relationship to the likelihood respondents reported environmental or social fears related to participation in outdoor recreation. The relationship between place of residence and reporting environmental fears was contingent on how place of residence was measured. In contrast, results related to social fears were more consistent such that residents of more urban and suburban settings were significantly more likely to report a social fear related to outdoor recreation activities regardless of how place of residence was operationalized. We present a programmatic approach to incorporating ethics into an undergraduatemajor in statistical and data sciences. We discuss departmental-level initiatives designed to meet the National Academy of Sciences recommendation for integrating ethics into the curriculum fromtop-to-bottomas our majors progress from our introductory courses to our senior capstone course, as well as from side-to-side through co-curricular programming. We also provide six examples of data science ethics modules used in five different courses at our liberal arts college, each focusing on a different ethical consideration. The modules are designed to be portable such that they can be flexibly incorporated into existing courses at different levels of instruction with minimal disruption to syllabi. We connect our efforts to a growing body of literature on the teaching of data science ethics, present assessments of our effectiveness, and conclude with next steps and final thoughts. The advancement of scientific knowledge increasingly depends on ensuring that data‐driven research is reproducible: that two people with the same data obtain the same results. However, while the necessity of reproducibility is clear, there are significant behavioral and technical challenges that impede its widespread implementation and no clear consensus on standards of what constitutes reproducibility in published research. We present fertile, an R package that focuses on a series of common mistakes programmers make while conducting data science projects in R, primarily through the RStudio integrated development environment. fertile operates in two modes: proactively, to prevent reproducibility mistakes from happening in the first place, and retroactively, analyzing code that is already written for potential problems. The OCA2-HERC2 locus is responsible for the greatest proportion of eye color variation in humans. Numerous studies extensively described both functional SNPs and associated patterns of variation over this region. The goal of our study is to examine how these haplotype structures and allelic associations vary when highly variable markers such as microsatellites are used. Eleven microsatellites spanning 357 Kb of OCA2-HERC2 genes are analyzed in 3029 individuals from worldwide populations. We found that several markers display large differences in allele frequency (10% to 35% difference) among Europeans, East Asians and Africans. In Europe, the alleles showing increased frequency can also discriminate individuals with (IrisPlex) predicted blue and brown eyes. Distinct haplotypes are identified around the variants C and T of the functional SNP rs12913832 (associated to blue eyes), with linkage disequilibrium r2 values significant up to 237 Kb. The haplotype carrying the allele rs12913832 C has high frequency (76%) in blue eye predicted individuals (30% in brown eye predicted individuals), while the haplotype associated to the allele rs12913832 T is restricted to brown eye predicted individuals. Finally, homozygosity values reach levels of 91% near rs12913832. Odds ratios show values of 4.2, 7.4 and 10.4 for four markers around rs12913832 and 7.1 for their core haplotype. Hence, this study provides an example on the informativeness of multiallelic markers that, despite their current limited potential contribution to forensic eye color prediction, supports the use of microsatellites for identifying causing variants showing similar genetic features and history. Until recently, national‐level data on criminal victimization in the United States did not include information on immigrant or citizenship status of respondents. This data‐infrastructure limitation has hindered scientific understanding of whether immigrants are more or less likely than native‐born Americans to be criminally victimized and how victimization may vary among immigrants of different statuses. We address these issues in the present study by using new data from the 2017–2018 National Crime Victimization Survey (NCVS) to explore the association between citizenship status and victimization risk in a nationally representative sample of households and persons aged 12 years and older. The research is guided by a theoretical framing that integrates insights from studies of citizenship with the literature on immigration and crime, as well as with theories of victimization. We find that a person's foreign‐born status (but not their acquired U.S. citizenship) confers protection against victimization. We also find that the protective benefit associated with being foreign born does not extend to those with ambiguous citizenship status, who in our data exhibit attributes similar to the known characteristics of undocumented immigrants. There are approximately 38 million people globally living with Human immunodeficiency virus 1 (HIV-1) and given the tremendous success of combination antiretroviral therapy (cART) this has dramatically reduced mortality and morbidity with prevention benefits. However, HIV-1 persists during cART within the human body and re-appears upon cART interruption. This HIV-1 reservoir remains a barrier to cure with cellular sites of viral