School health services have been described as the "hidden healthcare" system because of their isolation within schools and from other healthcare providers. This isolation may inhibit innovations in school healthcare delivery. Hence, there is a need to identify and characterize various delivery models. This review examines models to identify innovative approaches and formulate suggestions for public health departments, local and state educational agencies, and policymakers. Toward this goal, published and gray literature were studied and synthesized, identifying three delivery models: the traditional model of school-based nursing and two alternative models: school-based health centers and community partnerships. Mechanisms of delivery included telehealth, mobile clinics, and system-level care. Although no innovative, comprehensive approaches to school health services models were found, innovation generally focused on improving equitable delivery to vulnerable populations. Policies must be formulated and funded to integrate such innovations into a comprehensive, preventative approach, including improved care coordination and data sharing. [ABSTRACT FROM AUTHOR]

Ixodes ricinus is an important vector of infectious human and livestock diseases in Europe. Co-infections of pathogens in ticks and hosts have been reported. Tick cell lines offer a useful model system for study of co-infections. We present a review of the existing literature on co-infections in tick cell lines. Previous studies have demonstrated the usefulness of tick cell lines in studies on co-infection of different pathogens and their interaction with the tick microbiome. We also carried out a preliminary study to investigate the effects of co-culturing Borrelia burgdorferi and Anaplasma phagocytophilum on their growth and interactions with the Ixodes ricinus cell line IRE/CTVM19 over a 13-day period. Replication of both pathogens was quantified by real-time PCR. The presence of A. phagocytophilum appeared to have a slight inhibitory effect on the multiplication of B. burgdorferi, that were added subsequently. In contrast, the prior presence of B. burgdorferi appeared to have a stimulatory effect on A. phagocytophilum after 6 days in culture. We conclude that the IRE/CTVM19 tick cell line is suitable for simultaneous and continuous cultivation of both bacteria and can be applied in future research. [ABSTRACT FROM AUTHOR]

Estimating mortality of sea turtles incidentally captured in fishing gear is essential to understanding fishery impacts on sea turtle populations. Loggerhead turtles (Caretta caretta) are a component of bycatch in pelagic longline fisheries, including those operating in Atlantic Canada. In this study, we used pop-up satellite archival tags (PSATs) attached to 62 loggerhead turtles incidentally hooked in Atlantic Canadian shallow-set pelagic longline gear (2012-2018) to estimate associated post-release survival. Consistent with conventional fishing practices, hooks were not removed from turtles prior to release. Analysis of diving behaviour, ocean temperature, and ambient light level was used to assign fates to turtles. Post-release survival was calculated using the Kaplan-Meier estimator with right censoring. Based on this method, the annual survival probability of an individual loggerhead post-release is 0.877 ± SE 0.052. While it is commonly assumed that hook location is an important variable in predicting fates of turtles after release, there was no difference in survival of deep-hooked (swallowed hook) versus shallow-hooked turtles. These results indicate higher survivorship among hooked loggerheads captured in shallow-set pelagic longline gear than prevailing wisdom and suggest that at least some of the population impacts on loggerhead turtles conventionally attributed to pelagic longline interactions may be linked to other threats. Objectives This study seeks to comprehensively analyze the impact of smoking history on outcomes after endoscopic transsphenoidal hypophysectomy (TSH) for pituitary adenoma. Design This was a retrospective study. Setting This study was done at the tertiary care center. Participants Three hundred and ninety-eight adult patients undergoing TSH for a pituitary adenoma. Main Outcome Measures Clinical and tumor characteristics and operative factors were collected. Patients were categorized as never, former, or active smokers, and the pack-years of smoking history was collected. Years since cessation of smoking was obtained for former smokers. Specific outcomes included postoperative cerebrospinal fluid (CSF) leak, length of hospitalization, 30-day return to the operating room, and 30-day readmission. Smoking history details were comprehensively analyzed for association with outcomes. Results Any history of smoking tobacco was associated with return to the operating room (odds ratio [OR] = 2.67, 95% confidence interval [CI]: 1.05–6.76, p = 0.039), which was for persistent CSF leak in 58.3%. Among patients with postoperative CSF leak, any history of smoking was associated with need for return to the operating room to repair the CSF leak (OR = 5.25, 95% CI: 1.07–25.79, p = 0.041). Pack-years of smoking was positively associated with a return to the operating room (OR = 1.03, 95% CI: 1.01–1.06, p = 0.048). In all multivariable models, all negative outcomes were significantly associated with the covariate: occurrence of intraoperative CSF leak. Conclusion This is the first study to show smoking may have a negative impact on healing of CSF leak repairs after TSH, requiring a return to the operating room. This effect appears to be dose dependent on the smoking history. Secondarily, intraoperative CSF leak as covariate in multivariable models was significantly associated with all negative outcomes. This systematic review and meta-analysis aimed to compare the effect of photobiomodulation (PBM) therapy on implant stability and crestal bone loss placed