Anthocyanins are the most valuable pigments in Lycium ruthenicum Murray (L. ruthenicum). Although ultraviolet-B (UV-B) irradiation is a key environmental factor influencing anthocyanin biosynthesis in L. ruthenicum, the deep molecular mechanism remains unclear. Herein, we examined the changes in the total anthocyanin content and transcriptomic characteristics of L. ruthenicum leaves following UV-B irradiation treatment. The results showed a twofold increase in anthocyanin content in the leaves of L. ruthenicum after the treatment. The transcriptome analysis showed that the expression of 24 structural genes identified in the anthocyanin synthesis pathway was up-regulated. In particular, F3'H (Unigene0009145) and C4H (Unigene0046607) exhibit notable up-regulation, suggesting their potential roles in anthocyanin synthesis. Protein interaction network results revealed that MYB1 (Unigene0047706) had the highest connectivity, followed by bHLH (Unigene0014085). Additionally, UVR8 (Unigene0067978) and COP1 (Unigene0008780) were found to be highly involved in UV-B signal transduction. These findings provide new insights into the genetic and biochemical mechanisms that regulate anthocyanin production, and could guide agricultural practices to reduce environmental impacts and improve crop yield and quality. Photodynamic diagnosis and therapy (PDD and PDT) are emerging techniques for diagnosing and treating tumors and malignant diseases. Photoproducts of protoporphyrin IX (PpIX) used in PDD and PDT may be used in the diagnosis and treatment, making a detailed analysis of the photoproduct formation under various treatment and diagnosis conditions important. Spectroscopic and mass spectrometric analysis of photoproduct formation from PpIX dissolved in dimethyl sulfoxide were performed under commonly used irradiation conditions for PDD and PDT, i.e., wavelengths of 405 and 635 nm and fluence rates of 10 and 100 mW/cm2. Irradiation resulted in the formation of hydroxyaldehyde photoproduct (photoprotoporphyrin; Ppp) and formyl photoproduct (product II; Pp II) existing in different quantities with the irradiation wavelength and fluence rate. Ppp was dominant under 635 nm irradiation of PpIX, with a fluorescence peak at 673 nm and a protonated monoisotopic peak at m/z 595.3. PpIX irradiation with 405 nm yielded more Pp II, with a fluorescence peak at 654 nm. A higher photoproduct formation was observed at a low fluence rate for irradiation with 635 nm, while irradiation with 405 nm indicated a higher photoproduct formation at a higher fluence rate. The photoproduct formation with the irradiation conditions can be exploited for dosimetry estimation and may be used as an additional photosensitizer to improve the diagnostics and treatment efficacies of PDD and PDT. Differences in environmental conditions of the present study from that of a biological environment may result in a variation in the photoproduct formation rate and may limit their clinical utilization in PDD and PDT. Thus, further investigation of photoproduct formation rates in more complex biological environments, including in vivo, is necessary. Because of their sessile life style, plants have evolved the ability to adjust to environmentally harsh conditions. An important aspect of stress adaptation involves the reprogramming of the cell cycle to ensure optimal growth. The atypical E2F transcription factor DP-E2F-like 1 (E2Fe/DEL1) had been found previously to be an important regulator of the endocycle onset. Here, a novel role for E2Fe/DEL1 was identified as a transcriptional repressor of the type-II cyclobutane pyrimidine dimer-photolyase DNA repair gene PHR1. Upon ultraviolet-B (UV-B) treatment, plants knocked out for E2Fe/DEL1 had improved DNA repair abilities when compared with control plants, whereas those overexpressing it performed less well. Better DNA repair allowed E2Fe/DEL1 knockout plants to resume endoreduplication faster than control plants, contributing in this manner to UV-B radiation resistance by compensating the stress-induced reduction in cell number by ploidy-dependent cell growth. As E2Fe/DEL1 levels decreased upon UV-B treatment, we hypothesize that the coordinated transcriptional induction of PHR1 with the endoreduplication onset contributes to the adaptation of plants exposed to UV-B stress.

Plants respond to low levels of UV-B radiation with a coordinated photomorphogenic response that allows acclimation to this environmental stress factor. The key players in this UV-B response are COP1 (an E3 ubiquitin ligase), UVR8 (a β-propeller protein), and HY5 (a bZIP transcription factor). We have shown previously that an elevated UV-B-specific response is associated with dwarf growth, indicating the importance of balancing UV-B-specific signaling. Negative regulators of this pathway are not known, however. Here, we describe two highly related WD40-repeat proteins, REPRESSOR OF UV-B PHOTOMORPHOGENESIS 1 (RUP1) and RUP2, that interact directly with UVR8 as potent repressors of UV-B signaling. Both genes were transcriptionally activated by UV-B in a COP1-, UVR8-, and HY5-dependent manner. rup1 rup2 double mutants showed an enhanced response to UV-B and elevated UV-B tolerance after acclimation. Overexpression of RUP2 resulted in reduced UV-B-induced photomorphogenesis and impaired acclimation, leading to hypersensitivity to UV-B stress. These results are consistent with an important regulatory role for RUP1 and RUP2, which act downstream of UVR8-COP1 in a negative feedback loop impinging on UVR8 function, balancing UV-B defense measures and plant growth.

Long-term effects of ultraviolet (UV) radiation on flavonoid biosynthesis were investigated in Arabidopsis thaliana using the sun simulators of the Helmholtz Zentrum München. The plants, which are widely used as a model system, were grown (1) at high photosynthetically active radiation (PAR; 1,310 micromol m(-2) s(-1)) and high biologically effective UV irradiation (UV-B(BE) 180 mW m(-2)) during a whole vegetative growth period. Under this irradiation regime, the levels of quercetin products were distinctively elevated with increasing UV-B irradiance. (2) Cultivation at high PAR (1,270 micromol m(-2) s(-1)) and low UV-B (UV-B(BE) 25 mW m(-2)) resulted in somewhat lower levels of quercetin products compared to the high-UV-B(BE) conditions, and only a slight increase with increasing UV-B irradiance was observed. On the other hand, when the plants were grown (3) at low PAR (540 micromol m(-2) s(-1)) and high UV-B (UV-B(BE) 180 mW m(-2)), the accumulation of quercetin products strongly increased from very low levels with increasing amounts of UV-B but the accumulation of kaempferol derivatives and sinapoyl glucose was less pronounced. We conclude (4) that the accumulation of quercetin products triggered by PAR leads to a basic UV protection that is further increased by UV-B radiation. Based on our data, (5) a combined effect of PAR and different spectral sections of UV radiation is satisfactorily described by a biological weighting function, which again emphasizes the additional role of UV-A (315-400 nm) in UV action on A. thaliana.

