A one-pot multicomponent synthesis of unsymmetrical indole-substituted methane derivative based on two consecutive reactions of p-methylbenzaldehyde with N-methylindole and 5,5-dimethyl-1,3-cyclohexanedione is described as an experiment that was carried out by second-year undergraduate chemistry students individually in a laboratory classroom. The experiment provides an opportunity for students to carry out novel organic chemistry transformations for the synthesis of complex molecules from simple substrates, which is also the main pedagogical goal. Salient features of this teaching experiment include (a) multicomponent synthesis and good operability, (b) the absence of catalyst and any additives, (c) high yield and atomic economy, and (d) the use of inexpensive and green solvent. This laboratory experiment combines organic synthesis, TLC, NMR, and IR analysis. Several important concepts, such as green chemistry, chem- and regioselectivity, conjugated 1,4-addition, and atomic economy, are also discussed.

Abstract The advancement of Li-metal batteries is significantly impeded by the presence of unstable solid electrolyte interphase and Li dendrites upon cycling. Herein, we present an innovative approach to address these issues through the synergetic regulation of solid electrolyte interphase mechanics and Li crystallography using yttrium fluoride/polymethyl methacrylate composite layer. Specifically, we demonstrate the in-situ generation of Y-doped lithium metal through the reaction of composite layer with Li metal, which reduces the surface energy of the (200) plane, and tunes the preferential crystallographic orientation to (200) plane from conventional (110) plane during Li plating. These changes effectively passivate Li metal, thereby significantly reducing undesired side reactions between Li and electrolytes by 4 times. Meanwhile, the composite layer with suitable modulus (~1.02 GPa) can enhance mechanical stability and maintain structural stability of SEI. Consequently, a 4.2 Ah pouch cell with high energy density of 468 Wh kg−1 and remarkable capacity stability of 0.08% decay/cycle is demonstrated under harsh condition, such as high-areal-capacity cathode (6 mAh cm−2), lean electrolyte (1.98 g Ah−1), and high current density (3 mA cm−2). Our findings highlight the potential of reactive composite layer as a promising strategy for the development of stable Li-metal batteries.

BackgroundCuproptosis is a new form of cell death, which has great potential to be developed in tumors treatment. Our study aimed to explore the predictive value of cuproptosis-related genes (CRGs) in various cancers, with a focus on kidney renal clear cell carcinoma (KIRC).MethodA total of 9502 pan-cancer patients from TCGA cohort were enrolled. The relationships between CRGs and overall survival (OS) or disease-free survival (DFS) were analyzed. Gene Set Variation Analysis (GSVA) enrichment analysis was performed to explore the expression differences of CRGs. Multivariate Cox regression analysis was used to evaluate the association between GSVA scores and patient survival. KEGG and GO analyses were employed to identify the biological functions and pathways. The expression and prognostic characteristics of FDX1 were examined to evaluate the correlation between FDX1 and KIRC. Cell experiments were conducted to verify whether FDX1 was involved in cuproptosis of Caki-1 cells induced by Elesclomol.ResultsPositive cuproptosis signature genes(pos.cu.sig) exhibited the correlation with prognosis in KIRC, and all of these genes showed differential expression between KIRC and normal tissues. Manganese dioxide has been significantly utilized in zinc ion batteries (ZIBs). However, in the rechargeable battery system, the manganese dioxide cathode suffers from poor conductivity, volume expansion, and substance dissolution, resulting in low capacity and poor stability. Herein, a 3D frame structure MnO2@CNTs cathode is proposed. In this system, the electrodeposited spherical MnO2 is anchored and interlinked via the in‐situ growth carbon nanotubes (CNTs) onto the carbon cloth. Benefiting the unique 3D frame structure, the MnO2 structure crush problem and the pathway of the electrons and ions are dramatically improved. The optimized MnO2@CNTs cathode demonstrate a high capacity of 256.35 mAh g−1 at 0.1 A g−1 and exceptional cycling stability. Furthermore, in‐situ Raman spectroscopy elucidates the energy storage mechanism of aqueous ZIBs (AZIBs). Moreover, COMSOL finite elements analysis demonstrates that the petal edge‐rich nanostructures of MnO2@CNTs generate a localized high electric field under constant current, accelerating ion/electron transfer. This work explains the rationale for CNTs to improve the properties of MnO2 cathodes, providing a new perspective for the design of high‐performance batteries.

