Abstract

An alternative pathway, which involves several serine proteases for the production of angiotensin II (ANG II), exists in human detrusor smooth muscle. This serine protease in human detrusor smooth muscle changes ANG I to ANG II. Serial protease inhibitors such as soybean trypsin inhibitor and chymostatin completely block ANG II formation, which remains uninhibited by angiotension-converting enzyme.

23. Item-Level Analysis of Category Fluency Test Performance: A Systematic Review and Meta-Analysis of Studies of Normal and Neurologically Abnormal Ageing.

Abstract

While Category Fluency (CF) is widely used to help profile semantic memory, item-level scoring (ILS) approaches to this test have been proposed to obtain indices that are less influenced by non-semantic supportive functions. We systematically reviewed the literature to test the hypotheses that (1) compared with healthy adults, individuals with a clinical diagnosis suggestive of neurodegeneration generate words of lower semantic complexity; (2) compared with young adults, older adults generate words of higher semantic complexity. We searched six databases (date of search: 8 December 2023) for studies that relied on CF and ILS methods, in normal ageing and in age-associated neurodegeneration. Thirty-four studies were shortlisted: 27 on neurodegenerative conditions; 7 on normal ageing. Risk of bias was evaluated via a published checklist. Data were presented via qualitative synthesis. Most studies reported words of lower semantic complexity in relation to at least one item-level feature in individuals with mild cognitive impairment (MCI), Alzheimer's dementia (AD), and other neurodegenerative diseases. Post-hoc meta-analyses focussing on the MCI/AD continuum confirmed an effect on words' frequency (385 MCI/AD individuals and 350 controls; Hedges's G = 0.59) and age-of-acquisition (193 MCI/AD individuals and 161 controls; Hedges's G =  - 1.51). Studies on normal ageing, conversely, failed to demonstrate any overall effect. Most studies on MCI and AD have not relied on neurobiological diagnostic criteria. Moreover, only a small number of studies analysed ILS controlling for quantitative CF performance. Abstract

Within-individual blood oxygen level-dependent (BOLD) signal variability, intrinsic moment-to-moment signal fluctuations within a single individual in specific voxels across a given time course, is a relatively new metric recognized in the neuroimaging literature. Within-individual BOLD signal variability has been postulated to provide information beyond that provided by mean-based analysis. Synthesis of the literature using within-individual BOLD signal variability methodology to examine various cognitive domains is needed to understand how intrinsic signal fluctuations contribute to optimal performance. This systematic review summarizes and integrates this literature to assess task-based cognitive performance in healthy groups and few clinical groups. Included papers were published through October 17, 2022. Searches were conducted on PubMed and APA PsycInfo. Studies eligible for inclusion used within-individual BOLD signal variability methodology to examine BOLD signal fluctuations during task-based functional magnetic resonance imaging (fMRI) and/or examined relationships between task-based BOLD signal variability and out-of-scanner behavioral measure performance, were in English, and were empirical research studies. Data from each of the included 19 studies were extracted and study quality was systematically assessed. Results suggest that variability patterns for different cognitive domains across the lifespan (ages 7-85) may depend on task demands, measures, variability quantification method used, and age. As neuroimaging methods explore individual-level contributions to cognition, within-individual BOLD signal variability may be a meaningful metric that can inform understanding of neurocognitive performance. Further research in understudied domains/populations, and with consistent quantification methods/cognitive measures, will help conceptualize how intrinsic BOLD variability impacts cognitive abilities in healthy and clinical groups., Competing Interests: Declarations. Abstract

General cognitive function is substantially heritable across the human life course from adolescence to old age. We investigated the genetic contribution to variation in this important, health-and well-being-related trait in middle-aged and older adults. We conducted a meta-analysis of genome-wide association studies of 31 cohorts (N = 53 949) in which the participants had undertaken multiple, diverse cognitive tests. A general cognitive function phenotype was tested for, and created in each cohort by principal component analysis. We report 13 genome-wide significant single-nucleotide polymorphism (SNP) associations in three genomic regions, 6q16.1, 14q12 and 19q13.32 (best SNP and closest gene, respectively: rs10457441, P = 3.93 x 10(-9), MIR2113; rs17522122, P = 2.55 x 10(-8), AKAP6; rs10119, P = 5.67 x 10(-9), APOE/TOMM40). We report one gene-based significant association with the HMGN1 gene located on chromosome 21 (P = 1x10(-6)). These genes have previously been associated with neuropsychiatric phenotypes. Meta-analysis results are consistent with a polygenic model of inheritance. To estimate SNP-based heritability, the genome-wide complex trait analysis procedure was applied to two large cohorts, the Atherosclerosis Risk in Communities Study (N = 6617) and the Health and Retirement Study (N = 5976). The proportion of phenotypic variation accounted for by all genotyped common SNPs was 29% (s.e. = 5%) and 28% (s.e. = 7%), respectively. Using polygenic prediction analysis, similar to 1.2% of the variance in general cognitive function was predicted in the Generation Scotland cohort (N = 5487; P = 1.5 x 10(-17)). Abstract

