ABSTRACT: Omitting or shortening the dry period may result in a fairly constant ration throughout the transition period of dairy cows, reducing the need for adaptation of cow metabolism and rumen function to a new lactation. The objective of this study was to determine the effect of dry period length (DP) on rumen adaptation and cow metabolic state during the transition period. Twelve pregnant, rumen-cannulated Holstein Friesian dairy cows at the end of their first lactation were assigned to 1 of 3 treatments: a conventional (60 d), short (30 d), or no dry period (0 d). At dry-off, cows received a dry-cow ration until calving. Lactating cows received a lactation ration. Cows were monitored from 8 wk before calving until 8 wk after calving for milk yield and DMI. Rumen biopsies were taken from 3 locations in the rumen at 60, 40, and 10 d before calving and 3, 7, 14, 28, and 56 d after calving to assess papillae dimensions. Blood was sampled weekly from 3 wk before until 8 wk after calving, and liver biopsies were taken at wk −2, 2, and 4 relative to calving. Prepartum, DMI, and milk yield were greater for cows with a short or no dry period, compared with cows with a conventional dry period. Postpartum, DMI was greater for cows with a short dry period compared with cows with a conventional dry period. Plasma glucose concentration was greater for cows without a dry period, compared with the other DP postpartum. Plasma concentrations of nonesterified fatty acids and BHB, and liver triglyceride content, did not differ among dry period. Rumen papillae differed in size based on biopsy location, but there was no interaction between biopsy location and the effect of DP. Rumen papillae surface area for cows managed for a 30- or 60-d dry period decreased toward calving. At 40 d prepartum, papillae surface area was greater for short and no dry period treatment compared with a conventional dry period. At 10 d prepartum, papillae surface area was greater for the no dry period treatment compared with both other treatments, and this difference was still present 3 d postpartum. Cows managed for a short dry period showed faster increase in papillae dimensions after calving compared with cows managed for a conventional dry period. From d 28 onwards, no differences in papillae surface area were observed. The faster rumen adaptation postpartum may be related to the increased DMI during the first weeks postpartum for cows managed for a short dry period. However, this did not result in improved metabolic status or milk yield. The results from the present study demonstrate that the dietary changes related to a conventional DP affected rumen papillae development, not only prepartum but also early postpartum. Further optimization of DP as well as dietary composition throughout the transition period may support cows in their adaptation to a new lactation.

In this study, Senescence Accelerated Mice (SAMP8) were supplemented with exogenous DHA milk, endogenous DHA milk, normal milk, or 0.9 % saline solution. Enzyme-linked immunosorbent assay (ELISA), gas chromatography (GC), ultra-performance liquid chromatography-electrospray ionization-tandem mass spectrometry (UPLC-ESI MS/MS), and Morris water maze were used to characterize the effects of diet on oxidative stress and cognition in SAMP8 mice. Supplementation endogenous DHA milk or exogenous DHA milk can enhance the antioxidant capacity of mice organs. Endogenous DHA milk increased the superoxide dismutase (SOD) activity of mice brain and serum than normal milk and 0.9 % saline solution (P ≤ 0.05), as well as increased SOD activity of mice liver and glutathione peroxidase (GSH-Px) activity of mice brain than normal milk (P ≤ 0.05). Exogenous DHA milk increased SOD activity of mice brain than normal milk and 0.9 % saline solution, as well as increased SOD activity of mice serum than 0.9 % saline solution (P ≤ 0.05). Several polar lipid relative content, such as 18:0/18:2 PS, 17:0 Ceramide, and 20:4 LPC in mice brain was affected by dietary supplementation with DHA-containing milk. Lipid oxidation metabolites in mice brain were not affected by DHA-containing milk. Endogenous DHA milk increased the number of platform location crossing times of mice in the Morris water maze test, compared with Exogenous DHA milk, normal milk, and 0.9 % saline solution (P ≤ 0.05).

Abstract Lateral wall thickness is a known predictor for postoperative stability of trochanteric femoral fractures and occurrence of secondary lateral wall fractures. Currently, the AO/OTA classification relies on the absolute lateral wall thickness (aLWT) to distinguish between stable A1.3 and unstable A2.1 fractures that does not take interpersonal patient differences into account. Thus, a more individualized and accurate measure would be favorable. Therefore, we proposed and validated a new patient-specific measure—the relative lateral wall thickness (rLWT)—to consider individualized measures and hypothesized its higher sensitivity and specificity compared with aLWT. First, in 146 pelvic radiographs of patients without a trochanteric femoral fracture, the symmetry of both caput-collum-diaphyseal angle (CCD) and total trochanteric thickness (TTT) was assessed to determine whether the contralateral side can be used for rLWT determination. Then, data of 202 patients were re-evaluated to compare rLWT versus previously published aLWT. Bilateral symmetry was found for both CCD and TTT (p ≥ 0.827), implying that bone morphology and geometry of the contralateral intact side could be used to calculate rLWT. Validation revealed increased accuracy of the rLWT compared with the gold standard aLWT, with increased specificity by 3.5% (Number Needed to Treat = 64 patients) and sensitivity by 1% (Number Needed to Treat = 75 patients). The novel rLWT is a more accurate and individualized predictor of secondary lateral wall fractures compared with the standard aLWT. This study established the threshold of 50.5% rLWT as a reference value for predicting fracture stability in trochanteric femoral fractures.