in healed sites. The present systematic review and meta-analysis were conducted according to PRISMA (The Preferred Reporting Items for Systematic Reviews and Meta-Analyses) guidelines. Two investigators carried out the electronic search of Pubmed, Google Scholar, and Ebscohost for published literature from 2012 till March 2024. Handsearch for articles relevant to the topic was also carried out along with references. Only randomized controlled trials were included for systematic review. The assessment quality was done using the revised JBI checklist (Joanna Biggs Institute). After screening and eligibility assessment, Qualitative analysis was envisaged for 15 studies on implant stability and crestal bone loss, followed by quantitative analysis of 8 studies on implant stability and only 2 studies for crestal bone loss. Meta-analysis showed that overall implant stability was significantly higher in the control group (P < 0.00001). Crestal bone loss showed a significant difference after the therapy when observed after six months of implant placement (P = 0.03). Heterogeneity was high with the random effects model for implant stability, while it was acceptable for crestal bone loss. However, more studies with similar parameters concerning implant stability and PBM characteristics are required for further subgroup analysis and to reach conclusive results., Competing Interests: Declarations. Corpus Christi Bay is a shallow, wind-driven lagoon located on the semi-arid South Texas coast that has a rapidly urbanizing watershed. Projections indicate that this region will become warmer and drier and will support an increasing urban population over the next several decades. Here, a 27-month field study was undertaken to quantify phytoplankton biovolume, community composition, and relationships with environmental drivers. Phytoplankton biovolume varied unimodally with a peak in biovolume from spring through summer followed by a decline into fall and winter. Phytoplankton growth was related to nutrient availability during the spring and summer, while water temperature and factors affecting flushing were important during the fall and winter. Regions with more restricted circulation patterns (i.e., man-made canals) were found to support higher standing crops of phytoplankton and the occurrence of high biovolume blooms. Diatoms were dominant during the winter and spring, dinoflagellates were dominant during the summer and fall, and picophytoplankton groups were important during spring, summer, and fall. These results suggest that nutrient and physical conditions interact to determine phytoplankton biomass and community composition and contribute to our ability to project potential impacts of future increases in human populations in the watershed, decreasing precipitation due to climate change, and increasing frequency of short-lived flood events. Phosphoinositide‐3‐kinase/AKT (PI3K/AKT) signaling plays key roles in the regulation of cellular activity in both health and disease. In immune cells, this PI3K/AKT pathway is critically regulated by the phosphoinositide phosphatase SHIP1, which has been reported to modulate the function of most immune subsets. In this review, we summarize our current knowledge of SHIP1 with a focus on innate immune cells, where we reflect on the most pertinent aspects described in the current literature. We also present several small‐molecule agonists and antagonists of SHIP1 developed over the last two decades, which have led to improved outcomes in several preclinical models of disease. We outline these promising findings and put them in relation to human diseases with unmet medical needs, where we discuss the most attractive targets for immune therapies based on SHIP1 modulation. This article sets out to broaden our knowledge of the sociopolitical dynamics of Santarém (AD 1000–1600), a regional society in the Lower Amazon, by synthesizing the existing archaeological data relating to settlement patterns, ritual ceramics, prestige goods and chronology, as well as exploring aspects linked to the technology of ceramic production at the Carapanari site, a small-scale community located in the region during the late precolonial period. Using an integrated approach, the research combines a techno-functional analysis of a sample basically composed of ceramic fragments, providing information on the original forms and possible uses, with microscopy and compositional analyses of fragments based on instrumental neutron activation analysis. This enables the identification of technological choices, processes of innovation and behavioral changes, also present at other sites in the region that are expressed over time. The set of information presented here engages with recent debates on the emergence of complex societies, providing some insight into the historical development of this polity in Amazonia during the late precolonial period. Background: Systemic lupus erythematosus (SLE) is a chronic autoimmune disease. The programmed cell death 1 (PDCD1) gene, which encodes the PD1 protein, is one of the many genes involved in susceptibility to SLE. The PD1 protein is an inhibitory immunoreceptor that plays a key role in maintaining immunological tolerance to self-antigens. The PD1.5 C/T polymorphism (rs2227981, c.804 T > C) alters the expression and function of the PD-1 receptor. Objectives: This case-control study aimed to determine the association between the rs2227981 polymorphism of the PDCD1 gene and susceptibility to SLE in the northwestern population of Iran. Methods: The rs2227981 genotype was determined in 52 SLE patients and 53 controls using the PCR-RFLP method with the Pvu II restriction enzyme. Results: The study found a significant association between the rs2227981 polymorphism and susceptibility to SLE in the study population. Neuroblastoma (NB), a childhood cancer arising from the neural crest, poses significant clinical challenges, particularly