Simple Summary: Milk fat is one of the most important economic traits in dairy cow production. Numerous investigations have demonstrated the influence of the ruminal microbiome's composition and metabolic profile on milk production. However, no studies have directly connected the ruminal metabolome with bacterial and fungal communities as a function of milk fat content in dairy cows. The specific objective of the present study was to compare ruminal metabolome with bacterial and fungal communities among Chinese Holstein cows with contrasting milk fat content that were fed the same diet. Our results confirm that ruminal metabolome, bacterial, and fungal communities differ significantly in dairy cows with different milk fat contents. Among these different metabolites, lauric acid was enriched in fatty acid biosynthesis and selected for an in vitro study with bovine mammary epithelial cells, indicating that lauric acid promoted intracellular triglyceride synthesis via upregulation of the mRNA abundance of fatty acid uptake and activation, as well as lipogenesis. This study revealed microbial-mediated metabolic processes related to milk fat synthesis. The composition and metabolic profile of the ruminal microbiome have an impact on milk composition. To unravel the ruminal microbiome and metabolome affecting milk fat synthesis in dairy cows, 16S rRNA and internal transcribed spacer (ITS) gene sequencing, as well as ultra-high performance liquid chromatography-mass spectrometry (UHPLC-MS/MS) methods were used to investigate the significant differences in ruminal bacterial and fungal communities as well as metabolome among Chinese Holstein cows with contrasting milk fat contents under the same diet (H-MF 5.82 ± 0.41% vs. L-MF 3.60 ± 0.12%). Another objective was to culture bovine mammary epithelial cells (BMECs) to assess the effect of metabolites on lipid metabolism. Results showed that the acetate-to-propionate ratio and xylanase activity in ruminal fluid were both higher in H-MF. Microbiome sequencing identified 10 types of bacteria and four types of fungi differently abundant at the genus level. Metabolomics analysis indicated 11 different ruminal metabolites between the two groups, the majority of which were lipids and organic acids. Among these, lauric acid (LA) was enriched in fatty acid biosynthesis with its concentration in milk fat of H-MF cows being greater (217 vs. 156 mg per 100 g milk), thus, it was selected for an in vitro study with BMECs. Exogenous LA led to a marked increase in intracellular triglyceride (TG) content and lipid droplet formation, and it upregulated the mRNA abundance of fatty acid uptake and activation (CD36 and ACSL1), TG synthesis (DGAT1, DGAT2 and GPAM), and transcriptional regulation (SREBP1) genes. Taken together, the greater relative abundance of xylan-fermenting bacteria and fungi, and lower abundance of bacteria suppressing short-chain fatty acid-producing bacteria or participating in fatty acid hydrogenation altered lipids and organic acids in the rumen of dairy cows. In BMECs, LA altered the expression of genes involved in lipid metabolism in mammary cells, ultimately promoting milk fat synthesis. Due to global warming, late-spring coldness affecting wheat (LSCW) is one of the major abiotic adversities affecting wheat production. A combination of field and pot trials were conducted in this study. In the field experiment, 20 wheat varieties from the main wheat-producing areas in China were selected as experimental materials. By exploring the effects of LSCW on the spikelet characteristics and yields of different varieties, the evaluation methods and indexes of wheat varieties' resistance to LSCW were established. Three varieties with strong resistance to LSCW (Yannong19, Guomai9, and Shannong17) and five varieties sensitive to LSCW (Zhengmai895, Xinmai26, Zhengmai366, Zhengmai8329, and Fanmai5) were screened out. The wheat varieties Yannong19 (YN19), with a strong resistance to LSCW, and Xinmai26 (XM26), with LSCW sensitivity, were selected as the test materials for the pot experiment. The ultrastructure changes in the wheat in different low-temperature treatments during the anther differentiation period were observed using an ultra-low-temperature artificial climate incubator set to 4 °C and −4 °C for 4 h (1:00–5:00 a.m.). The average temperature of the field during the low-temperature treatment was 10 °C, which was the control temperature (CK). The results showed that the cell morphology and chloroplast and mitochondrial structures of the functional leaves, young ears, and internodes below the ears were damaged, and the degree of damage was related to the cell location, the extent of low-temperature stress, and the resistance of the varieties. The