Extending lactation length reduces the frequency of critical calving events for the cow and herewith reduces the frequency of periods with increased risk for health problems. Moreover, breeding is postponed until a moment later in lactation, which is associated with better conception rates and less days open after start of the breeding period in most studies. Potential risks of an extended lactation are that milk yield of cows at the end of the lactation may be too low which may lead to cows being overconditioned at the end of the extended lactation. Therefore, extending lactation length might not fit every cow. Individual cow characteristics like parity, milk yield level, or body condition determine the response of the cow to an extended lactation. These individual cow characteristics can be used in customized management strategies to optimize lactation length for individual cows. Customized lactation length for individual cows could limit the impact at herd level of disadvantages concerning milk losses and overconditioning and maintain benefits for improved cow health and fertility, reduced number of surplus calves and increased work satisfaction for the farmer. In conclusion, extending lactation length has interesting perspectives for health and fertility of high-producing dairy cows, although questions remain concerning management approaches to support lactation persistency of cows with an extended lactation, and consequences for calf health and development. ABSTRACT: Early lactation metabolic imbalance is an important physiological change affecting the health, production, and reproduction of dairy cows. The aims of this study were (1) to evaluate the potential of test-day (TD) variables with or without milk fatty acids (FA) content to classify metabolically imbalanced cows and (2) to evaluate the robustness of the metabolic classification with external data. A data set was compiled from 3 experiments containing plasma β-hydroxybutyrate, nonesterified FA, glucose, insulin-like growth factor-I, FA proportions in milk fat, and TD variables collected from 244 lactations in wk 2 after calving. Based on the plasma metabolites, 3 metabolic clusters were identified using fuzzy c-means clustering and the probabilistic membership value of each cow to the 3 clusters was determined. Comparing the mean concentration of the plasma metabolites, the clusters were differentiated into metabolically imbalanced, moderately impacted, and balanced. Following this, the 2 metabolic status groups identified were imbalanced cows (n = 42), which were separated from what we refer to as “others” (n = 202) based on the membership value of each cow for the imbalanced cluster using a threshold of 0.5. The following 2 FA data sets were composed: (1) FA (groups) having high prediction accuracy by Fourier-transform infrared spectroscopy and, thus, have practical significance, and (2) FA (groups) formerly identified as associated with metabolic changes in early lactation. Metabolic status prediction models were built using FA alone or combined with TD variables as predictors of metabolic groups. Comparison was made among models and external evaluations were performed using an independent data set of 115 lactations. The area under the receiver operating characteristics curve of the models was between 75 and 91%, indicating their moderate to high accuracy as a diagnostic test for metabolic imbalance. The addition of FA groups to the TD models enhanced the accuracy of the models. Models with FA and TD variables showed high sensitivities (80–88%). Specificities of these models (73–79%) were also moderate and acceptable. The accuracy of the FA models on the external data set was high (area under the receiver operating characteristics curve between 76 and 84). The persistently good performance of models with Fourier-transform infrared spectroscopy-quantifiable FA on the external data set showed their robustness and potential for routine screening of metabolically imbalanced cows in early lactation.

