Mistletoes are important co-contributors to tree mortality globally, particularly during droughts. In Australia, mistletoe distributions are expanding in temperate woodlands, while their hosts have experienced unprecedented heat and drought stress in recent years. We investigated whether the excessive water use of mistletoes increased the probability of xylem emboli in a mature woodland during the recent record drought that was compounded by multiple heatwaves. We continuously recorded transpiration ($T_{SLA}$) of infected and uninfected branches from two eucalypt species over two summers, monitored stem and leaf water potentials ($\Psi $) and used hydraulic vulnerability curves to estimate percent loss in conductivity (PLC) for each species. Variations in weather (vapor pressure deficit, photosynthetically active radiation, soil water content), host species and % mistletoe foliage explained 78% of hourly $T_{SLA}$. While mistletoe acted as an uncontrollable sink for water in the host even during typical summer days, daily $T_{SLA}$ increased up to 4-fold in infected branches on hot days, highlighting the previously overlooked importance of temperature stress in amplifying water loss in mistletoes. The increased water use of mistletoes resulted in significantly decreased host $\Psi _{\rm{leaf}}$ and $\Psi _{\rm{trunk}}$. It further translated to an estimated increase of up to 11% PLC for infected hosts, confirming greater hydraulic dysfunction of infected trees that place them at higher risk of hydraulic failure. However, uninfected branches of Eucalyptus fibrosa F.Muell. had much tighter controls on water loss than uninfected branches of Eucalyptus moluccana Roxb., which shifted the risk of hydraulic failure towards an increased risk of carbon starvation for E. fibrosa. The total solar eclipse of August 21, 2017 created a path of totality ~115 km in width across the United States. While eclipse observations have shown distinct responses in animal behavior often emulating nocturnal behavior, the influence of eclipses on plant physiology are less understood. We investigated physiological perturbations due to rapid changes of sunlight and air temperature in big sagebrush (Artemisia tridentata ssp. vaseyana), a desert shrub common within the path of eclipse totality. Leaf gas exchange, water potential, and chlorophyll a fluorescence were monitored during the eclipse and compared to responses obtained the day before in absence of the eclipse. On the day of the eclipse, air temperature decreased by 6.4 °C, coupled with a 1.0 kPa drop in vapor pressure deficit having a 9-minute lag following totality. Using chlorophyll a fluorescence measurements, we found photosynthetic efficiency of photosystem II (Fv'/Fm') recovered to near dark acclimated state (i.e., 87%), but the short duration of darkness did not allow for complete recovery. Gas exchange data and a simple light response model were used to estimate a 14% reduction in carbon assimilation for one day over sagebrush dominated areas within the path of totality for the Western United States.

Climate models predict widespread increases in both drought intensity and duration in the next decades. Although water deficiency is a significant determinant of plant survival, limited understanding of plant responses to extreme drought impedes forecasts of both forest and crop productivity under increasing aridity. Drought induces a suite of physiological responses; however, we lack an accurate mechanistic description of plant response to lethal drought that would improve predictive understanding of mortality under altered climate conditions. Here, proxies for leaf cellular damage, chlorophyll a fluorescence, and electrolyte leakage were directly associated with failure to recover from drought upon rewatering in Brassica rapa (genotype R500) and thus define the exact timing of drought-induced death. We validated our results using a second genotype (imb211) that differs substantially in life history traits. Our study demonstrates that whereas changes in carbon dynamics and water transport are critical indicators of drought stress, they can be unrelated to visible metrics of mortality, i.e. lack of meristematic activity and regrowth. In contrast, membrane failure at the cellular scale is the most proximate cause of death. This hypothesis was corroborated in two gymnosperms ( Picea engelmannii and Pinus contorta ) that experienced lethal water stress in the field and in laboratory conditions. We suggest that measurement of chlorophyll a fluorescence can be used to operationally define plant death arising from drought, and improved plant characterization can enhance surface model predictions of drought mortality and its consequences to ecosystem services at a global scale., (© 2017 American Society of Plant Biologists. Eddy covariance nighttime fluxes are uncertain due to potential measurement biases. Many studies report eddy covariance nighttime flux lower than flux from extrapolated chamber measurements, despite corrections for low turbulence. We compared eddy covariance and chamber estimates of ecosystem respiration at the GLEES Ameriflux site over seven growing seasons under high turbulence [summer night mean friction velocity (u*) = 0.7 m s(-1)], during which bark beetles killed or infested 85% of the aboveground respiring biomass. Chamber-based estimates of ecosystem respiration during the growth season, developed from foliage, wood, and soil CO2 efflux measurements, declined 35% after 85% of the forest basal area had been killed or impaired by bark beetles (from 7.1 ± 0.22 μmol m(-2) s(-1) in 2005 to 4.6 ± 0.16 μmol m(-2) s(-1) in 2011). Soil efflux remained at ~3.3 μmol m(-2) s(-1) throughout the mortality, while the loss of live wood and foliage and their respiration drove the decline of the chamber estimate. Eddy covariance estimates of fluxes at night remained constant over the same period, ~3.0 μmol m(-2) s(-1) for both 2005 (intact forest) and 2011 (85% basal area killed or impaired). Eddy covariance fluxes were lower than chamber estimates of ecosystem respiration (60% lower in 2005, and 32% in 2011), but the mean night estimates from the two techniques were correlated within a year (r(2) from 0.18 to 0.60). The difference between the two techniques was not the result of inadequate turbulence, because the results were robust to a u* filter of >0.7 m s(-1). The decline in the average seasonal difference between the two techniques was strongly correlated with overstory leaf area (r(2) = 0.92). Arid and semi-arid regions, which account for more than 30% of the Earth's land area, increasingly dominate the spatiotemporal trends in global carbon fluxes. The Horqin Sandy Land is a typical semi-arid fragile ecosystem in northern China. Understanding the components of the carbon budget in ecosystems under conditions of extreme soil moisture limitations provides a foundation for comprehending the carbon balance in semi-arid ecosystems. The seasonal and diurnal variations in soil respiration (Rs) in semi-mobile dune (SD) and meadow wetland (MW) ecosystems of the Horqin Sandy Land were examined, and the sources of CO2 emissions from Rs were identified using stable carbon isotopes. The responses of Rs and ecosystem respiration (Reco) to environmental temperature, moisture and leaf area index (LAI) were revealed. The results showed that on a seasonal scale, in SD with soil moisture content (Ms) below field capacity (FC), Ms had a greater influence on Rs than soil temperature (Ts) during the growing season. Changes in the LAI during the middle and late growth period affected Rs by altering root carbon supply. In MW, the most favorable Ms for Rs was near FC. The increase in LAI before mowing could effectively promote root and soil microbial respiration, and the decomposition of litter driven by Ts was the main form of Rs at this time. After mowing, root respiration and soil microbial respiration were the main processes contributing to CO2 emissions. On a daily scale, relative humidity (RH) dominated the Rs variation under dry conditions, whereas in other conditions, the Rs was adequately explained by temperature in SD and MW. The overall Reco was larger than Rs, but occasionally Rs was greater than Reco. The effects of temperature, moisture and LAI on Reco and Rs varied with growing season. Adding factors, such as ecosystem type, vegetation growth, water, and heat, to the carbon cycle model can improve predictions of carbon emissions, and aid in further management decisions in arid and semi-arid areas. Key message: Winter reddening of young Douglas-fir (Pseudotsuga menziesii Mirb. Franco), triggered by large thermal fluctuations in late winter, is a critical problem for European forestry. A literature review identified certain climatic conditions that are characteristic of 'reddening' years, including warm daily temperatures, high daily temperature amplitude, low relative humidity, moderate wind speeds, as well as the occurrence of freeze-thaw cycles with cold night temperatures. By describing the triggering environmental and stand factors, we propose three hypotheses for the physiological processes leading to winter reddening, namely (i) hydraulic failure due to winter drought stress, (ii) photo-oxidative stress in shade-acclimated trees, and (iii) early cold deacclimation during warm periods. i) Low soil temperature, by reducing root water uptake, combined with anticyclonic conditions, by increasing water losses, can induce hydraulic failure in the xylem. Hydraulic failure may be further accelerated by night frosts. ii) Winter reddening can occur when low temperature and high irradiance coincide, disrupting photostasis. Overwhelming of winter photo-protection may lead to photodamage and subsequent reddening. iii) Warm periods, by inducing cold deacclimation, make trees susceptible to frost damage. Finally, the three processes may interact under atypical anticyclonic conditions in late winter (e.g. cold or dry soils, warm days, high irradiance and/or freezing nights). Indeed, trees under water stress would develop a higher sensitivity to freezing night and photooxidative stress. Both mistletoes and their hosts are challenged by increasing drought, highlighting the necessity of understanding their comparative hydraulic properties. The high transpiration of mistletoes requires efficient water transport, while high xylem tensions demand strong embolism resistance, representing a hydraulic paradox. This study, conducted across four environments with different aridity indices in Yunnan, China, examined the xylem traits of 119 mistletoe-host species pairs. Mistletoes showed lower water use efficiency, indicating a more aggressive water use. They also showed lower hydraulic efficiency (lower vessel diameter and theoretical hydraulic conductivity) but higher safety (lower vulnerability index and higher conduit wall reinforcement, vessel grouping index, and wood density) compared with their hosts, supporting the trade-off between efficiency and safety. Environmental variation across sites significantly affected xylem trait comparisons between mistletoes and hosts. Additionally, the xylem traits of mistletoes were strongly influenced by host water supply efficiency. The overall xylem trait relationships in mistletoes and hosts were different. These findings stress the impact of host and site on the hydraulic traits of mistletoes, and suggest that mistletoes may achieve high transpiration by maintaining high stomatal conductance under low water potentials. Understanding the physiological and biochemical responses of tree seedlings under extreme drought stress, along with recovery during rewatering, and potential intra-species differences, will allow us to more accurately predict forest responses under future climate change. Here, we selected seedlings from four provenances (AH (Anhui), JX (Jiangxi), HN (Hunan) and GX (Guangxi)) of Schima superba and carried out a simulated drought-rewatering experiment in a field-based rain-out shelter. Seedlings were progressively dried until they reached 50% and 88% loss of xylem hydraulic conductivity (PLC) (i.e. P50 and P88), respectively, before they were rehydrated and maintained at field capacity for 30 days. Leaf photosynthesis (Asat), water status, activity of superoxide dismutase (SOD), and proline (Pro) concentration were monitored and their associations were determined. Increasing drought significantly reduced Asat, relative water content (RWC) and SOD activity in all provenances, and Pro concentration was increased to improve water retention; all four provenances exhibited similar response patterns, associated with similar leaf ultrastructure at pre-drought. Upon rewatering, physiological and biochemical traits were restored to well-watered control values in P50-stressed seedlings. In P88-stressed seedlings, Pro was restored to control values, while SOD was not fully recovered. The recovery pattern differed partially among provenances. There was a progression of recovery following watering, with RWC firstly recovered, followed by SOD and Pro, and then Asat, but with significant associations among these traits. Collectively, the intra-specific differences of S. superba seedlings in recovery of physiology and biochemistry following rewatering highlight the need to consider variations within a given tree species coping with future more frequent drought stress. Climate change and disturbance are altering forests and the rates and locations of tree regeneration. In semi-arid forests of the southwestern USA, limitations imposed by hot and dry conditions are likely to influence seedling survival. We examined how the survival of 1-year seedlings of five southwestern US conifer species whose southwestern distributions range from warmer and drier woodlands and forests (Pinus edulis Engelm. Pinus ponderosa Douglas ex C. Lawson) to cooler and wetter subalpine forests (Pseudotsuga menziesii (Mirb.) Franco , Abies concolor (Gord. & Glend.) Lindl. Ex Hildebr. and Picea engelmannii Parry ex Engelm.) changed in response to low moisture availability, high temperatures and high vapor pressure deficit