in cases featuring amplification of the MYCN oncogene. Epigenetic factors play a pivotal role in normal neural crest and NB development, influencing gene expression patterns critical for tumorigenesis. This review delves into the multifaceted interplay between MYCN and known epigenetic modifications during NB genesis, shedding light on the intricate regulatory networks underlying the disease. We provide an extensive survey of known epigenetic mechanisms, encompassing DNA methylation, histone modifications, non-coding RNAs, super-enhancers (SEs), bromodomains (BET), and chromatin modifiers in MYCN-amplified (MNA) NB. These epigenetic changes collectively contribute to the dysregulated gene expression landscape observed in MNA NB. Furthermore, we review emerging therapeutic strategies targeting epigenetic regulators, including histone deacetylase inhibitors (HDACi), histone methyltransferase inhibitors (HMTi), and DNA methyltransferase inhibitors (DNMTi). Background: Offspring of parents with affective disorders (OAD) are at risk of developing a wide range of mental disorders. Deficits in the rearing environment and high levels of stress are well-known risk factors for negative outcomes in OAD. Building on prior research, we aim to examine the longitudinal relationships between family dysfunction, stressful life events, and mental health in OAD and control offspring of parents with no affective disorder. In the present study, we hypothesized that high levels of family dysfunction would be associated with more internalizing and externalizing problems across time in OAD than in controls, and that family dysfunction would mediate the relationship between stressful life events in adolescence and poor mental health in adulthood, particularly in OAD., Methods: As part of the TRacking Adolescents' Lives Survey (TRAILS), 2230 participants (51% female, M age  = 11.1 years, SD = 0.6, at baseline) and their parents completed measures across six time points, spanning 15 years. Mental health, family dysfunction, and stressful life events were assessed with the Youth and Adult Self-Report, Family Assessment Device, and an in-house measure, respectively., Results: Multi-group structured equation modeling revealed that family dysfunction was linked to internalizing and externalizing problems in OAD, but not controls, across time. We investigated the potential protective effects of silymarin (SLY) on doxorubicin (DOX) induced chronic cardiotoxicity in mice. We used 30 male BALB/c mice assigned randomly to four experimental groups: control group administered normal saline orally daily and intraperitoneally (i.p.) twice/week during weeks 1, 2, 5 and 6; SLY group (was administered 100 mg/kg SLY daily by oral gavage for 6 weeks; DOX group was administered 3 mg/kg DOX i.p. twice/week during weeks 1, 2, 5 and 6; DOX + SLY group administered DOX and SLY corresponding to the DOX and SLY groups. At the end of the experiment, heart tissues were collected for analysis. Cardiomyopathy was observed in the DOX group; this damage was reduced by SLY treatment. SLY administration in DOX treated mice decreased topoisomerase IIβ (TopIIβ) expression as indicated by qPCR and immunostaining. Immunohistochemistry and western blot analysis revealed decreased phosphorylated histone-2AX (γH2Ax) expression in the SLY + DOX group. SLY administration combined with DOX increased cardiac troponin T and I (cTnT and cTnI) expression based on immunohistochemical and western blot analyses. SLY administration with DOX was cardioprotective by reducing double-strand DNA breakage by blocking the DOX-TopIIβ-DNA cleavage complex in response to down-regulation of TopIIβ expression. Evaluating estuary water quality responses to reductions (or increases) in nutrient loading attributed to on the ground management actions can be challenging due to the strong influence of environmental drivers on nutrient loads and non-linear relationships. This study applied generalized additive models to calculate watershed nutrient loads and assess responses in estuary water quality to seasonally-adjusted freshwater inflow and flow-adjusted nutrient loads in Lavaca Bay, Texas. Lavaca Bay is a secondary embayment on the Texas coast displaying early potential for eutrophication and water quality degradation. Use of flow-adjusted nutrient loads allowed the study to evaluate the response in water quality to changes in nutrient loads driven by anthropogenic sources. Cross-validation indicated that, despite data constraints, semiparametric models performed well at nutrient load prediction. Based on these models, delivered annual nutrient loads varied substantially from year to year. In contrast, minimal changes in flow-normalized loads indicate that nutrient loadings were driven by natural variation in precipitation and runoff as opposed to changes in management of nonpoint sources. Models indicated no evidence of long-term changes in dissolved oxygen or chlorophyll-a within Lavaca Bay. However, site specific long-term increases in both organic and inorganic nitrogen are concerning for their potential to fuel eutrophication. Further analysis found freshwater inflow had strong influences on nutrient and chlorophyll-a concentrations but there was no evidence that changes in watershed nutrient loading explained additional variation in dissolved oxygen and limited evidence that watershed nutrient loadings explained chlorophyll-a concentrations. In addition to providing a baseline assessment of watershed nutrient loading and water quality responses in the Lavaca Bay watershed, this study provides methodological support for the use of semiparametric models in load regression models and estuary assessments. Introduction: Malignant pleural mesothelioma (MPM) is a neoplasm with dismal prognosis and notorious resistance to the standard therapeutics cisplatin and pemetrexed. Chalcone derivatives are efficacious anti-cancer agents