degree of damage to the wheat cells was found to occur in the following order: young ears > internode belove young ears > functional leaves. The degrees of damage to the wheat cell, chloroplast, and mitochondrial structures increased with the intensification of the low-temperature stress. The damage to the XM26 variety was obviously greater than that inflicted on the YN19 variety. The anatomical mechanism of YN19 cells makes the cell structure more stable during late-spring coldness. Background: Vinegar-quenched Testudinis Carapax et Plastrum (TCP) and vinegar-quenched Trionycis Carapax (TC) are made from the shells of Chinemys reevesii and Trionyx sinensis. However, some cheap substitutes from non-medicinal species such as Trachemys scripta, Mauremys sinensis, Chelydra serpentina , and Apalone ferox are often seen on the market, which exposes public health to high risk. Conventional methods are not applicable for the identification of these two products since their intrinsic constituents are largely destroyed after being highly processed. Objectives: A new quantitative polymerase chain reaction (qPCR) method was developed to detect DNA components of Vinegar-quenched TCP and vinegar-quenched TC based on simple sequence repeat differential genes of animal-derived mitochondria. Materials and Methods: Six species-specific primer reactions were designed and further validated for specificity, sensitivity, and repeatability. Finally, the developed method was used to assess the authenticity of their commercial products. Results: The assay exhibited good specificity, and the limit of detection was above 1 × 102 copies/µL. There was good linearity in the concentration, ranging from 1 × 102 to 1 × 107 copies/µL. In addition, the method was validated through repeatability assessment (coefficient variation <1%). While qPCR was applied for the analysis of 36 batches of commercial products of these two tonics, it could effectively identify the authenticity of the products, and consequently, seven batches were fake or adulterated. Conclusion: This newly proposed method is promising for the quality evaluation of highly processed animal-derived Chinese medicines, which will assist in ensuring safety and efficacy in clinical practice and protect fair trade in the market. Simple Summary: The intestinal epithelium is one of the tissues that renews the fastest in the body. This process is driven by intestinal stem cells and is greatly influenced by adenosine triphosphate supply. Glutamine is the preferred energy substrate for the intestinal epithelium, unlike most tissues in the body that prefer glucose. However, there needs to be more understanding of how glutamine affects gene expression in the intestinal epithelium. To address this gap, this study aimed to identify the essential genes and signals involved in glutamine-induced growth of intestinal epithelial cells by exploring gene expression profiles. The results showed that glutamine might upregulate deoxyribonucleic acid replication licensing factors, increasing proliferation-related signaling and suppressing inflammation-related pathways. Consequently, this mechanism leads to the proliferation of intestinal epithelial cells and the expansion of intestinal stem cells. Understanding these crucial targets provides new insight into regulating the homeostasis of the intestinal epithelium, particularly in porcine production. The intestinal epithelium is known for its rapid self-renewal, and glutamine is crucial in providing carbon and nitrogen for biosynthesis. However, understanding how glutamine affects gene expression in the intestinal epithelium is limited, and identifying the essential genes and signals involved in regulating intestinal epithelial cell growth is particularly challenging. In this study, glutamine supplementation exhibited a robust acceleration of intestinal epithelial cell proliferation and stem cell expansion. RNA sequencing indicated diverse transcriptome changes between the control and glutamine supplementation groups, identifying 925 up-regulated and 1152 down-regulated genes. The up-regulated DEGs were enriched in the KEGG pathway of cell cycle and GO terms of DNA replication initiation, regulation of phosphatidylinositol 3-kinase activity, DNA replication, minichromosome maintenance protein (MCM) complex, and ATP binding, whereas the down-regulated DEGs were enriched in the KEGG pathway of p53 signaling pathway, TNF signaling pathway, and JAK-STAT signaling pathway and GO terms of inflammatory response and intrinsic apoptotic signaling pathway in response to endoplasmic reticulum stress. Furthermore, GSEA analysis revealed a significant up-regulation of the cell cycle, DNA replication initiation, ATP-dependent RNA helicase activity, and