Background: One bioassay for detecting acaricide resistance in livestock ticks is the adult immersion test (AIT), wherein engorged ticks are briefly immersed into a solution of a particular acaricidal compound and examined for mortality, their egg-laying capacity and offspring hatchability in vitro. Usually, the recommended label dose or an established discriminating dose of an acaricide is used to determine high mortality (≥ 95%) of susceptible tick strains. Such a test intends to detect the presence of resistance in a tick population. However, the adult immersion test does not directly translate the bioassay results to the predicted efficacy in the field. In this paper, we used the AIT as an initial screening bioassay supplemented with the resistance intensity test (RIT), a novel larval-based bioassay, wherein the resistance level can be determined and translated to the expected field efficacy. This was done by adopting World Health Organisation (WHO) guidelines for resistance detection in mosquitoes, which combines a 1 × recommended dose with 5 × and 10 × concentrated doses to reveal low, moderate and high resistance intensity, respectively. Methods: Engorged Rhipicephalus microplus ticks were collected from cattle at six different ranches across Rio Grande do Sul, Brazil, as part of the state's acaricide resistance surveillance program. Groups of adult ticks from each field collection were subjected to the AIT from each field sample. Additionally, engorged female ticks from each ranch were allowed to lay eggs, and their larval progeny aged 14 to 28 days were then used in the RIT. Deltamethrin and a combination of cypermethrin, chlorpyrifos and piperonyl butoxide were used in both tests, and the results were statistically analysed. Results: The in vitro efficacy of deltamethrin against adult ticks in the AIT ranged between 8.74% and 25.38%. The corresponding RIT results on their larval progeny indicated a high resistance level. In the immersion test, the in vitro efficacy of the combination of cypermethrin, chlorpyrifos, and piperonyl butoxide against adult ticks ranged between 49.31% and 100%. The corresponding RIT results on their larval progeny indicated a similar response ranging from fully susceptible to low or moderate resistance. The Pearson correlation coefficient (r = 0.883) showed a high correlation between tick mortality at the 1 × recommended concentrations of acaricides in both tests. Conclusions: The resistance intensity test is a valuable addition to the range of bioassays currently available for detecting acaricide resistance by determining the level of acaricide resistance. well known for its insecticidal, anti-inflammatory, antioxidant, anticancer, anti-rheumatic and antibiotic properties, Turmeric (Curcuma longa L.) Essential oil (EO), is effective in repelling and controlling many different pest species. The present study evaluates in vitro efficacy of turmeric EO against ectoparasitic tick, Rhipicephalus (Boophilus)annulatus collected from their natural host, domesticated cows. Repellency and toxicity of the EO on ectoparasitic ticks was analyzed with petri dish choice bioassay and AIT (Adult immersion test) against adult engorged females and nymphal stages of ticks. The Repellency Concentration (RC50) and Lethal Concentration (LC50) reproductive index, inhibition of oviposition and egg hatchability of treated ticks were estimated. GC-MS analysis identified Ar-turmerone as the major phytochemical, which induces adulticidal and repellant properties of EO. The nymphal stages of the ticks exhibited greater repellency on application of EO than the adult stage. Also, EO induced effective mortality in adult ticks. Further, this lethal concentration is capable of producing a decreased output in reproductive parameters namely, fecundity, % of oviposition and egg hatchability in surviving ticks thereby considerably reducing Rhipicephalus (Boophilus) R(B). annulatus population on its host. This study reveals the potency C. longa EO to be used as an effective pest control agent against R (B). annulatus infestation that can serve as an alternative to synthetic pesticides. A novel sanitizer tablet containing clove essential oil (CO) microemulsion was developed. A preformulation study using nuclear magnetic resonance and thermal analyses showed component compatibility. The main components of the samples remained intact despite a color change, probably due to a strong acid-base interaction between eugenol and diethanolamine. The CO microemulsion showed acaricidal and larvicidal activities superior to the commercial product, with product efficacy of 99.9% and larvae mortality of 94%. Optimal spray-drying conditions were achieved with inlet and outlet temperatures of 50°C and 40°C, respectively, an aspiration rate of 1 m3 min⁻1, and a 0.25 L h⁻1 injection flow. The feed suspension comprised 50% (v/v) liquid soap, 37.5% (v/v) water, 12.5% (v/v) ethanol, and 5.0% (w/v) silica. This formulation and processing parameters allowed for successful free-flow powder formation, providing a suitable matrix for incorporating the CO microemulsion via wet granulation without heating. Finally, sanitizer tablets produced from such granules resulted in a uniform product with low weight variation (coefficient of