Abstract To detect circulating tumor cells (CTCs) in the peripheral blood of patients with tumor, and to analyze the significance of CTC detection in tumor diagnosis and monitoring. In the present study, peripheral blood was collected from 125 patients with tumor, and CTCs were isolated and identified. Differences in CTC number and subtype detection were analyzed for different tumor diseases and stages. CTCs were detected in 122 of the 125 patients with tumor, with a positive rate of 97.6%. The number of CTCs increases in patients with vascular metastasis. The number of mesenchymal CTCs increases in patients with lymph node or vascular metastasis. The average ratio of epithelial CTCs in each positive sample decreases in the later stages of cancer compared with the earlier stages, while the average ratio of mesenchymal CTCs increases in the later stages of cancer compared with the earlier stages. The results showed that CTCs with mesenchymal phenotypes are closely related to lymph node or vascular metastasis. CTC detection can help with early diagnosis of tumor diseases. Continuous monitoring of changes in CTCs number and subtypes can assist clinical judgment of tumor disease development status and prognosis.

Objective: To establish the method for content determination of the five phenolic acids ingredients of Phyllanthus emblica L. in different harvest time, and to investigate its variation rules so as to determine the best harvest time. Methods: The contents of Epigallocatechin, Corilagin, Scorpion Biphenyl acid, Ellagic acid and Scorpion acid of Phyllanthus emblica L. in different harvest time were determined by HPLC, and the total amount of accumulation of the five active ingredients was calculated. The chromatographic column was Inertsil ODS-3 C18, acetonitrile−0.1% phosphoric acid aqueous solution as mobile phase for gradient elution. A diode array detector was used with full-wavelength scanning. Flow rate was 1.0 mL·min−1. Injection was 10 μL. Used SPSS 24.0 software for principal component analysis, three principal components were selected and the comprehensive score was calculated. Results: The above five ingredients had a good linear relationship, and the correlation coefficients was more than 0.998, while the average recovery rates were 98.30%~101.22% and RSD were 0.43%~2.66%. There were some differences in the contents of the five phenolic acids ingredients in the leaves of Phyllanthus emblica L. during different harvesting periods. The contents of above five ingredients were 0.2317~1.5074, 1.9517~4.6593, 2.5105~6.3325, 2.2440~5.3386, 13.5570~39.1897 mg·g−1, respectively. The change in content of the five phenolic acids ingredients in different harvest time was: July>May>August>June>January>September>April>February>October>December>March>November. The total content was the highest in July, which was (55.158±0.83) mg/g and showed a dynamic trend of decreasing first and then increasing. Principal component analysis results showed that the month with the highest comprehensive score was July. Conclusion: Established method can be used to determine the contents of the five phenolic acids ingredients of Phyllanthus emblica L. in different harvest time. July was the best harvest time for Phyllanthus emblica L.

Abstract Lung adenocarcinoma (LUAD) is the predominant type of non-small lung cancer (NSCLC) with strong invasive ability and poor prognosis. The drug resistance related genes are potentially associated with prognosis of LUAD. Our research aimed to identify the drug resistance related genes and explore their potential prognostic value in LUAD patients. The data used in this study were obtained from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases. Firstly, we screened drug resistance related genes in LUAD by differential gene analysis, univariate Cox regression and drug sensitivity analyses. Subsequently, we constructed a risk score model using LASSO Cox regression analysis, and verified whether the risk score can predict the survival of LUAD patients independent of other factors. Moreover, we explored the immune infiltration of 22 immune cells between high-risk and low-risk patients. Totally 10 drug-resistance positively related genes (PLEK2, TFAP2A, KIF20A, S100P, GDF15, HSPB8, SASH1, WASF3, LAMA3 and TCN1) were identified in LUAD. The risk score model of LUAD constructed with these 10 genes could reliably predict the prognosis of LUAD patients. 18 pathways were significantly activated in high-risk group compared with low-risk group. In addition, the infiltration proportion of multiple immune cells was significantly different between high-risk and low-risk groups, and the proportion of M1 phagocytes was significantly higher in the high-risk group compared with the low-risk group. The drug resistance related genes (PLEK2, TFAP2A, KIF20A, S100P, GDF15, HSPB8, SASH1, WASF3, LAMA3 and TCN1) could predict the prognosis of LUAD patients. Clarifying the roles and mechanisms of these 10 genes in regulating drug resistance in LUAD will help to improve individualized clinical treatment protocols and predict patient sensitivity to treatment.