persistence not fully understood. In this study we provide evidence corroborating a recently published article in STM demonstrating the role of platelets as a novel cellular disseminator of HIV-1 particles in the setting of viral suppression. Using classical transmission electron microscopy with and without immunogold labeling, we visualize HIV-1 in both platelets and monocytes in cART suppressed HIV donors. Our study suggests that due to the close proximity of platelets and monocytes an alternative life cycle of HIV-1 cycling within monocytes and platelets without the need of active replication under cART occurs. Our findings are supported by the lack of detectable HIV-1 particles in platelets derived from HIV uninfected donors or the 'Berlin' patient suggesting that platelets may serve as an underappreciated hidden bearer for HIV-1 and should be considered in HIV remission studies and trials. Summary Changes in chromosome numbers are a prominent driver of plant evolution, impacting ecological diversification, stress tolerance, and phenotypes. ChromEvol is a widely used software tool for deciphering patterns of chromosome‐number change along a phylogeny of interest. It evaluates the fit of alternative models to the data, estimates transition rates of different types of events, and infers the expected number of events along each branch of the phylogeny. We introduce ChromEvol v.3, featuring multiple novel methodological advancements that capture variation in the transition rates along a phylogeny. This version better allows researchers to identify how dysploidy and polyploidy rates change based on the number of chromosomes in the genome, with respect to a discrete trait, or at certain subclades of the phylogeny. We demonstrate the applicability of the new models on the Solanaceae phylogeny. Our analyses identify four chromosome‐number transition regimes that characterize distinct Solanaceae clades and demonstrate an association between self‐compatibility and altered dynamics of chromosome‐number evolution. Introduction: Localized provoked vulvodynia (LPV) is a chronic condition in which patients experience disabling pain in the vulvar vestibule. This condition's mechanisms are poorly understood, and the lack of adequate treatment reflects the misunderstanding of the disease's origins. Many LPV patients attempt to treat their pain with NSAIDS, but their use has been clinically shown to be ineffective against LPV associated vulvar pain. Recent evidence, utilizing fibroblasts isolated from vestibular and vulvar tissue of LPV patients, supports a link between inflammation and symptomology of LPV. These fibroblasts have shown to be sensitive to pro-inflammatory stimuli and overproduce inflammatory mediators such as Interleukin-6 (IL-6) and Prostaglandin E2 (PGE2). Increased production of IL-6 and PGE2 correlates with decreased pain thresholds, endorsing these levels as a surrogate measure for pain. Lipidomic data collected via mass spectroscopy also demonstrates LPV fibroblast strains have dysregulated arachidonic acid metabolism, resulting in an imbalance in pro-resolving lipids. Utilizing this fibroblast model, different cyclooxygenase (COX) inhibitors were used to better illuminate the mechanism behind fibroblast inflammation in LPV. Objective: Using a fibroblast cell model, we set out to better understand pathophysiology of LPV and use that information to guide development of effective and pragmatic medical treatments. Methods: Fibroblast strains were generated from vulvar and vestibular tissue samples of LPV patients and non-LPV age and race-matched controls. These cells were then treated with Interleukin 1 beta (IL-1β), selective cyclooxygenase-1 inhibitor (SC560), celecoxib (selective COX-2 inhibitor), and ibuprofen (nonselective COX inhibitor) to investigate differences in cellular response when blocking different enzymes in the inflammatory pathway. IL-1β is known to induce IL-6 and PGE2 which acted as a positive control. Results were then quantified via ELISA assay. Results: Vestibular and vulvar tissues showed an increase in IL-6 and PGE2 production following treatment with IL-1B. When concurrently treated with IL-1B (to stimulate inflammation) and SC560 (to inhibit COX-1 signaling) ELISA analysis showed a significant decrease in PGE2. A significant PGE2 decrease was shown with concurrent IL-1B/Ibuprofen treatment as well as with IL-1B/Celecoxib treatments. In all three cases, IL-6 signaling was generally not decreased, while PGE2 was decreased, but not to baseline levels. Ibuprofen treatment in fact showed a trend towards increased IL-6 production. Conclusions: Fibroblast response to inflammation in the case of LPV is complex. COX inhibitors do not fully inhibit the pain signaling that contributes to the symptomology of LPV patients (IL-6 production remains elevated). NSAIDs leave patients with dysregulated inflammation cascades, which does not remediate pain. Background: Motor impairment after stroke is due not only to direct tissue loss but also to disrupted connectivity within the motor network. Mixed results from studies attempting to enhance motor recovery with Transcranial Magnetic Stimulation (TMS) highlight the need for a better understanding of both connectivity after stroke and the impact of TMS on this connectivity. This study used TMS-EEG to map the causal information flow in the motor network of healthy adult subjects and define how stroke alters these circuits.Methods: Fourteen stroke patients and 12 controls received TMS to two sites (bilateral primary motor cortices) during two motor tasks (paretic/dominant hand movement vs. rest) while EEG measured the cortical response to TMS pulses. TMS-EEG