Abstract Background Human epidermal growth factor receptor 2-positive (HER2+) breast cancer (BC), which accounts for approximately one-fifth of all BCs, are highly invasive with a high rate of recurrence and a poor prognosis. Several studies have shown that growth factor receptor-bound protein 7 (GRB7) might be a potential therapeutic target for tumor diagnosis and prognosis. Nevertheless, the role of GRB7 in HER2+ BC and its underlying mechanisms have not been fully elucidated. The aim of this study was to investigate the biological function and regulatory mechanism of GRB7 in HER2+ BC. Methods Bioinformatics analysis was performed using the TCGA, GEO and CancerSEA databases to evaluate the clinical significance of GRB7. RT quantitative PCR, western blot and immunofluorescence were conducted to assess the expression of GRB7 in BC cell lines and tissues. MTT, EdU, colony formation, wound healing, transwell, and xenograft assays were adopted to explore the biological function of GRB7 in HER2+ BC. RNA sequencing was performed to analyze the signaling pathways associated with GRB7 in SK-BR-3 cells after the cells were transfected with GRB7 siRNA. Chromatin immunoprecipitation analysis (ChIP) and luciferase reporter assay were employed to elucidate the potential molecular regulatory mechanisms of GRB7 in HER2+ BC. Results GRB7 was markedly upregulated and associated with poor prognosis in BC, especially in HER2+ BC. Overexpression of GRB7 increased the proliferation, migration, invasion, and colony formation of HER2+ BC cells, while depletion of GRB7 had the opposite effects in HER2+ BC cells and inhibited xenograft growth. ChIP-PCR and luciferase reporter assay revealed that TCF12 directly bound to the promoter of the GRB7 gene to promote its transcription. GRB7 facilitated HER2+ BC epithelial-mesenchymal transition (EMT) progression by interacting with Notch1 to activate Wnt/β-catenin pathways and other signaling (i.e., AKT, ERK). Moreover, forced GRB7 overexpression activated Wnt/β-catenin to promote EMT progression, and partially rescued the inhibition of HER2+ BC proliferation, migration and invasion induced by TCF12 silencing. Conclusions Our work elucidates the oncogenic role of GRB7 in HER2+ BC, which could serve as a prognostic indicator and promising therapeutic target.

To replace explosive nitrate-based chemicals in MS medium, this study developed a new, safer, and more cost-effective method using fertilizer-grade calcium ammonium nitrate and ammonium sulfate. This approach replaces ammonium nitrate and potassium nitrate, ensuring both safety and cost efficiency for sugarcane propagation. Six local sugarcane varieties—Zhongtang1 (ZT1), Zhongtang3 (ZT3), Zhongtang6 (ZT6), Guitang42 (GT42), Guitang44 (GT44), and Guiliu 07150 (GT07150)—were used. In the control group (Ck), nitrate ions (NO3−) were 39.28 mM, and ammonium ions (NH4+) were 20.49 mM, with a 2:1 ratio. In the treatment groups, the concentrations of nitrate ions (NO3−) and ammonium ions (NH4+) included treatment 1 (19.69 mM NO3− and 10.3 mM NH4+), treatment 2 (29.54 mM and 15.44 mM), treatment 3 (39.38 mM and 20.59 mM), treatment 4 (49.225 mM and 25.74 mM), treatment 5 (59.07 mM and 30.89 mM), and treatment 6 (68.915 mM and 36.03 mM), respectively, all with the same 2:1 ratio. Fifty bottles per treatment, with three replicates, were used for each sugarcane plantlets treatment. After five subcultures, the optimal ratio was determined by assessing morphological and physiological parameters, nitrogen levels, and SOD enzyme activity. The results indicated that treatment 3 (39.38 mM and 20.59 mM) and treatment 4 (49.225 mM and 25.74 mM) had the best morphological and physiological indicators. The optimal doses of calcium ammonium nitrate and ammonium sulfate were found in treatments 3 and 4, as well as in the control, with no significant difference among them. However, treatment 3, due to its lower dose, was more cost effective. To improve cost efficiency in practical production, it is recommended to use the lower concentration ratio of treatment 3 for plant tissue culture plantlets.