in incubators. We used a Bayesian framework to construct discrete-time proportional hazard models that explained 55–75% of the species-specific survival variability. We applied these to the recent climate (1980–2019) of the southwestern USA as well as 1980–2099 CMIP5 climate projections with the RCP8.5 emissions pathway. We found that the more mesic species (i.e. P. menziesii , A. concolor and P. engelmannii) were more susceptible to the effects of hot and dry periods. However, their existing ranges are not projected to experience the conditions we tested as early in the 21st century as the more xeric P. edulis and P. ponderosa , leading to lower percentages of their existing ranges predicted to experience seedling-killing conditions. By late-century, extensive areas of each species southwestern range could experience climate conditions that increase the likelihood of seedling mortality. These results demonstrate that empirically derived physiological limitations can be used to inform where species composition or vegetation type change are likely to occur in the southwestern USA. A non-invasive method to estimate the number of Trypodendron lineatum holes on dead standing pines (Pinus sylvestris L.) was developed using linear and nonlinear estimations. A classical linear regression model was first used to analyze the relationship between the number of holes caused by T. lineatum on selected stem units and the total number of holes on an entire dead stem of P. sylvestris. Then, to obtain a better fit of the regression function to the data for the stem unit selected in the first step, piecewise linear regression (PLR) was used. Last, in an area used to evaluate wood decomposition (method validation), the total and mean numbers of T. lineatum holes were estimated for single dead trees and for a sample (n = 8 dead trees). Data were collected in 2009 (data set D1), in 2010–2014 (data set D2) and in 2020 (data set D3) in forests containing P. sylvestris located within Suchedniów–Oblęgorek Landscape Park, Poland. A model was constructed with three linear equations. An evaluation of model accuracy showed that it was highly effective regardless of the density of T. lineatum holes and sample size. Background: Survival rate (SR) is frequently used to compare drought tolerance among plant genotypes. While a variety of techniques for evaluating the stress status of plants under drought stress conditions have been developed, determining the critical point for the recovery irrigation to evaluate plant SR often relies directly on a qualitative inspection by the researcher or on the employment of complex and invasive techniques that invalidate the subsequent use of the tested individuals. Results: Here, we present a simple, instantaneous, and non-invasive method to estimate the survival probability of Arabidopsis thaliana plants after severe drought treatments. The quantum yield (QY), or efficiency of photosystem II, was monitored in darkness (Fv/Fm) and light (Fv'/Fm') conditions in the last phase of the drought treatment before recovery irrigation. We found a high correlation between a plant's Fv'/Fm' value before recovery irrigation and its survival phenotype seven days after, allowing us to establish a threshold between alive and dead plants in a calibration stage. This correlation was maintained in the Arabidopsis accessions Col-0, Ler-0, C24, and Kondara under the same conditions. Fv'/Fm' was then applied as a survival predictor to compare the drought tolerance of transgenic lines overexpressing the transcription factors ATAF1 and PLATZ1 with the Col-0 control. Conclusions: The results obtained in this work demonstrate that the chlorophyll a fluorescence parameter Fv'/Fm' can be used as a survival predictor that gives a numerical estimate of the Arabidopsis drought SR before recovery irrigation. The procedure employed to get the Fv'/Fm' measurements is fast, non-destructive, and requires inexpensive and easy-to-handle equipment. Fv'/Fm' as a survival predictor can be used to offer an overview of the photosynthetic state of the tested plants and determine more accurately the best timing for rewatering to assess the SR, especially when the symptoms of severe dehydration between genotypes are not contrasting enough to identify a difference visually. Extreme drought events during post-fire regeneration are becoming increasingly frequent in Mediterranean-type ecosystems. Understanding how plants with different traits and origins respond to such conditions during early life stages is therefore critical for assessing the effect of climate change. Here, seedlings of three Cistus (semi-deciduous malacophylls from the Mediterranean Basin) and three Ceanothus (evergreen sclerophylls from California) species, two post-fire seeder genera with contrasting leaf traits, were subjected to complete water deprivation for 3 months in a common garden experiment. The leaf and plant structure and plant tissue water relations were characterized before the drought, and the functional responses (water availability, gas exchange and fluorescence) were monitored during the drought. Both genera exhibited contrasting leaf structure and tissue water relations traits, with higher leaf area and specific leaf area as well as higher osmotic potential at maximum turgor and turgor loss point in Cistus than Ceanothus. During drought, Ceanothus showed a more conservative use of water than Cistus , with a water potential less sensitive to decreasing soil moisture and a strong decline in photosynthesis and stomatal conductance in response to water deficit, but also a level of fluorescence more responsive to drought than Cistus. However, we could not find a different degree of drought resistance between the genera. This was particularly clear between Cistus ladanifer L. and Ceanothus pauciflorus DC. the two most functionally contrasting species, but at the same time, the two most drought-resistant. Our findings demonstrate that species with different leaf traits and functional responses to water stress may not differ in their degree of drought resistance, at least during the seedling stage. This underlines the need to take general categorizations by genus or functional types with caution and to deepen our knowledge about the Mediterranean-type species ecophysiology, especially during early life stages, in order to anticipate their vulnerability to climate change. Summary: Nitrogen (N) fertilization increases biomass and soil organic carbon (SOC) accumulation in boreal pine forests, but the underlying mechanisms remain uncertain. At two Scots pine sites, one undergoing annual N fertilization and the other a reference, we sought to explain these responses.We measured component fluxes, including biomass production, SOC accumulation, and respiration, and summed them into carbon budgets. We compared the resulting summations to ecosystem fluxes measured by eddy covariance.N fertilization increased most component fluxes (P < 0.05), especially SOC accumulation (20×). Only fine‐root, mycorrhiza, and exudate production decreased, by 237 (SD = 28) g C m−2 yr−1. Stemwood production increases were ascribed to this partitioning shift, gross primary production (GPP), and carbon‐use efficiency, in that order. The methods agreed in their estimates of GPP in both stands (P > 0.05), but the components detected an increase in net ecosystem production (NEP) (190 (54) g C m−2 yr−1; P < 0.01) that eddy covariance did not (19 (62) g C m−2 yr−1; ns).The pairing of plots, the simplicity of the sites, and the strength of response provide a compelling description of N effects on the C budget. Summary: Water stress can cause declines in plant function that persist after rehydration. Recent work has defined 'resilience' traits characterizing leaf resistance to persistent damage from drought, but whether these traits predict resilience in whole‐plant function is unknown. It is also unknown whether the coordination between resilience and 'resistance' – the ability to maintain function during drought – observed globally occurs within ecosystems.For eight rainforest species, we dehydrated and subsequently rehydrated leaves, and measured water stress thresholds for declines in rehydration capacity and maximum quantum yield of photosystem II (Fv/Fm). We tested correlations with embolism resistance and dry season water potentials (ΨMD), and calculated safety margins for damage (ΨMD – thresholds) and tested correlations with drought resilience in sap flow and growth.Ψ thresholds for persistent declines in Fv/Fm, indicating resilience, were positively correlated with ΨMD and thresholds for leaf vein embolism. Safety margins for persistent declines in Fv/Fm, but not rehydration capacity, were positively correlated with drought resilience in sap flow.Correlations between resistance and resilience suggest that species' differences in performance during drought are perpetuated after drought, potentially accelerating shifts in forest composition. Measurements of resistance to embolism suggest that Cupressus sempervirens has a stem xylem that resists embolism at very negative water potentials, with 50% embolism (P 50 ) at water potentials of approximately -10 MPa. However, field observations in a semi-arid region suggest tree mortality occurs before 10% embolism. To explore the interplay between embolism and plant mortality, we conducted a controlled drought experiment involving two types of CS seedlings: a local seed source (S-type) and a drought-resistant clone propagated from a semi-arid forest (C-type). We measured resistance to embolism, leaf relative water content (RWC), water potential, photosynthesis, electrolyte leakage (EL), plant water loss, leaf hydraulic conductivity, and leaf non-structural carbohydrate (NSC) content during plant dehydration and before rewatering. All measured individuals were monitored for survival or mortality. While the S- and C-types differed in P 50 , transpiration, and mortality rates, both displayed seedling mortality corresponding to threshold values of 52-55% leaf RWC, 55% and 18.5% percent loss of conductivity (PLC) in the xylem, which corresponds to 48% and 37% average EL values for S and C types, respectively. Although C-type C. sempervirens NSC content increased in response to drought, no differences were observed in NSC content between live and dead seedlings of both types. Invasive alien species (IAS) threaten the biodiversity richness of the Mediterranean basin, a drought-prone region. However, our knowledge on the adaptive strategies of IAS for facing Mediterranean drought summers is still incomplete. The aim of the present study is to compare the water relations and the critical relative water content (RWC) values leading to loss of cell rehydration capacity of two Mediterranean basin IAS (i.e. Ailanthus altissima (Mill.) Swingle and Robinia pseudoacacia L.) versus two co-occurring native species (i.e. Fraxinus ornus L. and Quercus pubescens  Willd.). Study IAS showed higher values of water potential at turgor loss point and osmotic potential at full turgor, lower values of modulus of elasticity and leaf mass area but higher photosynthesis rate, even during the summer, with respect to the Mediterranean native species. These findings supported the hypothesis that IAS are characterized by a resource acquisitive strategy coupled with a safety-efficiency trade-off, compared with Mediterranean native species. However, similar leaf RWC thresholds leading to loss of cell rehydration capacity were recorded in the two groups of species. Moreover, IAS showed higher saturated water content and capacitance values compared with the co-occurring species. Overall, our results suggest that the success of Mediterranean IAS is driven by their ability to delay dehydration damage of mesophyll cells during Mediterranean summer drought, thereby supporting their distinctive high carbon assimilation rate. Summary: Xylem hydraulic failure (HF) has been identified as a ubiquitous factor in triggering drought‐induced tree mortality through the damage induced by the progressive dehydration of plant living cells. However, fundamental evidence of the mechanistic link connecting xylem HF to cell death has not been identified yet. The main aim of this study was to evaluate, at the leaf level, the relationship between loss of hydraulic function due to cavitation and cell death under drought conditions and discern how this relationship varied across species with contrasting resistances to cavitation.Drought was induced by withholding water from potted seedlings, and their leaves were sampled to measure their relative water content (RWC) and cell mortality. Vulnerability curves to cavitation at the leaf level were constructed for each species.An increment in cavitation events occurrence precedes the onset of cell mortality. A variation in cells tolerance to dehydration was observed along with the resistance to cavitation.Overall, our results indicate that the onset of cellular mortality occurs at lower RWC than the one for cavitation indicating the role of cavitation in triggering cellular death. Key message: Large-scale stem-girdling experiment reduced soil respiration for five consecutive years. Timing and magnitude of soil respiration declines are better explained by changes in leaf area rather than in soil microclimate. Using a decade of observations from the Forest accelerated succession experiment (FASET), we examined relationships among Rs, soil temperature, soil moisture, and leaf area index (LAI) following the stem girdling-induced mortality of 40% of all canopy trees within a 39-ha area. Mean annual Rs declined by about 20% relative to the control two years after disturbance, but recovered to near pre-disturbance values within five years; this reduction correlated with LAI losses and lower Rs temperature sensitivity (i.e., Q10), with the latter counteracting soil warming caused by partial canopy defoliation. These observations are consistent with progressive reductions in belowground labile C causing reductions in Rs. We conclude that the effects of stem girdling on Rs (1) were not immediate, occurring two years after