Summary: The interaction between animals and plants for seed dispersal and predation has received much attention; however, the underlying physiological mechanisms driving the responses of both seeds and animals remain unclear.We conducted a series of behaviour and physiology experiments to examine the role of plant hormones in regulating seed germination and rodent hoarding behaviour in the Quercus variabilis and Leopoldamys edwardsi systems.We found that acorns that were partially consumed by rodents had increased gibberellin (GA) levels and shortened germination time. Rodents preferred scatter‐hoarded abscisic acid (ABA)‐treated and intact acorns but consumed germinated and GA‐treated acorns; such treatment differences disappeared for inactivated acorns by boiling water. Moreover, we found that seven potential compounds may be linked to seed germination and rodent hoarding behaviour.Our results indicate that acorns of oak showed rapid germination when facing predation risk, while rodents could identify the germination status of seeds for hoarding; GA and ABA may play an important role in regulating seed germination of oak and hoarding behaviour of rodents. Abstract Our study aims to identify the mechanisms involved in regulating the response of Rhodoendron Chrysanthum Pall. (R. chrysanthum) leaves to UV-B exposure; phosphorylated proteomics and metabolomics for phenolic acids and plant hormones were integrated in this study. The results showed that UV-B stress resulted in the accumulation of salicylic acid and the decrease of auxin, jasmonic acid, abscisic acid, cytokinin and gibberellin in R. chrysanthum. The phosphorylated proteins that changed in plant hormone signal transduction pathway and phenolic acid biosynthesis pathway were screened by comprehensive metabonomics and phosphorylated proteomics. In order to construct the regulatory network of R. chrysanthum leaves under UV-B stress, the relationship between plant hormones and phenolic acid compounds was analyzed. It provides a rationale for elucidating the molecular mechanisms of radiation tolerance in plants.

Abstract Background Plant hormones profoundly influence cotton growth, development, and responses to various stresses. Therefore, there is a pressing need for an efficient assay to quantify these hormones in cotton. In this groundbreaking study, we have established QuEChERS-HPLC‒MS/MS method, for the simultaneous detection of multiple plant hormones in cotton leaves, allowing the analysis and quantification of five key plant hormones. Results Sample extraction and purification employed 0.1% acetic acid in methanol and C18 for optimal recovery of plant hormones. The method applied to cotton demonstrated excellent linearity across a concentration range of 0.05–1 mg·L−1, with linear regression coefficients exceeding 0.99. The limits of quantification (LOQs) were 20 μg·kg−1 for GA3 and 5 μg·kg−1 for the other four plant hormones. Recovery rates for the five plant hormones matrix spiked at levels of 5, 10, 100, and 1000 μg·kg−1 were in the range of 79.07% to 98.97%, with intraday relative standard deviations (RSDs) ranging from 2.11% to 8.47%. The method was successfully employed to analyze and quantify the five analytes in cotton leaves treated with plant growth regulators. Conclusion The study demonstrates that the method is well-suited for the determination of five plant hormones in cotton. It exhibits excellent selectivity and sensitivity in detecting field samples, thus serving as a robust tool for in-depth research into cotton physiology.

Exploring plant behavior at the cellular scale in a minimally invasive manner is critical to understanding plant adaptation to the environment. Phytohormones play vital regulatory roles in multiple aspects of plant growth and development and acclimation to environmental changes. Since the biosynthesis, modification, transportation, and degradation of plant hormones in plants change with time and space, their content level and distribution are highly dynamic. To monitor the production, transport, perception, and distribution of phytohormones within undamaged tissues, we require qualitative and quantitative tools endowed with remarkably high temporal and spatial resolution. Fluorescent probes are regarded as excellent tools for widespread plant imaging because of their high sensitivity and selectivity, reproducibility, real-time in situ detection, and uncomplicated mechanism elucidation. In this review, we provide a systematical overview of the progress in the sensing and imaging of phytohormone fluorescent probes and fluorescently labeled phytohormones to their receptors in plants. Moreover, forthcoming viewpoints and possible applications of these fluorescent probes within the realm of plants are also presented. We hold the conviction that the new perspective brought by this paper can promote the development of fluorescent probes, enabling them to have better detection performance in plant hormone imaging.