variation of 0.15%), eugenol content of 95.5% ± 3.3, and friability of 0.58%. Furthermore, the tablets showed rapid aqueous dispersion, forming a colloidal system with particle sizes of 221 nm and a zeta potential of -17.2 mV. Antimicrobial activity tests demonstrated the effectiveness of the sanitizer tablet against bacteria and fungi, exhibiting comparable antimicrobial potency to isolated CO. Hence, the sanitizer tablet developed represents a promising candidate as a practical and efficient solution for pest control, offering strong antimicrobial and acaricidal activity. Background: Acaricide resistance in cattle ticks is a significant concern in (sub)tropical regions, particularly Brazil. The Larval Packet Test (LPT) is the standard laboratory bioassay for resistance diagnosis, which requires triplicates of seven acaricidal dilutions plus controls to cover larval mortalities ranging between 0 and 100%. The value of the LPT lies in providing resistance ratios based on the ratio between the LC50 calculated with potentially resistant and susceptible ticks. However, LC50 ratios are difficult to translate into practical advice for farmers. Moreover, LPT requires laboratory facilities to maintain susceptible tick colonies, and it takes 6 weeks to obtain the larvae to be tested by LPT derived from engorged female ticks collected from cattle in the field. Our novel approach was twofold: first, we upgraded the LPT to the Resistance Intensity Test (RIT) by adopting the latest WHO guidelines for resistance detection in mosquitoes, which combines a 1 × recommended dose with 5 × and 10 × concentrated doses to reveal low, moderate and high resistance intensity, respectively. This reduced the number of test papers and tick larvae and, more importantly, provided relevant information on the resistance level. Our second innovative step was to abolish testing larvae entirely and expose partly engorged adult ticks to the same acaricidal doses immediately after removing them from cattle in the field. This resulted in the Rapid Tick exposure Test (RaTexT®), wherein partly engorged adult ticks were exposed to an acaricide-impregnated, specially designed matrix providing test results within 24 h. This approach directly compared resistance detection in tick larvae in the RIT with resistance in adult ticks in RaTexT®. Methods: Laboratory validation was conducted in Brazil with resistant and susceptible colonies of Rhipicephalus microplus ticks. For field validation, adult R. microplus ticks collected from different cattle farms in Brazil were evaluated for resistance to RaTexT®, and the results regarding their larval progenies were compared with those for the RIT. Partly engorged adult ticks derived from cattle infested with laboratory and field strains of R. microplus were exposed to deltamethrin in RaTexT® containers, which contained six rows of four interconnected compartments, accommodating five to eight semi-engorged female ticks with a preferred size ranging between 5 and 8 mm. The corresponding larvae of each strain were exposed in the RIT to the same deltamethrin concentrations in filter papers. Results: In RaTexT®, mortality in adult ticks from a resistant strain of R. microplus from Seropédica in Brazil was 38.4%, 54.2% and 75.0% at the 1 ×, 5 × and 10 × doses of deltamethrin, respectively. In RIT, mortality of larvae from the same resistant strain was 2.0%, 4.9% and 19.5% at 1 ×, 5 × and 10 × doses, respectively. The results of RaTexT® and RIT agreed since both tests identified a high level of resistance based on a cut-off of 90% mortality. In RaTexT®, mortality of adult ticks from a susceptible strain originating from Porto Alegre was 73.8%, 92.9% and 97.6% at the 1 ×, 5 × and 10 × doses, respectively. In RIT, mortality of larvae from the susceptible strain was 95.2%, 95.2% and 96.8% at the 1 ×, 5 × and 10 × doses, respectively. Interestingly, both tests identified a low number of unexpected resistant individuals in the susceptible strain since the mortality of neither larvae nor adults reached 100%. This effect remained unnoticed in the LPT, wherein a resistance ratio of 159.5 was found based on the LC50 of the resistant strain divided by the LC50 of the susceptible strain. Next, RaTexT® was compared with RIT using adult and larval ticks derived from three field strains of R. microplus in Brazil. RaTexT® detected high levels of resistance to deltamethrin in adult ticks in all strains, which was confirmed in larvae tested by the RIT. Both tests agreed on the same resistance level with significantly lower mortality rates in larvae than in adult ticks. Conclusions: RaTexT® is a novel rapid pen-site test for detecting acaricide resistance in adult livestock ticks. Purpose : Rhipicephalus (Boophilus) microplus is one the most significant ectoparasite in cattle farming in tropical and subtropical regions, causing problems to livestock health worldwide. The control of this ectoparasite primarily relies on the use of synthetic acaricides. However, the emergence of acaricide resistance has stimulated the search for new control alternatives, including phytocompounds with acaricidal and insecticidal potential. The aim of this study was to evaluate the acaricidal potential of Lavandula dentata essential oil against the engorged