the treatment, (2) were primarily influenced by biotic rather than soil microclimate changes, and (3) persisted for nearly a decade but were temporally dynamic, underscoring the value of long-term experiments. A specific, robust threshold for drought-induced tree mortality is needed to improve the prediction of forest dieback. Here, we tested the relevance of continuous measurements of stem diameter variations for identifying such a threshold, their relationship with hydraulic and cellular damage mechanisms, and the influence of growth conditions on these relationships. Poplar saplings were grown under well-watered, water-limited, or light-limited conditions and then submitted to a drought followed by rewatering. Stem diameter was continuously measured to investigate two parameters: the percentage loss of diameter (PLD) and the percentage of diameter recovery (DR) following rewatering. Water potentials, stomatal conductance, embolism, and electrolyte leakage were also measured, and light microscopy allowed investigating cell collapse induced by drought. The water release observed through loss of diameter occurred throughout the drought, regardless of growth conditions. Poplars did not recover from drought when PLD reached a threshold and this differed according to growth conditions but remained linked to cell resistance to damage and collapse. Background: This study aims to understand the influence of chlorophyll fluorescence parameters on yield of winter wheat in some areas of China. Nitrogen (N) application is believed to improve photosynthesis in flag leaf ultimately increase final yield. Methods and Results: To understand the response of chlorophyll fluorescence parameters of wheat, flag leaf and the effect of N fertilization was carried out at booting stage under greenhouse during year 2018-2019 using winter wheat cultivar "Yunhan-20410' 'Yunhan-618". The results showed that the maximum chlorophyll content of flag leaves occurred at booting stage. Under, Yunhan-20410 condition, maximum photochemical quantum efficiency (FV/Fm), potential activity (ΦPSII), potential activity of PSII (FV/FO), and photochemical quenching coefficient (qp) showed "high-low" variation, and the maximum values were observed between May 4 and May 12. However, Yunhan-20410 showed FV/Fm, FV/FO, and qp showed "low-high-low" curve at booting stage. Compared to Yunhan-618, Yunhan-20410 at booting stage significantly decreased FV/Fm, FV/FO, qp, and ΦPSII (P<0.05), and non-photochemical quenching (NPQ) significantly increased (P<0.05). Conclusion: The outcome of present investigation suggest that chlorophyll fluorescence parameters could be valuable insight to understand yield stability under stress condition. Investigating the responses of plant anatomical traits of trees to drought–rewatering cycles helps us to understand their responses to climate change; however, such work has not been adequately reported. In this study, Ginkgo biloba L. saplings were subjected to moderate, severe, extreme and lethal drought conditions by withholding water according to the percentage loss of hydraulic conductivity (PLC) and rewatering on a regular basis. Samples of phloem, cambium and xylem were collected to quantify their cellular properties including cambium and phloem cell vitality, xylem growth ring width, pit aspiration rates and pit membrane thickness using light microscopy and transmission microscopy. The results showed that the mortality rate of G. biloba saplings reached 90% at approximately P 88 (xylem water potential inducing 88% loss of hydraulic conductivity). The onset of cambium and phloem cell mortality might be in accordance with that of xylem embolism. Close negative correlations between xylem water potential and PLC and between xylem water potential and cambium and phloem mortality suggested that xylem hydraulic traits are coupled with anatomical traits under declining xylem water potential. Cambium and phloem cell vitality as well as xylem growth ring width decreased significantly with increasing drought conditions. However, xylem pit membrane thickness, cambial zone width and cambial cell geometry were not affected by the drought–rewatering cycles. The tracheid radial diameter, intertracheid cell wall thickness and tracheid density decreased significantly during both drought conditions and rewatering conditions. In addition to hydraulic traits, cambium and phloem cell vitality can be used as anatomical traits to evaluate the mortality of G. biloba under drought. Future work is proposed to observe the dynamics of pit aspiration rates under drought–rewatering cycles in situ to deepen our understanding of the essential role of bordered pits in the 'air-seeding' mechanism. Acoustic emission analysis is promising to investigate the physiological events leading to drought-induced injury and mortality. However, their nature and source are not fully understood, making this technique difficult to use as a direct measure of the loss of xylem hydraulic conductance. Acoustic emissions were recorded during severe dehydration in lavender plants (Lavandula angustifolia) and compared with the dynamics of embolism development and cell damage. The timing and characteristics of acoustic signals from two independent recording systems were compared by principal component analysis (PCA). Changes in water potential, branch diameter, loss of hydraulic conductance, and cellular damage were also measured to quantify drought-induced damages. Two distinct phases of acoustic emissions were observed during dehydration: the first one associated with a rapid loss of diameter and a significant increase in loss of xylem conductance (90%), and the second with slower changes in diameter and a significant increase in cellular damage. Based on PCA, a developed algorithm discriminated hydraulic-related acoustic signals from other sources, proposing a reconstruction of hydraulic vulnerability curves. Cellular damage preceded by hydraulic failure seems to lead to a lack of recovery. The second acoustic phase would allow detection of plant mortality. [ABSTRACT FROM AUTHOR]