with minimal toxicity and have, therefore, gained pharmaceutical interest. Here, we investigated the efficacy of CIT-026 and CIT-223, two indolyl-chalcones (CITs), to inhibit growth and viability of MPM cells and defined the mechanism by which the compounds induce cell death. Methods: The effects of CIT-026 and CIT-223 were analyzed in five MPM cell lines, using viability, immunofluorescence, real-time cell death monitoring, and tubulin polymerization assays, along with siRNA knockdown. Phospho-kinase arrays and immunoblotting were used to identify signaling molecules that contribute to cell death. Results: CIT-026 and CIT-223 were toxic in all cell lines at sub-micromolar concentrations, in particular in MPM cells resistant to cisplatin and pemetrexed, while normal fibroblasts were only modestly affected. Both CITs targeted tubulin polymerization via (1) direct interaction with tubulin and (2) phosphorylation of microtubule regulators STMN1, CRMP2 and WNK1. Formation of aberrant tubulin fibers caused abnormal spindle morphology, mitotic arrest and apoptosis. CIT activity was not reduced in CRMP2-negative and STMN1-silenced MPM cells, indicating that direct tubulin targeting is sufficient for toxic effects of CITs. Discussion: CIT-026 and CIT-223 are highly effective inducers of tumor cell apoptosis by disrupting microtubule assembly, with only modest effects on nonmalignant cells. CITs are potent anti-tumor agents against MPM cells, in particular cells resistant to standard therapeutics, and thus warrant further evaluation as potential small-molecule therapeutics in MPM. Tissue and blood gas embolism (GE) associated with fisheries bycatch are likely a widespread, yet underestimated, cause of sea turtle mortality. Here, we evaluated risk factors associated with tissue and blood GE in loggerhead turtles caught incidentally by trawl and gillnet fisheries on the Valencian coastline of Spain. Of 413 turtles (303 caught by trawl, 110 by gillnet fisheries), 54% (n  = 222) exhibited GE. For sea turtles caught in trawls, the probability and severity of GE increased with trawl depth and turtle body mass. In addition, trawl depth and the GE score together explained the probability of mortality (P[mortality]) following recompression therapy. Specifically, a turtle with a GE score of 3 caught in a trawl deployed at 110 m had a P[mortality] of ~50%. For turtles caught in gillnets, no risk variables were significantly correlated with either the P[GE] or GE score. However, gillnet depth or GE score, separately, explained P[mortality], and a turtle caught at 45 m or with a GE score between 3 and 4 had a P[mortality] of 50%. Differences in the fishery characteristics precluded direct comparison of GE risk and mortality between these gear types. Although P[mortality] is expected to be significantly higher in untreated turtles released at sea, our findings can improve estimates of sea turtle mortality associated with trawls and gillnets, and help guide associate conservation efforts. Climate change is a primary driver of tropical mangroves colonizing temperate salt marshes in the Gulf of Mexico. Studies indicate that threshold temperatures of adult and sapling Avicennia germinans survival range from −7 to −10 °C and survival can depend on life history stage during a freeze event. We performed a growth chamber study to explore responses to varying freezing temperature regimes when produced at two different latitudes with varying salinity and nutrient conditions. Propagules were collected from Corpus Christi and South Padre Island, Texas. In Corpus Christi Bay, propagules were harvested from fertilized maternal shrubs (nitrogen, phosphorus, and controls) in an ongoing fertilization experiment (2017 to present). A 2-h hard freeze treatment (−8 °C), resulted in 20% survival across latitude and nutrient treatments. The lower latitude propagules, which were harvested from the hypersaline South Padre Island A. germinans, had higher survival rates after –2 °C exposure, grew significantly taller as seedlings over 20 weeks, and produced more leaves after −8 °C exposure. Propagules from nitrogen-treated shrubs grew taller following −2 °C exposure. In all treatments, propagule weight was a significant covariate predictor of seedling height and leaf production. Avicennia germinans populations may not be decimated by a single −8 °C freeze, and populations could possibly experience rapid recruitment from surviving propagules after a freeze, leading to local adaptation. Growth benefits from fertilized parent plants indicate that propagules from high nutrient areas may be "hotspots" of future A. germinans expansion. [ABSTRACT FROM AUTHOR]

Urticaria is characterized by the occurrence of wheals and flares in response to vasoactive mediators, such as histamine. Various studies have suggested the involvement of basophils in the pathogenesis of chronic spontaneous urticaria (CSU). However, histamine release from peripheral basophils in response to stimuli acting on the high affinity IgE receptor (FcϵRI) is impaired in many patients with CSU (non/low responders). We previously demonstrated that tissue factor (TF)s expressed on vascular endothelial cells in response to a combination of various stimuli, such as that of histamine and lipopolysaccharide (LPS), activates the extrinsic coagulation pathway and produces anaphylatoxin, complement 5a (C5a), which then activates basophils and mast cells via the C5a receptor (C5aR). We have revealed that histamine release was induced in response to C5a and formyl-l-methionyl-l-leucyl-l-phenylalanine (fMLP), regardless of the response to anti-IgE antibody, the reduced numbers of basophils and severity of urticaria. Moreover, we found that spontaneous release of histamine ex vivo from basophils of patients with CSU is higher than that from healthy individuals. These results suggest that basophils and the complement system, which could be activated by