Peanut seeds are harvested at different development stages (early and late) due to their uneven maturation. At the time of harvest, approximately 30% of the seeds are still immature, meaning they are not completely filled with compounds (e.g., oil and minerals) and exhibit reduced vigor. Hypothetically, these compounds can be detected as a "chemical fingerprinting" to classify seed maturation stages. Here, we investigated whether non-destructive techniques such as benchtop nuclear magnetic resonance (NMR), laser-induced breakdown spectroscopy (LIBS), and energy-dispersive X-ray fluorescence (ED-XRF) can identify chemical patterns unique to mature seeds with superior vigor. Field-grown seeds were classified into early (R5 and R6) and late (R7, R8, and R9) stages. Seed weight, germination, vigor, H2O2, and MDA (oxidative stress) were analyzed. Oil, potassium (K), and calcium (Ca) were measured digitally using spectroscopy techniques. We found that: (i) oxidative stress and K levels were higher in seeds from the early stages; (ii) seed oil and Ca were proportional to high-vigor seedlings and successful plant establishment in the field; and (iii) the seed chemical composition could be identified autonomously with 87% to 100% accuracy. Cynomorium songaricum is a commonly used medicinal and culinary plant renowned for its effects in tonifying kidney yang, as well as nourishing essence and blood. Consequently, it has garnered considerable attention within the fields of pharmacology and chemistry. However, given the distinct geographical distribution of C. songaricum resources, establishing an accurate analytical method to differentiate its origin becomes indispensable in ensuring its quality and efficacy. In this study, high-performance liquid chromatography (HPLC) was utilized to obtain comprehensive information on the chemical composition of C. songaricum samples from various regions and establish HPLC fingerprints. Through chemometrics analysis, essential chemical markers for classifying them were successfully identified. Additionally, the Kruskal–Wallis rank sum test was employed to evaluate differences in the content of intrinsic constituents of C. songaricum from different origins. The results demonstrated that C. songaricum samples from Neimenggu, Gansu, and Xinjiang could be accurately classified, highlighting the significance of chemical markers in assessing the quality of C. songaricum. Piper aduncum L. is widely distributed in tropical regions and the ethnobotanical uses of this species encompass medicinal applications for the treatment of respiratory, antimicrobial, and gynecological diseases. Chemical studies reveal a diverse array of secondary metabolites, including terpenes, flavonoids, and prenylated compounds. Extracts from P. aduncum have shown antibacterial, antifungal, and larvicidal activities. Our study explores the activity of extracts and partitions against Mycobacterium tuberculosis H37Rv, as well as the chemical diversity of the bioactive partition. This marks the first investigation of the bioactive partition of P. aduncum from agroecological cultivation. The ethyl acetate partition from the ethanolic leaf extract (PAEPL) was found to be the most active. PAEPL was subjected to column chromatography using Sephadex LH-20 and the obtained fractions were analyzed using UHPLC-HRMS/MS. The MS/MS data from the fractions were submitted to the online GNPS platform for the generation of the molecular network, which displayed 1714 nodes and 167 clusters. Compounds were identified via manual inspection and different libraries, allowing the annotation of 83 compounds, including flavonoids, benzoic acid derivatives, glycosides, free fatty acids, and glycerol-esterified fatty acids. Introduction: Farfarae Flos is widely used as a traditional herbal medicine. Currently, its size has been the primary grading criterion used in market circulation. Whether this empirical criterion can accurately reflect the quality of the medicinal material has not been systematically studied., Objective: This study aimed to evaluate the quality of Farfarae Flos from different regions based on their grades., Methods: Ultra-high performance liquid chromatography coupled with electrospray ionization quadrupole time-of-flight mass spectrometry (UPLC-ESI-Q/TOF-MS/MS) were applied to study the chemical constituents of Farfarae Flos; high performance liquid chromatography (HPLC) was used to quantify the content of Farfarae Flos samples. Meanwhile, fingerprint analysis and chemometric methods, including principal component analysis (PCA) and analysis of variance (ANOVA), were used to evaluate the quality differences among 33 batches of Farfarae Flos samples of different grades., Results: A total of 95 individual components were identified in Farfarae Flos. Fingerprint analysis revealed 23 common peaks, with fingerprint similarity among the 33 batches ranging from 0.838 to 0.995. PCA divided the 33 batches of Farfarae Flos into three categories based on their grades. ANOVA indicated significant differences in five of the 14 main active components across different grades of Farfarae Flos, with two components showing extremely significant differences. HPLC content determination showed that the content of 11 main active components was positively correlated with the grades of Farfarae Flos., Conclusion: This method is straightforward, efficient, and reliable, offering a valuable reference for establishing quality grading standards and ensuring the quality control of Farfarae Flos., (© 2025 John Wiley & Sons Ltd.)