down-regulation of the TNF signaling pathway. The protein–protein association network of the intersecting genes highlighted the significance of DNA replication licensing factors (MCM3, MCM6, and MCM10) in promoting intestinal epithelial growth in response to glutamine. Based on these findings, we propose that glutamine may upregulate DNA replication licensing factors, leading to increased PI3K/Akt signaling and the suppression of TNF, JAK-STAT, and p53 pathways. Elabela (ELA), which is the second endogenous peptide ligand of the apelin receptor (APJ) to be discovered, has been widely studied for potential use as a therapeutic peptide. However, its role in ischemic stroke (IS), which is a leading cause of disability and death worldwide and has limited therapeutic options, is uncertain. The aim of the present study was to investigate the beneficial effects of ELA on neuron survival after ischemia and the underlying molecular mechanisms. Primary cortical neurons were isolated from the cerebral cortex of pregnant C57BL/6J mice. Flow cytometry and immunofluorescence showed that ELA inhibited oxygen–glucose deprivation (OGD) -induced apoptosis and axonal damage in vitro. Additionally, analysis of the Gene Expression Omnibus database revealed that the expression of microRNA-124-3p (miR-124-3p) was decreased in blood samples from patients with IS, while the expression of C-terminal domain small phosphatase 1 (CTDSP1) was increased. These results indicated that miR-124-3p and CTDSP1 were related to ischemic stroke, and there might be a negative regulatory relationship between them. Then, we found that ELA significantly elevated miR-124-3p expression, suppressed CTDSP1 expression, and increased p-AKT expression by binding to the APJ receptor under OGD in vitro. A dual-luciferase reporter assay confirmed that CTDSP1 was a direct target of miR-124-3p. Furthermore, adenovirus-mediated overexpression of CTDSP1 exacerbated neuronal apoptosis and axonal damage and suppressed AKT phosphorylation, while treatment with ELA or miR-124-3p mimics reversed these effects. In conclusion, these results indicated that ELA could alleviate neuronal apoptosis and axonal damage by upregulating miR-124-3p and activating the CTDSP1/AKT signaling pathway. This study, for the first time, verified the protective effect of ELA against neuronal injury after ischemia and revealed the underlying mechanisms. In Internet of Things (IoT), the freshness of status updates is crucial for mission-critical applications. In this regard, it is suggested to quantify the freshness of updates by using Age of Information (AoI) from the receiver’s perspective. Specifically, the AoI measures the freshness over time. However, the freshness in the content is neglected. In this paper, we introduce an age-based utility, named as Age of Changed Information (AoCI), which captures both the passage of time and the change of information content. By modeling the underlying physical process as a discrete time Markov chain, we investigate the AoCI in a time-slotted status update system, where a sensor samples the physical process and transmits the update packets to the destination. With the aim of minimizing the weighted sum of the AoCI and the update cost, we formulate an infinite horizon average cost Markov Decision Process. We show that the optimal updating policy has a special structure with respect to the AoCI and identify the condition under which the special structure exists. By exploiting the special structure, we provide a low complexity relative policy iteration algorithm that finds the optimal updating policy. We further investigate the optimal policy for two special cases. In the first case where the state of the physical process transits with equiprobability, we show that optimal policy is of threshold type and derive the closed-form of the optimal threshold. We then study a more generalized periodic Markov model of the physical process in the second case. Lastly, simulation results are laid out to exhibit the performance of the optimal updating policy and its superiority over the zero-wait baseline policy. [ABSTRACT FROM AUTHOR]