females of R. (B.) microplus. Methods: Engorged females were obtained from infested bovines in dairy farms in Pernambuco, Brazil. L. dentata essential oil was extracted, and adult immersion test assays were performed using the following oil concentrations: 0.2%, 0.4%, 0.6%, 0.8%, and 1%. Results : L. dentata essential oil at a concentration of 1% was lethal to all engorged females, and concentrations of 0.6% and 0.8% caused mortality of 98.6% and 99.1%, respectively. These concentrations disrupted the reproductive capacity of engorged females, reducing oviposition by more than 90% and preventing egg hatching by over 87%. Conclusion: The data revealed that L. dentata essential oil possesses effective pharmacological properties against R. Entomopathogenic fungi and eugenol have demonstrated great potential against ticks, particularly cattle tick Rhipicephalus microplus. In a pioneering study, Beauveria bassiana sensu lato (s.l.) CG 479 and eugenol were combined to assess their impact on the lipid profile and ovarian morphology of R. microplus. Germination tests revealed that regardless of concentration, the combination of B. bassianas.l. at 1 × 106 conidia ml-1 with eugenol at 0.18 mg ml-1 or 0.75 mg ml-1 exceeded 98%. Next, the engorged females were exposed by dipping to both agents individually or in combination. B. bassianas.l. combined with both eugenol concentrations reduced the reproductive parameters by 50%. For histopathology, tick females were exposed to fungus and eugenol, either alone or combined. The ovaries were extracted at 24 or 72 h and stained with hematoxylin and eosin. Groups exposed to the agents showed several modifications, including vacuolization of the pedicel and disorganization of the cytoplasm. The ovaries were also removed to determine the lipid profile by thin-layer chromatography. After 72 h of exposure, B. bassianas.l. combined with eugenol affected the levels of fatty acid and cholesterol. Furthermore, the reproductive parameters of the ticks were affected, which may explain the changes observed in the ovaries of R. microplus engorged females treated with both agents (combined or not). Purpose: Ticks infestation has a negative impact against human and animal health through blood sucking, transmission of blood-borne diseases and also caused economic losses. Methods: In the present study the adulticidal, ovicidal and larvicidal activity of d-limonene nanoemulsion (DLN) were evaluated against two tick species; Rhipicephalus annulatus and Rhipicephalus sanguineus. Nanoemulsion form of d-limonene was prepared, and its characteristics were evaluated using a UV spectrophotometer and zeta droplet size measurement. Acetylcholinesterase activity was determined. Results: The results revealed significant adulticidal effect with low LC50 and LC90 for d-limonene pure form (DL) against both adult tick spp. (R. annulatus and R. sanguineus) ((0.958 and 1.559%) and (2.26 and 3.51%), respectively). DLN LC50 and LC90 values were ((1.277 and 2.396) and (3.97 and 7.28), respectively) against R. annulatus and R. sanguineus, respectively. DL and DLN showed significant ovicidal effect against R. sanguineus at high concentrations (10 and 5%). In larval packet test, LC50 and LC90 values of DL were ((1.53 and 2.22%) and (6.81 and 12.07%), respectively) against R. annulatus and R. sanguineus, respectively, while LC50 and LC90 values of DLN were ((6.48 and 11.26%) and (7.82 and 13.59%), respectively) against R. annulatus and R. sanguineus, respectively. Significant acetylcholinesterase inhibition percentage was detected for both ticks spp. which treated by DL and DLN. Alternative treatments to control ticks are being increasingly investigated, including natural products to decrease parasitic resistance and environmental impact. The present study investigated the chemical profile of eucalyptus wood vinegar and evaluated its effect on the reproduction of Rhipicephalus sanguineus. Eucalyptus wood vinegar (WV) was tested at several concentrations. For the experiment, 500 engorged females were collected from canine hosts. The female ticks were distributed by weight in groups of ten per WV concentration, with five replicates per treatment. For all tests, the ticks were immersed for 5 min in a solution with each WV concentration. The positive control was amitraz and negative was water. The major components of the WV's organic fraction were furfural and phenolic compounds. A significant decrease in egg hatching was observed in the WV concentration range from 65 to 1,000 mg mL−1. When parameters such as the percentage of hatching, female weight, and egg mass were evaluated, the efficiency of the product was verified as being between 84.97% and 99.38% in the concentration range of 65 to 1,000 mg mL–1, respectively. Hyalomma dromedarii is the predominant tick species parasitizing camels in Egypt which leads to mortalities in young animals that result in economic losses. It can transmit a lot of pathogens to animals and humans, such as the Crimean-Congo hemorrhagic fever virus, the Dhori virus, Kadam virus, Theileria annulata and spotted fever rickettsia. The continuous use of chemical acaricides has negative impact on the environment and almost led to acaricidal resistance, and hence the plant extracts represent