Conventional testing methods provide sufficient information to evaluate human or ecological risk. However, contaminant concentrations patterns alone provide only limited resolution of important liability issues, such as when and where did contaminant releases originate. Over the past few decades, scientists explored the isotope applications to better identify, delineate, and manage contaminants in the environment. Advanced chemical fingerprinting and isotope technologies revealed important linkages between isotope ratios and contaminant origins (e.g., chemical feedstock and manufacturing process). Studies of environmental weathering distinguished abiotic and biotic changes in the chemical composition and isotope patterns. The combined application of chemical and isotopic fingerprints offers powerful complementary lines of evidence for delineating contaminants, assessing risk, and identifying historical sources. This manuscript provides an integrated forensic approach that systematically links conventional environmental investigation data with specialized chemical fingerprinting and carbon/chlorine isotope methods for identifying the sources of groundwater impacts especially when multiple potential point and non-point sources exist. This paper focusses on chlorinated solvents. Specifically, it features the synoptic use of chemical concentration patterns and compound-specific isotope analysis (CSIA) as effective tools for confirming organic contaminant sources, characterizing environmental weathering, and answering a growing list of site-specific questions. Unlike conventional isotope methods, which can be both time-consuming and expensive, this paper presents an optimized analytical method for chlorine CSIA using gas chromatography-quadrupole mass spectrometry (GC-qMS). Chlorine isotopic composition for multiple analytes (e.g., tetrachloroethylene [PCE], trichloroethylene [TCE], dichloroethylene [DCE], and vinyl chloride [VC]) can be determined in one acquisition thus reducing analysis time and cost. Precise CSIA isotope values were achieved for chloroethylene concentrations between approximately 5 micrograms per liter (ug/l) and 100 ug/l for carbon and between approximately 30 ug/l to 1,000 ug/l for chlorine. The gradual improvement in CSIA methods better addresses the wide concentration range encountered in typical samples collected from groundwater aquifers with significant chlorinated solvent impacts. There is a growing interest in standardizing the biocompatible, cost‐effective, and eco‐friendly manufacturing techniques for metallic nanostructures due to their widespread applications in the industrial and medical sectors. In recent decades, green synthesis has been proven as the most suitable technique for synthesizing metal nanoparticles. The present research study investigates the use of Cassia angustifolia (senna) leaves and Pistacia vera (Pistachio) nuts to prepare crude aqueous extracts, ethanolic extracts, and biogenic silver nanoparticles (AgNPs). The prepared aqueous extracts were used as reducing, stabilizing, and capping agents for the production of silver nanoparticles. These AgNPs were characterized by scanning electron microscopy (SEM), Fourier‐transform infrared spectroscopy (FTIR), and ultraviolet–visible (UV–Vis) spectroscopy. The outcomes validated the formation of stable AgNPs with bioactive functional components. In vitro antibacterial, anticancer, anti‐inflammatory, and antioxidant potentials were assessed by Kirby–Bauer disk diffusion test, MIC test, MBC test, MTT assay, BSA denaturation inhibition assay, and DPPH antioxidant assay, respectively. Results confirmed that the tested plant extract possesses a variety of bioactive compounds with various biological activities and is therapeutically effective. These findings verified that C. angustifolia and P. vera are promising bioresources for the synthesis of therapeutic extracts and