Introduction: SARS-CoV-2 is a new variant of virus that attacks the human respiratory tract which was declared by WHO as a Covid19 pandemic. Covid19 transmission through solid surfaces, air, and waste occurring in hospital areas require disinfection and sanitation monitoring to ensure that new clusters are not created and outpatients are not infected with the SARS-CoV-2 virus in hospitals. Discussion: Using literature review method, 11 relevant articles that meet the inclusion criteria were selected. In the results of the study, there was a collection of samples in the solid surfaces, air and waste in both medical and non-medical areas containing SARS-CoV-2 RNA. The common objects where SARS-CoV-2 was discovered include doorknobs, toilet bowls, toilet flush buttons, and medical devices. In non-medical areas, the objects include computer keyboards, mice, and monitors. Chlorine-based disinfection for surfaces, monitoring in environmental sanitation by changing pillow and bedsheets and cleaning tables regularly were subsequently implemented. Conclusion: The implementation of environmental sanitation and disinfection is effective in eliminating the SARS-CoV-2 virus in hospitals. Studies indicated that disinfection is 95% to 99.99% effective in eliminating the SARS-CoV-2 virus. This work studied the rheological, thermal, and mechanical properties of bitumen modified by reclaimed polyethylene (PE), and polypropylene (PP) from different recycling sources. The rutting resistance under high temperature of polymer modified bitumen (PMB) was investigated by rheologically temperature ramp test and multiple stress creep recovery (MSCR) test. It is found that for some modified bitumen, a plateau of complex modulus G* could be formed with temperature increment, revealing rheological stability. Furthermore, these samples with rheological stability revealed a higher creep recovery and lower creep compliance measured by the MSCR test. Glass transition temperature (Tg) measured by dynamic mechanical analysis was used to evaluate the crack resistance under a low temperature of PMB. The influence of recycled PE on the Tg of modified bitumen was different from that of recycled PP modified bitumen, as compared with corresponding virgin polymer modified bitumen. A possible reason for the various effect of recycling sources on the service property of modified bitumen was explored by crystallization and melting behaviors of polymer in bitumen since that polymer with higher crystallinity degree could endow the modified bitumen stiffness, which was closely, related to their service property especially the rutting resistance. Transition metal dichalcogenides (TMDCs) are increasingly being used for chemical sensing, biosensing, and tumor therapy on account of their diversity, biocompatibility, multifunctionality, adjustable bandgap, and excellent photoelectric characteristics. This review mainly discusses the effect of the elemental composition and structure of TMDCs on the performance of electrochemical, biofluorescence, and colorimetric biosensors. The applications of TMDCs in tumor therapies are reviewed here. Furthermore, the current challenges and future directions for developing TMDC‐based theranostics are summarized. As the main biological components of the air ecosystem, airborne microorganisms play an important role in ecosystem function, and most existing studies focus on their health and environmental effects, while ignoring ecological effects. In this study, air microbial samples from two sampling points in the coastal city of Qingdao were collected by a large volume air sampler. The Illumina MiSeq and PacBio high-throughput sequencing methods were used to analyze the microbial community structure and diversity of the two sampling points. The results showed that there were significant differences in the number and relative abundances of microorganisms identified by the two platforms, and with the refinement of the sample classification, the differences in the community became increasingly significant. Proteobacteria and Firmicutes were the dominant bacterial phyla among the airborne microorganisms. MiSeq sequencing showed that the relative abundances of unidentified bacteria were as high as 8.03–16.42% in MiSeq sequencing, while they were almost undetected in PacBio sequencing. Based on the different sequencing platforms, Bacilli, Gammaproteobacteria and Alphaproteobacteria could be seen as significant gaps in the two sampling points. Betaproteobacteria showed differences only in the coastal area, while the other 14 classes were differentially abundant in the urban area. Pseudomonas was the main genus in the aerosol, followed by Lactococcus. The relative abundances of bacterial taxa displayed statistically significant differences in the different functional areas. There were more diverse microbes in the urban area than in the coastal area; moreover, the relative abundances of dominant genera showed significant differences, such as Variovorax and Deinococcus. Objective: To explore the changes in microbial composition and the corresponding impact after lactitol treatment in constipated patients. Methods: Altogether 29 consecutive outpatients diagnosed with chronic constipation from three centers were recruited and stratified based on their history of diabetes mellitus. All patients were administered with oral