Phosphorescent probes often show sensitive response toward analytes at a specific wavelength. However, oxygen quenching usually occurs at the same wavelength and thus hinders the accurate detection of analytes. In this study, we have developed dual‐emissive iridium(III) complexes that exhibit phosphorescence responses to copper(II) ions at a wavelength distinct from that where oxygen quenching occurs. The complexes displayed colorimetric phosphorescence response in aqueous solutions under different copper(II) and oxygen conditions. In cellular imaging, variation in oxygen concentration over a large range from 5 % to 80 % can modulate the intensity and lifetime of green phosphorescence without affecting the response of red phosphorescence toward intracellular copper(II) ions. Parkinson's disease remains one of the leading neurodegenerative diseases in developed countries. Despite well-defined symptomology and pathology, the complexity of Parkinson's disease prevents a full understanding of its etiological mechanism. Mechanistically, α-synuclein misfolding and aggregation appear to be central for disease progression, but mitochondrial dysfunction, dysfunctional protein clearance and ubiquitin/proteasome systems, and neuroinflammation have also been associated with Parkinson's disease. Particularly, neuroinflammation, which was initially thought to be a side effect of Parkinson's disease pathogenesis, has now been recognized as driver of Parkinson's disease exacerbation. Next-generation sequencing has been used to identify a plethora of long noncoding RNAs (lncRNA) with important transcriptional regulatory functions. Moreover, a myriad of lncRNAs are known to be regulators of inflammatory signaling and neurodegenerative diseases, including IL-1β secretion and Parkinson's disease. Here, LncZFAS1 was identified as a regulator of inflammasome activation, and pyroptosis in human neuroblast SH-SY5Y cells following MPP+ treatment, a common in vitro Parkinson's disease cell model. Mechanistically, TXNIP ubiquitination through MIB1 E3 ubiquitin ligase regulates NLRP3 inflammasome activation in neuroblasts. In contrast, MPP+ activates the NLPR3 inflammasome through miR590-3p upregulation and direct interference with MIB1-dependent TXNIP ubiquitination. LncZFAS overexpression inhibits this entire pathway through direct interference with miR590-3p, exposing a novel research idea regarding the mechanism of Parkinson's disease. [ABSTRACT FROM AUTHOR]

Aspergillus flavus (A. flavus) is a frequent harmful fungal pathogen. It can infect traditional Chinese medicine materials and release aflatoxin, to cause both economic and human health effects. By comparing the inhibitory potential of Litsea cubeba oil and its active components to A. flavus CGMCC 3.4408, citral was confirmed to be the main component that inhibits the growth of A. flavus CGMCC 3.4408, and the EC50 was 163.65 mg L−1. The inhibitory effect of citral on A. flavus CGMCC 3.4408 was studied for colony growth rate, mycelium biomass, aflatoxin production, and microstructure. Citral slowed down the growth rate of colonies and reduced mycelium biomass and toxin production. Moreover, citral altered the morphology of fungal spores and mycelium. In addition, citral also has the inhibitory effects on the isolates of A. flavus from moldy traditional Chinese medicinal materials. A number of studies have demonstrated the crucial functions of GINS2 within the GINS complex in various types of cancer. However, the molecular mechanisms and prognostic value of GINS2 in breast cancer remain unknown. The present study used; BC-GenExMiner, COSMIC, UCSC Xena, The Human Protein Atlas, GEPIA, cBioPortal, GeneMANIA, TIMER and Oncomine, in order to investigate gene expression, co-expression, clinical parameters and mutations in GINS2 in patients with breast cancer. Furthermore, the present study assessed the prognostic value of GINS2 in patients with breast cancer via the Kaplan-Meier plotter database. The results of the present study demonstrated that the mRNA levels of GINS2 were significantly higher in breast cancer tissue compared with normal tissue. In addition, high mRNA expression levels of GINS2 were associated with high Scarff-Bloom-Richardson status grades, a basal-like status and age (≤51 years); however, it was not associated with lymph node metastasis. The survival analysis revealed that increased GINS2 mRNA levels were associated with a worse prognosis for relapse-free survival in all patients with breast cancer, particularly in those with estrogen receptor-positive and progesterone receptor-positive subtypes. In addition, a positive association between the GINS2, CENPM and MCM4 genes was confirmed. The results of the present study suggest that GINS2 could be used as a potential prognostic biomarker for breast cancer. The innate immune response is the first line defense against viral infections. Novel genes involved in this system are continuing to emerge. SLC15A3, a proton-coupled histidine and di-tripeptide transporter that was previously found in lysosomes, has been reported to inhibit chikungunya viral replication in host cells. In this study, we found that SLC15A3 was significantly induced by DNA virus herpes simplex virus-1(HSV-1) in monocytes from human peripheral blood mononuclear cells. Aside from monocytes, it can also be induced by HSV-1 in 293T, HeLa cells, and HaCaT cells. Overexpression of SLC15A3 in 293T cells inhibits HSV-1 replication and enhances type I and type III interferon (IFN) responses, while silencing SLC15A3 leads to enhanced HSV-1 replication with reduced IFN production. Moreover, we found that SLC15A3 interacted with MAVS and STING and potentiated MAVS- and STING-mediated IFN production. These results demonstrate that SLC15A3 participates in anti-HSV-1 innate immune responses by regulating MAVS- and STING-mediated signaling pathways. [ABSTRACT FROM AUTHOR]

The Pr α-sialon powders prepared by self-propagating high-temperature synthesis (SHS), consisting of 55 wt% Pr α-sialon and 45 wt% of β-sialon (abbreviated as α′ and β′), were hot-pressed at 1800°C for 1 h. The results showed that Pr α′ phase would transfer to β′ with the appearance of JEM phase (Pr(Si6-zAIz)(N10-zOz)) after sintering., thus resulting in the increase of β′ phase to 86 wt%. The addition of Y2O3 into SHS-ed Pr α′ powders as the starting materials restrains the transformation of α′ to β′ and prevents the formation of JEM phase as well. The nucleation mechanism of Pr α′ grain during hot-pressing was investigated in terms of transmission electron microscope and energy-dispersive spectrometer analysis. Two nucleation modes of Pr α′ grains were found, i.e., nucleating on the undissolved Pr α′ grains and on the nuclei of (Pr, Y) α′ grains precipitated from liquid phase. [ABSTRACT FROM AUTHOR]