coagulation factors, may play a critical role in the pathogenesis of CSU, especially in cases refractory to treatment involving the IgE/FcϵRI pathway. Introduction: Chronic inducible urticaria (CIndU) is a subgroup of chronic urticaria induced by a specific stimulus. We evaluated basophil characteristics in patients with CIndU and compared with those in patients with chronic spontaneous urticaria (CSU) and healthy controls (HCs). Methods: Blood was collected from patients, and a basophil activation test (BAT) was performed. Basophil responsiveness and surface marker expression in patients with CIndU were compared with those in patients with CSU and HCs. For some patients with CIndU, blood was collected before and after wheals were induced. In these cases, we compared the responsiveness of basophils before and after the appearance of wheals. Result: HCs (n=23) and patients with CIndU (n=24) or CSU (n=38) were enrolled in the study. The degree of basophil activation at steady state in patients with CIndU was higher than in HCs. Basophil responsiveness via highaffinity IgE receptor (FcϵRI) stimulation with anti-IgE or anti-FcϵRI antibody in patients with CIndU was equivalent to that in HCs, and higher than that in patients with CSU. No abnormalities in IgE and FcϵRI expressions on the surface of basophils in patients with CIndU were observed. When we induced wheals in some patients with CIndU and performed a BAT before and after the appearance of wheals, no significant changes were found. Conclusion: Peripheral blood basophils in CIndU were slightly activated at steady state, but no abnormalities in basophil responsiveness. Tick cell lines are important tools for research on ticks and the pathogens they transmit. Here, we report the establishment of ten new cell lines from European ticks of the genera Argas, Dermacentor, Hyalomma, Ixodes and Rhipicephalus originating from Germany and Spain. For each cell line, the method used to generate the primary culture, a morphological description of the cells and species confirmation by sequencing of the partial 16S rRNA gene are presented. Further molecular analysis of the two new Ixodes ricinus cell lines and three existing cell lines of the same species revealed genetic variation between cell lines derived from ticks collected in the same or nearby locations. Collectively, these new cell lines will support research into a wide range of viral, bacterial and protozoal tick-borne diseases prevalent in Europe. [ABSTRACT FROM AUTHOR]

Chronic spontaneous urticaria (CSU) is defined as recurrent episodes of spontaneous wheal development and/or angioedema for more than six weeks and at least twice a week. The core link in the pathogenesis of CSU is the activation of mast cells, T cells, eosinophils, and other immune cells infiltrating around the small venules of the lesion. Increased vascular permeability, vasodilatation, and recruitment of inflammatory cells directly depend on mast cell mediators' release. Complex regulatory systems tightly influence the critical roles of mast cells in the local microenvironment. The bias toward Th2 inflammation and autoantibodies derived from B cells, histamine expressed by basophils, and initiation of the extrinsic coagulation pathway by eosinophils or monocytes exerts powerful modulatory influences on mast cells. Cell-to-cell interactions between mast cells and eosinophils/T cells also are regulators of their function and may involve CSU's pathomechanism. This review summarizes up-to-date knowledge regarding the crosstalk between mast cells and other immune cells, providing the impetus to develop new research concepts and treatment strategies for CSU. Children with parents suffering from a psychiatric disorder are at higher risk for developing a mental disorder themselves. This systematic review and meta-analysis of randomized controlled trials aims to evaluate the efficacy of psychosocial interventions to prevent negative mental health outcomes in the offspring of parents with mental illness. Eight electronic databases, grey literature and a journal hand-search identified 14 095 randomized controlled trials with no backward limit to June 2021. Outcomes in children included incidence of mental disorders (same or different from parental ones) and internalizing and externalizing symptoms at post-test, short-term and long-term follow-up. Relative risks and standardized mean differences (SMD) for symptom severity were generated using random-effect meta-analyses. Twenty trials were selected (pooled n = 2689 children). The main therapeutic approaches found were cognitive-behavioural therapy and psychoeducation. A significant effect of interventions on the incidence of mental disorders in children was found with a risk reduction of almost 50% [combined relative risk = 0.53, 95% confidence interval (CI) 0.34–0.84]. Interventions also had a small but significant effect on internalizing symptoms at post-test (SMD = −0.25, 95% CI −0.37 to −0.14) and short-term follow-up (−0.20, 95% CI −0.37 to −0.03). For externalizing symptoms, a decreasing slope was observed at post-test follow-up, without reaching the significance level (−0.11, 95% CI −0.27 to 0.04). Preventive interventions targeting the offspring of parents with mental disorders showed not only a significant reduction of the incidence of mental illness in children, but also a diminution of internalizing symptoms in the year following the intervention. Sea turtles, like other air-breathing diving vertebrates, commonly experience significant gas embolism (GE) when incidentally caught at depth in fishing gear and brought to the surface. To better understand why sea turtles develop GE, we built a mathematical model to estimate partial pressures of N2 (PN2), O2 (PO2), and CO2 (PCO2) in the major body-compartments of diving loggerheads (Caretta caretta), leatherbacks (Dermochelys coriacea), and green turtles (Chelonia mydas). This model was