alternative methods for controlling ticks. The present study was carried out to assess the histopathological effects on the ovary of fed female Hyalomma dromedarii following immersion in the ethanolic extract of fruits of Citrullus colocynthis (100 mg/mL). Light, scanning and transmission electron microscopy observations provided evidence that Citrullus colocynthis caused extensive damage to oocytes. Destruction of the internal organelles of oocytes, along with delay and/or inhibition of vitellogenesis were demonstrated. This is the first histological study that points to damage in H. dromedarii ovaries following treatment with the ethanolic extract of fruits of C. colocynthis. The data presented suggest that the plant extract affects the ovary either directly by entering the oocytes and/or indirectly by damaging the gut cells and digestion of blood that interfere with the development of oocytes, so it can be used as a promising agent for tick control. Cattle tick control poses a significant challenge for livestock in tropical and subtropical regions. The objective of this study was to determine the most suitable timing to initiate a strategic tick control program and to identify the ideal number of acaricide treatments for adult taurine cattle (Bos taurus taurus) in a tropical region throughout the year. Three groups with 10 bovines each were performed: T01 (strategic treatment in late autumn/winter/late spring, every 28 days), T02 (strategic treatment to act in the "first tick generation" – early spring/summer/early autumn, every 28 days) and T03 (control). Tick counts (females 4–8 mm) were conducted every 14 days. If the tick burden in any group reached 30 or more during these counts, we applied an additional treatment. Over the course of a year, T02 required significantly fewer (p < 0.05) acaricide treatments than T01, with nine treatments for T02 and eleven for T01. Furthermore, during the tick counts, animals in T02 showed a lower tick burden compared to those in T01. Initiating the strategic tick control program in early spring, corresponding to the first tick generation, proved more effective than starting in autumn. This approach not only required fewer acaricide treatments but also resulted in a reduced tick burden. Nanoparticles have been shown to inhibit major life cycle stages of ticks, indicative of the promising application of nanomaterials against hard ticks. The study thus probed into one of the alternative options to curtail Hyalomma by employing nanocomposites consisting of pyrethroids (cypermethrin and deltamethrin) coated nanoparticles of iron oxides and iron sulfides keeping alongside the evaluation of their toxicity through plant and mammalian cell lines. The nanoparticles used in this study were roughly spherical in morphology and exhibited various size dimensions upon characterization using SEM, EDX, and FTIR. The application of nanomaterials on female ovipositioning tick showed a decline up to 15% (females ovipositioned) in deltamethrin-coated FeO NPs, whereas this decline was up to 18% in Cyp-FeS NPs and up to 5% in Cyp-FeO NPs. Similarly, the larval hatching was also impacted, leading to a hatching percentage of 5% and only 1% by application of Cyp-FeS NPs and Cyp-FeO NPs, respectively. Similarly, the larval groups had LC90 of 4.1 and 4.73 mg/L for the Cyp-FeO NPs and Cyp-FeS NPs groups. The delta-FeO NPs and delta-FeS NPs demonstrated a promising effect against adult ticks, showing LC50= 3.5 mg/L, LC90= 6.7 mg/L and LC50= 3.8 mg/L, LC90= 7.9 mg/L, respectively. MTT assay revealed that the pyrethroids coupled with iron oxide nanoparticles showed the least cytotoxicity even at the highest concentration (10–1 µL) among other nanomaterials. Background: The present study aimed to evaluate the effects of different treatment strategies using a new commercial formulation containing pour-on fluralaner on Rhipicephalus microplus infestation in cattle and in pastures in a tropical climate region where up to five generations of this tick species can occur per year. Methods: Forty-five naturally infested cattle were divided into three experimental groups: T01, treated with fluralaner (2.5 mg/kg) pour-on every 42 days; T02, the cattle received the first treatment with fluralaner on Day 0 but the next treatment involved a weekly visual evaluation; T03, control, received palliative treatment with a spray formulation when the group mean was ≥ 30 ticks. Counts of female R. microplus were performed weekly until day 343, and larval counts on pasture were performed on Days 0, 30, and 60 and every 30 days until Day 330. Results: Using fluralaner, six applications were performed in Group T01, and four were performed in Group T02. In the control group (T03), it was necessary to perform eight palliative acaricide treatments with the spray formulation. The animals in T01 and T02 showed lower mean tick counts (p ≤ 0.05) than the control group (T03) on 28 and 27 of the 49 evaluated dates, respectively. In the paddock where the animals were kept as controls, the R. microplus larvae counts increased to 1458. In the paddocks where the animals were treated with fluralaner, the number was ≤ 19 per paddock during the study. Conclusions: The different strategic treatment protocols performed with pour-on