ABSTRACT: The objective of this study was to investigate relations between feeding behavior (FB) variables focusing on intake of the basal ration during 3 wk prepartum until 3 wk postpartum, and postpartum onset of luteal activity (OLA) in dairy cows subjected to no (0-d) or a short (30-d) dry period (DP). Feeding behavior of the basal ration intake (concentrate intake excluded) was continuously recorded by computerized feeders for 123 dairy cows and analyzed from 3 wk prepartum to 3 wk postpartum. During early lactation, dairy cows have a negative energy balance since their energy demands exceed their energy intake: in this study, we aimed to investigate the association between diet and plasma metabolomics profiles and how these relate to energy unbalance of course in the early-lactation stage. Holstein-Friesian cows were randomly assigned to a glucogenic (n = 15) or lipogenic (n = 15) diet in early lactation. Blood was collected in week 2 and week 4 after calving. Plasma metabolite profiles were detected using liquid chromatography–mass spectrometry (LC-MS), and a total of 39 metabolites were identified. Two plasma metabolomic profiles were available every week for each cow. Metabolite abundance and metabolite ratios were used for the analysis using the XGboost algorithm to discriminate between diet treatment and lactation week. Using metabolite ratios resulted in better discrimination performance compared with the metabolite abundances in assigning cows to a lipogenic diet or a glucogenic diet. The quality of the discrimination of performance of lipogenic diet and glucogenic diet effects improved from 0.606 to 0.753 and from 0.696 to 0.842 in week 2 and week 4 (as measured by area under the curve, AUC), when the metabolite abundance ratios were used instead of abundances. The top discriminating ratios for diet were the ratio of arginine to tyrosine and the ratio of aspartic acid to valine in week 2 and week 4, respectively. For cows fed the lipogenic diet, choline and the ratio of creatinine to tryptophan were top features to discriminate cows in week 2 vs. week 4. For cows fed the glucogenic diet, methionine and the ratio of 4-hydroxyproline to choline were top features to discriminate dietary effects in week 2 or week 4. This study shows the added value of using metabolite abundance ratios to discriminate between lipogenic and glucogenic diet and lactation weeks in early-lactation cows when using metabolomics data. The application of this research will help to accurately regulate the nutrition of lactating dairy cows and promote sustainable agricultural development.

Background and Objectives: The surgical treatment of proximal humeral shaft fractures usually considers application of either long straight plates or intramedullary nails. By being able to spare the rotator cuff and avoid the radial nerve distally, the implementation of helical plates might overcome the downsides of common fixation methods. The aims of the current study were (1) to explore the biomechanical competence of different plate designs and (2) to compare their performance versus the alternative treatment option of using intramedullary nails. Materials and Methods: Twenty-four artificial humeri were assigned to the following four groups for simulation of an unstable proximal humeral shaft fracture and instrumentation: Group 1 (Straight-PHILOS), Group 2 (MULTILOC-Nail), Group 3 (45°-Helical-PHILOS), and Group 4 (90°-Helical-PHILOS). All specimens underwent non-destructive, quasi-static biomechanical testing under loading in axial compression, torsion in internal/external rotation, and pure bending in four directions, accompanied by motion tracking. Results: Axial stiffness/displacement in Group 2 was significantly higher/smaller than in all other groups (p ≤ 0.010). Torsional displacement in Group 2 was significantly bigger than in all other groups (p ≤ 0.017). Significantly smaller coronal plane displacement was identified in Group 2 versus all other groups (p < 0.001) and in Group 4 versus Group 1 (p = 0.022). Significantly bigger sagittal plane displacement was detected in Group 4 versus all other groups (p ≤ 0.024) and in Group 1 versus Group 2 (p < 0.001). Conclusions: Intramedullary nails demonstrated higher axial stiffness and smaller axial interfragmentary movements compared with all investigated plate designs. However, they were associated with bigger torsional movements at the fracture site. Although 90°-helical plates revealed bigger interfragmentary movements in the sagittal plane, they demonstrated improved resistance against displacements in the coronal plane when compared with straight lateral plates. In addition, 45°-helical plates manifested similar biomechanical competence to straight plates and may be considered a valid alternative to the latter from a biomechanical standpoint.

ABSTRACT: Feed is a major cost in dairy production, and substantial genetic variation in feed efficiency exists between cows. Therefore, breeders aim to improve feed efficiency of dairy cattle. However, phenotypic data on individual feed intake on commercial farms is scarce, and accurate measurements are very costly. Several studies have shown that information from Fourier-transformed infrared spectra of milk samples (milk infrared, milk IR) can be used to predict phenotypes such as energy balance and energy intake, but this is usually based on small data sets obtained under experimental circumstances. The added value of information from milk IR spectra for estimation of breeding values is unknown. The objectives of this study were (1) to develop prediction equations for dry matter intake (DMI) and residual DMI (rDMI) from milk IR spectra; (2) to apply these for a data set of milk IR spectra from commercial Dutch dairy farms; (3) to estimate genetic parameters for these traits; and (4) to estimate correlations between these predictions and other traits in the breeding goal. We used data from feeding trials where individual feed intake was recorded daily and for which milk IR spectra were determined weekly to develop prediction equations for DMI and rDMI with partial least squares regression. This data set contained over 7,600 weekly averaged DMI records linked with milk IR spectra from 271 cows. The equations were applied for a data set with test day information from 676 Dutch dairy herds with 621,567 records of 78,488 cows. Both milk IR-predicted DMI and rDMI were analyzed with an animal model to obtain genetic parameters and sire effect estimates that could be correlated with breeding values. A partial least squares regression model with 10 components from the milk IR spectra explained around 25% of DMI variation and less than 10% of rDMI variation in the validation set. Nearly all variation in the milk IR spectra was captured by 7 components; additional components contributed marginally to the spectral variation but decreased prediction errors for both traits. Accuracies of predictions of DMI and rDMI from milk IR spectra for a large feeding experiment were 0.47 and 0.26 on average, respectively, with small differences between ration treatments (ranging from 0.43 to 0.55 and from 0.21 to 0.34, respectively) and among lactation stages (ranging from 0.24 to 0.59 and from 0.13 to 0.36, respectively), with the highest prediction accuracies in early lactation. The estimated heritabilities for predicted DMI and rDMI were 0.3 and 0.4, respectively, which suggests genetic potential for both predicted traits. The correlations of sire estimates for milk IR-predicted DMI with official Dutch breeding values were strongest with milk production (0.33), longevity (0.26), and fertility (−0.27), indicating that cows that eat more produce more, live longer, and have poorer fertility. The correlations of sire estimates for predicted DMI and rDMI with the official breeding values for DMI were low (0.14 and 0.03, respectively). This implies that the added value of including milk IR-predicted DMI information in the estimation procedure of breeding values for DMI would be considered insufficient for practical application.