nanostructures with commendable therapeutic potency. The World Health Organization has emphasized the importance of consuming small fruits for the prevention of chronic health problems, including diabetes, cardiovascular diseases, cancer, and obesity, which are named chronic non-communicable diseases (NCDs). Azara serrata Ruiz & Pav., commonly called “aroma de Castilla”, is a shrub endemic to Chile from the Salicaceae family that produces an underutilized blue-grey berry that grows wild in southern Chile. The species is widely used as a medicinal plant by the Andean communities of southern Chile. In this work, a high-resolution mass spectrometric analysis of the methanolic extract revealed several phenolic compounds for the first time in the edible berry of this endemic species. Furthermore, several glycosylated anthocyanins were detected and quantified using UHPLC coupled with UV/Vis detection and trapped ion mobility mass spectrometry (UHPLC-DAD-TIMS-TOF) for the anthocyanin-rich extract, which was prepared using an optimized anthocyanin extraction protocol. The extract proved to be active in the inhibition of several enzymes linked to NCDs, such as acetylcholinesterase, tyrosinase, amylase, lipase, and glucosidase (IC50 = 3.92 ± 0.23, 12.24 ± 0.03, 11.12 ± 0.10, 32.43 ± 0.0, and 371.6 ± 0.0 μg/mL, respectively). Furthermore, the extract concentrated in anthocyanins showed good antioxidant activity evidenced by the bleaching of the radicals DPPH and ABTS, ferric-reducing antioxidant power (FRAP), and oxygen radical absorbance capacity (ORAC). The results show that these neglected endemic small berries can be a source of healthy phytochemicals. These Chilean berries can be used as functional food and their extracts are candidates for use as functional ingredients in naturally healthy products.

IntroductionImpact of processing on product characteristics, sustainability, traceability, authenticity, and public health along the food chain becomes more and more important not only to the producer but also to the customer and the trust of a consumer toward a brand. In recent years, the number of juices and smoothies containing so called super foods or fruits, which have been “gently pasteurized,” has increased significantly. However, the term “gentle pasteurization” related to the application of emerging preservation technologies such as pulsed electric fields (PEF), high pressure processing (HPP) or ohmic heating (OH) is not clearly defined.MethodsTherefore, the presented study investigated the influence of PEF, HPP, OH, and thermal treatment on quality characteristics and microbial safety of sea buckthorn syrup. Syrups from two different varieties were investigated under the following conditions HPP (600 MPa 4–8 min), OH (83°C and 90°C), PEF (29.5 kV/cm, 6 μs, 100 Hz), and thermal (88°C, hot filling). Analyses to test the influence on quality parameters like ascorbic acid (AA), flavonoids, carotenoids, tocopherols, antioxidant activity; metabolomical/chemical profiling (fingerprinting) via U-HPLC-HRMS/MS (here especially flavonoids and fatty acids); sensory evaluation, as well as microbial stability including storage, were conducted.Results and discussionIndependent from the treatment, the samples were stable over 8 weeks of storage at 4°C. The influence on the nutrient content [Ascorbic acid (AA), total antioxidant activity (TAA), total phenolic compounds (TPC), tocopherols (Vit E)] was similar for all tested technologies. Employing statistical evaluation Principal Component Analysis (PCA) a clear clustering based on the processing technologies was observed. Flavonoids as well as fatty acids were significantly impacted by the type of used preservation technology. This was obvious during the storage time of PEF and HPP syrups, where enzyme activity was still active. The color as well as taste of the syrups were found to be more fresh-like for the HPP treated samples.