Environmental stresses are the main constraints on agricultural productivity and food security worldwide. This issue is worsened by abrupt and severe changes in global climate. The formation of sugarcane yield and the accumulation of sucrose are significantly influenced by biotic and abiotic stresses. Understanding the biochemical, physiological, and environmental phenomena associated with these stresses is essential to increase crop production. This review explores the effect of environmental factors on sucrose content and sugarcane yield and highlights the negative effects of insufficient water supply, temperature fluctuations, insect pests, and diseases. This article also explains the mechanism of reactive oxygen species (ROS), the role of different metabolites under environmental stresses, and highlights the function of environmental stress-related resistance genes in sugarcane. This review further discusses sugarcane crop improvement approaches, with a focus on endophytic mechanism and consortium endophyte application in sugarcane plants. Endophytes are vital in plant defense; they produce bioactive molecules that act as biocontrol agents to enhance plant immune systems and modify environmental responses through interaction with plants. This review provides an overview of internal mechanisms to enhance sugarcane plant growth and environmental resistance and offers new ideas for improving sugarcane plant fitness and crop productivity.

Banana (Musa spp.) is one of the most economically important horticultural crops. There are many types of banana, with differing ploidy (usually diploid, triploid, or tetraploid) and genome types (most containing the A or/and B genome). Currently, observation and genome type detection are commonly used to identify banana germplasm resources. However, observation is tedious, while genome type detection cannot distinguish categories below genome types. It is, therefore, urgent to establish a simple and effective method for identifying banana germplasm resources. This study sequenced and analyzed the ribosomal DNA internal transcribed spacer (ITS) sequences of 62 banana germplasm resources and found that the sequencing peaks, especially the 20 bp region near the 420-bp position (referred to as the 420-bp region), exhibited relatively recognizable and repeatable polymorphism characteristics. Using the 420-bp region as a marker, we were able to quickly distinguish bananas belonging to different genome type groups or different subgroups in the same genome type group. Moreover, it appeared that Sanger sequencing of ITS could be used to identify hybrid banana offspring. In general, ITS sequencing simplifies the classification of banana germplasm resources and has potential application in several areas of Musa improvement.

[Objectives] Sugarcane is the most significant sugar cash crop in the tropical and subtropical regions of China. However, it is notable for its narrow genetic basis, limited trait improvement, weak adaptability of varieties, and poor planting efficiency. In order to accelerate the process of sugarcane variety replacement and expand the basis of genetic variation, interspecific hybridization and multiple mutagen-esis are the most effective methods for obtaining new sugarcane varieties. The evaluation and identification of phenotypic traits of germplasm represents a significant analytical method. The "Zhongtang" series of sugarcane varieties is distinguished by its high yield and high sugar content. New sugarcane germplasms with excellent agronomic and quality traits can be identified and developed as breeding parents and new breeding lines through comprehensive evaluation of the existing germplasm. [Methods] A total of 181 new lines were selected through genetic origin and orientation, and evaluated and appraised for growth, yield, and economic characteristics. The data on 6 important agronomic and quality traits, including tillering, initial plant height, plant height, stem diameter, number of effective stems, and brix at maturity, were used to discover superior lines. These traits were evaluated during the two production seasons of the participant lines. [Results] A comprehensive evaluation of seedling growth traits and economic traits at maturity of the experimental lines identified 30 excellent new germ-plasms of sugarcane. Among the selected lines, 1501 and 1701 exhibited superior agronomic and quality traits, rendering them suitable as parental lines for sugarcane breeding or breeding as new varieties. [Conclusions] The exemplary results obtained in this study provide a solid foundation for the improvement of sugarcane germplasm, with the goal of enhancing quality and efficiency. These findings are of great scientific and practical significance to the study of sugarcane yield and sugar-related genes, as well as the exploration of the evaluation and utilization of sugarcane germplasm resources. The effects of carbon content on the mechanical properties and deformation mechanisms of boron carbides were investigated by first-principles calculations, based on the density functional theory. The B12–CBC (13.33 at % C) and B10 C 2 P –CC (28.75 at % C) were studied and then compared with the deformation of regular B11CP–CBC (20.0 at % C). The results show the B10 C 2 P –CC, which has the lowest carbon content, has the highest strength and hardness as well as the lowest toughness. With the increase of carbon content, the rhombohedral symmetry will be broken and the three-atoms chains will be replaced by diatomic carbon chains. These changes may have an influence on their anisotropic deformation mechanisms. For the B12–CBC, the destruction of icosahedra without bending three-atom chains causes structural failure for compression along the c axis; while for compression along the a axis, new B–B bonds are formed, causing an unrecoverable deformation; then it is gradually destroyed until full destruction. For the B10 C 2 P –CC, the anisotropic deformation mechanism is not obvious. For both loading directions, the breakage of B–CP bonds causes the stress to drop, suggesting that the structure is beginning to be destroyed. Finally, the icosahedra are fully destroyed, resulting in structural failure.