lactitol for 2 weeks, and a symptoms diary of constipation was recorded. Fecal samples were collected before and after lactitol treatment, and were analyzed by 16S rRNA sequencing and real‐time polymerase chain reaction (PCR) to detect gut microbiota. Results: Twenty patients with diabetes mellitus and nine without, all with chronic constipation, were enrolled in this study. After 2‐week administration of lactitol, their subscale scores and constipation symptoms significantly decreased (P < 0.05). An analysis of fecal flora using 16S rRNA sequencing found an increasing trend of abundance of Bifidobacterium in the post‐lactitol group (P = 0.08). Actinobacteria, Actinobacteria, Bifidobacteriales, Bifidobacteriaceae and Bifidobacterium were significantly more abundant after lactitol administration. Real‐time PCR showed significantly high DNA copy numbers of Bifidobacterium after lactitol treatment (1.39 × 1010 vs 2.74 × 109 copies/μL, P = 0.01). The results of 16S rRNA sequencing and real‐time PCR illustrated an increasing trend of Bifidobacterium in both patients with and without diabetes. In addition, Bifidobacterium was negatively correlated with constipation subscale scores. Conclusions: Alterations in fecal flora composition after lactitol supplementation, especially in terms of an increasing trend of Bifidobacterium, alleviated constipation symptoms. The evolution of increased competitive ability (EICA) hypothesis states that, when introduced in a novel habitat, invasive species may reallocate resources from costly quantitative defense mechanisms against enemies to dispersal and reproduction; meanwhile, the refinement of EICA suggests that concentrations of toxins used for qualitative defense against generalist herbivores may increase. Previous studies considered that only few genotypes were introduced to the new range, whereas most studies to test the EICA (or the refinement of EICA) hypotheses did not consider founder effects.In this study, genetic and phenotypic data of Chromolaena odorata populations sampled across native and introduced ranges were combined to investigate the role of postintroduction evolution in the successful invasion of C. odorata.Compared with native populations, the introduced populations exhibited lower levels of genetic diversity. Moreover, different founder effects events were interpreted as the main cause of the genetic structure observed in introduced ranges. Three Florida, two Trinidad, and two Puerto Rico populations may have been the sources of the invasive C. odorata in Asia.When in free of competition conditions, C. odorata plants from introduced ranges perform better than those from native ranges at high nutrient supply but not at low nutrient level. The differences in performance due to competition were significantly greater for C. odorata plants from the native range than those from the introduced range at both nutrient levels. Moreover, the differences in performance by competition were significantly greater for putative source populations than for invasive populations.Quantities of three types of secondary compounds in leaves of invasive C. odorata populations were significantly higher than those in putative source populations. To alleviate the negative impacts of muscle fatigue on a force estimation model, a modification framework taking use of fatigue index was put forward in this paper. Muscle force and surface electromyography were first collected using high-density electrode grid and dynamometer. Then, multi-step signal pre-processing and a nonnegative matrix factorization-based signal optimization were conducted, with fatigue indices being extracted in the same time. Next, a degree 4 polynomial fitting model was employed to undertake the training process, and the relationship between the generated model parameters and fatigue indices was built up. In the end, the parameter-index relationship was applied on different testing sets to complete fatigue-modified force estimation. Significant improvement was found in most testing cases across different sexes and ages. Relative decreases of 36.5%, 20.7%, and 20.4% in the percentage root mean square error were achieved by young males, young females, and elderly males. The proposed method can boost the performances of force estimation models, thereby contributing to the development of a variety of fields including biomechanical study, rehabilitation treatment, and prosthesis research. Piled embankments, which offer many advantages, are increasingly popular in construction of high-speed railways in China. Although the performance of piled embankment under static loading is well-known, the behavior under the dynamic train load of a high-speed railway is not yet understood. In light of this, a heavily