Rho of Plants (ROPs) constitute a plant-specific subset of small guanine nucleotide-binding proteins within the Cdc42/Rho/Rac family. These versatile proteins regulate diverse cellular processes, including cell growth, cell division, cell morphogenesis, organ development, and stress responses. In recent years, the dynamic cellular and subcellular behaviours orchestrated by ROPs have unveiled a notable connection to hormone-mediated organ development and physiological responses, thereby expanding our knowledge of the functions and regulatory mechanisms of this signalling pathway. This review delineates advancements in understanding the interplay between plant hormones and the ROP signalling cascade, focusing primarily on the connections with auxin and abscisic acid pathways, alongside preliminary discoveries in cytokinin, brassinosteroid, and salicylic acid responses. It endeavours to shed light on the intricate, coordinated mechanisms bridging cell- and tissue-level signals that underlie plant cell behaviour, organ development, and physiological processes, and highlights future research prospects and challenges in this rapidly developing field. Abstract Plant hormones play pivotal roles in stress responses by modulating growth, development, stomatal movement, and the expression of stress-related genes, thereby aiding plants in adapting to and managing various environmental challenges. Each hormone exhibits distinct functions and regulatory mechanisms in stress response, with potential complex interactions among them. Brassinosteroids (BRs) represent a novel hormone that influences the expression of its target genes through a series of phosphorylated cascade reactions involving various transcription factors. This signaling pathway regulates diverse growth and development processes in plants. Additionally, BRs interact with other hormones to modulate physiological development. This review examines BRs biosynthesis and metabolism, elucidates the interactions between BRs and abscisic acid (ABA), jasmonic acid (JA), and gibberellins (GA), and explores their roles in regulating responses to drought, salt, cold, and heat. The review underscores the importance of BRs and other hormonal crosstalk in managing nutrient stress, which is vital for understanding plant growth regulation, enhancing crop stress resistance, advancing biotechnology applications, and furthering plant science research. Future research directions and the potential of BRs production and application to improve plant stress resilience are also discussed in the context of current findings.

Microbial competition within plant tissues affects invading pathogens' fitness. Metabolomics is a great tool for studying their biochemical interactions by identifying accumulated metabolites. Xylella fastidiosa, a Gram-negative bacterium causing Pierce's disease (PD) in grapevines, secretes various virulence factors including cell wall-degrading enzymes, adhesion proteins, and quorum-sensing molecules. These factors, along with outer membrane vesicles, contribute to its pathogenicity. Previous studies demonstrated that co-inoculating X. fastidiosa with the Paraburkholderia phytofirmans strain PsJN suppressed PD symptoms. Here, we further investigated the interaction between the phytopathogen and the endophyte by analyzing the exometabolome of wild-type X. fastidiosa and a diffusible signaling factor (DSF) mutant lacking quorum sensing, cultivated with 20% P. phytofirmans spent media. Liquid chromatography-mass spectrometry (LC-MS) and the Method for Metabolite Annotation and Gene Integration (MAGI) were used to detect and map metabolites to genomes, revealing a total of 121 metabolites, of which 25 were further investigated. These metabolites potentially relate to host adaptation, virulence, and pathogenicity. Notably, this study presents the first comprehensive profile of X. fastidiosa in the presence of a P. phytofirmans spent media. The results highlight that P. phytofirmans and the absence of functional quorum sensing affect the ratios of glutamine to glutamate (Gln:Glu) in X. fastidiosa. Additionally, two compounds with plant metabolism and growth properties, 2-aminoisobutyric acid and gibberellic acid, were downregulated when X. fastidiosa interacted with P. phytofirmans. These findings suggest that P. phytofirmans-mediated disease suppression involves modulation of the exometabolome of X. fastidiosa, impacting plant immunity.