ABSTRACT: Based on modeling studies, a 1-yr calving interval for dairy cows is generally considered optimal from an economic point of view. Recently some dairy farmers are deliberately extending the voluntary waiting period for insemination (VWP) to extend the calving interval. Reasons to extend the VWP are to reduce the frequency of transitions such as dry-off and calving to improve health, to reduce labor associated with these transitions, and to reduce the number of surplus calves. This study aimed to evaluate yearly revenues, yearly costs, and yearly net partial cash flow (NPCF) for individual cows with a VWP of 50, 125, or 200 d based on data from a randomized control trial. The NPCF included revenues and costs for milk yield, calves born, inseminations, concentrate supply, partial mixed ration (PMR) supply, veterinary treatments, discarded milk due to veterinary treatments, culling, and labor (for milking, calving cows, inseminations, and veterinary treatments). Holstein-Friesian dairy cows (n = 153) within one herd were blocked for parity, calving season, and expected (primiparous cows) or previous (multiparous cows) 305-d milk yield. Cows were randomly assigned within the blocks to 1 of 3 VWP (VWP50, VWP125, or VWP200) in wk 6 after calving, and monitored from wk 6 after calving until wk 6 after the next calving or until culling. Revenues and costs were calculated per individual cow and expressed per cow per year. Revenues from milk and costs for PMR and concentrate contributed most to the yearly NPCF. Total yearly revenues were greater in VWP50 compared with VWP200 (€3,169 vs. €2,832), mainly because of €334 greater milk revenues. Total yearly costs were also greater in VWP50 compared with VWP200 (€1,964 vs. €1,729), mainly because of €102 greater concentrate costs. The VWP was not significantly associated with the NPCF per cow per year. A change in milk, feed, or calf price, or a change in labor costs for calving cows or for inseminations had a greater effect on the yearly NPCF of cows in VWP50 compared with cows in VWP200. To investigate variation in NPCF, cows were grouped for yearly NPCF and categorized into 3 economic classes (EC): EC1 (€1,400/yr). Cows in EC3 had greatest lactation production per day in the experiment (i.e., kg of milk, protein, fat, lactose), and lowest number of veterinary treatments during the experiment.

ABSTRACT: Due to a combination of a relatively low energy intake and a high demand of energy required for milk production, dairy cows experience a negative energy balance (EB) at the start of lactation. This energy deficit causes body weight reduction and an increased risk for metabolic diseases. Severity and length of negative EB can differ among cows. Peripartum time profiles of EB for dairy cows are not described yet in the literature. Creating EB-derived time profiles with corresponding metabolic status and disease treatments could improve understanding the relationship between EB and metabolic status, as well as enhance identification of cows at risk for compromised metabolic status. In this research we propose a novel method to cluster EB time series and examine associated metabolic status and disease treatments of dairy cows in the peripartum period. In this study, data of 3 earlier experiments were merged and examined. Four dairy cow clusters for time profiles of EB from wk −3 until +7 relative to calving were generated by the global alignment kernel algorithm. For each cluster, mean of body weight prepartum was distinguishable, indicating this might be a possible on-farm biomarker for the peripartum EB profile. Moreover, cows with severe EB drop postpartum were more treated for milk fever and had high plasma nonesterified fatty acids and β-hydroxybutyrate concentration, and low IGF-1, insulin, and glucose concentration in the first 7 wk of lactation. Overall, this study demonstrated that cows can be clustered based on EB time profiles and that characteristics such as prepartum body weight, and postpartum nonesterified fatty acids and glucose concentration are promising biomarkers to identify the time profile of EB and potentially the risk for metabolic diseases.