Plants in alpine habitats are exposed to many environmental stresses, in particular temperature and radiation extremes. Recent field experiments on Arnica montana L. cv. ARBO indicated pronounced altitudinal variation in plant phenolics. Ortho-diphenolics increased with altitude compared to other phenolic compounds, resulting in an increase in antioxidative capacity of the tissues involved. Factors causing these variations were investigated by climate chamber (CC) experiments focusing on temperature and ultraviolet (UV)-B radiation. Plants of A. montana L. cv. ARBO were grown in CCs under realistic climatic and radiation regimes. Key factors temperature and UV-B radiation were altered between different groups of plants. Subsequently, flowering heads were analyzed by HPLC for their contents of flavonoids and caffeic acid derivatives. Surprisingly, increased UV-B radiation did not trigger any change in phenolic metabolites in Arnica. In contrast, a pronounced increase in the ratio of B-ring ortho-diphenolic (quercetin) compared to B-ring monophenolic (kaempferol) flavonols resulted from a decrease in temperature by 5 degrees C in the applied climate regime. In conclusion, enhanced UV-B radiation is probably not the key factor triggering shifts in the phenolic composition in Arnica grown at higher altitudes but rather temperature, which decreases with altitude.

The impact of UV-B radiation on 10 genotypically different barley and tomato cultivars was tested in a predictive study to screen for potentially UV-tolerant accessions and to analyze underlying mechanisms for UV-B sensitivity. Plant response was analyzed by measuring thermoluminescence, fluorescence, gas exchange and antioxidant status. Generally, barley cultivars proved to be much more sensitive against UV-B radiation than tomato cultivars. Statistical cluster analysis could resolve two barley groups with distinct differences in reaction patterns. The UV-B sensitive group showed a stronger loss in PSII photochemistry and a lower gas-exchange performance and regulation after UV-B radiation compared to the more tolerant group. The results indicate that photosynthetic light and dark reactions have to play optimally in concert to render plants more tolerant against UV-B radiation. Hence, measuring thermoluminescence/fluorescence and gas exchange in parallel will have much higher potential in identifying tolerant cultivars and will help to understand the underlying mechanisms.

The ultraviolet-B (UV-B) portion of the solar radiation functions as an environmental signal for which plants have evolved specific and sensitive UV-B perception systems. The UV-B-specific UV RESPONSE LOCUS 8 (UVR8) and the multifunctional E3 ubiquitin ligase CONSTITUTIVELY PHOTOMORPHOGENIC 1 (COP1) are key regulators of the UV-B response. We show here that uvr8-null mutants are deficient in UV-B-induced photomorphogenesis and hypersensitive to UV-B stress, whereas overexpression of UVR8 results in enhanced UV-B photomorphogenesis, acclimation and tolerance to UV-B stress. By using sun simulators, we provide evidence at the physiological level that UV-B acclimation mediated by the UV-B-specific photoregulatory pathway is indeed required for survival in sunlight. At the molecular level, we demonstrate that the wild type but not the mutant UVR8 and COP1 proteins directly interact in a UV-B-dependent, rapid manner in planta. These data collectively suggest that UV-B-specific interaction of COP1 and UVR8 in the nucleus is a very early step in signalling and responsible for the plant's coordinated response to UV-B ensuring UV-B acclimation and protection in the natural environment.

There is limited information on the impact of present-day ultraviolet-B (UV-B) radiation on a reprogramming of gene expression in crops. Summer wheat was cultivated in controlled environmental facilities under simulated realistic climatic conditions. We investigated the effect of different regimes of UV-B radiation on summer wheat (Triticum aestivum L.) cultivars Nandu, Star and Turbo. Until recently, these were most important in Bavaria. Different cultivars of crops often show great differences in their sensitivity towards UV-B radiation. To identify genes that might be involved in UV-B defence mechanisms, we first analyzed selected genes known to be involved in plant defence mechanisms. RNA gel blot analysis of RNA isolated from the flag leaf of 84-day-old plants showed differences in transcript levels among the cultivars. Flag leaves are known to be important for grain development, which was completed at 84 days post-anthesis. Catalase 2 (Cat2) transcripts were elevated by increased UV irradiation in all cultivars with highest levels in cv. Nandu. Pathogenesis-related protein 1 (PR1) transcripts were elevated only in cv. Star. A minor influence on transcripts for phenylalanine ammonia-lyase (PAL) was observed in all three cultivars. This indicates different levels of acclimation to UV-B radiation in the wheat cultivars studied. To analyze these responses in more detail, UV-B-exposed flag leaves of 84-day-old wheat (cv. Nandu) were pooled to isolate cDNAs of induced genes by suppression-subtractive hybridization (SSH). Among the initially isolated cDNA clones, 13 were verified by RNA gel blot analysis showing an up-regulation at elevated levels of UV-B radiation. Functional classification revealed genes encoding proteins associated with protein assembly, chaperonins, programmed cell death and signal transduction. We also studied growth, flowering time, ear development and yield as more typical agricultural parameters. Plant growth of young plants was reduced at increased UV-B radiation. Flowering and ear development were delayed concomitantly, whereas total grain weight was not influenced at any of the UV-B irradiation regimes.

UV-B radiation can substantially impact plant growth. To study UV-B effects, broadband UV-B tubes are commonly used. Apart from UV-B, such tubes also emit UV-A wavelengths. This study aimed to distinguish effects of different UV-B intensities on Arabidopsis thaliana wildtype and UVR8 mutant rosette morphology, from those by accompanying UV-A. UV-A promotes leaf-blade expansion along the proximal–distal, but not the medio-lateral, axis. Consequent increases in blade length: width ratio are associated with increased light capture. However, petiole length is not affected by UV-A exposure. This scenario is distinct from the shade avoidance driven by low red to far-red ratios, whereby leaf blade elongation is impeded but petiole elongation is promoted. Thus, the UV-A mediated elongation response is phenotypically distinct from classical shade avoidance. UV-B exerts inhibitory effects on petiole length, blade length and leaf area, and these effects are mediated by UVR8. Thus, UV-B antagonises aspects of both UV-A mediated elongation and classical shade avoidance. Indeed, this study shows that accompanying UV-A wavelengths can mask effects of UV-B. This may lead to potential underestimates of the magnitude of the UV-B induced morphological response using broadband UV-B tubes. [ABSTRACT FROM AUTHOR]