Morphology‐controlled nanostructures are important functional materials in multidisciplinary research fields, such as sensing, imaging, catalysis, and energy harvesting. However, existing nanofabrication methods have limitations in terms of controllability, diversity, cost, and efficiency. Here, a novel nanofabrication approach is proposed to construct Au nanostructures with high controllability, cost‐efficiency, and high‐performance sensing and energy harvesting. The fabrication incorporates electric field‐assisted hydrothermal growth of zinc oxide nanorods (ZnO NRs) with two‐step gold electroplating. The ZnO NRs serve as a dissolvable template during the electroplating formation of gold nanostructures for eventually synthesizing pure gold nanostructures on electrodes. The synthesized gold nanostructures take unique composite shape of nanospheres (hundreds of nanometers) covered with tinier nanostructures (tens of nanometers) like floccules, grains, lamellas, or rods, which are highly controlled by regulating electroplating parameters. The gold composite nanostructures provide high‐performance sensing and energy harvesting, such as enhanced electrochemical sensing interface (electric double‐layer impedance decrease up to 97.1%), excellent surface‐enhanced Raman scattering (SERS enhancement factor up to 5×107), and preferable visible–near‐infrared light harvesting (absorptivity near to 90%). For the first time, the application of tumor cell photothermal therapy with nanostructured electrodes and in situ bimodal monitoring of single‐cell impedance and SERS‐based molecular expression during therapy is demonstrated.

Ambrosia trifida is an invasive alien plant species, which has very high reproductive and environmental adaptability. Through strong resource acquisition ability and allelopathy, it could inhibit the growth and reproduction of surrounding plants and destroy the stability of an invasive ecosystem. It is very important to predict the change of suitable distribution area of A. trifida with climate change before implementing scientific control measures. Based on 106 A. trifida distribution data and 14 points of environmental data, the optimal parameter combination (RM = 0.1, FC = LQ) was obtained using the MaxEnt (version 3.4.1) model optimized by Kuenm package, and thus the potential suitable areas of A. trifida in Northeast China under three different climate scenarios (RCP2.6, RCP4.5, RCP8.5) with different emission intensities in the future (2050, 2070) were predicted. The changes of A. trifida suitable area in Northeast China under three climate scenarios were compared, and the relationship between the change of suitable area and emission intensity was analyzed. In general, the suitable area of A. trifida in Northeast China will expand gradually in the future, and the area of its highly suitable area will also increase with the increasing emission intensity, which is unfavorable to the control of A. trifida.

Abstract The June‐July Yangtze flooding in 1998 and 2020 drew incredible attention owing to the extreme precipitation events and devastating societal/economic damages. However, the quantitative estimation of the moisture transport mechanism is intensely discussed but still unresolved. Here we investigated two events from a unique perspective of Eulerian atmospheric water tracers that tries to explain the two events from model physics. The results showed that the moisture supplies from the Northwest Pacific decreased despite of different inducements, whereas the southwest summer monsoon (SWSM)‐related moisture supplies exhibited conspicuous enhancements in both events, suggesting that the SWSM‐related moisture supplies controlled the occurrence of Yangtze flooding. The physical processes of the two events were further compared. The 2020 flooding was more severe than the 1998 event, which was related to the stronger advective convergences and in‐stratus condensations of the SWSM‐related moisture.

