The Psoralen (Pso) molecule finds extensive applications in photo-chemotherapy, courtesy of its triplet state forming ability. Sulfur and selenium replacement of exocyclic carbonyl oxygen of organic chromophores foster efficient triplet harvesting with near unity triplet quantum yield. These triplet-forming photosensitizers are useful in Photodynamic Therapy (PDT) applications for selective apoptosis of cancer cells. In this work, we have critically assessed the effect of the sulfur and selenium substitution at the exocyclic carbonyl (TPso and SePso, respectively) and endocyclic oxygen positions of Psoralen. It resulted in a significant redshifted absorption spectrum to access the PDT therapeutic window with increased oscillator strength. The reduction in singlet-triplet energy gap and enhancement in the spin-orbit coupling values increase the number of intersystem crossing (ISC) pathways to the triplet manifold, which shortens the ISC lifetime from 10 -5  s for Pso to 10 -8  s for TPso and 10 -9  s for SePso. The intramolecular photo-induced electron transfer process, a competitive pathway to ISC, is also considerably curbed by exocyclic functionalizations. In addition, a maximum of 115 GM of two-photon absorption (2PA) with IR absorption (660-1050 nm) confirms that the Psoralen skeleton can be effectively tweaked via single chalcogen atom replacement to design a suitable PDT photosensitizer., (© 2024 The Authors. Intercellular heterogeneity occurs widely under both normal physiological environments and abnormal disease‐causing conditions. Several attempts to couple spatiotemporal information to cell states in a microenvironment were performed to decipher the cause and effect of heterogeneity. Furthermore, spatiotemporal manipulation can be achieved with the use of photocaged/photoactivatable molecules. Here, we provide a platform to spatiotemporally analyze differential protein expression in neighboring cells by multiple photocaged probes coupled with homemade photomasks. We successfully established intercellular heterogeneity (photoactivable ROS trigger) and mapped the targets (directly ROS‐affected cells) and bystanders (surrounding cells), which were further characterized by total proteomic and cysteinomic analysis. Different protein profiles were shown between bystanders and target cells in both total proteome and cysteinome. The palladium (II) catalyzed direct C3‐(sp2)−H alkenylation of five membered heterocyclic aldehydes has been developed using 3‐aminobutanoic acid as a catalytic transient directing group. A wide range of heterocyclic aldehydes reacted with various acrylates to afford highly site‐selective C3‐alkenylated products. This method features an exclusive E‐selective alkenylation at the C3‐position of heterocyclic aldehydes with a broad substrate scope. The dual‐ratiometric thermometry is one of highly accurate methods for microscopic thermal measurement in biological systems. Herein, a series of chromone derivatives with noncovalently intramolecular interactions (NIIs) were designed and synthesized for ratiometric thermometers. The triplet states of these organic compounds were systematically tuned upon regulating the conformation with NIIs to yield efficient room temperature phosphorescence and large wavelength difference between fluorescence and phosphorescence simultaneously. As a result, an unprecedent organic 3D dual‐ratiometric thermometer was established based on the intensity ratio and lifetime ratio of fluorescence/phosphorescence vs temperature, which was used for in vitro and in vivo bio‐thermometry with high accuracy. Dual‐emissive photoconvertible fluorophores (DPCFs) are powerful tools to unambiguously track labeled cells in bioimaging. We recently introduced a new rational mechanism called directed photooxidation‐induced conversion (DPIC) enabling efficient DPCFs to be obtained by conjugating a coumarin to aromatic singlet‐oxygen reactive moieties (ASORMs). Pyrrole was found to be a suitable ASORM as it provided a high hypsochromic shift along with a fast and efficient conversion. By synthesizing various pyrrole‐based styryl coumarin dyes, we showed that the photoconversion properties, including the quantum yield of photoconversion and the chemical yield of conversion can be tuned by chemical modification of the pyrrole. These modifications led to an improved dual emissive converter, SCP−Boc, which displayed a high brightness and an enhanced photoconversion yield of 63 %. Noninvasive treatments for terminal cancer patients constitute a new trend in tumor treatment. Therefore, phototherapy, including photodynamic therapy (PDT) and photothermal therapy (PTT), has attracted considerable interest. Light and a photosensitizer (PS) are employed in both treatment methods. For PDT, the PS generates reactive oxygen species (ROS) when exposed to light, whereas for PTT, the PS generates heat. In this