adapted from a published model for estimating gas dynamics in marine mammals and penguins. To parameterize the sea turtle model, we used values gleaned from previously published literature and 22 necropsies. Next, we applied this model to data collected from free-roaming individuals of the three study species. Finally, we varied body-condition and cardiac output within the model to see how these factors affected the risk of GE. Our model suggests that cardiac output likely plays a significant role in the modulation of GE, especially in the deeper diving leatherback turtles. This baseline model also indicates that even during routine diving behavior, sea turtles are at high risk of GE. This likely means that turtles have additional behavioral, anatomical, and/or physiologic adaptions that serve to reduce the probability of GE but were not incorporated in this model. Objective: To identify biomarkers related to head and neck squamous cell carcinoma (HNSCC) metastasis and establish a prognostic model for patients with HNSCC. Methods: HNSCC mRNA expression data of metastasis and non-metastatic samples were downloaded from The Cancer Genome Atlas (TCGA) and Gene Expression Omnibus (GEO) databases. After screening the differentially expressed genes (DEGs) in the two datasets, a prognostic model, including clinical factors and biomarkers, was established, and verified in 36 samples of HNSCC by quantitative real-time transcription (qRT)-PCR. Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), and Gene sets enrichment analysis (GSEA) were consulted to explore the functions of the DEGs. Results: In total, 108 DEGs were identified. GSEA, GO, and KEGG analyses showed that these DEGs were mainly involved in the proliferation and metastasis of HNSCC. Six genes that were significantly related to metastasis, immune cell infiltration and prognosis were further identified to construct a prognostic gene signature. The reliability of the gene signature was verified in 36 samples of HNSCC. A prognostic model, including tumor stage, risk level, and a nomogram for prediction were further established. Receiver operating characteristic (ROC) analysis, decision curve analysis (DCA), C-index, and calibration plots showed that the model and nomogram perform well. Background: We have focused on the alteration of the PD-1/PD-L1 pathway in celiac disease and discussed the roles of the PD1 pathway in regulating the immune response. We explored the idea that the altered mRNA splicing process in key regulatory proteins could represent a novel source to identify diagnostic, prognostic, and therapeutic targets in celiac disease. Methods: We characterized the PD1 mRNA variants' profile in CD patients and in response to gluten peptides' incubation after in vitro experiments. Total RNA from whole blood was isolated, and the coding region of the human PD-1 mRNA was amplified by cDNA PCR. Results: PCR amplification of the human PD-1 coding sequence revealed an association between the over-expression of the sPD-1 protein and the PD-1Δex3 transcript in celiac disease. Thus, we have found three novel alternative spliced isoforms, two of which result in a truncated protein and the other isoform with a loss of 14 aa of exon 2 and complete exon 3 (Δ3) which could encode a new soluble form of PD1 (sPD-1). Conclusions: Our study provides evidence that dietary gluten can modulate processes required for cell homeostasis through the splicing of pre-mRNAs encoding key regulatory proteins, which represents an adaptive mechanism in response to different nutritional conditions. Objectives: To systematically describe the characteristics and techniques of prevention programmes for children of parents with mood/anxiety disorders. In addition, recruitment approaches and difficulties were identified and a meta‐analysis was conducted to examine the efficacy of these prevention programmes. Methods: Randomized controlled trials assessing the efficacy of a prevention programme for children (6–25 years) of parents with mood and/or anxiety disorders were included. A systematic literature search was conducted in PubMed, PsychINFO, and CENTRAL from the earliest record to March 2019. In addition, programme manuals of identified prevention programmes were requested for a content analysis. Results: Twenty‐two articles containing eight unique prevention programmes involving 1,325 subjects were identified. Programmes varied in the number and types of techniques, but all provided psychoeducation. Results suggested that recruitment via clinicians was more successful than recruitment via health maintenance organization databases. In a meta‐analysis, a significant risk difference was found in favour of prevention programmes on the risk of developing a depressive/anxiety disorder in offspring at short‐term (9–18 months follow‐up; RR = 0.37, 95% CI [0.21; 0.66]) and long‐term follow‐up (24 months or longer follow‐up; RR = 0.71, 95% CI [0.57; 0.87] and on symptom levels in offspring at post‐intervention (SMD = −0.19, 95% CI [−0.36; −0.02]) and at 12‐months follow‐up (SMD = −0.31, 95% CI [−0.57; −0.06]). Conclusions: The prevention programmes combined psychoeducational elements with skills training and/or cognitive‐behavioural therapy elements. The recruitment process and the content of these programmes are sometimes insufficiently described. Nevertheless, they appear to be effective, indicating a need to further examine how these programmes exactly work and for whom. Practitioner points: Preventive interventions for children of parents with mood/anxiety disorders appear to be effective in preventing these disorders in offspring.Available preventive intervention programmes focus mostly on psychoeducation, cognitive‐behavioural therapy, and family processes.More effort should be made into describing preventive interventions so that they can be easily implemented by practitioners.Studies should further examine why and for whom