Plant hormones and their receptors play a crucial role in regulating plant growth and adapting to the stress environment. The exploration of interaction between plant hormones and their receptors is significant to comprehend the molecular mechanisms of plant growth and development, the response mechanisms of adaptation to environmental changes, and to optimize the traits and stress‐resistance of crops. Since the biosynthesis, transport, and metabolism of hormones in plants are closely relevant to spatio‐temporal changes, and their content and distribution are highly dynamic, there is an urgent need for a qualitative and quantitative tool to accurately, real‐time, and in situ monitor the dynamic changes of hormones in plants without injury. Fluorescent probes have been widely used in the sensing and imaging of plant hormones and their receptors, due to their high spatio‐temporal resolution, high selectivity, non‐invasive, high sensitivity, and tailored molecular structures. Here, this paper provides a systematical overview of the research progress in the sensing and imaging of plant hormones and their receptors using fluorescent probes. In addition, the potential prospects and remaining challenges are also discussed to design fluorescent probes with better performance and promote the development of this field. Abstract Utilizing the organic fraction of agri-food industry waste for fertilization represents one approach to waste management, with composting emerging as a popular method. Composts derived from this waste may contain plant hormones alongside primary macronutrients. This study aimed to evaluate the content of plant hormones in composts crafted from the organic fraction of agri-food industry waste. The presence of these substances was ascertained using liquid chromatography–mass spectrometry (LC-MS) analysis, applied to extracted samples from three composts produced in a bioreactor and three obtained from companies. The results indicate the presence of 35 compounds, which belong to six types of plant hormones: auxins, cytokinins, gibberellins, brassinosteroids, abscisic acid, and salicylic acid, in composts for the first time. The highest amount of plant hormones was noted in buckwheat husk and biohumus extract (35 compounds), and the lowest in hemp chaff and apple pomace (14 compounds). Brassinosteroids (e.g., brassinolide, 28-homobrassinolide, 24-epicastasterone, 24-epibrassinolide, and 28-norbrassinolide) and auxins (e.g., indolilo-3-acetic acid) are dominant. The highest concentration of total phytohormones was reported in biohumus extract (2026.42 ng g−1 dry weight), and the lowest in organic compost (0.18 ng g−1 dry weight).