fluralaner (2.5 mg/kg) over a year in taurine cattle in a tropical region with a history of up to five annual generations of cattle ticks were effective, maintaining levels of R. microplus infestations in animals and in pastures close to zero in most of the study. Depending on the retreatment criterion adopted, the number of applications per year may be lower, resulting in a reduction in the mean cost of acaricide treatment per year and lower exposure of R. microplus populations to the active ingredient, resulting in lower resistance and selection pressure. To develop an appropriate program for essential oils (EOs) used in pest control, it is important to acquire information about the effect of the EO when used at a low concentration. Previously, we demonstrated the enzymatic activity of Haemaphysalis longicornis (the main livestock pest in Australia and New Zealand) exposed to Cinnamomum cassia EO and (E)-cinnamaldehyde (its major compound) when applied at 50% lethal concentration (LC50). However, H. longicornis respond to toxic substances via different mechanisms and at different times of exposure. Herein, the enzymes involved in the defense response were studied by monitoring glutathione-S-transferase, acetylcholinesterase, and Na+/K+-adenosine triphosphatases (ATPase) activities in H. longicornis larvae exposed to C. cassia EO and (E)-cinnamaldehyde across different times. Further, the molecular changes related to the defense mechanisms were studied by monitoring adenosine triphosphate (ATP)-binding cassette (ABCB10) gene expression. In addition, we investigated the acaricidal activity of the test substances against the engorged larvae and nymphs of H. longicornis. Our results showed that the activities of these enzymes were significantly affected by the EO and/or (E)-cinnamaldehyde depending on the exposure time. We observed up-regulation of the ABCB10 gene in the larvae. Also, our results revealed that C. cassia EO (LC50 = 1.46 and 16.69 mg/mL) and (E)-cinnamaldehyde (LC50 = 3.86 and 17.96 mg/mL) were effective against the engorged larvae and nymphs, respectively. Our results provided a deeper understanding of the defense mechanisms of H. longicornis, which may provide an important clue for the development of EO-based acaricides that are cheaper and environmentally friendly. The current study examined the impact of ultraviolet (UV)‐B radiation in Metarhizium pingshaense blastospores' photolyase expression and their virulence against Rhipicephalus microplus. Blastospores were exposed to UV under laboratory and field conditions. Ticks were treated topically with fungal suspension and exposed to UV‐B in the laboratory for three consecutive days. The expression of cyclobutane pyrimidine dimmers (CPDs)‐photolyase gene maphr1‐2 in blastospores after UV exposure followed by white light exposure was accessed after 0, 8, 12, 24, 36, and 48 h. Average relative germination of blastospores 24 h after in vitro UV exposure was 8.4% lower than 48 h. Despite this, the relative germination of blastospores exposed to UV in the field 18 h (95.7 ± 0.3%) and 28 h (97.3 ± 0.8%) after exposure were not different (p > 0.05). Ticks treated with fungus and not exposed to UV exhibited 0% survival 10 days after the treatment, while fungus‐treated ticks exposed to UV exhibited 50 ± 11.2% survival. Expression levels of maphr1‐2 8, 12, and 24 h after UV‐B exposure were not different from time zero. Maphr1‐2 expression peak in M. pingshaense blastospores occurred 36 h after UV‐B exposure, in the proposed conditions and times analyzed, suggesting repair mechanisms other than CPD‐mediated‐photoreactivation might be leading blastospores' germination from 0 to 24 h. Rhipicephalus sanguineus sensu lato 'tropical lineage' (Acari: Ixodidae) is considered a sanitary concern due to its role as a disease vector. Tick strains resistant to synthetic acaricides have caused difficulties in their control, besides synthetic acaricides are harmful to the environment and to the health of non-target animals. The research of plants with acaricidal and repellent properties has proved to be an efficient alternative in tick control. The genus Tagetes spp. excels for its use as traditional pest control in households and plantations and also for its potential as an acaricide against R. sanguineus under laboratory conditions. The first aim of the present study was to evaluate the effect of different doses of Tagetes minuta essential oil (TMEO) on the central nervous system (synganglion) in unfed R. sanguineus adults. The histological analysis of synganglion exposed to the different concentrations of TMEO and amitraz 12.5% (50% of the recommended dose in the package insert) showed a significant effect with signs of cell damage including volume increase, loss of shape, and vacuolization, in addition to chromatin alterations such as condensation, margination, and fragmentation. TMEO were analyzed by gas chromatography coupled with mass spectrometry showing the presence of 21 compounds that according to their chemical structure are classified as terpenoids. The current work evaluated the efficacy of 10 commercial acaricides in different pHs (4.5, 5.5, and 6.5) in laboratory (adult immersion tests (AIT), pH evaluation over time) and field assays (tick counts and