Abstract

Umami peptides from the enzymatic hydrolysate of Stropharia rugosoannulata were separated by ultrafiltration (UF) and gel filtration chromatography (GFC) combined with sensory evaluation and electronic tongue analysis. Fraction F3, with the strongest umami intensity, were identified by reversed-phase high performance liquid chromatography-tandem mass spectrometry (RP-HPLC-MS/MS). The identified peptides were synthesized and evaluated for their taste characteristics. The results showed that eight umami peptides were identified in fraction F3, namely ELWR, RLVDR, KPDNR, AHLRF, LDWDR, LAEWR, DDWWR and EGHKGW, and all of them possessed umami characteristics and umami-enhancing effects with thresholds ranging from 0.30 to 1.33 mmol/L and from 0.53 to 2.43 mmol/L, respectively. All peptides except LDWDR showed different degrees of umami improvement in monosodium glutamate (MSG) solution, ranging from 0.18% to 61.12%. ELWR, RLVDR, KPDNR, AHLRF and EGHKGW showed good umami-enhancing effects at low concentrations. The peak concentration of RLVDR and KPDNR for synergistic umami-enhancing effect was 5 mg/mL. LDWDR, LAEWR, DDWWR and EGHKGW reached the umami peak at 4 mg/mL. The amino acid composition and the spatial structure of umami peptides could affect their taste properties. The results of this study provide a theoretical basis for the development and utilization of umami peptides from S. rugosoannulata.

Abstract

Given the existing COVID-19 pandemic worldwide, it is critical to systematically study the interactions between hosts and coronaviruses including SARS-Cov, MERS-Cov, and SARS-CoV-2 (cause of COVID-19). We first created four host-pathogen interaction (HPI)-Outcome postulates, and generated a HPI-Outcome model as the basis for understanding host-coronavirus interactions (HCI) and their relations with the disease outcomes. We hypothesized that ontology can be used as an integrative platform to classify and analyze HCI and disease outcomes. Accordingly, we annotated and categorized different coronaviruses, hosts, and phenotypes using ontologies and identified their relations. Various COVID-19 phenotypes are hypothesized to be caused by the backend HCI mechanisms. To further identify the causal HCI-outcome relations, we collected 35 experimentally-verified HCI protein-protein interactions (PPIs), and applied literature mining to identify additional host PPIs in response to coronavirus infections. The results were formulated in a logical ontology representation for integrative HCI-outcome understanding. Using known PPIs as baits, we also developed and applied a domain-inferred prediction method to predict new PPIs and identified their pathological targets on multiple organs. Overall, our proposed ontology-based integrative framework combined with computational predictions can be used to support fundamental understanding of the intricate interactions between human patients and coronaviruses (including SARS-CoV-2) and their association with various disease outcomes., Comment: 32 pages, 1 table, 6 figures

Abstract

Critical current density (Jc) is one of the major limiting factors for high field applications of iron-based superconductors. Here, we report that Mn-ions are successfully incorporated into nontoxic superconducting (Li,Fe)OHFeSe films. Remarkably, the Jc is significantly enhanced from 0.03 to 0.32 MA/cm^2 under 33 T, and the vortex pinning force density monotonically increases up to 106 GN/m^3, which is the highest record so far among all iron-based superconductors. Our results demonstrate that Mn incorporation is an effective method to optimize the performance of (Li,Fe)OHFeSe films, offering a promising candidate for high-field applications., Comment: 6 pages, 5 figures

Abstract

Recently, MnBi2Te4 has been discovered as the first intrinsic antiferromagnetic topological insulator (AFM TI), and will become a promising material to discover exotic topological quantum phenomena. In this work, we have realized the successful synthesis of high-quality MnBi2Te4 single crystals by solid-state reactions. The as-grown MnBi2Te4 single crystal exhibits a van der Waals layered structure, which is composed of septuple Te-Bi-Te-Mn-Te-Bi-Te sequences as determined by powder X-ray diffraction (PXRD) and high-resolution high-angle annular dark field scanning transmission electron microscopy (HAADF-STEM). The magnetic order below 25 K as a consequence of A-type antiferromagnetic interaction between Mn layers in the MnBi2Te4 crystal suggests the unique interplay between antiferromagnetism and topological quantum states. The transport measurements of MnBi2Te4 single crystals further confirm its magnetic transition. Moreover, the unstable surface of MnBi2Te4, which is found to be easily oxidized in air, deserves attention for onging research on few-layer samples. Abstract

Nickel-63 radioactive sources used in micro nuclear batteries require high surface emissivity, thin coating and small substrate. The method for the preparation of nickel-63 radioactive sources by direct and constant current electrodeposition was established by using nickel-63 solution with small volume, high specific activity and low Ni2+ concentration as electrodeposition liquid.Using this method, under the conditions of copper substrate, Ni2+ concentration with 1 g/L, electrodeposition pH with 3.5-4.5 and sulphamic acid concentration with 0.1 g/L, the electrodeposition efficiency can reach to 95.8% after 1 h . The coating is compact and bright with the thickness from 0.3 μm to 1.6 μm. The surface emissivity of the prepared nickel-63 radio-source film is up to 2.40×107 s-1·(2πSr)-1 while the activity is up to 1.89×109 Bq, which can meet the application requirements of micro nuclear batteries. At the same time, by recovering the residual electrodeposition solution and adjusting its composition, the reuse of electrodeposition liquid is realized, reducing the waste of nickel-63 raw material.

Abstract

Escherichia coli (E. coli), a major foodborne pathogen, poses significant risks to public health by causing gastrointestinal diseases. Among its virulence factors, Yersiniabactin (Ybt), a siderophore, plays a crucial role in iron acquisition and enhancing intestinal colonization. Despite previous studies highlighting E. coli -Ybt's involvement in inflammation, its exact mechanisms remain unclear. This study investigates how Ybt contributes to intestinal inflammation through ferroptosis, using both in vitro and in vivo models. Our findings demonstrate that Ybt promotes oxidative stress, lipid peroxidation, inflammation, and iron accumulation in intestinal epithelial cells, leading to ferroptosis. Mechanistically, Ybt suppresses the Keap1/Nrf2 pathway, amplifying reactive oxygen species (ROS) and activating the TNF/NF-κB pathway, which drives inflammation. Moreover, Ybt induces lipid peroxidation via the arachidonic acid pathway, producing 6-trans-leukotriene B4 (6-transLTB4), which exacerbates inflammation and ferroptosis. Exogenous 6-transLTB4 further intensifies this cascade. Additionally, Ybt disrupts iron efflux by suppressing FPN1 expression, causing excessive intracellular iron accumulation. Using tree shrews as an in vivo model, we confirm that Ybt-induced ferroptosis significantly aggravates intestinal inflammation. These findings underscore the pathogenic role of Ybt in E. coli -induced intestinal injury and highlight ferroptosis as a novel mechanism contributing to gut health disruption. This study provides new insights into the molecular pathways of E. coli infection, with implications for therapeutic strategies targeting ferroptosis in intestinal diseases. [ABSTRACT FROM AUTHOR]