Objectives To improve the yield and secondary metabolite content of medicinal plants and to further develop and utilize the medicinal and other functions of medicinal plants. #Methods] We used the sterile tissue culture method with Houtuynia cordata Thunb. a the research object. Different concentrations of 1-naphthalene acetic acid (NAA), auxin (indole-3-acetic acid, IAA) and gibberellin acid (GA3) were added to the group culture medium of H. cordata to investigate the effects of exogenous plant hormones on plant height, root length, fresh weight, morphological characteristics, four phenolics and 20 volatile compounds. [Results] The results showed that the exogenous plant hormone of 3 mg/L GA3 significantly increased plant height by 79. 9% over the control; the exogenous plant hormone of 3 mg/L IAA significantly increased root length by 52. 6% over the control; and the exogenous plant hormone of 1 mg/L GA3 significantly increased fresh weight of single plant by 458.2% over the control. In the treatment group of 1 mg/LNAA, chlorogenic acid content was significantly increased by 52.6% com¬pared with the control; in the treatment group of 1 mg/L IAA, chlorogenic acid, rutin, isodendrin and quercetin content were significantly in¬creased by 109.1%, 100. 6%, 173. 8%, and 198. 7% compared with the control, respectively; in the treatment of 3 mg/L GA3, chlorogenic acid, rutin, isoquercitin, and quercitin content were significantly increased by 65. 3%, 104.9%, 139.0% and191.2% over the control. In addition, the content of volatile compounds was significantly higher in all H cordata treated with exogenous plant hormones of 2 mg/L NAA, 1 mg/L IAA, and 3 mg/L GA3; however, the content of volatile compounds was lower in aH of the treatments with 2 mg/L GA3. # Conclusions ] Different exogenous plant hormones have certain effects on the growth morphology and secondary metabolic content of H cordata, which provides theoretical basis and technical support for the development and utilization of medicinal plants. Simple Summary: The current review article focused on the complex interaction between plant hormones and transcription factors in the response to salt stress, a pressing global issue that has a considerable effect on agricultural productivity. The study unveils the effect of salt stress on the ion equilibrium and triggers a cascade of molecular reactions that have a response to plant growth and development. Transcription factors are essential in regulating gene expression under salt stress. The transcription factors function in collaboration with hormones to regulate environmental stress responses. The study sheds light on the underlying regulatory networks, thus providing crucial knowledge for cultivating salt-tolerant crops through selective breeding and genetic engineering. This review significantly contributes to the establishment of sustainable agricultural techniques, which are essential in addressing the increasing problem of soil salinization and securing global food security. The negative impacts of soil salinization on ion homeostasis provide a significant global barrier to agricultural production and development. Plant physiology and biochemistry are severely affected by primary and secondary NaCl stress impacts, which damage cellular integrity, impair water uptake, and trigger physiological drought. Determining how transcriptional factors (TFs) and hormone networks are regulated in plants in response to salt stress is necessary for developing crops that tolerate salt. This study investigates the complex mechanisms of several significant TF families that influence plant responses to salt stress, involving AP2/ERF, bZIP, NAC, MYB, and WRKY. It demonstrates how these transcription factors (TFs) help plants respond to the detrimental effects of salinity by modulating gene expression through mechanisms including hormone signaling, osmotic stress pathway activation, and ion homeostasis. Additionally, it explores the hormonal imbalances triggered by salt stress, which entail complex interactions among phytohormones like jasmonic acid (JA), salicylic acid (SA), and abscisic acid (ABA) within the hormonal regulatory networks. This review highlights the regulatory role of key transcription factors in salt-stress response, and their interaction with plant hormones is crucial for developing genome-edited crops that can enhance agricultural sustainability and address global food security challenges. Safflower (Carthamus tinctorius L.) belongs to the composite family with various uses, from the stem to the seeds. In vitro flowering and embryo rescue are common techniques for improving safflower hybrid fertility by overcoming breeding constraints. However, anther culture expedites the production of superior types. The aim of the study was to evaluate the effect of plant hormones and genotypes on anther culture responses and to develop a suitable protocol. In both safflower genotypes, callus, shoot, and root formation were investigated using ten factorial hormone treatments. To promote callus induction and shoot regeneration, variable doses of Thidiazuron (TDZ) at 1 mg/l indole butyric acid (IBA) and variable benzylaminopurine (BAP) at 0.5 mg/l NAA were added to the MS medium, respectively. ½ MS media with varying IBA concentrations at a fixed 0.5 mg/l NAA were used for root regeneration. Results showed that Turkan was superior in all aspects of callus induction, and the highest degree of callus formation (46 %) was observed at 0.5 mg/l TDZ. On the other hand, the local safflower performed better for shoot and root regeneration. The highest shoot regeneration capacity (20 %) was shown at 2.0 mg/l BAP, while shoot rooting was reached (21 %) at 1.0 mg/l IBA of culture media. Despite successful in vitro regeneration, acclimated plantlets did not survive in the glasshouse. In this study, we demonstrated the higher capability for both shoot and root regeneration of calli produced from local safflower anthers. More effort is required to improve shoot and root development for both genotypes.

Crops are destroyed by extreme heat, which also limits their growth and yield. The present study sought to determine whether selenium (0, 15, or 30 mg l-1) impacted ‘Barhee’ date palm seedling’s development under heat stress (in the field and canopy temperature). The growth parameters, chlorophyll and relative water content, ascorbic acid, catalase activity, and phytohormones in seedlings were reduced under heat stress. At the same time, ascorbate peroxidase activity, proline, phenols, malondialdehyde, hydrogen peroxide, and abscisic acid in seedlings increased. Enhancing growth features, chlorophyll content, relative water content, ascorbic acid, catalase activity, plant hormones, proline, phenols, and ascorbate peroxidase activity with exogenous nano-Selenium (15 mg l-1) reduced the negative impacts of heat stress. Date palm seedlings can be protected from high temperatures by using nano-selenium. Selenium reverses heat-induced oxidative damage by enhancing the antioxidative mechanism, improving reactive oxygen species scavenging, lowering lipid peroxidation, and modulating plant hormone levels.