efficacy). In the AIT (n=70), higher efficacies were obtained when the acaricide emulsion had a more acidic pH (4.5), mainly for two combinations of pyrethroids + organophosphate (acaricide 3 and acaricide 9). For amidine, a higher pH (6.5) showed a higher efficacy. Over time, there was a trend in the pH of these emulsions increasing. When the efficacy of chlorpyrifos + cypermethrin + piperonyl butoxide (acaricide 3) at different pHs was evaluated over time (0, 6, 12, and 24h) by AIT, the less acidic pH (6.5) showed a strongly variation in the acaricide efficacy range. The mean pH of the water samples from different regions of Brazil was 6.5. In the field, the association of pyrethroid + organophosphates (acaricide 9) with pH of 4.5 and 5.5 were more effective in tick control than the emulsion prepared with this same spray formulation at pH 6.5. The pH of the acaricide emulsions is an important point of attention and is recommended that the veterinary industry start to develop/share information regarding how the pH can affect the acaricide efficacy. The cattle tick Rhipicephalus (Boophilus) microplus is a major problem of concern for cattle industry in tropical and subtropical areas. Control of cattle tick is based mainly on the use of chemical acaricides, which has contributed to the emerging problem of selection of resistant tick lineages. Plants have been used as an alternative to conventional acaricidal drugs. On the other hand, the acaricidal activity of hydroethanolic extract of Randia aculeata seed (EHRA) has been demonstrated against R. microplus under laboratory conditions. However, the utility of EHRA seed as a potential acaricidal needs to be determined under field conditions. For this reason, the aim of this study was to evaluate the efficacy of the EHRA against R. microplus sprayed on naturally infested calves, determine the effect of the EHRA seed on acetylcholinesterase activity in R. microplus larval and identify the chemical composition of EHRA. Forty-five male calves were divided in three groups and treated with: G1 water; G2 EHRA 20% w/v and G3 coumaphos 0.2% v/v. Acetylcholinesterase (AChE) activity in R. microplus larvae was determined by a colorimetric assay. The chemical composition of EHRA was accessed through HPLC/MS. Significantly fewer ticks were observed after 24 h on the treated group compared to control group. EHRA significantly inhibited in vitro AChE activity in R. microplus at all tested concentrations. Chlorogenic acid, vanillinic acid, p-coumaric acid, caffeic acid. rutin, quercetin, (-)-epicatechin, 4-hydroxybenzoic acid, quercetin, vanillin, 2,4-dimethoxy-6-methylbenzoic acid, scopoletin and ferulic acid were identified in the extract. The results provided new data for the elucidation of the mechanisms of EHRA acaricide action and to further evaluate the use as a new alternative control agent against R. microplus under in vivo conditions. Three ticks, indigenous to the United States, were assessed for their ability to maintain and transmit African swine fever virus (ASFV). Amblyomma americanum and A cajennense adults and nymphs maintained virus for 4 to 7 days after engorging on infected swine blood; however, virus was not carried through the subsequent molt of nymphs and it was not transmitted vertically to the eggs and larvae of the infected females. On refeeding of the ticks, infection was not transmitted to healthy swine. Ornithodoros coriaceus adult females also did not show transovarial transmission of two strains of ASFV. Nymphs, however, maintained virus for 77 to 118 days through a molting stage and transmitted one strain of the virus (Lisbon 60) to healthy swine.

In South Africa, acaricides are widely used for tick control but very few reports are available on resistance development of Rhipicephalus (Boophilus) decoloratus Koch to chemical control on commercial farming systems in Africa, south of the Sahara. Resistance to different acaricide classes reported over the years was mostly from localised communal farming systems. This report addresses the lack of available information on resistance development by reviewing results found during a National Tick Resistance Survey carried out from 1998 to 2001, laying the foundation for more recent research on resistance development, and the evolution of resistance over the years. One hundred and eighty R. decoloratus populations were randomly collected from commercial farming systems, covering most of the provinces of South Africa. Larval immersion tests were used to determine phenotypic resistance for each tick population and 6.6% of the populations tested were found to be resistant to amitraz, 35.5% to cypermethrin, and 36.1% to chlorfenvinphos. Multi-resistance to all three acaricides was found in 1.2% of populations and a further 25.8% of the populations were resistant to two acaricides. The detection of resistance of Rhipicephalus (Boophilus) species to currently used or new acaricides is an essential tool in resistance management. The acaricides tested for the resistance of R. decoloratus during the survey are currently still in use in South Africa and these historical results, never published before, can be valuable and can act as reference data to determine the evolution of resistance development to acaricides