The aim of this study was to clarify the role of p62-dependent mitochondrial apoptosis in the initiation of monosodium urate (MSU) crystal-induced inflammation in macrophages. The induction of mitochondrial apoptosis in RAW 264.7 murine macrophages by MSU crystals was measured using western blotting and quantitative real-time polymerase chain reaction for Bax, caspase-3, caspase-9, or PARP1, and by flow cytometric analysis. Immunoprecipitation and western blotting was applied to detect ubiquitination of p62, TRAF6, and caspase-9. Mitochondrial apoptosis, reactive oxygen species (ROS) generation, and cell proliferation were assessed in cells transfected with p62 small interfering RNA (siRNA). Treatment of RAW 264.7 cells with MSU crystals induced activation of Bax, caspase-3, caspase-9, and PARP1 at the early phase, in addition to enhancing IL-1β expression, but these findings were attenuated at the late phase. MSU crystals induced ubiquitination of p62, followed by ubiquitination of TRAF6 and caspase-9, which were significantly reversed by ascorbic acid. RAW 264.7 cells transfected with p62 siRNA showed attenuated expression of Bax, caspase-3, caspase-9, and PARP1, decreased ROS and IL-1β production, and increased cell proliferation, compared to controls. The antioxidant ascorbic acid inhibited p62, caspase-9, and IL-1β expression increased by MSU crystals. p62 may be a crucial mediator for the mitochondrial apoptosis pathway in MSU crystal-induced inflammation, which is linked to the acute inflammatory response during the early phase of gout. Acyl coenzyme A long-chain 5 synthetase (ACSL5), a member of the ACSL family, plays a key role in fatty acid beta- oxidation and fat synthesis in mammals. In this study, two SNPs g.33185918G>A and g.33186348C>T in the 5' flanking sequence of the bovine ACSL5 gene were identified by PCR-RFLP. Thus, we aimed to evaluate the association between the two SNPs and meat quality traits, in addition to the carcass quality of Chinese Simmental-cross steers. The two SNPs were genotyped, and 36 carcass and meat quality traits were measured in a population of 354 individuals. Statistical analysis revealed that the two SNPs were significantly associated with carcass and meat quality traits. g.33185918G>A was very significantly (p<0.01) associated with the weight of the omasum and the pH of beef. For g.33186348C>T, the weight of the liver, hind hooves, kidney, and testis and the fat coverage rate of the carcass were very significantly associated (p<0.01). The present work indicated that polymorphisms in ACSL5 might be important genetic factors affecting meat quality and carcass yield in Chinese Simmental-cross steers. Moreover, such polymorphisms may serve as practical markers for carcass and meat quality traits in future marker-assisted selection (MAS) programs in beef cattle breeding and production. The objective of this study was to estimate genetic parameters for carnitine, creatine, creatinine, carnosine, and anserine concentration in LM and to evaluate their associations with Warner-Bratzler shear force (WBSF) and beef palatability traits. Longissimus muscle samples from 2,285 Angus cattle were obtained and fabricated into steaks for analysis of carnitine, creatine, creatinine, carnosine, anserine, and other nutrients, and for trained sensory panel and WBSF assessments. Restricted maximum likelihood procedures were used to obtain estimates of variance and covariance components under a multiple-trait animal model. Estimates of heritability for carnitine, creatine, creatinine, carnosine, and anserine concentrations in LM from Angus cattle were 0.015, 0.434, 0.070, 0.383, and 0.531, respectively. Creatine, carnosine, and anserine were found to be moderately heritable, whereas almost no genetic variation was observed in carnitine and creatinine. Moderate positive genetic (0.25, P < 0.05) and phenotypic correlations (0.25, P < 0.05) were identified between carnosine and anserine. Medium negative genetic correlations were identified between creatine and both carnosine (-0.53, P < 0.05) and anserine (-0.46, P < 0.05). Beef and livery/metallic flavor were not associated with any of the 5 compounds analyzed (P > 0.10), and carnitine concentrations were not associated (P > 0.10) with any of the meat palatability traits analyzed. Carnosine was negatively associated with overall tenderness as assessed by trained sensory panelists. Similar negative associations with overall tenderness were identified for creatinine and anserine. Painty/fishy was the only flavor significantly and negatively associated with creatinine and carnosine. These results provide information regarding the concentration of these compounds, the amount of genetic variation, and evidence for negligible associations with beef palatability traits in LM of beef cattle. Number of calvings at 53 mo (NC53) was proposed as an alternative selection criterion to improve productive longevity of Nellore cows. This study was carried out to estimate variance components for NC53 by assuming different models, so that the potential for using this selection criterion to improve fertility of Nellore cows could be assessed. Genetic correlations between NC53, number of calvings at 89 mo (NC89), and 2 selection indexes used in this breed were also estimated. The NC53 trait is moderately heritable (posterior mean heritability ≈ 0.17) and selecting for this criterion could improve productive longevity of Nellore cows. Greater response to selection is expected by fitting a threshold animal model for this trait, rather than a linear animal model. Greater accuracy of prediction for this criterion could be achieved by fitting a threshold-linear model, considering this trait and a selection index composed by traits evaluated at weaning and long-yearling.