Background A prerequisite for applying short tandem repeat (STR) kits is obtaining population allele and/or haplotype frequencies and forensic parameters. Aim Firstly, we aimed to investigate the population data of 19 X-chromosomal STRs (X-STRs) included in the AGCU X19 STR kit in the Han people residing in Hubei Province, Central China, and the Zhuang people residing in the Guangxi Zhuang Autonomous Region of South China. Furthermore, we compared these population data with those for other Chinese populations. Subjects and methods In total, 509 unrelated Han males and 266 unrelated Zhuang males were genotyped using the AGCU X19 STR kit. Allele frequencies, haplotype frequencies, and forensic parameters were computed, and genetic differences among 15 Chinese populations were analysed. Results The 19 X-STRs showed a high power of discrimination and high mean chance of exclusion, whether calculated using allele or haplotype frequencies. Major differences were found between Han and Oroqen, Uyghur, Mongolian, Tibetan, Li, and Yi populations. Aberrant biallelic patterns at DXS10159, DXS10134, and DXS10079 and allelic dropouts at DXS10164 were observed. Conclusion The 19 X-STRs were highly polymorphic in the Hubei Han and Guangxi Zhuang populations, and the AGCU X19 STR kit was shown to be suitable for forensic casework.

Gain-switching techniques offer a route to ultrafast lasing at the microscale. Here, 13 ps single-mode lasing from inorganic perovskite CsPbBr3 microwires is demonstrated and low transient saturated gain is identified as a significant limitation for further pulse shortening.

Coxsackievirus B3 (CVB3) was one of the most common pathogens to cause viral myocarditis. Circular RNAs as novel non-coding RNAs with a closed loop molecular structure have been confirmed to be involved in virus infectious diseases, but the function in CVB3 infection was not systematically studied. In this study, we identified that hsa_circ_0063331 (circDDX17) was drastically decreased after CVB3 infection by circRNA microarray. In vivo and in vitro, when cells or mice were infected with CVB3, the expression of circDDX17 was significantly reduced, as demonstrated by quantitative real-time PCR assays. Additionally, circDDX17 enhanced CVB3 replication by downregulating the expression of miR-1248 in HeLa and HL-1 cells, and miR-1248 regulated CVB3 replication through interacting with the gene coding for NOTCH Receptor 2 (NOTCH2), and NOTCH2 could upregulate methyltransferase-like protein 3 (METTL3). Taken together, this study suggested that circDDX17 promoted CVB3 replication and regulated NOTCH2 by targeting miR-1248 as a miRNAs sponge.

Based on the observational data of national ordinary stations in Jiangsu and Anhui, the live sounding data, combined with the reanalysis data of ERA5, the abnormal characteristics of Meiyu in Jianghuai region in 2020 are analyzed and a study on the causes of the Meiyu anomaly is carried out. The results show:1) the Meiyu period in 2020 is longer than usual, the rainfall is significantly more, and the heavy rain process is frequent with the coverage wide. The Meiyu rain belt is basically centered on 30°-32°N, and experience 4 continuous north-south oscillations about biweekly cycle in the range of 28°N to 34°N. 2) The stable subtropical high at low latitudes, the stable double resistance situation at high latitudes in the early stage and the multi shallow trough fluctuations in the late stage are the key influencing systems for the super Meiyu. The stable maintenance of low-level jet is the key factor to cause the continuous heavy rainfall in Meiyu period, and the excess water vapor come from the South China Sea. 3) When the heavy rainfall located in the north of the Jianghuai region, the center of the cold vortex is generally located in the west of 120°E, and moves eastward or slightly northward; when the cold vortex is located in the east or south of 120°E, the precipitation is forced to the South. 4) The subtropical high experience 4 biweekly oscillations and 9 volatile jumps to the North during the Meiyu period, with 12 cold vortex activities. During the period of the northward rising of the subtropical high and the southward and eastward moving of the cold vortex, the anti-phase overlap and prolong, which provide a good system configuration for the formation of the super long Meiyu and the maintenance of the North-South oscillation of the rain belt in the Jianghuai region.