in more recent studies. Rhipicephalus microplus is the main ectoparasite of cattle and its parasitism can reduce weight gain of hosts, in addition to causing anemia, increasing the risk of myiasis, and resulting in the transmission of Babesia bovis and Anaplasma marginale, among other pathogens. The use of synthetic chemicals plays an important role in controlling these ticks. However, its frequent and indiscriminate use has contributed to the selection of resistant strains, resulting in greater interest in the search for natural-origin products. The shrub Callistemon viminalis (Myrtaceae), also known as weeping bottlebrush, is known for its antibacterial, acaricidal, repellent, and antifungal activities, but there are no reports in the literature about its effects on the internal morphology of ticks. This study aimed to extract and characterize the essential oil obtained from the leaves of C. viminalis. Additionally, its effects on the ovary morphology of engorged R. microplus were assessed through histological, histochemical, and morphometric techniques. Exposure to C. viminalis caused dose-dependent morphological changes, such as cellular alterations in the epithelial layer lining the ovary lumen and the pedicel, irregularity of the chorion and oocyte shape, changes in protein and carbohydrate content, decrease in oocyte size, reduction in the size of the nucleus as well as cytoplasmic and nucleolar vacuolation. Thus, C. viminalis essential oil exhibited a toxic effect on the reproductive system of R. microplus, which may result in the reproductive impairment of this tick species. [ABSTRACT FROM AUTHOR]

Is well known the taurine and zebuine susceptibility to Rhipicephalus microplus. Few are the reports regarding tick population dynamics between the same herd/breed, and because of this, two experiments were performed. In the 1st, the cattle tick population dynamics in dairy nursing calves (reared collective and individually), weaning calves (4–16 months), heifers (17–29 months), cows in lactation and dry cows (≥ 30 months) from the same herd, tick burden and milk production correlation were performed, for two years. R. microplus females (4.5-8.0 mm) counts and the milk production were performed every 28 and 14 days, respectively. In the 2nd experiment, bovines belonging to different categories/age (newborn without previous contact with tick; 12–13 months with tick contact since birth; and 23–24 months with tick contact since birth) were experimentally infested with 30,000 R. microplus larvae, to quantify the number of fully engorged females detached from these animals. In the 1st experiment, when the mean counts of tick were ≥ 30 all animals of the group were treated. Nursing calves showed 3–4 peaks of ticks, animals reared individually showed smaller (p ≤ 0.05) tick burden than those reared collectively. Weaning calves (4–8 months) showed 5 tick peaks/year and higher mean tick burden was found than other categories. On the other hand, animals with 17–29 months of age showed smallest (p ≤ 0.05) tick burden, with 3 tick peaks/year. When the animal become lactating the tick burden increase, and 5 peaks/year occurred, and decrease again in dry cows (p ≤ 0.05) showing 4–5 tick peaks/year. Weaning calves and lactating cows received more acaricide treatments (p ≤ 0.05), 18 and 15, respectively. Nursing calves reared individually, and heifers (21–29 months) were the categories that received two acaricide treatments. The more milk the cow produce, more ticks it has (p ≤ 0.05). In the 2nd experiment, more (p ≤ 0.05) fully engorged females were recovered from younger animals than older ones. So, different tick control strategies need to be adopted in different dairy cattle categories, and the tick burden should be considered, once the effect may be more inherent to the animal rather than the strategy adopted. The cattle fever tick, Rhipicephalus (Boophilus) microplus, is the most economically important tick worldwide. Infestations with this tick can lead to direct damage and cattle mortality due to the transmission of potentially deadly pathogens. Management of this tick species has been focused on the use of synthetical acaricides; however, the emergence of acaricide resistance to single or multiple active ingredients has resulted in a need for novel acaricide compounds. Among potential avenues for the discovery of novel acaricides are plant-derived compounds. The efficacy of five organic compounds (nootkatone, Stop the Bites®, BioUD®, lavender oil, and cedarwood oil) was evaluated using larval immersion tests (LITs), repellency assays, and adult immersion tests (AITs). The results from the LITs indicate that three of the organic compounds (NootkaShield™, Stop the Bites, BioUD) led to significant mortalities at low concentrations (0.2, 0.02, and 0.08%, respectively). By comparison, lavender and cedar oil led to around 90% mortality at 10 and 1% concentrations, respectively. Similarly, NootkaShield, Stop the Bites, and BioUD had strong repellent properties with over 90% repellency at the two highest concentrations tested. Using the FAO 2004 guidelines, we evaluated the effectiveness of these organic compounds at reducing the fecundity of R. (B.) microplus and show that Nootkatone, Stop the Bites, and BioUD