Abstract

FAM20C, a member of the family with sequence similarity 20, is involved in many physiological functions. Obesity, characterized by excessive accumulation of adipose tissue, has attracted more and more attention as a worldwide health problem. Here we generated adipocyte‐specific FAM20C knockout mice to investigate the role of FAM20C in adipose tissue expansion and obesity. Our results demonstrate that knockout mice are protected against high fat diet‐induced obesity, adiposity, and fatty liver disease. Additionally, knockout mice exhibited improved metabolic phenotypes, including enhanced glucose tolerance and insulin sensitivity compared with control mice. Furthermore, we observed reduced inflammatory infiltration and collagen deposition in the adipose tissues of knockout mice. Taken together, our results indicate that targeting FAM20C in adipocytes may be a promising strategy for the treatment of obesity and associated metabolic disorders. [ABSTRACT FROM AUTHOR]

Introduction: Diabetes and obesity are momentous risk factors threatening people's lives and health. Currently available incretin analogue glucagon-like peptide 1 (GLP-1) possesses huge hypoglycemic effect with the unsatisfactory effect of weight loss. Co-agonists targeting GLP-1R plus glucagon receptor (GCGR) or gastric inhibitory polypeptide receptor (GIPR) show synergistic benefits in glycaemic control and weight loss. Here, we describe a novel dual GIP and GLP-1 receptor agonist, DR10627, and performed a preclinical assessment of it. Methods: The agonistic ability of DR10627 was indirectly assessed by inducing cAMP accumulation in Chinese hamster ovary (CHO) cells transfected with GLP-1R or GIPR in vitro. Results: DR10627 had the capacity to activate both GLP-1R and GIPR in vitro. The terminal half-life of DR10627 was found to be approximately 4.19– 5.8 h in cynomolgus monkeys. DR10627 had a great improvement in oral glucose tolerance in SD rats. Moreover, DR10627 had a potent glucose-lowering effect in db/db mice, and the hypoglycemic effect of 18 nmol/kg DR10627 was better than that of 50 nmol/kg liraglutide. In addition, 10 and 30 nmol/kg DR10627 possessed the ability of potentiating the weight-loss, lipid-lowing efficacy and improving metabolism to a greater extent than 80 nmol/kg liraglutide. Conclusion: Preclinical assessment demonstrated that administration of DR10627 resulted in glucose lowering in SD rats and db/db mice, and substantial body weight reduction and metabolism improvement in DIO mice. Abstract

Three-dimensional (3D) layered tin oxide quantum dots/graphene framework (SnO2 QDs@GF) were designed through anchoring SnO2 QD on the graphene surface under the hydrothermal reaction. SnO2 QDs@GF have a 3D skeleton with a large number of mesopores and ultrasmall SnO2 QDs with a large surface area. The unique design of this structure improves the specific area and promotes ion transport. The mechanically strong SnO2 QDs@GF can directly be used as the anode of lithium-ion batteries (LIBs); it displays a high reversible capacity (1300 mA h g-1 at 100 mA g-1), excellent rate performance (642 mA h g-1 at 2000 mA g-1), and superior cyclic stability (when the current density is 10 A g-1, the capacity loss is less than 2% after 5000 cycles). This novel synthetic method can further be expanded for the production of other quantum dots/graphene composites with a 3D structure as high-performance electrodes for LIBs.

Abstract

Organic carbonyl polymers have been gradually used as the cathode in lithium-ion batteries (LIB). However, there are some limits in most organic polymers, such as low reversible capacity, poor rate performance, cycle instability, etc., due to low electrochemical conductivity. To mitigate the limits, we propose a strategy based on polyimide (PI)/graphene electroactive materials coated with reduced graphene oxide to prepare a flexible film (G@PI/RGO) by solvothermal and vacuum filtration processes. As a flexible cathode for LIB, it provides a reversible capacity of 198 mA h g−1 at 30 mA g−1 and excellent rate performance of 100 mA h g−1 at high current densities of 6000 mA g−1, and even a super long cycle performance (2500 cycles, 70% capacity retention). The excellent performance results in a special layer structure in which the electroactive PI was anchored and coated by the graphene. The present synthetic method can be further applied to construct other high-performance organic electrodes in energy storage.