Rubisco small subunit (RbcS), a core component with crucial effects on the structure and kinetic properties of the Rubisco enzyme, plays an important role in response to plant growth, development, and various stresses. Although Rbcs genes have been characterized in many plants, their muti-functions in soybeans remain elusive. In this study, a total of 11 GmRbcS genes were identified and subsequently divided into three subgroups based on a phylogenetic relationship. The evolutionary analysis revealed that whole-genome duplication has a profound effect on GmRbcSs. The cis-acting elements responsive to plant hormones, development, and stress-related were widely found in the promoter region. Expression patterns based on the RT-qPCR assay exhibited that GmRbcS genes are expressed in multiple tissues, and notably Glyma.19G046600 (GmRbcS8) exhibited the highest expression level compared to other members, especially in leaves. Moreover, differential expressions of GmRbcS genes were found to be significantly regulated by exogenous plant hormones, demonstrating their potential functions in diverse biology processes. Finally, the function of GmRbcS8 in enhancing soybean resistance to soybean mosaic virus (SMV) was further determined through the virus-induced gene silencing (VIGS) assay. Plant hormones are essential and structurally diverse molecules that regulate various aspects of plant growth, development, and stress responses. However, the precise analysis of plant hormones in complex biological samples poses a challenge due to their low concentrations, dynamic levels, and intricate spatial distribution. Moreover, the complexity and interconnectedness of hormone signaling networks make it difficult to simultaneously trace multiple hormone distributions. In this review, we provide an overview of the currently recognized small-molecule plant hormones, signal peptide hormones, and plant growth regulators, along with the analytical methods employed for their analysis. We delve into the latest advancements in mass spectrometry imaging and in situ fluorescence techniques, which enable the examination of the spatial distribution of plant hormones. The advantages and disadvantages of these imaging techniques are further discussed. Finally, we propose potential avenues for future research in this field to further enhance our understanding of plant hormone biology., (© The Author(s) 2024. The olive fruit fly, Bactrocera oleae Rossi, is an important monophagous and multigenerational pest of olive fruit. Considering the damage caused by this pest, in this research, the effect of some plant hormones to reduce the pest population on Fishmi variety was investigated in the form of a complete randomized block design during the years 2021 and 2022. Trees were sprayed with jasmonic acid (1 and 2 mg/liter), salicylic (1 and 2 mM) two times before flowering and fruit growth. The results of investigating the density of different immature growth stages of this pest (eggs, first-instar, second-instar, third-instar larvae and pupae) in two years of the experiment showed that the effect of the year was significant only in the egg trait, and the effect of the treatment in other traits with significant differences (except the eggs and larvae of the first ages) were at the probability level of one percent. Examining the morphological characteristics of the fruit showed that the effect of the treatment on the length, weight, volume, density and weight of the flesh of the fruit; And the effect of the treatment in the year, only in the length and width of the fruit and kernel, and the kernel weight were significant at the probability level of 1%. Therefore, jasmonic acid and salicylic acid compounds can disrupt the life cycle and reduce pest damage by affecting some morphological traits and increasing the amounts of secondary metabolites of the fruit, by creating antibiosis resistance.

To study the mechanisms of growth and development, it is necessary to analyze the dynamics of the tissue patterning regulators in time and space and to take into account their effect on the cellular dynamics within a tissue. Plant hormones are the main regulators of the cell dynamics in plant tissues; they form gradients and maxima and control molecular processes in a concentration-dependent manner. Here, we present DyCeModel, a software tool implemented in MATLAB for one-dimensional simulation of tissue with a dynamic cellular ensemble, where changes in hormone (or other active substance) concentration in the cells are described by ordinary differential equations (ODEs). We applied DyCeModel to simulate cell dynamics in plant meristems with different cellular structures and demonstrated that DyCeModel helps to identify the relationships between hormone concentration and cellular behaviors. The tool visualizes the simulation progress and presents a video obtained during the calculation. Importantly, the tool is capable of automatically adjusting the parameters by fitting the distribution of the substance concentrations predicted in the model to experimental data taken from the microscopic images. Noteworthy, DyCeModel makes it possible to build models for distinct types of plant meristems with the same ODEs, recruiting specific input characteristics for each meristem. We demonstrate the tool’s efficiency by simulation of the effect of auxin and cytokinin distributions on tissue patterning in two types of Arabidopsis thaliana stem cell niches: the root and shoot apical meristems. The resulting models represent a promising framework for further study of the role of hormone-controlled gene regulatory networks in cell dynamics.