The purpose of this study was to compare the biomechanical stability, especially graft slippage of an allograft screw and a conventional interference screw for tibial implant fixation in ACL reconstruction. Twenty-four paired human proximal tibia specimens underwent ACL reconstruction, with the graft in one specimen of each pair fixed using the allograft screw and the other using the conventional interference screw. Specimens were subjected to cyclic tensile loading until failure. The two fixation methods did not show any statistical difference in load at graft slippage (p = 0.241) or estimated mean survival until slippage onset (p = 0.061). The ultimate load and the estimated mean survival until failure were higher for the interference screw (p = 0.04, and p = 0.018, respectively). Graft displacement at ultimate load reached values of up to 7.2 (interference screw) and 11.3 mm (allograft screw). The allograft screw for implant fixation in ACL reconstruction demonstrated comparable behavior in terms of graft slippage to the interference screw but underperformed in terms of ultimate load. However, the ultimate load, occurring at progressive graft slippage, may not be considered a direct indicator of clinical failure.

Background and Objectives: Proximal humerus fractures (PHFs) are common in the elderly, but the treatment results are often poor. A clear understanding of fracture morphology and distribution of cortical bone loss is important for improved surgical decision making, operative considerations, and new implant designs. The aim of this study was to develop a 3D segmentation fracture mapping technique to create a statistical description of the spatial pattern and cortical bone loss of complex PHFs. Materials and Methods: Fifty clinical computed tomography (CT) scans of complex PHFs and their contralateral intact shoulders were collected. In-house software was developed for semi-automated segmentation and fracture line detection and was combined with manual fracture reduction to the contralateral template in a commercial software. A statistical mean model of these cases was built and used to describe probability maps of the fracture lines and cortical fragments. Results: The fracture lines predominantly passed through the surgical neck and between the tuberosities and tendon insertions. The superior aspects of the tuberosities were constant fragments where comminution was less likely. Some fracture lines passed through the bicipital sulcus, but predominantly at its edges and curving around the tuberosities proximally and distally. Conclusions: A comprehensive and systematic approach was developed for processing clinical CT images of complex fractures into fracture morphology and fragment probability maps and applied on PHFs. This information creates an important basis for better understanding of fracture morphology that could be utilized in future studies for surgical training and implant design.

Daily milk production, and fluctuations therein, can provide information on health and resilience of dairy cows. We studied variance and autocorrelation of deviations in daily milk yield in relation to the occurrence of clinical mastitis (no, early or later in lactation). Individual lactation curves were fitted to 305-d lactations of 414 dairy cows using quantile regression. Log-transformed variance (lnVar) and autocorrelation of the quantile residuals of daily milk yield (predicted – observed) were evaluated for intervals until 30 and until 305 days in milk (DIM). Cows were classified as having no mastitis (n = 249), early mastitis that first occurred before 30 DIM (n = 29); or later mastitis (n = 136). Subsequently, linear models were used to assess effects of mastitis and parity class (primiparous or multiparous) on lnVar and autocorrelations; and logistic regression analyses were performed to predict mastitis from lnVar or autocorrelation and parity. From 10 to 30 DIM, lnVar was greater for cows with early mastitis than for cows with no or late mastitis, and autocorrelation tended to be lower for cows with early mastitis than for cows with no mastitis. The lnVar and autocorrelation from 10 to 30 DIM were not predictive of late mastitis. From 10 to 305 DIM, lnVar was greater and autocorrelation was lower for both cows with early and late mastitis than for cows with no mastitis; and both were predictive of having mastitis in the 305-d lactation. Primiparous cows had lower lnVar than multiparous cows. In cows without mastitis, autocorrelation values were positively correlated with lnVar. Results confirm that increased lnVar is associated with clinical mastitis.