The major UV-B screening pigments of the epidermal layer of Scots pine (Pinus sylvestris) needles are flavonol 3-o-glycosides (F3Gs) esterified with hydroxycinnamic acids at positions 3" and 6". Acylation is the last step in biosynthesis and is catalysed by position-specific hydroxycinnamoyl transferases (3" and 6"HCT). The UV-B dependence of these enzyme activities was studied in primary needles of Scots pine seedlings grown under different UV-B conditions in environmentally controlled sun simulators. 6"HCT activity was induced upon UV-B irradiation while 3"HCT activity was not induced but showed high constitutive values. To investigate the biosynthesis of diacylated F3Gs during needle development under natural conditions, the HCT activities and metabolite contents were analysed in needles of field-grown mature pine trees. Accumulation of diacylated compounds as well as of 6"HCT activity occurred transiently in the first year of needle development only. In contrast, 3"HCT activity exhibited broad maxima in two consecutive years during needle growth. The data suggest that acylated F3Gs are first formed as soluble compounds which are then translocated into the cell wall to be bound by their hydroxycinnamoyl residues.

Polyamines have been described to protect against numerous oxidative stresses in plants. Increasing UV-B radiation (280-315 nm) in the biosphere may also induce an increase in radical formation in tissues. This study employed the tobacco cultivars Bel B and Bel W3 to describe possible protective functions of polyamines against UV-B radiation in sun light simulators (GSF/Munich) with natural diurnal fluctuations of simulated UV-B. Polyamine measurements on a whole leaf basis in isolated chloroplasts and thylakoids were paralleled to photosynthetic and respiration rates, photosynthetic efficiency, leaf thickness and photosynthetic pigment compositions. The study revealed that an increase of polyamines, and especially of putrescine level in thylakoid membranes upon elevated UV-B exposure comprises one of the primary protective mechanisms in the photosynthetic apparatus of the tobacco variety Bel B against UV-B radiation. The tobacco cultivar Bel W3, sensitive to ozone, was also proved to be sensitive to UV-B. This sensitivity is attributed to its incapability to enhance putrescine level in thylakoid membranes. After prolongation of UV-B exposure, when endogenous plant balances are being gradually restored, due to secondary responses, (e.g., biosynthesis of carotenoids and of additional flavonoids) and the plant is adapting to the altered environmental conditions, then the polyamine level is being reduced. Thus, we can discriminate the UV-B induced stress period from a UV-B acclimation period.

In plants, hydrogen peroxide (H(2)O(2)) plays a major signaling role in triggering both a defense response and cell death. Increased cellular H(2)O(2) levels and subsequent redox imbalances are managed at the production and scavenging levels. Because catalases are the major H(2)O(2) scavengers that remove the bulk of cellular H(2)O(2), altering their levels allows in planta modulation of H(2)O(2) concentrations. Reduced peroxisomal catalase activity increased sensitivity toward both ozone and photorespiratory H(2)O(2)-induced cell death in transgenic catalase-deficient Arabidopsis thaliana. These plants were used as a model system to build a comprehensive inventory of transcriptomic variations, which were triggered by photorespiratory H(2)O(2) induced by high-light (HL) irradiance. In addition to an H(2)O(2)-dependent and -independent type of transcriptional response during light stress, microarray analysis on both control and transgenic catalase-deficient plants, exposed to 0, 3, 8, and 23 h of HL, revealed several specific regulatory patterns of gene expression. Thus, photorespiratory H(2)O(2) has a direct impact on transcriptional programs in plants.

Photodegradation of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) and polycyclic aromatic hydrocarbons (PAHs) sorbed on surfaces of spruce (Picea abies (L.) Karst.) needles was investigated. The role of sunlight and, in particular, the role of short wavelength UV-B radiation were studied in a sun simulator under controlled environmental conditions. The photodegradation of PCDD/Fs and PAHs follows first-order kinetics. Simulated sunlight containing UV-B induced two phases of photodegradation kinetics for PCDD/Fs, whereas no distinct phases of the kinetics were observed for PAHs. UV-B radiation plays a much more important role on the photodegradation for PCDD/Fs than that for PAHs. Direct photolysis dominates the photodegradation of PCDD/Fs and PAHs.

Plant secondary metabolism significantly contributes to defensive measures against adverse abiotic and biotic cues. To investigate stress-induced, transcriptional alterations of underlying effector gene families, which encode enzymes acting consecutively in secondary metabolism and defense reactions, a DNA array (MetArray) harboring gene-specific probes was established. It comprised complete sets of genes encoding 109 secondary product glycosyltransferases and 63 glutathione-utilizing enzymes along with 62 cytochrome P450 monooxygenases and 26 ABC transporters. Their transcriptome was monitored in different organs of unstressed plants and in shoots in response to herbicides, UV-B radiation, endogenous stress hormones, and pathogen infection. A principal component analysis based on the transcription of these effector gene families defined distinct responses and crosstalk. Methyl jasmonate and ethylene treatments were separated from a group combining reactions towards two sulfonylurea herbicides, salicylate and an avirulent strain of Pseudomonas syringae pv. tomato . The responses to the herbicide bromoxynil and UV-B radiation were distinct from both groups. In addition, these analyses pinpointed individual effector genes indicating their role in these stress responses. A small group of genes was diagnostic in differentiating the response to two herbicide classes used. Interestingly, a subset of genes induced by P. syringae was not responsive to the applied stress hormones. Small groups of comprehensively induced effector genes indicate common defense strategies. Furthermore, homologous members within branches of these effector gene families displayed differential expression patterns either in both organs or during stress responses arguing for their non-redundant functions.

Polysulphone film is used as a personal UV dosemeter in dermatological or epidemiological studies. The relative efficiency of this detector does not exactly match the action spectrum as proposed by the International Commission on Non-Ionizing Radiation Protection (ICNIRP) and to which the UV dose and exposure limits refer. Therefore, the calibration of the dosemeter depends on the spetrum. In the present paper the variation of the calibration factor for terrestrial solar UV spectra is analysed on the basis of a two year observation period at a site near Munich. Germany. A detailed error estimation is included. It is shown that the variation of the calibration factor within this class of spectra is the main contribution to the total uncertainty of the dose determination, which can be up to 40%. The shape of the spectrum of terrestrial solar UV radiation is mainly determined by the total ozone column and the solar elevation angle. It is shown how the calibration depends on these two parameters and how this additional information can help to reduce the measurement error to a residual uncertainty of 17%. Exposure studies of terrestrial solar UV radiation using polysulphone film as a dosemeter would gain in accuracy if total ozone column values at the study's site could be measured or taken from satellite or weather service data. The interpretation of the magnitude of the dose uncertainty depends on the further use of these data.