based connectivity measurements were derived for each hemisphere and the change in connectivity (ΔC) between the two motor tasks was calculated. We analyzed if ΔC for each hemisphere differed between the stroke and control groups or across TMS sites, and whether ΔC correlated with arm function in stroke patients.Results: Right hand movement increased connectivity in the left compared to the right hemisphere in controls, while hand movement did not significantly change connectivity in either hemisphere in stroke. Stroke patients with the largest increase in healthy hemisphere connectivity during paretic hand movement had the best arm function.Conclusions: TMS-EEG measurements are sensitive to movement-induced changes in brain connectivity. These measurements may characterize clinically meaningful changes in circuit dynamics after stroke, thus providing specific targets for trials of TMS in post-stroke rehabilitation. Compounds that selectively boost prion Z remain unreported. Here, we synthesized compounds that amplified the negative charge of misfolded superoxide dismutase‐1 (SOD1) by acetylating lysine‐NH3+ in amyloid‐SOD1, without acetylating native‐SOD1. Compounds resembled a "ball and chain" mace: a rigid amyloid‐binding "handle" (benzothiazole, stilbene, or styrylpyridine); an aryl ester "ball"; and a triethylene glycol chain connecting ball to handle. At stoichiometric excess, compounds acetylated up to 9 of 11 lysine per misfolded subunit (ΔZfibril=−8100 per 103 subunits). Acetylated amyloid‐SOD1 seeded aggregation more slowly than unacetylated amyloid‐SOD1 in vitro and organotypic spinal cord (these effects were partially due to compound binding). The COVID-19 pandemic has highlighted structural inequalities and racism promoting health disparities among communities of color. Taking cardiovascular disease as an example, we provide a framework for multidisciplinary efforts leveraging translational and epidemiologic approaches to decode the biological impacts of inequalities and racism and develop targeted interventions that promote health equity. Youth involvement in crime has declined substantially over the past few decades, yet the reasons for this trend remain unclear. We advance the literature by examining the role of several potentially important shifts in individual attitudes and behaviors that may help to account for the observed temporal variation in youth delinquency. Our multilevel analysis of repeated cross‐sectional data from eighth and tenth grade students in the Monitoring the Future (MTF) study indicates that changes in youth offending prevalence were not associated with changes in youth attachment and commitment to school, community involvement, or parental supervision after school. In contrast, the study provides suggestive evidence that the significant reduction in youth offending prevalence observed since the early 1990s was significantly associated with a decrease in unstructured socializing and alcohol consumption and, to a lesser extent, with a decrease in youth preferences for risky activities. The sediment succession of Lake Emanda in the Yana Highlands was investigated to reconstruct the regional late Quaternary climate and environmental history. Hydro‐acoustic data obtained during a field campaign in 2017 show laminated sediments in the north‐western and deepest (up to ̃15 m) part of the lake, where a ̃6‐m‐long sediment core (Co1412) was retrieved. The sediment core was studied with a multi‐proxy approach including sedimentological and geochemical analyses. The chronology of Co1412 is based on 14C AMS dating on plant fragments from the upper 4.65 m and by extrapolation suggests a basal age of c. 57 cal. ka BP. Pronounced changes in the proxy data indicate that early Marine Isotope Stage (MIS) 3 was characterized by unstable environmental conditions associated with short‐term temperature and/or precipitation variations. This interval was followed by progressively colder and likely drier conditions during mid‐MIS 3. A lake‐level decline between 32.0 and 19.1 cal. ka BP was presumably related to increased continentality and dry conditions peaking during the Last Glacial Maximum (LGM). A subsequent rise in lake level could accordingly have been the result of increased rainfall, probably in combination with seasonally high meltwater input. A milder or wetter Lateglacial climate increased lake productivity and vegetation growth, the latter stabilizing the catchment and reducing clastic input into the lake. The Bølling‐Allerød warming, Younger Dryas cooling and Holocene Thermal Maximum (HTM) are indicated by distinct changes in the environment around Lake Emanda. Unstable, but similar‐to‐present‐day climatic and environmental conditions have persisted since c. 5 cal. ka BP. The results emphasize the highly continental setting of the study site and therefore suggest that the climate at Lake Emanda was predominantly controlled by changes in summer insolation, global sea level, and the extent of ice sheets over Eurasia, which influenced atmospheric circulation patterns. Solvent extracts of mammalian tissues and blood contain a large amount of co-extracted matrix components, in particular lipids, which can adversely affect instrumental analysis. Clean-up typically degrades non-persistent chemicals. Alternatively, passive sampling with the polymer polydimethylsiloxane (PDMS) has been used for a comprehensive extraction from tissue without altering the mixture composition. Despite a smaller fraction of matrix being co-extracted by PDMS than by solvent extraction, direct analysis of PDMS extracts was only possible with direct sample introduction (DSI) GC-MS/MS, which prevented