The early lactation period in dairy cows is characterized by complex interactions among energy balance (EB), disease, and alterations in metabolic and inflammatory status. The objective of this study was to cluster cows based on EB time profiles in early lactation and investigate the association between EB clusters and inflammatory status, metabolic status, oxidative stress, and disease. Holstein-Friesian dairy cows (n = 153) were selected and monitored for disease treatments during wk 1 to 6 in lactation. Weekly EB was calculated based on energy intake and energy requirements for maintenance and milk yield in wk 1 to 6 in lactation. Weekly plasma samples were analyzed for metabolic variables in wk 1 to 6, and inflammatory and oxidative stress variables in wk 1, 2, and 4 in lactation. Liver activity index (LAI) was computed from plasma albumin, cholesterol, and retino-binding protein concentration. First, cows were clustered based on time profiles of EB, resulting in 4 clusters (SP: stable positive; MN: mild negative; IN: intermediate negative; SN: severe negative). Cows in the SN cluster had higher plasma nonesterified fatty acids and β-hydroxybutyrate concentrations, compared with cows in the SP cluster, with the MN and IN cluster being intermediate. Cows in the SN cluster had a higher milk yield, lower dry matter intake in wk 1, lower insulin concentration compared with cows in the SP cluster, and lower glucose and IGF-1 concentration compared with cows in the SP and MN clusters. Energy balance clusters were not related with plasma haptoglobin, cholesterol, albumin, paraoxonase, and liver activity index (LAI). Second, cows were grouped based on health status [IHP: cows with treatment for inflammatory health problem (endometritis, fever, clinical mastitis, vaginal discharge or retained placenta); OHP: cows with no IHP but treatment for other health problem (milk fever, cystic ovaries, claw, and leg problems, rumen and intestine problems or other diseases); NHP: cows with no treatments, in the first 6 weeks after calving]. Energy balance was not different among health status groups. The IHP cows had lower nonesterified fatty acids and greater insulin concentration in plasma compared with OHP. The IHP cows had lower plasma albumin concentration, lower LAI and higher haptoglobin concentration compared with OHP and NHP. Overall, EB time profiles were associated with the metabolic status of dairy cows in early lactation, but were only limitedly related with markers of inflammation and oxidative stress status. Extending the voluntary waiting period (VWP) for insemination in dairy cows is of interest to reduce the frequency of calving events and inseminate at a moment with fewer fertility problems. Little is known about the calves born from dams with a different VWP followed by a different calving interval (CInt). The objective of the current study was to identify the effect of dam's CInt on body condition, metabolic status, and milk production of their offspring from birth till 100 DIM of the offspring's first lactation. Holstein Friesian dairy cows (n = 154, 41 primiparous, 113 multiparous) were blocked according to parity, milk yield, and somatic cell count (SCC), and randomly assigned to a VWP of 50, 125, or 200 d. Female calves (n = 62) from cows with different CInt were monitored from birth until their first calving event as heifer. Certain dams were not successfully inseminated soon after the planned VWP, resulting in differences between the intended VWP and the actual CInt. Calves were regrouped according to their dam's actual CInt (CInt&#95;1: 324 - 408 d; CInt&#95;2: 409 - 468 d; CInt&#95;3: 469 - 586 d). The dam's CInt did not affect calf birth weight. From birth to weaning, the calves born to dams in CInt&#95;1 (0.34 mmol/L (confidence interval (CI): 0.30, 0.37) had a higher plasma nonesterified fatty acids (NEFA) concentration than CInt&#95;2 (0.28 mmol/L (CI: 0.26, 0.31)) and CInt&#95;3 (0.26 mmol/L (CI: 0.24, 0.29)) calves. Calves born to dams with a shorter CInt (CInt&#95;1) had greater IgG and IgM against keyhole limpet hemocyanin (KLH) than CInt&#95;3 (IgG: 6.05 ± 0.30 vs. 4.64 ± 0.30; IgM: 6.45 ± 0.17 vs. 5.89 ± 0.16, respectively) before weaning. After weaning till calving, CInt&#95;1-calves tended to have greater plasma NEFA concentration than CInt&#95;3-calves. During the first 100 d in milk, a longer CInt of the dams resulted in lower plasma IGF&#95;1 (CInt&#95;2), lower milk lactose (CInt&#95;3) and fat and protein corrected milk (FPCM) (CInt&#95;2) in offspring, compared with shorter CInt of the dams (CInt&#95;1). Collectively, a longer CInt in dams did not affect birth weight of their calves or body weight during the weaning or rearing phase. From birth till weaning, a longer CInt in dams resulted in less IgG against KLH and lower plasma NEFA concentration in plasma of the calves. During the first lactation of their offspring, a longer CInt in dams can result in a lower plasma IGF&#95;1 and FPCM during the first 100 DIM, although effects were not present in all CInt categories., (The Authors. Blood metabolite and hormone concentrations are indicative of metabolic status, but blood sampling and analysis is invasive and time-consuming. Monitoring behavior can be done automatically, and behaviors may also be used as indicators of metabolic status. The aim of this study was to analyze the relationships between metabolic status and feeding behavior, lying behavior, motion index and steps of dairy cows in week 4 postpartum. Behavioral data from 81 Holstein-Friesian cows were collected using computerized feeders and accelerometers, and blood samples were collected for analysis of free-fatty acid (FFA), β-hydroxybutyrate (BHB), glucose, insulin, IGF-1 and growth hormone (GH) concentrations. First, cluster analysis was performed to categorize cows as having poor, average, good or very good metabolic status based on their plasma FFA, BHB, glucose, insulin, IGF-1 and GH concentration. Subsequently, the performance and behavior of cows in clusters with poor, average and good metabolic status were compared using GLM. Cows with a poor or average metabolic status tended to have greater fat-and-protein-corrected milk yield than cows with good metabolic status. Furthermore, cows with a poor metabolic status had a lower energy balance and dry matter intake (DMI) than cows with an average or good metabolic status and had a lower number of meals than cows with good metabolic status. Daily number of visits to the feeder and lying time tended to be positively related with metabolic status. Feeding rate (kg/min), daily meal time (min/day), number of lying bouts per day, steps and motion index were not related with metabolic status. In conclusion, better metabolic status in dairy cows in early lactation was associated with a greater DMI, increased feeding activity and a tendency to more time spent lying, compared with poor metabolic status. These results suggest that compromised metabolic status is reflected in altered cow’s behavior in week 4 of lactation.