A novel fingerprinting platform for multiplex detection of flavor molecules in Baijiu was developed by using a surface-enhanced Raman scattering (SERS) nanosensor array in combination with machine learning. The SERS sensors were constructed by core–shell Fe3O4@Ag nanoparticles modified with molecules carrying end-groups of hydroxyl, pyridyl, methyl, and amino, respectively, which interacted with flavors and led to changes in the sensors' spectra. All the Raman spectra acquired from the nanosensor array contacting with the sample were concatenated into a single SERS super-spectrum, representing the flavor fingerprint which was recognized through machine learning. Principal component analysis, support vector machine, and partial least squares were utilized to build classification and quantitation models for predictive analyses. The SERS nanosensor array was successfully used for fingerprinting ten typical flavors in Baijiu including four esters, three alcohols, and three acids, with an accuracy of 100%, linear detection ranges over two orders of magnitude, and limits of detection ranging from 3.45 × 10−3 mg/L of phenylethyl acetate to 1.21 × 10−2 mg/L of ethyl hexanoate. It was also demonstrated that satisfactory accuracies (recoveries) ranging from 96.2 to 104% and relative standard deviations ranging from 0.65 to 2.78% were obtained for the simultaneous quantification of 3-methylbutyl acetate and phenylethyl acetate in eighteen Baijiu samples of three flavor types including sauce flavor, strong flavor, and light flavor. Compared with the existing detection techniques, this chemical fingerprinting platform is easy to use, highly sensitive, and can perform multiplex detection, which has great potential for practical applications. Optimization of experimental conditions in chromatography is recognized as a wear-and-tear process involving hundreds of experiments in order to maximize the number of separated compounds for each sample. The critical thinking skills and practices entailed cannot be achieved with students in a conventional lecture class. This report provides a Microsoft Excel-based spreadsheet allowing the fast simulation of fingerprinting results and the number of separated peaks for a given sample analyzed using gas chromatography-mass spectrometry. The applications were demonstrated for the simulation of extracted ion chromatograms in order to find a peak for the target compound in a model kerosene sample containing 254 volatile compounds with the optimization of the separation of fatty acid methyl esters. Using the simulation applications, students are able to perform experimental design in a lecture class. Test score results reveal the selectivity of the approach toward students with laboratory practical experience and experimental skills. This approach is expected to be useful to all higher education students in chemistry.

We present a combined experimental and theoretical investigation of the radiationless decay spectrum of an O 1s double core hole in liquid water. Our experiments were carried out using liquid-jet electron spectroscopy from cylindrical microjets of normal and deuterated water. The signal of the double-core-hole spectral fingerprints (hypersatellites) of liquid water is clearly identified, with an intensity ratio to Auger decay of singly charged O 1s of 0.0014(5). We observe a significant isotope effect between liquid H2O and D2O. For theoretical modeling, the Auger electron spectrum of the central water molecule in a water pentamer was calculated using an electronic-structure toolkit combined with molecular-dynamics simulations to capture the influence of molecular rearrangement within the ultrashort lifetime of the double core hole. We obtained the static and dynamic Auger spectra for H2O, (H2O)5, D2O, and (D2O)5, instantaneous Auger spectra at selected times after core-level ionization, and the symmetrized oxygen-hydrogen distance as a function of time after double core ionization for all four prototypical systems. Processing of Chinese herbal medicines (CHMs) is a traditional pharmaceutical technology in Chinese medicine. Traditionally, proper processing of CHMs is necessary to meet the specific clinical requirements of different syndromes. Processing with black bean juice is considered one of the most important techniques in traditional Chinese pharmaceutical technology. Despite the long-standing practice of processing Polygonatum cyrtonema Hua (PCH), there is little research on the changes in chemical constituents and bioactivity before and after processing. This study investigated the influence of black bean juice processing on the chemical composition and bioactivity of PCH. The results revealed significant changes in both composition and contents during processing. Saccharide and saponin content