co-extracted matrix components entering the system. Limits of quantitation (LOQ) ranged from 4 to 20 pg μL−1 ethyl acetate (PDMS extract) for pesticides and persistent organic pollutants (POPs). The group of organophosphorus flame retardants showed higher LOQs up to 107 pg μL−1 due to sorption to active sites at the injection system. Intraday precision ranged between 1 and 10%, while the range of interday precision was between 1 and 18% depending on the analyte. The method was developed using pork liver, brain, and fat as well as blood and was then applied to analyze human post-mortem tissues where polychlorinated biphenyls (PCBs) as well as dichlorodiphenyltrichloroethane (DDT) and DDT metabolites were detected. Mammalian taste buds are comprised of specialized neuroepithelial cells that act as sensors for molecules that provide nutrition (e.g., carbohydrates, amino acids, and salts) and those that are potentially harmful (e.g., certain plant compounds and strong acids). Type II and III taste bud cells (TBCs) detect molecules described by humans as "sweet," "bitter," "umami," and "sour." TBCs that detect metallic ions, described by humans as "salty," are undefined. Historically, type I glial-like TBCs have been thought to play a supportive role in the taste bud, but little research has been done to explore their role in taste transduction. Some evidence implies that type I cells may detect sodium (Na+) via an amiloride-sensitive mechanism, suggesting they play a role in Na 1 taste transduction. We used an optogenetic approach to study type 1 TBCs by driving the expression of the light-sensitive channelrhodopsin-2 (ChR2) in type I GAD65+ TBCs of male and female mice. Optogenetic stimulation of GAD65+ TBCs increased chorda tympani nerve activity and activated gustatory neurons in the rostral nucleus tractus solitarius. "N neurons," whose NaCl responses were blocked by the amiloride analog benzamil, responded robustly to light stimulation of GAD65+ TBCs on the anterior tongue. Two-bottle preference tests were conducted under Na+-replete and Na+-deplete conditions to assess the behavioral impact of optogenetic stimulation of GAD65+ TBCs. Under Na+-deplete conditions GAD65-ChR2-EYFP mice displayed a robust preference for H20 illuminated with 470 nm light versus nonilluminated H20, suggesting that type I glial-like TBCs are sufficient for driving a behavior that resembles Na appetite. Palm fat is often used in baked goods because of its relatively low cost, and its positive impact on texture and shelf life. Demand for alternatives has risen in recent years due to concerns about the ecological and social sustainability. This is a challenge for the bakery industry since palm fat possesses unique properties. In this study, unhydrogenated rapeseed oil was processed using novel physical technologies, such as wax crystallisation, stabilised foaming and Pickering emulsions, in order to simulate palm fat properties. Analysis showed that while the initial viscosity of the fat substitute products was low compared to palm fat, the fat replacement products behaved very similarly to palm fat in the baking experiments. The resulting biscuits baked with emulsified rapeseed oil and rapeseed oil complemented with wax crystals were judged to be suitable replacements for palm fat in terms of processability, as well as analytical and sensory assessment. Biocatalytic redox reactions regularly depend on expensive cofactors that require recycling. For continuous conversions in flow chemistry, this is often an obstacle since the cofactor is washed away. Here, we present a quasi‐stationary recycling system for nicotinamide adenine dinucleotide phosphate utilizing an immobilized alcohol dehydrogenase. Four model substrates were reduced with high enantioselectivity as a proof of concept. The two‐phase system enables continuous production as well as quick substrate changes. This setup may serve as a general cofactor regeneration module for continuous biocatalytic devices employing (co‐)substrates being miscible in organic solvent. The system resulted in space‐time yields up to 117 g L−1 h−1 and total turnover numbers for nicotinamide adenine dinucleotide phosphate higher than 12,000 mol/mol are possible. Analysis of the brain's surface can potentially assist in providing a better understanding of structural brain differences, and may help characterize DS-specific brain development. We performed a study of 73 magnetic resonance imaging (MRI) examinations of DS patients (aged 1 day to 22 years) and compared them to a large cohort of 993 brain MRI examinations of neurotypical participants, aged 1 day to 32 years. A variety of measurements that characterize the surface of the brain were extracted from each brain region in each examination. Results demonstrate broad reductions in surface area and abnormalities of surface curvature measurements across the brain in DS. Findings suggest the presence of developmental abnormalities of the brain's surface in DS that can be characterized from clinical MRI examinations. Surface curvature measurements, absolute surface area measurements, and surface areas as a percentage of total brain surface area (%TBSA) were extracted from each brain region in each examination. Results demonstrate broad reductions in surface area and abnormalities of surface curvature measurements across the brain in DS. After adjusting our regional surface area measurements as %TBSA, abnormally increased presentation in DS relative to neurotypical controls was observed in the left precentral, bilateral entorhinal, left parahippocampal, and bilateral perirhinal cortices, as well as Brodmann's area 44 (left), and the right temporal pole.