An observed fact is proposed that the 2020 record-breaking Meiyu over the Yangtze–Huaihe River Basin (YHRB) is characteristic of significant subseasonal variation. The anomalously enhanced rainfall experiences a southward retreat from the northern YHRB in June to the southern YHRB in July. The meridional shift of abnormal rainbelt is closely related to the different anomalous Meiyu front circulations. The first stage features a warm Meiyu front with an extremely strong northward transport of warm-and-wet air, whereas a cold front forms with a convergence between the strong northeasterlies and southwesterlies in July. Another significant subseasonal variation of the 2020 Meiyu is that the Meiyu rainfall evolves in a periodic quasi-biweekly oscillation (QBWO). On the scale of the QBWO, the cold air activities over East Asia contribute significantly to Meiyu; each intense southward invasion of cold air corresponds well to the enhancement of Meiyu rainfall. However, the location of the western Pacific subtropical high, the subtropical East Asia summer monsoon (SEASM), and the intensity of the East Asia subtropical westerly jet exhibit opposite QBWO characteristics in June and July. For an increasing (decreasing) of Meiyu rainfall, the western Pacific subtropical high marches northward (retreats southward), the subtropical East Asia summer monsoon weakens (strengthens), and the East Asia subtropical westerly jet accelerates (decelerates) in June. The opposite holds true in July. Further analyses reveal that the filtered cyclone over Northeast China benefiting the southward intrusion of cold air is associated with the eastward propagation of a wave train over the mid-high latitudes in June, whereas it arises from both the westward propagation of a wave train over the mid-high latitudes and the northward propagation of cyclonic circulation from the tropics in July.

Based on 130 climate signal indexes provided by National Climate Center of China, this paper established a decision tree diagnostic prediction model for Spring Kuroshio Sea Surface Temperature (SST) from 1961 to 2015 (65 years) by using Chi-Squared Automatic Interaction Detector (CHAID) algorithm in data mining and obtained five rule sets to determine whether Spring Kuroshio SST is high or not. Considering the data of the 44 years from 1961 to 2004 as the training set of the model and the other years as the test set, the training accuracy of the model can reach to 95.45% and the test accuracy can reach to 81.82%. Three types of Spring Kuroshio SST are different in intensity and distribution. The results show that the prediction model of Spring Kuroshio SST based on CHAID algorithm has a high prediction accuracy, with the reasonable and effective model and the well-thought-out decision rules. Moreover, based on the results of decision classification, the SST anomalies correspond to different distribution characteristics of summer daily precipitation anomalies in eastern China, which can provide a new idea and method for climate prediction of regional summer precipitation.

Cultivated soybean (Glycine max (L.)), the world’s most important legume crop, has high-to-moderate salt sensitivity. Being the frontier for sensing and controlling solute transport, membrane proteins could be involved in cell signaling, osmoregulation, and stress-sensing mechanisms, but their roles in abiotic stresses are still largely unknown. By analyzing salt-induced membrane proteomic changes in the roots and leaves of salt-sensitive soybean cultivar (C08) seedlings germinated under NaCl, we detected 972 membrane proteins, with those present in both leaves and roots annotated as receptor kinases, calcium-sensing proteins, abscisic acid receptors, cation and anion channel proteins, proton pumps, amide and peptide transporters, and vesicle transport-related proteins etc. Endocytosis, linoleic acid metabolism, and fatty acid biosynthesis pathway-related proteins were enriched in roots whereas phagosome, spliceosome and soluble NSF attachment protein receptor (SNARE) interaction-related proteins were enriched in leaves. Using label-free quantitation, 129 differentially expressed membrane proteins were found in both tissues upon NaCl treatment. Additionally, the 140 NaCl-induced proteins identified in roots and 57 in leaves are vesicle-, mitochondrial-, and chloroplast-associated membrane proteins and those with functions related to ion transport, protein transport, ATP hydrolysis, protein folding, and receptor kinases, etc. Our proteomic results were verified against corresponding gene expression patterns from published C08 RNA-seq data, demonstrating the importance of solute transport and sensing in salt stress responses.