[Objectives] This study was conducted to investigate the mutagenic effects of space radiation mutagenesis on sugarcane hybrid seeds. [Methods] Sugarcane hybrid spikes were subjected to space radiation mutagenesis treatment to identify the effects on the germination, seedling growth and plant growth and development of sugarcane hybrid seeds. [ Results] The space radiation treatment had a great impact on the germination and growth of sugarcane hybrid seeds, especially on the germination of sugarcane seeds. The germination number per gram of seeds increased significantly, and die survival rate of seedlings increased significantly. The effect on die growth status was relatively small. The average number of tillers decreased slightly, the percentage of available stems increased, and the plants became higher, which resulted in a greater effect on sugarcane yield. Sugarcane sucrose was increased. The seed germination and late growth status indicated that space radiation promoted the germination and growth of sugarcane hybrid seeds. [ Conclusions] This study provides a basis for the space radiation mutagenesis breeding of sugarcane hybrid seeds and the promotion of sugarcane breeding process. [ABSTRACT FROM AUTHOR]

Prismatic members subjected to internal axial loads exist widely in engineering applications. Because the effects of internal loads and end loads on stability are quite different, the traditional effective length method is not applicable. Engineers require an efficient and convenient method for solving the stability problem of such members. A two-bar model is proposed to illustrate the concept that the effect of compressive axial loads on the stability of sway-inhibited columns can be understood as a negative lateral stiffness. After obtaining the expression of negative lateral stiffness, a relationship between end loads and internal loads can be established so that the internal loads can be considered equivalent to end loads. Then, the column subjected to internal loads can be analyzed as a normal column (without internal axial loads), and the critical buckling load can be obtained easily with the Euler formula. Eigenvalue buckling analyses were performed to examine the proposed method, and the comparison results indicate it has high accuracy. [ABSTRACT FROM AUTHOR]

Because loess underground erosion is one of the major engineering geological problems in loess areas of China, foundation loess of the Zichang-Yongping section of West-East Gas Pipelines is taken as the research object. The soil-water characteristic curve (SWCC) and hydraulic conductivity function (HCF) of samples have been obtained by laboratory tests, and on this basis, seepage field numerical simulations of loess foundation with and without the pipelines have been processed by the finite-element method. Critical groundwater seepage velocity of underground erosion and the relationship between groundwater seepage velocity and underground erosion velocity have been tested by self-made instruments. Results of tests and numerical simulation indicate that the pipelines not only change the original seepage field of a loess foundation, but also accelerate the underground erosion of a loess foundation. Loess on the top and upstream groundwater side of pipelines will be eroded first, finally causing the failure of foundation and the exposure of pipelines. The combination of soil solidified material and revegetation is suggested as an effective and durable solution to deal with the underground erosion problem of loess pipeline foundations. Objectives: To evaluate the effect of different endodontic sealers (epoxy resin-based and bioceramic-based) and the time of post-cementation on the bond strength of a fiber post cemented with resin cement. Methods: Forty human premolars were instrumented and divided into 4 groups. According to the type of sealer and the time of post-cementation: AH-IM (AH Plus, post-cemented immediately after root canal treatment), SP-IM (iRoot SP, post-cemented immediately after root canal treatment), AH-OW (AH Plus, post-cementation after one week), and SP-OW (iRoot SP, post-cementation after one week). In each group, the samples were submitted to push-out test, and failure mode was assessed. Levene's test, one-way ANOVA, and Kruskal-Wallis analysis were applied for statistical analysis (α = 5%). Results: The highest mean push-out bond strength was obtained from the SP-IM group in the apical part (10.45 ± 5.15MPa), while the lowest was observed in samples from the AH-OW group in the middle part (2.63 ± 1.54 MPa). One-way ANOVA showed that within the same root region, the time of post-cementation had a negative influence on the bonding strength in the SP groups in the middle and apical portion (P<0.05), however, when comparing the effect of type of sealers on bonding strength between the OW groups or IM groups within the same root region, no significant difference was observed regardless of the post cementation time (P>0.05). Conclusions: The bond strength of the fiber post was higher when the post was cemented immediately after root canal treatment when the bioceramic sealer was applied. Clinical relevance: The correct choice of an endodontic sealer and the proper time of post-cementation may help to obtain the best quality of post-and-core restoration. [ABSTRACT FROM AUTHOR]

