Abstract

Enabling supply chain transparency (SCT) is essential for regulatory compliance and meeting sustainability standards. Multi-tier SCT plays a pivotal role in identifying and mitigating an organization's operational, environmental, and social (ESG) risks. While research observes increasing efforts towards SCT, a minority of companies are currently publishing supply chain information. Using the Design Science Research approach, we develop a collaborative platform for supply chain transparency. We derive design requirements, formulate design principles, and evaluate the artefact with industry experts. Our artefact is initialized with publicly available supply chain data through an automated pipeline designed to onboard future participants to our platform. This work contributes to SCT research by providing insights into the challenges and opportunities of implementing multi-tier SCT and offers a practical solution that encourages organizations to participate in a transparent ecosystem.

Abstract

Estimating the speed of vehicles using traffic cameras is a crucial task for traffic surveillance and management, enabling more optimal traffic flow, improved road safety, and lower environmental impact. Transportation-dependent systems, such as for navigation and logistics, have great potential to benefit from reliable speed estimation. While there is prior research in this area reporting competitive accuracy levels, their solutions lack reproducibility and robustness across different datasets. To address this, we provide a novel framework for automatic real-time vehicle speed calculation, which copes with more diverse data from publicly available traffic cameras to achieve greater robustness. Our model employs novel techniques to estimate the length of road segments via depth map prediction. Additionally, our framework is capable of handling realistic conditions such as camera movements and different video stream inputs automatically. We compare our model to three well-known models in the field using their benchmark datasets. While our model does not set a new state of the art regarding prediction performance, the results are competitive on realistic CCTV videos. At the same time, our end-to-end pipeline offers more consistent results, an easier implementation, and better compatibility. Its modular structure facilitates reproducibility and future improvements.

Abstract

Electric pulses in biological cells (action potentials) have been reported to be accompanied by a propagating cell-surface deformation with a nano-scale amplitude. Typically, this cell surface is covered by external layers of polymer material (extracellular matrix, cell wall material etc.). It was recently demonstrated in excitable plant cells (Chara Braunii) that the rigid external layer (cell wall) hinders the underlying deformation. When the cell membrane was separated from the cell wall by osmosis, a mechanical deformation, in the micrometer range, was observed upon excitation of the cell. The underlying mechanism of this mechanical pulse has up to date remained elusive. Herein we report that Chara cells can undergo a pearling instability, and when the pearled fragments were excited even larger and more regular cell shape changes were observed (about 10 to 100 um in amplitude). These transient cellular deformations were captured by a curvature model that is based on three parameters: surface tension, bending rigidity and pressure difference across the surface. In this paper these parameters are extracted by curve-fitting to the experimental cellular shapes at rest and during excitation. Abstract

The existence of acoustic pulse propagation in lipid monolayers at the air-water interface is well known. These pulses are controlled by the thermodynamic state of the lipid membrane. Nevertheless, the role of acoustic pulses for intra- and intercellular communication are still a matter of debate. Herein, we used the dye di 4- -ANEPPDHQ, which is known to be sensitive to the physical state and transmembrane potential of membranes, in order to gain insight into compression waves in lipid-based membrane interfaces. The dye was incorporated into lipid monolayers made of phosphatidylserine or phosphatidylcholine at the air-water-interface. A significant blue shift of the emission spectrum was detected when the state of the monolayer was changed from the liquid expanded (LE) to the liquid condensed (LC) phase. This transition-sensitivity of di-4-ANEPPDHQ was generalized in experiments with the bulk solvent dimethyl sulfoxide (DMSO). Upon crystallization of solvent, the emission spectrum also underwent a blue shift. During compression pulses in lipid monolayers, a significant fluorescence response was only observed when in the main transition regime. The optical signature of these waves, in terms of sign and magnitude, was identical to the response of di-4-ANEPPDHQ during action potentials in neurons and excitable plant cells. These findings corroborated the suggestion that action potentials are nonlinear state changes that propagate in the cell membrane.

17. Sharp, localized phase transitions in single neuronal cells

Abstract

The origin of nonlinear responses in cells has been suggested to be crucial for various cell functions including the propagation of the nervous impulse. In physics nonlinear behavior often originates from phase transitions. Evidence for such transitions on the single cell level, however, has so far not been provided leaving the field unattended by the biological community. Here we demonstrate that single cells of a human neuronal cell line, display all optical features of a sharp, highly nonlinear phase transition within their membrane. The transition is reversible and does not origin from protein denaturation. Triggered by temperature and modified by pH here, a thermodynamic approach, strongly suggests, that similar nonlinear state changes can be induced by other variables such as calcium or mechanical stress. Abstract

Current standard techniques for investigating protein-protein interactions require volumes and concentrations significantly higher than those relevant under physiological conditions. Microfluidic diffusional sizing offers an alternative method for investigating protein-protein interactions and protein assembly under physiological conditions and has been applied to investigate a wide range of protein properties, functions, and interactions. In this thesis, current approaches are further extended and their applicability to various interactions related to human diseases is shown. Specifically, recent work is described which makes use of microfluidic techniques combined with chemical kinetic and thermodynamic analyses to study the mechanism of the disaggregation of α-synuclein fibrils with Hsc70 along with its co-chaperones DnaJB1 and Apg2 as well the interaction between BRICHOS and amyloid fibrils. These analyses provide mechanistic insight into the role of molecular chaperones in clearance of amyloid fibrils and inhibition of their formation. The second part focuses on the development of a diagnostic tool based on microfluidic diffusional sizing for detection of antibodies in pristine human serum and plasma samples. This allows to characterise affinity and concentration as well as neutralisation potential of patient antibodies. This technique is then further applied to characterisation of plasma samples from convalescent SARS-CoV-2 infected individuals, revealing that the affinities span more than two orders of magnitude while the concentrations are more constant. The thesis shows how advances physico-chemical technology enable answering emerging questions in biophysical chemistry and medicine.

50. Momentum Conservation in Biological Communication: Its not about the molecules

Abstract

Life is full of hydrated interfaces that all must obey physical principles. Neglecting conservation laws (unintentionally or not) is no misdemeanor but a serious violation of nature’s principles and can lead to an absurd picture of nature. I here present, what (biological) interfaces naturally “produce” once “fed” with momentum conservation and the 2nd law of thermodynamics. Data show the existence of linear and nonlinear pulses, which are capable to trigger and suppress enzymatic activity. Together with the 2nd law, momentum conservation allows to control biological processes from remote. Specificity of this control also arises from physics and is found in the phase diagrams of this interfaces near transitions (state-function relation). Together this leads to a physical perspective for the origin of biological communication and a concept for the integration of physical principles and biochemical reactions.