The Enhancing NeuroImaging Genetics through Meta-Analysis (ENIGMA) Consortium is a collaborative network of researchers working together on a range of large-scale studies that integrate data from 70 institutions worldwide. Organized into Working Groups that tackle questions in neuroscience, genetics, and medicine, ENIGMA studies have analyzed neuroimaging data from over 12,826 subjects. In addition, data from 12,171 individuals were provided by the CHARGE consortium for replication of findings, in a total of 24,997 subjects. By meta-analyzing results from many sites, ENIGMA has detected factors that affect the brain that no individual site could detect on its own, and that require larger numbers of subjects than any individual neuroimaging study has currently collected. ENIGMA's first project was a genome-wide association study identifying common variants in the genome associated with hippocampal volume or intracranial volume. Continuing work is exploring genetic associations with subcortical volumes (ENIGMA2) and white matter microstructure (ENIGMA-DTI). Working groups also focus on understanding how schizophrenia, bipolar illness, major depression and attention deficit/hyperactivity disorder (ADHD) affect the brain. We review the current progress of the ENIGMA Consortium, along with challenges and unexpected discoveries made on the way.

38. Leisure-time activity in old age as predictors of impending dementia: A 15-year prospective study.

Abstract

This study examined the relationship between leisure activities and risk of dementia in a sample of healthy older individuals, dementia free at the beginning of the project. Data were drawn from a population-based longitudinal study (the Betula project) and the participants were followed up for 15 years. At baseline, participants were asked about their frequency of participation in 15 selected leisure activities. When age, gender, education, APOE and other potential confounders were controlled for, results revealed quite moderate effects on dementia after analysis of the activities separately. However, by weighting each activity into a mental, social and physical dimension (based on valuation by the participants), and then summarizing into a score for each dimension, we further investigated if level of engagement could predict impending dementia. Preliminary results indicate that the dimensions may have influence on the risk of dementia for certain age groups. The study also showed that the strongest predictor of dementia is being a carrier of the APOE ɛ4 allele. The outcomes are discussed in terms of important methodological difference between studies concerning the effects of leisure activities in preventing dementia diseases.

41. Cognitive Status in Persons with Amalgam-related Complaints.

Abstract

Self-reported cognitive symptoms are frequent in persons with amalgam-related complaints, but few studies have focused on their cognitive function. The aim was to examine a symptom profile and whether participants with amalgam-related complaints have cognitive deficits in comparison with control individuals. We drew 342 participants with amalgam-related complaints and 342 one-to-one matched control individuals from a longitudinal population-based study. For 81 of the participants with amalgam-related complaints and controls, data were available approximately five years before the onset of complaints, making a longitudinal analysis possible. All participants were assessed by a self-reported health questionnaire and a comprehensive cognitive test battery. The participants with amalgam-related complaints reported more symptoms, mainly musculoskeletal and neuropsychological, compared with control individuals (p < 0.001). The results revealed no significant difference between the amalgam and control group, either cross-sectionally or longitudinally, for any of the cognitive tests. These results suggest that cognitive decline is not associated with amalgam-related complaints. [ABSTRACT FROM PUBLISHER]

Abstract

This paper presents the cognitive test battery used in the CASCADE Study (Cardiovascular Determinants of Dementia) for examining the consequences of cerebral white matter lesions and atrophy. The test battery includes nine different tasks assessing memory, executive function, and global cognitive function. Three episodic memory tasks were used in combinations to assess the role of attention and speed on encoding. Estimates of short and long-term memory capacity were also derived from these three memory tasks. Semantic memory production / frontal lobe functions were assessed by means of a word fluency test. The Letter Digit Substitution test and the Stroop test were used to assess speed of processing and attention. Motor speed was measured with the Purdue Pegboard test, and global cognitive function was assessed by the Mini Mental State Examination. Overall performance data for the whole CASCADE sample and for each of eight study centers are presented for each test. Possible reasons for performance differences among study centers are discussed. Abstract

Two representative samples of adult Norwegians (n=2000) were asked a set of general and specific questions regarding their beliefs and opinions about human memory. The results indicate that on many questions, such as time of the earliest memories, inhibiting effects of collaboration, and memory for dramatic versus ordinary events, the views of the general public concurred with current research findings, and people in general had realistic views about their own memory performance. On other questions, such as the reliability of olfactory as compared with visual and auditory memory, the memory of small children in comparison with that of adults, the likelihood of repression of adult traumatic memories, and on more general questions such as the possibility of training memory and the capacity limitations of long-term memory, a large proportion of the participants expressed views that are less supported by scientific evidence. Implications of these findings are briefly discussed. © 2006 Psychology Press Ltd.

Abstract

Malfunctioning of cyclin-dependent kinase 5 (CDK5) through aberrant proteolytic cleavage of its neuronal activators p35 and p39 is involved in neurodegeneration in Alzheimer's disease (AD) and other neurodegenerative brain diseases. By extensive genetic analysis of the genes encoding CDK5 (CDK5), p35 (CDK5RI) and p39 (CDK5R2), we excluded causal mutations in 70 familial early-onset AD patients. We performed an association study with five informative SNPs in CDK5 in two independent samples of early-onset AD patients and matched control individuals from The Netherlands and northern Sweden. Association was observed with g.149800G > C in intron 5 of CDK5, and a two times increased risk was observed in both patient samples for carriers of the C-allele. Our data are indicative for a role of the CDK5 molecular complex in the genetic etiology of early-onset AD, and suggest that a yet unknown functional variant in CDK5 or in a nearby gene might lead to increased susceptibility for early-onset AD.