Long-term depletion of the stratospheric ozone layer contributes to an increase in terrestrial solar ultraviolet-B radiation. This has deleterious effects on living organisms, such as DNA damage. When exposed to elevated ultraviolet-B radiation (UV-B; 280-315 nm), plants display a wide variety of physiological and morphological responses characterized as acclimation and adaptation. Here we show, using special sun simulators, that elevated solar UV-B doses increase the frequency of somatic homologous DNA rearrangements in Arabidopsis and tobacco plants. Increases in recombination are accompanied by a strong induction of photolyase and Rad51 gene expression. These genes are putatively involved in major DNA repair pathways, photoreactivation and recombination repair. In mutant Arabidopsis plants that are deficient in photoreactivating ultraviolet-induced cyclobutane pyrimidine dimers, recombination under elevated UV-B regimes greatly exceeds wild-type levels. Our results show that homologous recombination repair pathways might be involved in eliminating UV-B-induced DNA lesions in plants. Thus, increases in terrestrial solar UV-B radiation as forecasted for the early 21st century may affect genome stability in plants.

The role clouds play in the modification of global radiation is still a major uncertainty in the risk assessment of UV effects on ecological systems and human health. This study presents cloud transmission data obtained from measurements with Robertson-Berger meters and simultaneous cloud observations. The global transmission of erythemally weighted irradiance depends strongly on cloud amount and can be described by a cubic function. The comparison with results derived from long-term records of total global irradiance indicates no statistically significant difference between the attenuation of erythemal and total global radiation. The large variance of data results from lumping together data from different cloud types. Classification of data according to cloud forms yields a more satisfactory fit. The coefficient of the cubic term characterizes the ability of various cloud forms to attenuate UV radiation. It varies between 0.4 for high clouds and approximately 1.0 for cumulonimbus. This attenuation parameter allows a quantitative description of the cloud influence on irradiance and therefore a more accurate risk assessment.

The degradation of sensitizers used in photodynamic therapy (PDT) involves photooxidation either by molecular oxygen or by oxygen intermediates which leads to hydroxyaldehyde and formyl products or to ring opening. Our investigations focused on the spectroscopic changes which protoporphyrin-dimethylester (PP) exhibits upon irradiation. As the microenvironment strongly influences the effects, we used an aprotic organic solvent, L-alpha-phosphatidylcholine dioleoyl (DOPC) liposomes and isogenic fibrosarcoma cells (SSKII) as carriers for PP. Hydroxyaldehyde product isomers develop a new absorption band centred around 670 nm and a new emission band at 676 nm. These characteristics can be used to discriminate them from formyl products and intact PP. In organic solvents, the formation of the hydroxyaldehyde products dominates. In DOPC liposomes and cells, the hydroxyaldehyde yield drops and photooxidation results in attack of the macrocycle. Time-resolved fluorescence spectroscopy of monomeric PP in an organic solvent gives a monoexponential decay time tau of 10.1 +/- 1.3 ns. Upon irradiation a second component with a decay time of 4.9 +/- 0.6 ns, resulting from the hydroxyaldehyde product, was detected. In liposomes and cells the monomeric decay time was significantly longer (15 ns) due to the altered microenvironment. Additionally, we observed in liposomes and in cells a small contribution of a short component (1 ns) which is attributed to an aggregated sensitizer species. In irradiated cells the aggregated fraction doubles, indicating a change in the microenvironment caused by the photodynamic action of the sensitizer.

We studied the optical and color performance of Olympus fiber- and electronic endoscopes, including the EVIS 100 and EVIS 200 series. The measurements showed that the optical resolution of new generation videoendoscopes equals or exceeds that of fiberendoscopes. The close-up focusing ability has been extended. The color performance of the new EVIS 200 gastroscope enables clear distinction of all mucosal hues.

The effects of solvent pH on spectral properties and fluorescence decay kinetics were investigated in order to characterize the microenvironment of meso-tetraphenylporphine tetrasulphonate (TPPS4) taken up by cells. Steady-state absorption and fluorescence spectra of TPPS4 in buffer solutions of different pH were used to identify a ring protonated species at pH less than or equal to 4. This dictation could also be distinguished from the unprotonated form by its altered fluorescence decay time (3.5 vs. 11.4 ns). In addition, time-resolved spectroscopy gave some evidence of a monocationic species existing at pH 6-9. This was concluded from the occurrence of another component with a decay time of 5 ns. Measurements of the spectral and kinetic properties of the fluorescence emission of single epithelial cells (RR1022) incubated with TPPS4 indicated that the sensitizer was mainly localized in a microenvironment with a pH of 5, a value which occurs intracellularly only within lysosomes. Cells kept in the dark exhibited the characteristic spectra of both the dication and the neutral form. The fluorescence decay showed two components with decay times of 2.6 ns and 10.6 ns. Irradiation of the cells changed the decay times to 4.6 ns and 13.4 ns and the dication fluorescence emission peak vanished, which is in accordance with the results obtained from buffer solutions at pH greater than or equal to 6. Therefore, we deduce that the photodynamic action leads to a rupture of the lysosomes and that the sensitizer is released into the surrounding cytoplasm.

Porphyrin monomers, dimers and aggregates, which can be differentiated on the basis of their fluorescence lifetimes, are shown to possess different degrees of fluorescence polarization. This opens up new possibilities for microscopic imaging of these individual components in photosensitization and tumour detection. A rough estimate of the size of the porphyrin aggregates is obtained from the data of time-resolved fluorescence anisotropy.