The transition period is the most critical period in the lactation cycle of dairy cows. Extended lactations reduce the frequency of transition periods, the number of calves and the related labour for farmers. This study aimed to assess the impact of 2 and 4 months extended lactations on milk yield and net partial cash flow (NPCF) at herd level, and on greenhouse gas (GHG) emissions per unit of fat- and protein-corrected milk (FPCM), using a stochastic simulation model. The model simulated individual lactations for 100 herds of 100 cows with a baseline lactation length (BL), and for 100 herds with lactations extended by 2 or 4 months for all cows (All+2 and All+4), or for heifers only (H+2 and H+4). Baseline lactation length herds produced 887 t (SD: 13) milk/year. The NPCF, based on revenues for milk, surplus calves and culled cows, and costs for feed, artificial insemination, calving management and rearing of youngstock, was k€174 (SD: 4)/BL herd per year. Extended lactations reduced milk yield of the herd by 4.1% for All+2, 6.9% for All+4, 1.1% for H+2 and 2.2% for H+4, and reduced the NPCF per herd per year by k€7 for All+2, k€12 for All+4, k€2 for H+2 and k€4 for H+4 compared with BL herds. Extended lactations increased GHG emissions in CO2-equivalents per t FPCM by 1.0% for All+2, by 1.7% for All+4, by 0.2% for H+2 and by 0.4% for H+4, but this could be compensated by an increase in lifespan of dairy cows. Subsequently, production level and lactation persistency were increased to assess the importance of these aspects for the impact of extended lactations. The increase in production level and lactation persistency increased milk production of BL herds by 30%. Moreover, reductions in milk yield for All+2 and All+4 compared with BL herds were only 0.7% and 1.1% per year, and milk yield in H+2 and H+4 herds was similar to BL herds. The resulting NPCF was equal to BL for All+2 and All+4 and increased by k€1 for H+2 and H+4 due to lower costs for insemination and calving management. Moreover, GHG emissions per t FPCM were equal to BL herds or reduced (0% to −0.3%) when lactations were extended. We concluded that, depending on lactation persistency, extending lactations of dairy cows can have a positive or negative impact on the NPCF and GHG emissions of milk production.

The objective of this study was to evaluate the effect of prepartum dietary energy level on postpartum production and reproduction in Nili Ravi buffaloes (n = 21). The buffaloes were offered low energy (LE: 1.31 Mcal/kg DM NEL (net energy for lactation)), medium energy (ME: 1.42 Mcal/kg DM NEL) or high energy (HE: 1.54 Mcal/kg DM NEL) diet for 63 days prepartum, and received the same lactation diet (LD: 1.22 Mcal/kg DM NEL) during 14 weeks postpartum. The effects of dietary energy level and week were analyzed with Proc GLIMMIX model. Dry matter intake (DMI) was lower in buffaloes fed the LE diet compared with buffaloes fed the ME or HE diet. Calf birth weight (CBW) was higher in buffaloes fed the HE diet compared with buffaloes fed the ME or LE diet. Milk production was similar in buffaloes fed the HE, ME or LE diet within 14 weeks postpartum and throughout the lactation. Milk fat was higher in buffaloes fed the LE diet compared with buffaloes fed the ME or HE diet. Milk protein and lactose yields was high in buffaloes fed the HE diet compared with buffaloes fed the ME or LE diet. Body condition score was high in HE and was affected by diet × week interactions during pre and postpartum period. The concentrations of β-hydroxybutyrate (BHBA) and triglycerides in serum was lowest in buffaloes fed the HE diet compared with the buffaloes fed the ME or LE diet. The buffaloes fed the HE diet had early uterine involution (UI), first estrus, short dry days, and calving interval (CI) compared with buffaloes fed the ME or LE diet. None of buffaloes fed the LE diet exhibited estrus during the first 14 weeks postpartum compared with buffaloes fed the ME or HE diet. In conclusion, prepartum feeding of high energy diet can be helpful in improving the postpartum productive and reproductive performance in Nili Ravi buffaloes.