preventive interventions for children of parents with mood/anxiety disorders are effective. Tropical cyclones represent a substantial disturbance to water quality in coastal ecosystems via storm surge, winds, and flooding. However, evidence to date suggests that the impacts of tropical cyclones on water quality are generally short-lived (days-months) and that the magnitude of the disturbance is related to proximity to storm track. Discrete and continuous water samples were collected in three Texas estuaries before and after Hurricane Harvey made landfall in 2017. Of the three estuaries, the Guadalupe Estuary and its watershed received the highest rainfall totals and wind speeds. An ephemeral increase in salinity was observed (mean of 9.8 on 24 August 2017 to a peak of 32.1 on 26 August 2017) due to storm surge and was followed by a rapid decrease to < 1 as floodwaters reached the estuary. Salinity returned to pre-storm levels within 1 month. During the low salinity period, bottom water hypoxia developed and lasted for 9 days. In all three estuaries, there was an increase in inorganic nutrients post-Harvey, but the nutrients largely returned to pre-storm baseline levels by winter. The lack of long-term water quality impacts from Harvey despite its severity corroborates previous findings that estuarine water quality tends to return to baseline conditions within days to a few months after storm passage. The year 2017 was pivotal for women because of the #MeToo movement's global momentum. People were alerted to the abuses of power and sexual harassment by male authorities in different fields such as government, entertainment and industry. These scandals also helped bring attention to the objectification of women's normalised bodies and their subjugation in patriarchal cultures. Yet neither this disempowerment of women nor women's coming forward new, because they have been widely explored in contemporary women's writing. Examining Nuala Ní Dhomhnaill's poetry from the prism of #MeToo, this paper discusses women's imprisonment and silence, their harassment and victimisation and the docility of their bodies in traditional patriarchal discourse. This paper aims to investigate how women are monitored physically and mentally in male-centred contexts, how they are molested in traditional societies and how they can be empowered by coming forward and telling their own stories. Neuroblastoma (NB) is a pediatric cancer of the sympathetic nervous system and one of the most common solid tumors in infancy. Amplification of MYCN, copy number alterations, numerical and segmental chromosomal aberrations, mutations, and rearrangements on a handful of genes, such as ALK, ATRX, TP53, RAS/MAPK pathway genes, and TERT, are attributed as underlying causes that give rise to NB. However, the heterogeneous nature of the disease—along with the relative paucity of recurrent somatic mutations—reinforces the need to understand the interplay of genetic factors and epigenetic alterations in the context of NB. Epigenetic mechanisms tightly control gene expression, embryogenesis, imprinting, chromosomal stability, and tumorigenesis, thereby playing a pivotal role in physio- and pathological settings. The main epigenetic alterations include aberrant DNA methylation, disrupted patterns of posttranslational histone modifications, alterations in chromatin composition and/or architecture, and aberrant expression of non-coding RNAs. DNA methylation and demethylation are mediated by DNA methyltransferases (DNMTs) and ten-eleven translocation (TET) proteins, respectively, while histone modifications are coordinated by histone acetyltransferases and deacetylases (HATs, HDACs), and histone methyltransferases and demethylases (HMTs, HDMs). Circular RNAs (circRNAs) played pivotal roles in the initiation and progression of cancers. CircRNA cut like homeobox 1 (circ-CUX1; hsa_circ_0132813) has been reported to contribute to neuroblastoma (NB) development by previous study. Furthermore, previous works reported that microRNA-16-5p (miR-16-5p) was down-regulated while doublesex and mab-3 related transcription factor 2 (DMRT2) was up-regulated in NB. The interaction and functional association between miR-16-5p and circ-CUX1 or DMRT2 were investigated in this study. Cell proliferation, cell cycle progression, colony formation, migration and invasion of NB cells were examined by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, flow cytometry, colony formation assay and transwell migration and invasion assays. The glycolysis was analyzed through measuring the consumption of glucose and the production of lactate and ATP. Dual-luciferase reporter assay, RNA immunoprecipitation (RIP) assay and RNA-pull down assay were utilized to confirm the interaction between miR-16-5p and circ-CUX1 or DMRT2. Tumor xenograft assay was performed to explore the function of circ-CUX1 in xenograft tumor growth in vivo. Circ-CUX1 promoted the proliferation, migration, invasion and glycolysis of NB cells. miR-16-5p was a direct target of circ-CUX1, and miR-16-5p overexpression-mediated effects in NB cells were partly alleviated by the introduction of circ-CUX1 overexpression plasmid. DMRT2 was a target of miR-16-5p in NB cells, and the introduction of anti-miR-16-5p overturned the influences of DMRT2 interference on the proliferation, migration and invasion and glycolysis of NB cells. Circ-CUX1 silencing restrained xenograft tumor growth in vivo. In conclusion, circ-CUX1 accelerated the proliferation, migration, invasion and glycolysis of NB cells through targeting miR-16-5p/DMRT2 signaling cascade. [ABSTRACT FROM AUTHOR]