instrumented piled embankment model was set up, and a model test was carried out, in which a servo-hydraulic actuator outputting M-shaped waves was adopted to simulate the process of a running train. Earth pressure, settlement, strain in the geogrid and pile and excess pore water pressure were measured. The results show that the soil arching height under the dynamic train load of a high-speed railway is shorter than under static loading. The growth trend for accumulated settlement slowed down after long-term vibration although there was still a tendency for it to increase. Accumulated geogrid strain has an increasing tendency after long-term vibration. The closer the embankment edge, the greater the geogrid strain over the subsoil. Strains in the pile were smaller under dynamic train loads, and their distribution was different from that under static loading. Supramolecular materials, in which small organic molecules are assembled into regular structures by non-covalent interactions, attract tremendous interests because of their highly tunable functional groups and porous structure. Supramolecular adsorbents are expected to fully expose their abundant adsorptive sites in a dynamic framework. In this contribution, we introduced cucurbit[6]uril as a supramolecular capsule for reversible storage/delivery of mobile polysulfides in lithium-sulfur (Li-S) batteries to control undesirable polysulfide shuttle. The Li-S battery equipped with the supramolecular capsules retains a high Coulombic efficiency and shows a large increase in capacity from 300 to 900 mAh g−1 at a sulfur loading of 4.2 mg cm−2. The implementation of supramolecular capsules offers insights into intricate multi-electron-conversion reactions and manifests as an effective and efficient strategy to enhance Li-S batteries and analogous applications that involve complex transport phenomena and intermediate manipulation. The lithium-sulfur (Li-S) battery is a promising high-energy-density storage system. The strong anchoring of intermediates is widely accepted to retard the shuttle of polysulfides in a working battery. However, the understanding of the intrinsic chemistry is still deficient. Inspired by the concept of hydrogen bond, herein we focus on the Li bond chemistry in Li-S batteries through sophisticated quantum chemical calculations, in combination with 7Li nuclear magnetic resonance (NMR) spectroscopy. Identified as Li bond, the strong dipole-dipole interaction between Li polysulfides and Li-S cathode materials originates from the electron-rich donors (e.g., pyridinic nitrogen (pN)), and is enhanced by the inductive and conjugative effect of scaffold materials with π-electrons (e.g., graphene). The chemical shift of Li polysulfides in 7Li NMR spectroscopy, being both theoretically predicted and experimentally verified, is suggested to serve as a quantitative descriptor of Li bond strength. These theoretical insights were further proved by actual electrochemical tests. Multiple-input–multiple-output (MIMO) full-duplex relaying (FDR) has been considered an efficient technique to provide coverage to users where their direct links from the base station (BS) are too weak for reliable signal reception. However, when multiple MIMO full-duplex relays are deployed in a network, the signal reception quality relies on the effective suppression of multiple types of interference. In this paper, the distributed beamforming is studied for the MIMO FDR network by formulating a power minimization problem with a nonstrict convex objective function (total transmit power of both the BS and all the relays in the network) under individual user rate constraints. We come up with two iterative distributed beamforming algorithms, Algorithm 1 for relays equipped with single receive antenna and Algorithm 2 with multiple receive antennas. The former can yield a global optimal solution of the power minimization problem, whereas the latter can yield only a local optimal solution due to conservative successive convex approximations (SCAs) performed at each iteration, and a rigorous analysis on the upper bounds of step sizes is proposed to guarantee their convergence. The proposed two algorithms only require local information exchange between relays and hence are scalable for different network sizes and topologies. An “early termination” strategy in the operation of the proposed two algorithms is also presented to acquire an acceptable transmit power solution with less computation time consumption and thus suitable for realistic applications. Finally, some simulation results are provided to demonstrate that the proposed two algorithms perform well and significantly better than the existing state-of-the-art scheme reported in the work of Lee and Shin.