Abstract In early lactation, dairy cows typically have a negative energy balance which has been related to metabolic disorders, compromised health and fertility, and reduced productive lifespan. Assessment of the energy balance, however, is not easy on the farm. Our aims were to investigate the milk metabolic profiles of dairy cows in early lactation, and to obtain models to estimate energy balance from milk metabolomics data and milk production traits. Milk samples were collected in week 2 and 7 after calving from 31 dairy cows. For each cow, the energy balance was calculated from energy intake, milk production traits and body weight. A total of 52 milk metabolites were detected using LC-QQQ-MS. Data from different lactation weeks was analysed by partial least squares analysis, the top 15 most relevant variables from the metabolomics data related to energy balance were used to develop reduced linear models to estimate energy balance by forward selection regression. Milk fat yield, glycine, choline and carnitine were important variables to estimate energy balance (adjusted R 2: 0.53 to 0.87, depending on the model). The relationship of these milk metabolites with energy balance is proposed to be related to their roles in cell renewal.

Numerous methods and devices are available for implant fixation in anterior cruciate ligament (ACL) reconstruction. Biomechanical data indicate high variability in fixation stability across different devices. This study aims to provide a better insight into measuring the structural characteristics and mechanical behavior of ACL implant fixations. Fourteen human tibial specimens with reconstructed ACLs were subjected to progressively increasing dynamic loading until failure. The motions of the tibia, the proximal and distal graft ends, as well as the testing frame and actuator, were continuously recorded via a motion tracking system. Significantly higher displacements of the machine actuator (1.0 mm at graft slippage onset, and 12.2 mm at ultimate load) were measured compared to the displacements of the proximal (0.8 and 4.3 mm, respectively) and distal graft (0.1 and 3.4 mm, respectively) ends. The displacements measured at different sites showed significant correlations. The provided data suggest significant and systematic inaccuracies in the stiffness and slippage of the fixation when using machine displacement, as commonly reported in the literature. The assessment of the distal graft displacement excludes the artifactual graft elongation, and most accurately reflects the graft slippage onset indicating clinical failure. Considering the high displacement at the ultimate load, the ultimate load could be used as a standardized variable to compare different fixation methods. However, the ultimate load alone is not sufficient to qualitatively describe fixation stability.

Background and Objectives: Wound infections provoked by alterations in microcirculation are major complications in the treatment of trochanteric femur fractures. Surgical fracture fixation on a traction table is the gold standard for treatment, but the effect on tissue microcirculation is unknown. Microcirculation could be impaired by the pull on the soft-tissue or by a release of vasoactive factors. We hypothesized that intraoperative traction impairs soft-tissue microcirculation. Materials and Methods: In 22 patients (14 women, eight men), average age 78 years (range 36–96 ± 14), with trochanteric femur fractures, non-invasive laser-Doppler spectrophotometry was used to assess oxygen saturation, hemoglobin content, and blood flow in the skin and subcutaneous tissue before and after application of traction. Measurements were recorded in nine locations around the greater trochanter at a depth of 2, 8, and 15 mm before and after fracture reduction by traction. Results: No differences were found in any depth with traction compared to without (oxygen saturation: p = 0.751, p = 0.308, and p = 0.955, haemoglobin content: p = 0.651, p = 0.928, and p = 0.926, blood flow: p = 0.829, p = 0.866, and p = 0.411). Conclusion: In this pilot study, the application of traction does not affect skin and subcutaneous microcirculation in the surgery of proximal femur fractures.

Traditionally, a 1-yr calving interval is advised to farmers from an economical point of view, to realize a yearly peak in milk yield. A 1-yr calving interval, however, implies a yearly event of drying-off, calving and start of lactation, which are all associated with an increased risk for diseases and disorders. Deliberately extending the lactation length by extending the voluntary waiting period (VWP) for frst insemination reduces the frequency of these challenging events. This reduction in frequency of calvings can be benefcial for cow health and fertility, but also can be of interest to reduce the number of surplus calves and labor associated with drying off, calving, and disease treatments. Current concerns with respect to an extended lactation are that milk yield is too low in late lactation, which might be associated with an increased risk of fattening of cows in late lactation, and compromised economic returns at herd level. In addition, limited knowledge is available with respect to consequences for cow performance in the subsequent lactation and for calves born to cows with an extended lactation. Moreover, response of dairy cows to an extended VWP depends on individual cow characteristics like parity, milk yield level or body condition. A customized strategy based on individual cow characteristics can be a future approach to select high-producing cows with persistent lactation curves for an extended lactation to limit the risk for fattening and milk yield reduction at the end of the lactation while beneftting from a reduction in challenging events around calving.