Over their lifetime, trees are repeatedly exposed to droughts. It is therefore important to understand whether repeated drought exposure makes trees more or less drought tolerant. Here, we investigated the effect of repeated droughts on functional trait expression and tree function in Eucalyptus obliqua. Further, we tested whether previous drought exposure enabled trees to avoid leaf death for longer under a subsequent severe drought. Trees were subjected for 1 year to 2 drought–rewatering cycles (drought treatment) or well-watered conditions, before imposing a severe drought. Trees in the drought treatment reduced their overall leaf area and biomass, whereas leaf-level anatomical, morphological and physiological traits remained mostly unaffected. There were no differences in water potential at the turgor loss point, leaf xylem vulnerability to embolism, leaf size, maximum xylem vessel diameter or cell wall thickness between treatments after the conditioning period. When exposed to a subsequent severe drought, trees previously exposed to drought were more drought tolerant due to a lower water potential at leaf death and tree-level morphological rather than physiological adjustments. Trees previously exposed to drought were smaller and used less water, which delayed leaf death for 39 days compared with 22 days for the well-watered trees. Our study indicates that previous drought exposure can facilitate tree-level morphological adjustment, which potentially enhances survival of E. obliqua trees during subsequent drought events. Fusarium wilt caused by Fusarium oxysporum f. sp. lycopersici (Fol) is a major fungal disease of tomato (Solanum lycopersicum L.). Xylem sap protein 10 (XSP10) and Salicylic acid methyl transferase (SlSAMT) have been identified as putative negative regulatory genes associated with Fusarium wilt of tomato. Despite their importance as potential genes for developing Fusarium wilt disease tolerance, very little knowledge is available about their expression, cell biology, and functional genomics. Semi-quantitative and quantitative real-time PCR expression analysis of XSP10 and SlSAMT, in this study, revealed higher expression in root and flower tissue respectively in different tomato cultivars viz. Micro-Tom (MT), Arka Vikas (AV), and Arka Abhed (AA). Therefore, the highly up-regulated expression of XSP10 and SlSAMT in biotic stress susceptible tomato cultivar (AV) than a multiple disease resistant cultivar (AA) suggested the disease susceptibility nature of these genes for Fusarium wilt. Sub-cellular localization analysis through the expression of gateway cloning constructs in tomato protoplasts and seedlings showed the predominant localization of XSP10 in the nucleus and SlSAMT at the cytoplasm. A strong in vivo protein–protein interaction of XSP10 with SlSAMT at cytoplasm from bi-molecular fluorescent complementation study suggested that these two proteins function together in regulating responses to Fusarium wilt tolerance in tomato. Summary: Under prolonged drought and reduced photosynthesis, plants consume stored nonstructural carbohydrates (NSCs). Stored NSC depletion may impair the regulation of plant water balance, but the underlying mechanisms are poorly understood, and whether such mechanisms are independent of plant water deficit is not known. If so, carbon costs of fungal symbionts could indirectly influence plant drought tolerance through stored NSC depletion.We connected well‐watered Pinus ponderosa seedling pairs via ectomycorrhizal (EM) networks where one seedling was shaded (D) and the other kept illuminated (LD) and compared responses to seedling pairs in full light (L). We measured plant NSCs, osmotic and water potential, and transfer of 13CO2 through EM to explore mechanisms linking stored NSCs to plant water balance regulation and identify potential tradeoffs between plant water retention and EM fungi under carbon‐limiting conditions.NSCs decreased from L to LD to D seedlings. Even without drought, NSC depletion impaired osmoregulation and turgor maintenance, both of which are critical for drought tolerance. Importantly, EM networks propagated NSC depletion and its negative effects on water retention from carbon stressed to nonstressed hosts.We demonstrate that NSC storage depletion influences turgor maintenance independently of plant water deficit and reveal carbon allocation tradeoffs between supporting fungal symbionts and retaining water. This study investigates the isolated decomposition of spruce and lodgepole conifer needles to enhance our understanding of how needle litter impacts near-surface terrestrial biogeochemical processes. Harvested needles were exported to a subalpine meadow to enable a discrete analysis of the decomposition processes over 2 years. Initial chemistry revealed the lodgepole needles to be less recalcitrant with a lower carbon to nitrogen (C:N) ratio. Total C and N fundamentally shifted within needle species over time with decreased C:N ratios for spruce and increased ratios for lodgepole. Differences in chemistry correlated with CO2 production and soil microbial communities. The most pronounced trends were associated with lodgepole needles in comparison to the spruce and needle-free controls. Increased organic carbon and nitrogen concentrations associated with needle presence in soil extractions further corroborate the results with clear biogeochemical signatures in association with needle chemistry. Interestingly, no clear differentiation was observed as a function of bark beetle impacted spruce needles vs those derived from healthy spruce trees despite initial differences in needle chemistry. These results reveal that the inherent chemistry associated with tree species has a greater impact on soil biogeochemical signatures during isolated needle decomposition. By extension, biogeochemical shifts associated with bark beetle infestation are likely driven more by changes such as the cessation of rhizospheric processes than by needle litter decomposition. Once forests have achieved a full canopy, their growth rate declines progressively with age. This work used a global data set with estimates from a wide range of forest types, aged 20‒795 years, of their annual photosynthetic production (gross primary production, GPP) and subsequent above- plus below-ground biomass production (net primary production, NPP). Both GPP and NPP increased with increasing mean annual temperature and precipitation. GPP was then unrelated to forest age whilst NPP declined progressively with increasing age. These results implied that autotrophic respiration increases with age. It has been proposed that GPP should decline in response to increasing water stress in leaves as water is raised to greater heights as trees grow taller with age. However, trees may make substantial plastic adjustment in morphology and anatomy of newly developing leaves, xylem and fine roots to compensate for this stress and maintain GPP with age. This work reviews the possibilities that NPP declines with age as respiratory costs increase progressively in, any or all of, the construction and maintenance of more complex tissues, the maintenance of increasing amounts of live tissue within the sapwood of stems and coarse roots, the conversion of sapwood to heartwood, the increasing distance of phloem transport, increased turnover rates of fine roots, cost of supporting very tall trees that are unable to compensate fully for increased water stress in their canopies or maintaining alive competitively unsuccessful small trees. Widespread drought-induced forest mortality (DIM) is expected to increase with climate change and drought, and is expected to have major impacts on carbon and water cycles. For large-scale assessment and management, it is critical to identify variables that integrate the physiological mechanisms of DIM and signal risk of DIM. We tested whether plant water content, a variable that can be remotely sensed at large scales, is a useful indicator of DIM risk at the population level. We subjected Pinus ponderosa Douglas