8. MiR-449a regulates the cell migration and invasion of human non-small cell lung carcinoma by targeting ADAM10.

Abstract

Background: MicroRNAs (miRNAs) are non-coding small RNAs that have been shown to play a key role in the development of many tumors. However, its specific mechanism of action in non-small cell lung cancer (NSCLC) is not very clear. Purpose: This study was to identify the effect of miRNA-449a on NSCLC invasion and migration. Methods: We used quantitative real-time PCR experiments to demonstrate that miRNA-449a is down-regulated in NSCLC tissues and cell lines. We also used the Transwell assay to detect cell invasion and migration, and the Western Blot assay  was used to detect protein expression. The dual luciferase assay was used to detect the targeting relationship between miR-449a and A Disintegrin And Metalloproteinases 10 (ADAM10). Results: Our experiments demonstrated that miRNA-449a was down-regulated in NSCLC tissues and cell lines. When miRNA-449a was up-regulated in NSCLC cells, the invasion and migration ability of the cells was weakened, and the expression of ADAM10 was decreased. After down-regulation of miRNA-449a, the cell's invasion and migration ability was enhanced, and the expression of ADAM10 was increased. Through dual luciferase assays, we also found that miRNA-449a can target ADAM10 to delay the progression of epithelial-mesenchymal transition (EMT) and inhibit invasion and migration. Conclusion: Our experiments demonstrated that miRNA-449a acted as a tumor suppressor gene through inhibiting the expression of ADAM10 in NSCLC. [ABSTRACT FROM AUTHOR]

Abstract

Abstract: Submicron- and micron-sized lead iodide (PbI2) particles with well-controlled morphologies were successfully fabricated via a low-temperature hydrothermal process assisted by dodecyltrimethylammonium bromide (DTAB) as cationic surfactant. The as-synthesized powders were characterized by X-ray diffraction, scanning electron microscopy, transmission electron microscopy, and UV–vis spectroscopy. The effects of synthesis parameters (temperature, time, pH, and surfactant amount) were systematically investigated. The obtained results showed that the submicron structure was belt-like at 100–120°C, transformed to rod-like by increasing temperature to 140°C and it became a microtube-like at 160–200°C. By changing the pH of the synthesizing solution, it was found that a pure PbI2 phase could be obtained below 7. With the addition of increasing amount of surfactant, microparticles were converted to microrods→submicron belts→microtubes. The time-dependent experimental results revealed that the dissolution–recrystallization and dissolution–recrystallization–self-oriented-attachment were considered to be the possible mechanisms for the formation of the belt- and tube-like PbI2 submicron- and micron-sized particles, respectively. The optical properties of the PbI2 particles synthesized at 100–200°C for 8h under hydrothermal conditions were also studied. [Copyright &y& Elsevier]

Abstract

The Key to Chinese Civilization: The Explication and Exploration of Chinese Characters is a fascinating guide to the history of the Chinese civilization, which has been recorded not just by means of the Chinese characters but also in the characters themselves. It studies the long history of Chinese characters, the laws of their construction and development, and what their correct interpretation can mean for contemporary communication.