Reacting to the challenges presented by the evolving nexus of environmental change, defossilization, and diversified natural product bioprospecting is vitally important for advancing global healthcare and placing patient benefit as the most important consideration. This overview emphasizes the importance of natural and synthetic medicines security and proposes areas for global research action to enhance the quality, safety, and effectiveness of sustainable natural medicines. Following a discussion of some contemporary factors influencing natural products, a rethinking of the paradigms in natural products research is presented in the interwoven contexts of the Fourth and Fifth Industrial Revolutions and based on the optimization of the valuable assets of Earth. Following COP28, bioprospecting is necessary to seek new classes of bioactive metabolites and enzymes for chemoenzymatic synthesis. Focus is placed on those performance and practice modifications which, in a sustainable manner, establish the patient, and the maintenance of their prophylactic and treatment needs, as the priority. Forty initiatives for natural products in healthcare are offered for the patient and the practitioner promoting global action to address issues of sustainability, environmental change, defossilization, quality control, product consistency, and neglected diseases to assure that quality natural medicinal agents will be accessible for future generations. Ultraviolet (UV) radiation influences development and genome stability in organisms; however, its impact on meiosis, a special cell division essential for the delivery of genetic information across generations in eukaryotes, has not yet been elucidated. In this study, by performing cytogenetic studies, we reported that UV radiation does not damage meiotic chromosome integrity but attenuates centromere-mediated chromosome stability and induces unreduced gametes in Arabidopsis thaliana. We showed that functional centromere-specific histone 3 (CENH3) is required for obligate crossover formation and plays a role in the protection of sister chromatid cohesion under UV stress. Moreover, we found that UV specifically alters the orientation and organization of spindles and phragmoplasts at meiosis II, resulting in meiotic restitution and unreduced gametes. We determined that UV-induced meiotic restitution does not rely on the UV Resistance Locus8-mediated UV perception and the Tapetal Development and Function1- and Aborted Microspores-dependent tapetum development, but possibly occurs via altered JASON function and downregulated Parallel Spindle1. UVA and UVB are two main components of solar ultraviolet (UV) radiation. Some studies have reported the impacts of UV irradiation on plant secondary metabolism and plant growth. However, the role of UV irradiation in plant growth and quality in biochar-treated soils is poorly understood. This study aims to investigate the interactive effects of UV irradiation and biochar application on the growth and quality of Pseudostellaria heterophylla (Miq.) Pax, a popular medicinal plant. Three UV irradiation conditions (no UV irradiation, UVA irradiation at 2.5 W/m2 for 12 h/day and UVB irradiation at 0.05 W/m2 for 0.5 h/day) were considered in biochar-free and biochar (3%)-amended soils. The non-UV irradiated biochar-treated group exhibited the highest tuber yield. UVA irradiation reduced the yield improvement efficiency of biochar, while under UVB irradiation, biochar treatment reduced the tuber biomass. This were mainly because the application of high-nutrient biochar increased plant sensitivity to UV radiation. The UVA-irradiated group without biochar treatment exhibited the maximum polysaccharide concentration in the tuber, which was 38% higher than that of the control. Biochar application inhibited polysaccharide synthesis under UV treatment. The saponin concentration in the tuber was enhanced by up to 79% under UV (particularly UVB) irradiation. The coupled UVB and biochar treatment significantly enhanced saponin synthesis. These results indicate the interactive effects of UV irradiation and biochar treatment on P. heterophylla. Therefore, biochar amendment without UV irradiation is suggested to improve the tuber yield of P. heterophylla. UVA (2.5 W/m2 for 12 h/day) treatment is recommended for higher active ingredient concentration. However, the negative effects of UV irradiation on biochar-induced plant growth promotion should be also considered. Background: Artemisia annua is the major source for artemisinin production. The artemisinin content in A. annua is affected by different types of light especially the UV light. UVR8, a member of RCC1 gene family was found to be the UV-B receptor in plants. The gene structures, evolutionary history and expression profile of UVR8 or RCC1 genes remain undiscovered in A. annua. Results: Twenty-two RCC1 genes (AaRCC1) were identified in each haplotype genome of two diploid strains of A. annua, LQ-9 and HAN1. Varied gene structures and sequences among paralogs were observed. The divergence of most RCC1 genes occurred at 46.7 – 51 MYA which overlapped with species divergence of core Asteraceae during the Eocene, while no recent novel RCC1 members were found in A. annua genome. The number of RCC1 genes remained stable among eudicots and RCC1 genes underwent purifying selection. The expression profile of AaRCC1 is analogous to that of Arabidopsis thaliana (AtRCC1) when responding to environmental stress. Conclusions: This study provided a comprehensive characterization of the AaRCC1 gene family and suggested that RCC1 genes were conserved in gene number, structures, constitution of amino acids and expression profiles among eudicots. Integration of reactive oxygen species (ROS)-mediated signal transduction pathways via redox sensors and the thiol-dependent signalling network is of increasing interest in cell biology for their implications in plant growth and productivity. Redox regulation is an important point of control in protein structure, interactions, cellular location, and function, with thioredoxins (TRXs) and glutaredoxins (GRXs) being key players in the maintenance of cellular redox homeostasis. The crosstalk between second messengers, ROS, thiol redox signalling, and redox homeostasis-related genes controls almost every aspect of plant development and stress response. We review the emerging roles of TRXs and GRXs in redox-regulated processes interacting with other cell signalling systems such as organellar retrograde communication and gene expression, especially in plants during their development and under stressful environments. This approach will cast light on the specific role of these proteins as redox signalling components, and their importance in different developmental processes during abiotic stress. Sunlight regulates transcriptional programs and triggers the shaping of the genome throughout plant development. Among the different sunlight wavelengths that reach the surface of the Earth, UV-B (280–315 nm) controls the expression of hundreds of genes for the photomorphogenic responses and also induces the formation of photodamage that interfere with genome integrity and transcriptional programs. The combination of cytogenetics and deep-learning-based analyses allowed determining the location of UV-B-induced photoproducts and quantifying the effects of UV-B irradiation on constitutive heterochromatin content in different Arabidopsis natural variants acclimated to various UV-B regimes. We identified that UV-B-induced photolesions are enriched within chromocenters. Furthermore, we uncovered that UV-B irradiation promotes constitutive heterochromatin dynamics that differs among the Arabidopsis ecotypes having divergent heterochromatin contents. Finally, we identified that the proper restoration of the chromocenter shape, upon DNA repair, relies on the UV-B photoreceptor, UV RESISTANCE LOCUS 8 (UVR8). Background: Light spectra have