ex C. Lawson seedlings to experimental drought using a point of no return experimental design. Periodically during the drought, independent sets of seedlings were sampled to measure physiological state (volumetric water content (VWC), percent loss of conductivity (PLC) and non-structural carbohydrates) and to estimate population-level probability of mortality through re-watering. We show that plant VWC is a good predictor of population-level DIM risk and exhibits a threshold-type response that distinguishes plants at no risk from those at increasing risk of mortality. We also show that plant VWC integrates the mechanisms involved in individual tree death: hydraulic failure (PLC), carbon depletion across organs and their interaction. Summary: Given increasing water deficits across numerous ecosystems world‐wide, it is urgent to understand the sequence of failure of leaf function during dehydration.We assessed dehydration‐induced losses of rehydration capacity and maximum quantum yield of the photosystem II (Fv/Fm) in the leaves of 10 diverse angiosperm species, and tested when these occurred relative to turgor loss, declines of stomatal conductance gs, and hydraulic conductance Kleaf, including xylem and outside xylem pathways for the same study plants. We resolved the sequences of relative water content and leaf water potential Ψleaf thresholds of functional impairment.On average, losses of leaf rehydration capacity occurred at dehydration beyond 50% declines of gs, Kleaf and turgor loss point. Losses of Fv/Fm occurred after much stronger dehydration and were not recovered with leaf rehydration. Across species, tissue dehydration thresholds were intercorrelated, suggesting trait co‐selection. Thresholds for each type of functional decline were much less variable across species in terms of relative water content than Ψleaf.The stomatal and leaf hydraulic systems show early functional declines before cell integrity is lost. Summary: Drought‐induced tree mortality has major impacts on ecosystem carbon and water cycles, and is expected to increase in forests across the globe with climate change. A large body of research in the past decade has advanced our understanding of plant water and carbon relations under drought. However, despite intense research, we still lack generalizable, cross‐scale indicators of mortality risk. In this Viewpoint, we propose that a more explicit consideration of water pools could improve our ability to monitor and anticipate mortality risk. Specifically, we focus on the relative water content (RWC), a classic metric in plant water relations, as a potential indicator of mortality risk that is physiologically relevant and integrates different aspects related to hydraulics, stomatal responses and carbon economy under drought. Measures of plant water content are likely to have a strong mechanistic link with mortality and to be integrative, threshold‐prone and relatively easy to measure and monitor at large spatial scales, and may complement current mortality metrics based on water potential, loss of hydraulic conductivity and nonstructural carbohydrates. Dynamic process-based plant models capture complex physiological response across time, carrying the potential to extend simulations out to novel environments and lend mechanistic insight to observed phenotypes. Despite the translational opportunities for varietal crop improvement that could be unlocked by linking natural genetic variation to first principles-based modeling, these models are challenging to apply to large populations of related individuals. Here we use a combination of model development, experimental evaluation, and genomic prediction in Brassica rapa L. to set the stage for future large-scale process-based modeling of intraspecific variation. We develop a new canopy growth submodel for B. rapa within the process-based model Terrestrial Regional Ecosystem Exchange Simulator (TREES), test input parameters for feasibility of direct estimation with observed phenotypes across cultivated morphotypes and indirect estimation using genomic prediction on a recombinant inbred line population, and explore model performance on an in silico population under non-stressed and mild water-stressed conditions. We find evidence that the updated whole-plant model has the capacity to distill genotype by environment interaction (G×E) into tractable components. The framework presented offers a means to link genetic variation with environment-modulated plant response and serves as a stepping stone towards large-scale prediction of unphenotyped, genetically related individuals under untested environmental scenarios. In situ carbon flux studies are typically rare over the Himalaya but are important to understand carbon (C) balance. We investigated night-time canopy respiration (Rnc) and soil respiration (Rs) of a deciduous coniferous forest in response to environmental factors. A comprehensive investigation has been carried out on C balance indicators by employing systematic and concurrent measurements over an annual growth cycle of pine (Nov 2010-Dec 2011). The study site consists of uniformly distributed young deciduous Pinus roxburghii plantation having understory as Lantana camara (an invasive shrub). Results underlined that both Rnc and Rs fluxes were highest in the post-monsoon season. Evaporative fraction (EF) and temperature explained maximum variability of fluxes during warm-moist monsoon. Our key finding depicts an inverse significant correlation between day-time canopy photosynthesis (Ac) and Rnc across the seasons (r = 0.83-0.99). This can be explained by the mechanistic physiological phase of optimal anabolism (Ac) with favorable environmental conditions and minimum level of catabolism (Rnc). The respiration-photosynthesis ratio (Rnc/Ac) typically ranged from 0.25 ± 0.11 (peak growing season) to 0.71 ± 0.16 (winter season) with mean of 0.26 ± 0.10. The ratio Rs/Ac was highest during the winter season (2.69 ± 0.43), while minimum during peak growing season (0.64 ± 0.29). The Rnc/Ac ratio and night-time temperature (AT) also revealed that the ratio could increase when AT crossed 24 °C. These responses indicate that under climate warming, it may have a significant influence on net plant C uptake. Presence of understory shrub minimizes the Rnc/Ac ratio, and indicative of a more positive C-balance. Nevertheless, the observations could certainly lend useful insight into C-balance and ecological function in the region. Further, it may be useful in parameterizing and validating C-cycle models. The circadian clock is a molecular timekeeper that matches endogenous rhythms in diverse traits with 24-h cycles in the external environment. Although a lack of clock resonance to the environment is detrimental to performance, clock phenotypes in wild populations nevertheless deviate substantially from the predicted optimal cycle length of 24 h, and significant genetic variation exists for circadian parameters. Here, we describe covariation between 2 traits considered to reflect adaptation to different aspects of temporal