regard, the systematic design of new PSs has become an active area of phototherapy research in an effort to solve the problems associated with conventional PSs. In the past decade, activatable and heavy‐atom‐free PSs have become significant research areas. Therefore, we discuss our recent contributions to the PDT and PTT in this account. Fluorescent probes are essential for single-molecule imaging. However, their application in biological systems is often limited by the short photobleaching lifetime. To overcome this, we developed a novel thiolation strategy for squaraine dyes. By introducing thiolation of the central cyclobutene of squaraine (thio-squaraine), we observed a ≈5-fold increase in photobleaching lifetime. Our single-molecule data analysis attributes this improvement to improved photostability resulting from thiolation. Interestingly, bulk measurements show rapid oxidation of thio-squaraine to its oxo-analogue under irradiation, giving the perception of inferior photostability. This discrepancy between bulk and single-molecule environments can be ascribed to the factors in the latter, including larger intermolecular distances and restricted mobility, which reduce the interactions between a fluorophore and reactive oxygen species produced by other fluorophores, ultimately impacting photobleaching and photoconversion rate. We demonstrate the remarkable performance of thio-squaraine probes in various imaging buffers, such as glucose oxidase with catalase (GLOX) and GLOX+trolox. We successfully employed these photostable probes for single-molecule tracking of CD56 membrane protein and monitoring mitochondria movements in live neurons. CD56 tracking revealed distinct motion states and the corresponding protein fractions. Coumarins, due to their intense and persistent emission of fluorescence, structural flexibility, and excellent biocompatibility, have become a popular choice in developing new fluorescent chemosensors. Coumarins have long been used for sensing anions, cations, chemically reactive nitrogen, oxygen, sulfur‐containing species, and small molecules. They have also been shown to have applicability in imaging, biology, and medical research. Due to the widespread use and excellent biological activity, hundreds of such coumarin motifs have either been isolated from nature or synthesized in a laboratory. This review covers all the developments pertaining to the synthesis and use of coumarin for sensing a variety of metal ions (alkali, alkaline earth, transition, heavy, and noble metal ions). This comprehensive review includes an overview of the coumarin‐based chemosensor's pH limit, detection limit, and binding mode. Single molecule localization microscopy based on photoactivation is a powerful tool for investigating the ultrastructure of cells. We developed a general strategy for photoactivatable fluorophores, using 2,3‐dihydro‐1,4‐oxathiine group (SO) as a tag to attach to various skeletal structures, including coumarin, BODIPY, rhodamine, and cyanine. The conjugation of SO resulted in a significant loss of fluorescence due to photoinduced electron transfer (PeT). Under the irradiation of excitation light, singlet oxygen generated by the fluorophores converted the SO moiety into its ester derivative, terminated the PeT process, and restored the fluorescence. Single molecule localization imaging was achieved using a dual functional illuminating beam in the visible, acting as both the activating and the exciting source. We successfully applied these photoactivatable probes for time‐lapse super‐resolution tracking in living cells and super‐resolution imaging of microtubule structures in neurons. Aryl azide and diaryl tetrazole are both photoactive molecules, which can form nitrene and nitrile imine intermediates respectively by photolysis. Depending on the new finding that the azide can suppress the photolysis of tetrazole in the azide‐tetrazole conjugated system, we developed aryl azide‐tetrazole probes for the photoactivatable fluorogenic azide alkyne click (PFAAC) reaction, in which the aryl azide‐tetrazole probes were not phoroactivatable fluorogenic itself, but the triazole products after click reaction were prefluorophore that can be activated by light. Therefore, in PFAAC chemistry, the fluorescent probes can be activated by two orthogonal events: azide‐alkyne click reaction and light, which leads to spatiotemporal resolution and high signal‐to‐noise ratio. This PFAAC process was proved in vitro by copper‐catalyzed or strain‐promoted azide‐alkyne reactions and in live cells by spatiotemporally controlled organelle imaging. By incorporation a linker to the azide‐tetrazole conjugate, this PFAAC chemistry could covalently label extra probes to the biomolecules and spatiotemporally detecting this process by photoinduced fluorescence. The borondipyrromethene (BODIPY) chromophore is a versatile platform for the