significantly increased after processing. Moreover, the processed samples exhibited considerably stronger DPPH and ABTS radical scavenging capacity, as well as FRAP-reducing capacity, compared to the raw samples. The IC50 values for DPPH were 1.0 ± 0.12 mg/mL and 0.65 ± 0.10 mg/mL for the raw and processed samples, respectively. For ABTS, the IC50 values were 0.65 ± 0.07 mg/mL and 0.25 ± 0.04 mg/mL, respectively. Additionally, the processed sample demonstrated significantly higher inhibitory activity against α-glucosidase and α-amylase (IC50 = 1.29 ± 0.12 mg/mL and 0.48 ± 0.04 mg/mL) compared to the raw sample (IC50 = 5.58 ± 0.22 mg/mL and 0.80 ± 0.09 mg/mL). These findings underscore the significance of black bean processing in enhancing the properties of PCH and lay the foundation for its further development as a functional food. Angelicae Dahuricae Radix (ADR) holds a prominent place in traditional medicine for its remarkable antioxidative, anti-allergic, and antiproliferative capabilities. Recognized within the Korean Pharmacopoeia (KP 12th), Angelica dahurica (Hoffm.) Benth. and Hook.f. ex Franch. and Sav. (AD) and Angelica dahurica var. formosana (H. Boissieu) Yen (ADF) serve as the botanical origins for ADR. Differentiating these two varieties is crucial for the formulation and quality control of botanical drugs, as they are categorized under the same medicinal label. This research utilized two-dimensional high-performance thin-layer chromatography (2D-HPTLC) to effectively distinguish AD from ADF. Additionally, a quantitative analysis reveals significant differences in the concentrations of key active constituents such as oxypeucedanin, imperatorin, and isoimperatorin, with AD showing higher total coumarin levels. We further enhanced our investigative depth by incorporating a DPPH bioautography, which confirmed known antioxidant coumarins and unearthed previously undetected antioxidant profiles, including byakangelicin, byakangelicol, falcarindiol in both AD and ADF, and notably, 2-linoleoyl glycerol detected only in AD as an antioxidant spot. This comprehensive approach affords a valuable tool set for botanical drug development, emphasizing the critical need for accurate source plant identification and differentiation in ensuring the efficacy and safety of herbal medicine products.

The analytical methods used for herbal analysis are need to be economic, fast and also produce minimum quantities of hazardous chemical waste. Presently analytical community put interest in the research area of non‐hazardous and eco‐friendly practices to develop various green chromatographic methods for routine quality analysis. High cost of phytochemical analysis and uses of hazardous chemicals with high‐end sophisticated instrument, the attempt made to develop a simple analytical method for multiple samples with short time and less uses of solvents. A HPTLC method was developed for simultaneous determination of biological important constituents like β‐sitosterol, taraxerol, clitorienolactone B and β‐sitosterol glycoside from Clitoria ternatea Linn. The proposed method was validated and satisfied the ICH guidelines to demonstrate that the method is adaptable for its intended purpose. The method is simple, sensitive and economic it therefore embraces potential for detection, monitoring, and simultaneous quantification of the four bioactive compounds for C. ternatea and could also be apply to other species. Poisonous plants are widely distributed and may have risk of phytotoxicity upon mixing with medicinal plants. Several species of Cestrum genus are poisonous and linked with many serious health issues. In the present study, cross-mixing of a toxic plant, Cestrum diurnum with morphologically resembling medicinal plant, Adhatoda vasica was studied using chemical fingerprinting approach. LC-ESI-MS/MS tool was used to develop the chemical fingerprints of three toxic species of Cestrum, including, C. diurnum, C. nocturnum and C. parqui. Total forty-three compounds were identified using high-resolution LC-ESI-MS/MS data comparison. Chemometric analyses were done to compare the distribution of identified compounds present in these Cestrum species. One of the identified compounds, nornicotine (a toxic compound) was also quantified using LC-IT-MS/MS. Adulteration study was conducted by mixing toxic C. diurnum in A. vasica with various ratios (w/w) and five differentiable compounds were identified to detect the adulteration. The method was able to detect up to the limit of 5% mixing of toxic C. diurnum. Moreover, cytotoxicity of the methanolic extracts of these three species were also studied on normal human PBMC (peripheral blood mononuclear cells) and all found to be toxic, while the C. nocturnum showed the highest level of toxicity with the IC50 12.5 μg/mL.