Abstract Background Non-small cell lung cancer (NSCLC) is one of the leading causes of cancer death in the world, and has a relatively low survival rate. Long non-coding RNAs (lncRNAs) have been demonstrated to modulate cancer progression through a variety of molecular mechanisms. We sought to investigate the role and potential mechanism of MYC-induced long non-coding RNA (MINCR) in NSCLC. Methods Expression levels of MINCR was first identified using The Cancer Genome Atlas (TCGA), further confirmed with specimens from 29 NSCLC patients and three cell lines using qRT-PCR. Overexpression and knockdown of MINCR were performed in NSCLC cell lines through MINCR overexpression vectors and synthesized siRNAs, respectively. The roles of MINCR in NSCLC cell lines, such as cell proliferation, cell cycle arrest, and apoptosis, were identified by MTT, flow cytometry, and Western blot. The modulation of MINCR-regulated genes, including c-Myc and its downstream effectors, as well as apoptosis-associated genes, was analyzed using Western blot. Results MINCR expression was increased in NSCLC patients from TCGA datasets, and was also significantly increased in our collected specimens from NSCLC patients and NSCLC cell lines. Knocking down of MINCR greatly inhibited the growth of NSCLC cell lines PC9 and A549. In addition, silencing of MINCR induced cell cycle arrest and apoptosis. Furthermore, silencing of MINCR reduced the expression levels of oncogene c-Myc and its downstream cyclin A, cyclin D, CD4, and CDK2, as well as apoptosis-associated Bcl-2, while significantly increased the expression levels of cleaved PARP-1. In the meantime, overexpression of MINCR remarkably enhanced cell proliferation of PC9 cells and activated c-Myc and its downstream effectors. Conclusion MINCR exerted inhibitory effects on the cell cycle arrest and apoptosis of NSCLC cells by activating c-Myc and its downstream effectors, suggesting that this lncRNA could be used as a potential therapeutic target for the treatment of NSCLC.

Under a declining trend of fog days in China, the duration of fog events since the 1990s reached a significant peak in the late autumn of 2018 over Eastern China. The average anomalous fog days were 4.74 d in November 2018 over Jiangsu Province in Eastern China, with a 1.73 standard deviation departure from climatology. Those fogs can thus be identified as a significantly abnormal climatic event with long duration, strong intensity, and extensive coverage. Based on the daily evolutions and correlations of atmospheric parameters, the dense fogs are revealed to be well configured by favorable metrological conditions such as weak dynamic progress, strong inversion in the lower troposphere and saturated air near the surface. If not disturbed, the intensification or duration of these conditions will further promote and maintain the development of fogs. The anomalous atmospheric background associated with those favorable meteorological conditions is revealed by composing the standardized anomalies of circulation fields during the fog days. Over the fog areas, vortex activities or cold air invasion is effectively hampered and the atmosphere inclines to be stable, due to the anomalous circulation pattern composed of the broadened jet stream, weakened jet core over Eastern China, undermined East Asian trough, declined East Asian winter monsoon, and enhanced anomalous southerly flows that transport abnormal warm and wet air to Eastern China. The vapor supplement is intensified by both sustained anomalous northward wind at the lower troposphere and anomalous westward wind in the near-surface. Overall, the numbers of standardized anomalies of 1000–200-hPa height, temperature, wind, and moisture fields during these fog days all significantly depart from climatology for that locale and time of the season, further demonstrating that the persistent dense fogs over Eastern China in the late autumn of 2018 is an unusual weather event with extreme synoptic-scale departures from normal.

In rice endosperms, the Waxy (Wx) gene is important for amylose synthesis, and various Wx alleles control the amylose content and affect the taste of cooked rice. Herein, we report the cloning of the ancestral allele Wxlv of the Wx locus, which affects the mouthfeel of rice grains by modulating the size of amylose molecules. Using evolutionary analysis, we demonstrated that Wxlv originated directly from wild rice, and the three major Wx alleles in cultivated rice (Wxb, Wxa, and Wxin) differentiated after the substitution of one base pair at the functional sites. These data indicate that the Wxlv allele played an important role in artificial selection and domestication. The findings also shed light on the evolution of various Wx alleles, which have greatly contributed to improving the eating and cooking quality of rice.