Abstract

A new portable molecular imprinting polymer (MIP)-SERS nanoprobe is fabricated by a convenient electrochemical method. Single-layered MoS2 is electrochemically reduced on a screen-printed electrode as the scaffold. Functional monomers o-phenylenediamine (oPD), template theophylline (THP), and SERS-active Au nanoparticles (AuNPs) are then one-step electropolymerized on the scaffold. The morphology of the nanoprobe is found to be a three-dimensional and porous structure. The abundant AuNPs with the size of 45~50 nm are trapped within the growing MIP instead of being confined to the surface. The thickness of MIP film is calculated to 25.1 nm. The nanoprobe displays a strong SERS effect for THP using 532 nm as excitation wavelength with a detection limit (LOD) of 0.01 nM. The SERS peak intensity at 1487 cm−1 increases linearly with the concentration of THP in the range 0.1 nM to 0.1 mM. After the template is removed, the imprint-removed nanoprobe is generated for selective binding of THP. The re-binding kinetics study implies the portable MIP-SERS nanoprobe can reach the adsorption equilibrium within 8 min. Abstract

A nitrogen-doped carbon layer coating on a 3D graphene framework (NCL@GF) was produced by the in-situ polymerization of a uniform polyimide layer on the graphene framework surface and a carbonization process. The NCL@GF exhibited a 3D macroporous structure with uniform N-doped porous carbon layers and a high 5.4 at. % nitrogen content, which not only effectively enhanced the active sites but also facilitated fast ion and electron transport in the 3D pathways. Consequently, the NCL@GF as the anode of a sodium-ion battery (SIB) exhibited a discharge capacity of 357 mAh g-1 at 0.1 A g-1 after 200 cycles, a remarkable rate capability with a capacity of 122 mAh g-1 at 8 A g-1 , and a super long cycle life with a capacity retention of 70 % capacity after 2500 cycles at 0.5 A g-1 . Such performance is superior to that of previously reported carbon or graphene-based composites.

39. Aggregation Kinetics of TiO2Nanoparticles in Human and Artificial Sweat Solutions: Effects of Particle Properties and Sweat Constituents

Abstract

Dermal penetration potentials of titanium dioxide nanoparticles (TiO2NPs) may be affected by aggregation upon contact with sweat. This study investigated the aggregation kinetics of three TiO2NPs in thirty human sweat samples and four artificial sweat standards. Effects of particle concentration, sweat type, and inorganic (sodium chloride, disodium hydrogen phosphate, and sodium dihydrogen phosphate) and organic (l-histidine, lactic acid, and urea) constituents were examined. Three TiO2NPs remained colloidally stable in >20/30 human sweat samples and showed significant negative correlations (P< 0.01) between aggregation rates and |zeta potentials|. They aggregated rapidly over 20 min to >750 nm in three artificial sweat standards, while remained more stable in the International-Standard-Organization-pH-5.5 standard. Aggregation behaviors of three TiO2NPs mostly followed the Derjaguin–Landau–Verwey–Overbeek (DLVO) theory, allowing for determining their critical coagulation concentrations in inorganic constituents (15–491 mM) and Hamaker constants (3.3–7.9 × 10–21J). Higher concentrations of particles, inorganic constituents, and l-histidine destabilized three TiO2NPs, whereas urea inhibited aggregation. Three TiO2NPs adsorbed organic sweat constituents via complexation with amino or carboxyl groups, with isotherms following the Langmuir model. Correlation analyses further suggested that the adsorbed organic constituents may stabilize three TiO2NPs against aggregation in sweat by steric hindrance.