been demonstrated to result in different levels of comfort or stress, which affect plant growth and the availability of health-promoting compounds in ways that sometimes contradict one another. To determine the optimal light conditions, it is necessary to weigh the vegetable's mass against the amount of nutrients it contains, as vegetables tend to grow poorly in environments where nutrient synthesis is optimal. This study investigates the effects of varying light conditions on the growth of red lettuce and its occurring nutrients in terms of productivities, which were determined by multiplying the total weight of the harvested vegetables by their nutrient content, particularly phenolics. Three different light-emitting diode (LED) spectral mixes, including blue, green, and red, which were all supplemented by white, denoted as BW, GW, and RW, respectively, as well as the standard white as the control, were equipped in grow tents with soilless cultivation systems for such purposes. Results: Results demonstrated that the biomass and fiber content did not differ substantially across treatments. This could be due to the use of a modest amount of broad-spectrum white LEDs, which could help retain the lettuce's core qualities. However, the concentrations of total phenolics and antioxidant capacity in lettuce grown with the BW treatment were the highest (1.3 and 1.4-fold higher than those obtained from the control, respectively), with chlorogenic acid accumulation (8.4 ± 1.5 mg g− 1 DW) being particularly notable. Meanwhile, the study observed a high glutathione reductase (GR) activity in the plant achieved from the RW treatment, which in this study was deemed the poorest treatment in terms of phenolics accumulation. Conclusion: In this study, the BW treatment provided the most efficient mixed light spectrum to stimulate phenolics productivity in red lettuce without a significant detrimental effect on other key properties. CONSTITUTIVE PHOTOMORPHOGENIC 1 (COP1), an E3 ubiquitin ligase, functions as a central repressor of light signaling and regulates various light-mediated developmental and metabolic processes in plants. However, detailed mechanisms underlying COP1-regulated flavonoid biosynthesis and embryogenesis in rice seeds remain largely unknown. Here, we performed transcriptome analysis of the rice cop1 (yellowish-pericarp embryo lethal [yel]) null mutant, characterized by flavonoid accumulation in pericarp and abnormal development of embryo, to identify and profile the expression genes involved in flavonoid biosynthesis and embryo development. Comparative transcriptome analysis of yel-hc and wild-type seeds revealed 979 differentially expressed genes (DEGs), of which 577 were upregulated and 402 were downregulated in yel-hc seeds. Functional annotation of DEGs revealed that DEGs were mainly enriched in 'metabolism', 'transcription factors', 'secondary metabolites', and 'flavonoid biosynthesis'. The DEGs encoding AP2-EREBP, MYB, and bZIP transcription factors (TFs) were predominantly upregulated, whereas those encoding HB, bHLH, and ABI3VP1 TFs were downregulated in yel-hc seeds. Comparative gene expression analysis revealed that genes involved in the C-glycosyl flavone biosynthesis pathway, including OsP1, were activated, whereas anthocyanin biosynthesis genes showed no significant change in expression. In addition, transcript levels of embryo development-related genes, especially homeobox auxin regulation genes, as well as somatic embryogenesis-related genes, were significantly downregulated in yel-hc. Taken together, these results indicate that OsCOP1 plays a crucial role in regulation of flavonoid biosynthesis and embryo structure formation, and changes in the expression of light signal transduction-related genes could have a significant impact on flavonoid biosynthesis and embryogenesis in rice seed. Ground level UV-B (290–315 nm) and UV-A (315–400 nm) radiation regulates multiple aspects of plant growth and development. In a natural environment, UV radiation interacts in a complex manner with other environmental factors (e.g., drought) to regulate plants' morphology, physiology, and growth. To assess the interactive effects of UV radiation and soil drying on plants' secondary metabolites and transcript abundance, we performed a field experiment using two different accessions of Medicago truncatula (F83005-5 French origin and Jemalong A17 Australian origin). Plants were grown for 37 days under long-pass filters to assess the effects of UV short wavelength (290–350 nm, UVsw) and UV-A long wavelength (350–400 nm, UV-Alw). Soil–water deficit was induced by not watering half of the plants during the last seven days of the experiment. The two accessions differed in the concentration of flavonoids in the leaf epidermis and in the whole leaf: F83005-5 had higher concentration than Jemalong A17. They also differed in the composition of the flavonoids: a greater number of apigenin derivatives than tricin derivatives in Jemalong A17 and the opposite in F83005-5. Furthermore, UVsw and soil drying interacted positively to regulate the biosynthesis of flavonoids in Jemalong A17 through an increase in transcript abundance of CHALCONE SYNTHASE (CHS). However, in F83005-5, this enhanced CHS transcript abundance was not detected. Light is an environmental signal that modulates plant defenses against attackers. Recent research has focused on the effects of light on defense hormone signaling; however, the connections between light signaling pathways and the biosynthesis of specialized metabolites involved in plant defense have been relatively unexplored. Here, we show that Arabidopsis BBX29, a protein that belongs to the B-Box transcription factor (TF) family, integrates photomorphogenic signaling with defense responses by promoting flavonoid, sinapate and glucosinolate accumulation in Arabidopsis leaves. AtBBX29 transcript levels were up regulated by light, through photoreceptor signaling pathways. Genetic evidence indicated that AtBBX29 up-regulates MYB12 gene expression, a TF known to induce genes related to flavonoid biosynthesis in a light-dependent manner, and MYB34 and MYB51, which encode TFs involved in the regulation of glucosinolate biosynthesis. Thus, bbx29 knockout mutants displayed low expression levels of key genes of the flavonoid biosynthetic pathway, and the opposite was true in BBX29 overexpression lines. In agreement with the transcriptomic data, bbx29 mutant plants accumulated lower levels of kaempferol glucosides, sinapoyl malate, indol-3-ylmethyl glucosinolate (I3M), 4-methylsulfinylbutyl glucosinolate (4MSOB) and 3-methylthiopropyl glucosinolate (3MSP) in rosette leaves compared to the wild-type, and showed increased susceptibility to the necrotrophic fungus Botrytis cinerea and to the herbivore Spodoptera frugiperda. In contrast, BBX29 overexpressing plants displayed increased resistance to both attackers. In addition, we found that AtBBX29 plays an important role in mediating the effects of ultraviolet-B (UV-B) radiation on plant defense against B. cinerea. Taken together, these results suggest that AtBBX29 orchestrates the accumulation of specific light-induced metabolites and regulates Arabidopsis resistance against pathogens and herbivores. At present, video endoscopic systems (VE) are available from four companies. We have had an opportunity to test these instruments in clinical practice. In principle, all the VE's are similar in that they employ charge coupling devices (CCD-chips) as image sensors, but they differ significantly in CCD chip technology and in the color acquisition technique. The basic aspects of CCD chip and transmission technology of relevance to the endoscopist are discussed. The mechanical properties of the endoscopes are listed. Finally, we provide an outlook on the determinants of the optical performance of VE's. The optical performance of a VE can be characterized by its resolution and color performance.