Chronic spontaneous urticaria (CSU) is characterized by the presence of wheals, angioedema, or both for at least 6 weeks. It may persist for a long time—up to 50% of the patients have been reported to be symptomatic 5 years after the onset. Some patients can suffer more than one episode of CSU during their lifetime. Considering the recurrences, disabling symptoms, and significant impact on quality of life, proper and effective treatment of CSU is critical. The use of antihistamines (AHs) is still the mainstay of treatment. However, given the low rates of response to AHs (38.6% and 63.2% to standard doses and higher doses, respectively), the complete control of symptoms seems difficult to attain. The use of omalizumab for CSU has been a major breakthrough in the care of patients with CSU. However, the partial response and lack of response to omalizumab in a subgroup of patients, as high as 70% in some studies, make the development of alternative treatments desirable. Ever-increasing knowledge on the pathogenesis is making new target molecules available and enabling drug development for CSU. In addition to drug repurposing as in anti-IL-4/13, IL-5, and IL-17 antibodies, novel targeted therapy options such as ligelizumab and Bruton's tyrosine kinase inhibitors are currently undergoing clinical trials and will be available in the near future. This article reviews the current challenges in the treatment of CSU, the pathogenesis and potential target molecules, and the rationale for novel treatments and their rapidly developing status. Programmed death 1 ligand (PD-L1) is an immunomodulatory molecule expressed by cancer cells, and it has been widely demonstrated to inhibit host antitumor responses. The aim of the present study was to identify clinicopathological features associated with PD-L1 expression in the secondary solid cancers of patients after allogeneic hematopoietic stem cell transplantation. In this database of 530 patients who received allo-HSCT between 1990 and 2017, 15 developed solid cancers with a median interval of 3487 days after transplantation. Three patients had 2 different solid cancers. Eighteen solid cancer cases were identified. A multivariate analysis showed that chronic graft-versus-host disease (GVHD) was associated with an increased risk of solid cancer. The presence of chronic GVHD was observed in 8 out of 18 cases at the diagnosis of secondary malignancies. PD-L1 expression levels in cancers were significantly higher in patients with active chronic GVHD than in those without chronic GVHD (P = 0.020). Five cases of cancer that developed in the involved organs of chronic GVHD showed 30% or higher PD-L1 positivity. The present results revealed distinct PD-L1 expression in the secondary solid cancers of post-transplant patients with chronic GVHD. [ABSTRACT FROM AUTHOR]

Incidental capture by fisheries is one of the principal threats to sea turtles. This study analysed spatial and temporal patterns of sea turtle bycatch, and estimated the direct initial mortality rate of these animals, in the industrial double‐rig‐bottom trawl fishery in south‐eastern Brazil. This is also the first attempt to relate bycatch/at‐sea mortality in bottom trawling to stranded turtles found along the adjacent coast.The fishery was monitored from October 2015 to April 2018 through data collected voluntarily by the captains of eight industrial double‐rig trawlers. Two hundred and one sea turtles were captured during 9362 tows (43,657.52 trawling hours), resulting in a catch per unit effort (CPUE) of 0.0025 ± 0.0032 turtles h−1 with a standard net of 30.5 m headrope, with no significant difference between the estimated CPUEs for licensed shrimp and demersal fish trawlers.Caretta caretta (52.24%) and Lepidochelys olivacea (38.81%) were the most frequently captured species. According to Generalized Linear Models, C. caretta bycatch was significantly higher during winter, at lower latitudes (−24° to −23°) and higher longitudes (−42° to −40°), while the L. olivacea bycatch was significantly higher at higher latitudes (−23° to −21°). The direct initial mortality rate of sea turtles in the shrimp trawlers was 7.65 ± 3.85%. However, none of the dead individuals subsequently released with plastic tags (n = 10) were found stranded on the coast. Mortality was not significantly related to the depth or duration of the trawling.The results of this study suggest the need for improvements to the current management of the bottom trawl fishery in Brazil, moving from a species‐based to a spatial and seasonal‐based approach. Cryptosporidium spp. and Enterocytozoon bieneusi are two important zoonotic pathogens that can infect humans and a broad range of animal hosts. However, few studies have been conducted to study infection of the two pathogens in domestic geese until now. The aims of the present study were to determine the prevalence of natural infection, and the species or genotype distribution of Cryptosporidium and E. bieneusi in farm-raised and free-ranging geese from Hainan Province of China. In total, 266 fecal samples of geese were collected (142 farm-raised and 124 free-ranging geese). Cryptosporidium spp. and E. bieneusi were identified by nested PCR and sequencing analysis of the SSU rRNA and the ITS region of the rRNA genes. A total of 4.1% (12/226) of the geese were positive for Cryptosporidium spp., with 0.7% identified in the farm-raised geese and 7.0% in the free-ranging geese. Two bird-adapted species/genotypes were identified: C. baileyi (n = 1) and Cryptosporidium goose genotype I (n = 11). Meanwhile, E. bieneusi was found in 13.9% (37/266) of geese, with 8.9% identified in the farm-raised and 21.8% in the free-ranging geese. Eleven genotypes of E. bieneusi were identified constituted with six known genotypes: D (n = 13), I (n = 5), CHG2 (n = 1), CHG3 (n = 5), and CHG5 (n = 1), and five novel genotypes named HNE-I to V (one each). All of the genotypes identified in the geese here belonged to zoonotic Groups 1 or 2. This study is the first to demonstrate the presence of Cryptosporidium spp. and E. bieneusi in domestic geese from Hainan, China, and provides baseline data that will be useful for controlling and preventing these pathogens in goose farms.