may significantly decrease tick populations (Drummond's index > 90% at concentrations of 5%), highlighting their potential as alternatives to synthetic acaricides for the control of cattle fever ticks. Background Odocoileus virginianus (the white-tailed deer) is a key reproductive host for medically important tick species, including Ixodes scapularis and Amblyomma americanum. Orally administering a systemic acaricide to whitetailed deer has the potential to reduce tick reproduction, abundance and pathogen-infected tick bites. Prior studies have demonstrated considerable efcacy of a low-dose fpronil mouse bait in controlling larval I. scapularis parasitizing the pathogen reservoir, Peromyscus leucopus. No prior studies have evaluated the efcacy of a fpronil product in controlling ticks parasitizing white-tailed deer. Methods A pen study was conducted to evaluate the efcacy of a fpronil deer feed in controlling I. scapularis and A. americanum adult ticks. Individually housed deer (n=24) were exposed to deer feed containing 0.0025% fpronil (fpronil deer feed) for 48 h and 120 h, and a control group of deer were exposed to an untreated placebo. On postexposure day 7 and day 21, all deer were parasitized with 20 mating pairs of feeding capsule-enclosed I. scapularis and A. americanum. Post-attachment, engorgement and mortality of ticks were recorded. The concentrations of fpronil in plasma, feces and tissues from euthanized deer were estimated using liquid chromatography-mass spectrometry. Results The fpronil deer feed efcaciously controlled ticks parasitizing pen-reared white-tailed deer. Efcacy in reducing survivorship of blood-feeding female I. scapularis exceeded 90% in all instances except for when ticks parasitized 48-h treated deer at day 21 post-exposure (47.2%). Efcacy in reducing survivorship of A. americanum females exceeded 80% in all instances. In the 120-h exposure group there was 100% tick mortality at day 7 post-exposure for both tick species. A signifcant correlation was observed between reductions in tick survivorship and concentrations of fpronil sulfone in plasma. The results of tissue analysis suggest that a withdrawal period may be needed to allow for fpronil degradation prior to hunting season. Conclusions The results provide proof-of-concept for the use of a fpronil-based oral acaricide in controlling two medically important tick species infesting a key reproductive host. A feld trial is necessary to confrm the efcacy and toxicology of the product in wild deer populations. In Brazil, two tick species are mainly involved in public health and livestock issues: Amblyomma sculptum and Rhipicephalus microplus, respectively. Nootkatone, a sesquiterpenoid isolated from Chamaecyparis nootkatensis, has been described in the literature as an environmentally friendly alternative method of control, with repellent and toxic action against different insect and tick species from the Northern Hemisphere. The objective of the present study was to evaluate the effect of nootkatone on the in vitro mortality of R. microplus and A. sculptum. Different dilutions of nootkatone (0.5, 1, 2, 5 and 10%) were used on instars of both tick species. In A. sculptum engorged larvae, unfed nymphs, and unfed adults, 2% nootkatone resulted in 95% mortality in almost all tests. An in vitro mortality rate of 95% was observed for larvae and engorged females of R. microplus with 1% and 0.5% nootkatone, respectively. Additionally, 5% and 10% nootkatone were at least 97% effective against engorged larvae, unfed nymphs, and unfed adults of A. sculptum and larvae and engorged females of R. microplus. This was the first study to use nootkatone against ticks from Brazil and the data suggest that nootkatone has great potential as a tool for tick control. [ABSTRACT FROM AUTHOR]

Repellent and acaricidal activities of essential oils (EO) extracted from common yarrow (Achillea millefolium L.) and main chemical components were evaluated against Ixodes scapularis and Dermacentor variabilis adult ticks and nymphs. Flowers and leaves were collected from two locations, Harvest Moon trail (HMT) and Port Williams (PW) in Nova Scotia (Canada), and EO were extracted via hydro-distillation. Samples were analyzed using GC-MS, and differences in chemical composition and quantity of compounds detected were reported in relation to the collection site and plant parts. EO were both rich in germacrene D (HMT EO 21.5 ± 1.31% wt; PW EO 25.5 ± 0.76% wt); however, HMT flower EO has a higher concentration of camphor (9.9 ± 0.08% wt) compared to PW flower EO (3.0 ± 0.01% wt). Significant acaricidal activity was reported against I. scapularis adult ticks, particularly for HMT flower EO with a LD50 of 2.4% v/v (95% confidence interval = 1.74–3.35) at 24 h post-exposure. Germacrene D had the lowest LD50 of 2.0% v/v (95% CI 1.45–2.58) among the four compounds after 7 days. No significant acaricidal effect was observed on D. variabilis adult ticks. Yarrow PW flower EO exerted repellent activity towards I. scapularis nymphs (100% repellency up to 30 min); however, repellency significantly declined over time. Yarrow EO exert promising acaricidal and repellent properties, that may be used to manage Ixodes ticks and the diseases they vector. [ABSTRACT FROM AUTHOR]