The subject of this study was the optical performance of video-endoscopic systems (VE) in terms of maximal resolving power and resolvable picture elements. Olympus, Toshiba/Machida, Fujinon, and Welch Allyn video gastroscopes were tested. A GIF Q 10 fiberscope from Olympus was also included for comparison. The resolution measurements were made at various distances using two independent methods--electronic analysis of the TV signal, and visual evaluation of the resolution, of a standardized test target. The results obtained with the two methods were in perfect agreement. The resolution of fine details clearly depends on the distance between the distal end and the target because of decreasing image scale. Depending on the individual optical design, the various VE's show maxima at different distances. At shorter distances, the image is degraded by defocusing. An optimal distance which is as small as possible is desired for clinical routine. Apart from the fiberscope this requirement is best met by the Fuji system. The greatest resolution is obtained with the Toshiba system but at the cost of the viewing angle which is the smallest of all the systems. Fuji combines relatively high resolution with a large viewing angle. Because of the widely varying viewing angle a comparison based solely on resolution cannot represent the true imaging capability of the system. We therefore eliminated purely optical parameters and calculated the number of resolvable picture elements per line. We regard this number to be a fair characterization of both TV and fiber systems.

The article focuses on techniques and applications of time-resolved fluorescence spectroscopy in biology and medicine. Both novel methods and well-established ones are discussed and future trends are outlined. Applications including fluorescence detection of nucleic acids, proteins, coenzymes and plant pigments and fluorescence labelling of nuclei, membranes and antibodies are outlined. In addition the fluorescence properties of photosensitizers used in photodynamic therapy are discussed.

We have conducted a study aimed at testing the color performance of video endoscopes in quantitative terms for the first time. The video endoscopes investigated were from Fuji, Olympus, Toshiba/Machida, and Welch-Allyn. The tests were carried out with an opto-electronic color analyser using standardized color charts and a neutral grey chart. Hue and chroma of the reproduced colors were measured. The Olympus video endoscope reproduces the hue very well, showing only slight color desaturation. The Fuji video endoscope had a yellowish tint, whereas the picture obtained with the WA video endoscope produced an inhomogeneous picture with a pronounced red/purplish cast at the top of the screen and almost neutral reproduction at the lower half. The Toshiba video endoscope was the only one with a color chip, and reproduced hue fairly well, but its colors were weakly saturated.

Plant UV-B responses are mediated by the photoreceptor UV RESISTANCE LOCUS 8 (UVR8). In response to UV-B irradiation, UVR8 homodimers dissociate into monomers that bind to the E3 ubiquitin ligase CONSTITUTIVE PHOTOMORPHOGENIC1 (COP1). The interaction of the C27 domain in the C-terminal tail of UVR8 with the WD40 domain of COP1 is critical for UV-B signaling. However, the function of the last 17 amino acids (C17) of the C-terminus of UVR8, which are adjacent to C27, is unknown, although they are largely conserved in land plants. In this study, we established that Arabidopsis thaliana UVR8 C17 binds to full-length UVR8, but not to COP1, and reduces COP1 binding to the remaining portion of UVR8, including C27. We hypothesized that overexpression of C17 in a wild-type background would have a dominant negative effect on UVR8 activity; however, C17 overexpression caused strong silencing of endogenous UVR8, precluding a detailed analysis. We therefore generated YFP-UVR8 N423 transgenic lines, in which C17 was deleted, to examine C17 function indirectly. YFP-UVR8 N423 was more active than YFP-UVR8, suggesting that C17 inhibits UV-B signaling by attenuating binding between C27 and COP1. Light plays an important role in plants' growth and development throughout their life cycle. Plants alter their morphological features in response to light cues of varying intensity and quality. Dedicated photoreceptors help plants to perceive light signals of different wavelengths. Activated photoreceptors stimulate the downstream signaling cascades that lead to extensive gene expression changes responsible for physiological and developmental responses. Proteins such as ELONGATED HYPOCOTYL5 (HY5) and CONSTITUTIVELY PHOTOMORPHOGENIC 1 (COP1) act as important factors which modulate light-regulated gene expression, especially during seedling development. These factors function as central regulatory intermediates not only in red, far-red, and blue light pathways but also in the UV-B signaling pathway. UV-B radiation makes up only a minor fraction of sunlight, yet it imparts many positive and negative effects on plant growth. Studies on UV-B perception, signaling, and response in plants has considerably surged in recent times. Plants have developed different strategies to use UV-B as a developmental cue as well as to withstand high doses of UV-B radiation. Plants' responses to UV-B are an integration of its cross-talks with both environmental factors and phytohormones. Elicitors, irrigation regimes and harvest times influence the content, yield and compound of the essential oil (EO) in Salvia officinalis (sage), through changes in biomass dynamics and biosynthetic pathways. A two-year field experiment was conducted to determine if foliar application of putrescine under optimum and deficit stress conditions would favorably affect EO yield, content and profile of sage harvested in spring and summer. The response of dry weight, EO yield and content, myrcene and borneol concentrations to irrigation regime and putrescine concentration can be expressed by a quadratic model. The maximum dry weight (182.63 g m−2) and EO yield (1.68 g m−2) were predicted under irrigation regimes of 9.06% and 27.75% available soil water depletion (ASWD), respectively. The highest EO content (1.05%) was predicted under 3.04 mM of putrescine. Based on results obtained from GC/MS analyses, 25 compounds (mostly monoterpenes) were identified in the EO of sage. Among EO compounds, α-thujone (54.08%), 1, 8-cineole (17.87%), pinocarvone (14.30%), β-thujone (7.97%) and camphor (8.76%) in turn were the most abundant. The concentration of myrcene was higher in spring than summer under the irrigation regimes of 60% and 80% ASWD. The myrcene concentration reached its maximum (4.53%) under the irrigation regime of 86.5% ASWD. The irrigation regimes of 48.03% and 45.6% ASWD caused the highest borneol concentrations of 1.47% and 1.41% by application of 1.5 mM and 2.25 mM putrescine, respectively. All treatments tested on sage, particularly harvest time, can play an important role in the improvement of EO quality and quantity. Averaged over both years, the irrigation regime of nearly 30% ASWD resulted in the highest EO yield harvested with greater quantity and better quality in summer. Summary: Due to their sessile nature, plants cannot escape adverse environmental conditions and evolved mechanisms to cope with sudden environmental changes. The reaction to variations in abiotic factors, also summarized as acclimation response, affects all layers of cellular functions and involves rapid modification of enzymatic activities, the metabolome, proteome and transcriptome on different timescales. One trait of plants acclimating to high light (HL) is the rapid transcriptional activation of the flavonoid biosynthesis (FB) pathway resulting in the accumulation of photoprotective and antioxidative flavonoids, such as flavonols and anthocyanins, in the leaf tissue. Although enormous progress has been made in identifying enzymes and transcriptional regulators of FB by forward and reverse genetic approaches in the past, the signals and signalling pathways permitting the conditional activation of FB in HL are still debated. With this Tansley Insight, we summarize the current knowledge on the proposed signals and downstream factors involved in regulating FB and will discuss their contribution to, particularly, HL‐induced accumulation of anthocyanins. The aim of this investigation was to study the effect of physical (ultraviolet light) and chemical mutagen (methyl salicylate) on common bean (Phaseolus vulgaris L.) and how they had positive or negative effect on alpha-amylase activity, phenological plant traits (days to flowering, days to maturity, days of pod fill) and agronomic traits (number of pods per plant; weight of pods per plant; Number of seeds per pod; number of seeds per plant; weight of seeds per plant; plant height; pod harvest index), seed and pod morphological characteristics (seed length; seed width; pod length; pod width). Seeds of common bean were exposed to ultraviolet (UV) light for different periods of exposure time (3 and 5 h) and were soaked with two different methyl salicylate (MeSA) concentrations (0.01 and 0.05 mM). The results indicated that the 5 h exposure to UV light plants were indicated to positively affect alpha-amylase activity, phenological and agronomic traits, seed and pod morphological characteristics as compared to control and other treatments while 0.01 mM MeSA significantly negatively impacted them. Traits of plants exposed to UV light for 5 h were significantly improved. This research could provide a valuable source of materials for common bean breeding. [ABSTRACT FROM AUTHOR]