construction of photoresponsive dyes with unique properties. Specifically, its covalent connection to a photocleavable group can be exploited to engineer compounds with photoswitchable fluorescence. The resulting photoactivatable fluorophores can increase their emission intensity or shift their emission wavelengths in response to switching. Such changes permit the spatiotemporal control of fluorescence with optical stimulations and the implementation of imaging strategies that would be impossible to replicate with conventional fluorophores. Indeed, BODIPYs with photoactivatable fluorescence enable the selective highlighting of intracellular targets, the nanoscaled visualization of sub‐cellular components, the real‐time monitoring of dynamic events and the photochemical writing of optical barcodes. Unlike amyloid aggregates, amorphous protein aggregates with no defined structures have been challenging to target and detect in a complex cellular milieu. In this study, we rationally designed sensors of amorphous protein aggregation from aggregation‐induced‐emission probes (AIEgens). Utilizing dicyanoisophorone as a model AIEgen scaffold, we first sensitized the fluorescence of AIEgens to a nonpolar and viscous environment mimicking the interior of amorphous aggregated proteins. We identified a generally applicable moiety (dimethylaminophenylene) for selective binding and fluorescence enhancement. Regulation of the electron‐withdrawing groups tuned the emission wavelength while retaining selective detection. Finally, we utilized the optimized probe to systematically image aggregated proteome upon proteostasis network regulation. Overall, we present a rational approach to develop amorphous protein aggregation sensors from AIEgens with controllable sensitivity, spectral coverage, and cellular performance. Designing a potential protein–ligand pair is pivotal, not only to track the protein structure dynamics, but also to assist in an atomistic understanding of drug delivery. Herein, the potential of a small model thioamide probe being used to study albumin proteins is reported. By monitoring the Förster resonance energy transfer (FRET) dynamics with the help of fluorescence spectroscopic techniques, a twofold enhancement in the FRET efficiency of 2‐thiopyridone (2TPY), relative to that of its amide analogue, is observed. Molecular dynamics simulations depict the relative position of the free energy minimum to be quite stable in the case of 2TPY through noncovalent interactions with sulfur, which help to enhance the FRET efficiency. Finally, its application is shown by pairing thiouracils with protein. It is found that the site‐selective sulfur atom substitution approach and noncovalent interactions with sulfur can substantially enhance the FRET efficiency, which could be a potential avenue to explore in the design of FRET probes to study the structure and dynamics of biomolecules. This Minireview summarizes the recent progress of stimuli‐responsive purely organic phosphorescence materials. Organic phosphorescence is closely related to the intermolecular interactions, because such interactions are beneficial to promote spin orbital coupling (SOC) and boost intersystem cross (ISC) efficiency and finally are conducive to satisfactory phosphorescence. It is found that the intermolecular interactions, which are essential for organic phosphorescence, are easily disturbed by external stimuli such as mechanical force, photon, acid, chemical vapor, leading to the luminescence change. According to this principle, various purely organic phosphorescence materials sensitive to external stimuli have been developed. This Minireview categorizes reported stimuli‐responsive purely organic phosphorescence materials on the basis of different stimuli, including mechanochromism, mechanoluminescence, photoactivity, acid‐responsiveness and other stimuli. According to classical measurement theory, fundamental measurement necessarily requires the operation of concatenation qua physical addition. Quantities which do not allow this operation are measurable only indirectly by means of derived measurement. Since only extensive quantities sustain the operation of physical addition, measurement in psychology has been considered problematic. In contrast, the theory of conjoint measurement, as developed in representational measurement theory, proposes that the operation of ordering is sufficient for establishing fundamental measurement. The validity of this view is questioned. The misconception about the advantages of conjoint measurement, it is argued, results from the failure to notice that magnitudes of derived quantities cannot be determined directly, i.e., without the help of associated quantitative indicators. A detailed description is provided for the optimization of heat treatment and calibrating procedure for Idaho National Laboratory’s high temperature irradiation