The objective of this investigation was to assess the cytotoxic, antioxidant, and enzyme inhibitory properties of the aerial portions of Nitraria schoberi (family Nitrariaceae). Extracts were prepared by sequential maceration in hexane, dichloromethane (DCM), ethyl acetate and methanol (MeOH) and water extract was prepared by infusion. DCM and MeOH extracts accumulated the highest amount of metabolites where the former was characterized by the presence of octadecyl ferulate (16.96 mg/100 mg), methoxyluteolin (8.48 mg/100 mg) and isorhamnetin (5.35 mg/100 mg) and the alkaloids perlolyrine (5.03 mg/100 mg), (+)-5a-hydroxylemannine (2.43 mg/100 mg) and matrine (2.40 mg/100 mg). Results of antioxidant activity showed that the MeOH extract exerted the highest anti-2,2-diphenyl-1-picrylhydrazyl (DPPH) (28.13 mg Trolox equivalent (TE)/g) and Fe+++ (54.38 mg TE/g) reducing capacity. All extracts exerted significant acetylcholinesterase (3.71–4.37 mg galantamine equivalent (GALAE)/g) and tyrosinase (86.40–109.63 mg kojic acid equivalent (KAE)/g) inhibition activity. The best α-amylase and α-glucosidase inhibition was exhibited by the DCM (0.81 mmol ACAE/g) and MeOH (1.34 mmol ACAE/g) extracts respectively. The highest cytotoxic effect was exerted against the breast adenocarcinoma (MDA-MB-231) (IC 50 : 26.71 μg/mL) and cervix adenocarcinoma (HELA) (IC 50 : 29.10 μg/mL) cell lines by the MeOH and DCM extracts respectively. In addition, we performed molecular docking analysis to understand the interaction between chemical components and tested enzymes and cellular targets for cancer. Chrysoeriol-3-O-rutinoside bound strongly to AChE. Furthermore, a good interaction between vascular endothelial growth factor receptor 2 (VEGFR2) and isovitexin-2″-O-arabinoside was observed. We employed network pharmacology to understand the role of N. schoberi's compounds in neurological disorder like Alzheimer and multiple sclerosis. Therefore, the results of this study indicate that N. schoberi may serve as a viable starting point for the future creation of phytopharmaceuticals that target particular diseases associated with oxidative stress, such as diabetes and certain neurological disorders. Herbal medicines for managing oral diseases are considered an effective alternative to synthetic compounds due to their lower side effect. Acacia catechu, Piper nigrum, Piper longum, and Piper cubeba are vital components of Dasanakanthi Churnam used to control and prevent oral inflammations in dentistry. Tannic acid and Piperine are major markers for the chemical fingerprinting of Dasanakanthi Churnam. They serve as quality indicating parameters for Dasanakanthi Churnam because of their pharmacological action. Development and validation of a stability-indicating assay method using RP-HPLC become an integral part of simultaneous estimation of Tannic acid and Piperine in Dasanakanthi Churnam. Stability indicating the method is established by exposing markers to various stress conditions such as alkaline hydrolysis, photolytic, oxidative and thermal conditions. Stress testing of the markers can help identify the likely degradation products and validate the stability indicating the power of the analytical procedures used. Tannic acid and Piperine were effectively separated at 5.2 and 22.5 minutes respectively using a PDA detector without interference from any degradant and impurities. Osmanthus fragrans (Thunb.) Lour. flowers are important natural resource of phenolic compounds, which have antioxidant activities and have been consumed as functional food and dietary supplements for thousand years. Ultra-performance liquid chromatography coupled with electrochemical detection (UPLC-ECD) was developed to simultaneously analyze the eight phenolic compounds in Osmanthus fragrans flowers collected from different regions across China. The UPLC-ECD fingerprint was established for identification and comprehensive quality control.