Environmental factors including climate factors and soil parameters affect growth, physiological processes and production of primary and secondary metabolites in plants. In addition, the responses of plants are widely varied to environmental factors. In the current study the impacts of rainfall, temperature and several soil factors such as soil texture, pH, electrical conductivity, organic matter, organic carbon, calcium carbonate and soil elements on nutrient elements, antioxidative compounds, activity, and isoenzymes patterns of peroxidase and superoxide dismutase in Stachys byzantina were investigated. S. byzantina as a valuable medicinal species was collected from three regions in the north of Iran with different altitudes and ecological conditions. The results showed that the content of nutrient elements (phosphorus, sodium, potassium, calcium), anti-oxidative compounds (phenols, flavonol, flavonoids, and anthocyanin) as well as the activity of peroxidase and superoxide dismutase enzymes along with intensity of their isoenzymes bands were positively correlated with temperature and rainfall, however, almost not correlated with soil parameters. Furthermore, the higher density of S. byzantina in the region with lower temperature and rainfall indicated greater adaptability of the plant in higher altitude areas, which is consistent with a decrease in the antioxidant compounds and activity of oxidative enzymes. Summary: Light regulates the subcellular localization of plant photoreceptors, a key step in light signaling. Ultraviolet‐B radiation (UV‐B) induces the plant photoreceptor UV RESISTANCE LOCUS 8 (UVR8) nuclear accumulation, where it regulates photomorphogenesis. However, the molecular mechanism for the UV‐B‐regulated UVR8 nuclear localization dynamics is unknown.With fluorescence recovery after photobleaching (FRAP), cell fractionation followed by immunoblotting and co‐immunoprecipitation (Co‐IP) assays we tested the function of UVR8‐interacting proteins including CONSTITUTIVELY PHOTOMORPHOGENIC 1 (COP1), REPRESSOR OF UV‐B PHOTOMORPHOGENESIS 1 (RUP1) and RUP2 in the regulation of UVR8 nuclear dynamics in Arabidopsis thaliana.We showed that UV‐B‐induced rapid UVR8 nuclear translocation is independent of COP1, which previously was shown to be required for UV‐B‐induced UVR8 nuclear accumulation. Instead, we provide evidence that the UV‐B‐induced UVR8 homodimer‐to‐monomer photo‐switch and the concurrent size reduction of UVR8 enables its monomer nuclear translocation, most likely via free diffusion. Nuclear COP1 interacts with UV‐B‐activated UVR8 monomer, thereby promoting UVR8 nuclear retention. Conversely, RUP1and RUP2, whose expressions are induced by UV‐B, inhibit UVR8 nuclear retention via attenuating the UVR8–COP1 interaction, allowing UVR8 to exit the nucleus.Collectively, our data suggest that UV‐B‐induced monomerization of UVR8 promotes its nuclear translocation via free diffusion. Background Land plants commonly produce red pigmentation as a response to environmental stressors, both abiotic and biotic. The type of pigment produced varies among different land plant lineages. In the majority of species they are flavonoids, a large branch of the phenylpropanoid pathway. Flavonoids that can confer red colours include 3-hydroxyanthocyanins, 3-deoxyanthocyanins, sphagnorubins and auronidins, which are the predominant red pigments in flowering plants, ferns, mosses and liverworts, respectively. However, some flowering plants have lost the capacity for anthocyanin biosynthesis and produce nitrogen-containing betalain pigments instead. Some terrestrial algal species also produce red pigmentation as an abiotic stress response, and these include both carotenoid and phenolic pigments. Scope In this review, we examine: which environmental triggers induce red pigmentation in non-reproductive tissues; theories on the functions of stress-induced pigmentation; the evolution of the biosynthetic pathways; and structure–function aspects of different pigment types. We also compare data on stress-induced pigmentation in land plants with those for terrestrial algae, and discuss possible explanations for the lack of red pigmentation in the hornwort lineage of land plants. Conclusions The evidence suggests that pigment biosynthetic pathways have evolved numerous times in land plants to provide compounds that have red colour to screen damaging photosynthetically active radiation but that also have secondary functions that provide specific benefits to the particular land plant lineage. To counter the insect infestation, plants respond with wide-ranging and highly dynamic biochemical reactions. Of these, the anti-oxidative activity is poorly understood. The red palm weevil (RPW) Rhynchophorus ferrugineus (Oliver), one of the most widespread pests in Pakistan, prefers to infest date palm Phoenix dactylifera. Our present study investigated the feeding preference of RPW to 11 different date palm cultivars and the results suggested that the Hillawi cultivar was most preferred. Greater infestation rate, fecundity and hatching rate were also recorded from Hillawi and Mozawati than other cultivars. No significant decreases were observed in chlorophyll a, chlorophyll b, total chlorophylls and carotenoids of RPW-infested Hillawi cultivar over un-infested control. In contrast, the contents of enzymatic antioxidants including phenols, proline, hydrogen peroxide, anthocyanin, malondialdehyde, ascorbic acid and glycine betaine showed a drastic increase after RPW infestation, and there was enhanced superoxide dismutase, peroxidase and catalase activities. Furthermore, we recorded the increase of total protein and sugar contents in RPW-infested date palms. These findings offer valuable insight into the antioxidative molecular mechanism of date palms under RPW attack and may contribute to the breeding of insect-resistant crops. [ABSTRACT FROM AUTHOR]