This study examines the early career of the renowned Buddhologist Lü Cheng as an aspiring revolutionary. My findings reveal that Lü's rhetoric of "aesthetic revolution" both catapulted him into the center of the New Culture Movement and popularized a Buddhist idealism--Yogācāra (consciousness-only school)--among thinkers who sought alternatives social theories. Lü aimed to refute social Darwinism and scientific materialism, which portray humans as mechanized individuals bereft of moral agency. He theorized an antirealist social ontology, i.e., a social oneness grounded in intersubjective resonances, from which subjective interiority and objective exteriority arise. Lü turned to Buddhism to further his revolution. Buddhist soteriology supplied powerful tools for theorizing the social: The doctrine of no-self refuted philosophical solipsism and curtailed individualism; dependent-origination refashioned social evolution as collective spiritual progress. Lü's spiritual-evolutionism-cum-socialontology broadens the field of Buddhist philosophy that has a long-standing blind spot on social philosophies developed in the Global South. This study discussed the development and evolution of Chinese royal gardens from the initial generation period (Zhou, Qin and Han dynasties) to the late mature period (middle Qing Dynasty to the late Qing Dynasty). Through the specific analysis of the typical royal gardens in different periods, the development and changes of the gardening skills and overall layout of the royal gardens in each period were obtained, and the development characteristics of the whole historical period of the Chinese royal gardens were summarized. "Empathy" is a common educational expectation among lower-class families, as well as a natural response by children. Through analysis of the growth narratives of a group of rural children who were born after the Opening and Reforms and achieved higher educational attainment, this study found that their "empathy" carried plural connotations, including love, understanding, affection, independence, and recompense. Although "empathy" allowed them to integrate into the family community, it also framed their role in the family, limited their emotional expression, and produced a relational structure between family members in which love and resentment were intertwined. However, the estrangement between rural children with higher educational attainment and their families was not insurmountable: by creatively exploring new methods of communication, emotional barriers which might form in an instant could also vanish in an instant. To experimentally validate a recently developed theory for predicting the stability of cut slopes under unsaturated conditions, the authors measured increasing strain localization in unsaturated slope cuts prior to abrupt failure. Cut slope width and moisture content were controlled and varied in a laboratory, and a sliding door that extended the height of the free face of the slope was lowered until the cut slope failed. A particle image velocimetry tool was used to quantify soil displacement in the x-y (horizontal) and x-z (vertical) planes, and strain was calculated from the displacement. Areas of maximum strain localization prior to failure were shown to coincide with the location of the eventual failure plane. Experimental failure heights agreed with the recently developed stability theory for unsaturated cut slopes (within 14.3% relative error) for a range of saturation and cut slope widths. A theoretical threshold for sidewall influence on cut slope failures was also proposed to quantify the relationship between normalized sidewall width and critical height. The proposed relationship was consistent with the cut slope experiment results, and is intended for consideration in future geotechnical experiment design. The experimental data of evolution of strain localization presented herein provide a physical basis from which future numerical models of strain localization can be validated. [ABSTRACT FROM AUTHOR]

Main conclusions : Exogenously applied GR24 affected somatic embryo formation and morphogenesis of strigolactone-deficient tomato mutant through cross-talk with auxins and cytokinins indicating involvement of SLs in the embryogenic process. Strigolactones (SLs) mediate the regulation of plant responses to the environment through cross-talk with other plant hormones, especially auxins. Auxins play a crucial role in coordinating the morphogenesis and development of plant reproductive organs, including the signal-transduction cascade leading to the reprogramming of gene-expression patterns before embryo formation. SLs' role in these processes is unknown, in contrast to their proven involvement in auxin transport and distribution. We used tomato cv. M82 and its SL-deficient mutant SL-ORT1 to study the influence of SLs on hormone profile in tomato roots and shoots, and their involvement in somatic embryogenesis (SE) and morphogenesis (adventitious root formation). The synthetic SL GR24 had different effects on SE of M82 and SL-ORT1, indicating that SLs influence the cytokinin-to-auxin ratio in tomato SE. [ABSTRACT FROM AUTHOR]