environmental heterogeneity, circadian rhythms (adaptation to daily environmental cycles) and flowering time (adaptation to seasonal cycles), in a Rocky Mountain population of the mustard Boechera stricta, a North American relative of Arabidopsis thaliana. We found that 18 families that differ in circadian period in leaf movement by 3.5 h expressed genetic diversity in first-year growth, reproductive phenology, vegetative size at reproduction, and starch concentration following vernalization. The families exhibited a large (~90-day) range in mean flowering time, even though the spatial scale of population sampling covered only a few hundred meters. Circadian period covaried with other traits such that longer-period families flowered earlier and at a larger size, a trait combination predicted to yield a fitness benefit in the wild. Circadian clock research in model systems has previously shown that mutations in clock genes influence phenology. Our results widen the scope of this research by illustrating a link between naturally segregating clock variation and reproductive phenology among wild genotypes, suggesting that the causes of genetic diversity in the clock lie partly in adaptation to seasonal environmental heterogeneity. Background: An acceleration of model-data synthesis activities has leveraged many terrestrial carbon datasets, but utilization of soil respiration (R) data has not kept pace. Scope: We identify three major challenges in interpreting R data, and opportunities to utilize it more extensively and creatively: (1) When R is compared to ecosystem respiration (R) measured from EC towers, it is not uncommon to find R > R. We argue this is most likely due to difficulties in calculating R, which provides an opportunity to utilize R for EC quality control. (2) R integrates belowground heterotrophic and autotrophic activity, but many models include only an explicit heterotrophic output. Opportunities exist to use the total R flux for data assimilation and model benchmarking methods rather than less-certain partitioned fluxes. (3) R is generally measured at a very different resolution than that needed for comparison to EC or ecosystem- to global-scale models. Downscaling EC fluxes to match the scale of R, and improvement of R upscaling techniques will improve resolution challenges. Conclusions: R data can bring a range of benefits to model development, particularly with larger databases and improved data sharing protocols to make R data more robust and broadly available to the research community. Drought- and heat-driven tree mortality, along with associated insect outbreaks, have been observed globally in recent decades and are expected to increase in future climates. Despite its potential to profoundly alter ecosystem carbon and water cycles, how tree mortality scales up to ecosystem functions and fluxes is uncertain. We describe a framework for this scaling where the effects of mortality are a function of the mortality attributes, such as spatial clustering and functional role of the trees killed, and ecosystem properties, such as productivity and diversity. We draw upon remote-sensing data and ecosystem flux data to illustrate this framework and place climate-driven tree mortality in the context of other major disturbances. We find that emerging evidence suggests that climate-driven tree mortality impacts may be relatively small and recovery times are remarkably fast (~4 years for net ecosystem production). We review the key processes in ecosystem models necessary to simulate the effects of mortality on ecosystem fluxes and highlight key research gaps in modeling. Overall, our results highlight the key axes of variation needed for better monitoring and modeling of the impacts of tree mortality and provide a foundation for including climate-driven tree mortality in a disturbance framework. Background: The nonstructural carbohydrates (NSCs) of plants are posited to be crucial traits for the resistance and resilience of plants to climate change-induced drought and flooding. However, the potential effects of persistent drought and waterlogging on the dynamics of the NSCs and the underlying mechanisms are still poorly understood. Methods: We measured the NSCs concentrations and pool size, photosynthetic rate and biomass of Robinia pseudoacacia L. seedlings for both 2015 and 2016 under five soil water treatments: 3%, 8%, 17%, 24% and 34% soil water content, representing extreme drought (ED), moderate drought (MD), the control group (CG), field capacity (FC) and waterlogging (WL) stresses, respectively. We observed the relationship between the pool size of NSCs and the survival of seedlings under water stress (drought and waterlogging) for 30 days in greenhouse. Results: Compared with CG, the net photosynthetic rate decreased 91%, 67%, 34% and 71%, and the biomass decreased by 37%, 15%, 16% and 33% under ED, MD, FC and WL, respectively. The total NSC (TNSC) concentration was significantly increased by 154% under ED after 10 days and sharply decreased by 50% under ED after 30 days. The concentrations of soluble sugars (SS) were significantly increased by 100% under MD after 10 days and sharply decreased by 60% under ED after 30 days. Compared with GC, the response of NSCs, photosynthetic rate and biomass under ED were more dramatic than that under WL. The pool sizes of fructose and sucrose were larger under ED and WL, but the maximum pool size of starch occurred under the CG. The depletion of NSCs was not observed under ED at the end of the experiments in both 2015 and 2016. Conclusions: Our results indicate that the dynamics of NSCs is an important physiological feature of plant adaptation and resistance to drought and waterlogging. The relationship between leaf photosynthetic rate (A) in a vegetation canopy and the net ecosystem CO2 exchange (NEE) over an entire ecosystem is not well understood. The aim of the present study is to assess the coordinated changes in NEE derived with eddy covariance, A measured in leaf cuvette, and their associations in a rainfed maize field. The light response-curves were estimated for the carbon assimilation rate at both the leaf and ecosystem scales. NEE and A synchronically changed throughout the day and were greater around noon and persisted longer during rapid growth periods. The leaf A had a similar pattern of daytime changes in the top, middle, and bottom leaves. Only severe leaf ageing led to a significant decline in the maximum efficiency of photosystem II (PSII) photochemistry. The greater maximum NEE was associated with a higher ecosystem quantum yield. NEE was positively and significantly correlated with the leaf A averaged based on the vertical distribution of leaf area. The finding highlights the feasibility of assessing NEE by leaf CO2 exchange because of most of experimental data obtained with leaf cuvette methods; and also implies that simultaneously enhancing leaf photosynthetic rate, electron transport rate, net carbon assimilation at whole ecosystem might play a critical role for the enhancement of crop productivity.