resistant thermocouples (HTIR-TC). Also discussed is the implication of the procedures on the overall performance of the HTIR-TC finished product; in particular, for the case of long lead sections that are typical of sensors deployed in nuclear reactors. The effect on the measurement accuracy of fluctuations of the reference temperature and localized heating along the sensor lead is also investigated. A calibration graphical user interface (GUI) and accompanying script is presented for unifying the calibration process. A fifth-order polynomial function was found to be best for fitting HTIR-TC calibration data. The performance of the HTIR-TCs subject to the optimized heat treatment and calibration procedures is shown to be robust and consistent with other TC industry standards. Increased muscularity of small pulmonary vessels, involving enhanced proliferation and migration of pulmonary arterial smooth muscle cells (PASMCs), is a key component of the vascular remodeling underlying the development of pulmonary hypertension (PH). Stimuli such as growth factors and hypoxia induce PASMC alkalinization, proliferation, and migration through upregulation of the Na+/H+ exchanger (NHE), inhibition of which prevents the development of hypoxia-induced vascular remodeling and PH. We wanted to explore whether NHE was also necessary for pathologic PASMC proliferation and migration in a model of pulmonary arterial hypertension (PAH), a severe form of PH not associated with persistent hypoxia. PASMCs were isolated from rats exposed to SU5416-hypoxia (SuHx) followed by return to normoxia and from vehicle controls. We measured resting intracellular pH (pHi) and NHE activity using the pH-sensitive fluorescent dye BCECF-AM. PASMC proliferation and migration were assessed using BrdU incorporation and transwell filters, respectively. NHE activity was increased in SuHx PASMCs, although resting pHi was unchanged. SuHx PASMCs also exhibited increased proliferation and migration relative to controls, which was attenuated in the setting of pharmacologic inhibition of NHE. Our findings suggest that increased NHE activity contributes to pathologic PASMC function in the SuHx model of PAH, although this effect does not appear to be mediated by global changes in pHi homeostasis. Hypoxemia is seen in patients with pulmonary hypertension and hypoxic pulmonary vasoconstriction worsens their clinical condition. However, vasoconstriction is not the only aspect through which hypoxia induces the progression to pulmonary hypertension. Hypoxia‐inducible factor‐1α (HIF‐1α) is a transcription factor responding to hypoxic conditions by regulating hundreds of genes involved in angiogenesis, erythropoiesis, inflammation, and proliferation. We sought to determine the contribution of HIF‐1α in myeloid lineage cells to the pulmonary vascular response to chronic exposure to hypoxia. We generated myeloid‐specific HIF‐1α knockout (MyeHIF1KO) mice by using Cre‐lox P system, and exposed them to hypoxic conditions for 3 weeks to induce pulmonary hypertension. Macrophages from MyeHIF1KO and control mice were used for western blotting, RT‐qPCR, chemotaxis assay, and ATP assay. MyeHIF1KO mice exposed to hypoxia for 3 weeks exhibited a significant reduction in the right ventricular systolic pressure accompanied by a decrease in the ratio of the right ventricular weight to left ventricular weight, muscularization of the small pulmonary arteries, and infiltration of macrophages into the lung and right ventricle compared with control mice. HIF‐1α‐deficient peritoneal macrophages showed less migration toward monocyte chemoattractant protein‐1 and a decrease in intracellular ATP levels. These results indicate that HIF‐1α in macrophages contributes to the progression of pulmonary vascular remodeling and pulmonary hypertension induced by chronic exposure to hypoxic conditions. The inhibition of myeloid‐specific HIF‐1α may be a novel therapeutic strategy for the treatment of pulmonary hypertension. Accurate high temperature measurement has always been problematic for industry. The vast majority of industrial temperature measurements are performed with thermocouples. The development of any new thermocouple types requires the characterization of the relationship between thermocouple output and temperature, i.e. the reference function. An important thermocouple for measuring temperatures above about 1500 °C is the so-called Land–Jewell (Pt-40%Rh/Pt-20%Rh) thermocouple. This was used as a test thermocouple for a new European distributed facility for characterizing thermocouple reference function. The facility is described, and used to generate a demonstrator reference function for the thermocouple. It is found that the demonstrator reference function differs from the de-facto standard reference function of ASTM E1750-09 by several degrees above 1500 °C; a suggested explanation for this is given.
